Termites are one of the most damaging insect pests destroying the cellulose components of historical artifacts. Herbal extracts obtained from resin of Vateria copallifera (Retzius) Alston and seeds of Madhuca longifolia (Konig) Macbride have been used for centuries to preserve e.g. palm leaf manuscripts from insect attack. Herbal extractions of these traditional products for palm leaf manuscript were tested for their effect against the termite species Coptotermes formosanus Shiraki, 1909. Natural and artificial aged herbal extractions were tested to obtain a repellent index. Resin oil of V. copallifera caused slightly higher repellencies than M. longifolia. Artificially aged samples produced lower repellencies than naturally aged samples. The results indicate that the active ingredients are volatile. The potential for barrier treatment was tested only with V. copallifera. Tunnelling-behavior of C. formosanus workers through sand in the presence of V. copallifera resin oil was largely reduced. MIC describes the negative effects a material can experience due to the presence of microorganisms. In Europe, several research groups/ other industrial stakeholders are already dealing with MIC. Unfortunately, the discussions are fragmented and the exchange of information is limited. A true transdisciplinary approach is hardly ever experienced, although this would be logical for this material/biology related challenge. USA, Canada and Australia have strong networks, and develop methods, prevention measures and standards, which Europe is forced to use, since nothing similar exists for a network and combined knowledge to design them according to european standards. This makes Europe extremely dependent and, in some cases, the potential measures or standards cannot been used because the suggested solutions are prohibited by European laws (e.g. use of biocides). Therefore, it is important to initiate a new European MIC-network. Europe needs to combine the efforts as experts in different fields and develop prevention measures according to the European rules, in close cooperation with industry and plant operators and owners of critical infrastructure. This COST Action will provide the necessary interaction and communication, knowledge sharing, training of personnel and of researchers of different disciplines. This will bring Europe to a leading role in this process, bringing ideas on an equal level with other nations, considering the values which are important for Europe and attitudes (e.g.environmental protection) and representing greater protection for people, property and the environment. However, the interaction pathway between cells and the metal surface remains unclear. The development of the MIC-specific ICP-ToF-MS analytical method presented here at the single cell level, in combination with the investigation of steel-MIC interactions, contributes significantly to progress in instrumental MIC analysis and will enable clarification of the processes taking place. For this, a MIC-specific staining procedure was developed. It allows the analysis of archaea at a single cell level and provides information about the interaction of the cells with the staining agent which is extremely scarce compared to other well characterized organisms. In comparison to traditional static and macro flow chamber assays for biofilm formation studies, microfluidic chips allow in situ monitoring of biofilm formation under various flow regimes, have better environment control and smaller sample requirements. In this work, a novel microfluidic platform is developed to investigate biofilm adhesion under precisely controlled bacteria concentration, temperature, and flow conditions. This platform central unit is a single-inlet microfluidic flow cell with a 5 mm wide chamber designed and tested to achieve ultra-homogenous flow in the central area of chamber. Within this area, defined microstructures are integrated that will disturb the homogeneity of the flow, thus changing bacterial adhesion pattern. Here we present the monitoring of bacterial biofilm formation in a microfluidic chip equipped with a microstructure known as micro-trap. This feature is based on a 3D bacteria trap designed by Di Giacomo et al. and successfully used to sequester motile bacteria. At first, fluorescent particles similar in size to Escherichia coli (E. coli) are used to simulate bacteria flow inside the flow cell and at the micro-trap. The turbulences induced by the trap are analyzed by imaging and particle tracking velocimetry (PTV). Secondly, the model strain E. coli TG1, ideal and well described for biofilm studies, is used to analyze biofilm formation in the micro-trap. Therefore, a stable fluorescent strain E. coli TG1-MRE-Tn7-141 is constructed by using Tn7 transposon mutagenesis according to the method described by Schlechter et al. Sequestering of E. coli cells within the micro-trap was followed using epifluorescence microscopy. The novel microfluidic platform shows great potential for assessment of bacterial adhesion under various flow regimes. The performance of structural feature with respect to the generation of turbulences that promote or reduce bacterial adhesion can be systematically examined. The combination of flow analysis and fluorescent strain injection into the microfluidic chip shows that the micro-trap is useful for capturing bacteria at defined positions and to study how flow conditions, especially micro-turbulences, can affect biofilm formation. Efficacy of nano metal fluoride-treated wood was tested against brown-rot fungi Coniophora puteana and Rhodonia placenta. Untreated wood specimens had higher mass losses (∼30%) compared to treated specimens, which had average mass loss of 2% against C. puteana and 14% against R. placenta, respectively. Our approach combined historical analysis with scientific examination of reconstructed sizings. We reconstructed 15 sizing materials based on identified Persian historical recipes and analysed their physical, optical, and spectral characteristics. Additionally, we assessed their behaviour against the mould fungus Aspergillus flavus. The results revealed distinctive properties for each sizing material, shedding light on their potential applications in paper preservation. Furthermore, our investigation demonstrated variations in hygroscopicity, thickness, grammage, and ash content post-sizing. The sizing materials also exhibited different effects on paper reflectance properties. Additionally, our study revealed insights into the impact of sizing on burnished papers, indicating that the mechanical process of burnishing did not significantly alter the chemical composition or spectral properties of the paper, with only minor changes in brightness observed in specific cases. All tested sizing materials supported varying levels of mould growth, indicating potential implications for paper conservation. Our findings provide valuable insights into the historical practices of Iranian paper sizing and offer practical considerations for the preservation of paper manuscripts. It is generally accepted that the natural habitats of most pest insects can be found outside the synanthropic environment in layers of leaf litter, under bark, as well as in rodent or bird nests. Indeed, most of the common fabric pests have been reported as being facultative nidicolous. Therefore, infestation of commodities by pest insects out of these reservoirs is one possibility to be considered. However, the likelihood of a pest´s occurrence and survival outside of the synanthropic habitat largely depends on its ecological potential and competitiveness against other species of the same ecological guild. Some pest species are rarely found in wild habitats, especially in those regions where they are not native and where they have been introduced by man. The fabric pest Tineola bisselliella serves as a good example. Most likely originating from Central or Southern Africa, this insect was introduced into Europe probably not earlier than the late 18th century. Being more tolerant to dry environments than other fabric pests, its economical importance increased during the 20th century when the indoor climate changed because of central heating systems. Its occurrence in outdoor natural habitats must be regarded as accidental. Reported finds of webbing clothes moth larvae in bird nests e. g. have been largely overstated in the literature. T1 - Biosynthesis of zearalenone conjugates by fungi N2 - Zearalenone (ZEN) and its sulfate and glucoside conjugates have been detected in (a broad variety of) food and feed commodities1. Both conjugated derivatives are formed as part of fungal or plant secondary metabolism and thus, belong to the group of modified mycotoxins2. After consumption of contaminated foodstuff, the conjugates can be hydrolyzed by human intestinal microbiota leading to liberation of ZEN that implies an underestimation of the true ZEN exposure. In order to include ZEN conjugates in routine analysis, as well as for toxicological investigation reliable standards are needed. The objective of the present study was to develop a simple and economic method for biosynthesis of ZEN conjugates. Preceding experiments on the biotransformation of ZEN by Rhizopus and Aspergillus species showed a mixed metabolite formation3. Therefore, these known ZEN conjugating fungal strains were screened for their potential to selectively synthesize the ZEN derivatives ZEN-14-sulfate (Z14S), ZEN-14-glucoside (Z14G) and ZEN-16-glucoside (Z16G). The screening was conducted by adding ZEN to liquid fungal cultures. Cultivation conditions and ZEN incubation time were varied. All media samples were analyzed for metabolite formation by HPLC-MS/MS. Z14S was exclusively formed by A. oryzae. Under optimized conditions a specific biosynthesis of Z14G by R. oryzae and Z16G by R. oligosporus was achieved. After liquid-liquid-extraction and preparative chromatographic cleanup 1H-NMR purities of ≥ 73% for Z14S, ≥ 82% for Z14G and ≥ 50% for Z16G were obtained. In addition, a consecutive biosynthesis was developed by first using Fusarium graminearum for ZEN biosynthesis on rice based liquid medium. After inactivation of Fusarium the subsequent conjugation reaction was conducted utilizing Aspergillus and Rhizopus species under the various optimized conditions. In this study an easy and cost-efficient biosynthesis for Z14S, Z14G and Z16G was developed. The developed biosynthesis could be also used for other metabolites like ZEL conjugates. Our results of the in vitro screening indicate also the formation of a ZEL-glucoside and α ZEL-sulfate as major metabolites by R. oryzae. In sum, under optimized cultivation conditions fungi can be easily utilized for a targeted and stereospecific synthesis of ZEN conjugates. T1 - Exploiting termite nest microbiomes for antibiotic discovery by using an ultra-high throughput Microfluidics/FACS driven pipeline combined with a microtiter plate based cultivation strategy N2 - Infections with multi-resistant Gram negative pathogens are a major threat to our health system. In order to serve the needs in antibiotics development we selected untapped bioresources and implemented high throughput approaches suitable for the discovery of strains producing antibiotics with anti-Gram negative activity. Our approaches rely on the hypothesis that Termite associated bacteria are likely to produce potent antibiotics to defend their hosts against entomopathogenic microorganisms. Termite nests and guts harbor suitable, highly diverse microbiomes in which bacterial taxa are present known to potentially produce natural compounds. In a first step the diversity of Coptotermes species nest microbiomes was assessed carefully by using 16S rDNA amplicon sequencing on the Illumina MiSeq platform and nest material was selected to retrieve viable cells by using Nycodenz density gradient centrifugation. In order to analyze the diversity of the culturable termite nest microbiome, bacterial cells were either distributed in 384-well plates (approach 1) or encapsulated in small spheric agarose beads by an high throughput microfluidics technique (approach 2). Cultures obtained from approach 1 were scaled-up in 96-well Duetz-systems for characterization of diversity and for rapid supernatant screening using the bioluminescence-labeled E. coli pFU166. The generated droplets of approach 2 simultaneously received a small population of GFP-tagged Gram negative screening cells and were sorted for low fluorescence using FACS. After elimination of redundancy we performed a fast scale-up of active strains. Implementation of this pipeline allows us to prioritize antibiotics producing strains in a ultra-high throughput fashion and by cultivation of broad diversity in our approches. In 2016, the house longhorn beetle (Hylotrupes bajulus) breeding culture of the Federal Institute for Materials Research and Testing (BAM) in Berlin produced a second female beetle with a unique malformed right antenna similar to the one previously reported. As in the first case the flagellum following the pedical was bifurcated. This time, however, both branches were of similar length with 7 and 8 flagellomeres, respectively. The left antenna was normally developed with 11 antennal segments (scape, pedicel and 9 flagellomeres). This second female stands as F3-generation in direct maternal line to the previous one. A heritable cause for the malformation is likely. N2 - Rare occurrences of termites in Germany. Termites are typical elements of the tropical and subtropical insect fauna. In Europe, their most northern natural range is southern France, Italy and the South-Balkan region. Only occasionally are termites found in Germany. Their occurrence is always very closely associated with the synanthropic environment. Drywood termites like Cryptotermes brevis or Kalotermis flavicollis were accidentally imported with furniture, wooden artefacts or plant material into homes or indoor arboretums. Similar pathways of termite introduction are described for Coptotermes gestroi and Reticulitermes sp. into tropical botanical gardens and tropical theme parks. Although termites were introduced to Germany several times, successful spreading or long term establishment never occurred due to climatic barriers, namely cold winter temperatures. Only Reticulitermes flavipes formed sustainable colonies in the city of Hamburg for several decades. Originating from the eastern United States of America, this species was introduced in the 1930s with infested wood through the sea port. Small subterranean colonies could survive the winter in areas where pipes of the central heating system kept the ground from freezing. Control efforts of this potentially invasive species of economic importance started early on but failed until modern bait techniques were applied in the year 2000. Currently, termites have not been detected in Hamburg for the last five years. T1 - Microbial corrosion of iron coupled to methanogenesis by strains from different environments N2 - Microbially influenced corrosion (MIC) of iron is usually attributed to sulfate-reducing microorganisms (SRM) which act upon the metal by the reactiveness of hydrogen sulfide, and by withdrawal of the available electrons (Fe → Fe²⁺ + 2e⁻ ; E° = 0.47 V) in electrical contact through surface attachment. Also methanogenic archaea are supposed to cause MIC. Because they do not produce hydrogen sulfide, withdrawal of electrons may be their main corrosive mechanism; however, mechanistic details and kinetics of the overall process are poorly understood. Precipitation of siderite (4Fe + 5HCO₃⁻ + 5H⁺ → 4FeCO₃ + CH₄ + 3H₂O) can lead to an insulating layer on the metal surface and lower the corrosion rate. Still, the extent of FeCO₃ precipitation may be significantly influenced by environmental conditions such as pH and advective processes. To investigate the corrosive potential of methanogens, we studied strains isolated from marine sediments (Methanococcus maripaludis 14266, 2067, Methanobacterium-affiliated strain IM1), crude oil tanks (Methanococcus maripaludis Mic1c10, KA1) and the oral cavity (Methanobrevibacter oralis) in a closed (batch) culture, and in a sand-packed flow-through cell with pH control and simulation of a fluctuating environment. Results indicate that the rates of iron corrosion due to coupled methanogenesis (up to 0.3 mm/yr) are comparable to that caused by SRM. Surface analyses of the metal showed severe pitting. Such knowledge and deeper understanding also from an electrokinetic point of view may not only provide further models in microbial electrophysiology, but also contribute to mitigation strategies in MIC. Ectoine possesses protecting properties and stabilizes proteins as well as whole cells against stresses like ionizing radiation and cytotoxins. These properties make ectoine a highly demanded ingredient in cosmetics and pharmaceuticals. To date H. elongata is the industrial Producer strain of ectoine, but several metabolic factors for optimum ectoine production remain to be explored. In this work, we used up to date Metabolic engineering approaches following the ‘Push, Pull, Block – strategy’ to examine targets that contribute to ectoine synthesis. Firstly, the basics of glucose catabolism were inspected to PUSH and enhance carbon flow towards ectoine synthesis. Secondly, lysine biosynthesis was targeted to BLOCK a pathway that is competing for precursors with ectoine synthesis. Thirdly, the mechanosensitive (MS) channels of H. elongata have been examined as possible excretion routes for ectoine. An overexpression of the ectoine excretion channels potentially could PULL out product at the end of ectoine synthesis and increase overall ectoine flux. For the interrogation of central metabolic pathways, we established the new molecular tool CRISPR-mediated interference (CRISPRi) for targeted modulation of gene expression. PUSH Glucose catabolism through the Entner-Doudoroff (ED) and Emden-Meyerhof-Parnas (EMP) pathway was targeted with CRISPRi and examined on gene expression level for ist response to changing salinity and different carbon sources. Changing salinity did not influence gene expression levels of glucose catabolism but the carbon source glucose triggered glycolysis through the (ED) pathway. When gene expression of the ED pathway was downregulated with CRISPRi, the growth rates remained constant. The observations indicate a metabolic overflow mechanism for glycolysis, in which fluxes are constantly high - even at lower salinity when no resources are demanded for ectoine synthesis. The further analysis of glucose to product conversion rates will advise optimum conditions for future industrial cultivation processes. BLOCK Lysine biosynthesis was downregulated with CRISPRi, which led to a significant increase in ectoine production. Hence, the blockage of lysine biosynthesis would be a valuable strategy for the optimization of the industrial producer strain in future studies. PULL MS channels and ectoine regulation are inevitably connected in osmoadaptation. Therefore, ectoine excretion, growth performance and gene expression levels of the MS channels were monitored in steady state conditions and in response to osmotic shock in the wildtype strain and in a MS channel deletion mutant. We observed that the MS channels were essential for the survival of osmotic shock but surprisingly their presence reduced cell growth under high salinity. The MS channels were only partially responsible for ectoine excretion. Thus, alternative ectoine excretion channels must exist and remain to be explored.
