Ecotoxicological effects of potentially generated degradation products of plastic materials in the storage water (mineral water, pH: 7.2) were detected by algae growth inhibition tests with Desmodesmus suspicatus and photosystem II inhibition tests with Raphidocelis subcapitata. Results clearly indicate inhibitory effects on algae growth by contaminants in the storage water of stressed plastic materials with increasing growth inhibition of proceeding hydrolysis and UV stress times. Here, different plastics cause variable responses of algal growth. First chemical analyses indicate dissolved monomers and their oxidation products of plastic materials as possible driver of detected ecotoxicological effects, since detected microplastic particles do not seem to harm algae. Here we train neural networks to use raw measurement data as input and immediately return the desired output parameters. We discuss the example of X-ray reflectivity measurements of ultrathin films, which contain reciprocal space information and must traditionally be fitted with dynamic scattering theory. Instead, we train the neural network with simulated X-ray data of multilayer structures and then apply it to measurement data. The neural network yields high accuracy results, is robust against noise, and performs significantly faster than fitting algorithms in real-time experiments. The presented neural network data analysis is becoming increasingly attractive, because free software has become very accessible and specialized computer chips (NPU) are currently being rolled out. This paper presents recent advances from two projects that use Artificial Neural Networks (ANNs) to address the challenges of automation and performance-efficient realizations of MS and NMR. In the first part, a complete toolchain has been developed to develop simulated spectra and train ANNs to identify compounds in MS. In the second part, a limited number of experimental NMR spectra have been augmented by simulated spectra to train an ANN with better prediction performance and speed than state-of-theart analysis. These results suggest that, in the context of the digital transformation of the process industry, we are now on the threshold of a possible strongly simplified use of MS and MRS and the accompanying data evaluation by machine-supported procedures, and can utilize both methods much wider for reaction and process monitoring or quality control. Therefore, many studies focus on investigations regarding their toxicological potential. This includes investigations concerning uptake, distribution and excretion of the particles. So far, little attention was paid to changes of physical and chemical properties in the human body. During processes like digestion, the question arises whether they can pass this barrier in a nanoscale form. In this study we analytically monitored the changes in the size distribution of colloidal silver during an artificial digestion process with the help of small angle x-ray scattering (SAXS). Therefore, we synthesized polyacrylic acid stabilized ultra-small silver nanoparticles with a radius of 3 nm and a size distribution width of 18%. The artificial digestion process mimics the gastro-intestinal passage and simulates the oral, gastric and small intestinal conditions. Additionally, food components like oil, starch, glucose and skimmed milk powder are used to provide a preferably realistic environment. In absence of any food components the low pH initiates aggregation of the particles in the stomach. However, the particles unexpectedly stabilize in a defined cluster form with a mean radius of 12 nm. By the use of the food components oil and starch we observed that the particles are dispersed again. Now we found a bimodal size distribution of primary particles and aggregates. In contrast to that, with skimmed milk powder only a slight aggregation occurs in the stomach. In the gastric tract the particle distribution is stabilized at a mean volume weighted radius of 5 nm. Hence, skimmed milk powder acts as a colloidal stabilizer. For comparison we also used silver nitrate as a control substance. Surprisingly, we observed a formation of nanoparticles already in the saliva. During the digestion process the distribution narrows and finally in the intestine it shows a stable distribution with a mean volume weighted radius of 3 nm and a small fraction of aggregates. These results indicate that the silver nanoparticles can pass the digestion process in a nanoscale form but undergo a transformation in the size distribution. However, even from pure silver nitrate nanoparticle formation can be observed. This sketches a complex mechanism in which not only food components but also silver ions cause changes in nanoparticle size and aggregation. T1 - Artificial digestion of aluminium-containing nanomaterials and their effects on the gastrointestinal tract in vitro N2 - Although aluminium is one of the most common elements in the biosphere, up to now little is known about its impact on human health. aluminium and its chemical derivatives are highly abundant in food, food contact materials and consumer products. Humans are exposed to aluminium via the gastrointestinal tract (GI tract). Exposition can change substantially due to consumer behavior since aluminium is also a compound of numerous food additives. Recently, aluminium exposition is increasingly considered to cohere with cancer and neurodegenerative disorders. Lately, due to an increasing attentiveness on this topic, limiting values for food additives have been tightened by the EFSA. However, cellular effects of aluminium and especially aluminium-containing nanomaterials, that represent a significant part of chemicals found in food products, are widely unknown and in the focus of our research activities, for example in the bilateral SolNanoTOX project. We established an in vitro simulation system of the GI tract, where nanomaterials undergo the different physiological, chemical and proteinbiochemical conditions of saliva, gastric juice and the intestine. The artificially digested nanomaterials, as well as soluble aluminium chloride as ionic control substance, were subjected to several analytical and biochemical methods to characterize their change of appearance and their cytotoxic effects on intestinal cellular models. We observed the fate of the nanomaterials during typical pH-values of saliva, gastric and intestinal juice with Dynamic light scattering measurements and ICP-MS in the single particle mode. After observable disappearance at pH 2 the particles recovered in the simulated intestinal fluid. The simulation of the GI tract, mainly the change of pH settings, can lead to a certain chemical activation of aluminium that can increase bioavailability in the intestine after oral uptake of aluminium-containing food products. In vitro assays like CTB, MTT and cellular impedance measurements showed that there were no acute cytotoxic effects measurable after a period up to 48h after incubation, comparable to undigested particles. In contrast, high amounts of aluminium ions showed synergistic effects on cell viability compared to non-digested aluminium ions. Although toxicological potential of Al ions to healthy tissue appears to be low, increased hazardous potential cannot be ruled out to pre-damaged tissue and can have a relevance in risk assessment for special consumer groups with for example chronical intestinal inflammation or dietary eating behavior combined with high exposure to Al-containing food products. Die damit verbundenen Vorteile wie die aktive Fokussierung oder, die gezielte Manipulation des Einschallwinkels zur Anregung von geführten Wellen, finden im Bereich der luftgekoppelten Prüfung dabei noch wenig Anwendung. The aminated aroyl-S,N-ketene acetals are emissive in the solid state and in the aggregate, but not in solution, thus, they are AIEgens (aggregation induced emission chromogens). Aggregation is induced by fractional alternation of the solvent mixture, here by increasing the water fraction of ethanol/water mixtures. For most derivatives, the emission upon induced aggregation stems solely from the aroyl-S,N-ketene acetal chromophore, regardless whether excitation occurs at the absorption maximum of the triarylamine or the aroyl-S,N-ketene acetal. Therefore, a pronounced energy transfer from the triarylamine donor to the aroyl-S,N-ketene acetal acceptor can be inferred. The color of the emission can be controlled by choosing the para-aroyl substituent. A partial energy transfer could also be observed for some bichromophores, leading to aggregation-induced dual emission (AIDE). In addition, four examples of aminated diaroyl-S,N-ketene acetals were added to the compound library. The electron-withdrawing properties of the additional aroyl group provide a bathochromic shift of the emission band of the aroyl-S,N-ketene acetal. These bichromophores also show AIDE and in one case even aggregation-induced white light emission as a result of additive color mixing. This way, truffles can be harvested faster, cheaper and more efficiently compared to the current method using dogs. For this purpose, suitable marker substances in the aroma of ripe truffles were identified using analytical-chemical methods. The collected data was used to create aroma profiles of different truffle species. The specificity of possible marker substances was confirmed with a truffle dog study. The hypothesis “If trained truffle dogs recognize the substances as supposed truffles in the context of an experiment, they can be regarded as specific” was made. The results show that the truffle aroma is mainly characterized and perceived by dogs by dimethyl sulfide and dimethyl disulfide. For the concentration measurement of these markers in the ambient air, we are currently building our own detector. The Challenge is to get a fast measurement with a very low limit of detection at the same time. The device will resemble a metal detector in terms of shape and handling and be particularly suitable for mobile use. The R&D cooperation project between the Federal Institute for Materials Research and Testing (BAM) and sglux GmbH thus enables truffles to be harvested professionally and systematically. They range from writing material analysis, mostly with X-ray fluorescence (XRF), permitting non-invasive characterization of inks and pigments used, to the investigation of the origin of writing supports. In addition, XRF mapping has proved to be an invaluable tool for recovering erased text. Finally, computed-tomography (CT) has shown potential in virtually unrolling rolls, making text readable without using-damaging mechanical methods. Despite their growing use, little attention has been paid to the side effects of such analytical tools. We observed irreversible parchment colour changes during some experiments on dead-sea scrolls with synchrotron radiation sources. Furthermore, partial photo-reduction of iron under high intensity beam during X-ray absorption near edge structure spectroscopy (XANES) measurements of iron-gall ink on paper has been reported several times [5,6]. Such phenomena have mostly been overlooked so far, although there is an increasing awareness of the necessity to study them. We conducted experiments at the Deutsches Elektronen-Synchrotron (DESY) facilities to investigate X-ray induced structural alteration of paper and parchment to see whether the presence of absorption centres (ink and pigments) has an impact. In addition to better understanding degradation processes, we are aiming to define an appropriate methodology of analysis of manuscripts with a tolerable risk of damage. The first results concerning X-ray induced damage of cellulose materials have already been presented at the Synchrotron Radiation and Neutrons in Art and Archaeology (SR2A). We are focusing here on the results on parchment materials. Die Anforderungen an die Schweißzusätze für das Extrusionsschweißen wurden angepasst. Derzeit gibt es intensive Diskussion über die Entlassung von Deponien aus der Nachsorge. Sind Geokunststoffe in den Böschungen einer Deponie verwendet worden, wäre eine Entlassung aus der Nachsorge demnach im Moment nicht möglich. Die Deponie müsste dafür nach der Deponieverordnung (DepV) u. a. „insgesamt dauerhaft standsicher“ sein. Geokunststoffe erfüllten ihre Funktion über einen Zeitrahmen von mindestens hundert Jahren. Die Dauerhaftigkeit wird bei der Diskussion aber offenbar als „Ewigkeit“ verstanden. Dies stehe somit im Widerspruch – so die Argumentation. The plastic litter that pollutes our environment leads to microplastic particles. They can be found (nearly) everywhere. The processes that lead to microplastic can also form nanoplastic particles, which have a size below 1 µm. Because of the small size they can penetrate tissue more easily. Only few risk assessment studies of nanoplastics were carried out so far. Using polystyrene (PS) nanoparticles to test effects on organisms is easy because it is commercially available. However, this falls a little short, as the polyolefins i.e., polypropylene (PP) and polyethylene (PE), are produced in a larger proportion than PS. Moreover, these plastics are mainly used for disposable products, which means that they also account for a large proportion of plastic waste. Therefore, the percentage of polyolefins in environmental nanoplastic is presumably high. It is important to test the toxicological effects also with nanoplastics made of PP and PE to have more realistic results. Herein, we present an easy and repeatable method to prepare an aqueous dispersion of polypropylene nanoplastics (nano-PP). They are stabilized electrostatically, resulting in a strongly negative zeta potential of -43 mV (± 2 mV) and making no surfactant necessary to keep the dispersion stable. The size and the size distribution were determined via Dynamic Light Scattering (DLS) and gives a hydrodynamic diameter of 180.5 nm (± 5.8 nm) and a PDI of 0.084 (± 0.023). Finally, ca. 480 bottles of the dispersion with a volume of 10 mL each were prepared to serve as a potential reference material for further testing of detection methods or risk assessments. The potential dangers of these pollutants remain unpredictable. While risk assessment studies on microplastics are already popular, nanoplastic has not yet reached the same focus of investigation. The reason for this difference is simple: There is a "methodological gap" in the analytics of plastic particles with a diameter smaller than 1 µm. Submicron and nanoplastic particles are currently not detectable in environmental matrices. Therefore, it is important for researchers to have a well-characterized nanoplastic material, that serves as a reference for nanoplastics found in nature. Our aim was to synthesize nanoplastics made from the most common used plastics such as polypropylene (PP). We found an easy way to form nanoparticles consisting of PP (nano-PP), adapting and improving the method presented for polystyrene (PS). Nano-PP was formed via a top-down method where the polymer was dispersed to acetone and then transferred to water. No additional surfactant is needed to obtain a dispersion which is stable for more than 35 weeks. The success of forming nanoplastics and their size was detected via scattering methods, predominantly dynamic light scattering (DLS). The chemical analysis of the nanoplastics was performed via Fourier Transform Infrared spectroscopy. To examine the good stability of the nanoparticles, zeta potential measurements were performed, which revealed zeta potentials of -30 to -40 mV. Seit dem Erscheinen der Deponieverordnung (DepV) im Jahre 2009 bedürfen Geokunststoffe, Polymere und Dichtungskontrollsysteme einer Zulassung durch die BAM, wenn diese in deutschen Deponieabdichtungssystemen eingesetzt werden sollen. Die DepV stellt dabei allgemeine Anforderungen an die Geokunststoffe und den Zulassungsprozess für die Produkte auf. Geokunststoffe müssen danach ihre Funktion unter allen zu erwartenden äußeren und inneren Einflüssen über einen Zeitraum von mindestens 100 Jahren erfüllen. Dichtungskontrollsysteme müssen ihre Funktion über mindestens 30 Jahre erfüllen. Die Anforderungen der DepV werden durch die Zulassungsrichtlinien der BAM in konkrete technische Anforderungen für die Produkte aber auch für das Qualitätsmanagement und den Einbau etc. überführt. Bei der Erarbeitung der Zulassungsrichtlinien wird die BAM durch einen Fachbeirat, der sich aus Experten aus allen relevanten Bereichen zusammensetzt, beraten. Der Vorsitz des Fachbeirats liegt beim UBA. Es soll eine kurze Übersicht über die Produkte und die wichtigsten Anforderungen gegeben werden. As they are increasingly tuned and modified for specific analytes, a better understanding of the surface chemistry and morphology of the inner channel is necessary. With the aim of developing a comprehensive approach for characterisation of such nanopipettes, this research focuses on combining surface-sensitive analysis methods with advanced sample preparation techniques. Quartz substrates were modified by gas phase silanization, a well-utilised technique in the field to enhance performance of nanopipettes, and further functionalised with a metal bis thiolate complex, to aid in chemical analysis. The sample characterisation involved scanning electron microscopy (SEM), low-energy dispersive x-ray spectroscopy (EDX), time-of-flight secondary ion mass spectrometry (ToF-SIMS) and Auger electron spectroscopy (AES). Using focused ion beam (FIB) milling under gentle conditions, the inner surface of quartz nanopipettes was exposed whilst preserving the integrity of the overall structure (see figure). Owing to the challenging analysis conditions, modification and analysis of flat quartz substrates has been performed in parallel for optimisation purposes. The results demonstrate the first steps towards full characterisation of nanopipettes at the nanoscale, notably with access to the inner channel. The methods used here can be applied to gain further understanding of the response of these sensors to complex analytes and allow for the study of different surface functionalisations at the all-important sensing region. However, besides their current prominence in the field of medical diagnostics, rapid tests and assays have also become increasingly important in other areas ranging from food and feed via security and forensics to environmental management. The advantage is obvious: taking the assay to the sample instead of bringing the sample to the laboratory minimizes the time between first suspicion and first decision taking. Especially today, where mobile communication devices with powerful computing capabilities and onboard cameras are omnipresent, the majority of the global population possesses the basic skills of operating a powerful detector, ready at their fingertips. The stage is thus set for a much broader use of analytical measurements in terms of prognostics and prevention. Today, however, tests for single parameters are still prevailing, whether it is a SARS-CoV-2 biomarker, the glucose level in blood or the concentration of lead in a water sample. Also, for industrial use, many mobile analytical systems still target a single parameter, and several separate runs or tests have to be used even if one wants to screen for only a small number of key parameters. Current challenges in the field thus lie with the development of low-number multiplexing strategies while allowing for robust, reliable, fast, and user-friendly operation and while still reaching the required sensitivities. This lecture will give an overview of various generic approaches developed in BAM’s Chemical and Optical Sensing Division over the years to address these challenges. In particular, it will highlight how the combination of supramolecular (bio)chemistry, luminescence detection, hybrid (nano)materials and device miniaturization can result in powerful (bio)analytical assays that can be used at a point-of-need. The RM used for these kinds of PT are poorly regulated, nevertheless with a more and more general acceptance of accreditation in the field of RM production and PT provision, there is an ever increasing interest in assessing producers and providers according to rules already well accepted in the field of quantitative analysis. The basic governing document, ISO 17034:2016, is written in a form that, at least for the overwhelming majority of requirements, may be applied to both qualitative and quantitative RM. However, problems remain. In particular, the expression of uncertainty of a purely qualitative result is still unresolved, and under discussion, the latter now lasting already dozens of years. Following aspects have to be considered concerning the determination of an appropriate length of the bar to obtain maximum damage to the specimen: the design of the package, its drop orientation and the impact point as well as a pre-damage of the package resulting from a previous 9m drop test. According to the Regulations the minimum length of the bar is defined as 20 cm, which has to be adapted to the outer surface of the package, especially to packages with large impact limiters. In this context, two main aspects are important: First, the realisation of a maximum puncture load applied to the package while no other component of the package has contact with the impact target. Second, the reduction of the risk of buckling and maximising the stiffness of the bar by a length which is as short as possible. In order to optimize the bar length, finite element calculations are often done by simulating the global behaviour of the package during the puncture test. The evaluation of the conducted puncture test regarding IAEA compliance is done by analysing deceleration measurements. The paper presents a possible approach to determine the length of the bar and the construction of a form-fitted connection to the target showing various examples of puncture tests. N2 - Time series analysis is a well-established field within the machine learning community, with two prominent applications being time-series forecasting, i.e., surrogate models, predicting the next time step for the systems outputs, and time-series classification, where complete timeseries are mapped to discrete labels, e.g. a sensor is either working or defective. Time-Series Extrinsic Regression (TSER), however, is a method for predicting continuous, time-invariant variables from a time series by learning the relation between these underlying parameters and the complete dynamic time series of the outputs without focusing on the recent states. E.g., it can be used to predict the heart rate based on an ECG signal. TSER as a research field was only established in 2021, but it is gaining traction ever since and it is used e.g. in the field of manufacturing technology to predict steel surface roughness from laser reflection measurements. It is applied, when there are no models available. Parameter Estimation (PE) is a common task in chemical engineering. It is used to adjust model parameters to better fit existing dynamic models to experimental time series data. This becomes more challenging in higher dimensions and for dynamic systems, where sensitivity and identifiability may change over time. There already exists a multitude of algorithms to solve the problem, including second-order methods that leverage information from Jacobian and Hessian matrices, as well as gradient-free optimization techniques, such as particle swarm optimization (PSO) or simulated annealing. However, with the growing establishment of machine learning (ML) in an increasing number of domains, the question arises as to whether, and if so, how, ML in general and TSER in particular can be employed to solve PE problems. This study marks the first application of TSER to PE problems. A comparative analysis is conducted between TSER and PSO, in terms of prediction accuracy, computational cost and data efficiency. We investigate, whether it is viable to use TSER, when there is a model available. Our methodology to regress model parameters via ML builds on the typical assumption, that a structurally correct and rigorous model, which can be simulated at low cost, is available. At the beginning, the boundaries of the parameter space are defined. This space is then sampled using Sobol sequences and the model is simulated. The resulting trajectories, along with their corresponding parameters, constitute the training data set. These trajectories are transformed through application of the “RandOm Convolutional Kernel Transform” method resulting in novel features, which are subsequently used to train the regressor model. This regressor returns predictions for the parameters. In a case study, the method is applied to predict the heat transfer and kinetic parameters of a batch reactor based on simulated data. However, real measurements are often not continuously available, but are taken only at rare, discrete points in time, and different variables are measured at different, asynchronous intervals. This is also mimicked in the synthetic training data, so the influence of heterogeneity on the results can be shown and over- or undersampling strategies are applied to counteract the effect. The EU-funded project TREEADS and the so-called German pilot within the project are concentrating on wildfire in Europe. Extensive experimental research is done on local vegeta-tion in Germany and the experimental results are used to adjust the Fire Dynam-ics Simulator (FDS) to local vegetation as a fuel in the calculations. The particle model and the boundary fuel model are developed for the simulation of forest fires. Both models require the kinetic input for the dehydration, pyrolysis, and char oxidation process. This three-step simplified model describes the basic fuel combustion kinetic. Most published simulations use the default kinetic input of pine needles from FDS user manual. To adjust the simulation to local vegetation, the corresponding kinetic parame-ters have been experimental investigated. Samples of pine needles, moss and other falling leaves were collected, air dried and grinded into powder. The TG analysis of all these samples were done under different heating rate of 5, 10 and 20K/min. The activation energy and the corresponding pre-exponential factor were calcu-lated. The results show that fire spread depends significantly on the vegetation and comparison with small scale experiments show good agreement using the new kinetic parameters. The new model is applied to a larger scenario and will be compared to the results of large-scale experiments for further validation of the model.
