T1 - Termite resistance of pine wood treated with nano metal fluorides N2 - Fluorides are well-known as wood preservatives. One of the limitations of fluoride-based wood preservatives is their high leachability. Alternative to current fluoride salts such as NaF used in wood protection are low water-soluble fluorides. However, impregnation of low water-soluble fluorides into wood poses a challenge. To address this challenge, low water-soluble fluorides like calcium fluoride (CaF2) and magnesium fluoride (MgF2) were synthesized as nanoparticles via the fluorolytic sol−gel synthesis and then impregnated into wood specimens. In this study, the toxicity of nano metal fluorides was assessed by termite mortality, mass loss and visual analysis of treated specimens after eight weeks of exposure to termites, Coptotermes formosanus. Nano metal fluorides with sol concentrations of 0.5 M and higher were found to be effective against termites resulting in 100% termite mortality and significantly inhibited termite feeding. Among the formulations tested, the least damage was found for specimens treated with combinations of CaF2 and MgF2 with an average mass loss less than 1% and visual rating of “1”. These results demonstrate the efficacy of low water-soluble nano metal fluorides to protect wood from termite attack. As extensive usage of antibiotics and biocides can lead to the emergence of resistances, various strategies are currently developed, tested and improved to realize anti-bacterial surface properties through surface functionalization steps avoiding antibiotics. In this study, contact-less and aseptic large-area ultrashort laser scan processing is employed to generate different surface structures in the nanometer- to micrometer-scale on technical materials, i.e. titanium-alloy, steel, and polymer. The processed surfaces were characterized by optical and scanning electron microscopy and subjected to bacterial colonization studies with Escherichia coli test strains. For each material, biofilm results of the fs-laser treated surfaces are compared to that obtained on polished (non-irradiated) surfaces as a reference. Tailored surface textures, enabled by different laser-processing strategies that result in topographic scales ranging from nanometers to micrometers may provide a solution to this challenge. While chemical coatings have been widely used, their restricted biocompatibility for eukaryotic cells and attenuated antibacterial-effects due to compound release limit their areas of application and alternative strategies focus on modified surfaces topographies to impede bacterial adhesion. In this work, we used ns-UV laser treatment (wavelength 248 nm and a pulse duration of 20 ns) to generate laser-induced periodic surface structures (LIPSS) with different submicrometric periods ranging from ~210 to ~610 nm on commercial poly(ethylene terephthalate) (PET) foils. Following structurally and chemically analyses, PET samples were subjected to bacterial colonization studies with Escherichia coli TG1, a bacterial test strain with a strong biofilm formation capacity due to the formation of nanofiber-like cell-appendages (pili). Bacterial adhesion tests revealed that E. coli repellence decisively depends on the spatial periods of the LIPSS with the strongest reduction (~91%) in cell adhesion observed for LIPSS periods of 214 nm. Thus, techniques that may shed light on their surface chemistry are attractive tools for providing an understanding of bacterial interactions. One of these tools is Al Kα-excited photoelectron spectroscopy (XPS) with its estimated information depth of <10 nm. XPS-analyses of bacteria have been performed for several decades on freeze-dried specimens in order to be compatible with the vacuum in the analysis chamber of the spectrometer. A limitation of these studies has been that the freeze-drying method may collapse cell structure as well as introduce surface contaminants. However, recent developments in XPS allow for analysis of biological samples at near ambient pressure (NAP-XPS) or as frozen hydrated specimens (cryo-XPS) in vacuum. In this work, we have analyzed bacterial samples from a reference strain of the Gram-negative bacterium Pseudomonas fluorescens using both techniques. We compare the results obtained and, in general, observe good agreement between the two techniques. Furthermore, we discuss advantages and disadvantages with the two analysis approaches and the output data they provide. XPS reference data from the bacterial strain are provided, and we propose that planktonic cells of this strain (DSM 50090) are used as a reference material for surface chemical analysis of bacterial systems. This typically manifests in difficulties to control the processing of LIPSS and in limitations to ensure the long-term stability of the created surface functions. Although human activity contributes to the acceleration of the process, bacteria have a self-driven stabilisation mechanism to protect themselves from such and other external threats: biofilm formation. Nonetheless, it is the adhesion of a single bacterial cell to a surface that triggers the formation of such network of biomolecules and microorganisms, as well as its hazardous consequences. The main objective of this work was to quantify the adhesion force of a single E. coli cell on a Ti substrate via the AFM-related single-cell force spectroscopy, with both the cell and the substrate material being of high clinical relevance. A set of 25 x 25 force displacement curves was acquired with a maximum force of 3.2 nN without dwell time, yielding a topography map and an adhesion force map that showed to be correlated. A mean adhesion force of 0.85 ± 0.175 nN was measured and the presence of cell appendages on the bacterial cell wall was verified through individual force-displacement curves. The search for new materials and coatings with lower colonization potential and antibacterial activity is of great importance to reduce biofilm formation. However, there is no standardized procedure to examine the colonization characteristics of bacteria in the Biofilm state in situ. Here, we describe an automated epifluorescence microscopy system for the semi-quantitative analysis of three-dimensional (3D) biofilms on various surfaces. To analyze adherent bacteria, three materials (glass, steel and titanium) were incubated with bacteria in a flow chamber system. After fluorescence staining of the bacteria, automated image capturing, quantification of the bacteria, measurement of the colonized area and determination of the 3D biofilm height were carried out by using novel software. Furthermore, the materials were examined for their surface topography using white light scanning interferometry. Titanium compared to glass showed a significantly higher number of adherent bacteria. We argue that this was due to the higher microroughness of titanium. The colonized area was in accordance with the number of adherent bacteria and was also significantly larger on titanium coupons compared to glass. Maximum 3D biofilm height on glass coupons was significantly lower compared to the ones on steel and titanium. This novel method enables the standardized, automated investigation of the colonization with bacteria on different materials. This approach can considerably support the characterization of new material surfaces and their innovative coatings by analyzing the amount of attached Bacteria and thickness of biofilms in situ and eliminates the need of conventional cultivation. Nowadays, the emergence of resistances because of extensive usage of antibiotics and biocides in medicine, agriculture and private households have become one of the most important medical challenges with considerable economic consequences. In addition, aggravated biofilm eradication and prolonged cell-surface interaction can lead to increased biodeterioration and undesired modification of industrial and medical surface materials. Various strategies are currently developed, tested, and improved to realize anti-bacterial surface properties through surface functionalization steps avoiding antibiotics. In this study, contact-less and aseptic large-area short or ultrashort laser processing is employed to generate different surface structures in the nanometer- to micrometer-scale on technical materials such as titanium-alloy and polyethylene terephthalate (PET). The laser processed surfaces were subjected to bacterial colonization studies with Escherichia coli test strains and analyzed with reflected-light and epi-fluorescence microscopy. Depending on the investigated surfaces, different bacterial adhesion patterns were found, ranging from bacterial-repellent to bacterial-attractant effects. The results suggest an influence of size, shape and cell appendages of the bacteria and – above all – the laser-processed nanostructure of the surface itself, emphasizing the potential of laser-processing as a versatile tool to control bacterial surface adhesion. EPDM) in Sportböden gilt. In Deutschland gibt es laut DFB ca. 5100 Kunstrasenplätze. In der EU beläuft sich die Anzahl laut ECHA auf 13,000 Kunstrasen- und 47,000 Bolzplätze, Tendenz steigend. Das Hauptziel des Projektes ist daher, eine Massenbilanz für die Freisetzung von MP zu erstellen, um so eine Schätzung des MP-Austrags pro Kunstrasenplatz und Jahr zu ermöglichen. Dafür werden die MP-Emissionen von drei Kunstrasenszenarien in verschiedenen Zuständen (ungealtert, künstlich gealtert und in Echtzeit gealtert) verglichen: die Vergangenheit (Kunstrasen: fossilbasiert, EPDM-Füllung), die Gegenwart (der in Europa am häufigsten installierte Rasen) und die Zukunft (Rasen mit recycelten Grasfasern, keine synthetische Füllung sondern z.B. Quarzsand oder Korkschrott). Dazu werden fabrikneue Kunststoffrasen und EPDM-Granulat unter Laborbedingungen beschleunigt gealtert, um die Freiland-Beanspruchung eines Kunststoffrasens in circa 15 Jahren abzubilden. Die Verwendung spezieller Mikrofiltertiegel in Lysimeter-Experimenten ermöglicht die Abschätzung der Partikelgrößen des emittierten MP, was eine Grundbedingung für die Bewertung einer potenziellen Gesundheitsgefährdung für Menschen ist. Ergänzend werden weitere umweltrelevante Schadstoffausträge aus den Kunststoffrasen quantifiziert und so mögliche Transportpfade überprüft. In Germany, rubber granulate made of ethylene-propylene-diene-monomer rubber (EPDM) is currently predominantly used. So far, there is no sufficient database for estimating MP emissions from artificial turf pitches into the environment and thus their relevance as a source of MP pollution.3 This topic is controversially discussed due to the complexity of sampling and analytics. To close this research gap, this project has the goal to determine mass balances for the emissions of MP from artificial turf pitches to allow an estimation of the amount of MP released per artificial turf pitch and year. Within this study, MP emissions of three artificial turf scenarios at different time states (unaged, artificially aged and real-time aged) are compared: the past (old turf: fossil based, synthetic infill), present (most commonly installed in Europe: fossil based, EPDM infill), and the future (turf with recycled gras fibres, no synthetic infill). To simulate the outdoor weathering during the lifespan of an artificial turf of approx. 15 years, brand-new artificial turf and EPDM rubber granulate were accelerated aged by means of UV weathering and mechanical stress. Potential MP emissions into surface and groundwater are simulated by lysimeter and shake experiments. MP mass contents are subsequently determined by Thermal Extraction Desorption Gas Chromatography/Mass Spectrometry. Using special microfilter crucibles allows the estimation of the particle sizes of the emitted MP, which is a fundamental requirement for an assessment of potential health hazards for humans.
