On the other hand, extraterrestrial samples contain largely racemic compounds. The same is true for any common organic synthesis. Therefore, it has been a perplexing puzzle for decades how these racemates could have formed enantiomerically enriched fractions as a basis for the origin of homochiral life forms. Numerous hypotheses have been put forward as to how preferentially homochiral molecules could have formed and accumulated on Earth. In this article, it is shown that homochirality of the abiotic organic pool at the time of formation of the first self-replicating molecules is not necessary and not even probable. It is proposed to abandon the notion of a molecular ensemble and to focus on the level of individual molecules. Although the formation of the first self-replicating, most likely homochiral molecule is a seemingly improbable event, on a closer look, it is almost inevitable that homochiral molecules have formed simply on a statistical basis. In this case, the non-selective leap to homochirality would be one of the first steps in chemical evolution directly out of a racemic “ocean”. Moreover, most studies focus on the chirality of the primordial monomers with respect to an asymmetric carbon atom. However, any polymer with a minimal size that allows folding to a secondary structure, would spontaneously lead to asymmetric higher structures (conformations). Most of the functions of these polymers would be influenced by this inherently asymmetric folding. To summarize, simple and universal mechanisms may have led to homochiral self-replicating systems in the context of chemical evolution. A homochiral monomer pool is deemed unnecessary and probably never existed on primordial Earth. N2 - Homochiralität ist ein offenkundiges Merkmal des Lebens auf der Erde. Es wurden zahlreiche Hypothesen darüber formuliert, wie sich bevorzugt homochirale Moleküle auf der Erde gebildet und angereichert haben könnten. In diesem Artikel wird gezeigt, dass die Homochiralität des abiotischen organischen Pools zum Zeitpunkt der Bildung der ersten selbstreplizierenden Moleküle nicht notwendig und nicht einmal wahrscheinlich ist. As support, epoxy slides were manufactured by coating microscope glass slides with epoxy silanes. The manufactured epoxy slides showed similar capacity for protein immobilization as commercially available NEXTERION slides, but showed a higher autofluorescence background. On the epoxy slides, protein A, Cys-A, G and Cys-G were immobilized as antibody capturing coating. It could be shown that the Cys-tag considerably increased the immobilization of all compared proteins, especially of protein G. Protein Cys-G immobilized at pH 8-9 was considered the most suitable protein for antibody immobilization due to good spot uniformity and a high binding capacity for IgG. Additionally, in immobilization experiments it could be shown, that 10-500 fold excess of bovine IgG, introduced by two different fetal bovine serum (FBS) products, showed no significant competition on the immobilization of mice or goat IgG on protein Cys-G. To examine the captured antibodies for affinity to CBZ, the CBZ-TOTA-DY654 tracer was custom synthesized. It could be demonstrated, that the tracer antibody interaction was competitive with the tracer precursor, CBZ-TOTA-NH2 and CBZ. Furthermore, the synthesized tracer was also successfully used by externals to evaluate CBZ affine hybriodoma cells in flow cytometry. In the simulated screening, five antibodies with known affinity to CBZ were diluted in a cell culture medium. The screening method allowed to differentiate between affine antibodies (IC50 < 20 µg/ml) and unaffine antibodies (IC50 > 20 µg/ml), when concentrations of 0.1 µg/ml IgG or greater have been used. Of five applied antibodies, all highly affine clones (CE2 and B3212M) were reported as positives while no false-positive samples were observed. The screening included competitive, with around 89500 fold excess of CBZ, and non-competitive tracer incubation on different segments of the same chip, which resulted in total signal suppression on the competitive segment. The screening was performed in 20 hours, only a few nanoliters, of each simulated supernatant, were consumed in the screening process, furthermore in the method standard 96 well MTPs were used and no cleanroom facilities were required. Therefore, we have chosen porous glass filters that are available with different pore sizes as raw materials to prepare monolithic columns. We purchased VitraPOR 4 (10-16 μm pore size) and VitraPOR 5 (1.0-1.6 μm pore size) monolithic glass filters. The physical properties of these glass filters were characterized. The surface area, pore size distribution and the porosity were determined using mercury intrusion porosimetry and BET. These glass filters only exhibit flow through pores and therefore show no bimodal pore size distribution in the mercury intrusion curves. Due to their low permeability, the applied filters that exhibit an inner diameter of 8.0 mm and a length of 15.0±0.1 mm could be operated at flow rates more than 10 ml/min. High flow rates are favorable for fast separation experiments. Sequencing of hybridoma clone was performed using a cost-effective Sanger-based workflow that includes DNA-level subclass determination and peptide mass fingerprint confirmation. The κ-chain sequence was assigned to IGKV4-6101 / IGKJ101, while the heavy chain was assigned to IGHV2-601 / IGHD1-101 / IGHJ3*01 with IgG2c constant regions from the NOD strain. T1 - Simple Determination of Affinity Constants of Antibodies by Competitive Immunoassays N2 - The affinity constant, also known as the equilibrium constant, binding constant, equilibrium association constant, or the reciprocal value, the equilibrium dissociation constant (Kd), can be considered as one of the most important characteristics for any antibody-antigen pair. Many methods based on different technologies have been proposed and used to determine this value. However, since a very large number of publications and commercial datasheets do not include this nformation, significant obstacles in performing such measurements seem to exist. In other cases where such data are reported, the results have often proved to be unreliable. This situation may indicate that most of the technologies available today require a high level of expertise that does not seem to be available in many laboratories. In this paper, we present a simple approach based on standard immunoassay technology that is easy and quick to perform. It relies on the effect that the molar IC50 approaches the Kd value in the case of infinitely small concentrations of the reagent concentrations. A two-dimensional dilution of the reagents leads to an asymptotic convergence to Kd. The approach has some similarity to the well-known checkerboard titration used for the optimization of immunoassays. A well-known antibody against the FLAG peptide, clone M2, was used as a model system and the results were compared with other methods. This approach could be used in any case where a competitive assay is available or can be developed. The determination of an affinity constant should belong to the crucial parameters in any quality control of antibody-related products and assays and should be mandatory in papers using immunochemical protocols. We present a lightweight, fully autonomous framework enabling line-of-sight inter-robot Tunable Diode Laser Absorption Spectroscopy (TDLAS) measurements without such constraints. A sensor robot equipped with a gimbal-mounted TDLAS unit tracks a reflector robot bearing an illuminated, color-controllable target. Coarse localization is achieved via RTK-GNSS, with vision-based fine tracking and passive time synchronization handled onboard. The system, based on off-the-shelf Pixhawk controllers and ArduPilot firmware, was validated in a 15m × 7m outdoor trial. Despite GNSS inaccuracies and deliberate occlusion by a methane-filled bag, the system retained lock, recovered from visual loss in under one second, and captured a 2800 ppm·m plume signature. These results demonstrate robust, scalable methane sensing for mobile gas tomography or standalone leak detection. Core components are released open-source to support future deployment. Hepcidin-25, a key iron-regulatory peptide hormone discovered in 2000, has revolutionized the understanding of iron disorders and its quantitative determination in biological samples should advance the management of iron-related pathology (diagnosis, prognosis and treatment). This study applied LC-MS/MS, using the triple quadrupole (QqQ) mass spectrometer, in a rapid and robust analytical strategy for the quantification of hepcidin-25 in human serum, to be implemented in routine laboratories. For this purpose, two sample preparation strategies and two complementary chromatographic separation conditions were investigated, where the use of acidic mobile phases (0.1% trifluoroacetic acid) was compared with a novel approach involving solvents at high pH (containing 0.1% ammonia). The application of these LC-MS/MS methods to human samples in an intra-laboratory comparison, using the same hepcidin-25 calibrators, yielded a very good correlation of the results. The LC-MS/MS employing trifluoroacetic acid-based mobile phases was selected as a highly sensitive (limit of quantification LOQ of 0.5 μg/L) and precise (coefficient of variation CV<15%) method and was recommended as a reference method candidate for hepcidin-25 quantification in real samples (in the dynamic range of 0.5-40 μg/L). One of the novel aspects of the methodology was the use of amino- and fluoro-silanized autosampler vials to reduce the interaction of the 25-residue peptide to laboratory glassware surfaces. Moreover, this LC-MS/MS method was used for an international round robin study, applying a secondary reference material as a calibrator. By determining the degree of equivalence between the results of the ten participating methodologies, the performance of the method developed in this study was found to be in the optimal range as defined by the International Consortium for Harmonization of Clinical Laboratory Results (ICHCLR). In this work, the formation of hepcidin-25 complexes with copper(II) was investigated. The first reversed-phase chromatographic separation of hepcidin-25/Cu2+ and hepcidin-25 (copper “free”) was achieved by applying mobile phases containing 0.1% of ammonia (pH 11). In this tutorial, we provide the theoretical background and give design guidelines for SOI-based MRR as well as examples of surface functionalization procedures for label-free detection of molecules. After introducing the advantages and perspectives of MRR, fundamentals of MRR are described in detail, followed by an introduction to the fabrication methods, which are based on a complementary metal-oxide semiconductor (CMOS) technology. Optimization of MRR for chemical and biological sensing is provided, with special emphasis on the optimization of waveguide geometry. T1 - Cocaine detection by a laser-induced immunofluorometric biosensor N2 - The trafficking of illegal drugs by criminal networks at borders, harbors, or airports is an increasing issue in public health as these routes ensure the main supply of illegal drugs. The prevention of drug smuggling, including the installation of scanners and other analytical devices to detect ultra-small traces of drugs within a reasonable time frame, remains a challenge. The presented immunosensor is based on a monolithic affinity column with a large excess of immobilized hapten, which traps fluorescently labeled antibodies as long as the analyte cocaine is absent. In the presence of the drug, some binding sites of the antibody will be blocked, which leads to an immediate breakthrough of the labeled protein, detectable by highly sensitive laser-induced fluorescence with the help of a Peltier-cooled complementary metal-oxide-semiconductor (CMOS) camera. Liquid handling is performed with high-precision syringe pumps and microfluidic chip-based mixing devices and flow cells. The biosensor achieved limits of detection of 23 pM (7 ppt) of cocaine with a response time of 90 seconds and a total assay time below 3 minutes. With surface wipe sampling, the biosensor was able to detect 300 pg of cocaine. This immunosensor belongs to the most sensitive and fastest detectors for cocaine and offers near-continuous analyte measurement. T1 - Sintered glass monoliths as new supports for affinity columns N2 - A novel stationary phase for affinity separations is presented. This material is based on sintered borosilicate glass readily available as semi-finished filter plates with defined porosity and surface area. The material shows fast binding kinetics and excellent long-term stability under real application conditions due to lacking macropores and high mechanical rigidity. The glass surface can be easily modified with standard organosilane chemistry to immobilize selective binders or other molecules used for biointeraction. In this paper, the manufacturing of the columns and their respective column holders by 3D printing is shown in detail. The model system protein A/IgG was chosen as an example to examine the properties of such monolithic columns under realistic application conditions. Several specifications, such as (dynamic) IgG capacity, pressure stability, long-term performance, productivity, non-specific binding, and peak shape, are presented. It could be shown that due to the very high separation speed, 250 mg antibody per hour and column can be collected, which surpasses the productivity of most standard columns of the same size. The total IgG capacity of the shown columns is around 4 mg (5.5 mg/mL), which is sufficient for most tasks in research laboratories. The cycle time of an IgG separation can be less than 1 minute. Due to the glass material's excellent pressure resistance, these columns are compatible with standard HPLC systems. This is usually not the case with standard affinity columns, limited to manual use or application in low-pressure systems. The use of a standard HPLC system also improves the ability for automation, which enables the purification of hundreds of cell supernatants in one day. The sharp peak shape of the elution leads to an enrichment effect, which might increase the concentration of IgG by a factor of 3. The final concentration of IgG can be around 7.5 mg/mL without the need for an additional nanofiltration step. The purity of the IgG was > 95% in one step and nearly 99% with a second polishing run. We investigate lithium isotope fractionation (LIF) as an innovative probe of degradation in lithium cobalt oxide (LCO) coin cells aged over 0−700 cycles. High-precision multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS) quantified δ7Li variations across cell stages: non-cycled (0 cycles), newly formed (5 cycles), semi-aged (250 cycles), and fully aged (700 cycles). During early cycling (≤ 45 cycles), chemical processes drive 7Li enrichment at the anode (δ7Li vs LSVEC = +12 ‰) through solid electrolyte interphase (SEI) formation, while the cathode depletes in 7Li (δ7Li vs LSVEC = −0.7 ‰). Beyond 45 cycles, electric field-induced migration predominates, promoting 6Li intercalation into the anode and increasing the δ7Li of the cathode by 8.1 ‰. Mass balance verifies isotope conservation, attributing shifts to redistribution and trapping. Complementary electrochemical impedance spectroscopy (EIS) and X-ray absorption spectroscopy and diffraction confirm SEI expansion, cobalt oxidation, lattice shrinkage, and changes in electrode structure, corroborating LIF trends. Notably, a δ7Li inflection at approximately 270 cycles anticipates end-of-life by 70 cycles, surpassing traditional methods in sensitivity. LIF emerges as a predictive indicator of aging mechanisms, informing optimized designs for durable LIBs. First, a comprehensive mechanistic study of molecule formation in graphite furnaces is presented, which is a key step into the recovery of analytical signals. For this, the molecule formation of CaF was studied, which is used for the indirect analytical determination of fluorine in HR-CS-GFMAS. A zirconium coating catalyzes the CaF formation, and its structure was investigated. The kinetics of this reaction was established by monitoring its molecular spectrum at different atomisation temperatures. An Arrhenius plot showed a pseudo-first order reaction with respect to fluorine (n = 1). An intermediate state was isolated, and its structure was elucidated by spectroscopic methods: scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM-EDX), X-ray photoelectron spectroscopy (XPS), X-ray absorption spectroscopy (XANES and EXAFS), and Raman microspectroscopy. Here a mechanism is proposed, where ZrO2 works as a heterogeneous catalyst: after a pyrolytic step, an intermediate state of ZrO(OCaF) is activated, and at higher temperatures, CaF(g) is released from the zirconium-coated graphite surface. Second, analytical methods were developed by using HR-CS-MAS as detector for non-metals and isotope analysis. Therefore, the determination of organic absorbable chlorine in water, the quantification of fluorine in consume care products with declared perfluorinated ingredients, and the determination of sulfur content in crude oils were investigated. Finally, the high resolution of the instrumentation allows to measure isotopic shifts with high precision in some observed molecular spectra. Consequently, the molecular spectra of enriched isotopes of boron and magnesium were investigated, establishing so the potential of HR-CS-MAS for the accurate and precise determination of isotopic amount ratios. Zusammenfassung Die vorliegende Arbeit befasst sich mit zwei zentralen Aspekten der High-Resolution-Continuum-Source-Molekülabsorptionsspektrometrie mit Graphitrohrtechnik (HR-CS-GFMAS), einer Analysetechnik für elementare Spurenanalyse. Der erste Teil der Arbeit umfasst eine mechanistische Studie zur Molekülbildung auf Graphitoberflächen. Dies ist ein wichtiger Schritt, um analytische Signale zu entdecken. Dazu wurde die Molekülbildung von CaF analysiert, welches für die indirekte, analytische Bestimmung von Fluor in HR-CS-GFMAS genutzt wird. Die CaF Bildung wurde mittels einer Beschichtung aus Zirconium katalysiert und deren Struktur analysiert. Die Kinetik dieser Reaktion wurde durch Beobachtung des jeweiligen Molekülspektrums bei verschiedenen Atomisierungstemperaturen beobachtet. Ein Arrheniusplot zeigte für Fluor (n = 1) eine Reaktion Pseudo-erster Ordnung. Auf Grundlage dieser Ergebnisse wird ein Mechanismus vorgeschlagen, bei dem ZrO2 als heterogener Katalysator fungiert; in Folge einer Pyrolysestufe wird ein Übergangszustand des ZrO(OCaF) aktiviert, welcher bei höheren Temperaturen CaF(g) an der Zirconium-Graphitoberfläche freisetzt. Im zweiten Teil der Arbeit werden Analysemethoden entwickelt, in dem HR-CS-MAS als Detektor für Nichtmetalle und Isotopanalyse angewandt wird. Hierfür wurde organisch gebundenes Chlor im Wasser bestimmt, der Fluorgehalt in Pflegeprodukten mit perfluorierten Inhaltsstoffen quantifiziert und der Schwefelgehalt in Erdöl untersucht. Weiterhin ermöglicht die hohe Auflösung der Messgeräte eine präzise Bestimmung der Isotopenverschiebung einiger untersuchter Molekülspektren. Daher wurden die Molekülspektren angereicherter Bor- und Magnesiumisotope untersucht. Auf diese Weise wurde das Potential von HR-CS-MAS für die akkurate und präzise Bestimmung von Isotop-Mengenverhältnissen nachgewiesen. Um die Phosphorversorgung für Pflanzen in der Landwirtschaft zu gewährleisten, werden Phosphordünger eingesetzt. Die Nährstoffe werden hauptsächlich durch Wirtschaftsdünger (Gülle, Jauche, Stallmist) zurückgeführt. Zusätzlich werden in der konventionellen Landwirtschaft mineralische Phosphordünger eingesetzt, die vorwiegend aus dem fossilen Rohstoff Phosphorit gewonnen werden. Um die Abhängigkeit von fossilen Rohstoffen zu reduzieren, sollen nährstoffhaltige Rest- und Abfallstoffe zur Phosphordünger aufbereitet werden. In Abwasserkläranlagen fällt phosphorreicher Klärschlamm als Abfallstoff an. Der größte Anteil des Klärschlammes wird in Steinkohlekraftwerken, Zementwerken oder in Monoklärschlammverbrennungsanlagen verbrannt. Die Klärschlammaschen aus den Monoverbrennungsanlagen enthalten bis zu 12 Gew.-% Phosphor und sind daher für die Düngerherstellung geeignet. Ein geeignetes Verfahren zur Aufbereitung der Klärschlammaschen ist die thermochemische Behandlung im Drehrohrofen. Mit dem sogenannten AshDec®-Prozess, kann ein Phosphordünger hergestellt werden. Aktuell wird eine großtechnische Anlage in Altenstadt (Bayern) geplant, die im Jahr 2023 den Betrieb aufnehmen soll. Der zentrale Bestandteil ist die Phasenumwandlung von schlecht pflanzenverfügbaren Phosphaten in der Klärschlammasche (vorwiegend das Calciumphosphat Whitlockit und Aluminiumphosphat) zu gut pflanzenverfügbaren Calciumalkaliphosphaten im Produkt. Um dies zu erzielen, werden die Aschen mit Natrium- und/oder Kalium-Verbindungen gemischt und einer thermochemischen Behandlung bei 800-1000 °C zugeführt. Um die Pflanzenverfügbarkeit und Düngewirkung von Phosphaten abschätzen zu können, ist die chemische Extraktionsmethode mit neutraler Ammoniumcitratlösung geeignet. In der Dissertation werden die (Phosphor-)Phasenreaktionen und die Prozessbedingungen des thermochemischen Verfahrens untersucht, um das Verfahren gezielt zu modifizieren und die Wirtschaftlichkeit zu steigern. Die Zielphasen der thermochemischen Behandlung sind die Calciumalkaliphosphate CaNaPO4 und CaKPO4, und deren Mischphasen Ca(Na,K)PO4, welche in der Publikation 3.1 synthetisiert wurden. Für diese Calciumalkaliphosphate wurden die thermodynamischen Daten der Standardbildungsenthalpie, Standardentropie, Wärmekapazität und die Wärmemenge der Phasenumwandlung bestimmt (Publikation 3.1). Bei der thermochemischen Behandlung von Klärschlammaschen wurden als Additive Natrium- und Kaliumsulfat bei verschiedenen Temperaturen getestet. Der Einsatz von Kaliumadditiven ist erwünscht, um den Marktwert des Produktes durch die Produktion eines Phosphor-Kalium-Düngers zu erhöhen. Zusätzlich wird eine möglichst geringe Prozesstemperatur angestrebt, bei der die erwünschten Calciumalkaliphosphate gebildet werden. In den Publikationen 3.2 und 3.3 wird gezeigt, dass für Klärschlammaschen eine komplette Phasenumwandlung zu Calciumnatriumphosphaten ab 875 °C in Laborversuchen (Korundtiegel) mit dem Additiv Natriumsulfat erzielt werden kann. Der Einsatz von Kaliumsulfat erforderte notwendige Reaktionstemperaturen von über 1100 °C. Diese unterschiedlichen Reaktionstemperaturen hängen mit den Schmelzpunkten von Natriumsulfat (890 °C) und Kaliumsulfat (1070 °C) zusammen. Um Kaliumsulfat in den Prozess bei niedrigen Temperaturen zu integrieren, wurden Natriumsulfat und Kaliumsulfat vor der Behandlung gemischt. Dies führte zu einer Schmelzpunkterniedrigung und resultierte in Reaktionstemperaturen zwischen 900 °C bis 1000 °C für die untersuchten Mischungen von Natrium- und Kaliumsulfat (Publikation 3.3). Die Zusammensetzung der Calciumalkaliphosphate Ca(Na,K)PO4 war anders als erwartet. Bei der Phasenanalytik stellte sich heraus, dass bei geringem und mittlerem Anteil von Kaliumsulfat im Alkali-Additiv nur Calciumnatriumphosphate mit sehr geringen Kaliumgehalten gebildet wurden. Erst bei einem hohen Anteil von Kaliumsulfat in der Mischung der Alkalisulfate konnten kaliumhaltige Calciumalkaliphosphate nachgewiesen werden (u.a. (Ca0.9Mg0.1)(Na0.6K0.4)PO4). Dieser geringere Einbau von Kalium in die Calciumalkaliphosphate hängt mit den zusätzlich stattfindenden Reaktionen zwischen den Alkalien und den Silikaten zusammen. Es ist bekannt, dass die zugegebenen Alkalisulfate zuerst mit Silikaten und anschließend mit den Phosphaten reagieren. Silikate bauen bevorzugt Kalium ein, deswegen konnten erst kaliumhaltige Phosphate gebildet werden, wenn die Reaktion mit den Silikaten abgeschlossen war. Dies führte zu einem geringeren Kaliumanteil in den gebildeten Phosphaten im Vergleich zum Kaliumanteil der verwendeten Alkaliadditive. Das in Silikaten gebundene Kalium ist wahrscheinlich schlecht für die Pflanzen verfügbar. Dies könnte den Einsatz der produzierten Phosphor-Kalium-Dünger einschränken. Der bevorzugte Kaliumeinbau in Silikaten konnte auch in Kalkulationen mit den thermodynamischen Daten aus der Publikation 3.1 gezeigt werden. Mittels Phasenanalytik vor und nach der chemischen Extraktion, konnte die Zugehörigkeit zu verschiedenen Modifikationen der Calciumalkaliphosphaten sicher bestimmt werden und eine ungefähre Zusammensetzung dieser Calciumalkaliphosphate abgeschätzt werden. So konnte nachgewiesen werden, dass die Modifikation vom CaNaPO4 ungefähr 10 % Magnesium einbauen kann. Wenn mehr Magnesium eingebaut wird, bildet sich die Phase (Ca,Mg)NaPO4, die eine vergleichbare Struktur wie die bekannte Phase (Ca0.72Mg0.28)NaPO4 aufweist. Dieses magnesiumreiche Calciumnatriumphosphat entstand vermutlich, wenn entweder erhöhte Anteile an Kalium eingebaut wurden (Ca0.8Mg0.2)(Na0.85K0.15)PO4 (Publikation 3.3) oder die Phasenumwandlung vom Calciumphosphat Whitlockit zum Calciumalkaliphosphat CaNaPO4 noch nicht abgeschlossen war (Publikation 3.2). Wenn mehr Kalium eingebaut wird, dann entsteht eine Phase (u.a. (Ca0.9Mg0.1)(Na0.6K0.4)PO4) ähnlich zu den Mischphasen aus der Publikation 3.1. In den Publikationen 3.4 und 3.5 wurden keine Klärschlammaschen untersucht, sondern Biokohlen aus Modellklärschlämmen (Publikation 3.5) und Biomasseaschen (Publikation 3.4) aus der Vergasung oder Verbrennung einer Mischung aus Zuckerrohrbagasse und Hühnertrockenkot. Die thermochemischen Produkte der Biokohlen bzw. Biomasseaschen enthielten das gewünschte CaNaPO4 und hatten eine hohe Düngewirkung in Pflanzenwachstumsversuchen mit Sojapflanzen bzw. Gräsern. Such simulated patterns can be used to test holistic analysis models, and, since the diffraction intensity is on the same scale as the scattering intensity, may offer a novel pathway for determining the degree of crystallinity. The "Ultima Ratio" strategy is demonstrated on a 64-nm Metal Organic Framework (MOF) particle, calculated from Q < 0.01 1/nm up to Q < 150 1/nm, with a resolution of 0.16 Angstrom. The computations exploit a modified 3D Fast Fourier Transform (3D-FFT), whose modifications enable the transformations of matrices at least up to 8000^3 voxels in size. Multiple of these modified 3D-FFTs are combined to improve the low-Q behaviour. The resulting curve is compared to a wide-range scattering pattern measured on a polydisperse MOF powder. While computationally intensive, the approach is expected to be useful for simulating scattering from a wide range of realistic, complex structures, from (poly-)crystalline particles to hierarchical, multicomponent structures such as viruses and catalysts. T1 - Direct insight into the admixture activity during cement hydration using levitated droplets N2 - Chemical admixtures are widely used to adjust properties of the cementitious mixtures. Different superplasticizers (SP) are typically added to improve the flowability of the fresh cement paste. Their working period is the initial stage of the cement hydration from its beginning until stiffening. Currently, polycarboxylate-based SP (PCE-SP) are the most effective ones. They can act beyond their aimed function causing changes in the hydration course, e. g. retardation effects. The exact mechanisms and influencing factors are still under investigation. However, the occurrence of the retardation effect indicates the involvement of PCE in the hydration reactions. Some observations indicate, that the retardation depends on the type, molecular structure and concentration of PCE, and the presence of other admixtures (stabiliser, ST). These factors influence further the working time of admixture and its amount related efficacy in the flow improvement. We investigate the cement hydration process in situ in levitated droplets. High resolution synchrotron X-ray diffraction allows gathering temporal course of the hydration reactions. The measurement setup is based on an ultrasonic levitator allowing in situ investigation of the temporal changes of the phase composition in the hydrating cementitious system. Further, the hydration can be followed ab initio avoiding the time loss for the sample preparation and filling in the sample holder. The collected data allows to conclude about the mechanisms of the admixture action and their involvement into the ongoing hydration process. Since the properties of concretes are dominated by their key compound, cement, devising a way to determine the cement’s origin, known as provenancing, is of great importance for answering different provenance-related questions. These questions range from liability issues when damage occurs to concrete-made structures, to forensic investigations where cement particles are found at crime scenes. This thesis showcases the use of isotope techniques to answer these provenance-related questions. Conventional 87Sr/86Sr and 143Nd/144Nd isotope and elemental ratios consisting of Ca, Sr, K, Mn, Mg, and Ti are used as fingerprints for ordinary Portland cement (OPC) provenancing. The first part of this thesis describes research previously conducted in fingerprinting cementitious materials, providing an overview of provenance studies of cement and the main approaches commonly used. In several studies, the origin of clinker for certain locations was determined via different approaches. However, clinker is an intermediate product, which is available only at the production site and therefore, the practical relevance is rather limited. Furthermore, the use of Sr and Nd isotope systems, together with elemental fingerprints are presented as state of the art in the field. Therein, the principal approach for the overall study is sketched. For the second part of the study, a sample preparation technique for Sr isotopes in Portland cement was developed. The aim was to find the most appropriate sample preparation procedure for cement provenancing and selection was realised by comparing the 87Sr/86Sr isotope ratios of differently treated OPCs with those of the corresponding clinkers. Based on these findings, the third part of the study focused on the measurements of Sr and Nd isotope ratios, together with elemental ratios, to establish a reliable technique for OPC provenancing. The outcomes of this final stage are then used to establish a procedure for fingerprinting cements. This becomes possible with the use of Sr and Nd isotope ratios and geochemical profiles. To perform isotope ratio measurements and obtain reliable data, it was necessary to establish a quality control procedure. Thus, an interlaboratory comparison (ILC) was organised to characterise 87Sr/86Sr isotope ratios in geological and industrial reference materials by applying the conventional method for 87Sr/86Sr isotope ratios. As reference material, four cements (VDZ 100a, VDZ 200a, VDZ 300a, IAG OPC-1), one limestone (IAG/CGL ML-3) and one slate (IAG OU-6) were selected, thus covering a wide range of Sr isotope signatures. This contribution shows how an improvement in the measurement data situation can improve the significance and comparability of outdoor exposures. Photooxidative ageing is calculated using sample polymers for which the corresponding modelling data (Exposure Response Functions, ERF) is available. The mean square deviations are estimated, which result from using different spectral irradiances and different spectral sensitivities, both of the sensors and the polymers. It will be shown here how the significance of the irradiance values increases with increasing approximation of the spectral sensitivities of the sensor and polymer. Also, the mean square deviations are estimated, which result from using different activation energies and surface temperatures. A selection of different variables to describe the weathering exposure is created. The potential of either individual values or data pairs is evaluated in terms of significance or correlation to the calculated photooxidative ageing. Recording hourly data also enables subsequent evaluations of individual test specimens, adapted to the respective colour or to determine activation energies. An accumulated value, calculated from PIT-radiometer irradiance and a suited surface temperature, can provide a comparison of different outdoor exposures over different locations or years. Thus, a characterisation of the weathering exposure would be related to a reference spectral sensitivity (much closer to a polymer than that of a common UV radiometer), one reference activation energy, and different surface temperature scenarios with, on the one hand, graded solar absorptions (white / grey / black) and, on the other hand, different thermal couplings (coated metal / thick plastic). This results in three values each for plastic samples and coated metals (light / grey / dark). It will never be possible to accurately predict the weathering tests for individual samples (with their specific sensitivities), but the proposed approaches can go a long way towards the comparability of outdoor weathering exposures. But due to legal actions this license has not been finally confirmed before 2007. Subsequently, the Federal Office for Radiation Protection (BfS) began scheduling backfitting of the former iron ore mine into a repository. The licensed repository volume is 303,000 m3 based on estimations of expected waste volumes to be disposed off although the mine itself would offer a much larger volume. Once the repository is ready for Operation, waste packages can be disposed, but this is not expected before the end of this decade. Nevertheless, there is already today a great interest in qualified and certified waste conditioning and packaging for disposal requiring Containers, tested, evaluated and certified by BAM and BfS. Key points of the Konrad license concern comprehensive requirements for waste products and Containers. Details are described in BfS reports, which constitute the basic documents for the license. Following the final license confirmation a revision of these documents has been requested in order to include considerations of water poisoning substances. According to IAEA regulations type B(U) packages must withstand the defined accident conditions of transport. For the cask material DCI, it is necessary to determine the brittle fracture behaviour. Due to the complex structure of the cask body and the dynamic loading a fracture mechanical assessment in an analytical way is not always possible. Numerical calculations are necessary to determine the fracture mechanical load in the component. At the first step a numerical analysis has to be done to identify the loading state at the cask body. Secondly an analysis of a detail of the cask body is made considering the boundary conditions of the global model. An artificial flaw is considered in this detailed model to calculate the fracture mechanical loading state. The size of the artificial flaw is characterized by the ultrasonic inspection used for the quality assurance of the package. The applicant developed additional analysis tools for calculation of stress intensity factor and/or J-Integral. The radiation emission from a combination of four different types of fluorescent UV lamps was superimposed on the specimen surfaces in the climatic cabinet to match the UV part of solar radiation as closely as possible. Air temperature was controlled to within ± 1 °C, and relative humidity to within ± 5 %RH. As the lamps’ emission is focussed on the UV range almost no radiation heating of the sample occurs. Therefore, the temperature of the sample, which is the quantity of consideration for the degradation, is nearly identical with the chamber’s air temperature, which is the quantity to be controlled. Also, therefore, high humidity can be obtained on the sample surface. The limitation of the emission to the UV range is sufficient for the study of the polymer matrix. This type of weathering device offers an exactness of microclimatic control of the sample’s surface, which should be a standard for weathering devices. Even now it is the only weathering device which generates pure relative humidity (without aerosol). However, they have only a limited validity, mainly due to the limited reproducibility of the weather as well as of the bad adaptability to other exposure sites. Typically, an outdoor exposure is characterized by the measured radiant exposure; sometimes averaged temperature data are added. These can be – separately – related to long-term annual means, to get an idea on the severity of the exposure. However, as for polymeric ageing, irradiance and temperature act simultaneously, such severity of an outdoor exposure could be better described by a computed property change of a similar material, which is calculated from the hourly irradiance and temperature values on the base of established exposure response functions. In this way, various outdoor exposures are evaluated. For several exposure sites, annual deviations of the ageing amount are related to annual mean values, to evaluate the weathering reproducibility at the exposure site. Additionally, correlations to relevant weather parameters are illuminated. Also some specific exposure conditions, such as tilt angle, are tested. Compared to the use of weathering reference materials, this procedure unfavourably depends on the availability of a suited control as well as on the quality of the established exposure response function. In contrast, it enables a later exposure evaluation, on the base of documented weather data, if for instance no reference material was exposed. ED - Völzke, Holger T1 - Extended storage collaboration program - International subcommittee report - International perspectives on technical data gaps associated with extended storage and transportation of used nuclear fuel N2 - The US Electric Power Research Institute (EPRI) chartered a volunteer committee in November 2009 to share information and data related to extended storage and subsequent transportation of used nuclear fuel. This committee is named the Extended Storage Collaboration Program (ESCP) and was initially comprised of representatives from industry (fuel and cask vendors, utilities, the Nuclear Energy Institute, and EPRI), as well as government (e.g., US Department of Energy) and the US Nuclear Regulatory Commission. The primary motivation for establishing this committee was the recognition that used fuel will need to be stored past the regulatory licensing timeframe. In the US, this is mainly a result of the cessation of licensing activities for the planned Yucca Mountain repository. Recognizing that there is a global interest in extended storage and transportation, the International Subcommittee was established in December 2010 to extend the reach of ESCP internationally in order to fully address extended storage and transportation issues, as they apply to different systems, national policies, and regulations around the world. This Subcommittee serves to deepen the overall understanding of degradation mechanisms associated with storage and transportation systems, expand the access to data and programs related to extended storage and transportation, and strengthen the overall technical basis for programmatic decisions. The International Subcommittee established three basic objectives: 1. Conduct a technical data gap survey 2. Assess opportunities to leverage specific research to benefit the larger international group 3. Integrate our work with relevant International Atomic Energy Agency (IAEA) committee work This report constitutes completion of Objective #1. The International Subcommittee worked to identify and prioritize technical gaps that need to be addressed to support licensing activities within their own countries. Because there are varying types of storage and transportation system designs, national policies and regulations, there are identified commonalities as well as differences that stress the importance of this work on an international scale. Section 5 of this report identifies and prioritizes all of the identified data gaps for each participating organization. However, important data gaps were identified that had a consensus of opinion of being a high priority. These technical gaps, as a function of storage system component are: 1. Fuel – hydride effects on fuel cladding and retrievability of fuel after storage 2. Canister – general canister corrosion and stress corrosion cracking of canister closure welds, particularly in marine environments 3. Closure – degradation of bolts and seals (metallic and organic) 4. Overpack – concrete degradation Dry storage systems were originally designed for limited periods. With extended dry storage, it is generally believed that used fuel can be stored safely and securely past the regulatory time period. However, the technical arguments need to be made to justify extended licensing periods. Addressing the data gaps will provide the necessary arguments to justify license extensions. In the development of this report, the International Subcommittee met five times over a period of 18 months to work through the issues of identifying and prioritizing the data gaps, and discussing of technical issues as they pertain to the safety of used fuel storage and transportation. Here the main differences are discussed for: - Global solar radiation In weathering devices there is a constant spectral irradiance which is 5 times higher than the averaged spectral irradiance outdoors, which is strongly varying in its diurnal course. Intensified weathering using Fresnel reflectors concentrates only the direct solar radiation with a relatively low UV proportion. Problems of reciprocity may arise from the different mean of irradiance levels. - Temperatures at specimen surfaces. Because of the high irradiance levels mentioned above, artificial exposures may lead to high surface temperatures and large temperature differences between differently coloured specimen surfaces, if the artificial radiation source emits much VIS- and IR-radiation. Four corrected datasets were provided to participants ready for analysis. All datasets were measured on samples containing spherical scatterers, with two datasets in dilute dispersions, and two from powders. Most of the 46 participants correctly identified the number of populations in the dilute dispersions, with half of the population mean entries within 1.5 % and half of the population width entries within 40 %, respectively. Due to the added complexity of the structure factor, much fewer people submitted answers on the powder datasets. For those that did, half of the entries for the means and widths were within 44 % and 86 % respectively. This Round Robin experiment highlights several causes for the discrepancies, for which solutions are proposed. T1 - Supplementary data set for "Seeds of imperfection rule the mesocrystalline disorder in natural anhydrite single crystals" N2 - In recent years, we have come to appreciate the astounding intricacy of the formation process of minerals from ions in aqueous solutions. In this context, a number of studies have revealed that nucleation in the calcium sulfate system is non-classical, involving the aggregation and reorganization of nanosized prenucleation particles. In a recent work we have shown that this particle-mediated nucleation pathway is actually imprinted in the resultant single micron-sized CaSO4 crystals. This property of CaSO4 minerals provides us with an unique opportunity to search for evidence of non-classical nucleation pathways in geological environments. In particular, we focused on the quintessential single crystals of anhydrite extracted from the Naica mine in Mexico. We elucidated the growth history from this mineral sample by mapping growth defects at different length scales. Based on these data we argue that the nano-scale misalignment of the structural sub-units observed in the initial calcium sulfate crystal seed propagate through different length-scales both in morphological, as well as strictly crystallographic aspects, eventually causing the formation of large mesostructured single crystals of anhydrite. Hence, the nanoparticle mediated nucleation mechanism introduces a 'seed of imperfection', which leads to a macroscopic single crystal, in which its fragments do not fit together at different length-scales in a self-similar manner. Consequently, anisotropic voids of various sizes are formed with very well-defined walls/edges. But, at the same time the material retains its essential single crystal nature. These findings shed new light on the longstanding concept of crystal structure. The reason for this is that especially organic materials, such as food or many pharmaceutical, are subject to photochemical degradation. Of course, suited transparent packaging material may give protection against such impact for radiation sensitive fillings. But for this, it is necessary to know about the spectral range of the fillings sensitivity as well as about the radiation impact of the radiation sources that are relevant during the life time of the product. The spectral irradiance for characterizing the emission of various radiation sources is easily measured by means of a spectroradiometer. The spectral sensitivity of a filling's property can be determined by spectrally dispersed irradiation, where the positions on a sample are related to the different wavelengths. Thus, the damaging effect of the different wavelengths can be directly evaluated. A lateral measurement of the relevant property change shows the activation spectrum, which is the product of the spectral radiant exposure and the spectral sensitivity. By measuring the spectral irradiance for each sample position, the spectral sensitivity of the ageing property can be calculated. Comparing the fillings spectral sensitivity and the spectral irradiance of a potential radiation source during later transport and storage, conclusions can be drawn about necessary spectral absorption of a (partly) transparent packaging to give sufficient protection. Here, weathering methods are presented, which have been used for the failure prediction in specific outdoor conditions. Issues of uncertainty, reproducibility, or validation are discussed. For the prediction of the plastic’s fragmentation, much longer time scales have to be considered. To do this within a limited project life span, various processes as well as their acceleration potential have to be evaluated separately, such as temperature increase or spectral shift. The dataset was acquired using cost-effective halogen-lamp excitation (2kW per lamp) as an alternative to laser-based systems, with dual-camera thermal imaging (CMOS and bolometric) to enable performance comparison across imaging modalities. The specimens include both new and aged barrel types with controlled defects — FBHs, lines, crosses, triangles, and rectangles — simulating internal corrosion at varying scales (4mm to 60mm). Three heating regimes (both lamps, left only, right only) were systematically applied across multiple measurement regions per sample, yielding normalized thermal sequences. To lower the barrier for machine learning practitioners without thermography expertise, the dataset provides pre-computed features derived from principal component analysis, pulse phase thermography, and independent component analysis extracted using experimentally optimized time windows. Ground-truth binary masks mapping defect locations are included to enable supervised learning.
