T1 - Characterization of a series of absolute isotope reference materials for magnesium: ab initio calibration of the mass spectrometers, and determination of isotopic compositions and relative atomic weights N2 - For the first time, an ab initio calibration for absolute Mg isotope ratios was carried out, without making any a priori assumptions. All quantities influencing the calibration such as the purity of the enriched isotopes or liquid and solid densities were carefully analysed and their associated uncertainties were considered. A second unique aspect was the preparation of three sets of calibration solutions, which were applied to calibrate three multicollector ICPMS instruments by quantifying the correction factors for instrumental mass discrimination. Those fully calibrated mass spectrometers were then used to determine the absolute Mg isotope ratios in three candidate European Reference Materials (ERM)-AE143, -AE144 and -AE145, with ERM-AE143 becoming the new primary isotopic reference material for absolute isotope ratio and delta measurements. The isotope amount ratios of ERM-AE143 are n(25Mg)/n(24Mg) = 0.126590(20) mol/mol and n(26Mg)/n(24Mg) = 0.139362(43) mol/mol, with the resulting isotope amount fractions of x(24Mg) = 0.789920(46) mol/mol, x(25Mg) = 0.099996(14) mol/ mol and x(26Mg) = 0.110085(28) mol/mol and an atomic weight of Ar(Mg) = 24.305017(73); all uncertainties were stated for k = 2. This isotopic composition is identical within uncertainties to those stated on the NIST SRM 980 certificate. The candidate materials ERM-AE144 and -AE145 are isotopically lighter than ERM-AE143 by 1.6 ‰ and 1.3 ‰, respectively, concerning their n(26Mg)/n(24Mg) ratio. The relative combined standard uncertainties are ≤0.1 ‰ for the isotope ratio n(25Mg)/n(24Mg) and ≤0.15 ‰ for the isotope ratio n(26Mg)/ n(24Mg). In addition to characterizing the new isotopic reference materials, it was demonstrated that commonly used fractionation laws are invalid for correcting Mg isotope ratios in multicollector ICPMS as they result in a bias which is not covered by its associated uncertainty. Depending on their type, fractionation laws create a bias up to several per mil, with the exponential law showing the smallest bias between 0.1 ‰ and 0.7 ‰. In this chapter, exemplary welding processes are presented that reduce the environmental and social impacts of thick metal plate welding. Social and environmental Life Cycle Assessments for a thick metal plate joint are conducted for the purpose of expressing and analysing the social and environmental impacts of welding. The application of high strength steels allows smaller wall thicknesses compared to conventional steels. Therefore, the risk of buckling has to be considered carefully. Due to the lack of knowledge concerning the residual stresses present after welding in high strength steel components conservative assumptions of their level and distribution is typically applied. In this study I-girders made of steels showing strengths of 355 MPa and 690 MPa were welded with varying heat input. Due to the dimension of the I-girders and the complex geometry the accessibility for residual stress measurement using X-ray diffraction was limited. Therefore, saw cutting accompanied by strain gauge measurement has been used to produce smaller sections appropriate to apply X-ray diffraction. The stress relaxation measured by strain gauges has been added to residual stresses determined by X-ray diffraction to obtain the original stress level and distribution before sectioning. The combination of both techniques can produce robust residual stress values. From practical point of view afford for strain gauge application can be limited to a number of measuring positions solely to record the global amount of stress relaxation. X-ray diffraction can be applied after sectioning to determine the residual stresses with sufficient spatial resolution. The wide diversity of compounds that can act as compatible solute complicates the task of understanding the different strategies that halophilic bacteria use to cope with salt. This is specially challenging when attempting to go beyond the pathway that produces a certain compatible solute towards an understanding of how the metabolic network as a whole addresses the problem. Metabolic reconstruction based on genomic data together with Flux Balance Analysis (FBA) is a promising tool to gain insight into this problem. However, as more of these reconstructions become available, it becomes clear that processes predicted by genome annotation may not reflect the processes that are active in vivo. As a case in point, E. coli is unable to grow aerobically on citrate in spite of having all the necessary genes to do it. It has also been shown that the realization of this genetic potential into an actual capability to metabolize citrate is an extremely unlikely event under normal evolutionary conditions. Moreover, many marine bacteria seem to have the same pathways to metabolize glucose but each species uses a different one. In this work, a metabolic network inferred from genomic annotation of the halophilic bacterium Halomonas elongata and proteomic profiling experiments are used as a starting point to motivate targeted experiments in order to find out some of the defining features of the osmoregulatory strategies of this bacterium. This new information is then used to refine the network in order to describe the actual capabilities of H. elongata, rather than its genetic potential. Here we evaluate the extent of agreement between classification by electron microscopy (EM) and classification by VSSA on a large set of diverse particulate substances that represent all the anticipated challenges except mixtures of different substances. EM and VSSA are determined in multiple labs to assess also the level of reproducibility. Based on the results obtained on highly characterized benchmark materials from the NanoDefine EU FP7 project, we derive a tiered screening strategy for the purpose of implementing the definition of nanomaterials. We finally apply the Screening strategy to further industrial materials, which were classified correctly and left only borderline cases for EM. On platelet-shaped nanomaterials, VSSA is essential to prevent false-negative classification by EM. On porous materials, approaches involving extended Adsorption isotherms prevent false positive classification by VSSA. We find no false negatives by VSSA, neither in Tier 1 nor in Tier 2, despite real-world industrial polydispersity and diverse composition, shape, and coatings. The VSSA screening strategy is recommended for inclusion in a technical guidance for the implementation of the definition. A large experimental work is presented in which micro-cracks have been made visible by heat-tinting at successive stages of fatigue life of the welded specimens. Here the correlation between the number of initiation sites and the applied stress level has been also investigated. The criteria have been implemented in in-house software, which allows multiple fatigue crack propagation, and validated against selected experimental tests. Such metabolic specialization could be driven by stochastic gene expression and could provide individual cells with growth benefits of specialization. We measured the degree of phenotypic specialization in two parallel metabolic pathways, the assimilation of glucose and arabinose. We grew Escherichia coli in chemostats, and used isotope-labeled sugars in combination with nanometer-scale secondary ion mass spectrometry and mathematical modeling to quantify sugar assimilation at the single-cell level. We found large variation in metabolic activities between single cells, both in absolute assimilation and in the degree to which individual cells specialize in the assimilation of different sugars. Analysis of transcriptional reporters indicated that this variation was at least partially based on cell-to-cell variation in gene expression. Metabolic differences between cells in clonal populations could potentially reduce metabolic incompatibilities between different pathways, and increase the rate at which parallel reactions can be performed. The influence of the load type on CF behavior of the alloy was examined by applying push/pull and rotating bending cyclic load to failure. The hour-glass shaped specimens with both polished and precisely turned surface finish were tested in corrosion chambers allowing for the permanent circulation of Northern German Basin electrolyte heated to 369 K. Additionally, the influence of a protective cathodic potential on the CF life expectancy was investigated by employing a potentiostat. The rotating bending cyclic load led to a higher CF life expectancy, which decreased with increasing duration of the experiment due to a higher deterioration rate of the material. The application of a cathodic potential caused a significant increase of the CF lifespan from 4.7 x 105 (open-circuit potential (OCP)) to 107 cycles (preset threshold cycle number) for the potential range from USHE = – 450 to – 900 mV. Therefore, the Arrhenius equation is widely used due to its simple form and often leads to suitable results. Where, the Arrhenius equation presents a linear relation for the temperature-dependent shift factor in logarithmic scale ln(α) with the absolute inverse temperature (1/ϑ). However, in cases with a large temperature range which eventually include more complex reaction processes, the functional relation between ln(α) and (1/ϑ) is nonlinear in the ‘Arrhenius plot’. In those cases, the monotone change of the nonlinear range in the ‘Arrhenius plot’ can be interpreted as a transient range between two approximately linear or constant regions. An extended application of the modified Arrhenius equation from Nakamura (1989) is presented in this study for this transient range. The introduced method was applied to describe the time-temperature equivalence in the relaxation analysis of restoring seal force of metal seals, which are used in lid-systems of transport and interim storage casks for radioactive materials. Exposure under such dual-environment can lead to accelerated corrosion of the components on the air side. The oxidation behaviour of ferritic/martensitic steel T92 was investigated under dual-environment in a specially designed test equipment. The samples were exposed to dry oxyfuel flue gas (CO2–27%N2–2%O2–1%SO2) on one side and to steam on the other side up to 1000 h at 650°C. The formation of oxide scales was characterised by optical microscopy and scanning electron microscopy with attached energy-dispersive spectroscopy. Oxidation rate of specimens under dual-Environment condition was almost three times higher than that in single-environment condition. In this work, a methodology is proposed to identify multiple axial forces in members of a truss structure based on the modal parameters. Vibration test allows the identification of the natural frequencies and mode shapes, globally of the truss structure as well as locally of the individual bars. The method calibrates the numerical model of the truss structure using a genetic algorithm and strategic validation criteria. The validation criteria are based on the identified natural frequencies and global mode shapes of the truss structure as well as information of the axial forces in the individual bars of the truss, which are estimated from the natural frequencies and five amplitudes of the corresponding local mode shapes of the single bars based on an analytical-based algorithm. The calibration allows the identification of the axial forces in all bars of the truss structure. For mode pairing strategy, a technique makes use of the enhanced modal assurance criteria with the calculation of the modal strain energies. Moreover, the modal strain energies are also used to select the relevant local mode shape of the individual bars. The feasibility and accuracy of the proposed methodology is verified by laboratory experiments on several truss structures. In situ tests on existing trusses are intended. The results from one of the laboratory tested structures, i.e. a two-bar system, are included in this paper. Xenobiotics are able to sorb to polymer materials, and this process is further enhanced where they Encounter microplastics (plastic fragments <5 mm). In this work we studied the sorption of metformin, a type-2 diabetes drug, and difenoconazole, a fungicide, onto the virgin polymer materials polyamide (PA), polypropylene (PP), and polystyrene (PS). Additionally, PP was cryo-milled and PA was treated with acid to investigate the influence of an increase in surface area and chemical modification. The material properties were also studied by dynamic scanning calorimetry (DSC), gel permeation chromatography (GPC) and Fourier transform infrared spectroscopy (FTIR). Sorption experiments were performed on the basis of a full factorial design examining the effect of agitation, pH value, and salinity. Experimental results showed that difenoconazole sorbs readily to all microplastics, whereas the more polar analyte metformin did not show any affinity to the materials used. For difenoconazole the governing factor in all cases is agitation, while both pH and salinity exhibited only a slight influence. The modification of polymers leads to enhanced sorption, indicating that an increase in surface area (cryo-milled PP) or inner volume (acid-treated PA) strongly favours adsorption. Moreover, long-term experiments demonstrated that the time until equilibrium is reached depends strongly on the particle size. Die bisher in definierten Intervallen durchgeführten, wiederkehrenden Prüfungen zur Sicherstellung leckagefreier Rohrleitungen sind kostenintensiv und mit einem erhöhten Aufwand verbunden. Mit dem interdisziplinären Forschungsprojekt AGIFAMOR (Ageing Infrastructures – Akustisches Monitoring von Rohrleitungen) arbeitet die BAM an der Entwicklung und Erprobung einer örtlich und zeitlich kontinuierlichen Zustandsüberwachung von Rohrleitungen basierend auf einem faseroptischen akustischen Messsystem, welches einen vergleichsweise geringen Installationsaufwand der Sensoren erfordert. Mit diesem Verfahren sollen vor allem betriebsbedingte Schädigungen infolge von Korrosion sowie daraus entstehende Pittings oder Risse erkannt und damit Leckagen verhindert werden. Im Rahmen einer Machbarkeitsstudie zum Teilaspekt „Detektion von Risswachstum“ wurde ein 2,5 m langes, definiert vorgekerbtes Stahlrohr mit der faseroptischen Sensorik (FOS) ausgestattet und quasistatisch unter 4-Punkt-Biegung belastet. Mit fortschreitender Belastung wächst, ausgehend vom vorhandenen Außenumfangskerb, ein Riss im Stahlrohr bis zum Erreichen der Leckage – in der Praxis ein gefährliches Schadensszenario. In this study, a systematic step-wise approach is carried out to identify optimal sonication conditions in order to achieve a stable dispersion. This approach has been adopted and shown to be suitable for several nanomaterials (cerium oxide, zinc oxide, and carbon nanotubes) dispersed in deionized (DI) water. However, with any change in either the nanomaterial type or dispersing medium, there needs to be optimization of the basic protocol by adjusting various factors such as sonication time, power, and sonicator type as well as temperature rise during the process. The approach records the dispersion process in detail. This is necessary to identify the time Points as well as other above-mentioned conditions during the sonication process in which there may be undesirable changes, such as damage to the particle surface thus affecting surface properties. Our goal is to offer a harmonized approach that can control the Quality of the final, produced dispersion. T1 - Ultrasonic Phased Array Design Study for High Speed Axle Inspection using an Electronically Rotating Beam N2 - For in-service inspections on wheelset axles with a hollow drilling, mechanized ultrasound inspection systems with single element probes are typically used. The ultrasonic testing in the zones close to the external surface of the railway axles can be realized from the inside of the bore hole, without demounting the wheelset and without dismantling the wheels and the brake discs. The testing system must be able to find flaws in the external surface of the hollow shafts, whose surface lies in the radial-radial plane, these are called transversal flaw. Presently testing systems are used, where scanning is realized in the circumferential direction by mechanical rotation of the probe system in the actual drilling. The phased array probe system, which is presented here, can carry out the rotation scan electronically. The scan can be carried out by simply moving the system forward and backwards through the drilling without mechanical rotation. Manipulation becomes simpler and the inspection time can be shortened considerably. The ultrasonic beam can be inclined exactly and be focused in the plane vertical to the specimen axis. The probe is designed with help of indispensable simulations using especially designed software developed by BAM. The feasibility and the alignment between the simulated and experimental results were shown in earlier projects reported by Boehm et al. (2006) and Völz et al. (2012). The main task here is to optimize a probe for bore holes with a diameter of 65 mm with an increase in sensitivity and a high spatial resolution. This development will be carried out by use of extensive simulations and result in certain changes of the relevant probe parameters. Here, we explore the evolution of Marek’s disease virus (MDV), a herpesvirus commonly afflicting chickens and rarely other avian species. The history of MDV in the 20th century represents an important case study in the evolution of virulence. The severity of MDV infection in chickens has been rising steadily since the adoption of intensive farming techniques and vaccination programs in the 1950s and 1970s, respectively. It has remained uncertain, however, which of these factors is causally more responsible for the observed increase in virulence of circulating viruses. We conducted a phylogenomic study to understand the evolution of MDV in the context of dramatic changes to poultry farming and disease control. Our analysis reveals evidence of geographical structuring of MDV strains, with reconstructions supporting the emergence of virulent viruses independently in North America and Eurasia. Of note, the emergence of virulent viruses appears to coincide approximately with the introduction of comprehensive vaccination on both continents. The time-dated phylogeny also indicated that MDV has a mean evolutionary rate of ~1.6 × 10−5 substitutions per site per year. An examination of gene-linked mutations did not identify a strong association between mutational variation and virulence phenotypes, indicating that MDV may evolve readily and rapidly under strong selective pressures and that multiple genotypic pathways may underlie virulence adaptation in MDV. In the course of digitization, digital detector arrays (DDA) are finding their way into industrial applications and are replacing film radiography step by step. This study deals with the latest generation of DDAs, the photon counting and energy resolving detectors (PCD), and their characteristics compared to charge integrating detectors (CID). No matter which technology to use, radiography still lacks a general issue: A three-dimensional object is projected onto a two dimensional image. Of course, advanced computed tomography (CT) algorithms exist since many years, but if the object to investigate is too large to fit into the manipulation system or its shape is not appropriate, CT is not feasible or sensible to be applied. To overcome this limitation, numerous laminographic algorithms have been developed in the past. In this study, photon counting detectors are used in combination with co-planar translational laminography to gain reconstructed three-dimensional volumes. Both laminographic testing and PCDs require a serious knowledge of many parameters that can influence the image quality in the resulting datasets. These are e.g. the detector efficiency and calibration procedure, setting of energy thresholds, exposure data, number of projections, beam length correction and spatial resolution. The use of PCDs yields more variables to be considered compared to CIDs. The most important parameters in laminographic testing and in the use of PCDs are described in this study and limits are discussed. It is applied as a method of quality control, for online structural health monitoring, and for inspection of safety related components. Due to the ability of automation and a simple test setup ultrasonic testing is one major NDT technique next to several existing options. Whereas contact technique allows the use of higher frequencies of some MHz and phased array focusing, air-coupled ultrasonic testing (ACUT) shows different advantages. Most significant for ACUT is the absence of any coupling fluid and an economical test procedure respective time and costs. Both contact technique and ACUT have been improved and enhanced during the past years. One important enhancement is the development of airborne transducers based on ferroelectrets, like charged cellular polypropylene (cpp), which makes the application of any matching layers being mandatory in conventional piezoelectric transducers unnecessary. In this contribution we show ultrasonic inspection results of specimens made of carbon- and glass-fibre-reinforced plastic. These specimens include defects represented by drill holes and artificial delaminations of various size and depth. We compare inspection results achieved by using contact technique to those achieved by ACUT. For ACUT, conventional piezoelectric transducers and transducers based on cpp were used, both focused as well as non-focused types. Contact inspections were performed with a multi-channel matrix array probe. Once the inspection data is recorded it can be analysed in order to detect and evaluate defects in the specimen. We present different analysing strategies and compare these regarding detection rate and sizing of defects. However, agitating certain types of soil and then passing the solution through a membrane filter with 0.45-µm pores yields filtrates that have been colored by the colloidal particles. These colloids might affect the results of the inorganic substances obtained in the batch tests. In this study, we evaluated the effects of colloidal particles on the results and the reproducibility of batch tests on contaminated soil using different agitation methods and membrane filters with different pore sizes. The leaching behaviors of As, Pb, Se, F and Cl from three types of soil were studied. The As and Pb concentrations in the leachates of some types of soil were clearly affected by the amount of colloidal particles with a diameter of 0.10–0.45 µm and by the agitation method used. This was probably because As and Pb were present mainly in the particulate form in the leachate that had been passed through a membrane filter with 0.45-µm pores. This is not the case for every type of soil. The results of batch leaching tests showed that not only dissolved but also colloidal forms with a diameter of 0.10–0.45 µm might be included and that the existence of colloidal particles in the leachate decreases the batch leaching test reproducibility. T1 - Structure of melanins from the fungi Ochroconis lascauxensis and Ochroconis anomala contaminating rock art in the Lascaux Cave N2 - Two novel species of the fungal genus Ochroconis, O. lascauxensis and O. anomala have been isolated from the walls of the Lascaux Cave, France. The interest in these fungi and their melanins lies in the formation of black stains on the walls and rock art which threatens the integrity of the paintings. Here we report solid-state cross polarization magic-angle spinning 13C and 15N nuclear magnetic resonance (NMR) spectroscopy and surface-enhanced Raman spectroscopy (SERS) of the melanins extracted from the mycelia of O. lascauxensis and O. anomala in order to known their chemical structure. The melanins from these two species were compared with those from other fungi. The melanins from the Ochroconis species have similar SERS and 13C and 15N NMR spectra. Their chemical structures as suggested by the data are not related to 3,4-dihydroxyphenylalanine, 5,6-dihydroxyindole or 1,8-dihydroxynaphthalene precursors and likely the building blocks from the melanins have to be based on other phenols that react with the N-terminal amino acid of proteins. The analytical pyrolysis of the acid hydrolysed melanin from O. lascauxensis supports this assumption. The purpose of this methodology is to help scholars determine the provenance of fragments of the scrolls and sort and compare them more easily than in the past. Since our measurements allowed us to differentiate between the minerals originating from the production processes and the sediments accrued during storage in the caves in Qumran, we were able to reconstruct the treatment of hides and discovered that at least two distinct parchmentproduction techniques co-existed at the beginning of the common era. The reconstruction of workmanship on animal hides in Antiquity raised doubts about the validity of the current definition of parchment, which is based on a production technique known from the Middle Ages. We hope that our current work will also help researchers understand the characteristic properties of gewil, qelaf and dukhsustos better.¹ The evolution and socio-geographic distribution of writing inks in late Antiquity and the early Middle Ages are the second focus of our investigative work at BAM. We use X-ray fluorescence analysis (XRF) to determine the chemical composition of inks and NIR reflectography for their typology. Although Geniza manuscripts have attracted a great deal of scholarly attention, the material aspects of these documents seem to have been largely neglected. The current study aims to obtain information on the composition of the writing supports and the inks that were used, the production of the original manuscripts and the history of their degradation. The collected data should supplement the description based on Hebrew palaeography and codicology. Moreover, a characterisation of the writing materials would provide insights on trade, local production techniques and social structures. The tabulated results have the potential to be used as geo-chronological markers if sufficient data is collected. For our study, we chose Hebrew legal documents and letters from three communities that co-existed in Fustat (today Cairo) in the 11th century: the Jerusalemite (or ‘Palestinian’) community, the Babylonian community and the Karaites, three different communities with different scribal traditions. In addition to this, we studied non-biblical scrolls attributed to the Palestinian and Babylonian communities. The question we addressed was whether we can correlate the use of a specific type of ink to an objective criterion. For our analysis at Cambridge University Library, we used a number of protocols developed at BAM (Bundesanstalt für Materialforschung und -prüfung) in Berlin and at the Centre for Study of Manuscript Cultures, University of Hamburg (CSMC). These include reflectographic examination followed by non-destructive X-ray fluorescence (XRF) analysis using transportable instruments so that the analysis can be carried out in situ. We have illustrated our approach here by providing two examples. The first one describes our investigation of the leather fragment containing a portion of the Babylonian Talmud on its hair side (TS Misc.26.53.17). The second example illustrates our ability to compare iron-gall inks using the XRF method. In this case, we explored the question of the codicological attribution of three fragments (TS F17.4, TS 12.755 and TS 12.756) to the same Talmud manuscript. However, there still remains significant uncertainty regarding their long term performance when used in steel‐reinforced structures. The durability of AAMs in such applications depends strongly on the corrosion behaviour of the embedded steel reinforcement, and the experimental data in the literature are limited and in some cases inconsistent. This letter elucidates the role of the chemistry of AAMs on the mechanisms governing passivation and chloride‐induced corrosion of the steel reinforcement, to bring a better understanding of the durability of AAM structures exposed to chloride. The corrosion of the steel reinforcement in AAMs differs significantly from observations in PC; the onset of pitting (or the chloride ‘threshold’ value) depends strongly on the alkalinity, and the redox environment, of these binders. Classifications or standards used to assess the severity of steel corrosion in PC appear not to be directly applicable to AAMs due to important differences in pore solution chemistry and phase assemblage. T1 - Detection of the electronic structure of iron-(III)-oxo oligomers forming in aqueous solutions N2 - The nature of the small iron-oxo oligomers in iron-(III) aqueous solutions has a determining effect on the chemical processes that govern the formation of nanoparticles in aqueous phase. Here we report on a liquid-Jet photoelectron-spectroscopy experiment for the investigation of the electronic structure of the occurring iron-oxo oligomers in FeCl3 aqueous solutions. The only iron species in the as-prepared 0.75 M solution are Fe3+ monomers. Addition of NaOH initiates Fe3+ hydrolysis which is followed by the formation of iron-oxo oligomers. At small enough NaOH concentrations, corresponding to approximately [OH]/[Fe] = 0.2–0.25 ratio, the iron oligomers can be stabilized for several hours without engaging in further aggregation. Here, we apply a combination of non-resonant as well as iron 2p and Oxygen 1s resonant photoelectron spectroscopy from a liquid microjet to detect the electronic structure of the occurring species. Specifically, the oxygen 1s partial electron yield X-ray absorption (PEY-XA) spectra are found to exhibit a peak well below the onset of liquid water and OH (aq) absorption. The iron 2p absorption gives rise to signal centered between the main absorption bands typical for aqueous Fe3+. Absorption bands in both PEY-XA spectra are found to correlate with an enhanced photoelectron Peak near 20 eV binding energy, which demonstrates the sensitivity of resonant photoelectron (RPE) spectroscopy to mixing between iron and ligand orbitals. These various signals from the iron-oxo oligomers exhibit Maximum intensity at [OH]/[Fe] = 0.25 ratio. For the same ratio, we observe changes in the pH as well as in complementary Raman spectra, which can be assigned to the Transition from monomeric to oligomeric species. At approximately [OH]/[Fe] = 0.3 we begin to observe particles larger than 1 nm in radius, detected by small-angle X-ray scattering. Due to their stability, they are still considered an environmental problem even though the use of most of them has been phased out several decades ago. Soil is a matrix which can retain these contaminants to a great extent. This ability is often associated with the total organic carbon content (TOC). In order to judge the pollution status of soil and to make monitoring data more easily comparable a simple, yet robust extraction method is needed. Agitation solid-liquid-extraction is well suited for this purpose. However, the influence of TOC on the analyte recovery has to be known. For the presented study, 12 organochlorine pesticides and 7 polychlorinated biphenyls were spiked into four model soils with organic carbon contents between 1.6% - 13.3%. The matrices were extracted using solid-liquid extraction between 45 minutes and 16 hours. For comparison, all soils were also extracted using pressurised liquid extraction and Soxhlet extraction. After clean-up the extracts were measured using a gas chromatography-mass spectrometry (GC-MS) system. Statistical analysis of the results implied that the TOC content of the soils did not have significant influence on the extraction efficiency. A longer solid-liquid extraction time did not necessarily increase analyte recovery: Extraction for one hour resulted in 88% recovery while 16 hour extraction led to 89%. Thus, the efficiency of all the methods was comparable for all model soils. Additional investigations regarding GC liner performance highlighted the need for isotopically labelled standards during the analysis of thermolabile pesticides. We report a low–molecular weight monomer whose supramolecular polymerization is triggered by the incorporation of water. The presence of water molecules as comonomers is essential to the polymerization process. The supramolecular polymeric material exhibits strong adhesion to surfaces, such as glass and paper. It can be used as a water-activated glue, which can be released at higher temperatures and reused many times without losing its performance. Taking high-rate loading into account such as with accidental scenarios, different standards such as ASTM E 1820 or BS 7448-3 provide some information on dynamic fracture mechanics testing. Nevertheless, the designations differ so that a validation of the own material specific test method used for dynamic R-curve determination is mandatory. In order to address this for ductile cast iron materials an experimental method for the reliable determination of dynamic J-integral crack resistance curves at -40 °C following the multiple specimen approach has been established and validated. The experimental concept offers some additional valuable features. Single values of dynamic crack initiation toughness can be determined using a single specimen technique based on crack sensors. Furthermore, an experimentally independent method is provided according to which CTOD δ5 R-curves can be established. The focus of the present paper is on the validation of the experimental low blow technique using a drop tower test system. A drop tower test system was developed and set up to perform low blow tests at temperatures down to -40 °C. The system allows for a variation of the impact mass and height and was optimized for testing of ductile cast iron at stress intensity rates from approximately 5∙10⁴ to 3∙10⁵ MPa√ms⁻¹. This range of loading rate is characteristic for instance with crash scenarios of heavy sectioned DCI casks for radioactive materials. In order to address characteristic challenges of impact tests (test duration of microseconds up to milliseconds, inertial effects, signal oscillations), an appropriate full bridge strain gage method for the measurement of force directly on the specimen as well as a non-contact measurement of load line displacement using an optical extensometer have been developed and validated. The low blow test requires either to prevent bouncing strikes of the hammer by using the stop block technique or to catch the hammer after its first strike. Both options are not part of the experimental concept and setup which have been realized here. The paper describes investigations which have been performed in order to make sure that bouncing strikes of the hammer do not cause additional crack extension in the specimen. This is necessary to ensure a unique relation between the work done and the achieved crack extension. The investigations covered the analysis of limit loads of the specimen with respect to the measured force. The measured stiffness of the specimen was assessed and signals of crack sensors were analyzed. Furthermore, an analysis of the mechanical behavior of the loading system and the specimen by optical observation was performed. Corresponding results are discussed in the paper. It had finally been proven that additional crack extension in the specimen due to bouncing strikes of the hammer is not to be expected under the given conditions of test setup, material and loading. T1 - Structural and magnetic properties of multi-core nanoparticles analysed using a generalised numerical inversion method N2 - The structural and magnetic properties of magnetic multi-core particles were determined by numerical inversion of small angle scattering and isothermal magnetisation data. The investigated particles consist of iron oxide nanoparticle cores (9 nm) embedded in poly(styrene) spheres (160 nm). A thorough physical characterisation of the particles included transmission electron microscopy, X-ray diffraction and asymmetrical flow field-flow fractionation. Their structure was ultimately disclosed by an indirect Fourier transform of static light scattering, small angle X-ray scattering and small angle neutron scattering data of the colloidal dispersion. The extracted pair distance distribution functions clearly indicated that the cores were mostly accumulated in the outer surface layers of the poly(styrene) spheres. To investigate the magnetic properties, the isothermal magnetisation curves of the multi-core particles (immobilised and dispersed in water) were analysed. The study stands out by applying the same numerical approach to extract the apparent moment distributions of the particles as for the indirect Fourier transform. It could be shown that the main peak of the apparent moment distributions correlated to the expected intrinsic moment distribution of the cores. Additional peaks were observed which signaled deviations of the isothermal magnetisation behavior from the non-interacting case, indicating weak dipolar interactions. The phase transition temperature was found to increase with increasing AN content in the copolymer, concentration of the solutions and copolymer chain length. A significant effect was observed onto the phase transition temperature by addition of different electrolytes into the copolymer solution. The copolymer chains were aggregated below the phase transition temperature and disaggregated above it. The size of the aggregates increases with increasing AN contents and concentration of the copolymer solutions below the phase transition temperature. The copolymer chains were expanded and weekly associated in solution above the phase transition temperature. A model is proposed to explain such association–aggregation behaviour of poly(AAm-co-AN) copolymers depending on AN contents and concentration of the copolymer solutions as a function of temperature. HABs can produce cyanotoxins, many of which are hepatotoxins. An ultra-performance liquid chromatography tandem mass spectrometry method was developed and validated for nine cyanotoxins across three classes including six microcystins, nodularin, cylindrospermopsin and anatoxin-a. The method was used to analyse free cyanotoxin(s) in muscle (n = 34), liver (n = 17) and egg (n = 9) tissue samples of 34 fish sourced from aquaculture farms in Southeast Asia. Conjugated microcystin was analysed by Lemieux oxidation to ascertain the total amount of microcystin present in muscle. Some tilapia accumulated free microcystin-LR in the muscle tissue at a mean of 15.45 μg/kg dry weight (dw), with total microcystin levels detected at a mean level of 110.1 μg/kg dw, indicating that the amount of conjugated or masked microcystin present in the fish muscle accounted for 85% of the total. Higher levels of cyanotoxin were detected in the livers, with approximately 60% of those tested being positive for microcystin-LR and microcystin-LF, along with cylindrospermopsin. Two fish from one of the aquaculture farms contained cylindrospermopsin in the eggs; the first time this has been reported. The estimated daily intake for free and total microcystins in fish muscle tissue was 2 and 14 times higher, respectively, than the tolerable daily intake value. This survey presents the requirement for further monitoring of cyanotoxins, including masked microcystins, in aquaculture farming in these regions and beyond, along with the implementation of guidelines to safeguard human health. Seit Einführung des Modells während des ersten Workshops zur Zuverlässigkeit der ZfP (European-American Workshop on Reliability of NDE) wurde das Modell in weiteren fünf Workshops stetig weiterentwickelt, diskutiert und an den Stand der Wissenschaft angepasst. Das Ziel dieser Publikation ist es, den aktuellen Stand der Diskussion und die neusten Fragestellungen der internationalen Zuverlässigkeitsgemeinschaft vorzustellen. Im Einzelnen werden die Entwicklung fortgeschrittener Modelle zur Bewertung der Probability of Detection (POD), Bewertungsansätze im Bereich Structural Health Monitoring und Fragestellungen zu den menschlichen Faktoren dargestellt. Auch die zerstörungsfreie Prüfung (ZfP) – bisher hauptsächlich als Endkontrolle oder Wartungselement wahrgenommen – kann und sollte im Wandel der Digitalisierung und Industrie 4.0 einen neuen Stellenwert erhalten. Die ZfP kann schon während der Konstruktion helfen, sichere und zuverlässige Produkte zu erstellen. Jedoch müssen dafür eine adäquate Informationskultur und die notwendigen Kennzahlen vorhanden sein. Objektive, probabilistische Kennzahlen (wie die POD - Auffindwahrscheinlichkeit) sind hierbei der Schlüssel. Diese müssen jedoch unbedingt an Prüfverfahren, Prüfgegenstand und Prüfbedingung angepasst werden, da sonst die gesamte Aussage in Frage zu stellen ist. Im Vortrag werden Beispiele aus der Anwendung gezeigt, die die Aussagekraft der Kennzahl aber auch die Sorgfalt, mit denen die einzelnen Aussagen genutzt werden, unterstrichen werden soll. Bewertung von Daten von realen Fehler, Anpassung an die Bruchfestigkeit und die Betrachtung von mehreren Prüfparametern am Beispiel der Bewertung von radiographischen, Ultraschall- und Wirbelstromprüfung werden in dem Vortrag gezeigt. T1 - Talbot-Lau Röntgen-Interferometrie für die zerstörungsfreie Prüfung N2 - Im vergangenen Jahrzehnt sind gitterinterferometrische Röntgenabbildungen zunehmend in den Fokus des Interesses gerückt. Sie sind insbesondere dann Vorteil, wenn der in der Standardradiographie erreichbare Kontrast nicht genügt, schwach absorbierende Mikro- und Nanostrukturen abzubilden. Auf Kosten mehrerer Aufnahmen gestattet die Talbot-Lau Gitterinterferometrie, die Beiträge aus Absorption, Refraktion und (refraktiver) Streuung zu trennen. Dazu werden die Störungen der Interferenzmuster von Phasengittern ausgewertet. Im Gegensatz zu einer Vielzahl anderer refraktionsbasierter Abbildungsmethoden kann diese Technik mit konventionellen Röntgenröhren (mit divergenter, polychromatischer Strahlung) angewendet werden. Damit ist die Technik geeignet, typische ZfP-Fragestellungen zu lösen. Hier stellen wir Untersuchungen zur Effizienz von Phasengittern vor. Die Visibilität (d.h. die Höhe der Oszillation im Interferenzmuster) wird als Funktion verschiedener geometrischer Parameter verfolgt. Eine Rotation um die Achse parallel zu den Gitterstegen ändert die Form des Gitterprofils (d.h. die Verteilung der Phasenschiebungen). Die kontinuierliche Variation des Winkels und des Detektionsabstandes führt zur Identifikation von ausgezeichneten Kombinationen mit maximaler Visibilität, wobei die Abstände deutlich geringer sind als im Standardaufbau mit senkrechter Gitterbestrahlung. Unsere Studie umfasst Simulationen für monochromatische Quelle und den Vergleich zu experimentellen Ergebnissen mit der Synchrotronstrahlung. In der Literatur wurden die Vorteile der Talbot-Lau Interferometrie für die zerstörungsfreie Prüfung von Faserkompositen und Verbundwerkstoffen demonstriert. Ziel ist die Gewährleistung der Sicherheit und Zuverlässigkeit der eingesetzten Bauteile. Aktive Thermografieverfahren ermöglichen die effiziente Prüfung großer Flächen mit hoher Auflösung in wenigen Arbeitsschritten. Ein wichtiges Teilgebiet der Prüfungen ist die Ortung und Charakterisierung von Delaminationen, die sowohl bereits in der Fertigung als auch während der Nutzung eines Bauteils auftreten können, und dessen strukturelle Integrität schwächen. In diesem Beitrag werden CFK-Strukturen mit künstlichen und natürlichen Delaminationen mit Hilfe unterschiedlich zeitlich modulierter Strahlungsquellen experimentell untersucht. Verwendet werden dabei Anregungen mit Blitzlampen und mit frequenzmodulierten Halogenlampen. Mittels Filterfunktionen im Zeit- und Frequenzbereich wird das Kontrast-zu-Rausch-Verhältnis (CNR) der detektierten Fehlstellen optimiert. Verglichen werden anschließend die Nachweisempfindlichkeit, das CNR und die Ortsauflösung der zu charakterisierenden Delaminationen für die unterschiedlichen Anregungs- und Auswertungstechniken. T1 - Verbesserung und Qualifizierung der Ultraschallprüfung von Mischnähten im Primärkreis von KKW N2 - Die im Primärkreislauf von Kernkraftwerken anzutreffenden austenitischen Schweiß- und Mischnähte sind nicht nur extrem sicherheitsrelevant, sondern stellen auch sehr hohe Anforderungen an die Materialprüfung. Neben der eingeschränkten Zugänglichkeit ist das makroskopische Gefüge der Schweißnaht für die Prüfung mit Ultraschall von höchster Bedeutung. Um Materialfehler zuverlässig in Position und Größe bestimmen zu können, müssen die Kornorientierungen und die elastischen Konstanten des anisotropen Schweißnahtgefüges bekannt sein. Für die bildgebende Darstellung möglicher Materialfehler kommen folgende Arbeitsschritte zum Einsatz: Zunächst wird die Schweißnaht durchschallt, um mit einem inversen Verfahren wichtige Schweißnahtparameter wie beispielsweise die Kornorientierung bestimmen zu können. Auf der Basis dieser Parameter werden im nächsten Schritt die Schallwege mittels Raytracing (RT) simuliert. Zuletzt werden dieser RT-Simulation die Messdaten (A-Scans) von verschiedenen Sender- und Empfängerpositionen zugeordnet und nach der SAFT-Methode (Synthetic Aperature Focusing Technique) zeitaufgelöst überlagert. Die Kombination aus inversem Verfahren, RT und SAFT gewährleistet auch in anisotropen Werkstoffen eine ortsrichtige Visualisierung der Fehler. Wir erläutern diese drei Verfahren und stellen die Prüfanordnung von Prüfkörpern mit künstlichen Testfehlern vor. Biegebalken finden unter anderem Anwendung in Form von Brückenträgern oder Unterzügen. Die bei maximaler Beanspruchung auftretenden Versagensformen unterteilen sich dabei in Biege- und Querkraftversagen (Schubversagen). Ein Versagen des Balkens kann je nach Versagensmechanismus mit oder ohne Vorankündigung erfolgen. Gebräuchliche Bemessungskonzepte der Baupraxis zur Dimensionierung entsprechender Tragelemente zielen auf ein Versagen mit ausreichender Vorankündigung (große Verformungen oder breite Risse) ab, wie es z.B. beim sekundären Biegedruckversagen entsteht. Für die Zustandsüberwachung von Bauwerken (z.B. Brücken) mittels zerstörungsfreier Prüftechniken sind fundierte Kenntnisse zu den bei Biegebeanspruchung auftretenden Schadensmechanismen erforderlich. Im Rahmen eines BAM-internen Projektes sollen durch praxisnahe Untersuchungen an einer brückenähnlichen Referenzstruktur unterschiedliche auf das Tragwerk einwirkende thermische und mechanische Belastungen untersucht werden und in die Entwicklung eines Monitoringsystems einfließen. Anhand der aus Biegeversuchen vorliegenden Ergebnisse wird der Einsatz unterschiedlicher Methoden der zerstörungsfreien Materialprüfung gezeigt und deren Anwendbarkeit für die Analyse des Verformungsverhaltens von Stahlbetonbalken diskutiert. Hierzu wurde das Verformungsverhalten eines Stahlbetonbalkens (L = 2,75 m) in einem Vierpunkt-Biegeversuch im Labormaßstab untersucht. Das Hauptaugenmerk liegt dabei auf der Charakterisierung der ausschlaggebenden Versagensmodi durch die einzelnen zerstörungsfreien Prüfverfahren und der Vergleich zu klassischen Messmethoden (z.B. Depending on the annealing temperature, micro-Raman spectroscopy analyses evidenced different relaxation behaviours, associated to bridging and non-bridging oxygen structures present in the glass network. The results indicate that the mobility of lithium ions is an important factor during the glass modification with fs-laser pulses. Quantitative phase contrast imaging (spatial light interference microscopy) revealed that these fs-laser induced structural modifications are closely related to local changes in the refractive index of the material. The results establish a promising strategy for tailoring fs-laser sensitivity of glasses through structural mobility. T1 - Synchrotron-Refraktion mit in-situ Zugbelastung zur Charakterisierung der Schadensentwicklung in Metall-Matrix- Kompositen N2 - Die Synchrotron-Refraktions-Topographie basiert auf der Brechung von Röntgenstrahlung an den Grenzflächen zwischen Materialien mit unterschiedlichem Brechungsindex. Refraktionseffekte verändern die Ausbreitungsrichtung der Röntgenstrahlung, welche mit Hilfe der Methode des Analyser-Based Imaging analysiert wird. Dabei wird ein Silizium-Einkristall im Strahlengang zwischen Probe und Detektor platziert. Dieser Analysator-Kristall filtert, innerhalb seiner Streuebene, die Röntgenstrahlung gemäß ihrer Ausbreitungsrichtung. Durch das Rastern verschiedener Winkelpositionen des Analysator-Kristalls erhält man das Winkelspektrum der von der Probe gebrochenen Strahlung. Aus dieser Rocking Curve lässt sich die spezifische innere Oberfläche der Probe bestimmen. Diese Information wird über einen 2D-Detektor mit einer nominellen Pixelgröße von 3,6 μm gewonnen. Die Installation einer Zugprüfmaschine ermöglicht es die Proben in verschiedenen Lastzuständen insitu zu untersuchen. Damit ist es möglich die spezifische innere Oberfläche in Abhängigkeit der Last darzustellen und so Schädigungsmechanismen abzuleiten. Der Vorteil bei der Ausnutzung des Refraktionseffekts gegenüber der einfachen Durchstrahlung ist die höhere Sensitivität gegenüber Defekten mit geringem Volumen aber großer Oberfläche. Somit werden auch Mikrorisse und, speziell bei Kompositen, die Enthaftung zwischen Matrix und Verstärkung (Partikel, Fasern) erfasst. In dieser Studie wurden Metall-Matrix-Komposite bestehend aus einer Aluminium-Matrix verstärkt mit Aluminiumoxid-Partikeln untersucht. Hierzu wurden knochenförmige Zugproben präpariert, deren Messlänge einen rechteckigen Querschnitt von 1,7 mm × 1,3 mm aufwies. Die Proben wurden einem Zugversuch unterzogen, welcher in Intervallen unterbrochen wurde. Bei jeder Unterbrechung wurde unter quasi-statischer Last die Rocking Curve gemessen und ein Radiogramm der spezifischen inneren Oberflächen berechnet. Dabei konnte ab dem Erreichen einer Last von ca. 600 N ein Anstieg der spezifischen inneren Oberfläche beobachtet werden. Demgegenüber zeigen die zum Vergleich durchgeführten Durchstrahlungsaufnahmen keinerlei Defekte. They are used to enhance the absorption of light of a reaction center which is often represented by a single acceptor. Here, we present different light harvesting systems on DNA origami structures and analyze systematically the light harvesting efficiency. By changing the number and arrangement of different fluorophores (FAM as donor, Cy3 as transmitter and Cy5 as acceptor molecules) the light harvesting efficiency is optimized to create a broadband absorption and to improve the antenna effect 1 (including two energy transfer steps) from 0.02 to 1.58, and the antenna effect 2 (including a single energy transfer step) from 0.04 to 8.7, i.e. the fluorescence emission of the acceptor is significantly higher when the light-harvesting antenna is excited at lower wavelength compared to direct excitation of the acceptor. The channeling of photo energy to the acceptor proceeds by Förster Resonance Energy Transfer (FRET) and we carefully analyze also the FRET efficiency of the different light harvesting systems. Accordingly, the antenna effect can be tuned by modifying the stoichiometry of donor, transmitter and acceptor dyes, whereas the FRET efficiency is mainly governed by the spectroscopic properties of dyes and their distances. This is carried out exemplary on plate girders from two structural steel grades. The calculated residual stresses are compared with measurements by sectioning method. It has been repeatedly stated that present Eurocode models fail to approximate the residual stresses. Especially for high strength steel (HSS) only limited information is available on realistic occurring residual stresses in typical I-girders. The investigations are aimed to give further guidance on these values. A few proposals on advanced models are discussed.
