T1 - A chemical and microbial characterization of selected mud volcanoes in Trinidad reveals pathogens introduced by surface water and rain water N2 - Terrestrial mud volcanoes are unique structures driven by tectonic pressure and fluids from the deep subsurface. These structures are mainly found in active tectonic zones, such as the area near the Los Bajos Fault in Trinidad. Here we report a chemical and microbiological characterization of three mud volcanoes, which included analyses of multiple liquid and solid samples from the mud volcanoes. Our study confirms previous suggestions that at least some of the mud volcano fluids are a mixture of deeper salt-rich water and surficial/precipitation water. No apparent water quality differences were found between sampling sites north and south of a major geological fault line. Microbiological analyses revealed diverse communities, both aerobic and anaerobic, including sulfate reducers, methanogens, carbon dioxide fixing and denitrifying bacteria. Several identified species were halophilic and likely derived from the deeper salt-rich subsurface water, while we also cultivated pathogenic species from the Vibrionaceae, Enterobacteriaceae, Shewanellaceae, and Clostridiaceae. These microorganisms were likely introduced into the mud volcano fluids both from surface water or shallow ground-water, and perhaps to a more minor degree by rain water. The identified pathogens are a major health concern that needs to be addressed. After extraction and separation of inorganic and organic fluorinated compounds, organically bound fluorinated compounds were indirectly determined via the molecular absorption of gallium mono-fluoride at 211.248 nm. The implementation of the decisions of the “Stockholm Convention on persistent organic pollutants” as well as the “Protocol on Persistent Organic Pollutants of the Convention on Long-Range Transboundary Air Pollution” of the UNECE include the reduction respectively the elimination of PFASs in the environment. Currently, regulations aim to target single compounds (mostly C8-PFAS). But the PFAS group includes over 4700 substances, which are potentially persistent and toxic, thus target analytical approaches are not suitable for a holistic approach investigating the PFAS pollutant situation. Furthermore, forbidden PFASs are substituted by short-chain PFASs, thus the number of unknow substances is steadily increasing. For this reason, sum parameter approaches are more suitable to investigate and assess the pollution situation as well as derive exposure limits. Our developed analytical method was successfully applied to determine PFASs in soil samples from a polluted site. In view of steadily increasing numbers of PFAS substances, our method will make an important contribution in assessing the pollution situation as well as support policy makers in deriving exposure limits for PFAS in the future. To omit the bias of the solid phase extraction (SPE) step commonly used during the analysis of extractable organically bound fluorine (EOF) we optimized a fast and simple SPE-free extraction method. The developed extraction method consists of a liquid-solid extraction using acidified methanol without any additional SPE. Four extraction steps were representative to determine a high proportion of the EOF (>80% of eight extractions). Comparison of the optimized method with and without an additional SPE clean-up step revealed a drastic underestimation of EOF concentrations using SPE. Differences of up to 94% were observed which were not explainable by coextracted inorganic fluoride. Therefore, not only a more accurate but also a more economic as well as ecologic method (bypassing of unnecessary SPE) was developed. The procedural limit of quantification (LOQ) of the developed method was 10.30 μg/kg which was sufficient for quantifying EOF concentrations in all tested samples. For future PFAS monitoring and potential regulative decisions the herein presented optimized extraction method can offer a valuable contribution. Two green GC-solvents (dimethyl carbonate (DMC), diethyl carbonate (DEC)) and several internal standards (hexane, benzene, toluene, DEC) have been tested. The separation of the products at low retention times (methanol, isobutane, isobutene) could be significantly improved. For the liquid main products t-butyl hydroperoxide (TBHP), di-t-butyl peroxide (DTBP), t-butanol (TBA), propanone and most trace products (isopropanol, isobutanal, methyl formate, isopropyl formate, t-butyl formate and some acetates – 24 compounds in total), a coefficient of determination CoD > 0.999 has been achieved. For all weighing and GC/MS measurements uncertainties have been calculated. The peaks of the peak pair t-butyl formate (CoD >0.999) and isobutanol (CoD >0.99), have been simulated and used for calibrations. DEC gave a decomposition of TBHP on the column. With DMC, a problem of peak purity of TBHP appeared but could be resolved. The use of the green solvent DMC, compared to also studied traditional solvents like heptane and toluene, proved to be very beneficial, since it also permitted to determine polar compounds, e. g. carbon dioxide and formic acid. The elemental composition of this nutrient-containing dust depends largely on the exposure region and time as well as on climate. Therefore, in this paper we report an analysis of atmospheric pollutions with a self-made low-cost bulk deposition sampler directed at sampling deposition via air transport and rainfall. We used the device in diverse environments - thus comparing an urban region, an area surrounded by forest and an area mainly dominated by agriculture. The total organic carbon (TOC) and total nitrogen (TN) amounts were selected as indicator parameters and analyzed in a biweekly rhythm for three and a half and two years, respectively. The TOC value responded to particulate matter in the urban area, especially significant were the influences of the New Year's firework in urban and pollen in the rural forest area. In contrast, the TN value was more under the influence of the nitrogen emissions in the agriculture-dominated area. However, the TN value did not correlate with the NOx values in the urban area because the atmospheric nitrogen emissions in the city might originate from various emission sources. Summarizing, the TOC and TN values of the self-made low-cost bulk deposition sampler were in good agreement with environmental events of their immediate surrounding. Moreover, the selected containers and sampling procedures are universally applicable to monitor and analyze organic as well as inorganic parameters (e.g. metal ions) of atmospheric deposition. While material identification methods are well established in the analysis of historical papers, the identification of manufacturing technologies is still an under-researched topic, that requires the development of appropriate methods and measurement protocols. This paper reports on the results of a research project aimed at the application of synchrotron scanning small angle X-ray scattering (SAXS) method in the characterization of paper structure, with emphasis on the assessment of fibrillar orientation as a marker characteristic for different, historical papermaking technologies. The main objective of this study consists of the development of a measurement protocol involving the SAXS technique complemented by other analytical methods in the characterization of the fibrous paper structure. In theory, the uptake into the passive sampler is only dependent on the diffusion coefficient of the analyte in air and the geometry of the sampler (ideal adsorption). For characterization, the uptake rate (UR) is used, defined as the ratio of the uptaken mass and the product of ambient concentration and exposure time. Various reported effective uptake rates (UR,eff) differ considerably from ideal values (UR,id). Literature values in national and international standards, are essentially sorted by the applicable exposure time, while detailed information about the corresponding concentration range is lacking. Moreover the number of itemized substances is limited. The aim of this study was to review and expand this set of uptake rates by determination of own quality assured laboratory values.The determined uptake rates are considered accurate in terms of RSD and comparability to literature values and can be recommended for exposure times of seven days at 50 100 µg/m³ (approximately 100 300 ppm min). Seven day uptake rates in ISO16017 2 and ASTM D6196 are not generally suited for this purpose, as has been exemplarily shown for benzene. In der Theorie ist die Stoffaufnahme in den Passivsammler nur abhängig vom Diffusionskoeffizienten der Substanz in Luft und der Geometrie des Sammlers (ideale Adsorption). Um sie zu charakterisieren, wird die Aufnahmerate (UR), definiert als Quotient der Massenaufnahme und dem Produkt aus Konzentration und Expositionszeit, verwendet. Neun Labore nahmen an dem Ringversuch teil, der sich aus zwei Teilen zusammensetzte: Im ersten Teil wurde die analytische Leistungsfähigkeit beurteilt. Jedes beteiligte Labor erhielt zwei mit Analyten dotierte Tenax® TA Desorptionsröhrchen zur Qualitätskontrolle (QC). Im zweiten Teil führte jedes Labor Emissionsmessungen über einen Zeitraum von 48 Stunden in Mikro-Kammern mit Volumina von 44 oder 114 ml durch. Als Probenmaterial dienten Stücke eines Regenponchos, der Cyclohexanon, Phenol, 2-Ethylhexanol, Naphthalin und Isophoron in signifikanten Konzentrationen emittierte. Für den QC-Test wurden sehr geringe relative Standardabweichungen (9 14 %) erhalten. Die Ergebnisse der Emissionsmessungen waren ebenfalls sehr gut. EPDM) in Sportböden gilt. In Deutschland gibt es laut DFB ca. 5100 Kunstrasenplätze. In der EU beläuft sich die Anzahl laut ECHA auf 13,000 Kunstrasen- und 47,000 Bolzplätze, Tendenz steigend. Das Hauptziel des Projektes war es daher, eine Massenbilanz für die Freisetzung von MP zu erstellen, um so eine Schätzung des MP-Austrags pro Kunstrasenplatz und Jahr zu ermöglichen. Dafür werden die MP-Emissionen von drei Kunstrasenszenarien in verschiedenen Zuständen (ungealtert, künstlich gealtert und in Echtzeit gealtert) verglichen: die Vergangenheit (Kunstrasen: fossilbasiert, EPDM-Füllung), die Gegenwart (der in Europa am häufigsten installierte Rasen) und die Zukunft (Rasen mit recycelten Grasfasern, keine synthetische Füllung sondern z.B. Quarzsand oder Korkschrott). Dazu werden fabrikneue Kunststoffrasen und EPDM-Granulat unter Laborbedingungen beschleunigt gealtert, um die Freiland-Beanspruchung eines Kunststoffrasens in circa 15 Jahren abzubilden. Con l'obiettivo di creare una storia dettagliata degli inchiostri neri, abbiamo elaborato un protocollo non invasivo per raccogliere dati statisticamente rilevanti sugli inchiostri da manoscritti datati e localizzati che coprono un ampio arco temporale e diverse aree geografiche. Il primo passo del nostro protocollo consiste nella selezione effettuata mediante riflettografia NIR e IRR. Le differenze ottiche tra gli inchiostri a base di carbonio, tannino e ferro-gallio sono meglio riconoscibili confrontando la loro risposta alla luce infrarossa: l'inchiostro a base di carbonio ha un colore nero intenso, l'inchiostro ferro-gallico diventa trasparente al di sopra dei 1400 nm e l'inchiostro a base di tannino scompare a circa 750 nm. abbiamo semplificato l'analisi utilizzando un piccolo microscopio USB con LED NIR (940 nm) e UV (395 nm) integrati, oltre a una fonte di luce bianca esterna. Confrontando le immagini sotto illuminazione bianca e nel vicino infrarosso, determiniamo la tipologia di inchiostro osservando i cambiamenti nell'opacità dell'inchiostro. In questo caso, gli inchiostri a base di carbonio non mostrano alcuna variazione di opacità quando illuminati con lunghezza d'onda NIR, mentre l'opacità degli inchiostri ferro-gallici cambia notevolmente e gli inchiostri tanninici diventano trasparenti. La nostra recente scoperta che gli inchiostri misti carbonio/ferro-gallici erano piuttosto diffusi nella tarda antichità e nell'alto Medioevo ci ha suggerito di tornare al metodo IRR convenzionale. The corpus under study contains manuscripts with formulae, i.e. model letters and charters, besides numerous other types of texts with no clear separation between them (for example by titles). Sixty-three manuscripts have been studied with the aim of reconstructing the history of both their copying and their use. To this aim, we adopted a non-invasive approach that included near-infrared imaging and X-ray fluorescence spectroscopy for on-site measurements, supported by historical and palaeographic analysis. Coloured ruling with plummet or ink would make a Torah scroll invalid for liturgical use. Ruling lines on some leather fragments of early Oriental Torah scrolls give the impression of being produced with plummet or ink but may be the result of dust in the furrows of relief ruling. Erfurt 9, a fourteenth-century Ashkenazi Torah scroll, has coloured ruling, which is partial secondary re-ruling, on 13 of the 34 surviving sheets, perhaps added when the original relief ruling was difficult to see. The coloured ruling in Erfurt 9 is brown, giving it the visual appearance of iron-gall ink. However, XRF tests showed that the only meaningful metallic component in the coloured ruling was lead (Pb). Lead plummet ruling tends to have a greyish appearance. Black specks under near infrared light are consistent with minium, a lead-based red ink that can turn brown when it degrades. Partial secondary re-ruling added with minium to the original relief ruling comes as a surprise as it would have rendered Erfurt 9 unfit for liturgical use. The continuous use of PFAS in household products and the discharge of PFAS from industrial plants into the sewer system have been resulted in contaminated effluents and sewage sludge from wastewater treatment plants (WWTPs) which became an important pathway for PFAS into the environment. Because sewage sludge is often used as fertilizer its application on agricultural soils has been observed as significant input path for PFAS into our food chain. To produce high-quality phosphorus fertilizers for a circular economy from sewage sludge, PFAS and other pollutants (e.g. pesticides and pharmaceuticals) must be separated from sewage sludge. Normally, PFAS are analyzed using PFAS protocols typically with time-consuming extraction steps and LC-MS/MS target quantification. However, for screening of PFAS contaminations in wastewater-based fertilizers also the DGT technique can be used for the PFAS extraction. Afterwards, combustion ion chromatography (CIC) can be applied to analyze the “total” amount of PFAS on the DGT binding layer. The DGT method was less sensitive and only comparable to the extractable organic fluorine (EOF) method values of the fertilizers in samples with >150 µg/kg, because of different diffusion properties for various PFAS, but also kinetic exchange limitations. Comparative leaching tests were performed using four PFAS-contaminated soils whose concentration patterns of 10 selected perfluoroalkyl and polyfluoroalkyl substances (PFAS) differed due to the two types of contamination sources. This study aimed to evaluate the equivalence of two usual laboratory-scale leaching test procedures, batch and column percolation tests, at liquid-to-solid ratios (L/S) of 2 l/kg, which is the current practice within the German assessment framework, and 10 l/kg (relevant for some EU regulations such as the landfill directive). The differences between the replicates of leaching tests investigating PFAS were smaller for column percolation tests than for batch tests, probably mainly due to the greater sample size and the better representativeness of the sample portion analyzed. It was observed that batch tests overestimate the release of shorter-chain PFAS, whereby the effect was greater with carboxylic than with sulfonic acids. Currently, the limits of detection of analyses given by the DIN standard with regard to PFCA and PFSA in soils are partly not sufficient to detect very low contents, whereas the detection of selected PFCA and PFSA in eluates is more sensitive, in accordance with the available standards. T1 - Corundum Particles as Trypsin Carrier for Efficient Protein Digestion N2 - Reusable enzyme carriers are valuable for proteomic workflows, yet many supports are expensive or lack robustness. This study describes the covalent immobilization of recombinant trypsin on micrometer-sized corundum particles and assesses their performance in protein digestion and antibody analysis. The corundum surface was cleaned with potassium hydroxide, silanized with 3-aminopropyltriethoxysilane and activated with glutaraldehyde. Recombinant trypsin was then attached, and the resulting imines were reduced with sodium cyanoborohydride. Aromatic amino acid analysis (AAAA) estimated an enzyme loading of approximately 1 µg/mg. Non-specific adsorption of human plasma proteins was suppressed by blocking residual aldehydes with a Tris-glycine-lysine buffer. Compared with free trypsin, immobilization shifted the temperature optimum from 50 to 60 °C and greatly improved stability in 1 M guanidinium hydrochloride. Activity remained above 80 % across several reuse cycles, and storage at 4 °C preserved functionality for weeks. When applied to digesting the NISTmAb, immobilized trypsin provided peptide yields and sequence coverage comparable to soluble enzyme and outperformed it at elevated temperatures. Visual observation of objects under examination is always a necessary first step in the characterisation process. Several questions arise when undertaking to identify and count particles to measure their size and shape distribution. In addition to challenges with the dispersion and identification of the particles, more than one protocol for counting particles is in use. This paper focuses on precise rules for the counting of particles in EM micrographs, as this influences the measurement accuracy of the number of particles, thus implicitly affecting the size values of the counted particles. We review and compare four different, commonly used methods for counting, which we then apply in case studies. The impact of the selected counting rule on the obtained final particle size distribution is highlighted. One main aim of this analysis is to support the application of a specific, well-defined counting approach in accordance with regulatory requirements to contribute to achieving more reliable and reproducible results. Interaction pathways between cells and the metal surface remain unclear. The development of this novel single cell-inductively coupled plasma-time of flight-mass spectrometry analytical method and a MIC-specific staining procedure facilitate the investigation of steel-MIC interactions. With this it is possible to analyze the multi-elemental fingerprint of individual cells. The detection method revealed elemental selectivity for the corrosive methanogenic archaeal strain Methanobacterium-affiliated IM1. The interface between material and environmental analysis thus receives special attention, e.g., when considering MIC on solid steel. Hence, the possible uptake of individual elements from different steel samples is investigated. Results showed the cells responded at a single-cell level to the different types of supplemented elements and displayed the abilities to interact with chromium, vanadium, titanium, cobalt, and molybdenum from solid metal surfaces. The information obtained will be used in the future to elucidate underlying mechanisms and develop possible material protection concepts, thus combining modern methods of analytical sciences with materials research. Per- and polyfluoroalkyl substances (PFAS) are a large group of chemicals used in the formulations of thousands of consumer goods, including aqueous film-forming foams used to suppress aviation fires in training scenarios, non-stick cookware, fast-food wrappers, water-repellent fabrics, medical equipment, and plastic and leather products. Because of the recent regulations and restrictions on the use of long chain (≥C8) PFAS a significant shift in the industry towards short (C4-C7) and ultrashort (C1-C3) chain alternatives has been recognized the last years. Due to the high polarity and water solubility of ultrashort PFAS, the potential for bioaccumulation is low. However, the high persistence of ultrashort-chain PFAS will result in environmental accumulation, especially in aquatic environments, leading to potential risks for aquatic organisms and increased human external exposure through drinking water. Ultrashort PFAS like trifluoroacetic acid (TFA) are low to moderately toxic to a range of organisms. In addition, ultrashort PFAS can penetrate natural and anthropogenic barriers and eventually reach drinking water sources. Because common drinking water treatment techniques do not sufficiently remove them, they may reach human consumption. In the project we are focusing on detecting and removing PFAS, especially ultrashort-chain PFAS from contaminated groundwater. We are designing passive sampling devices, which can collect and monitor the temporal profile of PFAS species in groundwater. This will allow us to analyze PFAS contaminations in German and Israeli groundwater using state-of-the-art and novel analytical techniques and understand the extent of contamination. In addition to quantification, PFAS contaminated groundwater will be treated via a two-stage process to produce PFAS-free drinking water. As ultrashort-chain PFAS are difficult to analyze with the current target (LC-MS/MS) and sum parameter (AOF, EOF) analysis methods, we additionally using gas chromatography – mass spectrometry (GC-MS). Therefore, an analytical method based on GC-MS is in development to analyze the volatile ultrashort-chain PFAS (TFA, PFPrA, TFMS, PFEtS, PFPrS, trifluoroethanol, pentafluoropropanol and hexafluoro isopropanol) directly in contaminated groundwater samples with the headspace technique and in eluates of organic solvents from the developed passive sampler after direct injection. Moreover, a two-stages process is designed to increase the low concentrations found in groundwater using novel membranes processes such as closed-circuit reverse osmosis (CCRO) and mixed matrix composite nanofiltration membranes (MMCM). Next, the rejected streams containing higher concentrations of PFAS will be treated by coagulation, and the remaining PFAS adsorbed onto carbonaceous nanomaterials (CNMs). Extractable (EOF) and adsorbable (AOF) organically bound fluorine as well as total fluorine (TF) were measured in samples fromriver Spree in Berlin, Germany, to reveal the advantages and disadvantages of the two techniques used as well as the two established fluorine sum Parameters AOF and EOF. TF concentrations determined via HR-CS-GFMAS and CIC were comparable between 148 and 270 μg/L. On average, AOF concentrations were higher than EOF concentrations, with AOF making up 0.14–0.81% of TF (determined using CIC) and EOF 0.04–0.28% of TF (determined using HR-CSGFMAS). The results obtained by the two independent methods were in good agreement. It turned out that HR-CS-GFMAS is a more sensitive and precise method for fluorine analysis compared to CIC. EOF and AOF are comparable tools in Risk evaluation for the emerging pollutants per- and polyfluorinated alkyl substances; however, EOF is much faster to conduct. The potential release of PFAS from accumulating electronic waste and lithium-ion battery (LIB) recycling activities has gained increasing attention in the past years. This creates a need for analytical methods tailored for the determination of PFAS out of environmental matrices related to the named activities or directly out of the concerned materials. In this work, analytical strategies for the monitoring of PFAS in LIB recycling materials were explored for a group of legacy perfluoro sulfonic- and carbonic acids (PFSA and PFCA) and of fluorinated sulfonylimides suspected to be LIB electrolyte ingredients. These analytical strategies comprehend PFAS target approaches with a herein optimized liquid chromatography tandem mass spectrometry (LC–MS/MS) method equipped with a HILIC (hydrophilic interaction liquid chromatography) column in combination with the TOP (total oxidizable precursor) assay and an adapted sample preparation method for high-matrix LIB recycling materials. The validated target method was applied to a set of LIB recycling materials: end-of-life batteries, black masses from hydrometallurgical recycling, and gas absorption solutions from thermal treatment of black masses as part of the recycling procedure. Investigation results show that the LIB industry can be connected to the release of both “LIB”-PFAS, such as the target sulfonylimides, and “already-legacy” PFAS, like the PFSA and PFCA. Especially, the presence of trifluoroacetic acid (TFA) as an emerging pollutant in every investigated LIB material type underlines the threat of PFAS emissions from LIB waste and recycling activities. The suitability of different chromatography columns was addressed: The use of PLOT (Porous Layer Open Tubular) columns is well suitable for VVOC analysis. The recoveries of the 60 analytes on several adsorbents and their combinations were determined: A combination of a graphitized carbon black and a carbon molecular sieve showed great results for all analytes. Carbon molecular sieves adsorb water which can impair the analysis. Different options such as purging, the use of a drying system or splitting were investigated for water removal. However, the interaction pathway between cells and the metal surface remains unclear. The development of the MIC-specific ICP-ToF-MS analytical method presented here at the single cell level, in combination with the investigation of steel-MIC interactions, contributes significantly to progress in instrumental MIC analysis and will enable clarification of the processes taking place. For this, a MIC-specific staining procedure was developed. It allows the analysis of archaea at a single cell level and provides information about the interaction of the cells with the staining agent which is extremely scarce compared to other well characterized organisms. Additionally, the single cell ICP-ToF-MS is used for the analysis of archaea involved in MIC of steel. Hence, the possible uptake of individual elements from different steel samples is investigated - the information obtained will be used in the future to elucidate underlying mechanisms and develop possible material protection concepts, thus combining modern methods of analytical sciences with materials. N2 - The fourth-century Greek Bible manuscript Codex Vaticanus B (Vat. gr. 1209) contains pairs of horizontally aligned marginal dots known as distigmai, which correspond to textual variants in other manuscripts. The production of the distigmai has been variously dated to the 4th or 16th centuries. A fourth-century date would prove the early existence of hundreds of textual variants, many of which are otherwise only witnessed by later manuscripts. Near infrared microscope reflectography combined with micro-X-ray fluorescence spectroscopy proved that the distigmai, both those categorized as “apricot” (which are visually similar to the ink of the original main text) and “chocolate” (which are visually similar to the reinking of the main text) were written with ink(s) made from chemically purified vitriol, a process that only became standard in the 16th century. As a result, there is no reasonable chance that the distigmai were written in the fourth century. Horizontal lines that have been hypothesized to function as text-critical obeloi were written in the same ink as the original main text, which differs completely from that of the distigmai. In other words, the distigmai and horizontal lines tested were not produced during the same writing session and are separated by more than 1,000 years, making it impossible for them to have functioned as conjoining text-critical symbols in the 4th century. As a result of the perpetual use of PFAS containing products, effluents and sewage sludge from wastewater treatment plants (WWTPs) have been observed to be an important pathway for PFAS into the environment. In Germany, phosphorus and other nutrients from sewage sludge and wastewater should be recycled in WWTPs of cities with a large population. However, it is not clear if PFAS contamination from wastewater and sewage sludge end up in novel wastewater-based fertilizers. Normally, PFAS are analyzed using PFAS protocols typically with time-consuming extraction steps and LC-MS-MS quantification. However, for screening of PFAS contaminations in sewage sludge or wastewater-based fertilizers also passive sampler based on the Diffusive Gradients in Thin-films (DGT) technique can be used for the PFAS extraction. Afterwards, combustion ion chromatography (CIC) can be applied to analyse the “total” amount of PFAS on the passive sampler. Here, we show results from the DGT method in comparison to those of the extractable organic fluorine (EOF) method for a variety of wastewater-based fertilizers. Products, such as building materials and furniture, emit volatile organic compounds (VOCs), which are therefore ubiquitous in indoor air. VOC in combination may, under certain environmental and occupational conditions, result in reported sensory irritation and health complaints. Emission concentrations can become further elevated in new or refurbished buildings where the rate of air exchange with fresh ambient air may be limited due to improved energy saving aspects. A healthy indoor environment can be achieved by controlling the sources and by eliminating or limiting the release of harmful substances into the air. One way is to use (building) materials proved to be low emitting. Meanwhile, a worldwide network of professional commercial and non-commercial laboratories performing emission tests for the evaluation of products for interior use has been established. Therefore, comparability of test results must be ensured. A laboratory’s proficiency can be proven by internal and external validation measures that both include the application of suitable emission reference materials (ERM). For the emission test chamber procedure according to EN 16516, no artificial ERM is commercially available. The EU-funded EMPIR project MetrIAQ aims to fill this gap by developing new and improved ERMs. The goal is to obtain a material with a reproducible and temporally constant compound release (less than 10 % variability over 14 days). Two approaches were tested: the impregnation of porous materials with VOC, and the encapsulation of VOC in polymer microcapsules. Impregnation is performed with help of an autoclave and supercritical CO2. The encapsulation is done by interfacial polymerisation on VOC droplets. For both approaches, synthesis and/or material parameters were varied to obtain an optimal ERM. Currently, the flask method is frequently used in Europe for market surveillance purposes to assess formaldehyde release from toys, but its concordance to levels measured in emission test chambers is poor. Surveillance laboratories are unable to afford laborious and expensive emission chamber testing to comply with a new amendment of the European Toy Directive; they need an alternative method that can provide reliable results. Therefore, the application of miniaturised emission test chambers was tested. Comparisons between a 1 m3 emission test chamber and 44 mL microchambers with two particleboards over 28 days and between a 24 L desiccator chamber and the microchambers with three puzzle samples over 10 days resulted in a correlation coefficient r2 of 0.834 for formaldehyde at steady state. The correlation between the results obtained in microchambers vs. flask showed a high variability over 10 samples (r2: 0.145), thereby demonstrating the error-proneness of the flask method in comparison to methods carried out under ambient parameters. An exposure assessment was also performed for three toy puzzles: indoor formaldehyde concentrations caused by puzzles were not negligible (up to 8 μg/m3), especially when more conservative exposure scenarios were considered. Röntgenfluoreszenzanalyse an Handschriften des Spätwerks N2 - Erstmalig in der Hölderlin-Forschung wurde mit dem Projekt der geisteswissenschaftlichen Methodik – über die Beachtung der Wasserzeichen hinaus – ein materialwissenschaftlicher Ansatz zur Seite gestellt, um Arbeitsphasen im Werk Friedrich Hölderlins sichtbar zu machen. Wesentlich ist dabei die Verzahnung von philologischer und naturwissenschaftlicher Perspektive. Wesentlicher Bestandteil der Messungen bildet das sogenannte Homburger Folioheft. Weiterhin wurden Briefe und Manuskripte aus den Beständen der Württembergische Landesbibliothek sowie zwei eigenhändige Gehaltsquittungen aus dem Hessischen Hauptstaatsarchiv Wiesbaden untersucht. Zwei in mehreren Manuskripten klar unterscheidbare Tinten lassen sich mit einiger Wahrscheinlichkeit biografisch mit dem Zeitraum zwischen Hölderlins Rückkehr aus der Schweiz im April 1801 und seinem Weggang nach Frankreich im Dezember dieses Jahres in Verbindung bringen. Für den in den untersuchten Handschriften häufigsten Tintentyp liegen datierbare Belege zwischen Januar 1803 und März 1804 vor, anzunehmen ist seine Verwendung zwischen Oktober 1802 und Juni 1804. Er dominiert auch im Homburger Folioheft. Eine vierte mehrfach vertretene Tinte gebraucht Hölderlin nach seiner Übersiedlung von Nürtingen nach Homburg. Sie findet sich im Folioheft und in einigen Einzelhandschriften. Damit können die in der Forschung umstrittenen Fragen geklärt werden, a) ob dem Dichter das Homburger Folioheft bei seinem zweiten Aufenthalt in Homburg zur Verfügung stand und b) ob überhaupt Werkhandschriften aus dieser Zeit erhalten sind. Darüber hinaus lässt sich über die Röntgenfluoreszenzanalyse der Arbeitsstand in den untersuchten Manuskripten zum Zeitpunkt des Ortswechsels im Juni 1804 genau angeben. Auch Einzelbefunde singulärer oder in nur wenigen Stücken belegter Tinten geben gelegentlich Anhaltspunkte zur Datierung. Zur Bewertung des Wiederverwendungspotenzials gibt es nur wenige Studien hinsichtlich der Schadstoffbelastung und der Anreicherung von Salzen in Gebäuden aus dem historischen Bestand. Tonminerale, die in Lehmbaustoffen als Bindemittel fungieren, können Schadstoffe binden. Die Region Mitteldeutschland ist geprägt durch jahrzehntelange Einwirkung erheblicher Schadstoffemissionen aus der Chemie- und Braunkohleindustrie. Der Eintrag von Schadstoffen in Lehmbaustoffe ist außerdem aus geogenen Hintergrundbelastungen denkbar, z. B. in Auenböden beeinflusst durch Einträge aus dem Erzbergbau. Salzbildende Anionen wie Chloride oder Nitrate können aufgrund ihrer hohen Löslichkeit mittels kapillarer Kräfte durch die Porenstruktur transportiert werden. In Lehmwänden können sie vor allem in oberflächennahen Lagen, in denen höhere Verdunstungsraten vorherrschen, durch Bindung mit in Lösung vorliegenden oder baustoffeigenen Kationen auskristallisieren und bauschädliche Salze ausbilden. Auch durch spezifische Nutzungen akkumulierte Schadstoffe, z. B. durch die Einwirkung von Tabakrauch, könnten theoretisch einer Wiederverwendung von Lehm entgegenstehen. In Anbetracht der Herausforderungen hinsichtlich der klimatischen und gesellschaftlichen Veränderungen, sind wissenschaftliche Erkenntnisse im Lehmbau erforderlich, die eine Integration in eine nachhaltige Kreislaufwirtschaft ermöglichen. Bei bisherigen Gesetzgebungsverfahren zu rezyklierten Baustoffen wurden Lehmbaustoffe noch nicht berücksichtigt. Ziel einer Vorstudie im Rahmen der WIR!-Initiative GOLEHM ist die Überprüfung der Wiederverwendbarkeit Wiederverwendbarkeit von Lehmbaustoffen aus historischen Bauten an Stichproben hinsichtlich der Umwelt- und Nutzerverträglichkeit. Für den Raum Mitteldeutschland können damit Anhaltspunkte darüber gegeben werden, welche Schadstoffe und bauschädlichen Salze in welchem Maße in Lehmbaustoffen aus dem Gebäudebestand erwartet werden können. Daraus können Hinweise zum Umgang mit Lehmbaustoffen aus historischen Gebäuden abgeleitet werden. Darüber hinaus wurden exemplarisch an zwei Gebäuden mikrobiologische Untersuchungen zur Ermittlung einer möglichen Belastung mit Schimmelpilzen vorgenommen. N2 - Earth construction materials, including earth from historical building stock, offer high potential in terms of their recyclability and sparing use of natural resources. Yet, only a few studies have been conducted on the levels of pollutants and the accumulation of salts in buildings from the historical building stock to enable an assessment of the potential for recycling. A protocol for the use of line-scans introduced in the beginning of 2000 permitted large scale and extremely successful investigations of the medieval iron-gall inks. Here, we usually focus on a comparison of the characteristic metallic components in the inks. In our work, we encountered cases that could be adequately addressed only using large area imaging XRF. However, conducting an M6 scan on a bound manuscript or on a scroll of huge dimensions is not a simple and easy matter. T1 - Implementation of matrix-matched semiquantification of PFAS in AFFF-contaminated soil N2 - This study presents a novel semiquantification approach for nontarget screening (NTS), combining matrix-matched calibration and ionization class-specific average calibration curves (ACCs) to address the lack of analytical reference standards for most per- and polyfluoroalkyl substances (PFAS). Ionization class-specific ACCs for carboxylic and sulfonic acids, sulfonamides, and cationic PFAS result in high accuracy, with median absolute accuracy quotients below 2.27×. The approach was applied to soil impacted by aqueous film-forming foam (AFFF) contamination. A total of 96 tentatively identified PFAS were semiquantified in addition to 28 quantified compounds based on available standards. Semiquantified concentrations exceeded those of target analytes, demonstrating the critical role of this method in capturing broader PFAS contamination. In this case, validation against extractable organofluorine (EOF) showed a 102% closed mass balance. The innovative approach not only enables comprehensive PFAS contamination assessment in complex matrices but also expands the scope of the NTS for environmental monitoring, remediation, and risk assessment of AFFF-contaminated sites. Since consumers may be exposed to these VOCs immediately after e.g. unwrapping of newly purchased products, reliable analytical methods are needed to determine air concentration profiles in the early phase of VOC emissions. Concerning typical sample sizes of consumer products, micro-scale test chambers (μ-chambers) were considered suitable for this purpose. In addition, compared to medium- or large-scale test chambers, μ-chambers require lower initial investment and operational costs, and offer options for emissions screenings where higher sample throughput is essential. For these reasons, a μ-chamber-based method to determine initial concentrations of VOCs emitting within the first hours of simulated product use was developed. The chamber air was frequently sampled on desorption tubes loaded with Tenax® TA, followed by quantification of emitted VOCs via thermal desorption gas chromatography mass spectrometry (TD-GC-MS). The method was assessed in a round robin test involving eight participating laboratories from 2 countries. Pieces of a rain poncho (sample area A: 2.01·10-4 m2) emitting cyclohexanone, phenol, 2-ethylhexanol, naphthalene and isophorone in significant concentrations were distributed as sample material. Each laboratory conducted triplicate emission tests over a period of 48 h in μ-chambers with volumes of 44 or 114 mL. The loading factors L of 4.57 m2·m-3 (44 mL) or 1.76 m2·m-3 (114 mL) and air flow rates of 24 mL·min-1 resulted in area-specific air flow rates q of 7.17 m3·m-2·h-1 in both chamber types. The temperature was set to 23°C and, if possible, relative humidity was set to approx. 50%. The chamber air was sampled after 0.5, 1, 2, 4, 24 and 48 hours. In addition, to assess the analytical performance, each participating laboratory received two analyte-spiked Tenax® TA-loaded desorption tubes for quality control (QC). Very low relative standard deviations (11–17%) were obtained for the QC test. The results of the emissions tests in the early phase (0.5–4 h) were also adequate, with acceptable z scores (|z|≤2.00) received by six to seven participating laboratories. All eight laboratories obtained acceptable z scores after the longer sampling periods (24 and 48 h). Hence, the inter-laboratory comparability of the results obtained with the developed μ-chamber method was successfully confirmed. Black writing inks in the period under consideration can be divided into three different classes: soot, tannin and iron-gall ink. The first one is a fine dispersion of carbon pigments in a water-soluble binding agent, the second one is a solution of plant extract from tree barks or gallnuts, while the third one contains both soluble and insoluble phases. This last ink is based on metal (iron) and is produced by a chemical reaction of soluble iron (II) with tannin in aqueous solution. Tracing the transition from writing inks based on carbon pigments (soot or charcoal), common in Antiquity to the iron-gall inks commonly used in the Middle Ages builds a focus of our investigative work at the BAM (Federal Institute of Material Analysis and Testing). The Bologna Scroll has some unusual scribal features. According to one opinion in the Babylonian Talmud, a Torah scroll must be written with carbon ink to be valid for liturgical use.12 The main inks of the Bologna Scroll are very black, and the inks of the marginal corrections in tiny letters are even blacker. In this study, eh inks were tested by means of XRF-spectroscopy and NIR imaging. Conreary to the expectations, the current tests have definitively determined that the Bologna Scroll was both written and corrected using multiple iron-gall inks. However, standardized methods for organofluorine analysis in a broad suite of matrices are currently unavailable, including a method for extractable organofluorine (EOF) measured using combustion ion chromatography (CIC). Here, we report the results of an international interlaboratory comparison. Seven laboratories representing academia, government, and the private sector measured paired EOF and PFAS concentrations in groundwater and eel (Anguilla rostrata) from a site contaminated by aqueous film-forming foam. Among all laboratories, targeted PFAS could not explain all EOF in groundwater but accounted for most EOF in eel. EOF results from all laboratories for at least one replicate extract fell within one standard deviation of the interlaboratory mean for groundwater and five out of seven laboratories for eel. PFAS spike mixture recoveries for EOF measurements in groundwater and eel were close to the criterion (±30%) for standardized targeted PFAS methods. Instrumental operation of the CIC such as replicate sample injections was a major source of measurement uncertainty. Blank contamination and incomplete inorganic fluorine removal may introduce additional uncertainties. To elucidate the presence of unknown organofluorine using paired EOF and PFAS measurements, we recommend that analysts carefully consider confounding methodological uncertainties such as differences in precision between measurements, data processing steps such as blank subtraction and replicate analyses, and the relative recoveries of PFAS and other fluorine compounds. Due to their harmful (environmental) potential, further dispersion in the environment needs to be stopped, which can be achieved by appropriate absorption materials. In this work, the influence of the cross-linking agent epichlorohydrin (ECH) concentration on the perfluorooctanesulfonic acid (PFOS) adsorption capacity of chitosan gel was investigated. It was found that higher ECH concentration during the cross-linking step decreases the PFOS adsorption capacity of the cross-linked chitosan gel from 0% to 4% ECH solution by about 15%. Using a concentration of 1%, ECH resulted still in an acid-stable material, and a maximum PFOS loading capacity of 4.04 mmol/g was obtained, one of the highest described in the literature. Furthermore, we used a rapid small-scale column test to compare the PFOS adsorption capacity of chitosan and activated carbon, each in both milled and unmilled form. Unmilled chitosan showed the highest PFOS adsorption capacity considering adsorption material dry masses (>0.9 and <0.4 mmol/g for both types of chitosan and activated carbon, respectively). Milled activated carbon proved to be the better adsorption material, considering the fixed volume of the adsorber (>99.9% PFOS adsorbed). Overall, the cross-linking agent concentration in chitosan is a crucial factor influencing its PFOS absorption potential. Our results feature cross-linked chitosan as an effective economic and ecologic alternative for PFOS adsorption in aqueous solutions. The EOF values for ten sewage sludges from Germany and Switzerland range from 154 to 7209 mg kg−1. For thermal treated sewage sludge and struvite the EOF were lower with values up to 121 mg kg−1. Moreover, the application of PFAS targeted and suspect screening analysis of selected sewage sludge samples showed that only a small part of the EOF sum parameter values can be explained by the usually screened legacy PFAS. The hitherto unknown part of EOF sum parameter contains also fluorinated pesticides, pharmaceutical and aromatic compounds. Because these partly fluorinated compounds can degrade to (ultra-)short PFAS in wastewater treatment plants they should be considered as significant sources of organic fluorine in the environment. The combined results of sum parameter analysis and suspect screening reveal the need to update current regulations, such as the German fertilizer ordinance, to focus not solely on a few selected PFAS such as perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) but consider an additional sum parameter approach as a more holistic alternative. Moreover, diffusion gradient in thin-films (DGT) passive samplers were utilized as an alternative simplified extraction method for PFAS in solid wastewater-based fertilizers and subsequently quantified via combustion ion chromatography. However, the DGT method was less sensitive and only comparable to the EOF values of the fertilizers in samples with >150 mg kg−1, because of different diffusion properties for various PFAS, but also kinetic exchange limitations. Today these are also known as the Humboldt fragments and represent the most significant part of a collection that was augmented over the years by the purchase of various types of Mesoamerican manuscripts, mainly in indigenous languages with Latin script. During the two year period from April 2017 to March 2019, the Berlin State Library (SBB - SPK) and the Bundesanstalt für Materialforschung und -prüfung (BAM) carried out a joint scientific project funded by the Deutsche Forschungsgemeinschaft (DFG) entitled "Material-analytical and cultural-historical examinations of colonial-period manuscripts from Mexico in Berlin and Cracow." The main objectives of the project were to determine the provenance of the Manuscripta americana and, since they exhibit a high degree of fragmentation, to examine how they are related to each other and to objects outside the collection. Testing was conducted using the following techniques: 3-color photography, 3D-digital microscopy, scanning μ-X-ray fluorescence analysis (XRF), μ- Raman and FTIR spectroscopy. I will give a short explanation of the principles of the techniques and explain the choice of the instruments. The main task of our work at the BAM required characterization of the inks and comparison of the ink as well as the composition of parchment. For the ink characterization we have followed the protocol developed at the BAM. The standard protocol includes three non-invasive stages: a) near infrared photography (NIRR) to determine the ink type or its main component; b) X-ray fluorescence analysis (XRF) to determine a fingerprint for iron-gall inks or contaminants in carbon/plant inks; c) Raman spectroscopy if there is an indication that the inks are of a mixed type. For parchment characterization, we usually employ µ-XRF-mapping and FTIR to gain information on specific treatments experienced by the parchment during its preparation and history. T1 - Material Characterisation of 19–20th Century Manuscripts from Northern Thailand N2 - Material analysis was carried out on four manuscripts from Northern Thailand, which included two palm-leaf manuscripts and two paper manuscripts. The two palm-leaf manuscripts MS 6 and MS 7 were found to have been written in the traditional method, wherein text was incised on the surface of the leaves, and then soot applied to the surfaces, confirmed by the identification of carbon ink. MS 7 additionally showed the presence of trace levels of iron in the ink, either from soot paste or from the stylus used for inscribing. The paper manuscript MS 3 was written in iron-gall ink, with sections written using a methyl-violet based ink while MS 4 was written in carbon ink. The paper used in the case of MS 3 was found to be machine made, while khoi fibres (Streblus asper) were used for making the paper used for MS 4. A combination of traditional and modern pigments, like molybdenum orange, was used for decorating the edges and cover. The results improved our understanding of these manuscripts in particular, and also provided us with insights about the rapid adoption of modern materials and their incorporation into the production of written artefacts from Northern Thailand in the late 19th and early 20th centuries. T1 - Materials and materiality as keys to understanding a map of Mount Kailash N2 - While graphic and linguistic content is central to the identification of maps and can provide insights into the concepts and systems of rules, ideas and beliefs that led to their production, the materials that constitute maps may speak about manufacturing processes, the provenance of raw materials, the practical and technical knowledge of the mapmakers, and the use of the maps. This contribution underscores how the materials employed in Tibetan maps reveal aspects of their context and trajectory. Furthermore, materiality, conceptualized as a convergence of matter and imagination, can yield even more profound insights into artefacts like maps. Through a detailed case study of a map of Mount Kailash in the tangkha format and a specialized material-scientific analysis of the colourants employed in its creation, this contribution explores how a map can ‘talk’ through its materiality. With this case study it will show how Tibetan Studies can benefit from interdisciplinary collaboration. Crossing the field’s traditional boundaries and working with other disciplines, we argue, is absolutely essential for serious research. It also gives directions to adapt this methodology to very volatile organic compounds (VVOCs). Indeed, toxicologically based guideline values are being implemented for these compounds and it becomes necessary to measure them. But a comprehensive and robust measurement method is lacking. This work highlights the points that still need to be explored towards the standardisation of a suitable procedure: investigations on sorbent combinations, the suitability of chromatography columns and the use of gaseous standards are required. The biggest challenge remains in the fact that strong sorbents adsorb water together with VVOCs. Water may impair the analysis and the optimal approach to eliminate it is still to be found and integrated into the sampling strategy. Ziel des Vorhabens war es, durch die Auswertung vorhandener und durchzuführender Untersuchungen eine Einstufung von Bodenmaterialien (jährlich fallen weit über 100 Mio. Tonnen davon an) für bestimmte Verwertungswege vornehmen zu können. Es sollte herausgefunden werden, ob Veränderungen hinsichtlich des Verwertungspotentials nachgewiesen werden können. Basis für die Durchführung und Bewertung waren die neuen Regelungen (hier insbesondere die Untersuchung mit einem Wasser/Feststoffverhältnis von 2:1, neue nationale und europäische Elutions- und Untersuchungsverfahren) der novellierten BBodSchV. Der Fokus lag dabei auf den polyzyklischen aromatischen Kohlenwasserstoffen (PAK) sowie ausgewählte anorganische Stoffe (Schwermetalle und Anionen) unter der Berücksichtigung unterschiedlicher Bodenmaterialien zur Verwertung. Dabei handelte es sich um: - Böden aus urban geprägten Gebieten, - Oberböden aus dem Straßenrandbereich in Form von Bankettschälgut, - Baggergut aus Gewässern mit Verdacht auf eine Kontamination, - Bergematerial (Mischung von Abraum- und Bodenmaterial), - Auenböden mit vermuteten Verunreinigungen aus Industrie und Gewerbe. Anhand der vorgelegten Ergebnisse kann nun eine Versachlichung der Diskussion um Ressorcenschutz vs. Schadlosigkeit der Verwertung erfolgen. Sie zeigen für die untersuchten Stoffe, dass bei einer Einführung der §§ 6-8 Novelle BBodSchV mit einer Erhöhung der Verwertungsquote für Bodenmaterial gerechnet werden kann. Eine Erhöhung des Deponievolumens ist somit nicht zu befürchten. Neben den ermittelten Daten konnten Grundlagen für die Überarbeitung verschiedener DIN-Normen für Bodenuntersuchungen erarbeitet werden. T1 - Non-extractable PFAS in functional textiles − Characterization by complementary methods: oxidation, hydrolysis, and fluorine sum parameters N2 - Per- and polyfluoroalkyl substances (PFAS) are widely used for durable water-repellent finishing of different fabrics and textiles like outdoor clothing, carpets, medical textiles and more. Existing PFAS extraction techniques followed by target analysis are often insufficient in detecting widely used side-chain fluorinated polymers (SFPs) that are barely or non-extractable. SFPs are typically copolymers consisting of a non-fluorinated backbone with perfluoroalkyl side-chains to obtain desired properties. We compared the accessible analytical information and performance of complementary techniques based on oxidation (dTOP assay, PhotoTOP), hydrolysis (THP assay), standard extraction, extractable organic fluorine (EOF), and total fluorine (TF) with five functional textiles and characterized 7 further textiles only by PhotoTOP oxidation. The results show that when applied directly to textile samples, oxidation by dTOP and PhotoTOP and also hydrolysis by the THP are able to capture large fractions of the TF in form of perfluoroalkyl side-chains present in the textiles while methods relying on extracts (EOF, target and non-target analysis) were much lower (e.g., factor ~25-50 lower). The conversion of large fractions of the measured TF into PFCAs or FTOHs from fluorinated side chains is in contrast to previous studies. Concentrations ranged from
