Such seals are used in a wide area of applications including transport and storage casks for spent nuclear fuel and high level radioactive waste. The seal function is mainly based on the compression of the inner helical spring, which generates the necessary seal force to keep the sealing surfaces in close contact. This in turn leads to a plastic deformation of the outer jacket of the seal, comprised of highly ductile aluminum or silver that adapts to the sealing surfaces of cask body and bolted lid, thus providing high Level leak tightness. In Germany, those casks are licensed for Interim storage periods of up to 40 years or more if extended Interim storage would become necessary before a final repository is available. Thus, the sealing performance has to be evaluated, including factors like elevated temperature due to decay heat or mechanical loads due to transport under normal as well as accident conditions. Long-term investigations at BAM have been running over the last nine years to identify and evaluate the seal performance by measuring the remaining seal force, the useable resilience and the leakage rate after various time intervals at temperatures ranging from room temperature up to 150 °C. It was found that the seal force and useable resilience decrease with time and temperature, caused by creep deformation of the outer jacket. In order to obtain an analytical description for the seal behavior and to achieve more information on the material behavior under application conditions a comprehensive investigation program with Focus on aluminum as outer jacket material was launched. The program includes material investigations such as compression and tension creep tests with representative basic materials. An additional test setup allows for the continuous measurement of the remaining seal force at temperatures of up to 150 °C. Furthermore, seal segments are compressed and stored in heating chambers, thus producing segments at different stages of the aging process. The segments are investigated regarding the development of the contact area width, jacket thickness and microstructural changes. This data will be used to develop material models and an analytical description of the time and temperature dependent long-term sealing behavior. This paper explains the current status of gained test results and modelling approaches and closes with an outlook to the future Project plans. Samples of different mild and high alloyed stainless injection-pipe steels partially heat treated: 42CrMo4, X20Cr13, X46Cr13, X35CrMo4 as well as X5CrNiCuNb16-4 were kept at T=60 °C and ambient pressure as well as p=100 bar for 700 h - 8000 h in a CO2-saturated synthetic aquifer environment similar to possible geological on-shore CCS-sites in the northern German Basin. Main corrosion products are FeCO3 and FeOOH. Corrosion rates obtained at 100 bar are generally much lower than those measured at ambient pressure. Highest surface corrosion rates are 0.8 mm/year for 42CrMo4 and lowest 0.01 mm/year for X5CrNiCuNb16-4 in the vapour phase at ambient pressure. At 100 bar the highest corrosion rates are 0.01 mm/year for 42CrMo4, X20Cr13 (liquid phase), X46Cr13 and less than 0.01 mm/year for X35CrMo4 and X5CrNiCuNb16-4 after 8000 h of exposure with no regard to atmosphere. More and more methodologies for risk analysis quantify the consequences of these scenarios with complex models. Examples for complex models are the computational fluid dynamics model Fire Dynamics Simulator (FDS) and the microscopic evacuation model FDS+Evac. However, the high computational effort of complex models often limits the number of scenarios in practice. To balance this drawback, the scenarios are often simplified. Accordingly, there is a challenge to consider complex scenarios in risk analysis. To face this challenge, we improved the metamodel used in the methodology for risk analysis presented on ISTSS 2016. In general, a metamodel quickly interpolates the consequences of few scenarios simulated with the complex models to a large number of arbitrary scenarios used in risk analysis. Now, our metamodel consists of the projection array-based design, the moving least squares method, and the prediction interval to quantify the metamodel uncertainty. Additionally, we adapted the projection array-based design in two ways: the focus of the sequential refinement on regions with high metamodel uncertainties; and the combination of two experimental designs for FDS and FDS+Evac. To scrutinise the metamodel, we analysed the effects of three sequential refinement steps on the metamodel itself and on the results of risk analysis. We observed convergence in both after the second step (ten scenarios in FDS, 192 scenarios in FDS+Evac). In comparison to ISTSS 2016, we then ran 20 scenarios in FDS and 800 scenarios in FDS+Evac. Thus, we reduced the number of scenarios remarkably with the improved metamodel. In conclusion, we can now efficiently integrate complex scenarios in risk analysis. We further emphasise that the metamodel is broadly applicable on various experimental or modelling issues in fire safety engineering. Zu diesen Effekten zählen Wechselwirkungen zwischen den Polymeren und Ionen in der Porenlösung, frühe Phasenbildung, zeitabhängige und kompetitive Adsorption zwischen anionischen Polymeren und Sulfationen, Bildung von Phasen in der Porenlösung sowie Morphologieänderungen an Partikeloberflächen. Die frühe Hydratation von Zement, die durch Lösungs- und Fällungsprozesse angetrieben wird, beeinflusst diese Effekte erheblich. Das permanente Ungleichgewicht der Porenlösung führt zu Veränderungen der Partikeloberflächen, welches widerum zur Folge hat, dass rheometrische Messungen dieser Zementleime anfällig für Streuungen sind. Um die Einflüsse aus der Zementhydratation zu minimieren, wurden die rheometrischen Untersuchungen mit Zement in Porenlösung durchgeführt. Die Experimente wurden mit verschiedenen Feststoffvolumenfraktionen durchgeführt und mit den Ergebnissen identischer Systeme mit Wasser anstelle von Porenlösung verglichen. Zusätzlich wurden die gleichen Systeme mit Zugabe von Polycarboxylatethern untersucht. Die Ergebnisse zeigen, dass die Leime mit Wasser niedrigere Werte für Fließgrenze und plastische Viskosität aufweisen, als die Systeme mit Porenlösung. Während die Polymere eine Verminderung der Fließgrenze zur Folge hatte, war die Wirkung von Polymeren auf die plastische Viskosität vernachlässigbar. The tanks were installed in the space normally reserved for the spare tyre, in the car boot. No safety device was installed on the tank, in order to force critical failure of the cylinder. Two of the cars were equipped with a tank filled with liquefied propane to a level of 20 % (5.3 kg), the third one was filled completely (25.5 kg). The partially filled tanks failed critically within a time period of more than 20 min after the initiation of the fire. The fully-filled tank did not rupture; the propane was released continuously through a small leak that appeared during the fire. Comprehensive equipment was used to procure measurement data, enabling an analysis of potential consequences and hazards to humans and infrastructure within the vehicle surroundings. The inner status of the tank (pressure, temperature of the liquid phase and the steel casing), the development of the fire (temperature inside and around the vehicle) and the pressure induced in the near-field in case of tank rupture were recorded. The results were analysed in detail and compared against the data gained in tests involving sole, but identical LPG tanks. Diese Sensoren werden in der Industrie verwendet, um die Spurenfeuchte in unterschiedlichen Gasen zu messen. Normalerweise erfolgt eine Beschichtung der Sensoren mit Phosphorpentoxid und dessen Hydrolyseprodukte. In dieser Arbeit wurde stattdessen eine hygroskopische ionische Flüssigkeit benutzt. Die generierte Gasfeuchte für die Untersuchungen betrug bezogen auf die Frostpunkttemperatur -80 °C bis -30 °C, was gleichbedeutend mit einem Wasserdampf-Stoffmengenanteil von 0,5 μmol·mol-1 bis 376 μmol·mol-1 ist. Zusätzlich zu der Messung mit coulometrischen Sensoren erfolgte eine Referenzmessung der Gasfeuchte mittels eines Taupunkthygrometers. Erste Ergebnisse haben gezeigt, dass es möglich ist mit dieser Art der Beschichtung unterschiedliche Gasfeuchten zu messen. Humidified gas flow at adjusted gas pressure was split into two flows, one passed a coulometric sensor and the other one passed a reference hygrometer after decompression. Both signals were recorded and then compared after calculation of resulting frost point temperature. Calculation is based on a calibration function obtained at ambient pressure. Comparison showed that an increasing pressure resulted in a higher derivation between sensor signal (calculated frost point temperature) and reference frost point temperature. At an absolute pressure of 2 bar the differences were minor in consideration of the uncertainty, however at 15 bar the differences were 6.77 K. Nevertheless, it was possible to measure the gas humidity at higher pressure with coulometric trace humidity sensors. Ageing infrastructures – distributed acoustic monitoring of pipes” is an interdisciplinary research project at BAM internally financed from 2015 to 2018. Therefore, the quite young fibre optic sensing technology of distributed acoustic sensing (DAS) was investigated to possibly be extended towards a global condition monitoring system for pipelines operating in real time. DAS is a highly dynamic fibre optic sensing technology based on the method of coherent optical time domain reflectometry (C-OTDR). DAS allows capturing strain changes in the range of kHz. For the experimental work, the most suitable application yielding an optimum sensitivity was proven by wrapping a standard single-mode silica fibre around the pipe. The DAS sensitivity was investigated regarding the detection of 1) incidents that initiate propagation of acoustic waves in the pipe wall, 2) changes inside the pipeline causing altered flow and 3) damage development in the pipe wall. Therefore, several testing setups in laboratory as well as in real scale were realized. For comparison purposes, experiments were accompanied by acoustic emission analyses and by measurements with accelerometers. DAS was found to be very sensitive to gas ignition and its propagation across the pipe. Furthermore, the ability of DAS to detect and localize acoustic signals associated with pipeline leakage was demonstrated. The detection of crack formation and propagation within the pipe wall by means of DAS was studied during bending tests on several pipe segments, but was not proven so far with certainty. As expected, these studies turned out as the most difficult challenge due to the random occurrence and transient nature of microscopic damage phenomena. The most recent development in the field of fibre optic sensing is the distributed acoustic sensing (DAS), which allows to detect and to localize third party threats to pipelines. For this purpose, fibre optic telecommunication cables located close to the pipelines are usually used. However, DAS carries a far greater potential for continuous condition monitoring of pipelines. The interdisciplinary research project AGIFAMOR (Ageing Infrastructures – Fibre Optic Monitoring of Pipes) at BAM investigates a new technical approach to extend the application field of DAS towards the detection and localization of acoustic signals that indicate critical alterations and certain damage scenarios originated from within the pipeline or the pipe wall. Therefore, the optical fibre sensors are applied onto the pipe itself and the application procedure towards an optimal acoustic signal transduction is optimized. A number of laboratory scale experiments were performed focusing on the signal transmission of acoustic signals as well as the detection of damages in the pipe wall by means of DAS. Furthermore, real-scale tests on a pipeline DN100 of 38m length have been carried out at the BAM test site for technical safety (BAM-TTS) to study the detection and localization of leaks and of changing flow profiles due to corrosion or sedimentation processes. Im Ramen des Forschungsbelegs wurde bereits eine Anreicherungsmethode für ein Modellsystem einer Peptidbibliothek entwickelt. Das Modellsystem bestand dabei aus zwei verschiedenen Peptiden. Als Positivkontrolle wurde das FLAG-Peptid gewählt, welches selektiv gegen den Anti-FLAG-Antikörper bindet, und ein weiteres Peptid wurde als Negativkontrolle gewählt. Anhand dieses Modellsystems konnte die zuvor entwickelte Anreicherungsmethode von 20.000 auf 1.000.000 Beads vergrößert werden. Hierbei wurde ein Anreicherungsfaktor von 818 und eine gute Wiederfindungsrate von 82% erreicht. Zu Beginn standen dabei die Herstellung stabiler Dispersionen der einzelnen Nanopartikel und die Stabilität der gebildeten Konjugate im Vordergrund. Der Nachweis einer erfolgreichen Konjugatbildung, sprich der Beschichtung von Nanopartikel mit Proteinen, wurde sowohl qualitativ mittels DLS-Messungen als auch über quantitative Protein-Bestimmungen erbracht. Für die Quantifizierung konnten verschiedene Methoden eingesetzt werden. Neben der klassischen Vorgehensweise, welche indirekt über die Quantifizierung von ungebundenem Protein im Überstand erfolgt, konnten ihm Rahmen dieser Arbeit verschiedene direkte Bestimmungsmethoden entwickelt werden. So wurden mittels kolorimetrischer Tests, wie dem BCA-Assay und dem Bradford-Assay, Nanodiamantdispersionen mit Hilfe einer Korrekturwellenlänge vermessen und quantifiziert. Ebenso zum Einsatz kam die Methode der Aminosäureanalytik, welche aufgrund ihrer guten Rückführbarkeit auf Aminosäurestandards Ergebnisse mit hoher Richtigkeit generieren kann und ebenso die Detektion kleiner Proteinmengen möglich macht. Nach den erfolgten quantitativen Betrachtungen wurden die Protein-beschichteten Nanopartikel auf ihre Anwendbarkeit als Analoga von Virus-like Particles (VLP) bei einer Immunisierung zur Gewinnung von polyklonalen Antikörpern gegen humanes Ceruloplasmin in Kaninchen überprüft. Es konnte mittels ELISA gezeigt werden, dass die Konjugate erfolgreich für die Herstellung von Antikörpern eingesetzt werden können und im zeitlichen Verlauf einer Immunisierung eine Steigerung des Antikörper-Titers zu erreichen ist. T1 - Development of LC-MS/MS methods for the quantitative determination of hepcidin-25, a key regulator of iron metabolism N2 - Isotope-dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) is emerging in the field of clinical chemistry and laboratory medicine as an alternative to immunoassays and is acknowledged as the MS “gold standard” for small biomolecule quantification. Hepcidin-25, a key iron-regulatory peptide hormone discovered in 2000, has revolutionized the understanding of iron disorders and its quantitative determination in biological samples should advance the management of iron-related pathology (diagnosis, prognosis and treatment). This study applied LC-MS/MS, using the triple quadrupole (QqQ) mass spectrometer, in a rapid and robust analytical strategy for the quantification of hepcidin-25 in human serum, to be implemented in routine laboratories. For this purpose, two sample preparation strategies and two complementary chromatographic separation conditions were investigated, where the use of acidic mobile phases (0.1% trifluoroacetic acid) was compared with a novel approach involving solvents at high pH (containing 0.1% ammonia). The application of these LC-MS/MS methods to human samples in an intra-laboratory comparison, using the same hepcidin-25 calibrators, yielded a very good correlation of the results. The LC-MS/MS employing trifluoroacetic acid-based mobile phases was selected as a highly sensitive (limit of quantification LOQ of 0.5 μg/L) and precise (coefficient of variation CV<15%) method and was recommended as a reference method candidate for hepcidin-25 quantification in real samples (in the dynamic range of 0.5-40 μg/L). One of the novel aspects of the methodology was the use of amino- and fluoro-silanized autosampler vials to reduce the interaction of the 25-residue peptide to laboratory glassware surfaces. Moreover, this LC-MS/MS method was used for an international round robin study, applying a secondary reference material as a calibrator. By determining the degree of equivalence between the results of the ten participating methodologies, the performance of the method developed in this study was found to be in the optimal range as defined by the International Consortium for Harmonization of Clinical Laboratory Results (ICHCLR). In this work, the formation of hepcidin-25 complexes with copper(II) was investigated. The first reversed-phase chromatographic separation of hepcidin-25/Cu2+ and hepcidin-25 (copper “free”) was achieved by applying mobile phases containing 0.1% of ammonia (pH 11). LC-MS/MS and high-resolution mass spectrometry (Fourier-transform ion cyclotron resonance (FTICR) MS) were applied for the mass spectrometric characterization of the formed hepcidin-25-Cu(II) species at pH values of 11 and 7.4 respectively. A new species corresponding to hepcidin-25 complexed with two copper ions was identified at high pH. Nachdem man die spezifischen Parameter der Beanspruchung unter Anwendungsbedingungen und die spezifische Empfindlichkeit des Materials erfasst hat, beginnt ein Prozess, der als „Test Tailoring“ häufig abstrakt verwendet, auf den aber nur selten näher eingegangen wird. Genau dieser Prozess soll hier am Beispiel der Entwicklung eines künstlichen Bewitterungstests unter saurer Beaufschlagung, dem in der BAM entwickelten Acid Dew and Fog Test, beispielhaft erläutert werden.
