Bezüglich der Klimakrise wird eine grundlegende Änderung unseres Wirtschaftens als wesentlicher Lösungsweg gesehen. Die aktuell praktizierte lineare Wertschöpfung führte zu Ressourcenknappheit und -abhängigkeit und trug aktiv zur Verstärkung des Klimawandels bei. Wie wir in diesem Jahrzehnt unseren Absprung von gewohnten Produktions-, Konsum- und Verwertungsmustern gestalten, wird entscheidend für die nächsten Jahrzehnte sein. Die Frage, wie bis 2030 die Transformation hin zu einer zirkulären Wirtschaft gelingen kann, wurde u.a. von der Circular Economy Initiative Deutschland beantwortet. Sie nennt zehn Handlungsschwerpunkte, wobei „Standardisierung“ mit an wichtigster Stelle steht. Dieses beruht auf Gegenseitigkeit. Auf der diesjährigen G7 Standardisation High-level Conference wurde Circular Economy zu dem Themabereich mit dem größten Potential für Klimaschutzmaßnahmen gesehen. In 2022 haben DIN, DKE und VDI mit über 500 Fachexperten aus Wirtschaft, Wissenschaft und Politik an einer Normungsroadmap Circular Economy gearbeitet. Die BAM war in fünf von sieben Schwerpunktthemen beteiligt. Ziel der Roadmap ist es, einen Handlungsrahmen für die Normung und Standardisierung zu schaffen, der die Transformation hin zu einer Circular Economy fördert. Normen und Standards bilden den untergesetzlichen und technischen Rahmen, auf denen eine zirkuläre Wirtschaft aufwachsen kann. Sie definieren Schnittstellen und stellen eine klare Kommunikation zwischen den verschiedenen Phasen des zirkulären Kreislaufes sicher – sie machen einen Kreislauf erst möglich. In der Präsentation sind die Highlights der Normungsroadmap Circular Economy dargestellt. Herein, high resolution continuum source molecular adsorption spectrometry (HR-CS-GFMAS) is employed for fast and sensitive measurements. An environmental study was carried out on the River Spree. Samples were collected over the course of one year to allow a more detailed look into seasonal changes. Following this, a range of options are being investigated with a view to removing PFAS from the environment regarding water samples. Potential adsorber materials are explored based on Graphene (in cooperation with the Haag-Group) and COFs (in cooperation with the Emmerling-Group). Initial experiments were performed, and the preliminary results are presented. Furthermore, experiments for PFAS adsorption are being improved and PFAS specific adsorption will be analyzed. Sum Parameter approaches have the chance to identify Hot-Spots in the environment and to cover a broad range of PFAS in a single measurement. Due to their relevance in natural and industrial processes, the formation pathways of these calcium sulfate phases from aqueous solution have been the subject of intensive research1. The state-of-the-art of the calcium sulfate formation mechanisms builds upon and goes beyond what we have come to appreciate in the astounding intricacy of other mineral formation processes from ions in aqueous solutions. The original, and rather naive, 'textbook' image of these phenomena, stemming from the adaptation of classical nucleation and growth theories, has increased in complexity due to the discovery of a variety of precursor and intermediate species2. These include solute clusters (e.g. prenucleation clusters, PNCs), liquid(-like) phases, as well as amorphous and nanocrystalline solids etc.. In this context, a number of studies have already revealed that nucleation in the CaSO4-H2O system is non-classical, where the formation of the different crystalline phases involves several steps including a common amorphous precursor1, 3, 4. In this contribution we show that the formation of the amorphous phase involves the aggregation of small primary particles into larger disordered aggregates exhibiting a "brick-in-the-wall" structure5, 6. The actual crystallization occurs by the restructuring and coalescence of the particles ("bricks") into a given calcium sulfate phase depending on the physicochemical conditions of the solution. Importantly, the rearrangement process does not continue until a (nearly-)perfect homogeneous single crystal is obtained. Instead it comes to a stop, or at least significantly slows down. Such a process thus yields a final imperfect mesocrystal, composed of smaller domains rather than a continuous crystal structure, within which the domains are separated by an amorphous (i.e. less ordered) calcium sulfate phase. Hence, the non-classical crystallization process of CaSO4 yields a final imperfect mesocrystal with an overall morphology resembling that of a single crystal, yet composed of smaller nano-domains. Importantly, these observations reveal that organic-free calcium sulfate mesocrystals grown by a particle mediated-pathway preserve in the final crystal structure a “memory” or “imprint” of their non-classical nucleation process, something that has been overlooked until now. Furthermore, the nano-scale misalignment of the structural sub-units within these crystals can propagate through the length-scales, and be expressed macroscopically as misaligned zones/domains in large single crystals. Indeed, by considering large anhydrite crystals from the famous Naica Mine (“Cueva de los cristales”) we observed a suite of correlated self-similar void defects spanning multiple length-scales7. These flaws, in the macroscopic crystal, stem from “seeds of imperfection” originating from a particle-mediated nucleation pathway. Hence, building a crystal could be viewed as Nature stacking blocks in a game of Tetris, whilst slowly forgetting the games core concept and failing to fill rows completely. A second option is the utilization of a “pump and treat” process, cycling polluted soils through a washing plant leading to the concentration of the pollutants in the fine fraction. Both approaches are cost-intensive and not intended for the direct decomposition of all PFAS contaminants. Furthermore, the contaminated adsorbants have to be safely thermal treated for recovery. Hence, there is a great demand for innovative developments, dealing with new strategies of tackling the PFAS problem. As a result of continuous use of fluorinated consumer products, effluents and sewage sludge from wastewater treatment plants (WWTPs) have been shown to be an important source of PFAS contamination in the environment. While environmental exposure of organic pollutants like PFAS, pesticides and pharmaceuticals have to be avoided, phosphorus from sewage sludge must be used in Germany among other things to produce high-quality P-fertilizers for a circular economy. However, the fate of legacy and emerging PFAS compounds during sewage sludge incineration or pyrolysis is for the most parts still unknown. Using the ScanLag system, we found that exposure to GTA resulted in wide lag-time distributions across different bacterial isolates of E. coli, S. aureus, and P. aeruginosa. Importantly, this was associated with elevated levels of survival (i.e. tolerance) towards lethal doses of antibiotics. As revealed by RNAseq in E. coli, GTA exposure caused global transcriptome remodeling, with more than 1200 differentially expressed genes of diverse biological functions. Several of these genes that were not previously associated with antibiotic tolerance or persistence induced, when overexpressed alone, antibiotic tolerance without showing a lag phenotype. This suggests that exposure to GTA induces unspecific, lag-dependent and specific, lag-independent tolerance to antibiotics in clonal bacterial populations. These findings have implications for 1.) settings where disinfectants and antibiotics are used in close proximity, such as hospitals and animal husbandry, and 2.) for the selection dynamics of tolerant pheno- and genotypes in fluctuating environments because of the trade-off that arises from exiting lag and resuming growth as fast as possible and maintaining antibiotic tolerance. Sunlight-associated stresses are however multiple: high temperatures, UV radiation with associated DNA damage, accumulation of reactive oxygen species (ROS), desiccation and osmotic stresses. Ascomycota dominating light-flooded habitats accurately sense and respond to changes in light using it as a cue to coordinate growth, stress responses as well as to establish pathogenic or symbiotic relationships. Two species from two light-flooded habitats – phyllosphere and sun-exposed solid surfaces – were analysed for their photoreceptor distribution. In both habitats phototroph-associated and black [dihydroxynaphthalene (DHN) melanin-containing] fungi are prevalent. This diversity was sampled with the plant-associated fungus Botrytis cinerea (Leotiomycetes), while Knufia petricola (Eurotiomycetes) was included as a typical biofilm-former on sun-exposed solid surfaces e.g. rocks, building facades, roofs, and solar panels. The analysis has shown that genomes of black fungi contain more photoreceptors than animal pathogens and saprophytes such as Aspergillus nidulans and Neurospora crassa1,2. B. cinerea that causes the grey mould disease by infecting the above-ground parts of more than 200 dicots has a highly sophisticated photosensory and signalling system that helps to avoid light and to locate susceptible hosts1. Rock-inhabiting Dothideomycetes and Eurotiomycetes including Knufia petricola possess equal numbers of photoreceptors along with the same set of protective metabolites i.e. melanin, carotenoids and mycosporines2. This similarity between black fungi from plant and rock surfaces suggests that photoperception and -regulation are important for sun-stressed fungi that receive nutrients through cooperation with phototrophs. CRISPR/Cas9-based genetic tools for manipulating K. petricola were established3 and are currently used for elucidating the functions of the different photoreceptors in the biology of rock-inhabiting fungi. While combination effects have been extensively mapped for antibiotic-antibiotic combinations, the combination effects of antibiotics with antimicrobials used as biocides or antiseptics have not been systematically investigated. Here, we investigated the physiological and evolutionary consequences of combinations of antibiotics (meropenem, gentamicin and ciprofloxacin) and substances used as biocides or antiseptics (octenidine, benzalkonium chloride, cetrimonium bromide, chlorhexidine, povidone-iodine, silver) on growth and selection of Pseudomonas aeruginosa. We find prevalent physiological combination effects with synergy occurring 6 times and antagonism occurring 10 times. The effects are specific to the antibiotic-biocide combination with meropenem showing a tendency for antagonism with biocides (6 of 7), while gentamicin has a tendency for synergy (5 of 7). A particular strong antagonism is apparent for the meropenem-chlorhexidine combination, for which we conducted an in-depth study on the underlying molecular mechanism using RNASeq. Moreover, we find widespread effects of the biocide-antibiotic combinations on selection of P. aeruginosa strains resistant to the antibiotics, including cross-resistance and collateral sensitivity. In conclusion, antibiotics and biocides or antiseptics exert physiological and evolutionary combination effects on the pathogen P. aeruginosa. These effects have consequences for the efficacy of both types of substances and for the selection of antimicrobial resistant strains in clinical applications with combined exposure (e.g. wound care, coated biomaterials). The standard method to quantify PFCAs in water samples is liquid chromatography or, for additional quantification of ultrashort-chain PFCAs (carbon chain length of two or three carbon atoms) too, hydrophilic interaction liquid chromatography, in each case coupled with mass spectrometry. However, there are a lot of laboratories using MS coupled to headspace gas chromatography systems (HS-GC-MS), usually to quantify a wide range of volatile organic compounds. While PFCAs themselves are difficult to measure with HS-GC-MS in water samples, this challenge can be solved by derivatizing the acids to more volatile compounds, like esters. This approach could enable laboratories using HS-GC-MS to analyse and monitor PFCAs in environmental water samples. By following this idea, we successfully developed a HS-GC-MS method to quantify PFCAs in ultrapure water. Trifluoric acid (TFA), perfluoropropionic acid (PFPrA), perfluorobutanoic acid (PFBA) and perfluorooctanoic acid (PFOS) served as exemplary analytes of PFCAs, which were derivatized with methanol and concentrated sulfuric acid to form methyl esters. The method was optimized by varying different preparation and measurement parameters, leading to limits of detection and quantification in the two-digit ppt-range for all four analytes. The method was than tested on various real water samples, including tap and bottled water, groundwater, landfill leachate, treated wastewater and impinger water from a sewage sludge incineration experiment. Furthermore, snow samples from a ski slope were analysed too. TFA was the most frequently detected PFAA, with concentrations in the ppt and low ppb range. The detection and quantification of the remaining three PFCAs vary with the sample types and sampling location.Our results show that HS-GC-MS is suitable, and enables laboratories with this technique, to quantify PFCAs water samples. Even though their growth is generally unappreciated, prevention or inhibition thereof is still to be accomplished. We have chosen the genetically amenable, black fungus Knufia petricola as a model species to study these organisms. Our goal is simple: the quantitative description of its growth on various environmental conditions to create a mathematical model using the iDynoMiCS 2.0 platform which could allow the development of a mitigation strategy. The conditions to be tested are the quantity and type of carbon and nitrogen sources, the pH and the presence of neighbouring colonies. First results showed that colonies grown on agar have the highest extension rates at pH 5, 0.01M NO3 or NH4, and 0.1M glucose or sucrose. Interestingly, extension rates but also CO2 production was unaffected by an increase in the concentration of the C-source from 0.01M to 0.1M. On a microscale, we observed that medium containing more C resulted in a higher quantity of branches produced by a single cell and not in a higher single cell growth rate. This rapidly results in a more compact microcolony versus the more extended colonies on C-deficient media. We believe that such deeper knowledge on the growth of black fungi will help mitigate their material colonisation and deterioration. Here, the stabilizing extracellular biofilm matrix together with physiological changes on the single cell level leads to an increased resilience towards harsh environmental conditions, antimicrobials, the host immune response and established cleaning procedures. Persistent microbial adhesion on e.g., medical implants, in water supply networks or food-processing industry is often associated with chronic inflammation, nosocomial and foodborne infections, enhanced biofouling and product contamination. To prevent persistent microbial colonization, antibacterial surface strategies often target the initial steps of biofilm formation and impede adhesion of single cells before a mature biofilm is being formed. While chemical coatings have been widely used, their restricted biocompatibility for eukaryotic cells and attenuated antibacterial-effects due to compound release limit their areas of application and alternative strategies focus on modified surfaces topographies to impede bacterial adhesion. In this work, we used ns-UV laser treatment (wavelength 248 nm and a pulse duration of 20 ns) to generate laser-induced periodic surface structures (LIPSS) with different submicrometric periods ranging from ~210 to ~610 nm on commercial poly(ethylene terephthalate) (PET) foils. Following structurally and chemically analyses, PET samples were subjected to bacterial colonization studies with Escherichia coli TG1, a bacterial test strain with a strong biofilm formation capacity due to the formation of nanofiber-like cell-appendages (pili). Bacterial adhesion tests revealed that E. coli repellence decisively depends on the spatial periods of the LIPSS with the strongest reduction (~91%) in cell adhesion observed for LIPSS periods of 214 nm. Scanning electron microscopy and additional biofilm studies using a pili-deficient E. coli TG1 strain revealed the importance of extracellular appendages in the bacterial repellence observed here, thus, pointing out new antibiotics-free strategies for antibacterial surfaces by impeding nanofiber-mediated bacterial adhesion. The challenges in analytics are very complex, e.g. representative sampling, non-destructive sample preparation with concentrated particles and homogeneous distribution and true detection. All together lead to lacks in harmonization and results, which are hardly comparable. On the other hand, monitoring of microplastics is mandatory in the future strictly regulated by the EU commission in the Drinking water and Wastewater Framework Directive. One step to accurate and precise results will be the development of suitable reference materials mimicking particles in the environment. BAM developed test materials, which are produced by mixing a small portion of microplastic particles with a water-soluble matrix. After solid phase dilution and homogenisation small portions are pressed into tablets and bottled in glass vials (Figure 1). These tablets are well characterized with particle size distribution and SEM images. Additionally, they are tested as reference material candidate according to homogeneity and stability for particle number with µ-IR and µ-Raman as well as on particle mass with Py-GC/MS and TED-GC/MS after ISO Guide 35. Results are promising. The material passed the homogeneity control. No changes are observed within 6 months of storage. The same tested reference material is finally used in sample preparation experiments, where environmental suspended particular matter from surface water or baby milk powders are spiked with the tablets. However, concerns have been raised about the evolution and selection of resistance against disinfectants and AMCs. In turn, resistance against disinfectants and AMCs can be associated to antibiotic resistance due to cross-resistance and co-resistance. We need to understand the mechanisms and risks of disinfectants and AMCs for resistance and cross-resistance evolution to optimize their application and safeguard their long-term efficacy. We used adaptive laboratory evolution (ALE) experiments based on repeated exposure of bacteria to disinfectants. Our results show that repeated disinfection of E. coli with benzalkonium chloride in suspension results in a 2000-fold increase in survival within 5 exposure cycles. Adaption is linked to the initial presence of persister cells highly tolerant to benzalkonium chloride. We used the same approach to develop standardizable ALE experiments to determine resistance evolution to AMCs. The results highlight rapid adaptation of E. coli and P. aeruginosa towards copper surfaces. Moreover, there are multiple situations in the clinic or in the environment in which biocides and antibiotics co-occur and in which combination effects can shape their antimicrobial activity or their selective effects. Our work with P. aeruginosa shows prevalent combination effects of biocides and antibiotics, ranging from synergy to antagonism and resulting in the selection for or against antibiotic resistant strains. The combination effects are dependent on the biofilm mode-of-growth, manifesting in apparent differences in the structural arrangement of antibiotic sensitive and resistant strains in biofilms exposed to combinations. Furthermore, biocides affect rates of mutation and horizontal gene transfer, thereby having a potential facilitating effect on resistance evolution. Taken together, our work shows that the role of biocides as potential drivers of resistance evolution and selection deserves further study and regulative action. They dominate a range of hostile natural and man‑made environments, from desert rocks and salterns to dishwashers, roofs and solar panels. Due to their slow growth and a lack of genetic tools, the underlying mechanisms of black fungi’s phenotypic traits have remained largely unexplored. We chose to address this gap by genetically engineering the rock‑inhabiting fungus Knufia petricola (Eurotiomycetes, Chaetothyriales), a species that exhibits all characteristics of black fungi. The state‑of‑the‑art genetic toolkit, together with the annotated genome sequence of strain A95, firmly established K. petricola as a model for exploring microcolonial black fungi. In addition to unique identification, this also includes reliable quantification. Two different methodological approaches are used for the quantification. With vibrational spectroscopic techniques such as µ-FTIR and µ-Raman, results are obtained in the form of particle number, size and shape. Instead, with thermal analytical technics as TED-GC/MS and Py-GC-MS the results are expressed as mass concentration.. Both concepts offer different information variables. In terms of routine monitoring, it is necessary to obtain a rapid sequence from sampling to the detection result. For this reason, this study focuses on TED-GC/MS and Py-GC-MS. Even though thermal analytical methods generally require less sample preparation, some matrices require it. This is usually done if the analyte concentration is too low or if strong matrix effects such as signal suppression and false positive signals occur. This talk presents advanced sample preparation for baby milk powder as food example and density separation for microplastic analysis in surface waters. A preparation protocol based on citric acid was selected for the milk to remove as much as possible of the matrix. The low pH value leads to a fast and effective protein precipitation and minimizes filter cake formation, making filtration possible, reduces the number and the amount of compounds in detection and hence, simplifies the evaluation. Microplastics in surface waters and sediments must be concentrated not only because of their low microplastics mass content but also in terms of homogeneity. For this purpose, a density separation was carried out using a concentrated sodium iodide solution, which led to a high reduction of the inorganic mineral matrix and made possible to obtain a representative subsample of initial masses of up to 80 g. Their aim is to support existing hygiene procedures, and to help combat the increasing threat of antimicrobial resistance. However, concerns have been raised over the potential selection pressure exerted by such surfaces, which might drive the evolution and spread of antimicrobial resistance. In my presentation, I will highlight the risks and knowledge gaps associated with resistance on antimicrobial surfaces by different processes including evolution by de novo mutations and horizontal gene transfer, and species sorting of inherently resistant bacteria dispersed onto antimicrobial surfaces. The latter process has the potential to select for antibiotic resistance via cross-resistance between traits that confer resistance to both the antimicrobial surface coating and antibiotics. Conditions in which antibiotics and antimicrobial coatings are present simultaneously (e.g. implants) will lead to more complex interactions that can either result in the selection for or against antibiotic resistance. We mapped these interactions between several antimicrobials and antibiotics on growth and selection of Pseudomonas aeruginosa. We find prevalent physiological (i.e. synergy and antagonism) and evolutionary (i.e. cross-resistance and collateral sensitivity) combination effects. Understanding these interactions opens the door to tailor therapeutic interventions to select against resistance. In additions, we need new methods and translational studies that investigate resistance development to antimicrobial surfaces under realistic conditions. Therefore, I will present recent developments in our lab on the development of such a method based on existing efficacy standards. This contribution shows how an improvement in the measurement data situation can improve the significance and comparability of outdoor exposures. Photooxidative ageing is calculated using sample polymers for which the corresponding modelling data (Exposure Response Functions, ERF) is available. The mean square deviations are estimated, which result from using different spectral irradiances and different spectral sensitivities, both of the sensors and the polymers. It will be shown here how the significance of the irradiance values increases with increasing approximation of the spectral sensitivities of the sensor and polymer. Also, the mean square deviations are estimated, which result from using different activation energies and surface temperatures. A selection of different variables to describe the weathering exposure is created. The potential of either individual values or data pairs is evaluated in terms of significance or correlation to the calculated photooxidative ageing. Recording hourly data also enables subsequent evaluations of individual test specimens, adapted to the respective colour or to determine activation energies. An accumulated value, calculated from PIT-radiometer irradiance and a suited surface temperature, can provide a comparison of different outdoor exposures over different locations or years. Thus, a characterisation of the weathering exposure would be related to a reference spectral sensitivity (much closer to a polymer than that of a common UV radiometer), one reference activation energy, and different surface temperature scenarios with, on the one hand, graded solar absorptions (white / grey / black) and, on the other hand, different thermal couplings (coated metal / thick plastic). This results in three values each for plastic samples and coated metals (light / grey / dark). It will never be possible to accurately predict the weathering tests for individual samples (with their specific sensitivities), but the proposed approaches can go a long way towards the comparability of outdoor weathering exposures. Um dennoch die Ergebnisse einer Beanspruchung vergleichen zu können, ist eine aussagekräftige Charakterisierung der Freibewitterung wichtig. Traditionell werden Daten bekannter Einflussgrößen wie Globalstrahlung, Temperatur oder Feuchtewerte herangezogen. Obwohl man weiß, dass diese Größen an den exponierten Oberflächen wechselwirken und kombiniert als Mikroklima Einfluss auf Degradationsprozesse nehmen, werden schon aus Praktikabilitätsgründen diese separat als klimatische Messgrößen angegeben. Die Aussagekraft dieser Daten ist mäßig. Ziel dieses Beitrags ist es, die Aussagekraft und damit Vergleichbarkeit zu erhöhen, indem diese Messdaten als Stundendaten zu Wirkintegralen vorverarbeitet und jahresweise akkumuliert werden. Betrachtet wird dafür der Prozess der Photooxidation, da dieser Alterungs-mechanismus bei allen Kunststoffen auftritt. Für eine einfache Modellierung wird die wirksame UV-Bestrahlung, gewichtet mit einer Arrhenius-abhängigen Ober¬flächen-temperatur (TOF), über den Beanspruchungszeitraum von hier je einem Jahr akkumuliert. Die Idee einer solchen Messgröße für photooxidative Ober¬flächen¬alterung wurde bereits vorgestellt [1]. Dort wurde ein Gerät vor¬geschlagen, mit dem dieser Wert für verschieden absorbierende Oberflächen für die Dauer einer Exposition akkumuliert wird, und ergänzend eine Abschätzung der Betauungs¬zeiten dieser Referenz-Oberflächen erfolgen könnte. Hier soll es um eine rechnerisch generierte Zusammenstellung dieser photo¬oxidativen 1-Jahres-Alterungswerte für verschiedene Standorte gehen. Ein Temperaturfaktor erfasst dabei den Einfluss der Strahlungserwärmung auf eine weiße, graue bzw. schwarze Referenzoberflächen, spiegelt also die Kopplung von solarer Strahlung und Temperatur wider [1]. Ermittelt wird er aus Stunden¬daten der Globalstrahlung und der Lufttemperatur. Die spektrale Wirkung wird aus Sonnenhöhenständen entsprechend der geografischen Breite mit Hilfe der FastRT-Oberfläche [2] als PiT·h-Wert abgeschätzt [3]. Anhand der errechneten Jahreswerte können die Schweregrade der photo¬oxidativen Beanspruchung an den verschiedenen Standorten unmittelbar verglichen werden. In particular metals play important roles in all areas of our daily life - from building materials to high tech products. Due to the increasing consumption of metals and corresponding waste production, an elevated release into the environment takes place. Furthermore, metals in direct contact with the environment undergo corrosion leading to a release into the (aquatic) environment. Thus, lifespan of products/buildings are substantially reduced – hence unnecessary economic costs arise. Thus, research in this regard is needed within the force field of metal/material - environment. Evaluating the environmental impact of materials as well as developing “safe” materials, new analytical methods are highly needed. One promising powerful tool is single cell-ICP-ToF-MS for multi-elemental analysis on a single cell/organism level. Within this presentation the concept, strength as well as challenge of single cell-ICP-MS are briefly introduced. Two application examples are presented: (i) assessing the environmental impact of metals and (ii) the impact of the environment on metal-based materials and the derivation of potential environmental-friendly material protection strategies. These applications highlight the strength of new analytical approaches to explore the durability and safety of newly developed materials. Thus, analytical chemistry is one corner stone to transformation of modern society into circular economy (CEco). (i) Diatoms are located at the bottom of the food chain. Thus, toxicological relevant metals taken up by diatoms possibly accumulate within the food web causing harmful effects. Diatoms are common test system in ecotoxicology. To investigate potential metal uptake and effects, we developed an on-line single cell-ICP-ToF-MS approach for multi-elemental diatom analysis. Our approach is a new potential tool in ecotoxicological testing for metal-based materials. (ii) Next to classical corrosion processes, microorganisms are responsible for so called microbially influenced corrosion (MIC). MIC is a highly unpredictable process relying on interaction pathways between cells and the metal surface. Sheding light on MIC processes and derivate potential protection strategies, we applied single cell-ICP-ToF-MS for MIC research on a single bacteria/archaea level. It turned out that microorganism are taking up particular metals from alloys - thus, single bacteria-ICP-ToF-MS will enable development of corrosion protection strategies. T1 - Single-event analysis of discrete entities using microwave-induced nitrogen plasma–mass spectrometry N2 - ICP-MS has become a standard for (ultra)trace elemental analysis due to its excellent sensitivity and multi-element capabilities. However, its widespread use is hampered by spectral interferences—especially in the low mass range (<81 amu)—primarily arising from Ar-based polyatomic ions, such as ArO+ and Ar2+, which compromise the accurate quantification of key elements like Fe and Se.1 Over the years, improvements such as collision/reaction cells and sector-field configurations have helped mitigate these interferences, albeit often at the cost of instrument complexity and increased operational burden.The microwave-induced nitrogen plasma (MINP) source, sustained by microwaves at atmospheric pressure and using nitrogen as the plasma gas, offers a fundamentally different plasma environment. Nitrogen is not only more economical (40-60%) and readily available than Ar, but also avoids the generation of problematic Ar-based interferences. While MINP had been previously applied in optical emission and bulk mass spectrometry,2 its implementation in single-event detection had not yet been demonstrated. This study pioneers the application of MINP-MS in single-event mode for real-time, high-throughput characterization of NPs, cells, and MPs. The evaluation began with Fe2O3 NPs monitoring the 56Fe nuclide, with a limit of detection of 8.6 ag for Fe, equivalent to a particle size threshold of 19 nm—surpassing the detection capabilities of quadrupole-based ICP-MS systems. Size distribution results obtained by SP-MINP-MS for Fe2O3 NPs (20–70 nm) matched closely with transmission electron microscopy (TEM) and dynamic light scattering (DLS), confirming the method’s accuracy. For Se, despite its high ionization energy, metallic SeNPs (150 and 250 nm) were reliably quantified by monitoring 80Se. A calibration curve constructed using SeNP standards yielded excellent linearity (R2 = 0.9994). This approach was further extended to single-cell analysis, using Se-enriched yeast (SELM-1 CRM) as a model. A transport efficiency-independent calibration strategy was employed, relying on SeNPs to determine Se content per cell. The results showed strong agreement with data from conventional SC-ICP-MS, with average Se masses of ~65 fg per cell, validating the performance of SC-MINP-MS for biological systems. Additionally, the instrument's capability to handle large, low atomic number particles was demonstrated via the analysis of polystyrene (PS) and polytetrafluoroethylene (PTFE) MPs. These MPs (2.5–3.0 µm) were quantified by monitoring 12C+ signals and applying a calibration strategy using citric acid as a standard. The resulting size distributions closely matched nominal sizes, reinforcing the system’s robustness for micrometer-sized polymeric materials. Event durations ranging from 470 to over 900 µs were consistent with literature values for single-entity ICP-MS and correlated well with particle size. These findings establish single-event MINP-MS as a promising analytical platform for analyzing discrete entities. It provides significant advantages over conventional Ar-based ICP-MS, including reduced interferences, lower operational cost, and comparable or superior sensitivity for analytes such as Fe and Se. By avoiding the limitations of Ar-based plasmas and enabling accurate quantification across a wide range of particle types and sizes, MINP-MS in single-event mode opens new avenues for high-resolution, interference-free elemental analysis at the individual entity level. In marine ecosystems, OTCs can cause severe damage to biodiversity, leading up to the extinction of vulnerable species. Due to the extensive use of tributyltin (TBT) as a biocide on ship hulls, it has been considered one of the most hazardous substances intentionally introduced into the aquatic environment. This resulted in a global ban on TBT-containing products in the 2000s. However, recent studies indicate the emerging presence of organotin pollutants. OTCs are known to persist and accumulate in marine sediments, posing a long-term threat to the environment. These harmful substances can be set free and dispersed even after several decades. Therefore, analyzing sediment probes is imperative for a thorough monitoring of pollution. However, species-specific analysis of OTCs at required concentration levels in complex environmental matrices remains challenging. Chromatographic systems are commonly used for their analysis, but the required sample preparation is time-consuming and prone to contamination and analyte loss. The coupling of electrothermal vaporization and inductively coupled plasma-mass spectrometry (ETV/ICP-MS) demonstrates high potential as a rapid, convenient, and chemical-saving scanning tool for environmental samples. This method enables the direct on-line fractionation of organic compounds from an inorganic fraction and provides element-specific detection at ultra-trace levels without complex sample preparation. Since OTCs are generally more toxic than ionic or elemental tin, analyzing them as a sum parameter is advantageous. Additionally, the determination of both organic and inorganic tin, rather than just organic tin, reveals valuable information about the fate of OTCs. The main challenge in obtaining accurate quantitative data using direct solid sampling techniques like as ETV/ICP-MS is applying a suitable calibration strategy. Our isotope dilution approach overcomes matrix effects in ETV/ICP-MS analysis and is compatible with commercial systems. However, it is negatively impacted by the unwanted accumulation of marine organisms on the ship hulls. To prevent the so-called bio-fouling, antifoulant paints containing the organometallic compound tributyltin (TBT) have been used on ship hulls since the 1960s. Compared to the generally considered non-toxic inorganic tin, the endocrine disruptor TBT leads to the endangerment of non-target organisms. The most prominent effects are imposex in snails and shell deformation in oysters. This resulted in a global ban by the International Maritime Organization in 2008. Nevertheless, recent studies have found alarmingly high concentrations in sediment. Organotin compounds (OTCs) are known to persist and accumulate in marine sediments, posing a long-term threat to the environment. Once released from the ship hull, TBT gets effectively absorbed into the sediment. Under anaerobic conditions and the absence of light, the half-life is extended up to several decades. The resuspension and transport of sediment remobilizes butyltin compounds, making them bioavailable even in distant regions. The monitoring of OTCs in sediment is inevitable to conduct a comprehensive risk assessment and a thorough monitoring of pollution. Conventionally, species-specific analysis of OTCs involves (gas-)chromatographic separation after extraction and pre-concentration of the analytes. However, the sample preparation is time-consuming, chemical-intensive, and prone to species-transformation, respectively incomplete extraction. In this context, electrothermal vaporization coupled with inductively coupled plasma-mass spectrometry (ETV/ICP-MS) shows high potential as a rapid and chemical-saving screening tool for OTCs in environmental samples. The main challenge for obtaining correct, quantitative data with direct solid sampling techniques like ETV/ICP-MS is the application of a suitable calibration strategy. In this context, a new quantification method based on on-line isotope dilution analysis has been developed and validated by a reference material with certified OTC content. Our approach overcomes matrix effects while being compatible with commercially available ETV systems. Using an optimized multi-step ETV temperature program, the full separation of OTCs, inorganic tin, and major matrix components could be achieved. Since its use as a screening tool, analyzing OTCs as a sum parameter is beneficial and can complement chromatographic approaches. Small fibers and fragments (microplastics: 1-1000 µm) can be released from textiles by mechanical abrasion during application (e.g. wearing) or washing. To date, the occurrence, transport pathways, and impacts of such small fibers on the environment and human health remain unclear. The European Commission revised the Urban Wastewater Treatment Directive (Directive (EU) 2024/3019) and made monitoring of microplastics mandatory in the next years. Additionally, standards for microplastic detection are developed in the ISO standardization body ISO/TC 147/SC 2 with the documents ISO/DIS 16094-2 and ISO/DIS 16094-3. Detection mainly occurs by thermoanalytical methods for mass or vibrational methods with microscopes for numbers. Such standards and regulatory efforts need validated methods, which themselves need reference materials for validation. Reference materials are well characterized materials that are produced for a specific purpose and are homogeneous and stable within one property of interest. The shape and the composition of the reference particles/fibers will affect the detection, hence it is important to validate the instruments with fibers, if this is the task for monitoring campaigns. This work describes the production of realistic fibers as reference material for microplastic analysis. The individual samples contain of a small number (< 1000) and mass (< 20 µm) of fibres. To date, little is known about the effects of glutaraldehyde on the susceptibility of bacteria to antibiotics and its role in the selection of tolerant phenotypes. Objectives • To determine the effect of glutaraldehyde exposure on the survival of E. coli, S. aureus and P. aeruginosa to antibiotics • To find the mechanistic basis for antibiotic tolerance upon glutaraldehyde exposure Materials & Methods Four bacterial isolates were exposed to sub-inhibitory glutaraldehyde. Antibiotic tolerance was determined by time-kill assays. Regrowth dynamics (lag times) were determined with ScanLag. E. coli was further investigated, using RNAseq to identify genes and processes involved in antibiotic tolerance. Mutants of candidate genes were screened for their antibiotic tolerance and heterogeneous target gene expression under stressed and unstressed conditions. Results Short-term exposure to sub-inhibitory levels of glutaraldehyde induced tolerance to high doses of bactericidal antibiotics. Tolerance to antibiotics was associated with highly heterogeneous regrowth dynamics and global transcriptome remodeling. Differentially expressed genes represented diverse biological functions and cellular components, including antibiotic efflux, metabolic processes, and the cell envelope. The heterogeneous regrowth dynamics and the diversity of the differentially expressed genes are likely related to the unspecific mode-of-action of glutaraldehyde. Among the many differentially expressed genes, several genes were identified that were not previously associated with antibiotic tolerance or persistence, which, when overexpressed alone, increased antibiotic tolerance. Conclusion Our results highlight how the big advantage of a disinfectant, its unspecific mode-of-action, can induce transient tolerance to antibiotics in bacteria. Environments with high salinity (> 1.5 Meq of NaCl) are resided by halophiles (salt-loving microorganisms). To compensate for the intensive osmotic stress, they have resorted to two main adaptation strategies: 1) production of compatible solutes and 2) accumulation of intracellular KCl. Microbial community analysis of several high salinity environments revealed a number of recurring genera, including Halomonas and Halanaerobium. However, the impact of halophiles on the overall integrity and stability of the storage facilities remain largely unknown. To evaluate the suitability and stability of saline storage facilities, several model halophilic microorganisms, such as members of Halomonas, will be selected as testing subjects. First, the impact of halophiles on the infrastructure will be determined using an integrative approach by combining a number of techniques, including electrochemistry, TOF-SIMS, SEM/FIB/EDS and FIB-TEM. Second, the abilities of halophiles to alter the fuel composition (i.e. increase/decrease the fractions of H2) will be monitored using gas chromatography by growing them under high pressure. The European biocidal products regulation requires environmental risk assessments for biocidal products under service conditions. This is only possible, if processes that lead to biocide emissions into the environment are understood and can be reasonably predicted. Actual emissions due to leaching result from different processes that are affected by material properties and environmental conditions. Transformation of biocides can affect emissions considerably. Leaching and possible transformation of the film preservatives carbendazim, diuron, OIT and terbutryn in paints was observed under laboratory and field conditons in order to investigate the influence of different factors, e.g. water contact and radiation on biocide emissions. The paints contained either white titanium dioxide or a red iron oxide pigment to check whether pigments affect leaching processes. Methods. Test specimens of a water-based styrene acrylic dispersion paint on glass were exposed to UVA-radiation in a UV weathering device and/or exposed to water contact by immersion events on the basis of the European standard EN 16105. Plywood panels were coated and exposed to natural weather conditions in a semi-field experiment. Concentrations of biocides and transformation products were analysed in leachates from laboratory tests, runoff samples and remaining coatings. Results. The impact of weathering conditions on transformation processes differs for the investigated active substances. Water contact, radiation as well as pigments in the paints determine the pattern and amount of transformation products on coatings, leachates and runoff water. Observations from laboratory and field experiments were compared. It proved that results from laboratory tests support understanding of data from field experiments. Detailed knowledge on the fate of active substances under environmental conditions supports manufacturers to develop safe applications of film preservatives and authorities to evaluate its environmental impact. Family houses, multi-storey residential buildings, office buildings and office towers have been modelled in detail. The frequency-dependent response due to a free-field excitation has been evaluated for walls, columns and floors. The ratio of building amplitudes to free-field amplitudes starts with uB/u0 = 1 at zero frequency and is usually lower than 1 at 50 Hz, the end of the frequency range considered here. In between, amplifications occur due to several reasons. There are „soil resonances“ where the whole building is vibrating on the compliant soil, “column resonances” where the upper storeys are vibrating on the compliant columns, and the “floor resonances” where the floors are vibrating excited by their supports. Results are presented for all building types, but a special focus is set on office buildings. A parameter study shows the influence of the stiffness of the soil, the number of storeys, and the width of the building. It has been found that the “soil resonance” is strongly modified by the low-frequency floor resonances for the normal office building. The main resonance of a twenty-storey office tower is determined equally by the “soil mode” and the “column mode”. It is an important observation for these office buildings that the resonances can differ for different parts of the building such as the centre, the edge, the corner, and the core of the building. This leads to non-uniform vibration modes across the building, which look like another type of “floor resonance” and which have been observed in several real building projects. Experimental results will be shown which can confirm the calculated phenomena. How could behavior of junctions be included? To which extend is it necessary? On the conference Geo-Chicago 2016, Swan and Yuan presented an ultimate limit state equation for the short-term material resistance of junctions. We discuss limitations and suggest improvements to include long-term behavior of junctions. Nevertheless, the approach applies only to a specific mode of shear-tensile failure of junctions and only to strictly rigid geogrids. A new design concept presented in the same year by Jacobs on the conference GeoAmerica for the special case of the anchorage of geogrids, which reinforce capping system on long and steep slopes of landfills, tried to overcome these drawbacks. We discuss the aspects of this concept related to the long-term behavior of junctions and the interplay between the load applied to junctions and the flexibility of longitudinal ribs. This interplay and the long-term junction strength determine the long-term behavior of geogrids. These organisms either fix these pigments in their cell wall or secrete them into their extracellular environment to protect themselves against an array of physicochemical stresses (e.g., UV irradiation, desiccation, ...). Melanin can adsorb metals like Fe. How this affects fungal uptake of Fe and deterioration of Fe-containing minerals and metals is however less known. To study this, we use the model fungi Knufia petricola A95, a rock-inhabiting fungus known to deteriorate minerals and have melanised cell walls, and Amorphotheca resinae, able to contaminate fuel tanks, secrete melanin and corrode metals. In K. petricola, we have deleted genes involved in melanin production and Fe uptake using CRISPR/Cas. Through comparison of the geochemical signatures of these gene deletion mutants with those of the wild type (WT), we explore the specific mineral/metal deterioration mechanisms of melanised fungi. Fe isotope signatures of the biomass of melanin- and Fe uptake-deficient mutants of K. petricola revealed that Fe adsorbed either directly onto melanin or after being reduced by Fe reductases. Importantly, once adsorbed to melanin, Fe could not be mobilised and taken up into the cell: both the WT and its melanin-deficient mutant, previously grown at Fe replete conditions, showed similar growth at Fe deficient conditions. Olivine dissolution experiments revealed that Fe oxidation inhibits dissolution. K. petricola was able to enhance dissolution when this inhibition is strongest (at pH 6) and prevented dissolution when this inhibition is weakest (at pH 4). The fungus therefore dissolves olivine by interacting with the oxidised Fe at the olivine surface. However, Fe uptake did not seem to be involved: mutants deficient in various Fe uptake mechanism dissolved olivine at the same rate as the WT. This indicates that Fe adsorption onto melanin might play a key role. This is also shown by K. petricola’s ability to enhance olivine dissolution even further if secreting a melanin precursor and A. resinae’s corrosion of carbon steel whilst secreting melanin. Combined, our results imply that the Fe adsorbed to melanin cannot be taken up but enables fungi to deteriorate Fe-containing substrates at a higher rate. Due to their slow growth, robust cell walls and the lack of sexual cycles and genetic tools, the underlying mechanisms of their phenotypic traits have remained largely unexplored. Using recently developed genetic tools, it is now possible to manipulate the genome of the rock-inhabiting model fungus Knufia petricola. Thus, gene functions and the cell biology of black fungi can be studied using CRISPR/Cas9-based genome editing and live-cell imaging with genetically encoded fluorescent proteins. Here, we are addressing the question to which extent constitutive pigment formation (melanin and carotenoids) and responses mediated by the stress-activated mitogen-activated protein (MAP) kinase contribute to the observed extremotolerance of K. petricola. The mutations of pks1, phs1 and both genes result in melanin-free (pink), carotenoid-free (black) and pigment-free (white) strains, respectively. The other putative melanogenic genes were identified in the genome, deleted to confirm their involvement in DHN melanogenesis and co-expressed in Saccharomyces cerevisiae for reconstruction of the synthesis pathway. Sak1 encoding the stress-activated MAP kinase was deleted in the wild-type and different pigment-deficient backgrounds. Growth of the obtained single, double and triple deletion mutants was tested by droplet tests on media supplemented with different stress-inducing agents. The Δsak1 mutants show slightly reduced growth rates even without environmental pressure and are hypersensitive to different stresses: e.g. osmotic, oxidative, membrane, pH and heat stress. Melanin-free Δsak1 mutants are more sensitive than black Δsak1 mutants to some but not all stress conditions, suggesting that melanin and the SAK1 pathway have complementary roles in protecting K. petricola from stress. With it, a more complex ontogeny derived from an ancestral linear developmental pathway common to all cockroaches and a wide array of morphologically distinct castes carrying out the division of labour emerged. We sought to underpine the complexity of this evolutionary transition at the transcriptomic level. We present the gene co-expression network analysis of 305 RNA-seq samples from the brains of 15 species of cockroaches and termites from different instars and castes. With this dataset, we investigate how the network of gene evolved between solitary cockroaches, sub-social cockroach and eusocial termites. Moreover, we investigate the emergence of the two most striking evolutionary innovations in termites, the appearance of the soldier caste, common in nearly all termites and thought to have evolved once and subsequently lost in some species, and the true worker caste, the sterile workforce of the colony thought to have derived from the totipotent false worker caste in four different lineages of termites independently. Genetic studies are hampered by slow growth, lacking sexual cycles and difficulties to transform these fungi. Recently, we implemented CRISPR/Cas9-based genetic tools enabling efficient targeted mutations of the rock inhabitant Knufia petricola (Eurotiomycetes/Chaetothyriales) for studying stress responses, the unusual types of cell division, mineral weathering and symbiotic interactions with phototrophic microbes (Voigt, Knabe et al. 2020, Sci Rep; Erdmann et al. 2022, Front Fungal Biol). To further explore the genome of this fungus as representative of the polyphyletic group of black fungi and to assign functions to yet unknown genes, a forward genetics approach is aimed. We chose the two-component Activator/ Dissociation (Ac/Ds) transposon system from maize for generating a collection of insertional mutants by in-vivo mutagenesis. For the optimal use of this genetic tool, an inducible promoter for the expression of the Ac transposase and by this the regulatable transposition of the resistance cassette-containing Ds transposon is requested. However, endogenous promoters for nitrate assimilation and galactose catabolism that are often used in fungi for regulatable gene expression are not inducible by their substrates in K. petricola suggesting that the regulatory networks for nutrient acquisition differ significantly in the oligotrophic black fungi. Therefore, the metabolism-independent Tet-on system was adopted and validated using a Ptet::gfp construct. No GFP fluorescence was detected in absence of the inducer doxycycline (DOX) while fluorescence intensities increased with increasing inducer concentrations. Consequently, the Tet-on system is suitable for regulatable expression of endogenous and foreign genes in K. petricola. This study employs cutting-edge machine learning techniques, specifically SLEAP pose estimation software, to investigate the interactions of *Reticulitermes flavipes* within controlled environments. By leveraging advanced tracking methods, this research addresses challenges such as track breaks in movement data and uses custom coding solutions to ensure accurate reconnections, preserving the continuity and reliability of individual and group behavior analyses. Custom-designed 3D-printed habitats facilitate controlled observations, while machine learning enables the annotation and tracking of key body segments (mandible, thorax, and abdomen). This approach provides granular insights into colony dynamics, highlighting task differentiation and social clustering. Future applications include the development of automated, continuous recording setups and expanded experiments to examine the impact of varying environmental conditions. This interdisciplinary work, combining biology and data science, advances our understanding of termite social systems and paves the way for predictive simulations of their activity. Especially, (+) single-stranded RNA viruses like Deformed Wing Virus (DWV) have become prominent due to a novel vector from Asia, the ectoparasitic mite – Varroa destructor. The evolution of DWV may have been significantly affected by the establishment of a new transmission route, as represented by V. destructor. According to virulence-transmission trade-off theory, viruses found in vector-invaded hosts may evolve a level of virulence that maximizes transmission and subsequent spread in the colony. In this study, we tested this prediction by passaging DWV in white-eyed pupae by microinjection over 10 generations, to emulate the vectoring function of V. destructor, while subjecting the virus to 3 different transmission selection regimes: (i) Early: at 4 days, (ii) Optimal: at 6 days, and (iii) Late: at 8 days post-injection. The evolved viral genomes are analyzed by long-read (Oxford Nanopore) sequencing. The reconstructed viral population dynamics are compared with the inter-host transmission and viral load, to identify fixed mutations contributing to the adapted virulence. Using a site-/gene-specific test of natural selection, we explore the phenotypic effects of virulence among the evolved viruses. Weit verbreitet ist mittlerweile die „Fused Filament Fabrication (FFF)“ Technologie. Hier wird ein thermoplastisches Filament geschmolzen, durch eine Metalldüse extrudiert und anschließend schichtweise so auf ein Druckbett aufgetragen, dass ein 3D-Objekt entsteht. Das Filamentmaterial wird dabei thermisch stark belastet, was zur Emission von Aerosolen sowie flüchtigen organischen Verbindungen (VOC) führt. Dabei werden hauptsächlich ultrafeine Partikel (UFP, dP < 100 nm) freigesetzt werden, die sogar in manchen Fällen im sub-4nm Größenbereich einen signifikanten Anteil ausmachen können (Tang und Seeger 2024). Die gesundheitliche Relevanz eingeatmeter UFP ist durch sehr viele Studien gut belegt. Während eines i.d.R. mehrstündigen FFF-Druckvorgangs wird ein Anwender mit diesen Luftschadstoffen im Innenraum exponiert, häufig ohne eine Einschätzung des damit verbundenen Risikos zu haben. Die Exposition kann durch technische Faktoren (z.B. Druckerausstattung und -einstellung, Innenraumventilation), aber auch erheblich durch die Filamentauswahl beeinflusst werden. Unser Vorschlag zur Risikominderung besteht in der Auszeichnung emissionsarmer Filamentprodukte mit dem Umweltzeichen „Blauer Engel“, um so den Verbrauchern eine fundierte Auswahl zu ermöglichen. In unserem Projekt entwickeln wir dafür ein kammerbasiertes, standardisierbares und robustes Prüfverfahren zur vergleichenden Messung der Emission aus Filamenten. Mit dem Strangdruck-Verfahren (engl. Strand Printing Method, SPM) wird eine festgelegte Filamentlänge mit konstanter Rate extrudiert und auf dem Druckbett abgelegt, ohne dabei ein 3D-Objekt aufzubauen. Diese Vorgehensweise reduziert den Einfluss experimentell schlecht zu kontrollierender Faktoren sowie die Ausfallquote. Eine detaillierte Beschreibung und die Vorteile von SPM werden in Tang und Seeger (2022) erläutert. SPM wurde bereits für 44 Filamentprodukte aus unterschiedlichen Polymeren und Additiven als Vergleichstest angewendet. Alle Messungen wurden in einer klimatisierten 1 m³-Emissionsprüfkammer durchgeführt. Die Gesamtanzahl der emittierten Partikel (TP) dient als Beurteilungsmaß, in Anlehnung an die Vergabegrundlage DE-UZ-219 des Umweltzeichens Blauer Engel für Laserdrucker. Unter den getesteten Filamentprodukten variiert TP um ca. vier Größenordnungen (1E+9 ≤ TP ≤ 1E+13). Auch innerhalb jeder der untersuchten Polymergruppen variierte TP signifikant. Die Partikelemission kann daher nicht allein nach dem Basispolymer kategorisiert werden. Unsere Resultate belegen, dass die Stärke der Partikelemission eher eine charakteristische und individuelle Eigenschaft eines Filamentproduktes ist. Die Herkunft der Polymere, die herstellerspezifischen Additive sowie die nicht deklarierte Verunreinigungen können einen starken Einfluss auf die Emission haben. Die Auszeichnung von emissionsarmen Filament-produkten verspricht somit eine einfache, aber effektive Maßnahme zu sein, um Verbrauchern eine Minderung des Expositionsrisikos beim FFF-3D-Druck zu ermöglichen. It describes the production of microplastic test material for the use in an interlaboratory comparison. IWe report the first results of an ILC on microplastic detection methods organized under the pre-stantdardisation plattform of VAMAS (www.vamas.org/twa45/) as Project 2 “Development of standardized methodologies for characterisation of microplastics with microscopy and spectroscopy methods”, within the Technical Working Area TWA 45 “Micro and Nano Plastics in the Environment”. The ILC has gathered 84 participants all over the world representing all continents. Reference materials were developed by BAM to test the main methods for MPs detection as addressed in ISO/TR 21960:2020: thermo-analytical methods (Py-GC/MS and TED-GC/MS) and vibrational methods (µ-IR and µ-Raman). As far as the mass fraction is concerned, accurate results with quite a good precision were achieved with the thermo-analytical methods. Number-based results are not adressed here. The polyketide synthases (PKS) release the heptaketide YWA1, the hexaketide AT4HN or the pentaketide T4HN. The first two products are deacetylated by ‘yellowish-green’ hydrolases to T4HN, and T4HN is further converted by a core set of enzymes to DHN. Final polymerization steps are accomplished by multicopper oxidases. DHN melanogenesis is often regulated in a spatial and temporal fashion resulting e.g., in melanized reproduction and survival structures of the foliar plant pathogen Botrytis cinerea (Schumacher 2016, Mol Microbiol). In contrast, a polyphyletic group of Ascomycetes (microcolonial fungi/ black yeasts) dwelling in hostile habitats such as bare rock surfaces in hot and cold deserts, exhibits constitutive melanogenesis. Here, DHN melanin builds a protective layer around all vegetative cells thus contributing to the survival of diverse environmental stresses even without specialized reproduction structures. For studying the relevance of constitutive DHN melanogenesis for tolerance of abiotic and biotic stresses, adhesion to substrates and subsequent damage of colonized surfaces, the rock-inhabiting fungus Knufia petricola was chosen as gene functions in this fungus can be studied by CRISPR/Cas9-based genome editing. The putative melanogenic genes were identified in the genome of K. petricola, deleted to confirm their involvement in DHN melanogenesis and co-expressed in Saccharomyces cerevisiae for reconstruction of the synthesis pathway. Phenotypes of DHN-deficient mutants are studied. Here, we will discuss the role of the DHN melanin layer on the outer cell wall in tolerating UV irradiation. N2 - In den letzten 20 Jahren ist die Aufmerksamkeit vieler Forschender sowie der Politik und der Bürger selbst auf ein wachsendes Kunststoffproblem unserer Zeit stark gestiegen. Mangelnde Wertstoffkonzepte, Sammelstellen und unbedachtes Einbringen von Kunststoffprodukten in die Umwelt führen dort zu einer Anreicherung. Durch äußere Witterungseinflüsse können diese Produkte degradieren und fraktionieren, so dass heute in allen Teilen der Welt Mikroplastik (1-1000 µm, ISO/TR 21960:2020) unterschiedlicher Polymermaterialien nachgewiesen werden kann. Bei Mikroplastik gilt das Vorsorgeprinzip. Die Partikel können weiter zerfallen in Nanoplastik (< 1 µm, ISO/TR 21960:2020). Ob eine toxikologische Gefährdung von Mikro- oder Nanoplastik ausgeht, wird vielfältig untersucht. Bisher stehen valide Ergebnisse aus. Untersuchungen zur Häufigkeit, dem Transport und möglicher Senken und Eintragspfade muss aber Rechenschafft getragen werden, so dass ein Monitoring von Mikroplastik bereits in der Revision der Trinkwasserrahmen-Richtlinie und der Abwasserrahmen-Richtlinie gefordert wird. Bedenklich ist auch die Verordnung über Mindestanforderungen an die Wasserwiederverwendung (2020/741), in welcher nicht verbindlich gesagt wird, Wasser welcher Reinigungsstufe eines Klärwerks auf die Felder zur Wiederverwendung aufgebracht werden darf. Einer Regulierung über die EU-Plastikstrategie hinaus mit z.B. Verboten für Einwegplastik-Produkte und Rezyklingquoten kann nur entgegengewirkt werden, wenn valide Messmethoden, Referenzmaterialien und Standards existieren, die das tatsächliche Ausmaß von Mikroplastik-Konzentrationen in Luft, Wasser, Boden und Biota zuverlässig und genau aufzeigen.
