Fungi actively sense the environmental light conditions to induce protective mechanisms against the light-associated stresses and to regulate development. Since fungi adapted to habitats with different light regimes, the complexities of their ‘visual’ systems and photoresponses may vary significantly. Fungi that are associated with plants experience a special light regime because the host seeks optimum light conditions for photosynthesis – and the fungus must cope with them. Moreover, fungi living under the canopy are confronted with an altered spectrum enriched for green and far-red light. Plants sense light to coordinate growth and morphogenesis: a low red:far-red ratio indicates the presence of competitors (shading by other plants) and triggers the shade avoidance response which is accompanied by increased susceptibility to nectrophic pathogens. Botrytis cinerea, the causal agent of gray mold diseases on many plant species in moderate climate zones (high humidity, temperatures around 20°C, seasons), exhibits striking photoresponses accompanied by a broad action spectrum and a high number of photoreceptors. Light is the most important factor controlling morphogenesis: it induces conidiation for disease spreading (summer cycle) and represses sclerotial development for survival and/or sexual recombination (winter cycle). In the past years, we identified several cellular components involved in photoperception and regulation of photomorphogenesis and virulence indicating that the same signaling pathways are of relevance for both processes, propagation and infection. Notably, we recognized the role of the phytochromes (red/far-red light sensors) for coordinated responses to light and elevated temperatures (photo-/thermomorphogenesis) – similar to the function of Arabidopsis PhyB. BcPHY2-like thermosensors are likely restricted to the Leotiomycetes, which include several plant pathogens of the moderate climate zones. Biological strategies to survive excess light include protection, avoidance and active utilisation. Phototrophic organisms including cyanobacteria, green algae and plants use all three strategies. Fungi occupying phototrophic niches may profit from the surplus photosynthetic products which also obliges them to cope with the same light-induced stresses. One way of handling these stresses would be to use similar signalling pathways to sense the presence and to interact with their phototrophic partners. Obviously, the levels of adaptation and responses to light will depend on the environment as well as fungal genotype and phylogenetic position. Black fungi – a polyphyletic group – accumulate the dark pigment DHN melanin in their cell walls and often occupy light-flooded habitats from phyllosphere to rock surfaces. Here we compare two sequenced melanised fungi of different lifestyles in their response to light. The Leotiomycete Botrytis cinerea is an aggressive pathogen that primarily infects the above-ground parts of plants. It possesses large numbers of photoreceptors that respond to a broad spectrum of light. As a consequence, light controls morphogenesis in which it induces conidiation for disease spreading and represses sclerotial development for survival and/or sexual recombination. Cellular components involved in photo-perception and regulation of morphogenesis, stress responses and virulence have been identified and appear to regulate propagation, survival and infection. These include phytochromes, a group of photoreceptors which are particularly enriched in the Leotiomycetes and that mediate coordinated responses to light and elevated temperatures. Assuming that photo-regulation may be equally important for fungi that live in mutualistic relationships with phototrophs either by forming composite organisms or biofilms, we investigate the role of light in the rock-inhabiting Eurotiomycete Knufia petricola. Like other black yeasts, K. petricola grows slowly, does not form specialised reproductive structures and constitutively produces DHN melanin as well as carotenoids. Combining K. petricola with the cyanobacterium Nostoc punctiforme, we developed a model system for studying biofilm formation and bio-weathering. A genetic toolbox to manipulate this model system is being developed. K. petricola strain A95 possesses ten putative photoreceptors, more than found in filamentous Eurotiomycetes suggesting that light plays an important role for abiotic and biotic interactions in extremo-tolerant and symbiosis-capable fungi. This presentation will examine the inks used by Jews in the in different geographical zones try to correlate the results of the material analysis with written records and existing traditions. We will compare the inks proposed by Maimonides, who lived in 12th-century Egypt, with the considerations of Rashi, who lived in 11th-century northern France, and see that they both advocated use of the inks commonly known and produced in their respective regions. It is Maimonides who proposes to add tannins to the soot inks, but rejects the metallic salt, both of which were practices that were well attested in contemporary Arabic recipes for making ink. In contrast, Rashi was favourable to employing the plant inks in use in contemporary Northern Europe. Therefore, one should try to go beyond the understanding of the basic principles of the scientific analysis. We will start with a glance at the basic principles of the techniques used in the material science for determination of the elemental composition (X-ray emission) and molecular composition (FTIR & Raman). We will move then to the bench and mobile equipment commonly used in the field of cultural heritage. At the end we will choose a question to be answered and design an ideal experiment that will be modified according to the limitations dictated by on-site conditions. Our studies of the Dead Sea Scrolls writing surfaces show that they can be divided roughly into three groups: leather, parchments of a light tint, and those of various shades of brown. The latter ones are invariably tanned, whereas the middle group is characterized by the presence of various inorganic salts. Some of the pale parchments, among them the Temple Scroll (11Q19), are remarkably similar to medieval European parchment. Therefore we have formulated the working theory that in the Judaea of the Hellenistic period two different parchment-making traditions existed side by side: an ‘eastern’ one (represented by the tanned parchments of Qumran, closely resembling Aramaic documents from the fifth century BC, and a ‘western’ one (represented by the untanned/lightly tanned ones similar to early Christian Greek parchments). This division has found support during our studies of the Geniza fragments, in which Babylonian and Palestinian traditions seem to follow the “eastern” and “western” technologies, respectively. The first comprises materials that write themselves, producing script by rubbing their own material off onto the writing surface. It includes charcoal, graphite, chalk, raddle, and metal styluses. Depending on the material and consistency, these are cut or pressed to make styluses and then used for writing. The second group comprises all coloring liquids that are applied to the writing surface with a quill, pen, or printing block. It includes inks made from dye solutions (for example, tannin inks) and those made from pigment dispersions (for example, sepia, soot, and bister inks). The latter are sometimes also rubbed as pastes into letters incised into the writing surface, where they increase visual contrast. Due to the variety of recipes and the natural origin of raw materials, there is a wide range of different components and impurities in writing materials. Soluble inks (Tinten) Soluble inks are based mainly on dyes forming a water solution. Colored inks were manufactured with different plant or insect dyes (e.g. Brazil wood, kermes). To stabilize the volatile material, the dyes were mixed with a mordant (e.g., alum). Brown plant inks – best-known as blackthorn or Theophilus’ inks – are usually produced from the blackthorn bark and wine. In the early European Middle Ages, inks of this kind were widely used in the production of manuscripts in monasteries. Usually, they are light brown, so sometimes small amounts of iron sulfate were added, which led to what was called an “imperfect” iron gall ink. The difference between “classic” iron gall ink and such imperfect ink is therefore not clear: the distinction is not possible, especially with the naked eye. Dispersion inks (Tuschen) According to its generic recipe, one of the oldest black writing materials is produced by mixing soot with a binder dissolved in a small amount of water. Thus, along with soot, binders such as gum arabic (ancient Egypt) or animal glue (China) are among the main components of soot inks. From Pliny’s detailed account of the manufacture of various soot-based inks, we learn that, despite its seeming simplicity, producing pure soot of high quality was not an easy task in Antiquity. Therefore, we expect to find various detectable additives that might be indicative of the time and place of production. One such carbon ink requires the addition of copper sulfate . The experimental discovery of this ink in 1990 led to a misleading expression “metal ink” that is sometimes found in the literature. Colored dispersion inks based on pigments such as orpiment, cinnabar, or azurite have been known since Antiquity. Natural or artificially produced minerals are finely ground and dispersed in a binding medium. As in soot inks, water-soluble binders such as gum arabic or egg white were used. Iron gall ink (Eisengallustinten) Iron gall inks are a borderline case between these two groups. They are produced from four basic ingredients: galls, vitriol as the main source of iron, gum arabic as a binding media, and an aqueous medium such as wine, beer, or vinegar. By mixing gallic acid with iron sulfate, a water-soluble ferrous gallate complex is formed; this product belongs to the type “soluble inks”. Due to its solubility, the ink penetrates the writing support’s surface, making it difficult to erase. Exposure to oxygen leads to the formation of insoluble black ferric gallate pigment, i.e., “dispersion ink”. Natural vitriol consists of a varying mixture of metal sulfates. Since for ink making it was obtained from different mines and by various techniques, inks contain many other metals, like copper, aluminum, zinc, and manganese, in addition to the iron sulfate. These metals do not contribute to color formation in the ink solution, but possibly change the chemical properties of the inks. Methanogenic archaea can also cause MIC by directly withdrawing electrons from the iron surface for methanogenesis. However, the mechanistic details and kinetics of the overall process are poorly understood. Precipitation of siderite, a by-product of methanogenesis, (4Fe + 5HCO3 + 5H+  4FeCO3 + CH4 + 3H2O) can lead to an insulating layer on the metal surface and lower the corrosion rate. Regarding the predicted decrease of gypsum supply in Germany, particularly the recycling of gypsum (calcium sulfate) is of growing importance. Currently, the gypsum demand is fulfilled (at least 60%) by gypsum as side product from coal-fired power plants (FGD Gypsum). Germany’s natural gypsum deposits fulfil the remaining gypsum demand. Due to national climate protection goals the gypsum supply from coal power plants will decrease significantly in the future. In addition, the content of sulfates in other secondary building materials, in particular in recycled concrete aggregates, should be minimized for quality reasons. Separated gypsum can be used in gypsum production if the high quality requirements for recycled gypsum are met. Accordingly, there have been significant advancements in the processing of gypsum residues in the last years. Since almost all processing steps in the recycling process are associated with environmental impacts, an evironmental evaluation of the use of recycled gypsum as a substitute in gypsum production has to be carefully conducted. A1 - Adam, Christian T1 - Effect of Nitrification Inhibitor on Nitrogen Forms in Soil analyzed by Nitrogen K edge micro XANES Spectroscopy N2 - Specific co-fertilization of nutrients can enhance their plant-availability and thus the yield of plants. To investigate this effect, we performed a pot experiment with three different P-fertilizers and ammonium nitrate sulfate as a co-fertilizer, without and with a nitrification inhibitor (NI), and analyzed the form of nitrogen (N) in the soil via novel X-ray spectroscopic method. The application of NI with the N fertilizer led to a higher dry matter yield of maize. Novel N K-edge micro-X-ray absorption near-edge structure (micro-XANES) spectroscopy identified that the application of a NI promotes the temporary formation of a non-exchangeable N in detectable hot-spots in the soil. The subsequent slow release and prolonged availability of N during plant growth leads to higher yield. It can be concluded that NIs lead to a temporary fixation of ammonium-N in a pool that can be accessed by plant roots. A wide range of variables is taken into consideration depending on the quantity of samples analysed from a specific geological region and the methods applied. In order to provide evidence, a multi-technique analytical approach using µXRF, ESEM, PGAA and PIXE is applied to samples from the most famous deposits of lapis lazuli. Special elements determined as fingerprints are compared in relation to the forming conditions obvious in textural features. The results and statistical output allow a differentiation that enables an optimized local classification of the blue stone. An absolute requirement for all geo-tracing performed on blue colored cultural objects of unknown provenance is awareness of the limits of analysis. The possible sources of lapis lazuli are tested by analysing the blue pigment used as paint on murals and ink on manuscripts from the Silk Road. Since pure tricalcium-silicate is only stable above temperatures of 1250 °C, its decomposition has to be prevented technically by fast cooling after the sintering process. At room temperature, the decomposition velocity is very slow so that metastable tricalcium-silicate is obtained. Although the mechanisms of clinker phase formation during burning process of Portland cement in a rotary kiln were solved and improved over the years, in view of possible economic and ecological benefits current projects aim to produce clinker phases from metallurgical slags. Recent studies discovered that the mineral phase which remained after a reducing treatment and separation of formed metallic iron from molten Linz-Donawitz (LD-) slags contained about 60 wt.% Alite despite it was cooled slowly. Because the results could be verified using slags from different origins and varying cooling velocities a chemical stabilisation of the Alite can be assumed. First tests in mortars indicate that workability, hardening and solid state properties are comparable with an ordinary Portland cement. An application of the observed phenomenon in cement production requires enhanced knowledge about formation and stabilisation conditions of Alite during crystallisation from melts in contrast to the sintering reactions in conventional Portland cement production. Therefore, this study focuses on the stabilisation mechanisms of Alite in consolidating melts. Samples from different melting experiments are analysed to determine stabilising factors. However, these DGT results do not identify which compound of the specific element has the high bioavailability. Using various spectroscopic techniques (infrared, XANES and NMR spectroscopy) to analyze the dried DGT binding layers after deployment could allow us to determine the specific elements or compounds. Nutrients such as phosphorus and nitrogen are often, together with other elements, present as molecules in the environment. These ions are detectable and distinguishable by infrared and NMR spectroscopy, respectively. In addition, XANES spectroscopy allows for the specification of nutrients and pollutants (e.g. chromium) on the DGT binding layer. Furthermore, microspectroscopic techniques make it also possible to analyze compounds on the DGT binding layer with a lateral resolution down to 5 µm2. Therefore, species of elements and compounds of e.g. a spatial soil segment can be mapped and analyzed, providing valuable insight to understand the dynamics of nutrients and pollutants in the environment. However, these novel P-fertilizer can also contain toxic pollutants e.g. chromium (Cr) in the hexavalent state (Cr(VI)), which is regulated with low limit values in agricultural products (German fertilizer ordinance limit: 2 mg/kg Cr(VI)). The determination of Cr(VI) in these novel fertilizer matrices can be hampered by redox processes that lead to false results with the standard wet chemical extraction method (German norm DIN EN 15192). Therefore, we analyzed Cr(VI) in various P-fertilizers with the DGT technique. DGT devices equipped with a APA (polyacrylamide) diffusion layer and Cr(VI) selective N-methyl-D-glucamine (NMDG) binding layer were used for the study. After a 24 h conditioning period of the fertilizer at 60% of the water holding capacity (WHC), the fertilizers were brought to 100% WHC, transferred onto the DGT devices and deployed for 24 h at 25°C. The extraction of Cr from the DGT binding layer was carried out with 1 M HNO3 for 24 h. The Cr-concentrations of the extract were determined by means of ICP-MS. We found a good correlation between the standard wet chemical extraction and the DGT method for the whole range of P-fertilizers. However, partly soluble Cr(VI) compounds cannot be detected in full extent by the DGT method that is best suited for mobile Cr(VI). Furthermore, Cr K-edge XANES spectroscopy showed that the Cr(VI)-selective DGT binding layer also adsorbs mobile Cr(III) compounds from acid treatment of phosphates which can therefore cause an overestimation of Cr(VI). The DGT method was very sensitive and in most cases selective for the analysis of Cr(VI) in P-fertilizers made from recycled materials. However, the results of certain types of P-fertilizers containing mobile Cr(III) or partly immobile Cr(VI) show that still some optimization of the method is required to avoid over- or underestimation of Cr(VI). T1 - Development of a standard substrate to facilitate the use of DGT for the assessment of P recycling fertilizers N2 - P recycling fertilizers are gaining increasing importance in our efforts to close nutrient cycles. An unsatisfactory performance of standard chemical extraction methods to assess the fertilizing effects of such products was reported. They demonstrated that DGT extractions of incubated soil/fertilizer mixtures were able to predict the fertilizing effects of the respective products more accurately. Since DGT works with soil/fertilizer mixtures, its interpretation is soil-dependent. Therefore, in order to facilitate its use as a tool to predict fertilizer performance, it needs to be standardized based on a standard substrate. This research aims to develop a standard substrate based on which evaluation categories for the DGT fertilizer extraction can be derived. The substrate composition should allow to vary the most important soil properties determining the plant availability of fertilizer P. It must also be reproducible at any time and any place. Substrate variants with varying proportions of quartz sand, a clay mineral and sphagnum peat were prepared and set to pH-levels 5.5 and 7 by addition of CaCO3. 7 variants were incubated with a set of test fertilizers (2 recycling fertilizers based on sewage sludge ash and 2 conventional mineral fertilizers) for 2 weeks. Substrate/fertilizer mixtures were then extracted with DGT and an ANOVA was performed to test if the DGT extraction was able to depict significant differences between fertilizers and substrate variants. An 8-week pot trial with ryegrass (3 cuts) was set up with the same substrate variants and test fertilizers. P uptake was determined to assess the fertilizing effect and correlated with the results of the DGT extractions. Statistically significant differences were found between DGT results for the various test fertilizers and substrate variants, indicating that DGT is able to differentiate between P solubility of fertilizers in relation to substrate quality. DGT results showed a strong relationship with P uptake, confirming that this method is suitable to predict the fertilizing effect of P fertilizers. Further optimization of substrate composition and tests with a wider variety of crops and fertilizer types are needed, before evaluation categories for DGT values can be derived. Biodiversity is commonly estimated based on genetic variation. We investigated a new approach that defines and measures biodiversity in complex microbial communities. We used the variation in multiple functionally-relevant, phenotypic traits measured in parallel in single cells as a metric for microbial phenotypic diversity. We studied phenotypic diversity and ecosystem functioning throughout different photosynthetic layers dominated by divergent microbial communities in the gradient of Lago di Cadagno. We determined genetic diversity by 16S and 18S amplicon sequencing and bulk ecosystem functioning (photosynthesis). In addition, we determined phenotypic diversity using single-cell technologies such as nanometer-scale secondary ion mass spectrometry (NanoSIMS) correlated with confocal laser scanning microscopy (CLSM) and scanning flow-cytometry. We measured functional trait variation between individuals in 13CO2 fixation, 15NH4+ uptake, and variation in physio-morphological cell traits, such as cell size, shape, and auto-fluorescence for various pigments related to photosynthesis. We used the distances between individuals in a multidimensional trait space to derive phenotypic trait-based diversity indices, such as trait richness, trait evenness, and trait divergence. We find that phenotypic trait divergence associates with ecosystem functioning, whereas genetic diversity does not. Including activity-based, single-cell phenotypic measurements with NanoSIMS provided an additional accuracy to the trait-based diversity assessment and allowed us to formulate hypotheses on the mechanisms that shape the correlation between phenotypic diversity and eco-system function. Together, our results show that phenotypic diversity is a meaningful concept to measure microbial biodiversity and associate it with ecosystem functioning. Until now, there is a lack of systematic studies that investigate if bacterial populations establish persister subpopulations that tolerate disinfectant stress longer than most of the population. Objectives: Our objective is to investigate if persistence is a bacterial survival strategy against disinfectants. Furthermore, we investigate the mechanisms of disinfectant persistence and if persistence can evolve in the face of fluctuating exposure to disinfectants. Ectoine possesses protecting properties and stabilizes proteins as well as whole cells against stresses like ionizing radiation and cytotoxins. These properties make ectoine a highly demanded ingredient in cosmetics and pharmaceuticals. To date H. elongata is the industrial Producer strain of ectoine, but several metabolic factors for optimum ectoine production remain to be explored. In this work, we used up to date Metabolic engineering approaches following the ‘Push, Pull, Block – strategy’ to examine targets that contribute to ectoine synthesis. Firstly, the basics of glucose catabolism were inspected to PUSH and enhance carbon flow towards ectoine synthesis. Secondly, lysine biosynthesis was targeted to BLOCK a pathway that is competing for precursors with ectoine synthesis. Thirdly, the mechanosensitive (MS) channels of H. elongata have been examined as possible excretion routes for ectoine. An overexpression of the ectoine excretion channels potentially could PULL out product at the end of ectoine synthesis and increase overall ectoine flux. For the interrogation of central metabolic pathways, we established the new molecular tool CRISPR-mediated interference (CRISPRi) for targeted modulation of gene expression. PUSH Glucose catabolism through the Entner-Doudoroff (ED) and Emden-Meyerhof-Parnas (EMP) pathway was targeted with CRISPRi and examined on gene expression level for ist response to changing salinity and different carbon sources. Changing salinity did not influence gene expression levels of glucose catabolism but the carbon source glucose triggered glycolysis through the (ED) pathway. When gene expression of the ED pathway was downregulated with CRISPRi, the growth rates remained constant. The observations indicate a metabolic overflow mechanism for glycolysis, in which fluxes are constantly high - even at lower salinity when no resources are demanded for ectoine synthesis. The further analysis of glucose to product conversion rates will advise optimum conditions for future industrial cultivation processes. BLOCK Lysine biosynthesis was downregulated with CRISPRi, which led to a significant increase in ectoine production. Hence, the blockage of lysine biosynthesis would be a valuable strategy for the optimization of the industrial producer strain in future studies. PULL MS channels and ectoine regulation are inevitably connected in osmoadaptation. Therefore, ectoine excretion, growth performance and gene expression levels of the MS channels were monitored in steady state conditions and in response to osmotic shock in the wildtype strain and in a MS channel deletion mutant. We observed that the MS channels were essential for the survival of osmotic shock but surprisingly their presence reduced cell growth under high salinity. The MS channels were only partially responsible for ectoine excretion. Thus, alternative ectoine excretion channels must exist and remain to be explored. T1 - Auswirkungen der novellierten AbfKlärV auf die Kreislaufführung von Pflanzennährstoffen N2 - Because of the longstanding oversupply of nutrients in Germany, waste and fertilization legislation has been amended in 2017. As the new laws do still not meet the European nitrate ordinance, fertilization laws must be tightened again. In consequence, there is a rising competition for land between sewage sludge and organic fertilizers. Furthermore, the new sewage sludge ordinance (AbfKlärV) contains stricter thresholds and a ban on agricultural sewage sludge utilization for WWTPs > 50,000 p.e. from 2029. WWTPs which do not use their sludge for agriculture have to recover phosphorus from then on. However, sludges with less than 2% P content are not affected by that obligation. Every federal state of Germany must report amounts and quality of agricultural utilized sewage sludge yearly. An evaluation of raw data from 10 federal states shows that nearly 40% of both WWTPs and sewage sludge volume fall below the recovery limit of 2%. On the other hand, this means less than 30% of the P potential. In addition, there is no information whether the sludges fall reliably below the limit as required by the ordinance. This project aims to investigate this reliability by regarding the variations in the P content of sewage sludge. For this reason, samples will be taken monthly at 10 representative WWTPs over a year. Whereas phosphorus will still mostly be subject of recovery other nutrients are not affected by this. As a result, large amounts of N, K, Mg and Ca will be increasingly removed from the cycle. In 2017, WWTPs > 50,000 p.e. accounted for 30-60% of the nutrient potentials. These potentials will probably get lost to a large extent due to the fertilization ban. Many small WWTPs are also currently changing their utilization strategy from agriculture to incineration to ensure a safe disposal in future. Therefore, we can expect that even larger amounts of nutrients apart from phosphorus will be discharged from the circular economy. In comparison to the domestic sales volume of mineral fertilizers these other nutrients do only account for a very small fraction. A recovery could nevertheless replace parts of the mineral fertilizers, thus reducing environmental impacts. Energy consumption for technical nitrogen fixation is high and nutrient cycles are strongly anthropogenic influenced with diverse consequences such as eutrophication, acidification and endangered groundwater resources. Sewage sludge contains several micronutrients next to the macronutrients P, N, K, Mg, Ca and S. Because of lacking information on the actual German need for micronutrients contents and freights cannot evaluated. Moreover, former sewage sludge fertilization was probably not needs-based. It is therefore not possible to estimate a true loss in micronutrients due to the amended sewage sludge ordinance. N2 - Die landwirtschaftliche Klärschlammverwertung ist in der Bundesrepublik Deutschland seit Jahren rückläufig, hatte im Jahr 2017 jedoch noch einen Anteil von rund 18 % [20]. Diese Art der Verwertung wird kontrovers diskutiert, da der Klärschlamm im Rahmen der Abwasserreinigung eine Schadstoffsenke darstellt. In vergangenen Jahrzehnten wurden insbesondere die hohen Gehalte an Schwermetallen im Klärschlamm als kritisch für eine landwirtschaftliche Verwertung eingestuft. Diese Belastung konnte allerdings durch Absenkung der Einträge von Schmermetallen in das Abwasser signifikant gesenkt werden. Neben Schwermetallen kamen jedoch auch organische Schadstoffe wie Pharmazeutika, Kosmetika und Haushaltschemikalien in den Fokus der Diskussion über den Einsatz von Klärschlamm auf Agrarflächen. Heute geraten zunehmend weitere Inhaltsstoffe in den Fokus, deren Auswirkungen noch nicht abschließend geklärt sind, weshalb eine einheitliche Bewertung über Grenzwerte nicht darstellbar ist. Beispiele für diese neuen Stoffgruppen sind Mikroplastik, Nanopartikel, Antibiotikaresistenzen und persistente organische Spurenstoffe. Neben den angesprochenen Risiken gibt es in Deutschland Regionen mit einem stark erhöhten Nährstoffüberschuss. In der Kritik steht vor allem eine nicht ordnungsgemäße Düngung mit Stickstoff, welche mit Nitratauswaschungen in das Grundwasser einhergeht. Um diese Grundwasserbelastung zu verhindern wurde u.a. das Düngerecht erneut verschärft, was die Flächenkonkurrenz zwischen Klärschlamm und Wirtschaftsdünger weiter vergrößern wird. Auf Druck der EU-Kommission stehen zudem weitere Anpassungen im Düngerecht bevor, da die beschlossenen Änderungen aus Kommissionssicht nicht ausreichen, um die Nitratrichtlinie einzuhalten. Vor dem Hintergrund der angeführten Probleme wurde im Koalitionsvertrag der 18. Legislaturperiode (2013) festgeschrieben, „die Klärschlammausbringung zu Düngezwecken [zu] beenden und Phosphor und andere Nährstoffe zurück[zu]gewinnen“ [5]. Die novellierte AbfKlärV beinhaltet allerdings kein grundsätzliches Aufbringungsverbot und eine Rückgewinnungspflicht besteht lediglich für den Nährstoff Phosphor. Es ist davon auszugehen, dass sich die neuen Regelungen im Düngerecht und der Klärschlammverordnung massiv auf die landwirtschaftliche Verwertung von Klärschlamm und damit auf die Kreislaufwirtschaft verschiedener Nährstoffe auswirken werden. In the Department “Materials and the Environment” of the BAM new experimental platform is being developed. Despite the focus on fundamental research, the main results of this project proposal will be transferable into material technology and construction chemistry and will influence the development of standardization in this topic. Morphologically simple microbial biofilms were the first settlers on these inhospitable surfaces at times when the Earth was inhabited only by microorganisms and the solid substrates represented only by natural rock surfaces i.e. lithosphere. Miniature, self-sufficient microbial ecosystems continue to develop on subaerial (i.e. air-exposed) solid surfaces at all altitudes and latitudes where direct contact with the atmosphere and solar radiation occurs – on rocks, mountains, buildings, monuments, solar panels. All these sub-aerial biofilms develop under fluctuating and hostile conditions – and thus frequently harbour stress-tolerant black fungi inherently able to cope with the stresses of bright sunlight and constantly changing atmospheric conditions. Black fungi – a polyphyletic group of Ascomycetes– accumulate the dark pigment DHN melanin, diverse carotenoids and mycosporines in their cells and thus successfully colonise sunlight-flooded habitats from phyllosphere to rock surfaces. Various chemical and physical extremes and fluctuating environments belong to the challenges effectively mastered by black fungi. In our laboratory we isolate novel black fungi from man-made habitats like building materials and solar panels. Using Knufia petricola A95 as a model we conduct experiments to clarify interactions of black fungi with inorganic substrates. We use available mutants to determine the functional consequences of changes in the outer cell wall envelopes – from excreted EPS to layers of protective pigments. A genetic toolbox to manipulate this Chaetothyriales representative is in further development. Our long-term goal is to understand the fundamental mechanisms how black fungi are able (i) to adhere to dry atmosphere-exposed surfaces, (ii) to survive multiple stresses and (iii) to change the underlying substrates including rocks. In the presentation examples of how knowledge of these organisms can be of practical value (e.g., in biodeterioration studies, such as on stone markers in cemeteries) would be given. Among other stories, the example of how roof tiles amended with a titanium oxide layers as a biocide actually selected for the black fungi will be told. A hypothesis about how these organisms would likely be found on solar panels and some early BAM work in that area will be presented. This then brings the connection to a study ripe for investigation in North Carolina. The talk presents the development of a study aiming at deciphering the influence of microbial biofilm formation on the energy conversion efficiency of solar photovoltaic panels or modules at two facilities (one facility under the impact of high intensity of animal agriculture and high deposition of ammonia from atmosphere and the other with low animal agriculture and lower atmospheric ammonia deposition) in North Carolina. The main hypothesis of the study is that microbial biofilm formation on solar photovoltaic panels will lead to significant decreases in energy conversion efficiency of solar photovoltaic modules and biofilm formation will also be accelerated by high ammonia concentration in the ambient atmosphere and high nitrogen deposition. In a joint project between CSMC and the University of St Andrews, we aim to analyse papers and their decorations in Persian The analyses aim to determine the consistencies of papers, pigments and metals and the techniques used for their production and application. The support will be first described in its codicological characteristics such as dimensions, thickness, colour (expressed in colour coordinates), pattern left from the sieve (observed in transmitted light). The plant used as raw material in paper production will be studied by fibres analysis, while the sizing agents, the pigments, dyes and metals will be analysed by the application of several methods: in particular Reflectography, X-Ray Fluorescence and InfraRed, Raman and Visible Reflectance Spectroscopy. The results of the analyses will form an important resource for the material aspects of Persian manuscripts and be a starting point for establishing a first chronology of the materials and techniques used in Persian manuscript production. In this talk I will give an overview of the recipes – in particular those by ar-Rāzī –, their context and transmission in order to evaluate this double nature of papyrus, which is considered at the same time a writing support and a material to be recycled. Moreover, I will discuss what kinds of ink are most likely to be found, according to these literary sources, on Arabic manuscripts and documents written on papyrus and compare them to the typologies mentioned for paper and parchment. Unfortunately, the scientific investigation of Arabic books is still uncommon, focusing mainly on single case studies of illuminated manuscripts where the black media used to write the text is largely overlooked. This lack of knowledge prompted a recent research in which black ink recipes have been collected from written Arabic sources, their feasibility has been assessed, and some of them have been reproduced. These samples were artificially aged and analysed through an array of analytical techniques, mostly non-invasive and non-destructive, in order to build a database of Arabic inks and their ingredients and to verify the detection limits of the equipment employed. Finally, the results were compared to the data collected from historical manuscripts. In this talk I will focus on the ink typologies and the terminology (ancient and modern) used to describe them. In particular I will highlight how even the modern classification fails to capture the variety encountered in both recipes and manuscripts, particularly in the light of recent discoveries. I will address especially the terminological and conceptual issues of mixed inks – in the form of carbon inks and tannins, of carbon and iron gall inks, and of carbon and metallic salts –, iron gall inks made without vitriol and iron gall inks made with sources of tannins different from gall nuts. Recipes can also appear, with or without relation to the main text in the manuscript, in the form of lists or collections or even added in empty spaces as single entries. Why were these recipes written down? Were they used by the many professionals dealing with inks in their every-day work? Or were they part of the literary genre of adab with little or no practical application? To answer these questions a research project was set up: ink recipes have been collected from written Arabic sources, their feasibility has been assessed and some of them have been reproduced. These samples were artificially aged and analysed through an array of analytical techniques, most non-invasive and non-destructive, and the results compared to the data obtained by the application of the same analyses on concrete manuscripts. Possibly as a response to the import of tinted and gold-decorated paper from China, which was used for manuscript production in Greater Iran, Persian papers were also tinted and embellished with gold-decorated and marbled margins or frames. In a joint project between CSMC and the University of St Andrews, and in co-operation with the Golestan Palace Museum, we aim to analyse papers and their decorations in manuscripts in the holdings of the Golestan Palace Library. The analyses aim to determine the consistencies of papers, pigments and metals and the techniques used for their production and application. The support will be first described in its codicological characteristics such as dimensions, thickness, colour (expressed in colour coordinates), pattern left from the sieve (observed in transmitted light). The plant used as raw material in paper production will be studied by fibres analysis, while the sizing agents, the pigments, dyes and metals will be analysed by the application of several methods: in particular Reflectography, X-Ray Fluorescence and InfraRed, Raman and Visible Reflectance Spectroscopy. The results of the analyses will be published in an online database that will form an important resource for the material aspects of Persian manuscripts and be a starting point for establishing a first chronology of the materials and techniques used in Persian manuscript production. Preventive and therapeutic strategies against biofilm infections commonly involve applications of multiple antimicrobial substances: antimicrobial coatings on the implanted biomaterials in combination with systemically administered antibiotics. While this practice of combination therapy harbours the risk of developing cross-resistance, it might also provide the possibility to implement specific antimicrobial-antibiotic combinations (AACs) that can slow down the selection of antibiotic resistant strains. Hypothesis and aims: Specific AACs can exert combinatorial effects on the growth of susceptible and antibiotic-resistant Pseudomonas aeruginosa that either suppress or increase their individual effects. Our aim is to identify AACs with antagonistic or synergistic effects on pseudomonal biofilms and to understand their impact on selection of resistant strains. Specifically, we want to identify AACs that select for and against antibiotic resistance during biofilm formation. Methodology: We screened for AACs that cause antagonistic or synergistic effects on planktonic P. aeruginosa. To study the effect of antimicrobial-antibiotic exposure on resistance selection in bacterial biofilms, we will grow resistant and sensitive strains on PDMS surfaces with and without antimicrobial coatings and expose them to antibiotics. Results: Several combinations with synergistic or antagonistic interaction on the growth rate of P. aeruginosa were detected. We observed a strong antagonism when combining the antimicrobial substance chlorhexidine with the carbapenem drug meropenem. A meropenem-resistant mutant showed a selection advantage in low concentrations of chlorhexidine combined with a sub-inhibitory concentration of meropenem over the wild-type. No antagonistic effect was observed for the same combination when E. coli was exposed to chlorhexidine and meropenem, suggesting a non-chemical basis for the observed effect on P. aeruginosa. Conclusion: Gaining a better understanding about resistance selection during biofilm formation on biomedical surfaces will enable us to mitigate against biofilm-associated antimicrobial resistance. T1 - Thermoresponsive Amphiphilic Functionalization of Thermally Reduced Graphene Oxide to Study Graphene/Bacteria Hydrophobic Interactions N2 - An understanding of the interactions of 2D nanomaterials with pathogens is of vital importance to developing and controlling their antimicrobial properties. In this work, the interaction of functionalized graphene with tunable hydrophobicity and bacteria is investigated. Poly-(ethylene glycol)-block-(poly-N-isopropylacrylamide) copolymer (PEG-b-PNIPAM) with the triazine joint point was attached to the graphene Surface by a nitrene [2 + 1] cycloaddition reaction. By thermally switching between hydrophobic and hydrophilic states, functionalized graphene sheets were able to bind to bacteria. Bacteria were eventually disrupted when the functionality was switched to the hydrophobic state. On the basis of measuring the different microscopy methods and a live/dead viability assay, it was found that Escherichia coli (E. coli) bacteria are more susceptible to hydrophobic interactions than B. cereus bacteria, under the same conditions. Our investigations confirm that hydrophobic interaction is one of the main driving forces at the presented graphene/bacteria interfaces and promotes the antibacterial activity of graphene derivatives significantly. Dabei werden möglichst identische Proben mit identischen Verfahren oder mit unterschiedlichen Verfahren untersucht. Die Teilnahme an Ringversuchen ist für alle akkreditierten Laboratorien ein wesentlicher Baustein der institutsinternen und -externen Qualitätssicherung. Je mehr Faktoren Einfluss auf das Endergebnis haben können, desto wichtiger ist es diese Faktoren zu kennen und deren Einflüsse durch weitestgehende Vergleichbarkeit der Messverfahren zu minimieren. Für die Bewertung von Emissionen aus Bauprodukten haben sich Emissionsmess-kammer-Untersuchungen als Messverfahren etabliert. Um den Geruch auch für Innenraumanwendungen bewertbar zu machen sind in den letzten Jahren erste Normen entstanden (z. B. DIN ISO 16000-28 und VDI 4302-1). Um die Eignung dieser Normen und deren Anwendung zu untersuchen sind seit 2012 nunmehr 3 Ringversuche der BAM auch zum Thema Geruch durchgeführt worden. Die bislang durchgeführten Ringversuche mit dem Ziel der Geruchsbewertung wiesen Standardabweichungen von ca. 24 bis 30 % um den jeweiligen Mittelwert auf und wiesen somit ähnliche Werte auf, die auch bei den VOC-Vergleichen ermittelt wurden. This is especially true for P-fertilizers from recycled materials which often have a lower plant-availability compared to commercial P-fertilizers but are expected to play an increasingly important role into the future (Kratz et al. 2019). One promising way to increase the plant-availability of the fertilizer P is a co-fertilization with specific nitrogen (N) forms which can enhance the P uptake and make P-fertilizers from recycled material more competitive to commercial phosphate rock-based P-fertilizers (Rahmatullah et al. 2006; Vogel et al. 2018). To investigate this effect, we performed a pot experiment with three different P-fertilizers (sewage sludge-based, phosphate rock and triple superphosphate) and ammonium nitrate sulfate as a co-fertilizer, without and with a nitrification inhibitor (NI), and analyzed the form of N and P in soil via a suite of chemical and novel X-ray spectroscopic methods. The application of NI with the P and N fertilizers led to a higher dry matter yield and a higher P uptake of maize. Novel N K-edge micro-X-ray absorption near-edge structure (micro-XANES) spectroscopy identified that the application of a NI promotes the temporary formation of a non-exchangeable N in detectable hot-spots in the soil. The subsequent slow release and prolonged availability of N during plant growth leads to higher yield and nutrient uptake. It can be concluded that NIs lead to a temporary fixation of ammonium-N in a pool that can be accessed by plant roots. Those types of available nutrient pools meet the idea of so-called “next generation fertilizers” as plants have access to nutrients according to their current demand. N2 - The earliest known recipes for iron gall inks include four basic ingredients: oak galls – pathological growths of oak leaves; metal salts – usually referred to as vitriol; a binder such as gum Arabic; and water. The final product differs in the elemental composition due to the multitude of recipes as well as differences within the composition of the ink’s ingredients. Nowadays, based on the qualitative and semi-quantitative evaluation of X-ray fluorescence data, it is possible to distinguish inks on the basis of the so-called fingerprint model. The first goal of our study was to determine to what extent the type of XRF spectrometer affects the quality of the ink evaluation. We tested two types of spectrometers, semi-stationary machines equipped with polycapillary focusing optics and a handheld spectrometer with a diaphragm collimator and a relatively big interaction spot. The second goal was to address the issue of whether the ink composition might be affected by storage in a metal container. The presentation will discuss the role of the spectrometer type in the evaluation of a thin layer material such as ink. We have also learned that the iron-gall ink composition might depend on the type of vessel in which ink was being stored. Biodiversity is commonly estimated based on genetic variation. We investigated a new approach that defines and measures biodiversity in complex microbial communities. We used the variation in multiple functionally-relevant, phenotypic traits measured in parallel in single cells as a metric for microbial phenotypic diversity. We studied phenotypic diversity and ecosystem functioning throughout different photosynthetic layers dominated by divergent microbial communities in the gradient of Lago di Cadagno. We determined genetic diversity by 16S and 18S amplicon sequencing and bulk ecosystem functioning (photosynthesis). In addition, we determined phenotypic diversity using single-cell technologies such as nanometer-scale secondary ion mass spectrometry (NanoSIMS) correlated with confocal laser scanning microscopy (CLSM) and scanning flow-cytometry. We measured functional trait variation between individuals in 13CO2 fixation, 15NH4+ uptake, and variation in physio-morphological cell traits, such as cell size, shape, and auto-fluorescence for various pigments related to photosynthesis. We used the distances between individuals in a multidimensional trait space to derive phenotypic trait-based diversity indices, such as trait richness, trait evenness, and trait divergence. We find that phenotypic trait divergence associates with ecosystem functioning, whereas genetic diversity does not. Including activity-based, single-cell phenotypic measurements with NanoSIMS provided an additional accuracy to the trait-based diversity assessment and allowed us to formulate hypotheses on the mechanisms that shape the correlation between phenotypic diversity and eco-system function. Together, our results show that phenotypic diversity is a meaningful concept to measure microbial biodiversity and associate it with ecosystem functioning. The sample environment in the near-ambient pressure (NAP) XPS instrument EnviroESCA allows for measurements in up to 15 mbar water vapor, thus, sample preparation is no longer restricted to UHV-compatible techniques. For instance, biofilms grown in medium can be transferred directly from the medium to the measurements chamber, maintaining a humid environment throughout the measurements. Considering the complexity of bacterial samples, sample preparation must be carefully considered in order to obtain meaningful and reproducible results. In this talk, various strategies for sample preparation of bacteria and biofilms for NAP-XPS measurements will be discussed. Model systems of planktonic bacteria, artificial biofilms resembling the exopolysaccharide matrix and biofilms have been characterised in various conditions. The stability and homogeneity of the samples was assessed by monitoring the C1s core level peak at different sample locations. The quality of the XPS-spectra is also influenced by the gas environment, which will be exemplified by core level spectra of P. Fluorescens acquired in air, water vapor and ultra-high vacuum. A1 - Hahn, Oliver T1 - Non-invasive Identifizierung von Pigmenten in modernen Hinterglasbildern (1905-1955) - Highlights aus der Hinterglasforschung N2 - Im Rahmen des Projektes „Hinterglasmalerei als Technik der Klassischen Moderne von 1905 – 1955“, welches vom Museum Penzberg – Sammlung Campendonk, Penzberg (Gisela Geiger und Diana Oesterle) geleitet wurde und in Zusammenarbeit mit der freischaffenden Restauratorin Simone Bretz, der Bundesanstalt für Materialforschung und -prüfung (BAM) und dem Doerner Institut erfolgte, konnte im Rahmen einer Promotion erstmals ein Konvolut von mehr als 60 Hinterglasbildern mit nicht-invasiven Analysemethoden untersucht werden. Die eingesetzten Verfahren ermöglichten die Identifizierung der Pigmente und eine Klassifizierung der verwendeten Bindemittel. Die materialwissenschaftliche Beschäftigung mit der Hinterglaskunst in ihrer Vielfalt ist ein relativ junges Forschungsfeld. Lange wurde ihr mit Unkenntnis, Missverständnis und sogar Geringschätzung begegnet. Im kunsthistorischen Kontext wurde diese Art der Kaltmalerei zumeist mit dem Begriff der Glasmalerei belegt und nicht als eigenständige Kunstform anerkannt, da in beiden Fällen Glas als Bildträger seinen Einsatz findet. In der Hinterglasmalerei wird die Tafelrückseite mit organisch gebundenen Malfarben verziert und das Werk ausschließlich bei auffallendem Licht betrachtet. Die künstlerische Herausforderung liegt immer in der technischen Notwendigkeit eines umgekehrten Malvorganges: die oberste, durch das Glas sichtbare Schicht muss zuerst auf den Bildträger Glas aufgetragen werden, der optische Hintergrund wird zuletzt aufgebracht. Im Vergleich zum Tafelbild, bei dem die Malerei erst durch den Firnisauftrag Tiefenlicht erhält, entfalten die Farben in der Hinterglasmalerei bereits beim Malen ihre Intensität und erhalten gleichzeitig Schutz durch den gläsernen Bildträger (Bretz et al. 2016). Neben der kunstgeschichtlichen und kunsttechnologischen Bearbeitung der Hinterglasbilder spielten materialwissenschaftliche Untersuchungen an einer Auswahl von 66 Bildern im Rahmen des von der VolkswagenStiftung finanzierten Forschungsprojektes eine zentrale Rolle. Grundsätzlich wurde ein nicht-invasiver Ansatz gewählt, um die fragilen Kunstwerke in-situ zu messen und somit einen Transport zu vermeiden. Die Methodik umfasst neben VIS-Spektrometrie in Reflexion und energiedispersiver Röntgenfluoreszenzanalyse (RFA) auch Infrarotspektroskopie in diffuser Reflexion (DRIFTS) und Raman-Spektroskopie. Mit Hilfe des gewählten methodischen Ansatzes lassen sich sowohl anorganische als auch organische Pigmente identifizieren und die Zusammensetzung von Metallfarben und Folien sowie die Glaszusammensetzung qualitativ bestimmen. Ferner ermöglicht die DRIFT-Spektroskopie eine Klassifizierung der Bindemittel. Die im Rahmen des Forschungsprojektes analysierten Hinterglasbilder können folgendermaßen chronologisch gruppiert werden: 1910-1919 (17 Bilder), 1920-1929 (18 Bilder), 1930-1939 (6 Bilder), 1940-1949 (7 Bilder), 1950-1955 (13 Bilder) und >1955 (5 Bilder). Die Auswertung der Ergebnisse ermöglicht grundsätzliche Aussagen über die verwendeten Malmaterialien in Hinterglasbildern in der ersten Hälfte des 20. Jahrhunderts und zeigt Änderungen der verwendeten Pigmente im zeitlichen Kontext auf. Während der ersten Hälfte des 20. Jahrhunderts kamen viele neue Pigmente und Malmaterialien auf den Markt, wobei speziell die synthetischen organischen Pigmente (SOP) zu nennen sind. Eine Vielzahl an SOP konnte in Hinterglasbildern aus allen Zeitgruppen identifiziert werden, wobei hier vor allem Farblacke aus synthetischem Alizarin (PR83), Vertreter der roten und orangen β-Naphthol Pigmente [(z.B. PR3, PR4, PO5 (in Nächtliche Fahrt, 1921 von L. Hildebrandt nachgewiesen)], die blauen und grünen Kupferphthalocyanine [z.B. PG7, PB15 (taucht ab 1949 in Hinterglasbildern auf)] und einige gelbe Azopigmente [z.B. PY1, PY3, PY12 (in Geneigter Mädchenkopf, 1941 von Oskar Schlemmer gefunden)] aufzuführen sind (Steger et al. 2018; Steger et al. 2019a). Einige seltene SOP der Triarylcarboniumgruppe konnten ebenso nachgewiesen werden. Darunter fallen neben der Identifizierung von PR81 (Ohne Titel, 1954, M. Uhlenhuth) auch die erstmaligen Nachweise von PV2 (Hahn, um 1945, L.-G. Buchheim) und PG1 (Exzellente und einmalige Dressuren, 1945/46, L.-G. Buchheim) in Kunstwerken überhaupt (Steger et al. 2018; Steger et al. 2019a). Ferner konnten mit PB52 aus der Gruppe der Anthrachinone und PR60 aus der Gruppe der verlackten Naphthalin-Sulfonsäure-Pigmente weitere seltene SOP in den Bildern Rudern (um 1912) beziehungsweise Apokalyptischer Reiter II (1914) von Wassily Kandinsky nachgewiesen werden (Steger et al. 2019b). Neben den Entwicklungen der SOP können auch eindeutige Trends in den anorganischen Pigmenten veranschaulicht werden. Seltene anorganische Pigmente wie zum Beispiel Strontiumweiß (SrSO4) in der Kreuzabnahme (1914/15) von Carlo Mense (Steger et al. 2019c) wurden identifiziert. Neu entwickelte anorganische Pigmente wie zum Beispiel Cadmiumrot (Cd(S,Se); ab 1928 in 11 Hinterglasbildern nachgewiesen) oder Titanweiß (TiO2; Anatas kommt ab 1922 vor, Rutil konnte nur in Kissen der Träume, 1976 von Werner Schriefers identifiziert werden) tauchen in einigen Hinterglasbildern auf. Schweinfurter Grün (3Cu(AsO2)2·Cu(CH3COO)2) oder Strontiumgelb (SrCrO4) wurden in einigen Bildern bis etwa um 1925 gefunden; diese Pigmente konnten dagegen in späteren Bildern nicht mehr nachgewiesen werden. Als Beispiel sind hier vier frühe Hinterglasbilder (1909–1914) von Wassily Kandinsky zu nennen, wobei Strontiumgelb in vielen Bildpartien zu finden war (Steger et al. 2019b). Publizierte materialwissenschaftliche Ergebnisse von Kandinskys Leinwandgemälden aus späteren Werksperioden zeigen, dass Strontiumgelb zu dieser Zeit vollkommen aus seiner Palette verschwunden ist und vor allem durch ein anderes Gelbpigment, nämlich Cadmiumgelb, ersetzt wurde (McMillan et al. 2013). Eine materialtechnische Besonderheit in der Hinterglaskunst ist die Verwendung von Metallfolien und Metalleffektpigmenten. Metallfolien aus Silber, Zinn, Aluminium oder Messing wurden vielfach als künstlerisches Element in die Komposition integriert, wobei sie als finale Schicht auf, die bereits gestaltete, leicht transparente Malerei geklebt wurden. Durch die Betrachtung im auffallenden Licht erzeugten die Folien bemerkenswerte Glitzereffekte. Zinn- und Aluminiumfolien wurden zum Beispiel in Rudern (um 1912, W. Kandinsky) nachgewiesen, während Messingfolien häufig in Bildern von L.-G. Buchheim gefunden wurden. Die Zinnfolien wurden im 19. und frühen 20. Jahrhundert für Verpackungszwecke (z.B. Zigaretten, Schokolade) verwendet. Aluminiumfolie wurde erst ab 1910 industriell in der Schweiz hergestellt und zu Beginn vor allem für Schokoladenverpackungen eingesetzt (Skrabec 2016). Die frühe Verwendung solche Folien in Hinterglasbildern (z.B. Rudern, um 1912) zeigen eine rasche Verbreitung dieses Materials bei den Künstlern an. Unter Metalleffektpigmenten versteht man feingemahlene Pulver aus Metall (z.B. Zinn, Nickel, Kupfer) und Legierungen (z.B. Messing, Neusilber) mit unterschiedlichen Tönungen, welche bei seitlichem Lichteinfall auf Hinterglasbildern ein merkbares Funkeln erzeugen. Solche Metalleffektpigmente wurden vielfach bei Werken von Heinrich Campendonk nachgewiesen (Geiger und Bretz 2017) und konnten in allen chronologischen Gruppen gefunden werden. Grundsätzlich ermöglichen die Ergebnisse ein besseres Verständnis für die Technik der Hinterglasmalerei und helfen insbesondere, diese eigenständige Kunst im Hinblick auf die „klassische“ Tafel- bzw. Leinwandmalerei abzugrenzen. So zeigen Vergleiche mit Gemälden auf Leinwand von Wassily Kandinsky deutliche Unterschiede in den verwendeten Farbmitteln für beide Techniken. Die Anzahl von 66 gemessenen Hinterglasbildern von 39 Künstlern erscheint im Kontext von über 1200 Arbeiten, welche im Rahmen des Projektes lokalisiert werden konnten, gering. Allerdings können die durchgeführten Untersuchungen als Startpunkt für zukünftige Projekte in diesem Forschungsfeld, beispielsweise für einzelne Werkkomplexe verstanden werden. Die Forschungsergebnisse werden auf die Webseite www.hinterglas-klassischemoderne.de eingestellt. Im Sommer 2020 präsentiert das Museum Penzberg – Sammlung Campendonk Hinterglasbilder von 1910 bis 1960 als Abschlussausstellung des Forschungsprojektes. Die Ausstellung wird von einem ausführlichen Katalog über die fächerübergreifende Forschung begleitet. N2 - Brazilian farming industry consumed around 2.2 million tons of phosphorus (P) fertilizers in 2016. The agricultural industry depends on imported P fertilizers and over 98% of P fertilizers were used for sugarcane, soybean and maize production. An alternative is to use P from sugarcane bagasse. Bagasse is the fibrous plant material remaining after extraction of sugarcane juice, and it is combusted for energy production. Remaining ash con-tains up to 0.6 wt% P. The use of bagasse ash (BA) as P fertilizer could decrease the annual import of P fertilizers by 6% of the imported P fertilizer based on 2016 values. Since the bioavailability of P from BA to plants is poorly investigated, this study addresses the effects of (i) gasification tempera-ture (710-849°C), (ii) processing method (gasification vs. combustion), (iii) biomass modifications by co-processing bagasse with chicken manure (BA+CM), and (iv) the soil (Brazilian Oxisol soil vs. nutri-ent poor substrate) on the bioavailability of P from BA to soybeans (Glycine max). Gasification of BA at 806 °C resulted in significantly highest uptake of P by soybeans and was around 0.33 mmol after 51 days growing. The bioavailability of P significantly increased due to co-gasification of bagasse and chicken manure (BA+CM) and the soybeans took up around 16% more P. Compared to the nutrient-poor substrate, the bioavailability of P in BA+CM ash treated Oxisol soil was signifi-cantly lower by 46% and there was no significant effect of processing method on the bioavailability of P from the BA+CM ash to soybeans. Contrary to the Oxisol soil, the bioavailability of P from co-combusted BA+CM ash was significantly higher compared to co-gasified BA+CM ash. In conclusion, co-processing of bagasse with nutrient rich residues can increase the value of BA as P fertilizer. The bioavailability of P from ash to plants depends on the P forms. Mineralogical analyses of ash P forms by NMR and X-ray diffraction are in progress and will be presented at the conference. Um die potentiellen Emissionen zu minimieren wurde ein Messverfahren entwickelt, welches inzwischen als BEMMA-Schema (Bewertung von Emissionen aus Materialien für Museumsausstattungen) in die Museumswelt Einzug genommen hat. Die erfolgreiche Bewertung der für den Bau verwendeten Materialien gemäß BEMMA-Schema resultiert nicht in einer emissionsfreien Vitrine, sondern soll bei der Auswahl geeigneter Materialien zur Herstellung möglichst emissionsarmer Vitrinen helfen. Konkrete handwerkliche Durchführungen beim Bau, Dichtheits-anforderungen, wie auch die Aufstellbedingungen (Umgebung der Vitrine) beeinflussen die Emissionscharakteristik der fertigen Vitrine in der Praxis. Eine saubere Vitrine hilft nichts in einer belasteten Umgebung oder auch bei emittierenden Exponaten. Inzwischen liegen Erfahrungen mit dem Schema seit 2012 vor. Der Messablauf sieht vor, dass von den Materialien kleine Muster mit der Mikrokammer (Markes) innerhalb von 2 Tagen untersucht werden. The formation of hydrogen sulfide (HS-) is particularly concerning for the petroleum industry due to its corrosiveness. However, the activities of SRB are limited to the concentration of sulfate present in the environment, whereas methanogens can utilize substrates such as H¬2 for methanogenesis. MA is commonly found in sulfate-free environments, such as deep sediments, and are known to form interspecies electron transfer relationships with SRB. Recently, SRB and MA capable of microbiologically influenced corrosion (MIC) by using elemental iron as a direct electron source (EMIC) have gained increased attention. On the iron surface, EMIC-SRB can outcompete EMIC-MA in the presence of sulfate, but this changes as sulfate depletes. The formation of FeS on the metal surface can be further utilized by MA for methanogenesis as it provides a conductive path. However, the possible kinetics involved of the overall process are currently unknown. We obtained a co-culture of EMIC-SRB and EMIC-MA to investigate the growth rates and electrical potential changes under different environmental conditions, including changes in pH, temperature and salinity. Results indicate that under neutral conditions and using iron as the sole substrate, methane production (up to 5 mM) starts after sulfate was depleted. Electrochemical measurements will be conducted on the co-culture under different conditions to determine the changes in the electrical potential in correlation with the sulfate and methane concentration. Fluorescence and electron microscope images of the biofilm structure will be used to visualize cell distribution and morphology. This study embarks the first step of understanding the relationship between EMIC-SRB and EMIC-MA. In order to measure the particle size distributions, many techniques are available. 9 methods for particles and 2 methods for fibres have been tested in a prevalidation study and appropriate methods will be compared in an interlaboratory round robin test starting in February 2019. For the reliable determination and assessment of behaviour and effects of nanomaterials as well as for the determination of the exposure of humans and environment a comprehensive physical-chemical characterization of nanomaterials is essential. This is an important prerequisite to identify them as nanomaterials and to interpret and compare test results and - in future – to forecast interaction and effects of nanomaterials. In 2006, the OECD launched a sponsorship program for the testing of nanomaterials in which 11 nanomaterials were thoroughly investigated using a variety of methods. The aim of the project was, among other things, to find out where problems occur and where there are gaps in the measurement and test procedures and where are changes required. An important outcome of the sponsorship program was the finding that the OECD Test Guidelines should in several cases be extended to the specific needs in testing of nanomaterials. The existing standardized test methods of the OECD for physical-chemical characterization have not been developed for nanomaterials in particular. A high demand for an extension of the test guidelines was identified. Germany complied with the OECD's request in 2017 and has agreed to extend the “Test Guideline on Particle Size Distribution / Fiber Length and Diameter Distributions Test Guideline” for Manufactured Nanomaterials (MN). UBA commissioned BAM and BAuA with the preparation of the Test Guideline. The aim of the project is the development of a harmonized test protocol for a valid and reproducible determination of particle size and size distribution which is one of the most relevant physical-chemical properties for MNs. Different measuring methods provide different results for the size distribution of the particles. This is caused by the different measuring principles of the methods. Each method measures a specific parameter that ultimately determines particle size. First, the measured quantity differs for each method (Scattered light intensity, 2D image / projection, electric mobility, etc.). Second, the calculated diameters of the MN may differ (Feret Diameter, Area Projection, Mobility Diameter, Aerodynamic Diameter, Hydrodynamic Diameter). Third, a measuring method provides a size distribution which is measured either mass-based, surface-based or number-based. A conversion between the results requires additional parameters and thus possibly increases the measurement error. In addition to the technical differences, the individual parameters are strongly influenced by the structure and material of the nanoparticles. For example, a surface functionalization can lead to very different results in the size distribution. The suitability of measurement methods differs with the material of the MN. As a result, two very different results can be measured for the particle size distribution using two different methods, which are nevertheless both correct. Several large projects in recent years therefore concluded that nanomaterials should be characterized by at least two complementary method. Imaging techniques are regarded as one of these methods for the characterization, the complementary methods are supposed to be statistical methods. The different results for the size distribution of nanomaterials become problematic for the registration of new MN. A comparable and reproducible size distribution is a prerequisite for a standardized registration. In the future, the particle size distribution in the EU will also decide on the classification of a substance as a nanomaterial or as a non-nanomaterial. Especially in borderline cases, a standardized and comparable measurement methodology is therefore essential. In the data evaluation the length and diameter of each fiber will be determined concurrently to allow for application of different regulatory definitions. The quality of the results critically depends on the sample preparation as well as the data evaluation. In this step the classification rules have to be formulated and followed accurately in order to optimize reproducibility of the method. This MAD principle ensures that, if a chemical is tested under the Good Laboratory Practice (GLP) conditions accordingly to an OECD TG, the data should be accepted in all OECD countries. The TGs have been developed, harmonized, internationally validated (round robin tests) and adopted by OECD countries to be used for the physical-chemical characterisation, fate estimation, and hazard identification for risk assessment of various chemicals. In addition to the TGs, OECD Guidance Documents (GDs) usually provide guidance on how to use TGs and how to interpret the results. But are the existing TGs and the related GDs applicable and adequate for the regulatory testing of nanomaterials? To investigate the corrosion potential of methanogens, we studied the EMIC methanogenic strains isolated from marine sediments (Methanobacterium-affiliated strain IM1) and crude oil tanks (Methanococcus maripaludis Mic1c10), in an in-house developed flow-through cell to simulate a fluctuating environment. A co-culture of M. maripaludis and D. alaskensis was also established to study the effect of syntrophic growth on metal corrosion that may occur in industrial pipelines. Results indicate that the rates of iron corrosion due to coupled methanogenesis (up to 0.4 mm/yr) are higher to that caused by the marine SRM Desulfovibrio alaskensis (0.15 mm/yr). Surface analyses of the metal showed severe pitting with high methane production. Genomic analysis of the EMIC methanogen M. maripaludis Mic1c10 will provide an insight on the mechanisms of MIC. For example, it is intended to enable reusing of soil-like materials containing regulated substances in concentrations between the single and double precautionary values (PV), provided that the limit values for the eluate are complied with. The aim of a running project (funding code UFOPLAN 3716 74 203 0, German Federal Environmental Agency) is to fill data deficits for soil-like materials which meet the above-mentioned criteria to promote circular economy and conservation of natural resources. Different soil-like materials of various origin (for example dredged material, banquet peeling material, urban and meadow soils) are considered with a special focus on PAHs. A major challenge in sample acquisition is the relatively narrow target range of the PAH content between 3 and 6 mg/kg dm considering a high measurement uncertainty at this concentration level. The contents of PAHs, heavy metals and other inorganic elements in solid matter are determined following established standards. In addition, the soil-like materials are characterized concerning particle size, pH, conductivity, SOM, TOC and carbonate content. To determine the eluate concentration, column tests according to DIN 19528 and batch tests according to DIN 19529 at a liquid/solid ratio (L/S) of 2 l/kg and 10 l/kg are performed comparatively. The results show that the new requirements of the planned amendment to the Federal Soil Protection and Contaminated Sites Ordinance certainly opens additional possibilities for reuse of soil-like materials. Even most soil-like materials within 2-3 x PV comply with the eluate value for PAH (0.2 μg/l) both in the batch test and in the column test at L/S 2 l/kg. Selected inorganic substances lead to similar evaluations. Therefore, the application of such soil-like materials outside of rootable soil layers would be possible. Previously, those materials were often used for landfill construction. In the future, a utilization in the adjacent environment by backfilling, recultivation, renaturation, composting or landscaping would be conceivable. Die Paragraphen 6 bis 8 der vorgesehenen Novelle der BBodSchV enthalten neue Vorgaben hinsichtlich des Auf- und Einbringens von Bodenmaterial unterhalb oder außerhalb einer durchwurzelbaren Bodenschicht (4. Referentenentwurf Mantelverordnung, Stand: 06.02.2017). Es soll die Möglichkeit eingeräumt werden, Bodenmaterialien mit Feststoffgehalten von regulierten Stoffen bei Überschreitung des Vorsorgewertes einer weiteren Verwertung zuzuführen unter der Voraussetzung, dass die Eluatwerte eingehalten werden. Für die Auswirkung dieser neuen Regelungen auf die Verwertung von Bodenmaterialien bestehen bisher noch Datendefizite besonders für organische Stoffe. Durch die Umstellung des Elutionsverfahrens von einem W/F von 10 l/kg (bisherige Regelung in der LAGA M 20) auf ein W/F von 2 l/kg bestehen zusätzliche Unsicherheiten im Hinblick auf die Einstufung von Bodenmaterialien zu Verwertungszwecken. Im Rahmen eines UFOPLAN-Projektes (FKZ 3716 74 203 0) wurden unterschiedliche Bodenmaterialien verschiedenen Ursprungs (z.B. Baggergut, Bankettschälgut, Stadt- und Auenböden, Bergematerial) mit besonderem Fokus auf PAK untersucht. Von den unter die Kriterien fallenden, ausgewählten Bodenmaterialien wurden Feststoffgehalte, TOC-Gehalte sowie die Eluatwerte aus Säulenversuchen nach DIN 19528 und Schüttelversuchen nach DIN 19529 bei einem Wasser-/Feststoffverhältnis (W/F) von 2 l/kg und 10 l/kg ermittelt und gegenübergestellt. To improve conservation practices of Arabic manuscripts we have assessed the feasibility of the recipes collected from many Arabic sources on bookmaking and reproduced the most common inks. Ink samples were then artificially aged and analysed using an array of analytical techniques, mostly non-invasive, in order to build a database of Arabic inks and their ingredients and also to verify the detection limits of the portable equipment employed. First, we will provide an overview of the sources [1-2] employed in the study – dating from 9th to 14th century, although the manuscripts in which they can be found dates up to the 20th century – with an eye on the ink typologies (real and perceived by the compilers). Then we will show how, by reproducing the recipes, it was possible to shed light on some oddities in the procedures and the choice of ingredients. In the end we will discuss problems concerning the identification of ink types and ingredients using reflectography (in Vis, IR and UV), X-Ray Fluorescence, Infrared and Raman spectroscopy. The focus will be in particular on mixed inks – both in the form of carbon inks with the addition of tannins and of a mixture of carbon and iron gall inks – which are particularly difficult to detect [3]. We will also examine the results of iron gall inks made with iron filings or nails instead of vitriol and the various precursor used to extract gallic acid that are mentioned in the sources as surrogates of gall nuts. Unfortunately, the scientific investigation of Arabic books is still uncommon, focusing mainly on single case studies, often producing unclear results, especially for black inks. This lack of knowledge prompted a recent research in which black ink recipes have been collected from written Arabic sources on bookmaking, their feasibility has been assessed, and finally some of them have been reproduced. These samples were artificially aged and analysed through an array of analytical techniques, mostly non-invasive and non-destructive, in order to build a database of Arabic inks and their ingredients and also to verify the detection limits of the portable equipment employed. In this talk I will present some instances of how this approach can help textual criticism, for example in evaluating variants and determining the competence of authors and compilers. At the same time the research highlights how much this scientific work depends on textual studies, especially concerning the identification of ingredients. Moreover, I will focus on some of the problematics I faced both during the textual interpretation and the actual reproduction of the recipes and how they influenced the results of the scientific analyses. In particular I’ll look into the role played by the degree of purity of the components and the function (real and perceived) of some of the ingredients. Quarantine of potentially infested objects can be logistically challenging. Anoxia under controlled Nitrogen atmosphere is a most gentle but also time-consuming method to eradicate insect pests in all kinds of different materials. Treatment results are usually affected by duration, temperature, humidity and residual oxygen content. During a two-year research project, 34 relevant pest insect species of all developmental stages were tested in different materials (wood, paper, wool) to monitor treatment success and to determine optimum treatment parameters. Duration of treatment ranged from one to three weeks at temperatures of 20–27 °C. As expected, results showed significant differences in mortality among tested species. Highest tolerance of hypoxic conditions was found in elder larvae of Hylotrupes bajulus. Although this species is an unlikely museum pest, it may serve as an overall most tolerant reference. Anobiids and other wood boring beetles are more often an issue related to cultural heritage. A combination of three weeks exposure time at maximum 0.5% residual oxygen and 24 °C, alternatively 1% residual oxygen and 27 °C are recommended for infested artefacts. Imbedding materials in general had no influence on mortality. This study was funded by Deutsche Bundesstiftung Umwelt (DBU). Their management is essential to protect cultural ethnological heritage and natural history collections for future generations. Pest management strategies have changed over time. Today, intensive knowledge on pest biology and overall material science are key cornerstones in IPM concepts - also for the museum environment. The important first steps for sustainable pest management are risk assessment, early pest detection and identification of pathways of infestation. These steps are followed by physical and biological means of control, which have lately gained more importance than applying biocides. Several potentially effective biological enemies of clothes moths and woodworm have been known for a long time, but their promotion for pest control in museums and historic houses is just beginning. This short review summarizes current concepts of pest life cycle interruption by applying good quarantine and very specific biological measures. The lessons learned from recent faunistic surveys, life-history studies as well as behavioral observations of parasitoids and predators of clothes moths and woodworm may supplement the pest management tool box. The need for further research in this field is addressed. In their roles as mineral weathering agents and mutualistic partners of phototrophs, fungi supply primary producers like plants and phototrophic microorganisms with mineral-derived nutrients. The exact mechanisms behind fungus-induced mineral weathering processes are however not well understood. Progress can be achieved here by reproducible experimental simulations of the natural processes, using well-characterised model organisms and minerals. In this study, the weathering-affecting, rock-inhabiting fungus, Knufia petricola A95 and the Fe-bearing olivine (Fe0.2Mg1.8SiO4) were selected to investigate fungi-induced effects on mineral dissolution. The availability of a melanin-deficient mutant (ΔKppks) of K. petricola A95, that produced more extracellular polymeric substances (EPS) than the wild type (WT), enabled comparative studies of the role of melanin and EPS in weathering processes. Three experimental systems, which generate long-term microbiological stability, were developed to study the impact of the WT and ΔKppks on olivine weathering: (1) batch and (2) mixed flow dissolution experiments, and (3) biofilm cultivation experiments. In addition, state-of-the-art analytical techniques were used to monitor changes in the growth medium, as well as of the mineral surface and biofilm-mineral interface. Inductively coupled plasma optical emission spectrometry (ICP-OES) analysis of the Mg, Si and Fe concentrations in the reacted growth medium was used to quantify olivine dissolution. In abiotic controls, Mg and Si dissolved congruently, while Fe precipitated. The measured olivine dissolution rates at pH 6 were two orders of magnitude lower than previously reported, but similar at acidic pH. X-ray photoelectron spectroscopy (XPS) analyses of the olivine surface confirmed the presence of Fe (oxyhydr)oxide precipitates. Transmission electron microscopy (TEM) imaging of an abiotically reacted polished olivine section from the long-term cultivation experiment showed the presence of an amorphous layer enriched in Fe. All these observations indicate that the precipitation of Fe (oxyhydr)oxides on the olivine surface inhibits olivine dissolution. Both tested rock-inhabiting fungal strains affect Fe precipitation as well as olivine dissolution. Evaluation of the WT and ΔKppks revealed that the WT formed less biomass but could take up higher amounts of metals (e.g. Fe) and was more efficient in its attachment to olivine. The WT and ΔKppks enhanced olivine dissolution as demonstrated by higher Mg and Si concentration in the reacted growth medium. They furthermore prevented Fe precipitation by binding Fe and retaining it in solution, thereby allowing olivine dissolution to proceed. The WT cells that were attached to the olivine surface were particularly efficient at inhibiting Fe precipitation. By binding Fe directly at the olivine surface, the WT cells removed the inhibition of olivine dissolution almost completely. TEM analysis of polished olivine sections, colonised by a fungal biofilm for seven months, supported this hypothesis. After long-term fungus-olivine interaction, the Fe-enriched, amorphous layer did not develop, and the olivine surface was stronger etched compared to the abiotic control. To study the effect of mutualism on mineral weathering, K. petricola was grown with the cyanobacterium, Nostoc punctiforme ATCC 29133. Both partners showed an enhanced growth and formed a stratified biofilm which attached more strongly to olivine. Nevertheless, the olivine dissolution rate of the fungus-cyanobacterium consortium was moderate. Rock weathering simulation systems developed here are promising research instruments. The experimental conditions allow for the alteration of the studied mineral surface, while the clear definition of these conditions delivers a stable growth of microorganisms. The latter makes these systems universally applicable, especially in combination with integrative multidisciplinary analytics. Processes underlying environmental and biological effects on rock weathering, metal corrosion, plastic degradation, or the deterioration of any other substrate can be studied reproducibly and over a long period of time. The chemical and biological complexity of these simulation systems mimics natural rock weathering processes. The mineral dissolution rates generated in this study are therefore relevant to natural ecosystems. Their application in clinical l settings is an integral part of antibiotics stewardship by preventing the occurrence and spread of infections. However, improper use of disinfectants also harbours the risk for the evolution of tolerance and resistance to disinfectants, but also to antibiotics. It is therefore crucial to understand whether and how bacteria can survive chemical disinfection and which conditions facilitate the evolution of tolerance and resistance. Here, we study the heterogeneity in the response of isogenic E. coli populations exposed to different levels of commonly used disinfectants. At concentrations below the minimal inhibitory concentration (MIC), we find that certain disinfectants induce prolonged lag times in individual cells, a phenotype that has been associated with persistence against antibiotics. At concentrations above the MIC, we find heterogeneous killing for a range of the tested substances. Interestingly, for the three cationic surfactants that were tested, we find kill kinetics revealing the presence of a tolerant subpopulation that can withstand disinfection longer than most of the population. We will present results from an ongoing evolution experiment in which we test the potential for evolution of population-wide tolerance and resistance through intermittent exposure to lethal doses of a cationic surfactant. It is used as an osmolyte helping microorganisms to maintain an osmotic equilibrium. In addition, ectoine acts as a stabilizer and protects proteins, membranes and whole cells against detrimental effects such as freezing and thawing, drying and high temperatures. Its protective effect is explained by the preferential exclusion model, which postulates that ectoine does not directly interact with biomolecules but is excluded from their surface. Interestingly, details on the interaction of ectoine with DNA are still unknown. Therefore, we studied the influence of ectoine on DNA and the mechanisms by which ectoine protects DNA against ionizing radiation. To emulate biological conditions, we used a sample holder comprising a silicon chip with a Si3N4 membrane, which allows for electron irradiation of DNA in aqueous solution. Analysis by atomic force microscopy revealed that without ectoine, DNA was damaged by irradiation with a dose of 1,7 +/-0,3 Gy. With ectoine, DNA remained undamaged, even after irradiation with 15 Gy. Simulations with dsDNA and ectoine in water revealed a preferential binding of the zwitterionic ectoine to the negatively charged DNA. According to the simulations, binding of ectoine will destabilize dsDNA. Destabilizing is probably caused by the transition of B-DNA to A-DNA and will reduce the DNA melting temperature, which was experimentally proven. The preferential binding provides a stable ectoine shell around DNA, which allows ectoine to reduce OH-radicals and electrons near the DNA and thereby mitigating the damaging effect of ionizing radiation. Sophisticated mechanical methods for unrolling the best-preserved scrolls have been applied, with varying success. However, such processes have been abandoned, to prevent risk from irremediable damage or loss and to preserve the integrity of the extremely fragile rolls. Following the development of X-ray based non-invasive techniques, attempts to virtually unroll the scrolls were made. The most common ink in Antiquity was carbon-based, and the main element of carbonized papyrus is carbon, making these investigations difficult. However, some attempts with synchrotron X-ray phase-contrast tomography (XPCT) were successful. Recently, the identification of antique inks containing metals raised hope that if some of the inks contain metal the rolls can be virtually unrolled using conventional CT- technique. In addition to iron, calcium, carbon, and silicon they usually contain high concentrations of heavy metals such as zinc, lead, and cadmium that are potentially hazardous to the environment, rendering disposal in landfills ecologically problematic and costly. Consequently, pyrometallurgical, hydrometallurgical, and hybrid methods for selective elimination of non-ferrous heavy metals from BF sludge and EAF dust have been conceived, of which only the carbothermic reduction route taken in the so-called Waelz rotary kiln process has been proven to be economically successful. However, this process has several drawbacks regarding efficiency of heavy-metal removal and recovery of iron, and it does not allow processing of BF sludge. In this study, we investigated the efficiency and feasibility of selective chlorination and evaporation of non-ferrous heavy metals, particularly zinc and lead, in both BF sludge and EAF dust as an alternative, thermochemical processing route. To this end, hydrochloric acid and iron(II) chloride solution have been used as chlorinating agents, and the process of heavy-metal chlorination and evaporation has been investigated under inert operating conditions, at variable chlorine concentrations, and at temperatures between 500 and 1200 °C. High zinc and lead removal efficiencies of > 99.5 % were achieved with both chlorinating agents, but iron(II) chloride turned out to be overall more efficient for removal of zinc and lead from BF sludge and EAF dust. Interestingly, and in contrast to previous studies, the iron was completely retained in the processed solid residue, therefore rendering the processed residues virtually zinc- and lead-free raw materials that may either be used internally (e.g., feeding processed BF sludge and EAF dust back into the respective furnaces) or externally (e.g., for cement production). Series of melting experiments were carried out in a pilot-scale electric arc furnace to study how the amount, the grain size and the way of feeding affect the activity of carbon as a reducing agent. During the pyrometallurgical treatment refractory metals such as tantalum and niobium are reduced to their carbide form and enriched in the molten iron-based metal phase. The cooled down slag and metal phase were analysed to investigate thermodynamic and kinetic conditions of the carbide formation. FACT Sage simulations were also used to investigate the material system in state of thermodynamic equilibrium. Results show that mass transfer and kinetics may play an important role if compared to equilibrium analyses using FACT Sage. A1 - Hahn, Oliver T1 - A complementary spectroscopic approach for the non invasive in situ identification of synthetic organic pigments in modern reverse paintings on glass N2 - This work addresses the identification of synthetic organic pigments (SOP) in ten modern reverse paintings on glass (1912-1946) by means of an in-situ multi-analytical approach. The combination of the complimentary properties of mobile Raman spectroscopy and diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) enabled the detection of sixteen SOP even in complex mixtures with inorganic compounds and binders. For the β-naphthol pigments, both Raman and DRIFTS yield appropriate results. DRIFTS was the preferred method for the detection of synthetic alizarin (PR83). Its diagnostic band pattern even allows its detection in complex mixtures with mineral pigments, binders and fillers. Raman spectroscopy yielded distinctive spectra for the triaryl carbonium pigments (PG1, PV2, PR81) and the two-yellow azo SOP (PY3, PY12), whereas DRIFT spectra were affected by extensive band overlapping. This may also occur in Raman spectra, but in less problematic amounts. Fluorescence is the major problem with Raman and it significantly hampers the SOP spectra even with the 785 nm laser. On the one hand the big spot size of DRIFTS (10 mm) limits the technique to rather large sampling areas, whereas the use of a 50× objective for in-situ Raman measurements permits a focus on small spots and aggregated SOP flakes. Moreover, “environmental” factors like temperature changes, artificial light, limited space and vibrations when people pass by need to be considered for in-situ measurements in museums. Finally, the results show the experimental use of SOP in modern reverse glass paintings. Among several rare SOP (e.g. PB52, PR81), two of them (PG1, PV2) have never been reported before in any artwork. T1 - First insights into Chinese reverse glass paintings gained by non invasive spectroscopic analysis N2 - A non-invasive methodological approach (X-ray fluorescence (XRF), diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS), Raman spectroscopy) has been carried out to identify the pigments and classify the binding media in two Chinese reverse glass paintings (The Archer, Yingying and Hongniang) from the late 19th and early 20th centuries. The results reveal a combined use of traditional Chinese and imported European materials. Several pigments like cinnabar, lead white, orpiment, carbon black and copper-arsenic green (probably emerald green) were found in both paintings; red lead, artificial ultramarine blue, Prussian blue and ochre appear in at least one of the paintings. The presence of portlandite (Ca(OH)2) along calcite (CaCO3) in the fine-grained, white backing layer of Yingying and Hongniang indicates the presence of limewash. In Chinese tradition, limewash was produced from clamshells, and was then sold as clamshell white. In contrast to the Japanese pigment, Chinese clamshell white was made of finely grounded shells, which were heated over a low fire. The residue (CaO) forms portlandite (Ca(OH)2) when water is continuously added. This water-rich mixture is applied on the painting. Portlandite reacts with atmospheric CO2 during drying and forms fine-grained calcite (CaCO3) [1,2]. The identification of emerald green (The Archer) suggests an earliest manufacturing date in the 1830s [3] and promotes the sinological dating of the painting. Drying oil was classified as a binding media in most areas of both paintings. However, the orange background of The Archer yielded prominent bands of both proteinaceous and fatty binder. T1 - EDX-Analysis on medieval glasses and innovative protection of stained glass panels N2 - The church of Koszewko (Poland) is a brick building edified in the 15th century built on cobblestone foundations. There are five windows in the sanctuary. Three of them enclose heraldic panels from the Küssow’s family from the 15th century which are surrounded with Goethe glass from the 18th century to complete the windows. The colored heraldic panels are strongly damaged and corroded with massive paint layer losses, glass- and leadbreakages. Those medieval glass fragments have been shortly discovered and are of particular interest for Poland since only few medieval glazing have been conserved. The damages as well as the glass compositions have been investigated with ESEM/EDX. Two categories of medieval glass compositions have been identified. The blue glass is particularly sensible to corrosion because of his high content in K2O. The colorless and the red glass samples belong to a stable glass type. Due to the thickness of the gel layer, it is easy to see that the degradation is strongly proceeded. The protection of those medieval stained-glass panels is absolute necessary. The medieval panels have been restored and surrounded from a copper frame. Then they have been fixed on the wood frame in the church. The exterior glazing has been closed with a panel of Goethe glass. The gap between the Goethe- and the medieval glass is about 3 cm. The Goethe glass panel has been stabilized with a film based on polyester to protect the medieval glasses against any damages. In this way, a low cost protective glazing has been installed for a long-term conservation of each medieval stained-glass panels. The climate measurements over the period of one year on the restored windows are in process. The temperature and the relative humidity are recorded in the church interior, in the gap between the original and the Goethe glass and outdoors. In collaboration with the other two service projects, Z01 and Z03, it aims at bridging the gap between humanities and natural sciences and technology. To that purpose, we set up a laboratory with a range of high-end instruments, most of them mobile, allowing thorough non-destructive analysis of manuscripts. In addition to working on constantly improving the laboratory and the methods of analysis, a substantial part of our activities is dedicated to service, by supporting different research projects conducted at the centre. In this talk, we will present our equipment and the possibilities offered by the different techniques available regarding the different kinds of missions: typology and classification of inks, provenance studies, recovery of faded inscriptions and palimpsests, reconstruction of the history of manuscripts, authentication and dating. We will give a brief overview of our past and ongoing activities in the frame of the second phase of the CSMC. Finally, a selection of three projects will be presented in greater detail to highlight the possibilities of our laboratory and the diversity of missions which can be carried out. SRM act upon the metal by the re-activeness of hydrogen sulfide (HS-), and by withdrawal of the available electrons in electrical contact with the metal (EMIC). Methanogenic archaea (MA) can also cause MIC (Mi-MIC). Several MAs were identified to be corrosive by using elemental iron as the sole electron donor for methanogenesis, including Methanobacterium­-affliated IM1 and Methanococcus maripaludis Mic1c10. Currently, low corrosion rates were reported for MA, possibly due to the formation of siderite (4Fe + 5HCO3- + 5H+ ® 4FeCO3 + CH4 + 3H2O). Since MA do not produce HS-, withdrawal of electrons may be their main corrosive mechanism; however, mechanistic details and kinetics of the overall process are poorly understood. To investigate the corrosion potential of MA, we studied the EMIC methanogenic strains (IM1 and Mic1c10) individually or part of a syntrophic co-culture with SRM. Corrosion studies were conducted using an in-house developed flow-through system to simulate fluctuating environmental conditions. Results indicate that the rates of iron corrosion by MA (up to 0.4 mm/yr) are higher to that caused by the marine SRM Desulfovibrio alaskensis (0.15 mm/yr) and the co-culture (0.1 mm/yr). Scanning electron microscopy (SEM) images of the metal incubated with MA showed severe pitting corrosion. Genomic analysis of the EMIC MA was conducted to provide an insight on the possible cellular mechanisms that could be involved. Furthermore, low concentrations of MA-targeting biocides will be applied to EMIC MA in static and flow conditions to gain insights for possible mitigation strategies. Such knowledge and deeper understanding also from an electrokinetic point of view may not only provide further models in microbial electrophysiology, but also contribute to mitigation strategies in MIC. T1 - Influence of carbon feeding on a pyrometallurgical Ta and Nb recycling process and the formation of carbides N2 - An existing pyrometallurgical process for tantalum and niobium recovery, mainly from low grade pyrometallurgical residues, was investigated. Series of melting experiments were carried out in a pilot-scale electric arc furnace to study how the amount, the grain size and the way of feeding affect the activity of carbon as a reducing agent. During the pyrometallurgical treatment refractory metals such as tantalum and niobium are reduced to their carbide form and enriched in the molten iron-based metal phase. The cooled down slag and metal phase were analysed to investigate thermodynamic and kinetic conditions of the carbide formation. FACT Sage simulations were also used to investigate the material system in state of thermodynamic equilibrium. Results show that mass transfer and kinetics may play an important role if compared to equilibrium analyses using FACT Sage.
