Many works of art are UV- and light-sensitive. It is well known that especially UV radiation causes damage and discoloration in paintings, textiles, plastics, wood and other materials. In particular, the wavelengths between 280 and 410 nm cause color changes, embrittlement or destruction of e.g. plastics over time. Therefore, strongly UV-absorbing glasses are advantageous for architecture and are necessary for the protection of cultural assets. As restorers in all disciplines become more and more aware, the demands placed on buildings and their furnishings in terms of climate are becoming increasingly detailed and precise. The aim in each case is to preserve the valuable artwork. For some years now, the industry has been offering the protection of cultural objects by installing special UV-protective glasses. Currently, UV protection for church buildings is realized by laminated safety glass equipped with appropriate UV-protective plastic films. Technically, this always means a second pane of glass in front of the windows, which is installed outside the building. This creates climatic gaps that are difficult to control and deterioration due to ageing effects can be expected. At the same time, this protective glazing is not invisible and has a considerable aesthetic influence on the interior and exterior appearance of the building. Meanwhile, the preservation of historical monuments accepts such aesthetic cuts on buildings in order to protect the artwork in the interior from UV light. To this day, however, the long-term durability of UV protection provided by inserted plastic films is still controversial. To date the only available alternative on the market is a mouth-blown UV protection glass which uses a so-called overlay to provide UV protection. This shows that UV protection can also be achieved by glasses without plastic films thus realizing an exclusive inorganic protection which normally is more stable than a polymeric one. So far there are not enough studies to prove long-term durability. The aim of this project is to provide existing glazing or new glazing to be created with a highly transparent layer that ensures this UV-protective filter function below 400 nm. The glass coating is to be applied to the glass over a large area and fired into the surface like a classic ceramic enamel paint with the same technics. In addition, it should be long-term durable in its function. It means, that the glass has to be fused at temperatures below 630 °C during the firing process, its chemical durability has to be high, its coefficient of thermal expansion has to be as close as the one of the substrate (usually float glass) and the glass has to absorb the UV-radiation within a thin thickness (thinner as 100 µm). In this project, the long-term durability of commercial UV-protective glasses is examined. New low melting glasses containing UV-absorbing ions are being developed. Their UV-absorption as a thin layer is analyzed as well as their chemical durability and their thermal properties. Korrosion ist der Prozess, der von elektrochemischen Reaktionen an der Grenzfläche Metall/Patina kontrolliert wird. Runoff dagegen wird durch chemische Reaktionen an der Grenzfläche Patina/Atmosphäre begünstigt. Mit zunehmender Bewitterungszeit nimmt die Korrosionsgeschwindigkeit stetig ab, während postuliert wird, dass die Geschwindigkeit des Runoffs über die gesamte Zeit konstant bleibt. Dieses Verhalten des Runoffs muss aber zunehmend bezweifelt werden. Die Gesamtmenge an abgezehrtem Metall beinhaltet zwei Bestandteile. Ein Teil wird in die Schicht aus Korrosionsprodukten eingebaut, der andere wird aufgrund von Niederschlägen aus der Patina gelöst. Aus diesem Grund ist die Menge an Zink-Runoff stark abhängig von den Eigenschaften der Korrosionsprodukt- bzw. Patinaschicht und nicht anhand der Zusammensetzung des Metalls abschätzbar. Vorhandene Modelle können die Menge an Runoff, und damit verbunden die Abzehrung einer Schicht aus Zink und Kupfer, sowie den resultierenden Eintrag von Schwermetallionen in die Umwelt nicht mehr darstellen und erklären. Um den Korrosionsschutz an Bauwerken zum einen dauerhaft unter Beachtung von Kostenaspekten und Ressourcenschonung und zum anderen unter ökologischen Gesichtspunkten auslegen zu können, müssen deshalb neue wissenschaftliche Grundlagen erarbeitet und die Mechanismen beschrieben werden. Schwerpunkte waren die Bestimmung der Korrosivität der Atmosphäre von 3 ausgewählten Straßentunneln, die Analyse korrosionsrelevanter Ablagerungen und von Klimadaten. Die Auswirkungen dieser Einflussgrößen auf die Dauerhaftigkeit von Konstruktionen aus nichtrostenden Stählen wurde ermittelt und in Abhängigkeit verschiedener Einflussparameter dargestellt. Die Ergebnisse der systematischen Untersuchungen an Prüfköpern decken sich mit Beobachtungen an realen Bauteilen. Spaltbehaftete Konstruktionen aus Werkstoffen der CRC III führen demnach in Zeiträumen zwischen 10 und 15 Jahren zu Korrosionserscheinungen, welche die technische Sicherheit beeinträchtigen können und die erwartete Dauerhaftigkeit nicht erfüllen. Die Folge sind zusätzliche Sperrungen und ein vorzeitig notwendiger Unterhaltsaufwand, die erhebliche Kosten verursachen. These surfaces are a natural habitat of subaerial rock biofilms (SAB) that are an important stage of primary succession. As their metabolic processes have a great impact on the composition of the near-surface layers of the earth biofilms on natural and manmade hard substrates are a research focus of geomicrobiology. In technical systems immobilized algae biofilms are already used successfully for wastewater cleaning. Algae can eliminate bacteria from the water and at the same time increase the oxygen concentration and the pH value in the water through photosynthesis, which again allows phosphate elimination to take place. Nitrificants contained in the algae biofilm convert ammonium to nitrite and nitrate. Mineral claddings for building facades could be engineered to sustain either natural or previously established carefully selected algae-dominated biofilms and this way help to clean the air. For a targeted engineering of the mineral substrates and the biofilms attached to them basic knowledge is however missing. By systematic research in an interdisciplinary team of microbiologists and building material scientists the following questions shall be clarified • How can a curated biofilm be stabilized on the surface and designed to be durable without compromising the substrate • What exactly do the biofilms need in terms of moisture in their respective growth stages? Or how long water must be available in what form and quantity to enable growth. • What is the influence of the substrate on the availability and quality of the (rain) water (water quality, leaching of substances (alkalis) from the building materials, leaching of aerosol components, surface tension, suction tension, thickness of the water molecule layers adsorbed in the pore spaces....) There are several research groups working on bioreceptive building materials which emphasizes the general interest in the topic. The idea of speeding-up the process by the targeted establishment of biofilms on building façade panels is a unique approach that has so far only been researched at BAM. Architects are very interested in biofilm facades as a new co-designing method of humans and nature reflecting the changing awareness for the loss of biodiversity in cities. Die Akkumulation der nicht-reversiblen Effekte über die Zeit und allen Eigenschaftsänderungen entspricht der Alterung des Materials für den jeweiligen Expositionszeitraum. Die digitale Umweltsimulation gliedert sich hier in drei Teilaspekte. Ein erster, rein materialwissenschaftlicher Teil hat die Aufgabe, alle relevanten Materialempfindlichkeiten experimentell zu quantifizieren. Ein zweiter – unter Umständen numerischer Teil – hat die Aufgabe, das Bauteil und seine Einbaulage zu digitalisieren und aus den makroskopischen Umgebungsbedingungen die relevanten mikroklimatischen Umweltparameter für alle Oberflächen- oder Volumenelemente zu bestimmen. In einem dritten Teil werden die Einwirkungen über den betrachteten Zeitraum berechnet und kumuliert. Dieses Konzept, das auf den so genannten Expositions-Reaktions-Funktionen (ERF) basiert, wird an Beispielen der Photoxidation erläutert. Dieses Vorgehen wurde schon beim ViPQuali-Projekt als Numerische Umweltsimulation umgesetzt. Überprüft werden muss das Modell unbedingt an einer realitätsnahen Validierungsbeanspruchung. Herein, high resolution continuum source molecular adsorption spectrometry (HR-CS-GFMAS) is employed for fast and sensitive measurements. An environmental study was carried out on the River Spree. Samples were collected over the course of one year to allow a more detailed look into seasonal changes. Following this, a range of options are being investigated with a view to removing PFAS from the environment regarding water samples. Potential adsorber materials are explored based on Graphene (in cooperation with the Haag-Group) and COFs (in cooperation with the Emmerling-Group). Initial experiments were performed, and the preliminary results are presented. Furthermore, experiments for PFAS adsorption are being improved and PFAS specific adsorption will be analyzed. Sum Parameter approaches have the chance to identify Hot-Spots in the environment and to cover a broad range of PFAS in a single measurement. To monitor and regulate exposure to microplastics in the environment and in food for human health, it is crucial to ensure their comparability and to validate the analytical methods used in microplastics studies. Monitoring microplastics requires complex approaches for accurate determination otherwise it can lead to an over- or underestimation in environmental and food-relevant matrices. Moreover, to support the need of reliable measurement required by the European legislation directive on drinking water (EU 2020/2184) or the revision of the urban waste water directive, which will be finalised in 2024 the production and validation of polymeric reference materials with environmentally relevant concentrations, irregular shapes, defined aging status and different sizes is essential. The production of reference material with acceptable uncertainties in homogeneity need to be balanced, considering the low microplastics concentrations detected in the environment and in food. The aim of the project PLASTICTRACE and of this study, is to propose a “easy-to-use PET tablet ” (PET particle size mainly 1-100 µm) for routine applications as a candidate general reference material for detection methods. Batches of water-soluble matrix containing different concentrations of PET powder were produced. Results of imaging and counting methods (SEM, Laser diffraction), including production´s challenges; together with thermo-analytical methods (TED-GC/MS and Py-GC/MS) and vibrational spectroscopy (µ-Raman and µ-FTIR), will be presented. These data are useful to assess homogeneity and stability of the particles over time according to ISO 33405:2024. In addition, we will show some insights on the application of the PET-tablets in complex matrices, as milk powder and suspended solid from surface water, that could be easily used for routine microplastics analysis. In a laboratory test, transparent HDPE jerrycans have been exposed to both UV radiation and 55 wt-% nitric acid solution at (41 ± 2)°C, for up to 20 days. For comparison, UV radiant exposure (21 days) and nitric acid exposure (up to 6 weeks) were performed separately, at nearly equal temperatures. The respective damages are compared with FTIR spectroscopy in ATR and HT-gel permeation chromatography (GPC) on a molecular level and with hydraulic internal pressure testing as a component test. For the used jerrycans, relevant oxidation can only be found after the combined exposure. The gradual increase in oxidative damage shows the good reproducibility of the lab exposure. The decomposition of nitric acid into nitrous gases by UV radiation – as well as the jerrycan oxidation – is also observed at lower HNO3 concentration (28 wt- %). Similar results are obtained after outdoor tests. Again, the damage occurs only after combined exposure, in contrast to the exposures to UV only and to HNO3 only, which were conducted in parallel. Outdoor exposures are most readily accepted as they represent possible end-use conditions. However, the reproducibility of these exposures is poor due to the large temporal variations in weather. There are also several safety risks, which is why the number of replicates remains limited. Since the outdoor and lab exposure tests show the same qualitative results, it is appropriate to conduct systematic studies in the laboratory. After 6 days of lab exposure, the oxidation damage is rated as critical, which corresponds to about 1/10 year in Central Europe, according to the UV radiant exposure. This contribution shows how an improvement in the measurement data situation can improve the significance and comparability of outdoor exposures. Photooxidative ageing is calculated using sample polymers for which the corresponding modelling data (Exposure Response Functions, ERF) is available. The mean square deviations are estimated, which result from using different spectral irradiances and different spectral sensitivities, both of the sensors and the polymers. It will be shown here how the significance of the irradiance values increases with increasing approximation of the spectral sensitivities of the sensor and polymer. Also, the mean square deviations are estimated, which result from using different activation energies and surface temperatures. A selection of different variables to describe the weathering exposure is created. The potential of either individual values or data pairs is evaluated in terms of significance or correlation to the calculated photooxidative ageing. Recording hourly data also enables subsequent evaluations of individual test specimens, adapted to the respective colour or to determine activation energies. An accumulated value, calculated from PIT-radiometer irradiance and a suited surface temperature, can provide a comparison of different outdoor exposures over different locations or years. Thus, a characterisation of the weathering exposure would be related to a reference spectral sensitivity (much closer to a polymer than that of a common UV radiometer), one reference activation energy, and different surface temperature scenarios with, on the one hand, graded solar absorptions (white / grey / black) and, on the other hand, different thermal couplings (coated metal / thick plastic). This results in three values each for plastic samples and coated metals (light / grey / dark). Here, the stabilizing extracellular biofilm matrix together with physiological changes on the single cell level leads to an increased resilience towards harsh environmental conditions, antimicrobials, the host immune response and established cleaning procedures. Persistent microbial adhesion on e.g., medical implants, in water supply networks or food-processing industry is often associated with chronic inflammation, nosocomial and foodborne infections, enhanced biofouling and product contamination. To prevent persistent microbial colonization, antibacterial surface strategies often target the initial steps of biofilm formation and impede adhesion of single cells before a mature biofilm is being formed. While chemical coatings have been widely used, their restricted biocompatibility for eukaryotic cells and attenuated antibacterial-effects due to compound release limit their areas of application and alternative strategies focus on modified surfaces topographies to impede bacterial adhesion. In this work, we used ns-UV laser treatment (wavelength 248 nm and a pulse duration of 20 ns) to generate laser-induced periodic surface structures (LIPSS) with different submicrometric periods ranging from ~210 to ~610 nm on commercial poly(ethylene terephthalate) (PET) foils. Following structurally and chemically analyses, PET samples were subjected to bacterial colonization studies with Escherichia coli TG1, a bacterial test strain with a strong biofilm formation capacity due to the formation of nanofiber-like cell-appendages (pili). Bacterial adhesion tests revealed that E. coli repellence decisively depends on the spatial periods of the LIPSS with the strongest reduction (~91%) in cell adhesion observed for LIPSS periods of 214 nm. Scanning electron microscopy and additional biofilm studies using a pili-deficient E. coli TG1 strain revealed the importance of extracellular appendages in the bacterial repellence observed here, thus, pointing out new antibiotics-free strategies for antibacterial surfaces by impeding nanofiber-mediated bacterial adhesion. The large body of available data suggests that larger microplastics (MPs) in the natural environment are irregular shaped, partially degraded (chemically and physically), a continuum of sizes, shapes and densities, represent a wide range of polymer types, and acts as a reservoir for thousands of plastic chemicals. Although NPs have been described as being neither MPs nor engineered nanoparticles [2], and while direct measurement data of true NP in the environment remain elusive, they are expected to be as equally diverse as MPs [3]. Here, we present a strategy towards the development of a potential NP reference material based on nano-sized polyproplyene particles (nanoPP) developed within the EURAMET-funded PlasticTrace project (https://plastictrace.eu/). The crucial point is not only the reduction of microplastics pollution but also the introduction of legally binding, standardized procedures for their analysis. The limited availability of certified reference material as well as absence of standard operating procedures to identify, characterize and quantify microplastics according to number and mass have generated lack of comparability between different analytical methods and laboratories; but also, the need of time-consuming development of own protocols for each laboratory. Further on, harmonisation and standardisation of microplastics analysis requires specific reference materials that are used to validate the measurement and to ensure the comparability of results. In the first step, we developed tablets of a water-soluble matrix, containing defined numbers of microplastic particles or mass contents. These can be solved in water and can be handled as reference materials in bottled drinking water without additional sample preparation. These samples can be measured, using mass-based methods as thermo-analytical methods like thermal desorption-gas chromatography-mass spectrometry (TED-GC/MS) or pyrolysis–gas chromatography–mass spectrometry (Py-GC/MS) and using umber-based methods as vibrational spectroscopy like Infrared and Raman directly after filtration. The aim of this work was to delevop standard operating procedure to measure microplastics in more complex matrices and to harmonise and compare different detection and characterization methods. To bridge this gap, the EU-funded PlasticTrace project has developed water-soluble tablets containing precisely defined amounts of polyethylene terephthalate (PET) particles. Each tablet includes a specific mass (0.2, 18.5, or 400 µg) and particle count (151 or 1764, measured via Raman spectroscopy). Production follows a rigorous solid-phase dilution process, ensuring high reproducibility and quality control. The PET particles used in these tablets have an average size of approximately 40 µm, with a range between 10 and 200 µm, and an irregular, stone-like shape. All measurements align with ISO/TC 147/SC 2 standardization efforts and adhere to ISO/DIS 16094-2 and ISO/DIS 16094-3. To assess tablet homogeneity, the most widely accepted microplastics detection techniques, i.e. µ-FTIR, µ-Raman, Py-GC/MS, and TED-GC/MS, were applied. Different batches were produced, each containing varying particle numbers and masses, reaching the detection limits of the methods used. The results demonstrated excellent consistency between tablets, successfully passing homogeneity tests. Furthermore, no significant changes were observed in the tablets' composition or stability after six months of storage. This innovative reference material represents an important advancement in microplastics analysis, supporting laboratories in meeting EU Commission requirements for the monitoring of microplastics in drinking water and wastewater with greater accuracy and reliability. They can be identified all around our environment, even in air, soil and water. One of the major global MNPs polluters is the textile industry, which generates plastic particles (nylon, polyester, polyurethane, polyolefin, acrylic etc.) during the manufacturing processes. The study analyses the level of particle concentration outside a company that processes textile yarns, but also from the proximity of workplaces using an online air samplers with cascade and mono filter holders for silicon (Si) and quartz filters, respectively. A laser detector was used for real- time counting of particles of six size classes (PM0.3-PM10). Silicon (Si) filters with a diameter of 9 mm and pore sizes of 10 µm and 1 µm and quartz filters with a diameter of 25 mm and a pore size of 1.2 µm were used for particle sampling. A special holder was designed and made for selective sampling (PM10 and PM1.0) on Si filters. Quartz filters were pre-calcined (8000C) before collection. Sampling parameters: airflow (GilAir Plus pump) -2 l/m3, collection time -3 hours; starting time 7.00 am (T1)/2.00 pm (T2), intermittent natural ventilation, average temperature and humidity: 23.3 0C, RH: 52.5%. The weight of particles collected (µg) on the quart filters was 4.9E-05 (T1) and 5.1E-05 (T2) and on the Si filters 3,2 E-05(T1) and 5E-06(T2). The total number of the outside particles (PM0,3-PM10) was situated between 3584,58 (T1)-15165,05(T2) and inside 9901(T1) and 12921,61(T2). The highest number of PM 10 particles inside was 72/m3 and of PM0.3 particles 71979/m3. The samples were characterized using electron microscopy and µRaman spectroscopy. All samples showed a high concentration of particles on the filter. Samples show only particles with irregular shapes. The equivalent particle diameter was for PM10: 1-5 µm and PM1<1 µm. The total Ion Chromatogram shows the presence of PET-type polymer and the TED GS-MS analysis shows that the weight of the collected particles is PET. After extracting the particles from the quartz filters, they were tested for evaluation of the cytotoxic potential of preparations on human monocyte-macrophage cell cultures and peripheral lymphocytes. In the first case, the level of cytotoxicity was at the border between non-cytotoxic and cytotoxic and in the second test the result was moderate cytotoxic. Soluble inks are based on plant or insect dyes forming a water solution. In the early European Middle Ages brown plant inks were usually produced from the blackthorn bark and wine. In contrast to these soluble brown inks, one of the oldest black writing materials was produced by mixing soot with gum arabic or animal glue dissolved in a small amount of water. From Pliny’s detailed account of the manufacture of various black carbon inks, we learn that producing pure soot of high quality was not an easy task in Antiquity. Iron gall inks are a borderline case between these two groups. They are produced from four basic ingredients: gallic acid, an iron-containing component, gum arabic as a binding media, and an aqueous medium such as wine, beer, or vinegar. By mixing gallic acid with soluble iron ions, a water-soluble ferrous gallate complex is formed. Exposure to oxygen leads to the formation of insoluble black ferric gallate pigment. Due to the variety of recipes and the natural origin of raw materials, there is a wide range of different components and impurities in writing inks. The Bundesanstalt für Materialforschung und -prüfung (BAM) together with the Centre for the Study of Manuscript Cultures in Hamburg (CSMC) has developed a protocol for ink analysis. It consists of a primary screening to determine the type of the ink and a subsequent in-depth analysis using several spectroscopic techniques (X-ray fluorescence analysis and vibrational spectroscopy). Die Kombination aus geringer Dachneigung und einem pH-Wert von 4,5 führt zu den höchsten Run-Off-Raten für Zink und auch Kupfer. Um dennoch die Ergebnisse einer Beanspruchung vergleichen zu können, ist eine aussagekräftige Charakterisierung der Freibewitterung wichtig. Traditionell werden Daten bekannter Einflussgrößen wie Globalstrahlung, Temperatur oder Feuchtewerte herangezogen. Obwohl man weiß, dass diese Größen an den exponierten Oberflächen wechselwirken und kombiniert als Mikroklima Einfluss auf Degradationsprozesse nehmen, werden schon aus Praktikabilitätsgründen diese separat als klimatische Messgrößen angegeben. Die Aussagekraft dieser Daten ist mäßig. Ziel dieses Beitrags ist es, die Aussagekraft und damit Vergleichbarkeit zu erhöhen, indem diese Messdaten als Stundendaten zu Wirkintegralen vorverarbeitet und jahresweise akkumuliert werden. Betrachtet wird dafür der Prozess der Photooxidation, da dieser Alterungs-mechanismus bei allen Kunststoffen auftritt. Für eine einfache Modellierung wird die wirksame UV-Bestrahlung, gewichtet mit einer Arrhenius-abhängigen Ober¬flächen-temperatur (TOF), über den Beanspruchungszeitraum von hier je einem Jahr akkumuliert. Die Idee einer solchen Messgröße für photooxidative Ober¬flächen¬alterung wurde bereits vorgestellt [1]. Dort wurde ein Gerät vor¬geschlagen, mit dem dieser Wert für verschieden absorbierende Oberflächen für die Dauer einer Exposition akkumuliert wird, und ergänzend eine Abschätzung der Betauungs¬zeiten dieser Referenz-Oberflächen erfolgen könnte. Hier soll es um eine rechnerisch generierte Zusammenstellung dieser photo¬oxidativen 1-Jahres-Alterungswerte für verschiedene Standorte gehen. Ein Temperaturfaktor erfasst dabei den Einfluss der Strahlungserwärmung auf eine weiße, graue bzw. schwarze Referenzoberflächen, spiegelt also die Kopplung von solarer Strahlung und Temperatur wider [1]. Ermittelt wird er aus Stunden¬daten der Globalstrahlung und der Lufttemperatur. Die spektrale Wirkung wird aus Sonnenhöhenständen entsprechend der geografischen Breite mit Hilfe der FastRT-Oberfläche [2] als PiT·h-Wert abgeschätzt [3]. Anhand der errechneten Jahreswerte können die Schweregrade der photo¬oxidativen Beanspruchung an den verschiedenen Standorten unmittelbar verglichen werden. These hardy substrates accumulate rich polymeric matrices from their surroundings that in turn offer nutrition and protection to diverse communities and their theatre of activity, representing a new ecological niche. With our work, we aim to establish a holistic understanding of the interactions between microplastics and microorganisms within aquatic systems, and how these interactions shape their surrounding environment. In our research, we combine field sampling with in situ experiments, and apply high-throughput sequencing technologies to 16S genes and whole bacterial genomes. We demonstrate through our studies that biofilm communities exhibit no specificity to plastic polymer type but appear rather to be opportunistic colonizers more strongly shaped by spatial and temporal factors. While the polymer surface does not appear to elicit a selective effect on associated biofilms, our studies show that microplastics might shape communities in a different way. Based on our detection of carotenoid synthesis pathways and photosynthetic gene clusters among plastic biofilms, we postulate this new niche to shape a specific functional toolkit, adaptive among its members as a product of prolonged exposure to UV radiation at the sea’s surface. To fully assess the interactions between microplastics and microorganisms, however, one cannot consider plastics singularly but as a component within a more complex and interconnected system. It is known, for instance, that plastics can sorb significant concentrations of polycyclic aromatic hydrocarbons (PAH) in the environment. We show, in our work, that the PAH sorption patterns of microplastics correlate significantly with the structure of associated biofilm communities. Elevated concentrations of specific PAHs on microplastics coincided with the enrichment of selected taxa reportedly capable of hydrocarbon utilisation as well as a reduced diversity among associated biofilm communities. Here, we present a synthesis of our collective research on microplastic biofilms in the aquatic environment, the factors that shape them, and their interactions with nature. Genetic studies are hampered by slow growth, lacking sexual cycles and difficulties to transform these fungi. Recently, we implemented CRISPR/Cas9-based genetic tools enabling efficient targeted mutations of the rock inhabitant Knufia petricola (Eurotiomycetes/Chaetothyriales) for studying stress responses, the unusual types of cell division, mineral weathering and symbiotic interactions with phototrophic microbes (Voigt, Knabe et al. 2020, Sci Rep; Erdmann et al. 2022, Front Fungal Biol). To further explore the genome of this fungus as representative of the polyphyletic group of black fungi and to assign functions to yet unknown genes, a forward genetics approach is aimed. We chose the two-component Activator/ Dissociation (Ac/Ds) transposon system from maize for generating a collection of insertional mutants by in-vivo mutagenesis. For the optimal use of this genetic tool, an inducible promoter for the expression of the Ac transposase and by this the regulatable transposition of the resistance cassette-containing Ds transposon is requested. However, endogenous promoters for nitrate assimilation and galactose catabolism that are often used in fungi for regulatable gene expression are not inducible by their substrates in K. petricola suggesting that the regulatory networks for nutrient acquisition differ significantly in the oligotrophic black fungi. Therefore, the metabolism-independent Tet-on system was adopted and validated using a Ptet::gfp construct. No GFP fluorescence was detected in absence of the inducer doxycycline (DOX) while fluorescence intensities increased with increasing inducer concentrations. Melanin can adsorb metals like Fe. How this affects fungal uptake of Fe and deterioration of Fe-containing minerals and metals is however less known. To study this, we use the model fungi Knufia petricola A95, a rock-inhabiting fungus known to deteriorate minerals and have melanised cell walls, and Amorphotheca resinae, able to contaminate fuel tanks, secrete melanin and corrode metals. In K. petricola, we have deleted genes involved in melanin production and Fe uptake using CRISPR/Cas. Through comparison of the geochemical signatures of these gene deletion mutants with those of the wild type (WT), we explore the specific mineral/metal deterioration mechanisms of melanised fungi. Fe isotope signatures of the biomass of melanin- and Fe uptake-deficient mutants of K. petricola revealed that Fe adsorbed either directly onto melanin or after being reduced by Fe reductases. Importantly, once adsorbed to melanin, Fe could not be mobilised and taken up into the cell: both the WT and its melanin-deficient mutant, previously grown at Fe replete conditions, showed similar growth at Fe deficient conditions. Olivine dissolution experiments revealed that Fe oxidation inhibits dissolution. K. petricola was able to enhance dissolution when this inhibition is strongest (at pH 6) and prevented dissolution when this inhibition is weakest (at pH 4). The fungus therefore dissolves olivine by interacting with the oxidised Fe at the olivine surface. However, Fe uptake did not seem to be involved: mutants deficient in various Fe uptake mechanism dissolved olivine at the same rate as the WT. This indicates that Fe adsorption onto melanin might play a key role. This is also shown by K. petricola’s ability to enhance olivine dissolution even further if secreting a melanin precursor and A. resinae’s corrosion of carbon steel whilst secreting melanin. Combined, our results imply that the Fe adsorbed to melanin cannot be taken up but enables fungi to deteriorate Fe-containing substrates at a higher rate. Even more progress on assigning functions to yet unknown genes can be achieved by a forward genetics approach. We chose the two-component Activator/Dissociation (Ac/Ds) transposon system from maize for generating K. petricola insertional mutants by in-vivo mutagenesis. For the optimal use of this genetic tool, an inducible promoter i.e, from the metabolism-independent Tet-on system, was combined with the AcTPase-coding sequence enabling the regulatable transposition of the resistance cassette-containing Ds transposon. In total, six auxotrophic Ac/Ds starter strains containing the Ds transposon at different position of ade2, ura3 or ppt1 were generated. The cultivation of these strains with doxycycline for induction of TET::Ac and subsequent selection of cells on ADE/URA/LYS-lacking media resulted in prototrophic colonies (revertants) for most Ac/Ds strains. Amplicon sequencing of excision sites revealed characteristic footprint patterns, proving that the transposon jumped. For identifying unknown Ds re-insertions sites, the thermal asymmetric interlaced (TAIL)-PCR was successfully implemented. First identified Ds re-insertion sites suggest that the distribution pattern may depend on the excision site. Currently, transposition frequencies and genome-wide distribution of re-insertion sites are studied in different Ac/Ds starter strains to identify the best candidate for generating saturated mutant libraries. On e.g. medical implants, in water supply networks or food-processing industry, biofilms can be a fertile source of bacterial pathogens and are repeatedly associated with persisting, nosocomial and foodborne infections. Nowadays, the emergence of resistances because of extensive usage of antibiotics and biocides in medicine, agriculture and private households have become one of the most important medical challenges with considerable economic consequences. In addition, aggravated biofilm eradication and prolonged cell-surface interaction can lead to increased biodeterioration and undesired modification of industrial and medical surface materials. Various strategies are currently developed, tested, and improved to realize anti-bacterial surface properties through surface functionalization steps avoiding antibiotics. In this study, contact-less and aseptic large-area short or ultrashort laser processing is employed to generate different surface structures in the nanometer- to micrometer-scale on technical materials such as titanium-alloy and polyethylene terephthalate (PET). The laser processed surfaces were subjected to bacterial colonization studies with Escherichia coli test strains and analyzed with reflected-light and epi-fluorescence microscopy. Depending on the investigated surfaces, different bacterial adhesion patterns were found, ranging from bacterial-repellent to bacterial-attractant effects. The results suggest an influence of size, shape and cell appendages of the bacteria and – above all – the laser-processed nanostructure of the surface itself, emphasizing the potential of laser-processing as a versatile tool to control bacterial surface adhesion. We found that short-term exposure to sub-inhibitory levels of glutaraldehyde makes E. coli resistant to high doses of bactericidal antibiotics from different classes. This tolerance is associated with delayed, heterogeneous regrowth dynamics and global transcriptome remodeling. We identified over 1200 differentially expressed genes, including those related to antibiotic efflux, metabolic processes, and the cell envelope. The cells entered a disrupted state likely due to the unspecific mode-of-action of glutaraldehyde. Despite this unregulated response, we identified several differentially expressed genes not previously associated with antibiotic tolerance or persistence that induce antibiotic tolerance when overexpressed alone. These findings highlight how the unspecific mode-of-action of disinfectants can make bacteria temporarily resistant to antibiotics. They have implications for settings where disinfectants and antibiotics are used in close proximity, such as hospitals and animal husbandry, and for the selection dynamics of tolerant pheno- and genotypes in fluctuating environments where microorganisms are exposed to these substances, such as sewage systems. A trade-off arises from overcoming the disrupted state as quickly as possible and maintaining antibiotic tolerance. It is part of the RUNOFF-project, which studies and re-assesses the durability of roofing and facade materials under current atmospheric conditions in Germany. Studying building materials is important as they have a significant impact on achieving a variety of goals and targets within the sustainable developments goals (SDGs). The durability of materials is essential to reach sustainability. However, the durability of metallic materials is strongly depended on climate conditions, which have changed as a result of technical measures in industry, climate change and increasing urbanisation. In Germany at least, the data base is not up-to-date leading to prediction models regarding corrosion resistance and durability of metallic materials which can no longer be trusted and therefore need to be re-assessed and updated. Also, not only the demand for sustainable but also environmentally friendly building materials has increased dramatically. A number of construction materials produce chemical hazards, and therefore have negative impacts on water quality, soils health and ecosystems. To limit these impacts, environmental assessment methods and tools are needed to measure and quantify the inputs and outputs of building materials throughout their lifetime. The toxicity and transport dynamics of OTs depend on their specific chemical forms— i.e., the type and number of organic substituents—resulting in distinct toxicity profiles and varying affinities for environmental colloids. These colloids-species interactions collectively influence the mobility, bioavailability, and health impacts of OTs. To date, however, most studies addressed speciation and colloidal characterization separately; thus, the data on the combined determinations of the organometallic species in association with their carrier colloidal fractions remain largely elusive. Here, we present a comprehensive account of method development, application, and validation to quantitatively characterize the adsorption dynamics of 10 different OT species on natural colloidal particles (<500 nm). Our approach utilizes asymmetrical flow field-flow fractionation (AF4) coupled with inductively coupled plasma time-of-flight mass spectrometry (ICP-ToF-MS), achieving detection limits for Sn-equivalent concentrations as low as 6.0 ng/L. The method effectively separates free OT species from those bound to colloids, facilitates the fractionation of particles ranging from a few nm up to 500 nm, and enables the determination of fraction-specific OT interactions. This unique dataset offers comparative insights into the interactions of 10 OT species, representing a significant advancement in understanding species-colloid interactions. Our findings have important implications for assessing the distribution and mobility patterns of toxic organometallic species in surface waters, groundwater, sediments, and soils. Um darüber hinaus Schweißzeit, Schweißnahtvolumina und Schweißkosten zu minimieren, kam es zur Entwicklung moderner Lichtbogenprozesse mit erhöhter Abschmelzleistung, wie bspw. dem modifizierten Sprühlichtbogen. Das Schweißen mit modifiziertem Sprühlichtbogen bietet die Möglichkeit, schmalere Nahtfugen bzw. kleinere Nahtöffnungswinkel zu schweißen. Mit zunehmender Festigkeit der Baustähle steigen allerdings die Anforderungen an die schweißtechnische Verarbeitung. Hochfeste Feinkornbaustähle weisen ein erhöhtes Risiko gegenüber der wasserstoffunterstützten Kaltrissbildung auf. Diese Werkstofftrennungen entstehen unter gemeinsamer Wechselwirkung von lokalem risskritischem Gefüge, lokal erhöhter Wasserstoffkonzentration und lokal erhöhter Beanspruchung bzw. Dehnung. Untersuchungen haben gezeigt, dass der Einsatz des modifizierten Sprühlichtbogens in Verbindung mit schmalen Nahtfugen zu erhöhten Wasserstoffkonzentrationen im Schweißgut führt. Die vorliegende Arbeit gibt einen Aufschluss hinsichtlich der Kaltrissempfindlichkeit des vergüteten, hochfesten Feinkornbaustahls S960QL anhand der durchgeführten Kaltrissprüfung mittels selbstbeanspruchenden TEKKEN-Tests. Dazu wurden die Nahtöffnungswinkel der Prüfnähte zwischen 30° (Einsatz des modifizierten Sprühlichtbogens) und 60° (Einsatz des konventionellen Übergangslichtbogens) variiert. Ferner wurde die Wirksamkeit einer Nachwärmprozedur aus der Schweißwärme heraus zur Vermeidung der wasserstoffunterstützte Kaltrissbildung untersucht. Die Ergebnisse weisen in allen Proben (ohne Nachwärmung) Kerbrisse auf, welche als Kaltrisse identifiziert werden konnten. Durch eine Nachwärmung unmittelbar nach dem Schweißen können diese gänzlich vermieden werden. Das Schweißgut mit modifizierten Sprühlichtbogen weist die höchsten mittleren Wasserstoffkonzentrationen auf. How could behavior of junctions be included? To which extend is it necessary? On the conference Geo-Chicago 2016, Swan and Yuan presented an ultimate limit state equation for the short-term material resistance of junctions. We discuss limitations and suggest improvements to include long-term behavior of junctions. Nevertheless, the approach applies only to a specific mode of shear-tensile failure of junctions and only to strictly rigid geogrids. A new design concept presented in the same year by Jacobs on the conference GeoAmerica for the special case of the anchorage of geogrids, which reinforce capping system on long and steep slopes of landfills, tried to overcome these drawbacks. We discuss the aspects of this concept related to the long-term behavior of junctions and the interplay between the load applied to junctions and the flexibility of longitudinal ribs. Standard test specimens of steel, zinc, copper and aluminium were exposed to rural, city and marine atmosphere at test sites of the Bundesanstalt für Materialforschung und -prüfung (BAM). Test sites at city atmosphere comprised exposure on a rooftop, next to a highly frequented road and directly at a motorway. Exposure under marine conditions was carried out at Heligoland in the North Sea. The specimens were stored under sheltered and unsheltered conditions on each test site, respectively. Corrosion rates after the first year of field exposure were determined and used for the classification of the atmospheres and the test sites regarding the corrosivity category. Results after the first year of exposure were presented at the EUROCORR 2018 in Cracow. The current presentation will present an update of results of the systematic study after five years of exposure. The corrosion progress and the mass loss of the standard metals steel, zinc, copper and aluminium will be presented and discussed with respect to the atmospheric conditions and the time of wetness at the test sites. Nevertheless, the interaction of such contrast agents with tissue components is not yet fully understood. Typically, they go into extracellular space. The extracellular matrix (ECM) is a three-dimensional network of macromolecules providing structural and biochemical support of the surrounding cells in all mammalian tissues. It is composed of structural proteins (e.g., collagen, elastin) and proteoglycans, which consist of glycosaminoglycans (GAGs) covalently bound to a protein core. GAGs are long, linear polysaccharides composed of repeating disaccharide units that differ in molecular mass, disaccharide structure and degree of sulfation. Many diseases, including inflammation, fibrosis, and tumor invasion, are associated with characteristic ECM changes, especially at an early stage of disease development. As disease severity increases, the amount of one or more different GAG types in the ECM and the extent of GAG sulfation increases. Characteristic of GAGs is their ability to form complexes with cations, e.g., with lanthanides. Thus, GAGs could be a potential binding partner for GBCA molecules as a whole or for dechelated Gd. Currently, there are still many unanswered questions about the interaction of contrast agents with ECM components. In this study, therefore, the uptake and distribution of ionic Gd and various linear and macrocyclic GBCAs was investigated in spheroids as model systems mimicking the complex physiologically relevant tissue microenvironment. Chinese hamster ovary (CHO) cells and CRL-2242 cells, a CHO mutant that does not produce sulfated GAGs, were used to prepare spheroids. Afterwards the spheroids were incubated with gadolinium chloride and GBCAs. For elemental mapping laser ablation inductively coupled plasma time-of-flight mass spectrometry (LA-ICP-TOFMS) and synchrotron radiation nano X-ray fluorescence spectroscopy (syn-nanoXRF) were used. Although all spheroids were exposed to identical Gd concentrations, differences were observed in the spatial distribution and the amount of Gd taken up. After incubation with linear and macrocyclic GBCAs, Gd is detected in the interior of both types of spheroids. Furthermore, differences in the Gd amount were found depending on the GBCA used. In contrast, incubation with gadolinium chloride leads to an Gd enrichment in the outer regions of the spheroids as well as to much higher Gd contents compared to incubation with GBCAs. Both LA-ICP-TOFMS and syn-nanoXRF can make an important contribution to better understand the relationship between the affinity of GBCAs and ECM components. T1 - Inflammation-related key events stimulated by micro- and nanoplastics N2 - The presence of micro- and nanoplastic particles (MNP) in our environment has raised increasing public and political concern. Increasing evidence suggests that humans are exposed to these MNP, mostly via inhalation or ingestion. The EC-Horizon project POLYRISK (https://polyrisk.science) aims to establish an IATA/AOP-based tiered approach to assess human health risks of MNP. In our research, we focus on assessing immunotoxicological effects of MNP. We have tested a series of primary, secondary, environmentally aged (incubated in River Rine water) and chemically altered MNP for their potential to affect dendritic cells or macrophages. MNP used in this study include primary and weathered PS, secondary PVC, PA, PP (with or without talc) and PE. The secondary PE and PP (w/o talc) particles appeared to have oxidized groups on their surface. Data shows that MNP can be engulfed by macrophages (PMA-stimulated human THP1 cells) and human blood-derived dendritic cells (DCs). The effects of virgin as well as secondary MNP (0, 10 or 100 μg/ml) on THP1 macrophages were limited to decrease of mitochondrial activity (Alamar blue), increase of cellular leakage (LDH), and stimulation of lysosomal activity. None of these MNP stimulated gene expression of NFκB or release of pro-inflammatory cytokines (IL-6, TNFα, IL-1β). On the other hand, secondary PP and PE with oxidized surface groups did increase NFκB gene expression and release of cytokines by THP1 macrophages. Virgin and weathered PS particles were tested using DCs, and only weathered PS did stimulate DC activity (increased costimulatory molecules CD83, CD86) and as consequence allogeneic T cells. DC activation appeared to result from environmental contaminants. In conclusion, of all MNPs tested only those that contained active surface groups or environmental components appeared to be immunostimulatory, whereas primary and secondary MNP rather reduced macrophage activity and viability (at relatively high concentrations of 100 μg/ml). Further research is needed to reveal molecular mechanisms and to translate in vitro findings to real-world exposure scenarios. The challenges in analytics are very complex, e.g. representative sampling, non-destructive sample preparation with concentrated particles and homogeneous distribution and true detection. All together lead to lacks in harmonization and results, which are hardly comparable. On the other hand, monitoring of microplastics is mandatory in the future strictly regulated by the EU commission in the Drinking water and Wastewater Framework Directive. One step to accurate and precise results will be the development of suitable reference materials mimicking particles in the environment. BAM developed test materials, which are produced by mixing a small portion of microplastic particles with a water-soluble matrix. After solid phase dilution and homogenisation small portions are pressed into tablets and bottled in glass vials (Figure 1). These tablets are well characterized with particle size distribution and SEM images. Additionally, they are tested as reference material candidate according to homogeneity and stability for particle number with µ-IR and µ-Raman as well as on particle mass with Py-GC/MS and TED-GC/MS after ISO Guide 35. Results are promising. The material passed the homogeneity control. No changes are observed within 6 months of storage. The same tested reference material is finally used in sample preparation experiments, where environmental suspended particular matter from surface water or baby milk powders are spiked with the tablets. In addition to unique identification, this also includes reliable quantification. Two different methodological approaches are used for the quantification. With vibrational spectroscopic techniques such as µ-FTIR and µ-Raman, results are obtained in the form of particle number, size and shape. Instead, with thermal analytical technics as TED-GC/MS and Py-GC-MS the results are expressed as mass concentration.. Both concepts offer different information variables. In terms of routine monitoring, it is necessary to obtain a rapid sequence from sampling to the detection result. For this reason, this study focuses on TED-GC/MS and Py-GC-MS. Even though thermal analytical methods generally require less sample preparation, some matrices require it. This is usually done if the analyte concentration is too low or if strong matrix effects such as signal suppression and false positive signals occur. This talk presents advanced sample preparation for baby milk powder as food example and density separation for microplastic analysis in surface waters. A preparation protocol based on citric acid was selected for the milk to remove as much as possible of the matrix. The low pH value leads to a fast and effective protein precipitation and minimizes filter cake formation, making filtration possible, reduces the number and the amount of compounds in detection and hence, simplifies the evaluation. Microplastics in surface waters and sediments must be concentrated not only because of their low microplastics mass content but also in terms of homogeneity. For this purpose, a density separation was carried out using a concentrated sodium iodide solution, which led to a high reduction of the inorganic mineral matrix and made possible to obtain a representative subsample of initial masses of up to 80 g. However, due to a lack of standardized methods, MP analysis as well as its quality assurance, is still a challenge. The thermal extraction desorption-gas chromatography/-mass spectrometry (TED-GC/MS) as a novel innovative thermoanalytic method can detect MP particles in environmental samples based on their decomposition products. According to MP analysis, a distinctively larger polyethylene (PE) content in comparison to other synthetic polymers was detected in environmental samples. Various substances within the matrix which have a similar molecular structure to PE, like fats and oils, might interfere with its marker and lead to an overestimation. In this investigation an automated extraction method, the accelerated solvent extraction (ASE), is introduced as quality control tool before TED-GC/MS measurement. Thereby, not the extract like usually used, but the solid filtrate is analyzed. The aim is to clean the environmental samples from solvent removable compounds which could lead to false positive signals and thus receive a more accurate PE mass. Consequently, a sample mimicking environmental samples with a defined matrix and a known polymer mass was generated. The common polymers polyethylene, polypropylene, polystyrene and styrene-butadiene rubber, used as an indication for tire wear, were added to the matrix. Different parameters for ASE extraction were varied such as different solvents, filters and purge time. Finally, a PE recovery rate of 91 % after extraction could be reached.
