Halogenated organics are effective FRs but are controversially discussed due to the release of toxic gases during a fire or their persistence if landfilled. Phosphorus-containing compounds are effective alternatives to halogenated FRs and have potential lower toxicity and degradability. In addition, nitrogencontaining additives were reported to induce synergistic effects with phosphorus-based FRs. However, no systematic study of the gradual variation on a single phosphorus FR containing both P−O and P−N moieties and their comparison to the respective blends of phosphates and phosphoramides was reported. This study developed general design principles for P−O- and P−N-based FRs and will help to design effective FRs for various polymers. We synthesized a library of phosphorus FRs that only differ in their P-binding pattern from each other and studied their decomposition mechanism in epoxy resins. Systematic control over the decomposition pathways of phosphate (PO(OR)3), phosphoramidate (PO(OR)2(NHR)), phosphorodiamidate (PO(OR)(NHR)2), phosphoramide (PO(NHR)3), and their blends was identified, for example, by reducing cis-elimination and the formation of P−N-rich char with increasing nitrogen content in the P-binding sphere. Our FR epoxy resins can compete with commercial FRs in most cases, but we proved that the blending of esters and amides outperformed the single molecule amidates/diamidates due to distinctively different decomposition mechanisms acting synergistically when blended. Zur Untersuchung des Alterungsverhaltens solcher Composite-Druckgefäße wurde in der BAM das interdisziplinäre Themenfeldprojekt COD-AGE durchgeführt. Ziel des Projektes war die Entwicklung von Methoden und Modellen zur Beschreibung und Bestimmung des Alterungsverhaltens von Carbonfaser-Compositen am Beispiel von Druckgefäßen, um eine bessere Vorhersage der Alterung und der sicheren Gebrauchsdauer zu ermöglichen. Ein Schwerpunkt in diesem Projekt war die Bereitstellung geeigneter ZfP-Methoden. Dabei ging es sowohl um versuchsbegleitende Prüfungen, als auch um die mögliche Entwicklung von Prüfeinrichtungen für den späteren praktischen Gebrauch. In dem Vortrag werden Prüfergebnisse der alterungsbegleitenden Wirbelstromprüfung an Composite-Druckgefäßen vorgestellt. In Alterungsversuchen wurden Druckgefäße aus einem ca. 4 mm dicken Aluminiumliner und ca. 8 mm dicker CFK-Ummantelung untersucht. Typisches Einsatzgebiet solcher Druckgefäße sind Atemschutzgeräte der Feuerwehr. Die Druckgefäße wurden sowohl mit konventioneller Wirbelstromtechnik von außen und von innen als auch mit hochfrequenter Wirbelstromtechnik von außen geprüft. Es konnten sowohl Beschädigungen des metallischen Liners, als auch Strukturen des CFK-Geflechts nachgewiesen werden. Eine besondere mechanische Herausforderung bestand in der Prüfung der Innenseite der Liner, da hier eine zylindrische Fläche mit einem Innendurchmesser von 150 mm durch einen Zugang von nur 15 mm Durchmesser geprüft werden muss. Durch den unterschiedlichen Aufbau, im Vergleich zu integrierten Scherwellen-Winkelprüfköpfen, können jedoch während der Prüfung Fehlanzeigen generiert werden. Diese fehlerhaften Anzeigen beruhen auf Mehrfachreflexionen und Wellenumwandlungen innerhalb des Vorlaufkeils, aufgrund der Geometrie und können im Feld nur bedingt von realen Anzeigen unterschieden werden. Dieser Beitrag zeigt die geometrische Betrachtung bei der Verwendung von Vorlaufkeilen und wurde experimentell durch die Bestimmung der Richtcharakteristik und dem Einsatz von elektrodynamischen Sonden validiert. T1 - Cancer cell lipid class homeostasis is altered under nutrient-deprivation but stable under hypoxia N2 - Background: Cancer cells modify the balance between fatty acid (FA) synthesis and uptake under metabolic stress, induced by oxygen/nutrient deprivation. These modifications were shown to alter the levels of individual triglyceride (TG) or phospholipid sub-species. To attain a holistic overview of the lipidomic profiles of cancer cells under stress we performed a broad lipidomic assay, comprising 244 lipids from six major classes. This assay allowed us to perform robust analyses and assess the changes in averages of broader lipid-classes, stratified on the basis of saturation index of their fatty-acyl side chains. Methods: Global lipidomic profiling using Liquid Chromatography-Mass Spectrometry was performed to assess lipidomic profiles of biologically diverse cancer cell lines cultivated under metabolically stressed conditions. Results: Neutral lipid compositions were markedly modified under serum-deprived conditions and, strikingly, the cellular level of triglyceride subspecies decreased with increasing number of double bonds in their fatty acyl chains. In contrast and unexpectedly, no robust changes were observed in lipidomic profiles of hypoxic (2% O2) Cancer cells despite concurrent changes in proliferation rates and metabolic gene expression. Conclusions: Serum-deprivation significantly affects lipidomic profiles of cancer cells. Although, the levels of individual lipid moieties alter under hypoxia (2% O2), the robust averages of broader lipid classes remain unchanged. This work focuses on the processing of industrial grade purified rosin mixed with polyethylene (PE), polypropylene (PP), polylactic acid (PLA), polyamide (PA) and corn starch based biopolymer (CS). Homopolymer masterbatches were extrusion-compounded and melt-spun to form fibres for a wide range of products, such as filters, reinforcements, clothing and medical textiles. Due to the versatile chemical structure of rosin, it was observed compatible with all the selected polymers. In general, the rosin-blended systems were shear-thinning in a molten condition. The doped fibres spun of PE and PP indicated adequate melt-spinning capability and proper mechanical properties in terms of ultimate strength and Young's modulus. The antibacterial response was found dependent on the selected polymer. Especially PE with a 10 wt% rosin content showed significant antibacterial effects against Escherichia coli DH5α and Staphylococcus aureus ATCC 12598 when analysed in the Ringer's solution for 24 h. T1 - Preactivation Crosslinking - An Efficient Method for the Oriented Immobilization of Antibodies N2 - Crosslinking of proteins for their irreversible immobilization on surfaces is a proven and popular method. However, many protocols lead to random orientation and the formation of undefined or even inactive by-products. Most concepts to obtain a more targeted conjugation or immobilization requires the recombinant modification of at least one binding partner, which is often impractical or prohibitively expensive. Here a novel method is presented, which is based on the chemical preactivation of Protein A or G with selected conventional crosslinkers. In a second step, the antibody is added, which is subsequently crosslinked in the Fc part. This leads to an oriented and covalent immobilization of the immunoglobulin with a very high yield. Protocols for Protein A and Protein G with murine and human IgG are presented. This method may be useful for the preparation of columns for affinity chromatography, immunoprecipitation, antibodies conjugated to magnetic particles, permanent and oriented immobilization of antibodies in biosensor systems, microarrays, microtitration plates or any other system, where the loss of antibodies needs to be avoided, and maximum binding capacity is desired. This method is directly applicable even to antibodies in crude cell culture supernatants, raw sera or protein-stabilized antibody preparations without any purification nor enrichment of the IgG. This new method delivered much higher signals as a traditional method and, hence, seems to be preferable in many applications. A research project financed by the German Federal Environment Agency was initiated to finalize some of these CEN standard drafts, since fully validated standard procedures are crucial for the evaluation of their reliability in the context of implementation in legislation on environmental health. Approach: Appropriate test materials (< 2mm particle size) were prepared and homogenized from contaminated soils, sludge and treated biowaste containing polycyclic aromatic hydrocarbons (PAH), polychlorinated biphenyls (PCB), dioxins, furans and dioxin-like-PCB and served, along with reference solutions, as the basis for international interlaboratory comparisons. Performance data of three analytical standard procedures were obtained by the statistical evaluation of results received from 11 to 29 participants per test material. Results: The overall variation coefficients of reproducibility (between-lab standard deviations) for the sum parameters were roughly between 10 and 35 %. The variation coefficients of repeatability (within-lab standard deviations) range between 3 % and 8 % and show no trend considering the substance groups or matrices. The highest coefficients of reproducibility were found for the analysis of PAHs, which were between 26 and 35 %, depending on the matrix, whereas 7-17 % reproducibility was observed for toxicity equivalents (TEQ) comprising dioxins, furans and dl-PCB. Conclusions: Overall, the results confirm that the procedures described in the Technical Specifications are fit for purpose for all three matrices and that the feasibility of the HORIZONTAL approach, to cover several matrices with one standard per analyte, was thereby proven. The multicomponent system was designed to improve flame retardancy and the mechanical properties of the rubber compounds, while simultaneously reducing the amount of filler. PANI was applied at low loadings (7 phr) and combined with the phosphorous APP (21 phr) and the mineral flame retardant ATH (50 phr). A comprehensive study of six EPDM rubbers was carried out by systematically varying the fillers to explain the impact of multicomponent flame retardant systems on mechanical properties. The six EPDM materials were investigated via the UL 94, limiting oxygen index (LOI), FMVSS 302, glow wire tests, and the cone calorimeter, showing that multicomponent flame retardant systems led to improved fire performance. In cone calorimeter tests the EPDM/APP/ATH/PANI composite reduced the maximum average rate of heat emission (MARHE) to 142 kW·m-2, a value 50% lower than that for the unfilled EPDM rubber. Furthermore, the amount of phosphorus in the residues was quantified and the mode of action of the phosphorous flame retardant APP was explained. The data from the cone calorimeter were used to determine the protective layer effect of the multicomponent flame retardant systems in the EPDM compounds. Increasing biofuels use would bring some benefits, such as a reduction in oil demands and greenhouse gas emissions, and an improvement in air quality. Materials compatibility is a major concern whenever the fuel composition is changed in a fuel system. The objective of this research was to determine the resistance of frequently used sealing materials such as CR (chloroprene rubber), CSM (chlorosulfonated polyethylene), EPDM (ethylene-propylene-diene rubber), FKM (fluorocarbon rubber), FVMQ (methyl-fluorosilicone rubber), IIR (butyl rubber), NBR (acrylonitrile-butadiene rubber), PA (polyamides), PUR (polyester urethane rubber) and VMQ (methyl-vinyl-silicone rubber), in heating oil with admixtures of biogenic sources such as E10 (fuel with max. 10 % ethanol), E85 (fuel with 85 % ethanol), non-aged and aged biodiesel, diesel fuel with 5 % biodiesel, non-aged and aged B10 (heating oil with 10 % biodiesel) at 20 °C, 40 °C and 70 °C. Mass, tensile strength and breaking elongation of the test specimens were determined before and after the exposure for 84 days in the fuels. The visual examination of some elastomer test specimens clearly showed the great volume increase until break or partial dissolution. Shore hardness A and D (for PA) were determined before and after exposure of the test specimens in the biofuels for 42 days. There is not determined a threshold for the reduction in tensile properties and Shore hardness in the international standards. Therefore, a threshold of 15 % was set for the evaluation of the compatibility. The sealing materials CR, CSM, EPDM, IIR and NBR were generally not resistant to biodiesel and B10. In summary, it can be therefore stated that the chemical resistance of the fluoropolymers FKM and FVMQ in fuels and biofuels is the best one.
