T1 - Disclosing a new aspect in modern European art: multidisciplinary analysis of modern reverse paintings on glass (1905-1955) N2 - The technique of painting on the reverse side of a glass panel was rediscovered by German artists at the beginning of the 20th century. They appreciated the impressive gloss, luminosity, and depth of colours in this genre. Compared to stained glass, the distinctive properties of this technique are: (1) the paint is applied “cold”, hence, it doesn’t involve a firing step, (2) reverse paintings on glass are framed and always viewed in reflected light and (3) the reverse paint stratigraphy is different from canvas paintings, starting with the front most layer and ending with the backing layer. In 1908 several artists, including Gabriele Münter, Wassily Kandinsky, Heinrich Campendonk, August Macke and Franz Marc of the “Der Blaue Reiter” (the Blue Rider) collective took an interest in this technique and started to share their interest with other colleagues in Europe. Our pioneering project is tracing this transfer of knowledge by a multidisciplinary approach in terms of art history, paint technology and material science. More than 100 artists and >1000 reverse paintings on glass were identified during the project. This high number of objects clearly points out that this technique was by far more important for modern art than previously assumed. In-situ, non-invasive measurements (XRF, Raman, VIS, DRIFTS) on a well-considered selection of 67 paintings reveal the broad palette of colorants ranging from traditional to experimental. Special attention is paid to the impact of synthetic organic pigments (SOP) in artists palette. Demonstrative examples by W. Kandinsky, L. G. Buchheim and F. A1 - Hahn, Oliver A1 - Rabin, Ira T1 - Cairo Genizah documents of the 11th century: analysis of writing materials N2 - The Cairo Geniza is an “archive” discovered in the 19th century in Ben Ezra Synagogue in Fustat, a district in Old Cairo (Egypt). The collection of mostly Jewish documents that vary in genres, languages and writing supports contains a large number of early medieval Hebrew manuscripts. The Geniza provides sources for the literary, linguistic, historical studies of the various aspects Jewish life. As the documents attest, at least two Jewish communities co-existed in Fustat up to 11th century: a so-called Babylonian and Palestinian. These communities had different leaders, different traditions and lived independently. The differences seem to manifest themselves also in the paleographical, codicological and some material properties of the manuscripts produced by each community. The aim of this project is to compare the inks used in the Jewish documents depending on different variables: support (paper, parchment), purpose of the manuscript (legal, private, religious), provenance of the scribe. For the determination of the inks type and composition we had to choose non-invasive, non-destructive and portable techniques to analyze the corpus directly on-situ. The analyses were carried out with a micro-X-ray spectrometer ArtTAX® (Bruker GmbH) and a digital stereomicroscope features built-in LED illumination at 395 nm and 940nm, Dino Lite. We would like to stress that though the methods of material analysis listed above have been successfully employed in the field of cultural heritage and conservation including ancient and medieval manuscripts they have not yet been used to study fragments from the Cairo Genizah. In addition to working on constantly improving the laboratory and the methods of analysis, a substantial part of our activities is dedicated to service, by supporting different research projects conducted at the centre. In this talk, we will present our equipment and the possibilities offered by the different techniques available regarding the different kinds of missions: typology and classification of inks, provenance studies, recovery of faded inscriptions and palimpsests, reconstruction of the history of manuscripts, authentication and dating. We will give a brief overview of our past and ongoing activities in the frame of the second phase of the CSMC. These viruses are predominantly positive single stranded (+ss) RNA viruses belonging to the Picornavirales. Managed honeybees are often infested with the invasive mite, Varroa destructor, which vectors RNA viruses including Deformed wing virus (DWV, family Iflaviridae): a leading culprit of colony losses. Many bee viruses have been sequenced and structural features are now available for viruses such as DWV. DWV consists of at least 3 distinct genotypes, two of which have been shown to be differentially virulent in honeybees. Molecular studies have demonstrated that DWV has a mean evolutionary rate of 1.35 x 10-3 per site per year. For such viruses – in contrast to their eukaryotic hosts – ecological and evolutionary timescales significantly overlap. This rapid evolution allows RNA viruses to adapt quickly to novel host environments with recombination representing an additional key source of genetic variation. Interestingly, recombination between genotypes of DWV has recently been shown to be a common occurrence in honeybees. A challenge has been to develop bioinformatics tools that can accurately reconstruct viral haplotypes – including recombinants – from heterogenous high-throughput sequence data. The impact of blood-to-blood Varroa destructor transmission on virus evolution represents an important question in bee virus research. Due to the nature of the V. destructor life cycle, predictions can be made about the potential impact of the mite on virus virulence evolution. Specifically, the developing honeybee host pupa should remain alive until close to the completion of metamorphosis to provide sufficient time for successful mite reproduction, including offspring mating. For optimal transmission, any virus found in a mature and mated daughter mite will hold a significant selective advantage over a virus found in an immature or unmated daughter mite – placing a cost on virus virulence that impacts honeybee pupae before mites can mate. On the other side, viruses replicating too slowly and with delayed virulence effects will hold a selective handicap because fewer transmission units will be found in mated mites. I have hypothesized that the evolution of virus virulence shifted following the arrival of V. destructor, with viruses, including recombinants and/or specific viral genotypes being selected for a level of virulence in pupae (and likely also in adults) that maximises R0, which represents the basic reproductive number of the virus in the host population. R0 is defined by the number of subsequent infections caused by a single infection and it must be greater than 1 for an infection to spread in a population. It is enhanced by maximising the number of transmission units passed to uninfected susceptible vectors, and ultimately hosts (Figure 1). Honeybee viruses are also shared with sympatric wild bees and viral prevalence and sequence data indicate frequent virus transmission between managed and wild bee species. In addition to infecting the western honeybee (Apis mellifera), DWV can infect other Asian honeybee species such as Apis ceranae. Outside of honeybees, DWV has been found widely in bumblebees, including solitary bees and wasps and there is evidence that it can actively replicate in several Bombus and solitary bee species. Whether the arrival of the V. destructor mite in A. mellifera has driven viral emergence in non-Apis bees is a target of ongoing research. This represents a considerable gap in understanding, given the growing role of insects as a source of food and feed, as well as the more general relevance of insects in agriculture and health. Illness due to entomophagy is rare but well documented, including fatal cases following the consumption of termites. Termites are eaten commonly in tropical Asia, Africa and South America, and are among the insects with the highest recorded fat content. There are many species of termites, with a wide range of diets and habitats centering around the consumption of wood and soil substrates. In this study, we report the results from a survey of more than 30 cockroach and termite transcriptomes, with the aim of understanding the diversity and evolution of RNA viruses as well as other potentially pathogenic organisms that are associated with this relevant but somewhat overlooked group of insects. We discuss our results in the context of the possible zoonotic risk posed by insects, as well as in the context of emerging viral and other disease threats that may face insects being reared at industrial scales. Understanding the proximal and ultimate drivers of this major evolutionary transition represents an important goal in biology. One outstanding question concerns the evolution of termite immunity, which is thought to have undergone broad-sweeping adaptations in order to enhance group-level immune protection. To understand the evolutionary origins of termite immunity, we conducted qualitative and quantitative transcriptome analyses along a gradient of sociality. Firstly, we aimed to identify large-scale genetic shifts in immune traits linked to eusociality by comparing immune gene repertoires in solitary and subsocial cockroaches and across a range of eusocial termite lineages. Secondly, we compared the responses of a solitary cockroach, a subsocial wood-roach and different castes of a lower termite species to a non-specific immune challenge, in order to understand how sociality may have influenced the evolution of immune gene regulation. Firstly, we found that termites have a broadly representative repertoire of canonical immune genes as compared to gregarious cockroaches and subsocial wood-roaches. Secondly, with respect to immune challenge, the solitary cockroach and the subsocial wood-roach displayed a similarly comprehensive induced response, while the termite response was considerably dampened by comparison and strongly influenced by caste; with reproductives displaying a generally higher constitutive level of immune-gene expression compared to sterile castes. In summary we did not find termite eusociality to be associated with significant changes in immune gene diversity, but rather to be linked with significant modifications to the regulation of immunity following the origin of division of labour. Polymeric fuel storage tanks containing diesel and biodiesel provide environmental conditions for microbial growth. Several studies demonstrated that bacteria, which were found in contaminated fuel systems, can use fuels as macronutrient; but such bacteria can also cause microbiologically influenced corrosion and fouling. The aim of this study is to investigate the initial attachment behavior of bacteria, isolated from a diesel contamination, on neat and photooxidized high-density polyethylene (PE-HD). Two common PE-HD’s, less- and biodiesel-stabilized, were radiated to UV light representing a tank exposed to sunlight. Bacteria growing in biofilms produce a protective, extracellular matrix, which enables them to tolerate much higher antimicrobial concentrations than free-living bacteria and survive long enough to acquire antimicrobial resistance. Preventive and therapeutic strategies against biofilm infections in clinical settings commonly involve the application of multiple antimicrobials: antimicrobial coatings on the biomaterials and systemically administered antibiotics. This frequent practice harbors the risk of the development of cross-resistance via shared resistance mechanisms between antimicrobials used in material coatings and administered antibiotics. Our goal is to determine how population dynamics within biofilms affect the transmission of resistance mutations. Specifically, we want to identify antimicrobial-antibiotic-combinations that select for and against antibiotic resistance in biofilms by following the population dynamics of resistant and susceptible strains in competition assays. Gaining a better understanding about resistance development and spread in persistent biofilm infections will enable us to provide clinical recommendations for improved administration of antibiotics/antimicrobials in combination with medical device materials to mitigate against biofilm associated antimicrobial resistance. The most well-known culprit for MIC is the sulfate-reducing microorganisms (SRM), such as members from the genus Desulfovibrio. It has been widely accepted that SRM can contribute significantly to MIC through the production of hydrogen sulfide (HS-) or in some cases a direct electron uptake from the metal surface. However, in a real environmental system, SRM is not exclusive and often involved with other microorganisms that may also contribute to MIC, such as methanogens. Methanogenic archaea can produce methane (CH4) using H2+CO2, formate, methylated amines or acetate. Methanogens are highly abundant in the environment and many are found in very extreme conditions, such as high temperature and high salinity. Previous researches have demonstrated that methanogens are capable of MIC, though the specific mechanisms are still under investigation. In the oil and gas industry, methanogens are not considered as the main contributor for MIC since the corrosion rates are often too low. However, the tests for methanogen-induced MIC are usually performed at static conditions, which cannot represent the system accurately. Here, we developed a novel anaerobic system to evaluate the corrosion potential of methanogens under flow conditions. We will use the Methanococcus maripaludis KA1 strain, which was isolated from a crude oil tank, as the organism of interest. A separate system for Desulfovibrio alaskensis will be established for corrosion rate comparisons. Furthermore, we will study the synergistic effects of M. maripaludis and D. alaskensis on MIC under flow. In contrast to other paint techniques (e.g. panel and mural painting), the paint layers are applied in reverse succession starting with the foremost paint layer and ending with the primer (backmost layer). The paintings are viewed in reflected light, thus revealing an impressive gloss, luminosity and depth of colour. The artist group “Der Blaue Reiter” (the Blue Rider) around W. Kandinsky and F. Marc got in touch with this technique in the summers of 1908 and 1909 and spread their knowledge in different regions. Our pioneering project is tracing this transfer of knowledge by a multidisciplinary approach in terms of art history, painting technology and material science. More than 100 artists and >1000 reverse paintings on glass (1905-1955) were identified during the project. This numbers clearly point out that this technique was by far more important for modern art than previously assumed. In-situ, non-invasive measurements (XRF, Raman, VIS, DRIFTS) on a well-considered selection of 67 paintings reveal the broad palette of colorants ranging from traditional to experimental materials. Untersuchungen des Lehr- und Forschungsgebiets Technologie der Energierohstoffe (TEER) der RWTH Aachen University haben gezeigt, dass die Bedingungen für eine energetische Nutzung von CF-Abfällen in Abfallverbrennungsanlagen nicht geeignet sind. Zudem können CF Betriebsprobleme verursachen. Des Weiteren können bei einer unzureichenden thermischen Behandlung sogenannte WHO-Fasern entstehen, die als gesundheitsschädlich einzustufen sind. Eine Alternative können Hochtemperaturverfahren mit höheren Verweilzeiten sein. Es kommt die Herstellung von Calciumcarbid in Frage, bei der CF als Kohlenstoffträger und der Kunststoff als Energieträger genutzt werden. Systematic study of Coptik inks N2 - While studying the socio-geographic history of inks, division 4.5 of the BAM (Bundesanstalt für Materialforschung und Prüfung) together with the Centre for the Study of Manuscript Cultures in Hamburg has developed a protocol for ink analysis. It consists of a primary screening to determine the type of the ink, and a subsequent in-depth analysis using several spectroscopic techniques. The XRF elemental analysis aims primarily at understanding the fingerprints of inks containing metals, making it possible to distinguish among different inks. Our own research aims primarily at recreating a socio-geographic history of inks, parchment, and papyrus and includes the comparative analysis of the writing materials of the Dead Sea Scrolls, ink and papyrus in Ancient and Hellenistic Egypt, and inks in documents from various contemporary medieval communities in Fustat (first nucleus of Cairo). During many years of study, we concluded that the continuous production of Coptic manuscripts from late Antiquity to the Middle Ages offers a unique opportunity for historical study of the ink in a large geographic area. Thanks to the collaboration with the ERC project “PAThs” (www.paths.uniroma1.it), based at the University of Rome La Sapienza, and within the activities of a PhD research dedicated to this topic, we therefore created a new branch of our project focused entirely on the analysis of Coptic inks, pigments, and dyes. This pioneering systematic study of writing materials coming from a specific area and time frame (5th-10th century) aims not only at a better understanding of the complex Coptic multicultural and plurilingual society, but also and mainly at clarifying the links among the Coptic and other societies between the ancient and medieval eras. The main certification requirements for the products as well as conclusions for their use are presented. The Genizah provides sources for the literary, linguistic, historical studies of the various aspects Jewish and non-Jewish life. As the documents attest, at least two Jewish communities co-existed in Fustat up to 11th century: a so-called Babylonian and Palestinian. These communities had different leaders, different traditions and lived independently. Neuste Untersuchungen zeigen, dass nach der reduzierenden Behandlung schmelz-flüssiger Linz-Donawitz-(LD)-Schlacken und Abscheidung des gebildeten metallischen Eisens eine mineralische Schlacke entsteht, die unabhängig von ihren Abkühlbedin-gungen ca. 50–60 Gew.% des wichtigsten Klinkerminerals Alit (C3S) enthält und eine hohe hydraulische Reaktivität aufweist. Die Stabilität des Alits auch nach langsamer Abkühlung deutet auf eine Stabilisierung des Minerals durch Nebenelemente aus den LD-Schlacken hin. Due to the scarcity of P and the increasing world population an efficient and sustainable recycling management is required. A few biogenic waste materials are high in P contents such as sewage sludge and meat and bone meal. Thus, they are suitable for P recycling and fertilizer production. But besides the high P content sewage sludge is often highly contaminated with organic pollutants and toxic heavy metals which have to be eliminated before agricultural field application. In this presentation we show the potential of sewage sludge as secondary resource for fertilizers. This includes our developments in thermochemical processes for the production of novel P-fertilizers from recycled materials. Furthermore, for these novel P-fertilizers common extraction tests to determine the plant-available P are often unusable. Dieser Stoff ist selbst in geringen Konzentrationen nicht nur eine Geruchsbelästigung, sondern auch eine Substanz mit ökologischer und klimatischer Relevanz. Die Bestimmung von Ammoniak in relevanten Konzentrationen von kleiner 25 μg m-3 erfolgt im Allgemeinen über NH4+ als Analyten, spektralfotometrisch oder mit der Ionenchromatographie nach Überführung in die flüssige Phase. Wegen der niedrigen Konzentrationen an Ammoniak in der Außenluft wird meist eine gesammelte Probe verwendet oder die Bestimmung erfolgt nach einer aktiven, anreichernden Probenahme. Die eigenen Arbeiten sollen dazu beitragen, die Ammoniak-Bestimmung in der Außenluft präziser, sowie schneller und kostengünstiger zu machen. Die BAM prüfte daher kommerzielle elektrochemische und Metalloxid-basierte Sensoren, die für diesen Konzentrationsbereich aber nur bedingt geeignet sind. Deshalb wurden alternativ eigene Entwicklungen zum Nachweis von Ammoniak im Spurenbereich aufgenommen, wobei der Analyt über die Änderung der Fluoreszenz eines BODIPY-Farbstoffs bei 550 nm mittels eines portablen Fluoreszenz-Sensors direkt aus der Gasphase gemessen wird. Zur Kalibrierung von Ammoniak-Sensoren und -Messgeräten steht ein stationäres System basierend auf der Mischung von zertifizierten Prüfgasen aus Druckflaschen mit kalibrierten Massendurchfluss-regler (MFC) zur Verfügung. Darüber hinaus erfolgt eine chemische Analyse der verwendeten Gasgemische mittels eines Massenspektrometers. Für die Kalibrierung und Prüfung von Sensoren und Messgeräten vor Ort wurde ein mobiles Prüfsystem entwickelt. Die Generierung von Ammoniak-haltigen Gasen im Spurenbereich von 0,5 nmol/mol bis 500 nmol/mol erfolgt durch das Permeationsverfahren nach ISO 6145-10. Für die Realisierung der Rückführbarkeit der Ammoniakbestimmung werden von den Nationalen Metrologischen Instituten sowie designierten Instituten Standards bereitgestellt und auch weiterentwickelt. Kulturgüter mit Anteilen von Holz, Federn, Wolle, Haare, etc. wie ethnologische oder naturkundliche Sammlungsstücke, aber auch Bilderahmen oder Freilichtmuseen werden von ihnen gerne besucht. Dabei sind es die Jugendstadien (Larven) dieser Insekten, die sich von ungeschützten Exponaten oder Depotstücken, die diese natürlichen Materialien enthalten, ernähren. Die Entwicklung der Larven findet verborgen im Inneren der Materialien statt. Die geflügelten erwachsenen Formen, die sich im freien Raum bewegen, sich fortpflanzen und den Neubefall herbeiführen, treten saisonal sehr begrenzt auf. Eine frühzeitige Detektion der Schädlinge, als erwachsenes Insekt oder als Larve, ist dadurch erschwert. Es gilt folglich deren Präsenz und erste Anzeichen eines Befalls zuverlässig und vor allem frühzeitig feststellen zu können, um geeignete Maßnahmen zum Schutz des Kulturgutes einzuleiten. Neben dem Wissen um die Materialeigenschaften des Kulturgutes sind umfängliche Kenntnis über die Biologie der Schädlinge, zu ihren grundsätzlichen Lebensansprüchen und insbesondere zum Verhalten, das zum Neubefall führt, wichtig. Die Larvenstadien von Holz- und Textilschädlingen zeichnen sich durch hohe Toleranzen gegenüber extremen Temperaturen und Luftfeuchten aus. Für sie ungünstige Bedingungen können sie sehr lange in Ruhephasen überdauern. Nicht erkannter, versteckter Befall von Materialien sind häufige Infektionsquellen für Neubefall von Sammlungen. Verborgene natürliche Befallsquellen im eigenen Museumssystem spielen bei Neubefall von Exponaten eine größere Rolle als der Zuflug von Schadinsekten aus Reservoiren von außen. This is in the same order of magnitude as the thickness of the outer bacterial membrane of gram-negative bacteria, as well as outer membrane molecules as exopolysaccharides and lipopolysaccharides, commonly attached to the cell surface. With the development of near-ambient pressure (NAP)-XPS, bacteria can be analysed with minimal sample preparation. EnviroESCA is a laboratory based NAP-XPS instrument, equipped with a monochromated Al Kα radiation source and a differentially pumped energy analyser connected to an exchangeable sample environment. Biodiversity is commonly estimated based on genetic variation. We investigated a new approach that defines and measures biodiversity in complex microbial communities. We used the variation in multiple functionally-relevant, phenotypic traits measured in parallel in single cells as a metric for microbial phenotypic diversity. We studied phenotypic diversity and ecosystem functioning throughout different photosynthetic layers dominated by divergent microbial communities in the gradient of Lago di Cadagno. We determined genetic diversity by 16S and 18S amplicon sequencing and bulk ecosystem functioning (photosynthesis). In addition, we determined phenotypic diversity using single-cell technologies such as nanometer-scale secondary ion mass spectrometry (NanoSIMS) correlated with confocal laser scanning microscopy (CLSM) and scanning flow-cytometry. We measured functional trait variation between individuals in 13CO2 fixation, 15NH4+ uptake, and variation in physio-morphological cell traits, such as cell size, shape, and auto-fluorescence for various pigments related to photosynthesis. We used the distances between individuals in a multidimensional trait space to derive phenotypic trait-based diversity indices, such as trait richness, trait evenness, and trait divergence. We find that phenotypic trait divergence associates with ecosystem functioning, whereas genetic diversity does not. Including activity-based, single-cell phenotypic measurements with NanoSIMS provided an additional accuracy to the trait-based diversity assessment and allowed us to formulate hypotheses on the mechanisms that shape the correlation between phenotypic diversity and eco-system function. Together, our results show that phenotypic diversity is a meaningful concept to measure microbial biodiversity and associate it with ecosystem functioning. Quarantine of potentially infested objects can be logistically challenging. Anoxia under controlled nitrogen atmosphere is a most compatible but also time-consuming method to eradicate insect pests in all kinds of different materials. Treatment results are usually effected by duration, temperature, humidity and residual oxygen content. During a two-year research project, 34 relevant pest insect species of all developmental stages were tested in several different materials (wood, paper, wool) to monitor treatment success and to determine optimum treatment parameters. Duration of Treatment ranged from one to three weeks at temperatures of 20 - 27 °C. As expected, results showed significant differences in mortality among tested species. Highest tolerance of hypoxic conditions was found in older larvae of Hylotrupes bajulus. However, this species is an unlikely museum pest. Anobiids and other wood boring beetles are more often an issue related to cultural heritage. Tested imbedding materials in general had no mortality lowering influence. A combination of three weeks exposure time at up to 0.5 % residual oxygen and at 24 °C and 50 % RH is recommended for infested artefacts. Mit dem entwickelten Verfahren soll es ermöglicht werden, schwermetallhaltige Filterstäube und -schlämme, die als Abfälle während der Produktion von Roheisen und Rohstahl anfallen und die derzeit größtenteils deponiert werden, zu recyceln und die darin enthaltenen Wertstoffe (v.a. Eisen, Kohlenstoff und Zink) wieder in den Rohstoffkreislauf zurückzugeben. Dies wird durch eine Chlorierung der in den Filterstäuben und -schlämmen enthaltenen Schwermetalloxiden erreicht, die bei Temperaturen zwischen 650 und 1100 °C in Form von Schwermetallchloriden verdampft und so selektiv separiert werden. Erste experimentelle Ergebnisse zeigen, dass die Gehalte an Schwermetallen wie Zink, Blei und Cadmium um bis zu 99,7% reduziert werden können und die thermochemisch behandelten Filterstäube und -schlämme durch das Verfahren in sekundäre Rohstoffe für die Roheisenproduktion umgewandelt werden können. The diversity of the written fragments and their shape presume few accordance concerning purpose, place or time of production. Even before the papers came to Berlin, manuscripts were copied, divided and collected by famous scholars also in Mexico. Back in Europe, Humboldt visited further collections and published the detected hieroglyphic writings altogether with cultural objects and landscapes in the book `Vues des Cordillères et Monuments des Peuples Indigènes de l`Amerique, voyage de Humboldt et Bonpland´, labelled corresponding to the cultural background. By scientific and culture historical analyses, the Humboldt codices are revised and related to each other and others. In especial, the compilation of fragments shown on plate 36 in the book is looked at more closely. Whereas most of the plates represent only one sheet of painted amate paper, here segments with details of seven fragments are put together. It is tried to comprehend why these pieces are on one plate. To find out a common feature of the documents, their content and represented form are compared. A correlation between some pieces is obvious and thus is also expected in the manufacture of the codices, in the kind of material used. For this the inks and colors are determined by scientific analysis including XRF-, Raman-, FTIR- and VIS- spectroscopy. Special material is detected and related to results of former analysis of codices written during the colonial period of America. Herewith a clear cultural assignment of the manuscripts is performed. A relation of the fragments represented on plate 36 among each other and to other codices of the collection Manuscripta Americana in Berlin is discussed. Also, if the manuscripts are unique `originals´ or represent fragments of documents, whose corresponding pieces maybe found- in the best case- incorporated in other collections. We detected, that at least two of them can be put into relation to similar manuscripts deposited in Mexico. Further material analysis should clarify, if the corresponding pieces are identical- from one document, or if one of them is a copy. Herewith an approximation to authenticity features and history is issued. SRM act upon the metal by the reactiveness of hydrogen sulfide (HS-), and by withdrawal of the available electrons (Fe --> Fe2+ + 2e-; E° = -0.47 V) in electrical contact with the metal (EMIC). However, methanogenic archaea can also cause MIC. Because they do not produce HS-, withdrawal of electrons may be their main corrosive mechanism; however, mechanistic details and kinetics of the overall process are poorly understood. To investigate the corrosion potential of methanogens, we studied the EMIC methanogenic strains isolated from marine sediments (Methanobacterium-affiliated strain IM1) and crude oil tanks (Methanococcus maripaludis Mic1c10), in an in-house developed flow-through cell to simulate a fluctuating environment. A co-culture of M. maripaludis and D. alaskensis was also established to study the effect of syntrophic growth on metal corrosion that may occur in industrial pipelines. Results indicate that the rates of iron corrosion due to coupled methanogenesis (up to 0.4 mm/yr) are higher to that caused by the marine SRM Desulfovibrio alaskensis (0.15 mm/yr). The standard method for the determination of volatile organic compounds (VOC) is the sampling onto Tenax-tubes followed by thermal desorption (TDS) and gas chromatography-mass spectrometry (GC-MS) analysis. The EU-LCI list includes some very volatile organic compounds (VVOC) and some VOC for which there are limitations when using the standard method. For VVOC additional sampling is required using stronger absorbers like Carbotrap or multi-bed adsorption tubes. The analysis of VVOC also requires a different GC oven program and a different column for the separation. For the determination of formaldehyde and other low boiling aldehydes (e.g. acetaldehyde, acetone, propanal, propenal) DNPH-cartridges are used which are extracted with acetonitrile followed by liquid chromatography (HPLC-UV) analysis. The derivatisation of propenal and other unsaturated aldehydes (e.g. 2-butenal) with DNPH might lead to lower findings due to incomplete derivatization and forming of by-products. For a better quantification of acetic acid the use of ion chromatography (IC) is recommended because the analysis of acetic acid with the standard method (TDS) leads to lower findings due to break through during sampling. A novel home-made dynamic gas mixture preparation system, operating according to ISO 6145-4 (continuous injection method), has recently been developed and validated. Thanks to the stable temperature control up to 100 oC, the system can prevent condensation of the SVOCs in air at indoor air concentration levels. The in-situ obtained SVOC gas standards can be sampled in sorbent tubes to obtain SVOC transfer standards. A study was performed to determine the optimal sorbent material and storage conditions. This study will be presented together with the results of the 2018 Round Robin test for emission test chamber measurements organised by BAM. Using the novel system, VSL prepared transfer standards with known amounts of VOCs and SVOCs for participants to evaluate their analytical performance. T1 - Quantitative elemental analysis of ambient aerosol particles using portable TXRF N2 - A reliable analysis of aerosol particle is curial for enforcing EU air quality regulations to protect human health, and for research on climate change effects [1]. Although metrics such as PM10 and PM2.5 are currently in use, the level of uncertainty of aerosol metrics is too high and the traceability is insufficient. Within the AEROMET project [2] procedures are developed aiming at reducing the uncertainties of particle mass, size, and number concentration measurements including the characterization of regulated components in airborne particles. Here, we present an approach how to improve the uncertainties of the particle mass by mobile total reflection x-ray fluorescence (TXRF) analysis. The combination of TXRF and aerosols sampling techniques supported by reference-free synchrotron radiation-based XRF enables a quantitative real-time analysis of particle mass. During in-field campaigns, the procedure was tested, monitoring the size dependent mass concentrations of specific elements in ambient aerosols under dynamic conditions. This approach allows a direct time and size-resolved analysis without laborious digestion steps and a reduced risk of contamination. Aerosol particles were sampled in a 13-stage DLPI impactor on acrylic discs. TXRF analysis was performed on-site with the transportable spectrometer S2 PICOFOX (Bruker Nano GmbH). The TXRF quantification was based on internal standardization. At moderate air pollution levels (PM10 20 µg/m³) sampling times of less than 2 hours were enough to detect elements in different particle size bins. The on-site approach and the high sensitivity of TXRF enables the observation of rather quick changes in the quantity and distribution of elements in an ambient aerosol on the day of sampling. The analysis of the morning and afternoon sampling shifts reveals the occurrence of the elements Fe, Ca and Si in different size bins as well as their temporal change in respective mass concentrations over the day while the distributions of several other elements remain unchanged. Exhibit display cases should protect cultural objects from dust as well as from mechanical and physical damage. To ensure a stable climate inside the display cases, a low air exchange rate is maintained. Typically air exchange rates are often smaller than 0.1 d 1, which can result in rising concentrations of potential harmful immissions inside of the display cases due to emissions from materials. Especially high concentrations of organic acids, which can emit from e.g. sealing materials, can produce damage of cultural objects. In 2012 BAM introduced a procedure witch is called: BEMMA-Scheme (Bewertung von Emissionen aus Materialien für Museumsausstattungen) which stands for: “Assessment of Emissions from Materials for Museum Equipment”. Micro chambers are used for VOC emission tests of display case construction materials, e.g. textiles, plastics, sealing material, coatings and others. Each sampling procedure is carried out in duplicate. Emissions like formic acid, acetic acid, formaldehyde and oximes are excluded and the sum of emissions of VVOCs, VOCs and SVOCs is limited. For a positive assessment all listed criteria must be fulfilled; otherwise the display construction material fails the BEMMA scheme. The BEMMA scheme is not a guarantee for an emission free display case, but a necessary requirement for the choice of suitable materials for emission and immission reduced display cases. Holzwerkstoffe, die unter den Prüfbedingungen der DIN EN 717-1 die Formaldehydklasse E1 einhalten, können insbesondere bei großflächigem Einsatz im Innenraum zu Überschreitungen des Richtwertes für die Innenraumluft führen. Zusätzlich zur Anpassung des Luftwechsels ist auch der Parameter Beladung bei einer Überarbeitung der Norm anzupassen. In der DIN EN 16516 werden produktspezifische (am Einsatz der Produkte orientierte) Beladungsfaktoren vorgegeben. Für den Einsatz von Holzwerkstoffplatten ergäben sich Beladungen von 0,4 (Boden oder Decke), 1,0 (Wände), 1,4 (Wände plus Boden oder Decke) und 1,8 m²/m³ (Wände, Boden und Decke). Um die realen Bedingungen in modernen Gebäuden abzubilden, ist deshalb eine Beladung von 1,8 m²/m³ für die Prüfkammermessung erforderlich. Hierbei ist darauf hinzuweisen, dass für die Beladung im Referenzraum Möbel nicht berücksichtigt wurden. Damit soll sichergestellt werden, dass die Einhaltung des Schutzniveaus bei realen Innenraumbedingungen auch unter den Randbedingungen eines niedrigen Luftwechsels sowie bei sommerlichen Bedingungen und heutigem Lüftungsverhalten der Menschen soweit wie möglich gewährleistet ist. Das Papier besteht aus Fasern meist pflanzlicher Herkunft und wird durch Entwässerung einer Fasersuspension auf einem Sieb gebildet. Der Abdruck des Schöpfsiebes (die Papierstruktur) und ein vorhandenes Wasserzeichen bieten Anhaltspunkte für Provenienz und Datierung; daneben erlaubt die Analyse der Fasern, der Füllstoffe und der Leimung eine weitere Charakterisierung des Schriftträgers. Spätestens seit dem Mittelalter sind es Eisengallustinten, die das schwarze bis bräunliche Erscheinungsbild vieler Manuskripte auf Papier hervorrufen. Daneben lassen sich auch schwarze Rußtuschen oder farbige Auszeichnungstuschen nachweisen. Eisengallustinten werden durch Mischung einer eisenhaltigen Komponente mit Gerbstoffen hergestellt. Eisensulfat ist die am häufigsten genannte Eisen liefernde Zutat, daneben sind aber auch eisenhaltige Minerale, Nägel oder Rost denkbar. Die Gallussäure leitet sich ab aus den Galläpfeln, der krankhaften Veränderung einzelner Planzenteile verschiedener Eichenarten. Diese werden durch die Eiablage von Schlupfwespen hervorgerufen. Wie schon beim Papier ermöglicht der materialanalytische Nachweis charakteristischer Beimischungen oder Verunreinigungen innerhalb der Tinten die Beantwortung kulturhistorischer Fragestellungen – gemeint sind hier die Unterscheidung von Original und Korrektur oder die Sichtbarmachung späterer Ergänzungen. T1 - Strategien für die Nutzung von Schlacken zukünftiger wasserstoffbasierter Stahlproduktionsrouten – Herstellung von Stahlwerksschlacke basierten Ausgangsstoffen für die Zementindustrie N2 - Die Europäische Kommission präsentierte am 11. Dezember 2019 den European Green Deal, ein Konzept mit dem Ziel, bis zum Jahr 2050 als erster Kontinent Klimaneutralität zu erreichen. Mit Inkrafttreten der Verordnung (EU) 2021/1119 des Europäischen Parlaments und des Europäischen Rates am 29. Juli 2021 zur Schaffung des Rahmens für die Verwirklichung der Klimaneutralität („Europäisches Klimagesetz“), ist dieses, sowie das Ziel der Reduzierung der Nettotreibhausgasemissionen um mindestens 55 % bis 2030 in der Gesetzgebung der Europäischen Union verankert. Die Stahlindustrie in der Europäischen Union mit ca. 57 Mio. t Nettotreibhausgasemissionen im Jahr 2020, die annähernd 2 % der Emissionen der Europäischen Union ausmachen (Inklusive Energie: 127 Mio. t; 4 %), bekennt sich zur erheblichen Reduzierung der CO2-Emissionen bis zum Jahr 2030 und zum Erreichen der Klimaneutralität bis zum Jahr 2050. Als Schlüsselwege zur Verwirklichung dieser Zielstellung hat sie zum einen Smart Carbon Usage (SCU) und zum anderen Carbon Direct Avoidance (CDA) in einer Roadmap formuliert. SCU umfasst CO2-reduzierende Maßnahmen in bestehenden Eisen- und Stahlproduktionsprozessen, Carbon Capture and Storage- (CCS) sowie Carbon Capture and Usage-Technologien (CCU). Wohingegen bei CDA CO2-Emissionen durch erneuerbare elektrische Energie sowie wasserstoffbasierte Reduktion des Eisenerzes gar nicht erst entstehen sollen. Der CDA-Weg, der voraussichtlich beschritten werden muss, um eine klimaneutrale Stahlproduktion zu erreichen [38], geht mit einer kostenintensiven Transformation der Stahlproduktion einher, die bereits von vielen Unternehmen der Stahlindustrie eingeleitet wurde und von der im Wesentlichen die beiden in Europa vorherrschenden Stahlproduktionsrouten Hochofen-Konverter-Route und Elektrolichtbogenofen-Route betroffen sind. Die CO2-Emissionen der Hochofen-Konverter-Route, von denen der überwiegende Anteil auf den Hochofen fällt, liegen laut einer für das Jahr 2015 erstellten Studie bei 1921 kg CO2/t Rohstahl inklusive der CO2-Last der Pellets als Einsatzmaterial sowie der Emissionen der nachgelagerten Stranggießanlagen und Walzwerke. Im Hochofen wird das Eisenerz durch Kohlenstoff (C) bzw. Kohlenstoffmonoxid (CO) – welche größtenteils aus dem Koks, der dem Hochofen zusammen mit dem Möller über die Gicht zugeführten wird, stammen – unter Entstehung von Roheisen und Kohlenstoffdioxid (CO2) reduziert. Das neben dem CO2 im Prozessgas enthaltene CO wird zum Beispiel zur Energiegewinnung in peripheren Prozessen ebenfalls unter Bildung von CO2 verbrannt. Zusätzlich zur Reduktion hat der Koks unter anderem physikalische Aufgaben, wie die Errichtung eines Stützgerüstes und die Gewährleistung der Permeabilität im Hochofenprozess. Somit ist eine vollkommene Substitution des Kokses zur Vermeidung der CO2-Emissionen im Hochofen nicht denkbar. Viele Strategien der Stahlunternehmen zum Erreichen der Klimaziele verfolgen daher den Wechsel von der Rohstahlproduktion aus der Hochofen-Konverter-Route hin zur Rohstahlproduktion im Elektrolichtbogenofen (EAF – Electric Arc Furnace) mit dem Einsatz von direkt reduziertem Eisen (Eisenschwamm, DRI – Direct Reduced Iron) aus vorgelagerten Direktreduktionsanlagen, die mit Wasserstoff (H2) als Reduktionsmittel betrieben werden können. Dabei ist die Elektrostahlerzeugung kein neuer Prozess und nimmt in der europäischen Union bereits jetzt einen Anteil von ca. 44 % der Rohstahlproduktion ein, jedoch überwiegend mit Schrott als Einsatzmaterial. Bei der Elektrostahlerzeugung mit Schrotteinsatz fallen laut der oben genannten Studie von Lüngen für das Jahr 20152 410 kg CO2-Emissionen pro Tonne Rohstahl an, von denen der Großteil auf die CO2-Last der fremd bezogenen elektrischen Energie zurückzuführen ist und daher nicht im Produktionsprozess selbst entsteht. Auch die Direktreduktion wird schon seit 1972 in der weltweit zweiten und bis heute in Westeuropa einzigen Direktreduktionsanlage in Hamburg betrieben. Während hier noch mit einem Reduktionsgas gearbeitet wird, das hauptsächlich aus einem aus Erdgas gewonnenen Wasserstoff-Kohlenstoffmonoxid-Gemisch besteht, liegt eine aktuelle Kernherausforderung in der Bereitstellung des in der Größenordnung der von der Transformation betroffenen Rohstahlproduktionskapazitäten notwendigen, mit erneuerbarer elektrischer Energie hergestellten Wasserstoffs. Wird zusätzlich der Strom für das Einschmelzen im Elektrolichtbogenofen aus erneuerbaren Quellen gewonnen und werden schließlich als Brennstoff verwendete Kohle und als Brennstoff verwendetes Erdgas durch CO2-neutrale Bio-Brennstoffe ersetzt, sind sowohl für den Einsatz von mit Wasserstoff reduziertem und auf Pellets ohne CO2-Last basiertem Eisenschwamm als auch für den Einsatz von Schrott CO2-Emissionen von jeweils nur 60 kg CO2/t Rohstahl erreichbar, was eine Reduktion der Emissionen, ausgehend von der Konverter-Hochofen-Route um 97 % und ausgehend von der Schrott-Elektrolichtbogenofen-Route um 85 % bedeuten würde. Der Betrieb von Elektrolichtbogenöfen macht die an den Standorten vorhandenen Konverter sowie die Infrastruktur für Behandlung und Transport des Roheisens überflüssig. Die fortwährende Nutzung dieser bestehenden Anlagenstruktur dient daher unter anderem als Motivation für die Entwicklung einer zweiten Transformationsstrategie, die einen sogenannten Einschmelzer (SAF – Submerged Arc Furnace) als der Direktreduktionsanlage nachgelagerten Prozess vorsieht, in dem wiederum Roheisen erschmolzen wird, das anschließend im Konverter zu Rohstahl veredelt wird. Die Transformation der Stahlindustrie beeinflusst einen weiteren CO2-intensiven Industriesektor, nämlich die Zementindustrie die verantwortlich ist für rund 4 % der CO2-Emissionen in der EU. Etwa zwei Drittel dieser Emissionen entfallen als Prozessemissionen auf die Entsäuerung des Rohstoffs Kalkstein (CaCO3), also auf die Abspaltung von CO2 unter Entstehung des für die Herstellung von Zementklinker notwendigen Branntkalks (CaO) als Zwischenstufe bei der Bildung der hydraulischen Phasen Tri- und Dicalciumsilikat. Als eines der fünf Themenfelder (5C-Ansatz: Clinker, Cement, Concrete, Construction, Carbonation), welche die europäische Zementindustrie entlang ihrer Wertschöpfungskette zum Erreichen der gesetzlichen Vorgaben der europäischen Union erarbeitet hat, umfasst das Feld "Cement" zum größten Teil die Reduzierung des Klinker-Zement-Faktors. Im Jahr 2017 betrug er 77 %, das heißt, dass 23 % des Klinkers durch alternative Materialien ersetzt wurden. Bis zum Jahr 2030 wird ein Faktor von 74 % und bis zum Jahr 2050 von 65 % angestrebt. Den größten Anteil an den Klinkersubstituten hat mit 33 % der Hüttensand aus der Stahlindustrie. Bei der Roheisenproduktion im Hochofen entsteht als Nebenprodukt Hochofenschlacke, die unter Zugabe von Schlackebildnern die Gangart des Möllers und mineralische Koksbestandteile abbindet, sowie weitere unerwünschte Begleitelemente wie Schwefel und Alkalien aufnimmt. Allein in Deutschland wurden im Jahr 2021 6,90 Mio. t – das entspricht 90 % der gesamten produzierten Hochofenschlacke – zu glasig erstarrtem Hüttensand granuliert. Dieser Hüttensand besitzt latent hydraulische Eigenschaften und wird daher seit mehr als 140 Jahren als Zementbestandteil eingesetzt, wodurch in Deutschland jährlich rund 4,5 Mio. t CO2-Emissionen eingespart werden. Alle Transformationsstrategien der Stahlindustrie beinhalten die notwendige Außerbetriebnahme der CO2-intensiven Hochöfen. Dies hat unter anderem zur Folge, dass die Hochofenschlacke als Nebenprodukt der Rohstahlerzeugung im Hochofen nicht mehr zur Verfügung stehen wird, wodurch die weitere Reduzierung des Klinker-Zement-Faktors erhebliche Zusatzmengen alternativer Materialien benötigen wird. Derzeit ist noch unklar, ob z.B. calcinierte Tone im benötigten Umfang erzeugt werden können. Der Fachbereich Thermochemische Reststoffbehandlung und Wertstoffrückgewinnung der Bundesanstalt für Materialforschung und -prüfung (BAM) und das Institut für Baustoffforschung FEhS forschen zusammen mit Industriepartnern und weiteren Forschungseinrichtungen im Rahmen zweier Projekte in den Kooperationssphären der Stahl- und Zementindustrie, die zur Bewältigung dieser Herausforderungen beitragen. Im Projekt SlagCEM wird die Produktion eines hydraulischen Zementbestandteils durch Reduktion von Konverter- bzw. Linz-Donawitz-Schlacke (LDS) untersucht. Die mit den zusätzlich aufgenommen Lichtbogenofenkapazitäten steigenden Mengen an Elektrolichtbogenofenschlacke (EOS, engl. electric arc furnace slag, EAFS) werden im Projekt DRI-EOS behandelt, welches die Produktion eines latent hydraulischen Hüttensandsubstituts zum Ziel hat. The disadvantage of their metabolic activity (e.g. sulfate reduction) results in the formation of toxic sulfide that leads to microbial influenced corrosion. SRB have been responsible for biocorrosion of ferrous metal. One of mitigation strategy is the use of biocides. However, it has been shown that various bacteria develop antimicrobial resistance due to excessive use of biocides. Thus, a deeper understanding of the evolution of biocide resistance of SRB is necessary. Three commonly used biocides, THPS, BAC, and GLUT were applied to investigate the susceptibility of Desulfovibrio alaskensis G20.The minimum inhibitory and bactericidal concentration and the killing kinetics of the three biocides was determined. These results will be used to conduct evolution experiments to determine the evolution of resistance towards biocides of SRBs. The outcome of this work can be helpful to improve the management of MIC treatments. Die Objekte werden mit nicht-invasiven bildgebenden Verfahren untersucht, in dem die Wechselwirkung zwischen Röntgen-, sichtbarem und infraroten Licht und den Zeichnungen studiert wird. Dier Analysen liefern nicht nur generell Erkenntnisse über die Art und Zusammensetzung und damit das Alter der verwendeten Farbmittel. Durch die Auffindung von Vorzeichnungen, die Bestimmung von Überarbeitungen und Korrekturen werden Erkenntnisse über die Genese von Zeichnungen gewonnen. Auf der Basis dieser materialwissenschaftlichen Erkenntnisse werden die Begriffe Original, Kopie, Fälschung diskutiert. As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. While liquid chromatography tandem mass spectrometry (LC-MS/MS) is commonly used technique to characterize targeted PFAS in environmental samples, there are more than 10,000 different PFAS known, which have various headgroups and properties. Therefore, several analytical techniques are available to analyse various groups or pools of PFAS or “all” PFAS as a sum parameter. Current decontamination strategies of PFAS-burdened soils mainly consist of adsorption methods using adsorbents for fixation of PFAS in the ground. A second option is the utilization of a “pump and treat” process, cycling polluted soils through a washing plant leading to the concentration of the pollutants in the fine fraction. Both approaches are cost-intensive and not intended for the direct decomposition of all PFAS contaminants. Hence, there is a great demand for innovative developments and chemical treatment technologies, dealing with new strategies of tackling the PFAS problem. Previously, mechanochemical treatment of polychlorinated organic compounds in soils showed an efficient dechlorination. Thus, we investigated mechanochemical treatment of PFAS contaminated soils with various additives in a ball mill and analyzed the PFAS defluorination with gas chromatography mass spectrometry (GC-MS) and liquid chromatography tandem mass spectrometry (LC-MS/MS), respectively, as well es the fluoride mineralization by ion chromatography (IC) and fluorine K-edge X-ray absorption near-edge structure (XANES) spectroscopy. Lately also silverfish have gained importance. Protection of our cultural heritage for future generations is not a new task but the methods have changed. Conventional use of pesticides in the past has preserved precious objects up to the presence, which otherwise probably would have been lost over time due to the destruction by insects and microbes. However, several of the objects were contaminated with poisons in such a manner that they became unsafe to be handled and thus are useless for exhibitions or scientific studies. Today, the concept of Integrated Museum Pest Management (IMPM) provides guidance for non-residue treatments of infested objects, followed by save storage or display under pest free (or maximum pest reduced) environments. Inert fumigants, physical and biological control measure, precise monitoring, threshold evaluations and detailed knowledge of the pest`s biology are key elements within IMPM. Woodworms, clothes moths and termites, however, are not always considered as only pests. Some professionals have also used them to produce new pieces of art or for the installation of ephemeral displays. Under more or less controlled conditions, the insects´ destructive nature on wood or textiles has been turned around to a process of creation with aesthetic or philosophical messages. The emergence of sociality in termites was associated with the acquisition of a diverse range of social structures. Previous work has found evidence for a caste-specific social defence system in termites leading to an immune system that may favour group over individual defence. While preliminary work suggests a correlation between social transitions and a reduction of immune gene family diversity, the lack of available high-quality termite genomes hampers complete knowledge of the true diversity of immune gene evolution across termite phylogeny. Here, we report on the sequencing and near-chromosome level assembly of 48 high-quality long-read-based genomes across major termite and cockroach sister-branch lineages. Defence strategies against such harmful threats led to a variety of molecular mechanisms, ecological shifts, and genetic innovations. Gene families underlying the molecular basis of the immune responses have evolved within the boundaries given by the species ecology. Here, we explore the evolution of some emblematic immune gene families throughout the Insecta phylum shedding light on remarkable genetic events such as gene duplication or gene loss. We followed a workflow based on the Hidden Markov model to search for immune genes in 55 high-quality genomes of insects. We highlight the emergence of group defence in social species as an ecological shift that released selection pressure on immune gene families notably in Blattodea. Further, we draw attention to certain gene families and the link between their diversity and the specificities of the species’ microbiota. Overall, we report data on immune gene diversity in insects. AKH acts by binding to a rhodopsin-like G protein-coupled receptor (GPCR), the adipokinetic hormone receptor (AKHR). In this study, we tackle AKH ligand and receptor gene evolution as well as the evolutionary origins of AKH gene paralogues from the order Blattodea (termites and cockroaches). Phylogenetic analyses of AKH precursor sequences point to an ancient AKH gene duplication event in the common ancestor of Blaberoidea, yielding a new group of putative decapeptides. In total, 16 different AKH peptides from 90 species were obtained. Two octapeptides and seven putatively novel decapeptides are predicted for the first time. AKH receptor (AKHRs) sequences from 18 species, spanning solitary cockroaches and subsocial woodroaches as well as lower and higher termites, were subsequently acquired using classical molecular methods and in silico approaches employing transcriptomic data. Aligned AKHR open reading frames revealed 7 highly conserved transmembrane regions, a typical arrangement for GPCRs. Phylogenetic analyses based on AKHR sequences support accepted relationships among termite and cockroach lineages to a large extent, while putative post-translational modification sites do not greatly differ between solitary and subsocial roaches and social termites. This study provides important information not only for AKH and AKHR functional research but also for further analyses interested in their development as potential candidates for biorational pest control agents against invasive termites and cockroaches. Diese Produkte werden aus einem Kunststoff mit relativ komplexer Zusammensetzung hergestellt. Um eine Aussage über die Haltbarkeit von KDB aus PE-HD treffen zu können, muss das Materialverhalten berücksichtigt werden. In diesem Zusammenhang sind die wichtigsten Aspekte der Dauerhaftigkeit die Spannungsrissbeständigkeit und die Beständigkeit gegen Oxidation. Verschiedene PE-Sorten weisen eine unterschiedliche Spannungsrissbeständigkeit auf. Um dieser Eigenschaft von PE-HD zu begegnen, werden nur KDB aus sehr hochwertigen Rohstoffen zugelassen. Auf der anderen Seite werden hohe Anforderungen an die umgebenden Schichten, wie Auflager und Schutzschicht gestellt, um möglichst geringe Spannungen zu erzeugen und damit Dehnungen die Dehnungen der KDB gering zu halten. Bestimmte Medien, die bei den betreffenden Prüfungen verwendet werden, dürfen nicht mehr in der EU in Verkehr gebracht werden. Im Folgenden sollen die Zusammenhänge in Kürze dargestellt werden. It is mainly used in road construction, earthwork and hydraulic engineering. A smaller part is returned to the metallurgical circle, used as fertilizer or landfilled. With this use, iron oxides still contained in steelmaking slag are lost. In addition, the possibility of producing higher-grade products from steelmaking slag is foregone. In recent decades, many researchers have investigated the production of Portland cement clinker and crude iron from basic oxygen furnace slags (BOFS) via a reductive treatment. Carbothermal treatment of liquid BOFS causes a reduction of iron oxides to metallic iron, which separates from the mineral phase due to its higher density. Simultaneously, the chemical composition of the reduced slag is adapted to that of the Portland cement clinker. In this study, German BOFS was reduced in a small-scale electric arc furnace using petrol coke as a reducing agent. The resulting low-iron mineral product had a similar chemical composition to Portland cement clinker and was rich in the tricalcium silicate solid solution alite (Ca3SiO5). Based on its chemical and mineralogical composition, similar to that of Portland cement clinker, the reduced BOFS has the potential to react comparably. In our study, the reduced BOFS produced less heat of hydration compared to OPC and its hydraulic reaction was delayed. However, adding gypsum has been shown to accelerate the hydration rate of the reduced BOFS compared to that known from the calcium silicates of Portland cement clinker. Further research to improve the hydraulic properties of the reduced slag is essential. If successful, producing a hydraulic binder and crude iron from BOFS could have economic and ecological benefits for both the cement and steel industry. These organisms either fix these pigments in their cell wall or secrete them into their extracellular environment to protect themselves against an array of physicochemical stresses (e.g., UV irradiation, desiccation, ...). Melanin can adsorb metals like Fe. How this affects fungal uptake of Fe and deterioration of Fe-containing minerals and metals is however less known. To study this, we use the model fungi Knufia petricola A95, a rock-inhabiting fungus known to deteriorate minerals and have melanised cell walls, and Amorphotheca resinae, able to contaminate fuel tanks, secrete melanin and corrode metals. In K. petricola, we have deleted genes involved in melanin production and Fe uptake using CRISPR/Cas. Through comparison of the geochemical signatures of these gene deletion mutants with those of the wild type (WT), we explore the specific mineral/metal deterioration mechanisms of melanised fungi. Fe isotope signatures of the biomass of melanin- and Fe uptake-deficient mutants of K. petricola revealed that Fe adsorbed either directly onto melanin or after being reduced by Fe reductases. Importantly, once adsorbed to melanin, Fe could not be mobilised and taken up into the cell: both the WT and its melanin-deficient mutant, previously grown at Fe replete conditions, showed similar growth at Fe deficient conditions. Olivine dissolution experiments revealed that Fe oxidation inhibits dissolution. K. petricola was able to enhance dissolution when this inhibition is strongest (at pH 6) and prevented dissolution when this inhibition is weakest (at pH 4). The fungus therefore dissolves olivine by interacting with the oxidised Fe at the olivine surface. However, Fe uptake did not seem to be involved: mutants deficient in various Fe uptake mechanism dissolved olivine at the same rate as the WT. This indicates that Fe adsorption onto melanin might play a key role. This is also shown by K. petricola’s ability to enhance olivine dissolution even further if secreting a melanin precursor and A. resinae’s corrosion of carbon steel whilst secreting melanin. Combined, our results imply that the Fe adsorbed to melanin cannot be taken up but enables fungi to deteriorate Fe-containing substrates at a higher rate. A1 - Rurack, Knut T1 - Dipstick coated with polystyrene-silica core-shell particles for the detection of microbiological fuel contamination N2 - Microbial contamination of fuels by fungi or bacteria poses risks such as corrosion and fuel system fouling, which can lead to critical problems in refineries and distribution systems and has a significant economic impact at every stage of the process. Many factors have been cited as being responsible for microbial growth, like the presence of water in the storage tanks. In fact, only 1 % water in a storage system is sufficient for the growth of microorganisms like bacteria or yeasts, as well as for the development of fungal biomass at the oil/water interface. This work presents a rapid test for the accurate determination of genomic DNA from aqueous fuel extracts. The detection is based on the use of polystyrene-mesoporous silica core-shell particles onto which modified fluorescent molecular beacons are covalently grafted. These beacons contain in the hairpin loop a target sequence highly conserved in all bacteria, corresponding to a fragment of the 16S ribosomal RNA subunit. The designed single-stranded molecular beacon contained fluorescein as an internal indicator and a quencher in its proximity when not hybridized. Upon hybridization in presence of the target sequence, the indicator and the quencher are spatially separated, resulting in fluorescence enhancement. To perform the assay the developed particles were deposited on different glass fibre strips to obtain a portable and sensitive rapid test. The assays showed that the presence of genomic DNA extracts from bacteria down to 50–70 μg L–1 induced a fluorescence response. The optical read-out was adapted for on-site monitoring by fitting a 3D-printed case to a conventional smartphone, taking advantages of the sensitivity of the CMOS detector. Such embedded assembly enabled the detection of genomic DNA in aqueous extracts down to the mg L–1 range and represents an interesting step toward on-site monitoring of fuel contamination. These systems have become particularly widespread since the Industrial Revolution, i.e., since the 17th century. At the same time, these technical systems – buildings, monuments, energy production, transformation and transmission, water purification and supply systems - serve as new habitats for living organisms. Life is ubiquitously present on our planet since a very long time: the Earth is 4.54 billion years old and microbial communities have played a key role on our planet for 3.7 billion years. Once human-made system appeared, microorganisms became an integral part of all types of technosphere infrastructure as well. Here we will illustrate biosphere-technosphere interactions using a specific example of the black fungi and their impact on the efficiency of solar (photovoltaic) panels. This expanding renewable infrastructure for electricity generation is growing on all continents - and create a specific, arid habitat for stress-tolerant black fungi. Black fungi were once discovered in hot and cold natural deserts – and now belong to the persistent colonisers of human-made deserts of solar parks. This new niche is evolving an impressive biodiversity. So far more than 60 isolates of black fungi belonging to Arthoniomycetes, Eurotiomycetes and Dothideomycetes were obtained from solar panels in Europe and Americas. Here we will present the analysis of this emerging anthropogenic biodiversity. Opportunities for future research in the field include quantification of the microbial load on technosphere surfaces – along with characterisation of the corresponding microbial diversity. The strategy of precise measurement and characterisation will enable us to reliably determine the beneficial and harmful functions that living microorganisms play in the functioning of energy-generating systems – and technosphere in general. T1 - Generation of Cryomyces antarcticus mutants to explore the importance of DHN melanin for survival N2 - Cryomyces antarcticus, a cryptoendolithic melanized fungus endemic to Antarctica (phylum Ascomycota, class Dothideomycetes incertae sedis), has demonstrated high capability to survive extreme environmental conditions like those found in space (e.g., ionizing radiation, vacuum, microgravity), thus fueling fundamental astrobiological questions like “the search for life beyond Earth”. Its extraordinary resilience has been attributed to the presence of thick, highly melanized cell walls, which may contain both DHN and DOPA melanins. To better understand the contribution of DHN melanin to the overall resilience of C. antarcticus, we decided to generate melanin-deficient mutants by genetic engineering. For this, the melanin-PKS (polyketide synthase)- encoding ortholog was identified in the C. antarcticus CBS 116301 genome and used to design primers for re-sequencing of the capks1 locus in the strain CCFEE 515. Based on the genetic toolkit developed for the black fungus Knufia petricola, we designed a strategy for mutating capks1. Protoplasts of C. antarcticus CCFEE 515 were generated and co-transformed with a circular AMA-containing plasmid for expression and in-vivo assembly of Cas9 and two capks1-specific sgRNAs and a PCR-generated donor DNA i.e., a hygromycin resistance cassette flanked by ~75-bp-long sequences homologous to the capks1 locus. Transformation of C. antarcticus is challenging because of its very slow growth and uncertain gene copy number. It took six months from obtaining enough biomass for cell wall lysis to transferring the putatively resistant transformants for genotyping, but we eventually managed to generate three independent non-melanized (whitish) Δcapks1 mutants! We are now awaiting first complemented strains as ultimate genetic transformation control. They are considered to have a wild-type-like pigmentation due to the reintroduction of the wild-type capks1 into the Δcapks1 mutant. The plastic fragments are home to a diverse community of microorganisms, and while researchers have gained a better understanding of these marine plastic biofilms, we lack insights into the physiology and genomic potential of the bacteria that colonize them. Examining the lifestyle of plastic colonizers from the GPGP is particularly intriguing, as the high concentration of plastics in this accumulation zone might allow for a microbial adaptation to this unique man-made habitat. A range of pigmentation was observed in 67 isolated strains obtained directly from plastic pieces sampled from the GPGP surface water. Sequence comparison between the cultivated bacterial strains and the 16S rRNA gene amplicon dataset confirmed that most of the cultivates could also be captured through DNA-only methods. Whole genome analysis of four taxonomically diverse representatives revealed multiple carotenoid pathways, including those to produce less common glycosylated carotenoids, like sarcinaxanthin glucoside. Further, we identified a potentially new Rhodobacteraceae species containing a photosynthetic gene cluster (PGC). Absorption analysis confirmed the actual production of the carotenoids and bacteriochlorophyll a. Floating plastics represent a habitat with strong UV-light exposure, making the protection with antioxidant carotenoids as well as the ability to use light as an energy source highly beneficial traits for plastic colonizers. Our findings indicate, that the production of pigments is a common adaption mechanism for plastic-associated bacteria, and that plastic biofilms present a so far overlooked source of rare carotenoids and light-harvesting mechanisms. There are two possibilities to explore a genome and its gene functions. The hypothesis-based method proves the function of already existing gene/allele candidates by targeted mutagenesis - so called reverse genetics. The basis of forward genetics approaches is the random mutagenesis of the genome, followed by screening of obtained mutants for the phenotype of interest, and identification of the mutated genes in the respective mutants. This strategy is hypothesis-generating, means it is necessary to verify the relationship between the detected mutations and the observed phenotype by targeted mutagenesis of the identified gene. We developed a toolbox for editing the genome of the rock inhabitant Knufia petricola [Eurotiomycetes, Chaetothyriales] that allows the study of the phenotypic characteristics of black fungi such as the regulation of pigment synthesis, general stress responses, oligotrophy, and the unusual modes of cell division by advanced reverse and forward genetics approaches. The toolbox includes the annotated genome sequence of strain A95, efficient strategies for CRISPR/Cas9-based genome editing and live-cell imaging using genetically encoded fluorescent proteins, as well as protocols for -omics approaches and for simulation of mineral weathering. A forward genetics approach using transposon mutagenesis is currently developed for identifying essential genes. The established protocols and knowledge gained from K. petricola form a starting point for making other fungi from extreme environments accessible to genetic manipulation. The polyketide synthases (PKS) release the heptaketide YWA1, the hexaketide AT4HN or the pentaketide T4HN. The first two products are deacetylated by ‘yellowish-green’ hydrolases to T4HN, and T4HN is further converted by a core set of enzymes to DHN. Final polymerization steps are accomplished by multicopper oxidases. DHN melanogenesis is often regulated in a spatial and temporal fashion resulting e.g., in melanized reproduction and survival structures of the foliar plant pathogen Botrytis cinerea (Schumacher 2016, Mol Microbiol). In contrast, a polyphyletic group of Ascomycetes (microcolonial fungi/ black yeasts) dwelling in hostile habitats such as bare rock surfaces in hot and cold deserts, exhibits constitutive melanogenesis. Here, DHN melanin builds a protective layer around all vegetative cells thus contributing to the survival of diverse environmental stresses even without specialized reproduction structures. For studying the relevance of constitutive DHN melanogenesis for tolerance of abiotic and biotic stresses, adhesion to substrates and subsequent damage of colonized surfaces, the rock-inhabiting fungus Knufia petricola was chosen as gene functions in this fungus can be studied by CRISPR/Cas9-based genome editing. The putative melanogenic genes were identified in the genome of K. petricola, deleted to confirm their involvement in DHN melanogenesis and co-expressed in Saccharomyces cerevisiae for reconstruction of the synthesis pathway. Phenotypes of DHN-deficient mutants are studied. Here, we will discuss the role of the DHN melanin layer on the outer cell wall in tolerating UV irradiation. To promote genetic studies, CRISPR/Cas9-based genome editing was implemented in the rock-inhabiting fungus Knufia petricola (Eurotiomycetes/ Chaetothyriales). Now efficient targeted mutagenesis of K. petricola - as a representative of the polyphyletic group of black fungi - enables the elucidation of extremotolerance, oligotrophism, unusual types of cell division, mineral weathering and symbiotic interactions. Even more progress on assigning functions to yet unknown genes can be achieved by a forward genetics approach. We chose the two-component Activator/Dissociation (Ac/Ds) transposon system from maize for generating K. petricola insertional mutants by in-vivo mutagenesis. For the optimal use of this genetic tool, an inducible promoter i.e, from the metabolism-independent Tet-on system, was combined with the AcTPase-coding sequence enabling the regulatable transposition of the resistance cassette-containing Ds transposon. In total, six auxotrophic Ac/Ds starter strains containing the Ds transposon at different position of ade2, ura3 or ppt1 were generated. The cultivation of these strains with doxycycline for induction of TET::Ac and subsequent selection of cells on ADE/URA/LYS-lacking media resulted in prototrophic colonies (revertants) for most Ac/Ds strains. Amplicon sequencing of excision sites revealed characteristic footprint patterns, proving that the transposon jumped. For identifying unknown Ds re-insertions sites, the thermal asymmetric interlaced (TAIL)-PCR was successfully implemented. First identified Ds re-insertion sites suggest that the distribution pattern may depend on the excision site. Currently, transposition frequencies and genome-wide distribution of re-insertion sites are studied in different Ac/Ds starter strains to identify the best candidate for generating saturated mutant libraries. This transposon mutagenesis strategy is also interesting for studying other black fungi, because once the Ac and Ds components are integrated in the genome, the fungus ‘only’ needs to be cultivated for generating insertional mutants. The emergence of sociality in the termites was associated with the acquisition of a diverse range of social structures and differential expression of specific gene network. Previous work has found evidence for a caste-specific social defence system in termites leading to an immune system that may favour group over individual defence. While preliminary work suggests a correlation between social transitions and a reduction of immune gene family diversity, the lack of available high-quality termite genomes hampers complete knowledge of the true diversity of immune gene evolution across termite phylogeny. Here, we report on the sequencing and assembly of 50 high-quality long-read-based genomes and 180 caste- and sex-specific brain transcriptomes across major termite and cockroach sister-branch lineage. We investigate the diversity and evolutionary history of immune genes across genomes, and the potential gene networks that have evolved with the emergence of termite sociality and some aspect of immune related behaviours. Um diese Schwermetalle aus den Materialien zu entfernen, werden drei thermochemische Verfahren zur Aufarbeitung von Gichtgasschlamm, Elektroofenstaub und ähnlichen Stahlwerksstäuben vorgestellt. Zum einen kann die Mischung von Elektroofenstaub mit chloridhaltigen Lösungen, wie sie z. B. in der Titandioxidproduktion anfallen, gemischt werden, um bei der thermischen Behandlung Schwermetallchloride zu effektiv verdampfen. Weiterhin kann der im Gichtgasschlamm enthaltene Kohlenstoff genutzt werden, um in Mischungen aus Gichtgasschlamm und Elektroofenstaub bei der thermischen Behandlung als Reduktionsmittel zu wirken. Dieses Verfahren führt zur Rückgewinnung von Zink und metallischem Eisen und wird im Technikums- und Pilotmaßstab erprobt. Abschließend wird die Nutzung von Wasserstoff als Reduktionsmittel für Elektroofenstaub vorgestellt. Hierbei lässt sich der Reststoff potenziell klimaneutral in Eisenschwamm umwandeln, wobei die chemischen Grundlagen anhand von DTA-Experimenten und thermodynamischen Simulationen ergründet werden.
