TY - CONF A1 - Jiang, Shixiong T1 - Adipokinetic hormones and their receptors in Blattodea: a comparative analysis N2 - Adipokinetic hormone (AKH) is a neuropeptide produced in the insect corpora cardiaca that plays an essential role in mobilizing carbohydrates and lipids from the fat body to the hemolymph. AKH acts by binding to a rhodopsin-like G protein-coupled receptor (GPCR), the adipokinetic hormone receptor (AKHR). In this study, we tackle AKH ligand and receptor gene evolution as well as the evolutionary origins of AKH gene paralogues from the order Blattodea (termites and cockroaches). Phylogenetic analyses of AKH precursor sequences point to an ancient AKH gene duplication event in the common ancestor of Blaberoidea, yielding a new group of putative decapeptides. In total, 16 different AKH peptides from 90 species were obtained. Two octapeptides and seven putatively novel decapeptides are predicted for the first time. AKH receptor (AKHRs) sequences from 18 species, spanning solitary cockroaches and subsocial woodroaches as well as lower and higher termites, were subsequently acquired using classical molecular methods and in silico approaches employing transcriptomic data. Aligned AKHR open reading frames revealed 7 highly conserved transmembrane regions, a typical arrangement for GPCRs. Phylogenetic analyses based on AKHR sequences support accepted relationships among termite and cockroach lineages to a large extent, while putative post-translational modification sites do not greatly differ between solitary and subsocial roaches and social termites. This study provides important information not only for AKH and AKHR functional research but also for further analyses interested in their development as potential candidates for biorational pest control agents against invasive termites and cockroaches. Within this paper, already existing front-runner approaches and recent and ongoing product policy-making processes were reviewed. Based on the results, an EU front-runner approach is outlined. The presented approach (i) refers to performance levels of the best products already available on the market, (ii) aggregates information in existing databases, and (iii) works semi-automated. Together, all three attributes have a high potential to facilitate and accelerate the specification of appropriate minimum requirements for products at the EU level. This way, EU policymakers can deliver on the core objectives of the Ecodesign legislative framework much better. The basic mechanism and its legal entrenchment of the approach are illustrated for the energy efficiency of energy-related products. In addition, the Front-Runner Approach can be applied to any product group in the scope of the upcoming Ecodesign for Sustainable Products Regulation and to a wide range of product-related minimum requirements, such as durability, reparability, or recycled content. The study’s objective is to suggest a tailor-made and dynamic approach to keep the EU product legislation up to date using innovative technology based on the investigation of current regulations and identify the gap. Experiences from three international case studies suggest that a front-runner approach to setting energy-performance standards can drive innovation and reduce energy consumption via promoting energy-efficient products; transparency about available products is one of the key factors and can be established by a database. The EU front-runner approach comprises extending the existing energy label database (or making use of the digital product passport) and introducing a legislative procedure that triggers changes in the energy efficiency requirements in the specific EU regulations if the database shows that a certain threshold value is reached. Challenges such as limited EU staff capacities and opportunities such as increased dynamic are discussed. The motivation is to reduce the energy consumption in the milling of the iron ore by utilizing coarser iron ore particles (+0.05 mm) and to select a suitable binder for improving pellet properties. Iron ore fines in the range of 0.05–0.25 mm was selected and classified into three size ranges. Fluxed iron ore pellets were prepared using lime as a binder for the basicity of 0, 1, and 2. Reduction of these pellets with a packed bed of coal fines was performed in the temperature range of 900–1200 °C for a duration of 30–120 min. The direct reduction kinetics of the iron ore pellets were studied by employing diffusion and chemical reaction control models to the experimental data. The results show that pellets made with coarser iron ore particles have improved reduction behavior and kinetics. The reduction reaction is found to be a mixed control. The activation energy for the reduction reaction varies from 44.3 to 74.76 kJ mol−1 as iron ore particle size decreases from 0.25 to 0.05 mm and basicity increases from 0 to 2. To analyze the emissions reduction potential, we conducted a comparative life cycle assessment of a proton exchange membrane fuel cell in a residential application and a conventional system with a stand-alone gas condensing boiler and electricity from a grid mix. The period under review was referred to as the service life of the PEMFC and is assumed to be 10 years (83,038 h of PEMFC). The applicability of this in a single-family house built between 1991 and 2000 under German climatic conditions was investigated. The functional unit is set to the thermal energy demand of 16,244 kWh/a and electricity demand of 4919 kWh/a of a single-family house. The impact assessment method “CML 2001–August 2016” was used in this investigation. The manufacturing phase of the proton exchange membrane fuel cell showed disadvantages, whereby the use phase had significant advantages in most of the environmental impact categories as compared to the conventional energy supply system. Considering the whole life cycle, the advantages from the use phase could outperform the disadvantages from the manufacturing phase in most of the impact categories, except for ADP elements and TETP. Due to the strength of the C-F bond, they do not decompose but accumulate in the environment posing a risk for nature and humans alike. While the use of some PFAS, like perfluorooctanoic acid (PFOA) is already regulated in the EU, new ‘precursor’ substances are used to replace them. However, these precursors may be equally harmful to the environment. In recent years, many ‘hot spot’ sites with high PFAS contaminations in soil have been distinguished, but no German-wide background values have been determined yet. Knowing these background values is crucial to understand the degree of PFAS contamination, underpinning future regulatory decisions. Due to the complexity and variety of different PFAS compounds, one needs to apply a broad spectrum of different techniques to capture most of the PFAS content in one sample. In the framework of this project, 600 soil samples are taken at different sites all over Germany. The concentration of 30 PFAS - 13 carboxylic acids (C4-C18), 5 sulfonic acids (C4-C10) and 12 precursor substances) - is determined in these soil samples using three different sample preparation approaches: 1) ultrasonic extraction of the soil samples with Methanol; 2) preparation of eluates to test the leaching behaviour of the PFAS and 3) TOP assays following the method published by Houtz & Sedlak. Targeted LC-MS/MS is used to determine PFAS concentration after all three preparation steps. For the TOP-assay, the concentration of all 30 PFAS is compared prior to and after the oxidation reaction. Besides the ‘classic’ TOP-Assay, two other TOP-Assay approaches, the dTOP Assay and the photoTOP-Assay, are tested and the results of all three approaches are compared. First, all three TOP-Assay approaches will be tested on a reference soil spiked with a solution including all 30 PFAS measured in the project. In addition, initiatives on recyclability and other aspects of sustainable products can be observed in France under the Ecomodulation Approach for some electrical and electronic products. Neither the Commission's ambitions nor the Ecomodulation Approach have so far led to a crucially better ecodesign of products. Therefore, at the EU level, indices for reparability (already developed for smartphones and tablets), recyclability and recycled content of products are being discussed within the Ecodesign legislative framework in an attempt to achieve better ecodesign through advanced consumer purchasing decisions. The product standards requested by the European Commission to assess the recyclability (EN4555) and recycled content (EN4557) of energy-related products have already been published, but are purely based on a mass-based approach. Material-specific aspects such as the environmental impact or the financial, technical and redesign efforts required by manufacturers to facilitate the recycling and use of recycled materials are not addressed. Materials that are easy to recycle and use and have a high recycled content, such as copper, are treated in the same way as plastics that are difficult to recycle and use in products as post-consumer recycled plastics. The circularity is still low so that enhanced use of secondary building material is required to close loops. Three different secondary building materials are discussed based on exemplary research results: construction and demolition waste (C&D waste), soil-like material and incineration bottom ash (IBA). Focus was laid on the environmental compatibility of the materials examined mainly by standardized leaching tests. C&D waste was investigated after a wet treatment using a jigging machine, soil-like material and IBA were characterized with respect to their material composition. The environmental compatibility in particular was studied using standard leaching tests (batch tests and column tests). It was concluded that soil-like material can mostly be utilized even when the precautionary limit values set are exceeded by a factor of less than 2. For C&D waste the fine fraction below 2 mm and the content of brick material is problematic. IBA fulfills quality level HMVA-2 following German regulation. Higher quality levels of utilization might be achievable with better treatment technologies. Many companies run own lab procedures for mycotoxin analysis, but it is a major goal to simplify the analysis. The enzyme-linked immunosorbent assay using horseradish peroxidase as enzymatic label, together with 3,3',5,5' tetramethylbenzidine (TMB)/H2O2 as substrates allows sensitive mycotoxin detection with optical detection methods. For the miniaturization of the detection step, an electrochemical system for mycotoxin analysis was developed. To this end, the electrochemical detection of TMB was studied by cyclic voltammetry on different screen-printed electrodes (carbon and gold) and at different pH values (pH 1 and pH 4). A stable electrode reaction, which is the basis for the further construction of the electrochemical detection system, could be achieved at pH 1 on gold electrodes. An amperometric detection method for oxidized TMB, using a custom-made flow cell for screen-printed electrodes, was established and applied for a competitive magnetic bead-based immunoassay for the mycotoxin ochratoxin A. A limit of detection of 150 pM (60 ng/L) could be obtained and the results were verified with optical detection. The applicability of the magnetic bead-based immunoassay was tested in spiked beer using a handheld potentiostat connected via Bluetooth to a smartphone for amperometric detection allowing to quantify ochratoxin A down to 1.2 nM (0.5 µg/L). Based on the developed electrochemical detection system for TMB, the applicability of the approach was demonstrated with a magnetic bead-based immunoassay for the ergot alkaloid, ergometrine. Under optimized assay conditions a limit of detection of 3 nM (1 µg/L) was achieved and in spiked rye flour samples ergometrine levels in a range from 25 to 250 µg/kg could be quantified. All results were verified with optical detection. The developed electrochemical detection method for TMB gives great promise for the detection of TMB in many other HRP-based assays. A new sensing approach, based on an enzymatic electrochemical detection system for the mycotoxin fumonisin B1 was established using an Aspergillus niger fumonisin amine oxidase (AnFAO). AnFAO was produced recombinantly in E. coli as maltose-binding protein fusion protein and catalyzes the oxidative deamination of fumonisins, producing hydrogen peroxide. It was found that AnFAO has a high storage and temperature stability. The enzyme was coupled covalently to magnetic particles, and the enzymatically produced H2O2 in the reaction with fumonisin B1 was detected amperometrically in a flow injection system using Prussian blue/carbon electrodes and the custom-made wall-jet flow cell. Fumonisin B1 could be quantified down to 1.5 µM (≈ 1 mg/L). The developed system represents a new approach to detect mycotoxins using enzymes and electrochemical methods. N2 - Zur Entwicklung von einfach zu bedienenden Vor-Ort-Geräten, welche für die Analytik von wichtigen Parametern in der Lebensmittelindustrie eingesetzt werden können, sind elektrochemische Methoden besonders vielversprechend. Viele Unternehmen führen bereits Mykotoxinanalytik in eigenen Laboren am Produktionsstandort durch, dennoch gibt es große Bestrebungen Analysenmethoden weiter zu vereinfachen. Der Enzyme-linked Immunosorbent Assay (ELISA), welcher häufig mit der Meerrettichperoxidase als enzymatischem Label und 3,3',5,5'-Tetramethylbenzidine (TMB)/H2O2 als Enzymsubstraten arbeitet, ermöglicht den sensitiven Mykotoxinnachweis mithilfe optischer Detektionsmethoden. Zur Miniaturisierung des Detektionsschrittes wurde in dieser Arbeit der Aufbau von elektrochemischen Detektionssystemen für die Mykotoxinanalytik untersucht. Dazu wurde zunächst die elektrochemische Reaktion von TMB an verschiedenen Materialien siebgedruckter Elektroden (Kohlenstoff und Gold) sowie bei verschiedenen pH-Werten (pH 1 und pH 4) untersucht. Eine stabile Elektrodenreaktion, welche die Grundlage für den weiteren Aufbau des elektrochemischen Detektionssystems darstellt, konnte bei pH 1 an Goldelektroden erzielt werden. Basierend darauf wurde eine amperometrische Detektionsmethode für oxidiertes TMB entwickelt, wofür eine maßgefertigte Durchflusszelle verwendet wurde. Die amperometrische TMB-Detektion wurde für einen kompetitiven Magnetpartikel-basierten Immunoassay für Ochratoxin A eingesetzt. Mit diesem Assay wurde eine Nachweisgrenze von 150 pM (60 ng L-1) erreicht und die Ergebnisse konnten durch optische Detektion verifiziert werden. Die Anwendbarkeit des Assays konnte in Ochratoxin A dotiertem Bier demonstriert werden, wobei für die Detektion ein tragbarer Potentiostat verwendet wurde, welcher über Bluetooth mit einem Smartphone verbunden war. Hiermit konnten niedrige Ochratoxin A Konzentration von bis zu 1.2 nM (0.5 µg L-1) bestimmt werden. Aufbauend auf dem entwickelten elektrochemischen Detektionssystem für TMB wurde die Anwendbarkeit des Ansatzes auf einen Magnetpartikel-basierten Immunoassay für das Ergotalkaloid Ergometrine, evaluiert. Unter optimierten Bedingungen konnte mit dem Assay eine Nachweisgrenze von 3 nM (1 µg L-1) erreicht werden. In mit Ergometrin versetztem Roggenmehl konnten Konzentrationen von 25 bis 250 µg kg-1 nachgewiesen werden. Die entwickelte elektrochemische Nachweismethode für TMB bietet einen vielversprechenden Ansatz für den Einsatz in vielen anderen Meerrettichperoxidase-basierten Assays. Für das Mykotoxin Fumonisin B1, wurde ein neues sensorisches System entwickelt, welches auf einem enzymatischen elektrochemischen Nachweis basiert. Hierfür wurde eine Aspergillus niger Fumonisin Aminoxidase (AnFAO) rekombinant als Fusionsprotein mit dem Maltose-bindenden Protein exprimiert. AnFAO katalysiert die oxidative Deaminierung von Fumonisinen, wobei H2O2 gebildet wird. Es konnte gezeigt werden, dass AnFAO eine hohe Lagerungs- und Temperaturstabilität hat. Für den Nachweis von Fumonisin B1 wurde das Enzym kovalent an Magnetpartikel gekoppelt. Das enzymatisch produzierte H2O2 konnte anschließend amperometrisch mithilfe von Preußisch Blau/Kohlenstoff-Elektroden in der maßgefertigten Durchflusszelle detektiert werden. Fumonisin B1 konnte bis zu einer Konzentration von 1,5 µM (≈ 1 mg L-1) quantifiziert werden. The surface waves are generated by strong blasts of 25 kg of explosives at a dedicated test site. A 10 km dark fiber link in the vicinity of the test site connected to the test setup records the resulting strain signals. The spike-free and low-noise strain data thus obtained minimize post-processing requirements, making the setup a candidate for real-time seismic monitoring. An analysis of the dispersion characteristics of the generated surface waves is performed using a recently reported optimised seismic interferometric technique. Based on the dispersion characteristics, the shear wave velocities of the surface waves as a function of the depth profile of the Earth’s crust are determined using an optimised evolutionary algorithm. The observation period was up to 281 days. The aging processes proceeded faster in the outdoor storage of the samples. The dominant factor in the leaching behavior is the pH, which starts at values above 12 and decreases to values below 10 (outdoors, <11 indoors). Most heavy metals exhibit minimum solubility in this pH range. The solubility of Sb depends on the prevailing Ca concentration, due to the formation of low-soluble Ca antimonate. The very low sulfate concentrations observed in the leaching tests with fresh IBA could be explained by the presence of ettringite. In the course of the aging reaction, ettringite is transformed into gypsum. The results from batch tests were compared with those from column tests, showing reasonable agreement. Leaching dynamics can be better followed with column tests. All results confirm that the use of IBA is possible under German law. For example, patients suffering from phenylketonuria should be able to monitor their phenylalanine (PHE) level in a rapid, decentralized, and affordable manner to avoid high levels of PHE in the body which can lead to a profound and irreversible mental disability. Herein, a magnetic and conductive Iron based metal–organic framework (MOF) is prepared using tetrahydroxyquinone and benzene dicarboxylic acid, dual ligands. The uncoordinated carboxyl groups provided the binding sites for PHE. Physicochemical properties of MOF were characterized by XRD, SEM, DLS and FTIR analyses. The effects of different operational parameters: pH (2, 4, 6, 7.2, 8.6, 10, 12), contact time and MOF concentration were optimized for best properties using cyclic voltametric technique. The 10 µl of 1mg/ml MOF (pH 8.6) in deionized water was used as working electrode on screen-printed gold electrode. The presence of new well-defined oxidation and reduction current peak in electrochemical signal of MOF-modified SPGE sensor vs. Ag/AgCl in phosphate buffer solution (pH 1.9) remarks the quantification of PHE. The analytical calibration curve for PHE showed a linear response with concentration in the oxidation and reduction peak current range from 50 to 1000 µM, with a limit of detection (LOD) of 35.16 µM. In diesem Beitrag wird eine neuartige GSL-Strategie für Schwärme vorgestellt, die auf Pheromonkommunikation und Intermittenz der Gasverteilung basiert. Die Agenten, d.h. die Nanodrohnen, emittieren Pheromonmarker in einer virtuellen Umgebung, wenn sie eine neue Gaswolke feststellen. Die Agenten werden durch virtuelle Kräfte gesteuert und nutzen abwechselnd das Wissen des Schwarms, indem sie dem Pheromongradienten folgen, oder erkunden den Suchraum, indem sie einen Zufallspunkt ansteuern. Due to the lack of analytical standards and the enormous numbers of these compounds, current target-based methods (e.g., LC-MS/MS) are not suitable for the analysis of new/unknown PFAS and transformation products. Therefore, PFAS sum parameter analysis is becoming increasingly important. For PFAS determination of surface waters, solid phase extraction (SPE) is commonly implemented for clean-up and pre-concentration of samples. Thus, within this work, single-layer SPE methods (based on Strata™-X, Strata™-X-AW, Strata™-NH2 and Oasis-HLB) were investigated for maximum PFAS coverage utilizing extractable organically bound fluorine (EOF) analysis. Thereby, the optimization procedure relied on the analysis of 3 surface water samples in Berlin, Germany, which were affected by the effluent discharge of wastewater treatment plants. To analyse the SPE elution profiles for EOF and inorganic fluorine, high resolution-continuum source-graphite furnace molecular absorption spectroscopy (HR-CS-GFMAS) and ion chromatography (IC) were used, respectively. Highest EOF concentrations were achieved by using Strata-X/Strata-XAW as SPE sorbents and methanol as eluent. Furthermore, combinations of the most promising SPE sorbents were selected (X/XAW, XAW/X, HLB/X and WAX/GCB) to extract a wider range of PFASs. By comparing multi-layer SPE methods, lower EOF concentrations were obtained for all investigated combination phases compared to the analysed single phases. For the commercially available combination phase Strata-PFAS (WAX/GCB) for all three river water samples, lower EOF concentrations (1.5 times lower) were determined compared to Strata-X. The results have shown that single-layer SPE systems are currently superior compared to combination phases for PFAS sum parameter analysis. The multi-layer SPE methods need further optimization regarding appropriate sorbent combinations, loading volumes and elution conditions. The common structural motif of this compound class is the fluorine – carbon bond on an alkyl backbone, which are known for their chemical inertness and combination of hydrophobicity and oleophobicity. The downside of those properties is the general lack of usable molecular interactions for adsorption to extract them from the environment. The notable exception to that inertness are fluorine – fluorine interactions, which are stronger, the longer the perfluorinated chain length is. Therefore, in this project we investigated the effect of fluorination, length and presence of a charge on the alkyl chain on an adsorber. Towards that goal, in total 16 different adsorber materials were synthesized (fluorinated/non-fluorinated; C4/ C6/ C8/ C10 ; positive charge/no charge) and tested in regard to their potential to adsorb PFOA and perfluoralkylic acids of differing chain lengths. The PFAS concentration was tested via LC-MS/MS to determine the adsorption rate on the various materials. In marine ecosystems, OTCs can cause severe damage to biodiversity, leading up to the extinction of vulnerable species. Due to the extensive use of tributyltin (TBT) as a biocide on ship hulls, it has been considered one of the most hazardous substances intentionally introduced into the aquatic environment. This resulted in a global ban on TBT-containing products in the 2000s. However, recent studies indicate the emerging presence of organotin pollutants. OTCs are known to persist and accumulate in marine sediments, posing a long-term threat to the environment. These harmful substances can be set free and dispersed even after several decades. Therefore, analyzing sediment probes is imperative for a thorough monitoring of pollution. However, species-specific analysis of OTCs at required concentration levels in complex environmental matrices remains challenging. Chromatographic systems are commonly used for their analysis, but the required sample preparation is time-consuming and prone to contamination and analyte loss. The coupling of electrothermal vaporization and inductively coupled plasma-mass spectrometry (ETV/ICP-MS) demonstrates high potential as a rapid, convenient, and chemical-saving scanning tool for environmental samples. This method enables the direct on-line fractionation of organic compounds from an inorganic fraction and provides element-specific detection at ultra-trace levels without complex sample preparation. Since OTCs are generally more toxic than ionic or elemental tin, analyzing them as a sum parameter is advantageous. Additionally, the determination of both organic and inorganic tin, rather than just organic tin, reveals valuable information about the fate of OTCs. The main challenge in obtaining accurate quantitative data using direct solid sampling techniques like as ETV/ICP-MS is applying a suitable calibration strategy. Entry 19 01 11* describes a waste material containing hazardous substances. It was shown that only the hazardous property HP14 (ecotoxic) is of relevance for the classification. If substances with the hazard statement code (HSC) H400, H410, H411, H412 and H413 are present in incineration bottom ash (IBA) above certain level the waste is classified as hazardous, otherwise it is non-hazardous (in this case 19 01 12). It was further shown that only Cu, Zn and Pb compounds have an impact on the classification using a summation rule: 100xΣ c(H410) + 10xΣ c(H411) + Σ c(H412). Limit value is 25 percent. It is obvious that H410 substances have the highest impact. Therefore, the knowledge of the elemental composition of IBA is not sufficient because the content of distinct chemical compounds (with the respective molecular weight) is needed. When the exact speciation is not known a worst-case approach has to be applied. A practical guideline for the estimation of HP14 was developed based on the definition of 4 substance groups: 1. Metals and alloys, 2. Water-soluble substances, 3. Oxides and carbonates and 4. Substances not soluble in weak acids. In substance group 1 only fine Zn powder has the HSC H410. Water-soluble substances are assumed to be H410 substances. Their concentrations are, however, regularly below the cut-off value of 0.1 %. The amount of substance group 3 is estimated by a selective extraction with maleic acid. According to the worst-case approach it is assumed that CuCO3xCu(OH)2 and ZnO represent the respective Cu and Zn species in IBA because these substances have the highest stochiometric factor. Even with this worst-case approach results from operators of IBA treatment plant showed that the limit value of 25 % is not exceeded. The requirements for utilization of IBA in Germany are defined in the Secondary Building Materials Decree (Ersatzbaustoffverordnung EBV) which was set into force in August 2023. Leaching methods (batch test or column test) with a liquid-to-solid ratio of 2 l/kg are applied. IBA fulfills the requirements for class HMVA-2. Actually, even a classification of IBA as hazardous (i.e 19 01 11*) would not affect utilization but handling and transport. In this presentation, a novel ultrasonic testing method is explored in order to acquire part thickness information in the turbine blade’s airfoil. In established industry processes, the measurements are mainly carried out manually and only at a few specific positions of the inspected parts. The proposed method scans the part using a robot arm guiding an ultrasonic array sensor. For ultrasonic coupling to the complex-shaped surface geometry, the inspected part and sensor are immersed into water. A two-step TFM[1, 2] (Total Focusing Method) approach is used to reconstruct the outer and inner surfaces subsequently from the ultrasonic raw data, which are acquired using the FMC[3] (Full Matrix Capture) measurement principle. For each sensor position, the location and geometry of the outer surface is first identified and then used to create an image of an area inside the material. From that image, the inner surface is reconstructed. Finally, part thickness information is deducted from merging location data of inner and outer surface. The result is a high resolution, high precision mapping of the inspected part’s wall thickness. A vast number of publications focus on the identification of warfarin and its major cytochrome P450-mediated phase I metabolites by liquid chromatography (LC) with mass spectrometry (MS) and tandem mass spectrometric (MS/MS) detection techniques. For the first time, electron ionization (EI) induced high-resolution quadrupole time-of-flight mass spectrometric (HR-qToF-MS) data of in-liner derivatized warfarin and selected hydroxylated species is provided in this study as an alternative to LC-MS/MS approaches. Furthermore, the characteristic fragments and fragmentation pathways of the analyzed methyl ethers are concluded. The obtained data of analytical standards, specific deuterated and 13C-labeled compounds prove inductive cleavage of the acyl or acetonyl side chain, methyl migration, and H-migration, along with consequential inductive cleavage as predominant fragmentation routes. Based on the HR-spectral data, commonalities and differences between the analyzed compounds and fragment groups were evaluated with future applicability in structure elucidation and spectra prediction of related compounds. Research has shown that warfarin and other rodenticides are present in the environment and food chain. However, emerging contaminants are subject to degradation by biotic and abiotic processes and advanced oxidation processes. In some cases, detecting the parent compound may not be possible due to the formation of structurally changed species. This approach aims to identify hydroxylated transformation products of warfarin in a laboratory setting, even after the parent compound has undergone degradation. Therefore, the Fenton reaction is utilized to insert hydroxylation into the parent compound, warfarin, by hydroxyl and hydroperoxyl radicals generated by Fe2+/Fe3+ redox reaction with hydrogen peroxide. Using multiple reaction monitoring, a GC–MS/MS method, incorporating isotopically labeled reference compounds, is used to quantify the expected derivatized species. The analytes are derivatized using trimethyl-3-trifluoromethyl phenyl ammonium hydroxide, and the derivatization yield of warfarin is determined by using isotopically labeled reference compounds. The method has a linear working range of 30 to 1800 ng/mL, with detection limits ranging from 18.7 to 67.0 ng/mL. The analytes are enriched using a C18-SPE step, and the recovery for each compound is calculated. The Fenton reaction generates all preselected hydroxylated transformation products of warfarin. The method successfully identifies that 4′-Me-O-WAR forms preferentially under the specified experimental conditions. By further optimizing the SPE clean-up procedures, this GC–MS-based method will be suitable for detecting transformation products in more complex matrices, such as environmental water samples. Overall, this study provides a better understanding of warfarin’s degradation and offers a robust analytical tool for investigating its transformation products. The system employs a wavelength-scanning laser and an imbalanced Mach-Zehnder interferometer to acquire the reflection spectra and the phase of the detected light, respectively. Fading-free and low-frequency measurements are realized via the crosscorrelation of the reflection spectra. The discrete crosscorrelation is used to circumvent the nonlinear frequency sweeping of the laser. Based on the phase of the backscattered light, it is possible to quantify fast environmental variations. The whole system requires no hardware modification of the existing system and its functionality is experimentally validated. We exploited the stepwise infection dynamics of an entomopathogenic fungus in a termite to explore how care-kill transitions occur, and identify the chemical cues behind these shifts. We found collective responses towards pathogen-injected individuals to vary according to severity and timing of pathogen challenge, with elimination, via cannibalism, occurring sooner in response to a severe active infection. However, injection with inactivated fungal blastospores also resulted in increased albeit delayed cannibalism, even though it did not universally cause host death. This indicates that the decision to eliminate an individual is triggered before pathogen viability or terminal disease status has been established. We then compared the surface chemistry of differently challenged individuals, finding increased amounts of long-chained methyl-branched alkanes with similar branching patterns in individuals injected with both dead and viable fungal blastospores, with the latter showing the largest increase. This coincided with the highest amounts of observed cannibalism as well as signs of severe moribundity. Our study provides new mechanistic insight into the emergent collective behaviors involved in the disease defense of a termite society. For the first time, the EU Energy Label will depict, among energy efficiency, also a reparability score, battery endurance, battery lifetime, drop resistance and dust and water ingress protection. However, does the multitude of parameters bear the risk to confuse consumers instead of triggering environmentally sustainable purchase decisions? In a survey in Germany, consumers were asked to make a choice when the label does not clearly identify the environmentally better product. Instead, energy efficiency has to be valued against reparability, reliability against energy efficiency, and reparability against reliability. The sustainability lever of the different aspects is not the same, as e.g. lifetime extension has a much more positive environmental impact than energy efficiency in case of mobile devices. Therefore, the question remains, whether consumers intuitively make the right choice. Results of the survey indicate that consumers interpret the complex label in the desired manner and value those aspects higher, which indeed have a more positive effect on the environment. In this paper, an approach how to maintain a comprehensive MSE-centric terminology composing a mid-level ontology–the Platform MaterialDigital Core Ontology (PMDco)–via MSE community-based curation procedures is presented. The illustrated findings show how the PMDco bridges semantic gaps between high-level, MSE-specific, and other science domain semantics. Foremost among these is the insufficient availability of extensive high-quality data sets essential for robust training. Within SHM, high-quality data is defined by its accuracy, relevance, and fidelity in representing real-world structural scenarios (pristine as well as damaged). Although methods like data augmentation and creating synthetic data can add to datasets, they frequently sacrifice the authenticity and true representation of the data. Sharing real-world data encapsulating true structural and anomalous scenarios offers promise. However, entities are often reluctant to share raw data, given the potential extraction of sensitive information, leading to trust issues among collaborating entities. Our study introduces a novel methodology leveraging Federated Learning (FL) to navigate these challenges. Within the FL framework, models are trained in a decentralized manner across different entities, preserving data privacy. In our research, we simulated several scenarios and compared them to traditional local training methods. Employing guided wave (GW) datasets, we distributed the data among different parties (clients) using IID (independent, identically distributed or in other words, statistically identical) mini batches of dataset, as well as non-IID configurations. This approach mirrors real-world data distribution among varied entities, such as hydrogen refueling stations. In our methodology, the initial round involves individualized training for each client using their unique datasets . Subsequently, the model parameters are sent to the FL server, where they are averaged to construct a global model. In the second round, this global model is disseminated back to the clients to aid in predictive tasks. This iterative process continues for several rounds to reach convergence. Our findings distinctly highlight the advantages of FL over localized training, evidenced by a marked improvement in prediction accuracy . This research underscores the potential of FL in GW-based SHM, offering a remedy to similar challenges tied to data scarcity in other SHM approaches and paving the way for a new era of collaborative, data-centric monitoring systems. In Deutschland wird die Rostasche durch Nassentschlackungssysteme aus dem Feuerraum ausgetragen. Durch den Kontakt mit Wasser setzen direkt hydraulische Reaktionen ein, die eine vollständige Abtrennung der Feinfraktion unmöglich machen. Das wurde bereits in den 1990er Jahren erkannt und führte zur Entwicklung des ABB InRec Verfahrens mit Trockenentaschung als ersten Verfahrensschritt. Auch damals war das Ziel eine möglichst inerte Rostasche für die Verwertung zu erzeugen. In den letzten Jahren wurden in der Schweiz Trockenentaschungssystem installiert, hier vor allem um die Rückgewinnung von elementaren Metallen auch in der Feinfraktion zu verbessern. Welche Potenziale sich dabei erschließen lassen, wurde in einem anderen UBA-Projekt untersucht. Im Rahmen unserer Arbeiten wurde versuchsweise trockene Rostasche an einer Anlage in Süddeutschland genommen und im Labor untersucht. Daher findet man diese Tiere auch an und in vom Menschen angelegten Nahrungsvorräten wie z. B. Getreidespeicher, an Kleidungs- und an Gebrauchsgegenständen aus Wolle, Pelzen oder Fellen sowie an oder in natürlichen Baumaterialien wie Holz. Aus wirtschaftlicher Sicht werden sie dann als Vorrats- bzw. Materialschädlinge bezeichnet. Vorrats- und Materialschädlinge haben sehr ähnliche Lebensstrategien. Sie verfügen über eine breite Toleranz gegenüber abiotischen Umweltparametern und sind in der Regel Nahrungsopportunisten. Sie können längere Zeit ohne Nahrungsaufnahme überdauern und haben unter optimalen Bedingungen hohe Reproduktionsraten. Es stellt sich die Frage, ob die auffälligen Anpassungen von Vorrats- und Materialschädlingen an einen menschennahen Lebensraum das Resultat von Prädispositionen und Selektion hin zu einem Lebensformtyp „Lager- oder Materialschädling“ sind, oder ob die synanthropen künstlichen (Öko)Systeme nur als überdimensionierte natürliche Reservoire fungieren. Ökologische und ethologische Prädispositionen und Adaptationen sowie Verbreitungs- und Fortpflanzungsstrategien von Schadinsekten werden hier diskutiert, um phylogenetische und evolutionsbiologische Szenarien beim Übergang von ursprünglichen, also natürlichen Lebensräumen auf relativ jungzeitliche, von Menschen geprägte Habitate, zu rekonstruieren. The proposed two/two configuration ensures comprehensive coverage of WWTPs, overcoming obstacles. To evaluate the suitability of this method we analyzed plant extracts with molecular absorption spectrometry (MAS) as well as liquid chromatography–tandem mass spectrometry (LC–MS/MS) for mass balance studies (w(F)). French bean plants (Phaseolus vulgaris) were grown on soil spiked using eight PFAS substances that vary in chain length and functional group composition. Specifically, these include three short-chained (C4–C5), five long-chained (C7–C10) carboxylic acids, one sulfonic acid and one sulfonic amide moieties. To investigate substance-specific PFAS uptake systematically, PFAS were spiked as single substance spike. Additionally, we studied one mixture of the investigated substances in equal proportions regarding w(F) and four PFAS mixtures of unknown composition. After 6 weeks, the plants were separated into four compartments. We analyzed the four compartments as well as the soil for extractable organically bound fluorine (EOF) by high resolution-continuum source-graphite furnace-molecular absorption spectrometry (HR-CS-GFMAS) as well as for sum of ten target-PFAS by LC–MS/MS. All three short-chained PFAS perfluorobutanoic acid (PFBA), perfluorobutanoic sulfonic acid (PFBS) and perfluoropentanoic acid (PFPeA) were determined in high concentrations mainly in the fruits of the investigated plants while long-chained PFAS perfluorononanoic acid (PFNA) and perfluorodecanoic acid (PFDA) were mainly determined in roots. PFBS was determined in remarkably high concentrations in leaves compartment by both quantification methods. Overall, comprehensive results of single substance spikes were in good agreement for both methods except for a few cases. Hence, two phenomena were identified: for mixed PFAS spikes of unknown composition huge differences between EOF and sum of target PFAS were observed with systematically higher EOF values. Overall, both methods indicate comparable results with MS being more reliable for known PFAS contamination and MAS being more valuable to identify PFAS exposure of unknown composition. T1 - High polarity analyte(s) in aqueous media: determination of L-PFOA and L-PFOS in ground water N2 - The CCQM-K156 comparison was coordinated by TUBITAK UME on behalf of the Organic Analysis Working Group (OAWG) of the Comité Consultatif pour la Quantité de Matière (CCQM) for National Measurement Institutes (NMIs) and Designated Institutes (DIs) which provide measurement services in organic analysis under the 'Comité International des Poids et Mesures' Mutual Recognition Arrangement (CIPM MRA). Perfluoro alkyl substances (PFAS) such as PFOS and PFOA have been used in numerous industrial applications and products. Because of their high stability and resistance to biodegradation, atmospheric photooxidation, direct photolysis and hydrolysis, they are extremely persistent in the environment. The European Union (EU) Water Framework Directive lists PFOS as a priority hazardous substance that poses a significant risk to the aquatic environment. The use of PFOS-containing Aqueous Film-Forming Foams (AFFFs) has been banned since June 2011 in the EU. As relatively water-soluble, effectively non-degradable compounds, PFOS and PFOA migrate to ground water. They are not removed in the conventional drinking water treatment, and therefore cause health risks in polluted areas. The EU Drinking Water Directive and the European Commission has proposed a limit value of 100 ng/L for the sum of 20 PFAS, including PFOS and PFOA. This study provides the means for assessing measurement capabilities for determination of high polarity measurands in a procedure that requires extraction, clean-up, analytical separation and detection. Successful participation in CCQM-K156 demonstrates measurement capabilities in determining mass fraction of organic compounds, with a molecular mass of 200 g/mol to 700 g/mol, having high polarity pKow -2, in a mass fraction range from 0.5 ng/kg to 500 ng/kg in aqueous media. Nine NMIs and DIs participated in the CCQM-K156 key comparison. Seven institutes reported their results. SPE was applied in the sample pre-treatment and LC-MS was applied for detection. All participating laboratories applied isotope dilution mass spectrometry (IDMS) techniques for quantification. Participants established the metrological traceability of their results using certified reference materials (CRMs) from NMIs with stated traceability; where commercially available high purity materials were used the purity was determined in-house. The CCQM-K156 results for L-PFOA and L-PFOS range from 2.75 ng/kg to 5.50 ng/kg with a % RSD of 19.5 % for L-PFOA and from 2.04 ng/kg to 4.45 ng/kg with a % RSD of 21.3 % for L-PFOS. The KCRV was assigned using a Hierarchical Bayesian Random Effects Model (HB REM) estimator from the values reported by six of the participants. One participant result of L-PFOS and one result of L-PFOA were excluded from the KCRV for technical reasons. The KCRV was 4.9 ng/kg ± 0.4 ng/kg for L-PFOA and 3.8 ng/kg ± 0.4 ng/kg for L-PFOS. The six institutes that were included in the assignment of consensus KCRV all agreed within their standard uncertainties. To reach the main text of this paper, click on Final Report. As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. While liquid chromatography tandem mass spectrometry (LC-MS/MS) is commonly used technique to characterize targeted PFAS in environmental samples, there are more than 10,000 different PFAS known, which have various headgroups and properties. Therefore, several analytical techniques are available to analyse various groups or pools of PFAS or “all” PFAS as a sum parameter. Current decontamination strategies of PFAS-burdened soils mainly consist of adsorption methods using adsorbents for fixation of PFAS in the ground. A second option is the utilization of a “pump and treat” process, cycling polluted soils through a washing plant leading to the concentration of the pollutants in the fine fraction. Both approaches are cost-intensive and not intended for the direct decomposition of all PFAS contaminants. Hence, there is a great demand for innovative developments and chemical treatment technologies, dealing with new strategies of tackling the PFAS problem. Previously, mechanochemical treatment of polychlorinated organic compounds in soils showed an efficient dechlorination. Thus, we investigated mechanochemical treatment of PFAS contaminated soils with various additives in a ball mill and analyzed the PFAS defluorination with gas chromatography mass spectrometry (GC-MS) and liquid chromatography tandem mass spectrometry (LC-MS/MS), respectively, as well es the fluoride mineralization by ion chromatography (IC) and fluorine K-edge X-ray absorption near-edge structure (XANES) spectroscopy. Especially accredited metrology services providers are often bound to paper certificates by regulatory constrictions and sometimes lack of IT-infrastructure. This analog form of metrology data transfer, however, implies hands-on work, in particular for the certificate receiver, as its key values need to be translated into the device- and quality management (QM) system, manually. With thousands of certificates transferred from accredited calibration labs until the industrial regime, this binds a significant amount of human resources and is a well-known source of errors, thus constricting cost-effective and secure workflows. This contribution highlights the status of the advancing digital transformation in metrology, on the example of force calibration. Although this appealing initiative is to a large extend driven by German stakeholders, it currently evolves to extend across all Europe and far beyond.
