In the steel construction industry, e.g. production of windmill sections, welding accounts for a main part of the manufacturing costs and resource consumption. Moreover, social issues attached to welding involve working in dangerous environments. This aspect has unfortunately been neglected so far, in light of a predominant focus on economics combined with a lack of suitable assessment methods. In this chapter, exemplary welding processes are presented that reduce the environmental and social impacts of thick metal plate welding. Social and environmental Life Cycle Assessments for a thick metal plate joint are conducted for the purpose of expressing and analysing the social and environmental impacts of welding. Furthermore, it is shown that state-of-the-art technologies like Gas Metal Arc Welding with modified spray arcs and Laser Arc-Hybrid Welding serve to increase social and environmental performance in contrast to common technologies, and therefore offer great potential for sustainable manufacturing. The application of high strength steels allows smaller wall thicknesses compared to conventional steels. Therefore, the risk of buckling has to be considered carefully. Due to the lack of knowledge concerning the residual stresses present after welding in high strength steel components conservative assumptions of their level and distribution is typically applied. In this study I-girders made of steels showing strengths of 355 MPa and 690 MPa were welded with varying heat input. Due to the dimension of the I-girders and the complex geometry the accessibility for residual stress measurement using X-ray diffraction was limited. Therefore, saw cutting accompanied by strain gauge measurement has been used to produce smaller sections appropriate to apply X-ray diffraction. The stress relaxation measured by strain gauges has been added to residual stresses determined by X-ray diffraction to obtain the original stress level and distribution before sectioning. The combination of both techniques can produce robust residual stress values. From practical point of view afford for strain gauge application can be limited to a number of measuring positions solely to record the global amount of stress relaxation. X-ray diffraction can be applied after sectioning to determine the residual stresses with sufficient spatial resolution. The wide diversity of compounds that can act as compatible solute complicates the task of understanding the different strategies that halophilic bacteria use to cope with salt. This is specially challenging when attempting to go beyond the pathway that produces a certain compatible solute towards an understanding of how the metabolic network as a whole addresses the problem. Metabolic reconstruction based on genomic data together with Flux Balance Analysis (FBA) is a promising tool to gain insight into this problem. However, as more of these reconstructions become available, it becomes clear that processes predicted by genome annotation may not reflect the processes that are active in vivo. As a case in point, E. coli is unable to grow aerobically on citrate in spite of having all the necessary genes to do it. It has also been shown that the realization of this genetic potential into an actual capability to metabolize citrate is an extremely unlikely event under normal evolutionary conditions. Moreover, many marine bacteria seem to have the same pathways to metabolize glucose but each species uses a different one. In this work, a metabolic network inferred from genomic annotation of the halophilic bacterium Halomonas elongata and proteomic profiling experiments are used as a starting point to motivate targeted experiments in order to find out some of the defining features of the osmoregulatory strategies of this bacterium. This new information is then used to refine the network in order to describe the actual capabilities of H. elongata, rather than its genetic potential. Here we evaluate the extent of agreement between classification by electron microscopy (EM) and classification by VSSA on a large set of diverse particulate substances that represent all the anticipated challenges except mixtures of different substances. EM and VSSA are determined in multiple labs to assess also the level of reproducibility. Based on the results obtained on highly characterized benchmark materials from the NanoDefine EU FP7 project, we derive a tiered screening strategy for the purpose of implementing the definition of nanomaterials. We finally apply the Screening strategy to further industrial materials, which were classified correctly and left only borderline cases for EM. On platelet-shaped nanomaterials, VSSA is essential to prevent false-negative classification by EM. On porous materials, approaches involving extended Adsorption isotherms prevent false positive classification by VSSA. We find no false negatives by VSSA, neither in Tier 1 nor in Tier 2, despite real-world industrial polydispersity and diverse composition, shape, and coatings. The VSSA screening strategy is recommended for inclusion in a technical guidance for the implementation of the definition. Here the correlation between the number of initiation sites and the applied stress level has been also investigated. The criteria have been implemented in in-house software, which allows multiple fatigue crack propagation, and validated against selected experimental tests. The results have shown that the modelling of multiple crack propagation and interaction is crucial for the prediction of the fatigue strength of weldments, both in finite and infinite life regime. Such metabolic specialization could be driven by stochastic gene expression and could provide individual cells with growth benefits of specialization. We measured the degree of phenotypic specialization in two parallel metabolic pathways, the assimilation of glucose and arabinose. We grew Escherichia coli in chemostats, and used isotope-labeled sugars in combination with nanometer-scale secondary ion mass spectrometry and mathematical modeling to quantify sugar assimilation at the single-cell level. We found large variation in metabolic activities between single cells, both in absolute assimilation and in the degree to which individual cells specialize in the assimilation of different sugars. Analysis of transcriptional reporters indicated that this variation was at least partially based on cell-to-cell variation in gene expression. Metabolic differences between cells in clonal populations could potentially reduce metabolic incompatibilities between different pathways, and increase the rate at which parallel reactions can be performed. The influence of the load type on CF behavior of the alloy was examined by applying push/pull and rotating bending cyclic load to failure. The hour-glass shaped specimens with both polished and precisely turned surface finish were tested in corrosion chambers allowing for the permanent circulation of Northern German Basin electrolyte heated to 369 K. Additionally, the influence of a protective cathodic potential on the CF life expectancy was investigated by employing a potentiostat. The rotating bending cyclic load led to a higher CF life expectancy, which decreased with increasing duration of the experiment due to a higher deterioration rate of the material. The application of a cathodic potential caused a significant increase of the CF lifespan from 4.7 x 105 (open-circuit potential (OCP)) to 107 cycles (preset threshold cycle number) for the potential range from USHE = – 450 to – 900 mV. Microstructural analysis of the CF damage revealed horizontal grain attack within corrosion pit cavities, multiple fatigue cracks and preferable deterioration of the austenitic phase. Therefore, the Arrhenius equation is widely used due to its simple form and often leads to suitable results. Where, the Arrhenius equation presents a linear relation for the temperature-dependent shift factor in logarithmic scale ln(α) with the absolute inverse temperature (1/ϑ). However, in cases with a large temperature range which eventually include more complex reaction processes, the functional relation between ln(α) and (1/ϑ) is nonlinear in the ‘Arrhenius plot’. In those cases, the monotone change of the nonlinear range in the ‘Arrhenius plot’ can be interpreted as a transient range between two approximately linear or constant regions. An extended application of the modified Arrhenius equation from Nakamura (1989) is presented in this study for this transient range. The introduced method was applied to describe the time-temperature equivalence in the relaxation analysis of restoring seal force of metal seals, which are used in lid-systems of transport and interim storage casks for radioactive materials. But, the method is widely valid and can be used for different objectives which are characterized by thermorheologically simple behavior with nonlinear sensitivity to inverse temperature. Exposure under such dual-environment can lead to accelerated corrosion of the components on the air side. The oxidation behaviour of ferritic/martensitic steel T92 was investigated under dual-environment in a specially designed test equipment. The samples were exposed to dry oxyfuel flue gas (CO2–27%N2–2%O2–1%SO2) on one side and to steam on the other side up to 1000 h at 650°C. The formation of oxide scales was characterised by optical microscopy and scanning electron microscopy with attached energy-dispersive spectroscopy. Oxidation rate of specimens under dual-Environment condition was almost three times higher than that in single-environment condition. This is explained based on hydrogen transport through the bulk alloy from the steam side to the flue gas side. In this work, a methodology is proposed to identify multiple axial forces in members of a truss structure based on the modal parameters. Vibration test allows the identification of the natural frequencies and mode shapes, globally of the truss structure as well as locally of the individual bars. The method calibrates the numerical model of the truss structure using a genetic algorithm and strategic validation criteria. The validation criteria are based on the identified natural frequencies and global mode shapes of the truss structure as well as information of the axial forces in the individual bars of the truss, which are estimated from the natural frequencies and five amplitudes of the corresponding local mode shapes of the single bars based on an analytical-based algorithm. The calibration allows the identification of the axial forces in all bars of the truss structure. For mode pairing strategy, a technique makes use of the enhanced modal assurance criteria with the calculation of the modal strain energies. Moreover, the modal strain energies are also used to select the relevant local mode shape of the individual bars. The feasibility and accuracy of the proposed methodology is verified by laboratory experiments on several truss structures. In situ tests on existing trusses are intended. The results from one of the laboratory tested structures, i.e. a two-bar system, are included in this paper. Xenobiotics are able to sorb to polymer materials, and this process is further enhanced where they Encounter microplastics (plastic fragments <5 mm). In this work we studied the sorption of metformin, a type-2 diabetes drug, and difenoconazole, a fungicide, onto the virgin polymer materials polyamide (PA), polypropylene (PP), and polystyrene (PS). Additionally, PP was cryo-milled and PA was treated with acid to investigate the influence of an increase in surface area and chemical modification. The material properties were also studied by dynamic scanning calorimetry (DSC), gel permeation chromatography (GPC) and Fourier transform infrared spectroscopy (FTIR). Sorption experiments were performed on the basis of a full factorial design examining the effect of agitation, pH value, and salinity. Experimental results showed that difenoconazole sorbs readily to all microplastics, whereas the more polar analyte metformin did not show any affinity to the materials used. For difenoconazole the governing factor in all cases is agitation, while both pH and salinity exhibited only a slight influence. The modification of polymers leads to enhanced sorption, indicating that an increase in surface area (cryo-milled PP) or inner volume (acid-treated PA) strongly favours adsorption. Moreover, long-term experiments demonstrated that the time until equilibrium is reached depends strongly on the particle size. T1 - Dispersion of nanomaterials in aqueous media: Towards protocol optimization JF - Journal of Visualized Experiments N2 - The sonication process is commonly used for de-agglomerating and dispersing nanomaterials in aqueous based media, necessary to improve homogeneity and stability of the suspension. In this study, a systematic step-wise approach is carried out to identify optimal sonication conditions in order to achieve a stable dispersion. This approach has been adopted and shown to be suitable for several nanomaterials (cerium oxide, zinc oxide, and carbon nanotubes) dispersed in deionized (DI) water. However, with any change in either the nanomaterial type or dispersing medium, there needs to be optimization of the basic protocol by adjusting various factors such as sonication time, power, and sonicator type as well as temperature rise during the process. The approach records the dispersion process in detail. This is necessary to identify the time Points as well as other above-mentioned conditions during the sonication process in which there may be undesirable changes, such as damage to the particle surface thus affecting surface properties. Our goal is to offer a harmonized approach that can control the Quality of the final, produced dispersion. Such a guideline is instrumental in ensuring dispersion quality repeatability in the nanoscience community, particularly in the field of nanotoxicology. T1 - Ultrasonic Phased Array Design Study for High Speed Axle Inspection using an Electronically Rotating Beam JF - Procedia Structural Integrity N2 - For in-service inspections on wheelset axles with a hollow drilling, mechanized ultrasound inspection systems with single element probes are typically used. The ultrasonic testing in the zones close to the external surface of the railway axles can be realized from the inside of the bore hole, without demounting the wheelset and without dismantling the wheels and the brake discs. The testing system must be able to find flaws in the external surface of the hollow shafts, whose surface lies in the radial-radial plane, these are called transversal flaw. Presently testing systems are used, where scanning is realized in the circumferential direction by mechanical rotation of the probe system in the actual drilling. The phased array probe system, which is presented here, can carry out the rotation scan electronically. The scan can be carried out by simply moving the system forward and backwards through the drilling without mechanical rotation. Manipulation becomes simpler and the inspection time can be shortened considerably. The ultrasonic beam can be inclined exactly and be focused in the plane vertical to the specimen axis. The probe is designed with help of indispensable simulations using especially designed software developed by BAM. The feasibility and the alignment between the simulated and experimental results were shown in earlier projects reported by Boehm et al. (2006) and Völz et al. (2012). The main task here is to optimize a probe for bore holes with a diameter of 65 mm with an increase in sensitivity and a high spatial resolution. This development will be carried out by use of extensive simulations and result in certain changes of the relevant probe parameters. This group of gold and silver figurines includes typical specimens of the Inca production, such as the four silver figurines from the Island Coati in Lake Titicaca and the two gold figurines from Pachacamac, as well as specimens that are seldom present in collections, which makes them atypical. In addition to the tall figurines with atypical hair or standing pose, one bicolour figurine half silver, half gold) and another with coloured inlays were included in this study for comparison. The large majority of the silver specimens are made from very high quality alloys and the gold figurines are made from alloys containing low copper contents and silver ranging from 30wt% to 52wt%. Here, we explore the evolution of Marek’s disease virus (MDV), a herpesvirus commonly afflicting chickens and rarely other avian species. The history of MDV in the 20th century represents an important case study in the evolution of virulence. The severity of MDV infection in chickens has been rising steadily since the adoption of intensive farming techniques and vaccination programs in the 1950s and 1970s, respectively. It has remained uncertain, however, which of these factors is causally more responsible for the observed increase in virulence of circulating viruses. We conducted a phylogenomic study to understand the evolution of MDV in the context of dramatic changes to poultry farming and disease control. Our analysis reveals evidence of geographical structuring of MDV strains, with reconstructions supporting the emergence of virulent viruses independently in North America and Eurasia. Of note, the emergence of virulent viruses appears to coincide approximately with the introduction of comprehensive vaccination on both continents. The time-dated phylogeny also indicated that MDV has a mean evolutionary rate of ~1.6 × 10−5 substitutions per site per year. An examination of gene-linked mutations did not identify a strong association between mutational variation and virulence phenotypes, indicating that MDV may evolve readily and rapidly under strong selective pressures and that multiple genotypic pathways may underlie virulence adaptation in MDV. In the course of digitization, digital detector arrays (DDA) are finding their way into industrial applications and are replacing film radiography step by step. This study deals with the latest generation of DDAs, the photon counting and energy resolving detectors (PCD), and their characteristics compared to charge integrating detectors (CID). No matter which technology to use, radiography still lacks a general issue: A three-dimensional object is projected onto a two dimensional image. Of course, advanced computed tomography (CT) algorithms exist since many years, but if the object to investigate is too large to fit into the manipulation system or its shape is not appropriate, CT is not feasible or sensible to be applied. To overcome this limitation, numerous laminographic algorithms have been developed in the past. In this study, photon counting detectors are used in combination with co-planar translational laminography to gain reconstructed three-dimensional volumes. Both laminographic testing and PCDs require a serious knowledge of many parameters that can influence the image quality in the resulting datasets. These are e.g. the detector efficiency and calibration procedure, setting of energy thresholds, exposure data, number of projections, beam length correction and spatial resolution. The use of PCDs yields more variables to be considered compared to CIDs. The most important parameters in laminographic testing and in the use of PCDs are described in this study and limits are discussed. It is applied as a method of quality control, for online structural health monitoring, and for inspection of safety related components. Due to the ability of automation and a simple test setup ultrasonic testing is one major NDT technique next to several existing options. Whereas contact technique allows the use of higher frequencies of some MHz and phased array focusing, air-coupled ultrasonic testing (ACUT) shows different advantages. Most significant for ACUT is the absence of any coupling fluid and an economical test procedure respective time and costs. Both contact technique and ACUT have been improved and enhanced during the past years. One important enhancement is the development of airborne transducers based on ferroelectrets, like charged cellular polypropylene (cpp), which makes the application of any matching layers being mandatory in conventional piezoelectric transducers unnecessary. In this contribution we show ultrasonic inspection results of specimens made of carbon- and glass-fibre-reinforced plastic. These specimens include defects represented by drill holes and artificial delaminations of various size and depth. We compare inspection results achieved by using contact technique to those achieved by ACUT. For ACUT, conventional piezoelectric transducers and transducers based on cpp were used, both focused as well as non-focused types. Contact inspections were performed with a multi-channel matrix array probe. Once the inspection data is recorded it can be analysed in order to detect and evaluate defects in the specimen. We present different analysing strategies and compare these regarding detection rate and sizing of defects.
