Hauptnutzen ist eine gegenüber der natürlichen Beanspruchung erhöhte Reproduzierbarkeit der Umweltbedingungen. Hat man sein Laborsetup entwickelt, ist es auf verschiedene Materialien anwendbar. Unter solchen Laborbedingungen ermittelte Lebensdauern sind dabei nicht auf die typischerweise sehr variablen Real-Umweltbedingungen übertragbar. (ii) Parametrisierter Ansatz - Ermittlung einzelner Materialempfindlichkeiten Hierbei werden im Labor die Wirkungen separater Umweltparameter auf Materialien nachgestellt. Für eine solche Separation der Einflussfaktoren ist insbesondere die Aufschlüsselung möglicher Wechselwirkungen der Umwelt-parameter (z.B. Mikroklima an bestrahlten Oberflächen) erforderlich. Einzelne (meist Alterungs-) Empfindlichkeiten können qualitativ nachgewiesen werden oder sogar – als Beanspruchungs-Wirkungs-Funktionen – quantifiziert werden, was einen wesentlichen Schritt in Richtung der Digitalisierung der Material¬prüfung darstellt. Insbesondere ist dann auch eine Lebensdauer-vorhersage für vorgegebene Zeitreihen der Beanspruchungs¬parameter umsetzbar. (iii) Rückwirkungen auf die Umwelt Umweltbeanspruchungen können zur Freisetzung von Schadstoffen in die Umwelt führen. Durch die Nachstellung kritischer, aber realitätsnaher Einsatzszenarien kann die Menge an freigesetzten Substanzen abgeschätzt werden. Egal, welcher Ansatz verfolgt wird – ein Vergleich mit der oder einer Real-beanspruchung ist unerlässlich, ebenso wie die Messdatenaufzeichnung (data logging) aller potenziell relevanten Beanspruchungsparameter während dieser Realbeanspruchung. The use of gaseous standards is necessary and a standard gas mixture containing 60 substances was successfully generated and employed for further investigations. The suitability of different chromatography columns was addressed: The use of PLOT (Porous Layer Open Tubular) columns is well suitable for VVOC analysis. The recoveries of the 60 analytes on several adsorbents and their combinations were determined: A combination of a graphitized carbon black and a carbon molecular sieve showed great results for all analytes. Carbon molecular sieves adsorb water which can impair the analysis. Different options such as purging, the use of a drying system or splitting were investigated for water removal. This contribution will present experimental results supporting the standardization of a method for VVOC analysis. At the same time, phosphorous and transition metals constitute highly valuable resources. Typically, separate pathways have been considered to extract hazardous transition metals or phosphate independently from each other. Investigations on the simultaneous removal of multiple components have been carried out only to a limited extent. Here, we report the synthesis routes for Ni- and Co-struvites (NH4MPO4·6H2O, M = Ni2+ and Co2+), which allow for P, ammonia, and metal co-precipitation. By evaluating different reaction parameters, the phase and stability of transition metal struvites as well as their crystal morphologies and sizes could be optimized. Ni-struvite is stable in a wide reactant concentration range and at different metal/phosphorus (M/P) ratios, whereas Co-struvite only forms at low M/P ratios. Detailed investigations of the precipitation process using ex situ and in situ techniques provided insights into the crystallization mechanisms/crystal engineering of these materials. M-struvites crystallize via intermediate colloidal amorphous nanophases, which subsequently aggregate and condense to final crystals after extended reaction times. However, the exact reaction kinetics of the formation of a final crystalline product varies significantly depending on the involved metal cation in the precipitation process: several seconds (Mg) to minutes (Ni) to hours (Co). The achieved level of control over the morphology and size makes precipitation of transition metal struvites a promising method for direct metal recovery and binding them in the form of valuable phosphate raw materials. Under this paradigm, the crystals can be potentially up-cycled as precursor powders for electrochemical or (electro)catalytic applications, which require transition metal phosphates. In theory, the uptake into the passive sampler is only dependent on the diffusion coefficient of the analyte in air and the geometry of the sampler (ideal adsorption). For characterization, the uptake rate (UR) is used, defined as the ratio of the uptaken mass and the product of ambient concentration and exposure time. Various reported effective uptake rates (UR,eff) differ considerably from ideal values (UR,id). Literature values in national and international standards, are essentially sorted by the applicable exposure time, while detailed information about the corresponding concentration range is lacking. Moreover the number of itemized substances is limited. The aim of this study was to review and expand this set of uptake rates by determination of own quality assured laboratory values.The determined uptake rates are considered accurate in terms of RSD and comparability to literature values and can be recommended for exposure times of seven days at 50 100 µg/m³ (approximately 100 300 ppm min). Seven day uptake rates in ISO16017 2 and ASTM D6196 are not generally suited for this purpose, as has been exemplarily shown for benzene. N2 - Die passive Probenahme von VOC (Volatile Organic Compounds) mit Tenax® TA und Thermodesorption GC/MS Analytik ist eine einfache, kosteneffiziente und repräsentative Methode zur Bestimmung der VOC-Konzentrationen in der Innenraumluft. In der Theorie ist die Stoffaufnahme in den Passivsammler nur abhängig vom Diffusionskoeffizienten der Substanz in Luft und der Geometrie des Sammlers (ideale Adsorption). Um sie zu charakterisieren, wird die Aufnahmerate (UR), definiert als Quotient der Massenaufnahme und dem Produkt aus Konzentration und Expositionszeit, verwendet. The European biocidal products regulation requires environmental risk assessments for biocidal products under service conditions. This is only possible, if processes that lead to biocide emissions into the environment are understood and can be reasonably predicted. Actual emissions due to leaching result from different processes that are affected by material properties and environmental conditions. Transformation of biocides can affect emissions considerably. Leaching and possible transformation of the film preservatives carbendazim, diuron, OIT and terbutryn in paints was observed under laboratory and field conditons in order to investigate the influence of different factors, e.g. water contact and radiation on biocide emissions. The paints contained either white titanium dioxide or a red iron oxide pigment to check whether pigments affect leaching processes. Methods. Test specimens of a water-based styrene acrylic dispersion paint on glass were exposed to UVA-radiation in a UV weathering device and/or exposed to water contact by immersion events on the basis of the European standard EN 16105. Plywood panels were coated and exposed to natural weather conditions in a semi-field experiment. Concentrations of biocides and transformation products were analysed in leachates from laboratory tests, runoff samples and remaining coatings. Results. The impact of weathering conditions on transformation processes differs for the investigated active substances. Water contact, radiation as well as pigments in the paints determine the pattern and amount of transformation products on coatings, leachates and runoff water. Observations from laboratory and field experiments were compared. It proved that results from laboratory tests support understanding of data from field experiments. Detailed knowledge on the fate of active substances under environmental conditions supports manufacturers to develop safe applications of film preservatives and authorities to evaluate its environmental impact. Die Verwendung gasförmiger Standards ist notwendig und es wurde ein stabiles Standardgasgemisch aus 60 Substanzen erfolgreich erzeugt und auf Stabilität und Reaktivität überprüft. Die Eignungsuntersuchung verschiedener Chromatographiesäulen ergab, dass PLOT-Säulen (Porous Layer Open Tubular) für die VVOC-Analytik gut geeignet sind. Für die 60 Analyten wurden Wiederfindungsraten auf mehreren Adsorbenzien und deren Kombinationen bestimmt. Dabei zeigte eine Kombination aus einem graphitierten Ruß und einem Kohlenstoff-Molekularsieb hervorragende Ergebnisse für alle Zielkomponenten. Nachteilig an Kohlenstoff-Molekularsieben ist, dass sie Wasser adsorbieren, was die Analyse beeinträchtigen kann. T1 - A Novel Dry Treatment for Municipal Solid Waste Incineration Bottom Ash for the Reduction of Salts and Potential Toxic Elements N2 - The main obstacle to bottom ash (BA) being used as a recycling aggregate is the content of salts and potential toxic elements (PTEs), concentrated in a layer that coats BA particles. This work presents a dry treatment for the removal of salts and PTEs from BA particles. Two pilotscale abrasion units (with/without the removal of the fine particles) were fed with different BA samples. The performance of the abrasion tests was assessed through the analyses of particle size and moisture, and that of the column leaching tests at solid-to-liquid ratios between 0.3 and 4. The results were: the particle-size distribution of the treated materials was homogeneous (25 wt % had dimensions <6.3 mm) and their moisture halved, as well as the electrical conductivity of the leachates. A significant decrease was observed in the leachates of the treated BA for sulphates (44%), chlorides (26%), and PTEs (53% Cr, 60% Cu and 8% Mo). The statistical analysis revealed good correlations between chloride and sulphate concentrations in the leachates with Ba, Cu, Mo, and Sr, illustrating the consistent behavior of the major and minor components of the layer surrounding BA particles. In conclusion, the tested process could be considered as promising for the improvement of BA valorization. All organisms must protect themselves from the harmful effects of light such as UV radiation, ROS accumulation, heat, and desiccation. Finally, light qualities and quantities can be used for decision making, timing and as guide for directed growth when they are sensed and transduced into intracellular signals. Botrytis cinerea and other plant pathogens infecting the sun-exposed parts of the plant must cope with the high light conditions the host plant seeks. Further they experience an altered light spectrum (‘green gap’) when they colonize shaded parts of the plant; it is depleted for blue and red light that is absorbed by the plant chlorophyll and enriched for green and far-red light that is reflected or transmitted by the plant tissue. As these ambient light conditions trigger the shade avoidance response in the plant, the pathogens may trigger their own ‘shading response’ such as the upregulation of virulence determinants and inoculum production. B. cinerea maintains a highly sophisticated light signaling machinery that senses different light qualities to trigger a variety of responses, that are protection, morphogenesis, positive and negative tropisms, and entrainment. These characteristics render B. cinerea a valuable model to enlighten the role of light in parasitic fungus-plant interactions and beyond. The vegetative mycelium – the core of all infection and developmental programs – is not visibly pigmented and thus considered to be sensitive to biotic and abiotic stresses. However, the vegetative hyphae have a very limited half-life and are usually restricted to the invasive growth phase in which they are protected from light by the plant tissue. Fast colonization of host tissues and by this proper nutrient acquisition enables the rapid formation of long-lasting reproduction structures (melanized conidiophores with conidia, sclerotia) on the surfaces of rotted plant tissues. Depending on the light and temperature conditions, conidiation or sclerotial development is initiated. But to cope with sunlight-associated stresses [e.g. high temperatures, UV radiation with associated DNA damage, accumulation of reactive oxygen species (ROS), desiccation and osmotic stresses] it is important for fungi to accurately sense and respond to changes in light. The genomes of black [dihydroxynaphthalene (DHN) melanin-containing] fungi from phyllosphere and exposed solid surfaces contain multiple photoreceptors (PRs). The plant pathogen Botrytis cinerea (Leotiomycetes) has a highly sophisticated photosensory and signalling system that helps to avoid light and to locate susceptible hosts. Rock-inhabiting Dothideomycetes and Eurotiomycetes including Knufia petricola possess equal numbers of PRs along with the same set of protective pigments. This similarity between black fungi from plant and rock surfaces suggests that photoperception and -regulation are important for fungi that receive nutrients through cooperation with phototrophs. T1 - Counting Small Particles in Electron Microscopy Images — Proposal for Rules and Their Application in Practice N2 - Electron microscopy (EM) is the gold standard for the characterisation of the morphology (size and shape) of nanoparticles. Visual observation of objects under examination is always a necessary first step in the characterisation process. Several questions arise when undertaking to identify and count particles to measure their size and shape distribution. In addition to challenges with the dispersion and identification of the particles, more than one protocol for counting particles is in use. This paper focuses on precise rules for the counting of particles in EM micrographs, as this influences the measurement accuracy of the number of particles, thus implicitly affecting the size values of the counted particles. We review and compare four different, commonly used methods for counting, which we then apply in case studies. The impact of the selected counting rule on the obtained final particle size distribution is highlighted. One main aim of this analysis is to support the application of a specific, well-defined counting approach in accordance with regulatory requirements to contribute to achieving more reliable and reproducible results. It is also useful for the new harmonised measurement procedures for determining the particle size and particle size distribution of nanomaterials. T1 - Test Guideline No. 125 - Nanomaterial Particle Size and Size Distribution of Nanomaterials N2 - The OECD Working Party on Manufactured Nanomaterials (WPMN) has actively worked towards understanding possible safety issues for manufactured nanomaterials and has contributed significantly to resolving these by developing Test Guidelines, Guidance Documents, Test Reports and other publications with the aim of a safe use of manufactured nanomaterials. To address the specific needs of manufactured nanomaterials, the OECD Test Guideline No. 110 “Particle Size Distribution/Fibre Length and Diameter Distributions” was identified as one of the test guidelines (TGs) to require an update. The current TG 110 (adopted in 1981) is only valid for particles and fibres with sizes above 250 nm. The WPMN prioritised to either update TG 110 to be applicable also to particles at the nanoscale or draft a new nanomaterial specific (TG). Eventually, it was decided to develop a new TG that covers the size range from 1 nm to 1000 nm, intended for particle size and particle size distribution measurements of nanomaterials. Paragraph 11 provides further justification on the need for such measurements for nanomaterials. This TG overlaps with TG 110 in the size range from 250 nm to 1000 nm. When measuring particulate or fibrous materials, the appropriate TG should be selected depending on the size range of particles tested. In line with TG 110, the new TG for nanomaterials includes separate parts for particles and fibres. For the part of this TG which addresses particles, several methods applicable to nanomaterials were reviewed and included to take into account developments since 1981 when the TG 110 was adopted. This TG includes the following methods: Atomic Force Microscopy (AFM), Centrifugal Liquid Sedimentation (CLS)/Analytical Ultracentrifugation (AUC), Dynamic Light Scattering (DLS), Differential Mobility Analysis System (DMAS), (Nano)Particle Tracking Analysis (PTA/NTA), Small Angle X-Ray Scattering (SAXS), Scanning Electron Microscopy (SEM), and Transmission Electron Microscopy (TEM). The method Single Particle Inductively Coupled Plasma Mass Spectrometry (sp-ICP-MS) could not be sufficiently validated within the interlaboratory comparison (ILC) carried out for the different methods in this TG (see also paragraph 6 for further details on the ILC). Applicability of sp-ICP-MS is strongly limited to nanomaterials with high mass values in combination with a sufficiently high particle size. However, the general method ICP-MS is widely used and the sp-mode for the size measurement of specific nanomaterials was successfully performed in ILCs elsewhere. The method is therefore included in the Appendix Part C of this TG, which further details the limitations of sp-ICP-MS. For measuring the diameter and length of fibres, analysing images captured with electron microscopy is currently the only method available. This TG includes Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM). To test the validity of this TG, an ILC was performed. Test materials were chosen to reflect a broad range of nanomaterial classes, e.g. metals, metal oxides, polymers and carbon materials. Where possible, well-characterised test materials were used. Additionally, the test materials were chosen to reflect a broad range of sizes representing the size range 1 nm to 1000 nm. Specifically for fibres, a broad range of aspect ratios was included (length/diameter of 3 to > 50). Some of the test materials used are commercially available and further references are given in the validation report of the ILC. Sample preparation for physical chemical characterisation is critical for all listed methods. Due to the differences between individual nanomaterials and due to the wide range of individual material properties it is impossible to have a generic protocol to obtain the best possible sample preparation for every nanomaterial. Therefore, a generic protocol on sample preparation is not part of this TG. Information on sample preparation is given in the paragraphs 25-29, 33, 34 and 39 for particles and in paragraphs 159) for fibres. Da die hervorragenden Bindemitteleigenschaften des bisher hauptsächlich in Zementen eingesetzten Portlandklinkers jedoch auf Calciumverbindungen wie Tri- und Di-calciumsilikaten basieren, stellt die Substitution des Kalks derzeit für die Zementindustrie die größte Herausforderung dar. Neben vollständig neuen Bindemittelsystemen ohne Verwendung von Kalk und CCS/CCU-Technologien zur Vermeidung der CO2-Emissionen bei Verwendung von Kalk, können Abfälle und Nebenprodukte wie etwa Betonbrechsand, verschiedene Schlacken und weitere Massenreststoffe sinnvoll zur Einsparung von Rohstoffen und zur Vermeidung von CO2-Emissionen eingesetzt werden. Es gilt diese Potentiale von Sekundärrohstoffen branchenübergreifend zu identifizieren und Synergien zwischen den Branchen nutzbar zu machen. Hierbei werden neben der chemischen Zusammensetzung der Abfälle und Nebenprodukte auch die Verfügbarkeit und der Massenstrom relevante Kriterien sein. Die potenziellen Sekundärrohstoffe und deren Gemische mit chemisch ähnlicher Zusammensetzung wie die bisher verwendeten Bindemittel, können durch verschiedene technische Ansätze wieder in reaktive Bindemittel umgewandelt und damit nutzbar gemacht werden. Eine gezielte Umwandlung der nicht reaktiven mineralischen Phasen in neue mineralische Phasen mit guten Bindemitteleigenschaften, kann u. a. durch Schmelzprozesse erzielt werden. In Schmelzen können bei hohen Temperaturen die bestehenden, nicht reaktiven mineralischen Phasen zerstört und neue mineralische Phasen mit guten Bindemitteleigenschaften erzeugt werden. Hohe Freiheitsgrade durch gezielte Einstellung der stofflichen Zusammensetzung der Schmelze, durch Stoffseparationen und durch geregelte Abkühlregimes können eine maßgeschneiderte Erzeugung neuer Bindemitteltypen ermöglichen. Aufgrund der hohen erforderlichen Temperaturen von Schmelzprozessen ist die Umsetzung dieser neuen Verfahren nach Möglichkeit in bestehenden pyrometallurgischen Produktionsrouten wie etwa der Stahlproduktion vorzunehmen. So kann etwa der Einsatz von Betonbrechsand als Schlackebildner in der Stahlproduktion zielführend sein, wenn die Schlacke wieder zu einem Bindemittel prozessiert wird und in Zementen Verwendung findet. Eine stoffliche Kreislaufführung wäre hiermit gegeben, wobei der Einsatz von Kalkstein als Schlackebildner oder Rohstoff für die Zementproduktion, und damit die entsprechenden CO2-Emissionen, vermieden werden. Die Entwicklung neuartiger Bindemittelsysteme auf Basis von Sekundärrohstoffen sollte von erforderlichen Anpassungen und neuen Entwicklungen in der Regelsetzung und Normung begleitet werden. Vor allem muss gewährleistet sein, dass die mit neuen Bindemitteln hergestellten Betone den hohen Anforderungen an Dauerhaftigkeit und Umweltverträglichkeit genügen. Des Weiteren sollte mit den etablierten Methoden, wie etwa der Lebenszyklusanalyse, untersucht werden, inwiefern und in welchem Umfang die neuen technischen Entwicklungen zu Nachhaltigkeit und Klimaschutz beitragen. The dynamic corrosion process is dependent on the conditions present in the DGR which are influenced and/or controlled by geochemical parameters (e.g., redox potential, pH, presence of and ionic concentration in (pore-)water), physical parameters (e.g., pressure), and the influence of metabolically active microorganisms. Cast iron corrosion will occur at the intersection of container and its decontaminable coating with the bentonite backfill material which contains natural microbial populations. The conditions in a DGR are simulated in microcosm experiments to investigate the impact of microbiologically influenced corrosion (MIC); the microcosms contain: B27 bentonite, synthetic pore water, N2 or N2-CO2 atmosphere, cast iron coupons, as well as the bacterium Desulfosporosinus burensis (isolated from repository depth in Buré, France). Three coupon configurations will be used: untreated, coated with decontaminable coating, and coated with decontaminable coating which has been damaged to simulate possible damages. The microcosms will be examined for bio- and geochemical parameters, such as pH, redox potential, mineral phases, sulphate concentration, Fe(II):Fe(III), changes in microbial populations, and the corrosion process for formation of corrosion products, and potential microbial influence, after a 270-day incubation period at 25°C under anaerobic conditions. Auch wenn in beiden Fällen Glas das Trägermaterial ist, bezeichnet Hinterglasmalerei jedoch eine völlig andere Technik. Während bei der Glasmalerei die Farben auf den Bildträger eingebrannt werden, handelt es sich bei der Hinterglasmalerei um eine Kaltmalerei. Selbsttrocknende Farben werden auf die Rückseite der Glastafel aufgetragen, zuweilen wird die Malweise durch aufwendige Veredelungstechniken ergänzt. Breeding success of K. caeruleus was low, but basic parameters of this species’ developmental cycle were identified. At 21 °C and 75 % relative humidity and a four-month cold period at 4 °C, the development of K. caeruleus from egg to adult appearance lasted 2 years. The pupal stage may be reached and completed after one and a half years. Feeding on larvae of A. punctatum by larvae of K. caeruleus was observed and consisted of a combination of sucking haemolymph and consuming body parts. The sickle-like mandibles of larvae of K. caeruleus penetrate the cuticle of prey larvae followed by pumping and sucking body movements. Adult beetles of A. punctatum were not attacked by K. caeruleus larvae. Feeding behavior of adult K. caeruleus was not investigated. One aspect of this includes self-medication, where infected individuals consume substances such as toxins or alter their macronutrient consumption to enhance immune competence. Another widely adopted animal response to infection is illness-induced anorexia, which is thought to assist host immunity directly or by limiting the nutritional resources available to pathogens. Here, we recorded macronutrient preferences of the global pest cockroach, Blatta orientalis to investigate how shifts in host macronutrient dietary preference and quantity of carbohydrate (C) and protein (P) interact with immunity following bacterial infection. Results: We fnd that B. orientalis avoids diets enriched for P under normal conditions, and that high P diets reduce cockroach survival in the long term. However, following bacterial challenge, cockroaches signifcantly reduced their overall nutrient intake, particularly of carbohydrates, and increased the relative ratio of protein (P:C) consumed. Surprisingly, these behavioural shifts had a limited efect on cockroach immunity and survival, with minor changes to immune protein abundance and antimicrobial activity between individuals placed on diferent diets, regardless of infection status. Conclusions: We show that cockroach feeding behaviour can be modulated by a pathogen, resulting in an illness-induced anorexia-like feeding response and a shift from a C-enriched to a more P:C equal diet. However, our results also indicate that such responses do not provide signifcant immune protection in B. orientalis, suggesting that the host’s dietary shift might also result from random rather than directed behaviour. The lack of an apparent beneft of the shift in feeding behaviour highlights a possible reduced importance of diet in immune regulation in these invasive animals, although further investigations employing pathogens with alternative infection strategies are warranted. As a result of the perpetual use of PFAS containing products, effluents and sewage sludge from wastewater treatment plants (WWTPs) have been observed to be an important pathway for PFAS into the environment. In Germany, phosphorus and other nutrients from sewage sludge and wastewater should be recycled in WWTPs of cities with a large population. However, it is not clear if PFAS contamination from wastewater and sewage sludge end up in novel wastewater-based fertilizers. Normally, PFAS are analyzed using PFAS protocols typically with time-consuming extraction steps and LC-MS-MS quantification. However, for screening of PFAS contaminations in sewage sludge or wastewater-based fertilizers also passive sampler based on the Diffusive Gradients in Thin-films (DGT) technique can be used for the PFAS extraction. Afterwards, combustion ion chromatography (CIC) can be applied to analyse the “total” amount of PFAS on the passive sampler. Here, we show results from the DGT method in comparison to those of the extractable organic fluorine (EOF) method for a variety of wastewater-based fertilizers. Altogether, there are currently more than 4700 known but only partly characterized fluorinated compounds identified by the Organization for Economic Cooperation and Development (OECD), as shown by a recently published classification proposal. The ongoing production of new, yet unrestricted PFAS alternatives has become a major challenge for environmental routine analytics, since the state-of-the-art method LC-MS/MS relies on structural information and availability of isotope standards of the targeted compound. First reported by Miyake et al., fluorine sum parameters like adsorbable organic fluorine (AOF), extractable organic fluorine (EOF) and total fluorine (TF) can be applied to survey and detect the presence of large amounts of unidentified organofluorine compounds in environmental matrices. Today fluorine sum parameters have been established as a useful supplement to classic target-analytical approaches of PFAS and were implemented for the first time as a sum value “PFAS-total” in the recently revised Drinking Water Directive (2020/2184) by the European Commission. In contrast, X-ray absorption near-edge structure (XANES) spectroscopy has been widely applied to identify low concentration of element-specific contamination without pre-treatment in environmental samples in the past. The XANES approach enables a penetration depth of approx. 1 µm (at fluorine K-edge energy; depending on the matrix) which is significantly deeper than for X-ray photoelectron spectroscopy (XPS; penetration depth approx. 10 nm). Furthermore, the method is fast, non-destructive and only simple preparation of the samples is required. In our work, we combined both F-XANES spectroscopy and combustion ion chromatography (CIC) in order to present a new analytical perspective on the investigation of PFAS in environmental media and material samples. T1 - A brief survey on open questions about laser-induced periodic surface structures N2 - The processing of laser-induced periodic surface structures (LIPSS) represents a simple and robust way for the nanostructuring of solids that allows creating a wide range of surface functionalities featuring applications in optics, tribology, medicine, energy technologies, etc. While the currently available laser and scanner technology already allows surface processing rates at the m2/min level, industrial applications of LIPSS are sometimes hampered by the complex interplay between the nanoscale surface topography and the specific surface chemistry. This typically manifests in difficulties to control the processing of LIPSS and in limitations to ensure the long-term stability of the created surface functions. This presentation aims to identify some unsolved scientific problems related to LIPSS, discusses the pending technological limitations, and sketches the current state of theoretical modelling. Here, the stabilizing extracellular biofilm matrix together with physiological changes on the single cell level leads to an increased resilience towards harsh environmental conditions, antimicrobials, the host immune response and established cleaning procedures. Persistent microbial adhesion on e.g., medical implants, in water supply networks or food-processing industry is often associated with chronic inflammation, nosocomial and foodborne infections, enhanced biofouling and product contamination. To prevent persistent microbial colonization, antibacterial surface strategies often target the initial steps of biofilm formation and impede adhesion of single cells before a mature biofilm is being formed. While chemical coatings have been widely used, their restricted biocompatibility for eukaryotic cells and attenuated antibacterial-effects due to compound release limit their areas of application and alternative strategies focus on modified surfaces topographies to impede bacterial adhesion. In this work, we used ns-UV laser treatment (wavelength 248 nm and a pulse duration of 20 ns) to generate laser-induced periodic surface structures (LIPSS) with different submicrometric periods ranging from ~210 to ~610 nm on commercial poly(ethylene terephthalate) (PET) foils. Following structurally and chemically analyses, PET samples were subjected to bacterial colonization studies with Escherichia coli TG1, a bacterial test strain with a strong biofilm formation capacity due to the formation of nanofiber-like cell-appendages (pili). Bacterial adhesion tests revealed that E. coli repellence decisively depends on the spatial periods of the LIPSS with the strongest reduction (~91%) in cell adhesion observed for LIPSS periods of 214 nm. Scanning electron microscopy and additional biofilm studies using a pili-deficient E. coli TG1 strain revealed the importance of extracellular appendages in the bacterial repellence observed here, thus, pointing out new antibiotics-free strategies for antibacterial surfaces by impeding nanofiber-mediated bacterial adhesion. Mutants deficient in melanin and/or carotenoid synthesis were grown as air-exposed biofilms. Extracted EPS were quantified and characterised using a combination of analytical techniques. The absence of melanin affected the quantity and composition of the produced EPS: mutants no longer able to form melanin synthesised more EPS containing fewer pullulan-related glycosidic linkages. Moreover, the melanin-producing strains attached more strongly to the mineral olivine and dissolved it at a higher rate. We hypothesise that the pullulan-related linkages, with their known adhesion functionality, enable fungal attachment and weathering. The released phenolic intermediates of melanin synthesis in the Δsdh1 mutant might play a role similar to Fe-chelating siderophores, driving olivine dissolution even further. These data demonstrate the need for careful compositional and quantitative analyses of biofilm-created microenvironments. Previous studies showed that de-novo mutagenesis and horizontal gene transfer (HGT) by conjugation or transformation – important processes underlying resistance evolution and spread - are affected by antibiotics, metals and pesticides. However, natural microbial communities are also frequently exposed to biocides used as material preservatives, but it is unknown if these substances induce mutagenesis and HGT. Here, we show that active substances used in material preservatives can increase rates of mutation and conjugation in a species- and substance-dependent manner, while rates of transformation are not increased. The bisbiguanide chlorhexidine digluconate, the quaternary ammonium compound didecyldimethylammonium chloride, the metal copper, the pyrethroid-insecticide permethrin, and the azole-fungicide propiconazole increase mutation rates in Escherichia coli, whereas no increases were identified for Bacillus subtilis and Acinetobacter baylyi. Benzalkonium chloride, chlorhexidine and permethrin increased conjugation in E. coli. Moreover, our results show a connection between the RpoS-mediated general stress and the RecA-linked SOS response with increased rates of mutation and conjugation, but not for all biocides. Taken together, our data show the importance of assessing the contribution of material preservatives on AMR evolution and spread. Vaccines' efficacy, in inducing immunity, varies in different societies due to economic, social, and biological conditions. One of the influential biological factors is gut microbiota. Cross-talks between gut bacteria and the host immune system are initiated at birth during microbial colonization and directly control the immune responses and protection against pathogen colonization. Imbalances in the gut microbiota composition, termed dysbiosis, can trigger several immune disorders through the activity of the adaptive immune system and impair the adequate response to the vaccination. The bacteria used in probiotics are often members of the gut microbiota, which have health benefits for the host. Probiotics are generally consumed as a component of fermented foods, affect both innate and acquired immune systems, and decrease infections. This review aimed to discuss the gut microbiota's role in regulating immune responses to vaccination and how probiotics can help induce immune responses against pathogens. T1 - Release of substances from joint grouts based on various binder types and their ecotoxic effects N2 - Background: The leaching of substances and the ecotoxic effects of eluates were studied for joint grouts that are based on various types of binders. Eight products, two of them containing either epoxy resin, polybutadiene or polyurethane binders, or modifed cement, were investigated using harmonized leaching tests for construction products in combination with ecotoxicity tests on algae, daphnia, luminescent bacteria, fish eggs and mutagenicity in accordance with CEN/TR 17105. In addition to basic parameters, such as pH, TOC, and inorganic components, organic substances in the eluates were analysed by gas and liquid chromatography in combination with mass spectrometry. Quantitative analyses in combination with ecotoxicity data on selected substances were used to deduce which substances cause the observed ecotoxic effects. Results: Different patterns of ecotoxic effects were observed in joint grouts with different binder types. The most ecotoxic effects were observed in epoxy resin-based products, followed by polybutadiene-based products. Fewer ecotoxic effects were observed in polyurethane-based products and modifed cements. Some of these showed no ecotoxicity. Some of the substances in the eluates were identified and related to ecotoxic effects. 4-Tert-butylphenol and amines probably contributed to the ecotoxic effects of at least one of the epoxy resin-based renders, whereas cobalt is assumed to contribute to the toxic effect on algae of one of the polybutadiene-based products. However, only some of the leached substances could be identifed, and only some of the ecotoxic effects can be explained by the available information on the composition of eluates and known ecotoxic profiles of the identified substances. Conclusions: Ecotoxicity tests on eluates from leaching tests indicate whether environmentally hazardous substances can be leached from construction products. Combined ecotoxicity tests and chemical analysis of eluates from EU-wide harmonized leaching tests for construction products can provide information on substances that cause these effects. This supports the identifcation and development of environmentally friendly construction products. This study confirmed that ecotoxicity tests in accordance with CEN/TR 17105 are a tool well-suited to support the implementation of the European Commission’s zero pollution vision for 2050 and to reduce pollution to levels no longer considered harmful to health and natural ecosystems. For this, a MIC-specific staining procedure was developed, which ensures the analysis of intact cells. It allows the analysis of archaea at a single cell level, which is extremely scarce compared to other well-characterized organisms. The detection method revealed elemental selectivity for the corrosive methanogenic strain Methanobacterium -affiliated IM1. Hence, the possible uptake of individual elements from different steel samples was investigated and results showed the cells responded at a single-cell level to the different types of supplemented elements and displayed the abilities to uptake chromium, vanadium, titanium, cobalt, and molybdenum from solid metal surfaces. The methods developed and information obtained will be used in the future to elucidate underlying mechanisms, compliment well-developed methods, such as SEM-EDS, and develop novel material protection concepts. These processes are impacted by pollution with antibiotics. However, biocides used as disinfectants and material preservatives are major pollutants by far excceding the market for antibiotics in terms of mass. Our work shows that biocides have the potential to affect evolutionary processes towards AMR by increasing the rates of de-novo mutation and conjugation. These effects depend on the species and biocidal substance. Importantly, chlorhexidine and quaternary ammonium compounds (QACs) affect rates of mutation and conjugation at environmentally relevant concentrations in E. coli. Moreover, our results show a connection between the RpoS-mediated general stress and the RecA-linked SOS response with increased rates of mutation and conjugation, but not for all biocides. Furthermore, our work highlights the potential of biocides to contribute to selection and transmission of AMR. We show that the application of biocides, especially QAC disinfectants, leads to the rapid evolution of tolerance (i.e. increased survival) in adaptive laboratory evolution (ALE) experiments. The evolved tolerant strains have a selective advantage in the presence of environmentally-relevant concentrations of antibiotics, which could lead to the stabilization of biocide tolerance in environments where biocides and antibiotics co-occur (e.g. wastewater, animal stables). ALE experiments with biocide tolerant strains indicate a decreased evolvability of resistance to antibiotics. Taken together, our work shows the importance of assessing the contribution of biocides on evolution, selection and transmission of AMR in the environment. Contact with water causes hydrolysis and leaching of substances. Ultraviolet radiation may induce photolysis. As a result, a mixture of biocidal active substances and transformation products can be emitted into the environment. In a semi-field study, leaching of the biocidal active substances terbutryn, diuron, octylisothiazolinone, carbendazim, and selected transformation products was observed for two paints containing either a white or a red pigment. Painted test panels were exposed to natural weathering for about 1.5 years. Runoff samples were analyzed during the course of the experiment. At the end of the study, residues of biocidal active substances and transformation products were determined in sections of the test panels. Emissions of substances were mainly observed during the first few months of the experiments. Increased emissions of transformation products were observed during periods of increased global radiation and subsequent periods with relatively high amounts of driving rain. As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. As a result of the perpetual use of PFAS containing products, effluents and sewage sludge from wastewater treatment plants (WWTPs) have been observed to be an important pathway for PFAS into the environment. In Germany, phosphorus and other nutrients from sewage sludge and wastewater should be recycled in WWTPs of cities with a large population. However, it is not clear if PFAS contamination from wastewater and sewage sludge end up in novel wastewater-based fertilizers. Normally, PFAS are analyzed using PFAS protocols typically with liquid chromatography tandem mass spectrometry (LC-MS/MS) quantification. To get a better overview of the amount of “total PFAS,” we applied sum parameter methods based on combustion ion chromatography (CIC) to screen the PFAS contaminations in various sewage sludge and wastewater-based fertilizers. Furthermore, current decontamination strategies of PFAS-burdened soils mainly consist of adsorption methods using adsorbents for fixation of PFAS in the ground. A second option is the utilization of a “pump and treat” process, cycling polluted soils through a washing plant leading to the concentration of the pollutants in the fine fraction. Only a subsequent, high-energy consuming pyrolysis process guarantees the total destruction of all fluorinated organic contaminants. Understanding plant strategies under current conditions is important to assess and predict responses of natural ecosystems to future climate and environmental changes. Nitrogen (N) and potassium (K) (re-)utilization from topsoil and their acquisition from subsoil and saprolite were evaluated in a continental transect, encompassing three study sites – an arid shrubland, a mediterranean woodland, and a temperate rainforest – on similar granitoid parent material in the Chilean Coastal Cordillera. The short-term (<1 year) plant N and K acquisition was traced with 15N and the K analogs rubidium and cesium. To do so, the tracers were either injected into topsoil, subsoil, or saprolite, in the immediate vicinity of eight individual plants per study site and injection depth. The long-term (>decades) K uplift by plants was investigated by the vertical distribution of exchangeable K+ and Na+. Recoveries of 15N and K analogs by arid shrubland plants were similar from topsoil, subsoil, and saprolite. Mediterranean woodland shrubs recovered the tracers primarily from topsoil (i.e., 89 % of recovered 15N and 84 % of recovered K analogs). Forest plants recovered the tracers from topsoil (15N = 49 %, K analogs = 57 %) and partially from greater depth: 38 % of recovered 15N and 43 % of recovered K analogs were acquired from subsoil and saprolite, respectively. Low nutrient accessibility in the topsoil (e.g., because of frequent droughts) drives shrubland plants to expand their N and K uptake to deeper and moister soil and saprolite. Woodland and forest plants dominantly recycled nutrients from topsoil. In the forest, this strategy was complemented by short-term uplift of N and K from depth. The vertical distribution of exchangeable K indicated long-term uplift of K by roots in all three sites. This highlighted that long-term K uplift from depth complements the nutrient budget across the continental transect. Insights into the painting technique along with art historical comparisons provided the opportunity for a better understanding of the painting’s genesis, in particular concerning an early reworking of the background by the artist. A recovery and reconstruction of the earlier version of the background is now possible. The discovery of zinc in distinct passages of the painting, as well as the grey-black pigment stibnite and glass particles used as a supplement in paint layers are of special interest. The technological investigation initiated a discussion about the circumstances of the revision, as well as the painting’s relation to Vasari’s Lives of Artists. Those natural microbial communities are frequently exposed to biocides used as material preservatives. Previous studies have shown that antibiotics, metals and pesticides affect the underlying processes of resistance evolution and spread; namely de novo mutagenesis and horizontal gene transfer by conjugation and transformation in microbial communities. However, it is unknown if active substances used in material preservatives are involved in these processes. We show that biocides used as material preservatives affect rates of mutation and conjugation in microorganism in a species- and substance-dependent manner, while rates of transformation are not directly affected. However, concerns have been raised about the possibility for the evolution and selection of resistance against disinfectants and AMCs. In turn, resistance against disinfectants and AMCs can be associated to antibiotic resistance due to cross-resistance - a single mechanism conferring resistance to a disinfectant and an antibiotic- and co-resistance - two distinct mechanisms physically linked on e.g. a plasmid. The risk for resistance and cross-resistance during use of biocides (including disinfectants and AMCs) must be evaluated during authorization according to the EU biocidal product regulation. However, to date there is a lack of standardized methods that support risk assessment during the authorization process. Methods We used adaptive laboratory evolution (ALE) experiments which are based on repeated exposure of bacteria to disinfectants or AMCs. The experiments are followed by phenotypic (antimicrobial susceptibility testing) and genotypic (whole genome sequencing) characterization of the evolved strains. The basic idea of these experiments is to expose bacteria to lethal conditions and select for mutants with increased survival. This approach is fundamentally different to other ALE experiments, which commonly select for increased growth at subinhibitory concentrations. However, selection for increased survival represents a selective pressure that more realistically reflects selection under in-use conditions of disinfectants and AMCs. Results First, we studied adaptation of E. coli during repeated disinfection with benzalkonium chloride in a suspension assay. The experiments showed a 2000-fold increase in survival within 5 exposure cycles. The adaptive changes are linked to highly parallel mutations in genes related to lipid A biosynthesis, less negative cell surface charge, reduced growth rate and increased competitive ability in the presence of certain antibiotics. We use the same approach to develop standardizable ALE experiments based upon accepted standards that are used to determine the efficacy of disinfectants (EN 13697) and antimicrobial surfaces (ISO 22196). The results highlight pronounced adaptation of different test strains towards surface disinfection (benzalkonium chloride and isopropanol) and AMCs (copper). Conclusion Bacteria can adapt with increased survival towards lethal stress imposed by disinfectants and AMCs. These exposure pathways need to be investigated and included in risk assessments to safeguard water quality and promote the sustainability of construction materials. The applied materials, exposure conditions, distribution routes and resilience of receiving compartments vary considerably. This demonstrates the need for a consistent concept that integrates knowledge of emission sources, leaching processes, transport pathways, and effects on targets. Such a consistent concept can serve as the basis for environmental risk assessment for several scenarios using experimentally determined emissions. Typically, a source–path–target concept integrates data from standardized leaching tests and models to describe leaching processes, the distribution of substances in the environment and the occurrence of substances at different points of compliance. This article presents an integrated concept for assessing the environmental impact of construction products on aquatic systems and unravels currently existing gaps and necessary actions. Biocides are antimicrobials used as disinfectants, antiseptics and preservatives. They find application on a large scale in the industrial and medical sector, but also in private households. In terms of mass, the worldwide use of biocides exceeds that of antibiotics. Thus, despite their important role in preventing the spread of pathogens, due to their ubiquity, biocides are suspected to be drivers of the antimicrobial resistance crisis. In our work at BAM we try to understand how biocides contribute to the emergence of AMR, what the underlying adaptation principles and mechanisms are and how they compare to those found for antibiotics. Within our group, I mainly focus on the following two questions: How does phenotypic heterogeneity in bacteria affect the ability to survive treatment with biocides? And what are the consequences of phenotypic heterogeneity for the evolution of resistance to biocides and antibiotics? I will share published and unpublished results which demonstrate that phenotypic heterogeneity can enable the survival of biocide treatment and, through this, facilitate the evolution of AMR. Im Falle des Portlandklinkers besteht die Schwierigkeit darin, dass zwei Drittel der CO2-Emissionen rohstoffbedingt durch die Entsäuerung des Kalksteins (CaCO3) anfallen, die durch die beschriebenen Maßnahmen nicht weiter abgesenkt werden können. Vielmehr wird es erforderlich sein, noch zu entwickelnde oder zu optimierende Technologien anzuwenden und ggf. alle relevanten Ebenen wie Produktion, Weiterverarbeitung und Anwendung von Zement und Beton neu zu denken. Da die hervorragenden Bindemitteleigenschaften des bisher hauptsächlich in Zementen eingesetzten Portlandklinkers jedoch auf Calciumverbindungen wie Tri- und Di-calciumsilikaten basieren, stellt die Substitution des Kalks derzeit für die Zementindustrie die größte Herausforderung dar. Neben vollständig neuen Bindemittelsystemen ohne Verwendung von Kalk und CCS/CCU-Technologien zur Vermeidung der CO2-Emissionen bei Verwendung von Kalk, können Abfälle und Nebenprodukte wie etwa Betonbrechsand, verschiedene Schlacken und weitere Massenreststoffe sinnvoll zur Einsparung von Rohstoffen und zur Vermeidung von CO2-Emissionen eingesetzt werden. Es gilt diese Potentiale von Sekundärrohstoffen branchenübergreifend zu identifizieren und Synergien zwischen den Branchen nutzbar zu machen. Hierbei werden neben der chemischen Zusammensetzung der Abfälle und Nebenprodukte auch die Verfügbarkeit und der Massenstrom relevante Kriterien sein. Die potenziellen Sekundärrohstoffe und deren Gemische mit chemisch ähnlicher Zusammensetzung wie die bisher verwendeten Bindemittel, können durch verschiedene technische Ansätze wieder in reaktive Bindemittel umgewandelt und damit nutzbar gemacht werden. Eine gezielte Umwandlung der nicht reaktiven mineralischen Phasen in neue mineralische Phasen mit guten Bindemitteleigenschaften, kann u. a. durch Schmelzprozesse erzielt werden. In Schmelzen können bei hohen Temperaturen die bestehenden, nicht reaktiven mineralischen Phasen zerstört und neue mineralische Phasen mit guten Bindemitteleigenschaften erzeugt werden. Hohe Freiheitsgrade durch gezielte Einstellung der stofflichen Zusammensetzung der Schmelze, durch Stoffseparationen und durch geregelte Abkühlregimes können eine maßgeschneiderte Erzeugung neuer Bindemitteltypen ermöglichen. Aufgrund der hohen erforderlichen Temperaturen von Schmelzprozessen ist die Umsetzung dieser neuen Verfahren nach Möglichkeit in bestehenden pyrometallurgischen Produktionsrouten wie etwa der Stahlproduktion vorzunehmen. So kann etwa der Einsatz von Betonbrechsand als Schlackebildner in der Stahlproduktion zielführend sein, wenn die Schlacke wieder zu einem Bindemittel prozessiert wird und in Zementen Verwendung findet. Eine stoffliche Kreislaufführung wäre hiermit gegeben, wobei der Einsatz von Kalkstein als Schlackebildner oder Rohstoff für die Zementproduktion, und damit die entsprechenden CO2-Emissionen, vermieden werden. Die Entwicklung neuartiger Bindemittelsysteme auf Basis von Sekundärrohstoffen sollte von erforderlichen Anpassungen und neuen Entwicklungen in der Regelsetzung und Normung begleitet werden. Vor allem muss gewährleistet sein, dass die mit neuen Bindemitteln hergestellten Betone den hohen Anforderungen an Dauerhaftigkeit und Umweltverträglichkeit genügen. Sophisticated mechanical methods for unrolling the best-preserved scrolls have been applied, with varying success. However, such processes have been abandoned, to prevent risk from irremediable damage or loss and to preserve the integrity of the extremely fragile rolls. Following the development of X-ray based non-invasive techniques, attempts to virtually unroll the scrolls were made. The most common ink in Antiquity was carbon-based, and the main element of carbonized papyrus is carbon, making these investigations difficult. However, some attempts with synchrotron X-ray phase-contrast tomography (XPCT) were successful. Recently, the identification of antique inks containing metals raised hope that if some of the inks contain metal the rolls can be virtually unrolled using conventional CT- technique. We investigated the inks of a selection of partially unrolled fragments stored at the Biblioteca Nazionale di Napoli with X-ray fluorescence in order to select the best candidates for tomography. Despite the many difficulties (analysis of several layers sticking together, letters barely visible, difficulty to separate contribution from the ink and from the papyrus, inhomogeneity of the support, fragility of the fragments…), encouraging results were found, with a number of inks from Greek fragments found to contain additions to the soot (Fe, Pb, Cu P). Um die Phosphorversorgung für Pflanzen in der Landwirtschaft zu gewährleisten, werden Phosphordünger eingesetzt. Die Nährstoffe werden hauptsächlich durch Wirtschaftsdünger (Gülle, Jauche, Stallmist) zurückgeführt. Zusätzlich werden in der konventionellen Landwirtschaft mineralische Phosphordünger eingesetzt, die vorwiegend aus dem fossilen Rohstoff Phosphorit gewonnen werden. Um die Abhängigkeit von fossilen Rohstoffen zu reduzieren, sollen nährstoffhaltige Rest- und Abfallstoffe zur Phosphordünger aufbereitet werden. In Abwasserkläranlagen fällt phosphorreicher Klärschlamm als Abfallstoff an. Der größte Anteil des Klärschlammes wird in Steinkohlekraftwerken, Zementwerken oder in Monoklärschlammverbrennungsanlagen verbrannt. Die Klärschlammaschen aus den Monoverbrennungsanlagen enthalten bis zu 12 Gew.-% Phosphor und sind daher für die Düngerherstellung geeignet. Ein geeignetes Verfahren zur Aufbereitung der Klärschlammaschen ist die thermochemische Behandlung im Drehrohrofen. Mit dem sogenannten AshDec®-Prozess, kann ein Phosphordünger hergestellt werden. Aktuell wird eine großtechnische Anlage in Altenstadt (Bayern) geplant, die im Jahr 2023 den Betrieb aufnehmen soll. Der zentrale Bestandteil ist die Phasenumwandlung von schlecht pflanzenverfügbaren Phosphaten in der Klärschlammasche (vorwiegend das Calciumphosphat Whitlockit und Aluminiumphosphat) zu gut pflanzenverfügbaren Calciumalkaliphosphaten im Produkt. Um dies zu erzielen, werden die Aschen mit Natrium- und/oder Kalium-Verbindungen gemischt und einer thermochemischen Behandlung bei 800-1000 °C zugeführt. Um die Pflanzenverfügbarkeit und Düngewirkung von Phosphaten abschätzen zu können, ist die chemische Extraktionsmethode mit neutraler Ammoniumcitratlösung geeignet. In der Dissertation werden die (Phosphor-)Phasenreaktionen und die Prozessbedingungen des thermochemischen Verfahrens untersucht, um das Verfahren gezielt zu modifizieren und die Wirtschaftlichkeit zu steigern. Die Zielphasen der thermochemischen Behandlung sind die Calciumalkaliphosphate CaNaPO4 und CaKPO4, und deren Mischphasen Ca(Na,K)PO4, welche in der Publikation 3.1 synthetisiert wurden. Für diese Calciumalkaliphosphate wurden die thermodynamischen Daten der Standardbildungsenthalpie, Standardentropie, Wärmekapazität und die Wärmemenge der Phasenumwandlung bestimmt (Publikation 3.1). Bei der thermochemischen Behandlung von Klärschlammaschen wurden als Additive Natrium- und Kaliumsulfat bei verschiedenen Temperaturen getestet. Der Einsatz von Kaliumadditiven ist erwünscht, um den Marktwert des Produktes durch die Produktion eines Phosphor-Kalium-Düngers zu erhöhen. Zusätzlich wird eine möglichst geringe Prozesstemperatur angestrebt, bei der die erwünschten Calciumalkaliphosphate gebildet werden. In den Publikationen 3.2 und 3.3 wird gezeigt, dass für Klärschlammaschen eine komplette Phasenumwandlung zu Calciumnatriumphosphaten ab 875 °C in Laborversuchen (Korundtiegel) mit dem Additiv Natriumsulfat erzielt werden kann. Der Einsatz von Kaliumsulfat erforderte notwendige Reaktionstemperaturen von über 1100 °C. Diese unterschiedlichen Reaktionstemperaturen hängen mit den Schmelzpunkten von Natriumsulfat (890 °C) und Kaliumsulfat (1070 °C) zusammen. Um Kaliumsulfat in den Prozess bei niedrigen Temperaturen zu integrieren, wurden Natriumsulfat und Kaliumsulfat vor der Behandlung gemischt. Dies führte zu einer Schmelzpunkterniedrigung und resultierte in Reaktionstemperaturen zwischen 900 °C bis 1000 °C für die untersuchten Mischungen von Natrium- und Kaliumsulfat (Publikation 3.3). Die Zusammensetzung der Calciumalkaliphosphate Ca(Na,K)PO4 war anders als erwartet. Bei der Phasenanalytik stellte sich heraus, dass bei geringem und mittlerem Anteil von Kaliumsulfat im Alkali-Additiv nur Calciumnatriumphosphate mit sehr geringen Kaliumgehalten gebildet wurden. Erst bei einem hohen Anteil von Kaliumsulfat in der Mischung der Alkalisulfate konnten kaliumhaltige Calciumalkaliphosphate nachgewiesen werden (u.a. (Ca0.9Mg0.1)(Na0.6K0.4)PO4). Dieser geringere Einbau von Kalium in die Calciumalkaliphosphate hängt mit den zusätzlich stattfindenden Reaktionen zwischen den Alkalien und den Silikaten zusammen. Es ist bekannt, dass die zugegebenen Alkalisulfate zuerst mit Silikaten und anschließend mit den Phosphaten reagieren. Silikate bauen bevorzugt Kalium ein, deswegen konnten erst kaliumhaltige Phosphate gebildet werden, wenn die Reaktion mit den Silikaten abgeschlossen war. Dies führte zu einem geringeren Kaliumanteil in den gebildeten Phosphaten im Vergleich zum Kaliumanteil der verwendeten Alkaliadditive. Das in Silikaten gebundene Kalium ist wahrscheinlich schlecht für die Pflanzen verfügbar. Dies könnte den Einsatz der produzierten Phosphor-Kalium-Dünger einschränken. Der bevorzugte Kaliumeinbau in Silikaten konnte auch in Kalkulationen mit den thermodynamischen Daten aus der Publikation 3.1 gezeigt werden. Mittels Phasenanalytik vor und nach der chemischen Extraktion, konnte die Zugehörigkeit zu verschiedenen Modifikationen der Calciumalkaliphosphaten sicher bestimmt werden und eine ungefähre Zusammensetzung dieser Calciumalkaliphosphate abgeschätzt werden. So konnte nachgewiesen werden, dass die Modifikation vom CaNaPO4 ungefähr 10 % Magnesium einbauen kann. Wenn mehr Magnesium eingebaut wird, bildet sich die Phase (Ca,Mg)NaPO4, die eine vergleichbare Struktur wie die bekannte Phase (Ca0.72Mg0.28)NaPO4 aufweist. Dieses magnesiumreiche Calciumnatriumphosphat entstand vermutlich, wenn entweder erhöhte Anteile an Kalium eingebaut wurden (Ca0.8Mg0.2)(Na0.85K0.15)PO4 (Publikation 3.3) oder die Phasenumwandlung vom Calciumphosphat Whitlockit zum Calciumalkaliphosphat CaNaPO4 noch nicht abgeschlossen war (Publikation 3.2). Wenn mehr Kalium eingebaut wird, dann entsteht eine Phase (u.a. (Ca0.9Mg0.1)(Na0.6K0.4)PO4) ähnlich zu den Mischphasen aus der Publikation 3.1. In den Publikationen 3.4 und 3.5 wurden keine Klärschlammaschen untersucht, sondern Biokohlen aus Modellklärschlämmen (Publikation 3.5) und Biomasseaschen (Publikation 3.4) aus der Vergasung oder Verbrennung einer Mischung aus Zuckerrohrbagasse und Hühnertrockenkot. Bisher wird dieses Potential der Klärschlammaschen für die Phosphor-Rückgewinnung jedoch kaum genutzt, da die in den Aschen enthaltenen Phosphate für Pflanzen kaum verfügbar und die Aschen teilweise mit toxischen Schwermetallen belastet sind. Im AshDec®-Verfahren wird die Klärschlammaschen im Drehrohrofen im Temperaturbereich 800-1000°C unter Zugabe von Alkali-Additiven (bsp. Na2CO3) thermochemisch behandelt, um ein wirksames und schadstoffarmes Phosphatdüngemittel herzustellen. Aktuell bereitet die Firma Emter GmbH den Bau der ersten großtechnischen AshDec®-Anlage mit einer Kapazität von 30.000 Jahrestonnen Klärschlammasche am Standort ihrer Klärschlammverbrennungsanlage (Altenstadt / Oberbayern) vor. Herbal extracts obtained from resin of Vateria copallifera (Retzius) Alston and seeds of Madhuca longifolia (Konig) Macbride have been used for centuries to preserve e.g. palm leaf manuscripts from insect attack. Herbal extractions of these traditional products for palm leaf manuscript were tested for their effect against the termite species Coptotermes formosanus Shiraki, 1909. Natural and artificial aged herbal extractions were tested to obtain a repellent index. Resin oil of V. copallifera caused slightly higher repellencies than M. longifolia. Artificially aged samples produced lower repellencies than naturally aged samples. The results indicate that the active ingredients are volatile. The potential for barrier treatment was tested only with V. copallifera. Tunnelling-behavior of C. formosanus workers through sand in the presence of V. copallifera resin oil was largely reduced. BOFS is mostly used in road construction, earthwork and hydraulic engineering. However, in this use, the iron bound in BOFS is lost and the opportunity to produce higher value products from BOFS is forgone. In recent decades, many researchers have investigated a thermochemical process to reduce iron oxides to metallic iron in molten BOFS. The metallic iron formed separates from the reduced slag due to its higher density and can be recovered. An advantage of this process is that simultaneously the chemical composition of the reduced slag is adapted to that of the Portland cement clinker and the hydraulic reactive mineral alite is formed. In this study, BOFS was reduced in a small-scale electric arc furnace using petrol coke as reducing agent, and the hydraulic properties of the reduced, low-iron BOFS were investigated. Despite a chemical and mineralogical composition similar to that of Portland cement clinker, the reduced BOFS produced less heat of hydration, and its reaction was delayed compared to Portland cement. However, the addition of gypsum, as is also done in cement production from Portland cement clinker, has been found to accelerate the hydration rate of reduced BOFS. Further research to improve the hydraulic properties of the reduced slag is essential. If successful, the production of a hydraulic binder and crude iron from BOFS could have economic and ecological benefits for both the cement and steel industry. In this use, the iron bound in the steelmaking slags (< 30 wt.%) is lost. Recovery of iron from steelmaking slags is possible by thermochemical reductive treatment. The reductive treatment of liquid steelmaking slags causes iron oxides to be reduced to metallic iron, which separates from the mineral phase due to its higher density. The chemical composition of the mineral phase is thus adapted to that of the Portland cement clinker and the mineral alite, the most important component of Portland cement, is formed. The same techniques with sorbent-based active sampling, thermal desorption and gas chromatography coupled with mass spectrometry (TD-GC/MS) should be used. However, VVOCs require gaseous standards, an adapted GC column and a reliable sampling adsorbent. This work presents experimental results to tackle those three experimental gaps. A stable standard gas mixture containing 47 VVOCs, 13 VOCs and an internal standard was successfully generated. It was employed to study the suitability of seven types of chromatography columns. The use of PLOT (Porous Layer Open Tubular) columns such as PoraBOND Q is well suitable for VVOC analysis. The recoveries of the 60 analytes on a total of 16 adsorbents and their combinations were determined: A combination of the graphitized carbon black Carbograph 5TD 40/60 and the carbon molecular sieve CarbosieveTM SII showed great recoveries for all analytes. Carbon molecular sieves adsorb water which can impair the analysis. A dry purge of the multi-sorbent in the sampling direction led to a complete water removal and promising recoveries of the analytes. Despite being in use since the 1940s, there remain multiple open questions regarding their detailed mode-of-action and the mechanisms, including phenotypic heterogeneity, that can make bacteria less susceptible to QACs. To facilitate studies on resistance mechanisms towards QACs, we synthesized a fluorescent quaternary ammonium compound, namely N-dodecyl-N,N-dimethyl-[2-[(4-nitro-2,1,3-benzoxadiazol-7-yl)amino]ethyl]azanium-iodide (NBD-DDA). NBD-DDA is readily detected by flow cytometry and fluorescence microscopy with standard GFP/FITC-settings, making it suitable for molecular and single-cell studies. As a proof-of-concept, NBD-DDA was then used to investigate resistance mechanisms which can be heterogeneous among individual bacterial cells. Our results reveal that the antimicrobial activity of NBD-DDA against Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa is comparable to that of benzalkonium chloride (BAC), a widely used QAC, and benzyl-dimethyl-dodecylammonium chloride (BAC12), a mono-constituent BAC with alkyl-chain length of 12 and high structural similarity to NBD-DDA. Characteristic time-kill kinetics and increased tolerance of a BAC tolerant E. coli strain against NBD-DDA suggest that the mode of action of NBD-DDA is similar to that of BAC. As revealed by confocal laser scanning microscopy (CLSM), NBD-DDA is preferentially localized to the cell envelope of E. coli, which is a primary target of BAC and other QACs. Leveraging these findings and NBD-DDA‘s fluorescent properties, we show that reduced cellular accumulation is responsible for the evolved BAC tolerance in the BAC tolerant E. coli strain and that NBD-DDA is subject to efflux mediated by TolC. Overall, NBD-DDA’s antimicrobial activity, its fluorescent properties, and its ease of detection render it a powerful tool to study resistance mechanisms of QACs in bacteria and highlight its potential to gain detailed insights into its mode-of-action. However, because of the continuous use of fluorinated consumer products, also effluents and sewage sludge from wastewater treatment plants (WWTPs) have been shown to be an important source of PFAS contamination into the aquatic environment. Resulting from recent stricter regulations and restrictions in the last years on the use of long chain (≥C8) PFAS, there is a significant shift in the chemical industry towards production of short (C4-C7) and ultrashort (C1-C3) chain alternatives. With the amendment of the Sewage Sludge Ordinance in 2017 the German legislation banned sewage sludge application on agricultural land, and by 2029/2032 sewage sludge will be completely prohibited from agricultural application. While environmental exposure of organic pollutants like PFAS, pesticides and pharmaceuticals are no longer desirable, phosphorus (P) from sewage sludge must still be used to produce high-quality P-fertilizers for a circular economy. However, most deoxyribonucleic acid (DNA) viruses employ DNA polymerases (Pol) capable of exonucleolytic proofreading to limit mutation rates during DNA replication. The relative genetic stability produced by high-fidelity genome replication can make studying DNA virus adaptation and evolution an intensive endeavor, especially in slowly replicating viruses. Here, we present a proofreading-impaired Pol mutant (Y547S) of Marek’s disease virus that exhibits a hypermutator phenotype while maintaining unimpaired growth in vitro and wild-type (WT)-like pathogenicity in vivo. At the same time, mutation frequencies observed in Y547S virus populations are 2–5-fold higher compared to the parental WT virus. We find that Y547S adapts faster to growth in originally non-permissive cells, evades pressure conferred by antiviral inhibitors more efficiently, and is more easily attenuated by serial passage in cultured cells compared to WT. Our results suggest that hypermutator viruses can serve as a tool to accelerate evolutionary processes and help identify key genetic changes required for adaptation to novel host cells and resistance to antiviral therapy. Similarly, the rapid attenuation achieved through adaptation of hypermutators to growth in cell culture enables identification of genetic changes underlying attenuation and virulence, knowledge that could practically exploited, e.g. in the rational design of vaccines. Antimicrobial resistance (AMR) is a global health problem and the environment is an important compartment for the evolution and transmission of AMR. Soil is an environment with a large reservoir of natural microbial communities and antimicrobial resistance (AMR) genes. Those natural microbial communities are frequently exposed to biocides used as material preservatives. Previous studies have shown that antibiotics, metals and pesticides affect the underlying processes of resistance evolution and spread; namely de novo mutagenesis and horizontal gene transfer by conjugation and transformation in microbial communities. However, it is unknown if active substances used in material preservatives are involved in these processes. We show that biocides used as material preservatives affect rates of mutation and conjugation in microorganism in a species- and substance-dependent manner, while rates of transformation are not directly affected. Our data highlights the importance of assessing the contribution of material preservatives on AMR evolution and spread in the environment. Im Rahmen des Forschungsprojektes »Kennerschaft heute« wurde der Bestand der über 1.500 Zeichnungen erstmals wissenschaftlich erschlossen. Das Handbuch führt anhand bedeutender Werkgruppen, darunter Zeichnungen von Lucas van Leyden, Dirck Vellert, Roelant Savery und Rembrandt, den Reichtum der Weimarer Sammlung vor Augen, die zum großen Teil auf die Sammelleidenschaft von Johann Wolfgang Goethe zurückgeht. T1 - Role of light in the life cycle of Botrytis cinerea N2 - The fungus Botrytis cinerea (Botryotinia fuckeliana) infects more than 500 plant species and causes a wide range of symptoms: soft rots, accompanied by collapse and water-soaking of tissues followed by the appearance of gray masses of conidia on leaves and soft fruits (gray mold), and spots that may turn brown to full-scale soft rotting on flower petals (Botrytis blight). In general, B. cinerea is responsible for severe economic losses that are either due to the damage of growing plants in the field or the rot of harvested fruits, flowers, and vegetables during storage under cold and humid conditions. B. cinerea has adapted to the plant host and its environment by evolving strategies to use plant tissues for proliferation in terms of a necrotrophic lifestyle, and to survive biotic stresses (host responses) as well as abiotic factors of the host’s environment such as sunlight and concomitant stresses. B. cinerea maintains a complex regulatory network of light-sensitive proteins and signal transduction pathways to use light for coordinating stress responses, virulence, and reproduction. Different light-controlled reproduction cycles enable B. cinerea to live in moderate climate zones by infecting and propagating in summer and resting in winter when green host tissues are unavailable. This topic is reviewed here with an emphasis on the following questions that remain unanswered: What is the quantitative contribution of bacteria and fungi to weathering? What are the associated mechanisms and do they leave characteristic imprints on mineral surfaces or in the geological record? Does biogenic weathering fulfill an ecological function, or does it occur as a side effect of unrelated metabolic functions and biological processes? An overview of efforts to integrate the contribution of living organisms into reactive transport models is provided. Here we show how both colonisation and stress resistance are linked to the properties of the fungal cell surface. By deleting genes involved in the synthesis of melanin and carotenoid pigments of the model rock-inhabiting fungus Knufia petricola via CRISPR-Cas and comparing the behaviour of the gene-deletion mutants with the wild type (WT), we studied the role of these genes in mineral colonisation and stress sensitivity. The extracellular polymeric substances (EPS) of biofilms of the WT and mutants were extracted, quantified and chemically characterised. We observed that the absence of melanin affected the quantity and composition of the produced EPS: melanin-deficient mutants synthesised more EPS containing fewer pullulan-related glycosidic linkages. Moreover, in mineral dissolution experiments, these mutants showed a lower ability to colonise the mineral olivine. We hypothesise that not melanin, but the pullulan-related linkages enabled the melanin-producing strains to attach more strongly to the mineral. Moreover, ICP-OES analysis of the aqueous mineral-derived solutes showed that biofilms of the K. petricola mutants which could attach were able to dissolve the olivine faster than those that could not. The same mutants were also characterised by their sensitivity to desiccation stress: only the mutant deficient in both melanin and carotenoid synthesis was more sensitive to desiccation compared to the WT, indicating that a combination of both pigments is critical to withstand desiccation. Overall, these results show the critical role of the cell surface in the specific capacities of rock-inhabiting fungi. Hierbei wurden insbesondere bei den Fugenmörteln und Korkgranulaten sehr hohe Ökotoxizitäten (bis LID = 16384 bei den Fugenmörteln und LID = 24578 bei den Korkgranulaten) beobachtet. Algen- und Leuchtbakterientests waren in der Regel deutlich empfindlicher als der Daphnien- und Fischeitest. Die untersuchten Dachbahnen, Lacke, Wood-Plastic-Composites und Schaumglasschotter zeigten hingegen keine oder nur sehr geringe Ökotoxizitäten. Zur Qualitätssicherung und Validierung wurde ein Europäischer Ringversuch gemäß den Vorgaben der DIN ISO 5725 organisiert, durchgeführt und ausgewertet. The environment has been regocnized as an improtant compartment for the occurance, evolution and transmission of AMR. Biocides used as material preservatives are in contact with the environment and natural microbial communities through direct application and passive leaching from protected materials. It has been shown that environmental contaminants, such as antibiotics, metals and pesticides, can affect resistance evolution and spread by modifying the underlying processes of de novo mutagenesis, horizontal gene transfer and selection. However, it is unknown if material preservatives are involved in these processes and thereby can also drive AMR in the environment. Here, we investigate the effect of material preservatives on rates of de novo mutation and horizontal gene transfer (HGT) in Escherichia coli and the model soil microorganisms Acinetobacter baylyi and Bacillus subtilis. To this end, we determined the effects of material preservatives on the mutation rates and HGT with the Luria-Delbrück fluctuation assay and a conjugation assay with the broad host-range plasmid pKJK5 and a transformation assay. Our data shows that the quaternary ammonium compound DDAC, copper, the pyrethroid insecticide permethrin and the azole fungicide propiconazole significantly increase mutation rates in E. coli, whereas A. baylyi and B. subtilis are not significantly affected. Moreover, we show that the carbamate IPBC and the insecticide permethrin affect HGT in a concentration dependent manner. Investigations with reporter strains for bacterial stress response pathways show that induction of the general stress response (rpoS) and components of the SOS response (recA) underlie the effects of most biocides on mutation rates and HGT. Taken together, our data is important for assessing the contribution of biocides on AMR evolution and spread in the environment. Within the following 10 years, clandestine and legal excavations revealed some 900 highly fragmented manuscripts from the late Second Temple period. This collection is generally known as Scrolls of the Judea Desert or Dead Sea Scrolls (DSS). For many years after their discovery, text analysis and fragmeents attribution were the main concern of the scholars dealing with the scrolls. The uncertain archaeological provenance of the larger part of the collection added an additional difficulty to the formidable task of sorting some 19000 fragments. After 60 years of scholar research the question of origin, archaeological provenance and correct attribution of the fragmenst are still hotly debated. To dtermine a possible contribution to the debate from the point of view of writing materials, we used otpical and electron microscopy, various X-ray based techniques as well as vibratiional sprectroscopy. We validated our approach with SY - based studies using the advantages of the synchroton radiation source with respect to the benchtop devices. Our laboratory studies showed that often production and storage locality could be distinguished thenks to the specific residues ("fingerprint") they left on the material. Moreover, we have diescovered that diferrent parchment production processses coexisted in the antiquity, and the resulting writing materials can readily be distinguished. However, this technique cannot give information on the plant-available P species in soil. Therefore, we combined DGT with solution 31P nuclear magnetic resonance (NMR) spectroscopy. This was achieved by using a modified DGT device in which the diffusive layer had a larger pore size, the binding layer incorporated an adsorption material with a higher capacity, and the device had a larger exposure area. The spectroscopic investigation was undertaken after elution of the deployed DGT binding layer in a NaOH solution. Adsorption tests using solutions of known organic P compounds showed that a sufficient amount of these compounds could be adsorbed on the binding layer in order for them to be analyzed by solution 31P NMR spectroscopy. Furthermore, various intermediates of the hydrolysis of trimetaphosphate in soil could be also analyzed over time. However, there is little evidence of dust-derived P-containing minerals in soils. Here we studied P forms along a well-described climatic gradient on Hawaii, which is also a dust deposition gradient. Soil mineralogy and soil P forms from six sites along the climatic gradient were analyzed with bulk (X-ray diffraction and P K-edge X-ray absorption near edge structure) and microscale (X-ray fluorescence, P K-edge X-ray absorption near edge structure, and Raman) analysis methods. In the wettest soils, apatite grains ranging from 5 to 30 μm in size were co-located at the micro-scale with quartz, a known continental dust indicator suggesting recent atmospheric deposition. In addition to colocation with quartz, further evidence of dust-derived P included backward trajectory modeling indicating that dust particles could be brought to Hawaii from the major global dust-loading areas in central Asia and northern Africa. Although it is not certain whether the individual observed apatite grains were derived from long-distance transport of dust, or from local dust sources such as volcanic ash or windblown fertilizer, these observations offer direct evidence that P-containing minerals have reached surface layers of highly-weathered grassland soils through atmospheric deposition. They dominate a range of hostile natural and man‑made environments – from desert rocks and salterns to dishwashers, roofs, and solar panels. Due to their slow growth and the lack of sexual cycles and genetic tools, the underlying mechanisms of black fungi’s phenotypic traits have remained largely unexplored. We consider the rock inhabitant K. petricola [Eurotiomycetes, Chaetothyriales] a suitable model for studying the phenotypic characteristics of black fungi. With K. petricola the regulation of pigment synthesis, general stress responses and the unusual modes of cell division can be dissected by advanced reverse and forward genetics approaches. The genome of K. petricola strain A95 was sequenced using a combination of short high quality Illumina reads and long PacBio reads. The final assembly consists of twelve contigs: five complete chromosomes and six contigs with one telomer each. Gene annotation supported by transcriptomics and proteomics data was manually curated. Recently, we developed a set of genetic tools to manipulate the genome for analyzing gene functions and studying the cell biology. This set includes CRISPR/Cas9-based genome editing and live-cell imaging using genetically encoded fluorescent proteins, as well as protocols for -omics approaches and for simulation of mineral weathering in the laboratory. Mutants defective in DHN melanogenesis, carotenogenesis or both processes are currently studied to elucidate the role of these protective pigments in tolerance of natural and man-made stresses, weathering of olivine, penetration of marble, and adhesion to surfaces. Further, the established protocols and knowledge gained from K. petricola form a starting point for making other extremotolerant black fungi accessible to genetic manipulation. They exhibit slow yeast-like or meristematic growth, do not form specialized reproduction structures and accumulate the black pigment 1,8-dihydroxynaphthalene (DHN) in the multilayered cell walls. We chose the rock inhabitant Knufia petricola of the Chaetothyriales as a representative for developing methods for genetic manipulation, simulation of mineral weathering and study of symbiotic interactions. Here, we report on the expansion of the genetic toolkit by more efficient multiplex CRISPR/Cas9 using a plasmid-based system for expression of Cas9 and multiple sgRNAs and three additional resistance selection markers. The targeted integration of expression constructs by replacement of essential genes for pigment synthesis allows for an additional color screening of the transformants. The black-pink screening due to the elimination of pks1 (melanin) was applied for promoter studies using GFP fluorescence as reporter, while the black-white screening due to the concurrent elimination of pks1 (melanin) and phs1 (carotenoids) was used to identify transformants that contain the two expression constructs for co-localization or bimolecular fluorescence complementation (BiFC) studies. In addition, two intergenic regions (igr1, igr2) were identified in which expression constructs can be inserted without causing obvious phenotypes. For this study, wet-discharged bottom ash fine-fraction samples from full-scale treatment plants in Germany and Sweden were analyzed. The potential for the recovery of heavy metal compounds was investigated through wet density-separation with a shaking table. The feed materials were processed without any pre-treatment and the optimum processing conditions were determined by means of design of experiments. Tilt angle and stroke frequency were identified as the most relevant parameters, and the optimum settings were − 7.5° and 266 rpm, respectively. The obtained balanced copper enrichments (and yields) were 4.4 (41%), 6.2 (28%) and 2.4 (23%). A maximum copper enrichment of 14.5 with 2% yield was achieved, providing a concentrate containing 35.9 wt.% relevant heavy metal elements. This included 26.3 wt.% iron, 4.3 wt.% zinc and 3.8 wt.% copper. In conclusion, density separation with shaking tables can recover heavy metals from bottom ash fine fractions. Medium levels of heavy metal enrichment (e.g., for Cu 2.7–4.4) and yield (Cu: 26–41%) can be reached simultaneously. However, the separation performance also depends on the individual bottom ash sample. Circulating materials in anthropogenic processes come with the risk of accumulating hazardous substances and compounds. In this concept, waste incineration or waste-to-energy (WtE) is a necessary technology to remove these compounds from the life cycle. In this mini-review, contaminants of major importance in the flue gas from waste incineration plants and their environmental impact are discussed. Air pollution of WtE is often seen as the most relevant environmental impact of this treatment option. The emission values parameter set for different countries is presented and compared. The most stringent legally set of emission values could be found in parts of Europe and South Korea. Japan also permits similar strict values when authorising individual incineration plants. In North America, the values are partially less strict as the best available technologies in Europe suggest being possible. Emerging economies, such as India and China, have shown efforts to improve their environmental protection standards but still have room to improve. This could be set in relation to other industrial emitting processes and therefore could be used to assess the relevance of this industry sector to the national emission inventories. They exhibit slow yeast-like or meristematic growth, do not form specialized reproduction structures and accumulate the black pigment 1,8-dihydroxynaphthalene (DHN) melanin in the multilayered cell walls. To understand how black fungi live, survive, colonize mineral substrates, and interact with phototrophs genetic methods are needed to test these functions and interactions. We chose the rock inhabitant Knufia petricola of the Chaetothyriales as a model for developing methods for genetic manipulation. Here, we report on the expansion of the genetic toolkit by more efficient multiplex CRISPR/Cas9 using a plasmid-based system for expression of Cas9 and multiple sgRNAs and the implementation of the three resistance selection markers genR (geneticin/nptII), baR (glufosinate/bar), and suR (chlorimuron ethyl/sur). The targeted integration of expression constructs by replacement of essential genes for pigment synthesis allows for an additional color screening of the transformants. The black-pink screening due to the elimination of pks1 (melanin) was applied for promoter studies using GFP fluorescence as reporter. The black-white screening due to the concurrent elimination of pks1 and phs1 (carotenoids) allows to identify transformants that contain the two expression constructs for co-localization or bimolecular fluorescence complementation (BiFC) studies. The co-localization and interaction of the two K. petricola White Collar orthologs were demonstrated. Two intergenic regions (igr1, igr2) were identified in which expression constructs can be inserted without causing obvious phenotypes. Plasmids of the pNXR-XXX series and new compatible entry plasmids were used for fast and easy generation of expression constructs and are suitable for a broad implementation in other fungi. This variety of genetic tools is opening a completely new perspective for mechanistic and very detailed study of expression, functioning and regulation of the genes/proteins encoded by the genomes of black fungi. T1 - Improving consistency at testing cementitious materials in the Dynamic Surface Leaching Test on the basis of the European technical specification CENTS 16637–2 – Results of a round robin test N2 - The environmental impact assessment of materials is usually based on laboratory tests, mostly in combination with models describing the longterm fate of the substances of interest in the targeted environmental compartment. Thus, laboratory tests are the fundamental link to achieve appropriate assessment conclusions which makes it essential to generate consistent results. This just as applies to the leaching of cementitious materials. In Europe, the leaching behavior of monolithic building materials is tested in the Dynamic Surface Leaching Test following the specification CEN/TS 16637–2. An interlaboratory comparison on European level regarding this technical specification showed relatively high intra- and interlaboratory variations for the tested materials (monolithic copper slag and cement stabilized coal fly ash). Therefore the German Committee for Structural Concrete (DAfStb) framed a guideline to specify additional testing conditions for cementitious materials. To assess the possible improvement by this guidelines measures, a round robin test with 11 participants from Germany and the Netherlands was conducted. This work aims to provide insight into the factors to be considered in the testing of alkaline materials, including sample preparation, and highlights crucial procedures and their manifestation in the results. All evaluated parameters showed improved results compared to the earlier round robin test. The relative standard deviations for repeatability (RSDr) and reproducibility (RSDR) of the elements calcium, barium, antimony, chromium, molybdenum and vanadium, which are the parameters evaluated in both round robin tests, were RSDr = 4%, 4%, 2%, 5%, 5%, and 5% respectively (4% in average) for this work, in comparison to the European round robin test with an average RSDr of 29% (17%, 17%, 20%, 40%, 36%, and 42%). The RSDR improved from 41% (30%, 36%, 29%, 57%, 40%, and 56%) to 14% (12%, 8%, 6%, 28%, 15%, and 12%). CO2 ingress during testing and the inaccuracy of eluate analytics for concentrations close to the determination limits were identified as the main sources of error. T1 - Waste-derived fertilizers can increase phosphorus uptake by sugarcane and availability in a tropical soil N2 - The use of highly water-soluble phosphorus (P) fertilizers can lead to P fixation in the soil, reducing fertilization efficiency. Waste-derived, low water-solubility sources can potentially increase sugarcane’s P uptake compared to triple superphosphate by reducing adsorption to the soil. Aims:We aimed to test struvite, hazenite, and AshDec® for their agronomic potential as recycled fertilizers for sugarcane production in a typical tropical soil.We hypothesize that these sources can reduce P fixation in the soil, increasing its availability and sugarcane’s absorption. Methods: In a greenhouse pot experiment, two consecutive sugarcane cycles, 90 days each, were conducted in a Ferralsol. The recovered sources struvite, hazenite, AshDec®, and the conventional triple superphosphate were mixed in the soil in three P doses (30, 60, and 90 mg kg–1), aside a control (nil-P). At both harvests, sugarcane number of sprouts, plant height, stem diameter, dry mass yield, shoot phosphorus, and soil P fractionation were investigated. Results: At 90 days, struvite and hazenite performed better for dry mass yield (70.7 and 68.3 g pot–1, respectively) than AshDec® and triple superphosphate (59.8 and 57.4 g pot–1, respectively) and for shoot P, with 98.1, 91.6, 75.6, and 66.3 mg pot–1, respectively. At 180 days, struvite outperformed all treatments for dry mass yield (95.3 g pot–1) and AshDec® (75.5 mg pot–1) for shoot P. Struvite was 38% and hazenite 21% more efficient than triple superphosphate in P uptake, while AshDec® was 6% less efficient. Soil had higher labile P under struvite, hazenite, and AshDec® than triple superphosphate by the end of the first cycle, while only the later increased nonlabile P by the end of the experiment (180 days). Conclusions:Waste-derived P sources were more efficient in supplying P for sugarcane and delivering labile P in 180 days than triple superphosphate.
