It is also essential to take into account the necessary measurement equipment for the respective property change, in order to allow timely intervention. What are the key issues on choosing a weathering reference material? What can be learned from the weathering reference materials, investigated so far? T1 - Ecodesign spinning towards the circular Economy - The contribution of new standards on the material efficiency N2 - Whilst the concept of ecodesign is intended to account for the environmental impacts of the product across its whole lifecycle, European ecodesign policy to date has been heavily focused upon energy efficiency in the use phase. The Ecodesign Directive and Energy Labelling Regulation have achieved substantial energy savings since their inception. For many products, the magnitude of savings available from further tightening existing energy efficiency requirements is now often outweighed by savings that can be achieved by material efficiency requirements. Further, there are some products such as smartphones for which there are limited gains to be made in energy efficiency but for material efficiency. In the Commission’s Circular Economy Action Plan from 2015, a new direction was defined for ecodesign policy, to systematically examine considerations such as reparability, durability, upgradability, recyclability, or the identification of certain materials or substances. The Commission consequently issued a standardisation request (M/543, 2015) to the European Standardization Organisations (CEN / CENELEC / ETSI) to develop generic/horizontal standards relevant to energy-related products that address these major aspects of material efficiency. In this study we evaluate the potential contribution of these new standards (with specific focus on the standards for repair, durability and recyclability) to future developments under the European Ecodesign Directive. Previous studies have shown that antibiotics, metals and pesticides affect the underlying processes of resistance evolution and spread; namely de novo mutagenesis and horizontal gene transfer by conjugation and transformation in microbial communities. However, it is unknown if active substances used in material preservatives are involved in these processes. We show that biocides used as material preservatives affect rates of mutation and conjugation in microorganism in a species- and substance-dependent manner, while rates of transformation are not directly affected. Our data highlights the importance of assessing the contribution of material preservatives on AMR evolution and spread in the environment. It is used as an osmolyte helping microorganisms to maintain an osmotic equilibrium. In addition, ectoine acts as a stabilizer and protects proteins, membranes and whole cells against detrimental effects such as freezing and thawing, drying and high temperatures. Its protective effect is explained by the preferential exclusion model, which postulates that ectoine does not directly interact with biomolecules but is excluded from their surface. Interestingly, details on the interaction of ectoine with DNA are still unknown. Therefore, we studied the influence of ectoine on DNA and the mechanisms by which ectoine protects DNA against ionizing radiation. To emulate biological conditions, we used a sample holder comprising a silicon chip with a Si3N4 membrane, which allows for electron irradiation of DNA in aqueous solution. Analysis by atomic force microscopy revealed that without ectoine, DNA was damaged by irradiation with a dose of 1,7 +/-0,3 Gy. With ectoine, DNA remained undamaged, even after irradiation with 15 Gy. Simulations with dsDNA and ectoine in water revealed a preferential binding of the zwitterionic ectoine to the negatively charged DNA. According to the simulations, binding of ectoine will destabilize dsDNA. Destabilizing is probably caused by the transition of B-DNA to A-DNA and will reduce the DNA melting temperature, which was experimentally proven. The preferential binding provides a stable ectoine shell around DNA, which allows ectoine to reduce OH-radicals and electrons near the DNA and thereby mitigating the damaging effect of ionizing radiation. Prior to the formation of a biofilm on a substrate, planktonic cells attach on the surface. Hence, the properties of the surface play a key role in biofilm formation and are of great importance for the development of strategies to prevent bacterial attachment and biofilm formation. This project aims at clarifying to which extent surface micro-/nanostructuring and chemical functionalization affects bacterial attachment and whether a synergistic combination of the two can be used to control bacterial adhesion. To answer these questions, model surfaces with regular patterns of 5-10 micrometers in size have been prepared, which provide distinct zones differing in terms of their chemistry or nano-roughness. This was achieved by micro contact printing of self-assembled monolayers with different functional groups and deposition of patterned ZnO nanorod arrays for studying the effect of surface chemistry and morphology, respectively. Typical contrasts studied were combinations of positively/negatively charged, hydrophobic/hydrophilic or flat/rough. The attachment behavior of bacteria on tailored surfaces were studied in a flow chamber as a function of time. The strain Pseudomonas fluorescens SBW25 was chosen as a model organism. DNA-intercalating dyes such as Syto9 have a high affinity to adsorb on ZnO nanorods. To overcome this limitation a genetic modification was performed by introducing a gene which expresses a green fluorescent protein in P. fluorescens SBW25 enabling the quantitative evaluation of the flow chamber studies by means of fluorescence microscopy. Further analysis of the attachment behavior was performed by means of scanning electron microscopy. The presentation will summarize the results of our systematic study on the role of individual parameters on bacterial attachment and highlight synergistic combinations, showing an inhibition or enhancing effect. As the investigations with model substrates enable a precise control of the surface parameters, this approach can be applied to different microorganisms and material systems to achieve a correlative description of bacterial adhesion on solid surfaces. Um darüber hinaus Schweißzeit, Schweißnahtvolumina und Schweißkosten zu minimieren, kam es zur Entwicklung moderner Lichtbogenprozesse mit erhöhter Abschmelzleistung, wie bspw. dem modifizierten Sprühlichtbogen. Das Schweißen mit modifiziertem Sprühlichtbogen bietet die Möglichkeit, schmalere Nahtfugen bzw. kleinere Nahtöffnungswinkel zu schweißen. Mit zunehmender Festigkeit der Baustähle steigen allerdings die Anforderungen an die schweißtechnische Verarbeitung. Hochfeste Feinkornbaustähle weisen ein erhöhtes Risiko gegenüber der wasserstoffunterstützten Kaltrissbildung auf. Diese Werkstofftrennungen entstehen unter gemeinsamer Wechselwirkung von lokalem risskritischem Gefüge, lokal erhöhter Wasserstoffkonzentration und lokal erhöhter Beanspruchung bzw. Dehnung. Untersuchungen haben gezeigt, dass der Einsatz des modifizierten Sprühlichtbogens in Verbindung mit schmalen Nahtfugen zu erhöhten Wasserstoffkonzentrationen im Schweißgut führt. Die vorliegende Arbeit gibt einen Aufschluss hinsichtlich der Kaltrissempfindlichkeit des vergüteten, hochfesten Feinkornbaustahls S960QL anhand der durchgeführten Kaltrissprüfung mittels selbstbeanspruchenden TEKKEN-Tests. Dazu wurden die Nahtöffnungswinkel der Prüfnähte zwischen 30° (Einsatz des modifizierten Sprühlichtbogens) und 60° (Einsatz des konventionellen Übergangslichtbogens) variiert. Ferner wurde die Wirksamkeit einer Nachwärmprozedur aus der Schweißwärme heraus zur Vermeidung der wasserstoffunterstützte Kaltrissbildung untersucht. Die Ergebnisse weisen in allen Proben (ohne Nachwärmung) Kerbrisse auf, welche als Kaltrisse identifiziert werden konnten. Durch eine Nachwärmung unmittelbar nach dem Schweißen können diese gänzlich vermieden werden. Das Schweißgut mit modifizierten Sprühlichtbogen weist die höchsten mittleren Wasserstoffkonzentrationen auf. Ferner zeigt das Schweißgut unter reduziertem Nahtöffnungswinkel eine vermehrte Mikrorissbildung auf. Petroleum-, On- and off-shore, infrastructure. It is estimated that 20% of all corrosion damage is caused by microorganisms or microbiologically influenced corrosion (MIC). Several microorganisms are known to cause corrosion, including sulfate-reducing bacteria, nitrate-reducing bacteria, methanogens etc. For several years, methanogens were regarded as a mild corroder (~0.065 mm/yr), largely due to a lack of detailed investigation on the corrosion mechanism under real-environment simulated conditions. Resulting in the common belief that siderite, a non-conductive compound, is the sole corrosion product (CP) of methanogen-induced MIC (Mi-MIC). To simulate natural environmental conditions, we developed and introduced the multiport flow column system (MFC), a multi-sectional corrosion flow-cell. Using the MFC, we obtained ten times higher corrosion rates than previously reported. With a combination of several analytical techniques, such as ToF-SIMS, SEM-EDS and FIB-SEM, we found strong indication that siderite is not the sole corrosion product of Mi-MIC. The corrosion layers contained phosphorus, oxygen, magnesium, calcium and iron. The differences in the CP between static and dynamic environments demonstrated the impact of testing procedures on the corrosive potential of methanogens. To further verify and deepen our understanding of Mi-MIC, we are currently studying the influence of additional environmental parameters (e.g. pH, salinity, flow rate) on Mi-MIC. Overall, results of this study will expand the current understanding of MIC from both analytical and mechanistic points of view, thus aiding the development of different mitigation strategies for various industry sectors. For a long time, the impact of methanogen-induced MIC (Mi-MIC) was underestimated. This was mainly due to the rather low published corrosion rates and the presumed corrosion product siderite, which is not electrically conductive. In our laboratory, we were able to show that this trivialization or underestimation of Mi-MIC was due to the nature of the testing. The static systems used so far do not provide environmentally relevant information about the corrosion rate and the corrosion product, especially for methanogens To further illustrate the importance of the environment on the MIC process, we established a multiport flow column (MFC), to simulate the natural environment. With this method, and in contrast to published results using the static conditions, we obtained ten times higher corrosion rates. We analyzed the corrosion products with a combination of techniques, like ToF-SIMS, SEM-EDS and FIB-SEM, and found strong evidence that siderite is not the sole corrosion product of Mi-MIC. The corrosion layers contain phosphorus, oxygen, magnesium, calcium and iron and lacked on carbon-related species. It is hypothesized that methanogens may have influenced the nucleation process of siderite, converting bicarbonate into carbon dioxide for methanogenesis. This results in increased localized corrosion and reduced siderite formation. To verify and deepen our understanding of Mi-MIC, we are currently studying the influence of additional environmental parameters (e.g. pH, salinity, flow rate etc.) on Mi-MIC and the subsequent impacts on corrosion rates and the corrosion products. Plastics are in the focus of environmental politics due to their long-term behaviour and therefore to their persistence. Not only that they appear as visible contaminants in the sea and on the beach, but their unknown behaviour concerning their additives as well as the related transformation products are anxious. Therefore, we wish to establish a certified reference method to provide a method for testing plastics. Aim of this project is the development of fast motion standard reference methods for testing plastics regarding to their environmental compatibility. To establish these testing methods, we use polystyrene (PS) and polypropylene (PP) with environmental relevant brominated flame retardants, known for their persistent bioaccumulative and toxic (PBT) properties. In case of PS the material contains 1 wt% of 1,2,5,6,9,10-hexabromocyclododecan (HBCD) and in case of PP 0.1 wt% bromodiphenylether (BDE-209), which is known as a substance of very high concern (SVHC). Furthermore, we use polycarbonate (PC), which is still used as material in baby flasks and releases Bisphenol A (BPA), an estrogenic active substance. As an additional material PTFE is used for its importance as a source for two ubiquitous environmental substances (PFOS and PFOA), whose toxicological effects are still incompletely known. The focus in this current work is set on the transfer of potential pollutants out of applied materials mentioned above into environmental compartments like water or soil. Here an accelerated aging concept should be developed to shortened time consuming natural processes. For these resulting simulations we use a programmable weathering chamber with dry and wet periods and with high and low temperatures. All studied polymer samples were exposed to a defined weathering schedule in a climate chamber in accordance to regulation EN ISO 4892-3:2006. For the determination of BDE-209 in the collected raining water the samples were prepared in accordance to a validated protocol. Before the analyses each sample was spiked with isotopically labeled BDE-209. Subsequently the samples were extracted with isooctane. The obtained extracts were concentrated, and the resulting solutions were analyzed by GC/MS. Additionally, the total bromine content was monitored for the weathered and untreated samples using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) and X-ray fluorescence analysis (XRF) as a non-destructive and rapid method. In general, the resulting data from the accelerated weathering will be compared to those from the natural weathering experiments. Here, the surfaces of the test pieces were analyzed by LA-ICP-MS and XRF as well. Moreover, soil bed tests were conducted in a well characterized model soil. This soil (boulder-clay, sand with 12 % loam, particle size in total 0.2-4 mm) is filled in a free-draining basin inside of an air-conditioned cellar. In this manner, TOC, water capacity and humidity are recorded parameters. To induce a leaching process from the samples by the raining water, the target water content is calculated to 8%. The actual humidity is measured by a tensiometer, assuring the duration of the raining period. A defined humidity is a fundamental parameter for a biological activity in the soil. The test polymers were placed up to the half in the soil. Microbial activity of the soil is monitored by a reference polymer (polyurethane) and should induce the release of HBCD and BDE-209 out of the test materials. These released analytes will be captured by passive samplers (silicone tubes) placed in a distinct distance to the polymer samples in the soil. Bodenmaterial kann in Abhängigkeit seiner vorherigen Nutzung oder Herkunft mit Schadstoffen belastet sein. Seine weitere Verwertungsmöglichkeit hängt nicht nur vom Schadstoffgehalt im Feststoff, sondern auch von der Mobilisierbarkeit dieser Stoffe ab. Die Paragraphen 6 bis 8 der vorgesehenen Novelle der BBodSchV enthalten neue Vorgaben hinsichtlich des Auf- und Einbringens von Bodenmaterial unterhalb oder außerhalb einer durchwurzelbaren Bodenschicht (4. Referentenentwurf Mantelverordnung, Stand: 06.02.2017). Es soll die Möglichkeit eingeräumt werden, Bodenmaterialien mit Feststoffgehalten von regulierten Stoffen bei Überschreitung des Vorsorgewertes einer weiteren Verwertung zuzuführen unter der Voraussetzung, dass die Eluatwerte eingehalten werden. Für die Auswirkung dieser neuen Regelungen auf die Verwertung von Bodenmaterialien bestehen bisher noch Datendefizite besonders für organische Stoffe. Durch die Umstellung des Elutionsverfahrens von einem W/F von 10 l/kg (bisherige Regelung in der LAGA M 20) auf ein W/F von 2 l/kg bestehen zusätzliche Unsicherheiten im Hinblick auf die Einstufung von Bodenmaterialien zu Verwertungszwecken. Im Rahmen eines UFOPLAN-Projektes (FKZ 3716 74 203 0) wurden unterschiedliche Bodenmaterialien verschiedenen Ursprungs (z.B. Baggergut, Bankettschälgut, Stadt- und Auenböden, Bergematerial) mit besonderem Fokus auf PAK untersucht. Von den unter die Kriterien fallenden, ausgewählten Bodenmaterialien wurden Feststoffgehalte, TOC-Gehalte sowie die Eluatwerte aus Säulenversuchen nach DIN 19528 und Schüttelversuchen nach DIN 19529 bei einem Wasser-/Feststoffverhältnis (W/F) von 2 l/kg und 10 l/kg ermittelt und gegenübergestellt. The common structural motif of this compound class is the fluorine – carbon bond on an alkyl backbone, which are known for their chemical inertness and combination of hydrophobicity and oleophobicity. The downside of those properties is the general lack of usable molecular interactions for adsorption to extract them from the environment. The notable exception to that inertness are fluorine – fluorine interactions, which are stronger, the longer the perfluorinated chain length is. Therefore, in this project we investigated the effect of fluorination, length and presence of a charge on the alkyl chain on an adsorber. Towards that goal, in total 16 different adsorber materials were synthesized (fluorinated/non-fluorinated; C4/ C6/ C8/ C10 ; positive charge/no charge) and tested in regard to their potential to adsorb PFOA and perfluoralkylic acids of differing chain lengths. The PFAS concentration was tested via LC-MS/MS to determine the adsorption rate on the various materials. Further work includes the transition from a target analytics to sum parameter analysis for future evaluation of adsorber materials in PFAS environmental samples like river samples. Sie verfügen über eine breite Toleranz gegenüber abiotischen Umweltparametern und sind in der Regel Nahrungsopportunisten. Sie können längere Zeit ohne Nahrungsaufnahme überdauern und haben unter optimalen Bedingungen hohe Reproduktionsraten. Es stellt sich die Frage, ob die auffälligen Anpassungen von Vorrats- und Materialschädlingen an einen menschennahen Lebensraum das Resultat von Prädispositionen und Selektion hin zu einem Lebensformtyp „Lager- oder Materialschädling“ sind, oder ob die synanthropen künstlichen (Öko)Systeme nur als überdimensionierte natürliche Reservoire fungieren. T1 - Factors Influencing Droplet Corrosion in Dense Phase CO2 N2 - This work examined the factors that influence the droplet corrosion of CO2 pipeline steels caused by oxyfuel flue gases in dense phase CO2 at 278 K, simulating the underground transport conditions. The wetting properties were studied by contact angle measurement, revealing pH and time dependency on the reactive wetting behaviors of carbon steel X70. Exposure tests with CO2 saturated water droplet on steel surface showed that the impurities (220 ppmv SO2 and 6700 ppmv O2) diffused into the droplet and then reacted with metal surfaces in dense phase condition, forming the corrosion products. The corrosion rate was confirmed strongly depending on the droplet volume as well as the SO2 concentration. Condensation experiments carried out on freshly polished coupons in CO2 with 200 ppmv H2O, 220 ppmv SO2 and 6700 ppmv O2, showed that the formation and aggregation of droplets is time and temperature dependent. At 278 K, condensation happened stronger and the corrosion products, mainly consisted of dense hydrated FeSO3/FeSO4. While at 288 K, more fluffy corrosion products consisting of iron oxide/hydroxide and hydrated FeSO3/FeSO4 were found. Further exposure tests on carbon steel coupons with different surface roughness did not reveal the difference in weight loss/gain and therefore the corrosion rate. Aflatoxin B1 (AFB1) is a toxic low-molecular-weight secondary fungal metabolite produced mainly by Aspergillus flavus and Aspergillus parasiticus not only at pre-harvest time but also at post-harvest stages including storage. AFB1 was classified as a Group I carcinogen by the World Health Organization for Research on Cancer in 1993. AFB1 is able to naturally contaminate medicinal plants and therefore causing serious health issues for humans consuming the related medicine. This study aimed to develop an efficient fluorescence polarization immunoassay (FPIA) and for the first time a rapid (5-10 min), low-cost, and simple membrane-based flow-through immunoassay (MBA) for determination of AFB1 in medical herbs based on a monoclonal antibody. Methods. Two different techniques for AFB1detection in medical herbs (Herba Orígani vulgáris, Folia Urticae, Fructus Rubi idaei) were developed and compared, namely an easy-to-use semi-quantitative flow-through membrane-based enzyme immunoassay (MBA), and a homogeneous method which needs no separation or washing steps (assay time 10 min), a quantitative fluorescence polarization immunoassay (FPIA). Results. A cut-off level of the developed MBA can vary wide from 0.8 ppb to 1 ppb. The FPIA method showed a linear working range of 8.6 ppb to 64 ppb, IC50 of FPIA is 24 ppb. The results were in good correlation with the ELISA results (IC50 = 0.1 ppb). LC–MS/MS was used to confirm the results, too. Conclusion. MBA can be used for a quick on-site AFB1 detection in medical herbs (Herba Orígani vulgáris (Oríganum vulgáre), Folia Urticae (Urtíca dióica), Fructus Rubi idaei (Rúbus idáeus)), the developed test does not require special equipment and is not time-consuming (5-10min), includes several simple steps and can be performed directly on-site. The major advantages of the developed FPIA are its simplicity and suitability for a rapid screening of a large number of samples. Mit zunehmender Bewitterungszeit nimmt die Korrosionsgeschwindigkeit stetig ab, während postuliert wird, dass die Geschwindigkeit des Runoffs über die gesamte Zeit konstant bleibt. Dieses Verhalten des Runoffs muss aber zunehmend bezweifelt werden. Die Gesamtmenge an abgezehrtem Metall beinhaltet zwei Bestandteile. Ein Teil wird in die Schicht aus Korrosionsprodukten eingebaut, der andere wird aufgrund von Niederschlägen aus der Patina gelöst. Aus diesem Grund ist die Menge an Zink-Runoff stark abhängig von den Eigenschaften der Korrosionsprodukt- bzw. Patinaschicht und nicht anhand der Zusammensetzung des Metalls abschätzbar. Vorhandene Modelle können die Menge an Runoff, und damit verbunden die Abzehrung einer Schicht aus Zink und Kupfer, sowie den resultierenden Eintrag von Schwermetallionen in die Umwelt nicht mehr darstellen und erklären. Um den Korrosionsschutz an Bauwerken zum einen dauerhaft unter Beachtung von Kostenaspekten und Ressourcenschonung und zum anderen unter ökologischen Gesichtspunkten auslegen zu können, müssen deshalb neue wissenschaftliche Grundlagen erarbeitet und die Mechanismen beschrieben werden. The critical point is the lack of fast, harmonised methods for sampling, sample preparation and sample analysis. These three analytical steps are dependent on one another and must be optimised. In recent years, we developed a method for representative sampling and fast detection of MP in aqueous systems. The sampling in different freshwater bodies is performed in the field with a fractionated filtration system using mesh sizes of 500, 100 and 50 µm. For water with an intermediate or high content of suspended particular matter a minimum of 1000 L has to be filtered. In the lab, mesh sizes of 10 and 5 µm are used for further filtration. Subsequently, the water filtrates of the different particle size classes are sterilised, dried, weighed and homogenised, if necessary. Conventional methods for MP analysis are infrared and raman spectroscopy, giving information on the shapes and numbers of individually identified MP particles. Our focus is on the determination of mass contents of various polymers potentially contained in environmental samples. For qualitative and quantitative MP detection TED-GC-MS is used, a two-step method based on gas chromatography-mass spectrometry (GC-MS) with previous thermal extraction. This method not only enables us to screen the samples for characteristic marker-molecules, thus identifying single polymers, but furthermore allows the calculation of mass contents of individual polymers. In addition, initiatives on recyclability and other aspects of sustainable products can be observed in France under the Ecomodulation Approach for some electrical and electronic products. Neither the Commission's ambitions nor the Ecomodulation Approach have so far led to a crucially better ecodesign of products. Therefore, at the EU level, indices for reparability (already developed for smartphones and tablets), recyclability and recycled content of products are being discussed within the Ecodesign legislative framework in an attempt to achieve better ecodesign through advanced consumer purchasing decisions. The product standards requested by the European Commission to assess the recyclability (EN4555) and recycled content (EN4557) of energy-related products have already been published, but are purely based on a mass-based approach. Material-specific aspects such as the environmental impact or the financial, technical and redesign efforts required by manufacturers to facilitate the recycling and use of recycled materials are not addressed. Materials that are easy to recycle and use and have a high recycled content, such as copper, are treated in the same way as plastics that are difficult to recycle and use in products as post-consumer recycled plastics. Here we show how both colonisation and stress resistance are linked to the properties of the fungal cell surface. By deleting genes involved in the synthesis of melanin and carotenoid pigments of the model rock-inhabiting fungus Knufia petricola via CRISPR-Cas and comparing the behaviour of the gene-deletion mutants with the wild type (WT), we studied the role of these genes in mineral colonisation and stress sensitivity. The extracellular polymeric substances (EPS) of biofilms of the WT and mutants were extracted, quantified and chemically characterised. We observed that the absence of melanin affected the quantity and composition of the produced EPS: melanin-deficient mutants synthesised more EPS containing fewer pullulan-related glycosidic linkages. Moreover, in mineral dissolution experiments, these mutants showed a lower ability to colonise the mineral olivine. We hypothesise that not melanin, but the pullulan-related linkages enabled the melanin-producing strains to attach more strongly to the mineral. Moreover, ICP-OES analysis of the aqueous mineral-derived solutes showed that biofilms of the K. petricola mutants which could attach were able to dissolve the olivine faster than those that could not. The same mutants were also characterised by their sensitivity to desiccation stress: only the mutant deficient in both melanin and carotenoid synthesis was more sensitive to desiccation compared to the WT, indicating that a combination of both pigments is critical to withstand desiccation. Overall, these results show the critical role of the cell surface in the specific capacities of rock-inhabiting fungi. The genomes of black [dihydroxynaphthalene (DHN) melanin-containing] fungi from phyllosphere and exposed solid surfaces contain multiple photoreceptors (PRs). The plant pathogen Botrytis cinerea (Leotiomycetes) has a highly sophisticated photosensory and signalling system that helps to avoid light and to locate susceptible hosts. Rock-inhabiting Dothideomycetes and Eurotiomycetes including Knufia petricola possess equal numbers of PRs along with the same set of protective pigments. T1 - Generation of Cryomyces antarcticus mutants to explore the importance of DHN melanin for survival N2 - Cryomyces antarcticus, a cryptoendolithic melanized fungus endemic to Antarctica (phylum Ascomycota, class Dothideomycetes incertae sedis), has demonstrated high capability to survive extreme environmental conditions like those found in space (e.g., ionizing radiation, vacuum, microgravity), thus fueling fundamental astrobiological questions like “the search for life beyond Earth”. Its extraordinary resilience has been attributed to the presence of thick, highly melanized cell walls, which may contain both DHN and DOPA melanins. To better understand the contribution of DHN melanin to the overall resilience of C. antarcticus, we decided to generate melanin-deficient mutants by genetic engineering. For this, the melanin-PKS (polyketide synthase)- encoding ortholog was identified in the C. antarcticus CBS 116301 genome and used to design primers for re-sequencing of the capks1 locus in the strain CCFEE 515. Based on the genetic toolkit developed for the black fungus Knufia petricola, we designed a strategy for mutating capks1. Protoplasts of C. antarcticus CCFEE 515 were generated and co-transformed with a circular AMA-containing plasmid for expression and in-vivo assembly of Cas9 and two capks1-specific sgRNAs and a PCR-generated donor DNA i.e., a hygromycin resistance cassette flanked by ~75-bp-long sequences homologous to the capks1 locus. Transformation of C. antarcticus is challenging because of its very slow growth and uncertain gene copy number. It took six months from obtaining enough biomass for cell wall lysis to transferring the putatively resistant transformants for genotyping, but we eventually managed to generate three independent non-melanized (whitish) Δcapks1 mutants! We are now awaiting first complemented strains as ultimate genetic transformation control. They are considered to have a wild-type-like pigmentation due to the reintroduction of the wild-type capks1 into the Δcapks1 mutant. There are two possibilities to explore a genome and its gene functions. The hypothesis-based method proves the function of already existing gene/allele candidates by targeted mutagenesis - so called reverse genetics. The basis of forward genetics approaches is the random mutagenesis of the genome, followed by screening of obtained mutants for the phenotype of interest, and identification of the mutated genes in the respective mutants. This strategy is hypothesis-generating, means it is necessary to verify the relationship between the detected mutations and the observed phenotype by targeted mutagenesis of the identified gene. We developed a toolbox for editing the genome of the rock inhabitant Knufia petricola [Eurotiomycetes, Chaetothyriales] that allows the study of the phenotypic characteristics of black fungi such as the regulation of pigment synthesis, general stress responses, oligotrophy, and the unusual modes of cell division by advanced reverse and forward genetics approaches. The toolbox includes the annotated genome sequence of strain A95, efficient strategies for CRISPR/Cas9-based genome editing and live-cell imaging using genetically encoded fluorescent proteins, as well as protocols for -omics approaches and for simulation of mineral weathering. A forward genetics approach using transposon mutagenesis is currently developed for identifying essential genes. The established protocols and knowledge gained from K. petricola form a starting point for making other fungi from extreme environments accessible to genetic manipulation. They dominate – often together with bacteria and algae in sub-aerial biofilms – a range of hostile environments including natural and man-made ones, from salterns to dishwashers, roofs and solar panels. Because of lacking genetic tools and the slow growth of most isolates, the genetic bases for these specific properties are largely unknown. The rock-inhabiting fungus Knufia petricola (Eurotiomycetes, Chaetothyriales) exhibits all characteristics of microcolonial black fungi and was selected as recipient for genetic engineering to study gene functions and genetic interactions. Different variants of green and red fluorescent proteins were successfully expressed indicating that fluorescence microscopy using genetically encoded fluorescent proteins and fluorescent dyes enables various cell biology approaches. Furthermore, genes of biosynthetic pathways (DHN melanin, carotenoids, uracil, adenine) were successfully mutated by applying traditional gene replacement and plasmid-based or ribonucleoprotein (RNP)-based CRISPR/Cas9 or silenced by RNA interference (RNAi). Momentan regelt sie primär die Energieeffizienz von energieverbrauchsrelevanten Produkten, doch der Fokus wird aktuell um Anforderungen an den nachhaltigen Einsatz von Ressourcen erweitert. Bei den gegenwärtigen Aktivitäten im Ökodesign steht der Normungsauftrag M/543 im Zentrum. Es wurde 2015 von der Europäischen Kommission an CEN/CENELEC vergeben und später stieß auch ETSI dazu. Bislang sind acht generische EN-Normen und zwei Fachberichte (Technical Report (TR)) geplant. Sie sollen grundlegende Aspekte der Nachhaltigkeit von energieverbrauchsrelevanten Produkten, die von der Ökodesign-Richtlinie betroffen sind, abdecken. This test standard was developed based on the mandate M/366 of the European commission and is a horizontal reference method for the determination of volatile organic compounds (VOC) from different classes of construction (building) products. Specific test conditions are to be selected by the product TCs (technical committees) in a way that a product is tested under its intended condition of use. The test is based on the use of emission test chambers which are operated at constant air change rate and climate (23°C, 50 % r.h.) over 28 days. The standard defines the conditions and requirements for the measurement including loading factor, air change rate, sampling, analysis and calculation of emission rates of the substances. A 30 m³ reference room is described which is used to calculate air concentrations from the determined emission rates. The standard EN 16516 enables the evaluation of construction products regarding their emissions into indoor air under defined and comparable conditions. Jedoch sind diese Studien häufig nicht untereinander vergleichbar, da verschiedene Konzepte der Probennahme, der Aufbereitung und unterschiedliche Detektionsverfahren verwendet werden. Um diesen Mangel an vergleichbaren und aussagekräftigen Daten zu beheben, wird aktuell versucht, eine Harmonisierung der Mikroplastikanalytik voranzutreiben. Neue, schnellere Methoden zur MP Quantifizierung wurden entwickelt. Gleichzeitig werden neue Strategien zur Probenahme und Probenvorbereitung konzipiert, da hier die Fehlerquellen besonders große Auswirkungen auf das Ergebnis haben. Fungi actively sense the environmental light conditions to induce protective mechanisms against the light-associated stresses and to regulate development. Since fungi adapted to habitats with different light regimes, the complexities of their ‘visual’ systems and photoresponses may vary significantly. Fungi that are associated with plants experience a special light regime because the host seeks optimum light conditions for photosynthesis – and the fungus must cope with them. Moreover, fungi living under the canopy are confronted with an altered spectrum enriched for green and far-red light. Plants sense light to coordinate growth and morphogenesis: a low red:far-red ratio indicates the presence of competitors (shading by other plants) and triggers the shade avoidance response which is accompanied by increased susceptibility to nectrophic pathogens. Botrytis cinerea, the causal agent of gray mold diseases on many plant species in moderate climate zones (high humidity, temperatures around 20°C, seasons), exhibits striking photoresponses accompanied by a broad action spectrum and a high number of photoreceptors. Light is the most important factor controlling morphogenesis: it induces conidiation for disease spreading (summer cycle) and represses sclerotial development for survival and/or sexual recombination (winter cycle). In the past years, we identified several cellular components involved in photoperception and regulation of photomorphogenesis and virulence indicating that the same signaling pathways are of relevance for both processes, propagation and infection. Notably, we recognized the role of the phytochromes (red/far-red light sensors) for coordinated responses to light and elevated temperatures (photo-/thermomorphogenesis) – similar to the function of Arabidopsis PhyB. BcPHY2-like thermosensors are likely restricted to the Leotiomycetes, which include several plant pathogens of the moderate climate zones. The plastic fragments are home to a diverse community of microorganisms, and while researchers have gained a better understanding of these marine plastic biofilms, we lack insights into the physiology and genomic potential of the bacteria that colonize them. Examining the lifestyle of plastic colonizers from the GPGP is particularly intriguing, as the high concentration of plastics in this accumulation zone might allow for a microbial adaptation to this unique man-made habitat. A range of pigmentation was observed in 67 isolated strains obtained directly from plastic pieces sampled from the GPGP surface water. Sequence comparison between the cultivated bacterial strains and the 16S rRNA gene amplicon dataset confirmed that most of the cultivates could also be captured through DNA-only methods. Whole genome analysis of four taxonomically diverse representatives revealed multiple carotenoid pathways, including those to produce less common glycosylated carotenoids, like sarcinaxanthin glucoside. Further, we identified a potentially new Rhodobacteraceae species containing a photosynthetic gene cluster (PGC). Absorption analysis confirmed the actual production of the carotenoids and bacteriochlorophyll a. Floating plastics represent a habitat with strong UV-light exposure, making the protection with antioxidant carotenoids as well as the ability to use light as an energy source highly beneficial traits for plastic colonizers. Our findings indicate, that the production of pigments is a common adaption mechanism for plastic-associated bacteria, and that plastic biofilms present a so far overlooked source of rare carotenoids and light-harvesting mechanisms.
