T1 - Supplementary comparison study - measurement capabilities for the quantification of ethanol in water N2 - The accurate quantification of ethanol in water is essential for forensic applications such as blood and breath alcohol testing and for commercial applications such as the assessment of alcoholic beverages. The intercomparison EURAMET.QM-S14 is part of a capacity building project named ALCOREF “Certified forensic alcohol reference materials” that is running within the European Metrology Programme for Innovation and Research (EMPIR). The intercomparison should allow project partners and other interested National Metrology Institutes (NMIs) and Designated Institutes (DIs) to benchmark their analytical methods for the quantification of ethanol in water. The study plan was agreed by the European Association of National Metrology Institutes (EURAMET) Subcommittee Bio- and Organic Analysis (SCBOA) and the Organic Analysis Working Group (OAWG) of the Comité Consultatif pour la Quantité de Matière (CCQM) in February and April 2019, respectively. The intercomparison was coordinated by BAM. Two concentration levels relevant for the calibration and verification of evidential breath alcohol analysers were distributed to study participants. Fifteen institutes from 15 countries registered for the intercomparison and returned results. Participants mostly applied gas chromatography with flame ionisation detection (GC-FID) or mass spectroscopy (GC-MS), one participant used titrimetry and one participant employed a test bench for breath analyser calibration (“bubble train”). Participants did either in-house purity assessment of their commercial ethanol calibrants by Karl-Fischer titration, chromatographic methods, quantitative nuclear magnetic resonance spectroscopy (qNMR) and/or density measurements; or they used ethanol/water Certified Reference Materials (CRMs) from NMIs/DIs for calibration. CCQM OAWG agreed to use a consensus value from participants results that utilizes the reported uncertainties as Key Comparison Reference Value (KCRV). The Gaussian Random effects model with Hierarchical Bayesian solution (HB-REM) is a reasonable approach in this case. The KCRVs and Degrees of Equivalence (DoEs) were calculated with the NIST consensus builder version 1.2 Hierarchical Bayes procedure. Successful participation in the interlaboratory comparison has demonstrated the capabilities in determining the mass fraction of ethanol in aqueous matrices in the range 0.1 mg/g to 8 mg/g. Fourteen out of 15 participants have successfully quantified both samples, one participant successfully quantified only the lower-level (0.6 mg/g) sample. T1 - Key comparison CCQM-K78.b - non-polar analytes in organic solvent: methoxychlor and trifluralin in acetonitrile N2 - The CCQM-K78.b key comparison was coordinated by the Bureau International des Poids et Mesures (BIPM) on behalf of the CCQM Organic Analysis Working Group (OAWG) of the 'Comité Consultatif pour la Quantité de Matière' (CCQM), for National Measurement Institutes (NMIs) and Designated Institutes (DIs) providing measurement services in organic analysis under the 'Comité International des Poids et Mesures' (CIPM) Mutual Recognition Arrangement (MRA). This key comparison was conducted as a 'Track A' comparison within the OAWG's 10-year strategic plan. The goal of CCQM-K78.b was to underpin capabilities for the value assignment of calibration solutions containing low polarity/non-polar organic analytes in organic solvents. The selected model system consisted of a two-component pesticide solution in acetonitrile, comprising methoxychlor and trifluralin. Participants were tasked with assigning the mass fractions, in units of μg/g, of methoxychlor and trifluralin in acetonitrile solution. The mass fraction levels and analytical challenges of the selected analytes were representative of those encountered for calibration solutions of non-polar organic analytes. Participation in CCQM-K78.b allowed for the benchmarking of capabilities for assigning the mass fraction of non-polar organic compounds (pKow < -2) in solution, at mass fractions above 5 μg/g, in an organic solvent. Additionally, the comparison assessed the capabilities for the quantitative assignment of thermally labile compounds. Participants were provided by the BIPM with ampoules containing methoxychlor and trifluralin in acetonitrile. Each participant reported the mass fraction content of each analyte in μg/g. All participants ensured the metrological traceability of their results through the use of a Primary Reference Material (PRM), which was used to prepare a primary calibrator solution for each analyte using a gravimetric procedure. The twenty participating institutes primarily used analysis procedures based on GC-MS, -IDMS, -MS/MS, -ECD, and -FID, with some participants also using LC-UV for the value assignment. The analysis of methoxychlor and trifluralin in acetonitrile solution presented several challenges, including the thermal stability of the analytes under selected analytical techniques, control of solvent volatility, and considerable variation in some results using MS-based quantification methods. The mass fraction assignments for methoxychlor and trifluralin, consistent with the key comparison reference values (KCRVs), were achieved with associated relative standard uncertainties of (0.38 - 2.9) % for methoxychlor and (0.35 - 2.5) % for trifluralin. Rice yields must therefore increase to feed ever larger populations. By colonising rice and other plants, Herbaspirillum spp. stimulate plant growthand productivity. However the molecular factors involved are largely unknown. To further explore this interaction, the transcription profiles of Nipponbare rice roots inoculated with Herbaspirillum seropedicae were determined by RNA-seq. Mapping the 104 million reads against the Oryza sativa cv. Nipponbare genome produced 65 million unique mapped reads that represented 13,840 transcripts each with at least two-times coverage. About 7.4% (1,014) genes were differentially regulated and of these 255 changed expression levels more than two times. Several of the repressed genes encoded proteins related to plant defence (e.g. a putative probenazole inducible protein), plant disease resistance as well as enzymes involved in flavonoid and isoprenoid synthesis. Genes related to the synthesis and efflux of phytosiderophores (PS) and transport of PS-iron complexes were induced by the bacteria. These data suggest that the bacterium represses the rice defence system while concomitantly activating iron uptake. Transcripts of H. seropedicae were also detected amongst which transcripts of genes involved in nitrogen fixation, cell motility and cell wall synthesis were the most expressed. T1 - A review on the performance of açaí fiber in cementitious composites: Characteristics and application challenges N2 - This paper aimed to perform an exploratory literature review, focusing on the aspects of hydration and rheological properties of cementitious composites with açaí fibers. The use of natural fibers as a reinforcement in cementitious matrices has been widely studied, and açaí fiber is a promising option due to its unique characteristics. However, there is still a lack of knowledge regarding the correlation between fiber parameters and the technological properties of composites, which limits the development of new applications, such as 3D printing. The review provided an overview of the main characteristics of açaí fibers and the parameters for their application in cementitious matrices. Alkali treatment with 5% NaOH solution for 1 h shows increased mechanical and physical properties in açaí fibers. Despite the potential advantages of these composites, they pose challenges due to the different fiber surface treatment and interfacial transitions and the processing conditions of the fibers. Although small addition of açaí fibers slightly decreases the cement hydration process, keeping the cross-compatibility index high (>95). To overcome these challenges, it is necessary to evaluate the rheological behavior of cementitious materials with açaí fibers, which has not been reported in the literature. By doing so, it will be possible to optimize the processing conditions and develop new applications, such as additive manufacturing, which has never been explored with açaí fibers. T1 - In-Plane behaviour of earthen materials: A numerical comparison between adobe masonry, rammed earth and cob N2 - The paper presents a comparison between different numerical modelling approaches aiming to simulate the in-plain behaviour of three types of earthen materials, namely adobe masonry, rammed earth and cob. For this purpose, uniaxial and diagonal compression tests were carried out, which allowed determining important mechanical parameters, such as compressive strength, Young’s modulus, Poisson’s ratio, shear strength and shear modulus. Furthermore, the tests allowed assessing the level of non-linear behaviour of the respective stress–strain relationships as well as the failure modes. The experimental results were then used for the calibration of numerical models (based on the finite element method) for simulating the non-linear behaviour of the earth materials under in-plane shear loading. Both macro- and micro-modelling approaches were considered for this purpose. The procedures adopted for model calibration established the reliability of various modelling strategies for the different loading conditions. The simplified Approach based on macromodelling shows a satisfactory accuracy and low computational costs. The results reproducing the uniaxial compression are in good correspondence with the post-elastic behaviour observed in the experimental campaign. The micro-modelling approach adopted to reproduce the shear behaviour, even with higher computational cost, represents a suitable tool to predict the adobe masonry and rammed earth collapse mechanisms. In the first part of the study the mechanical characterisation of wallets tested under uniaxial compression and diagonal compression and walls tested under in-plane cyclic shear-compression loading is presented. The results were used to implement and validate the finite element simulations. The numerical modelling of the rammed earth samples tested is then discussed in the second part. A non-linear constitutive law based on the total strain rotating crack model (TSRCM) was employed as implemented in the DIANA® software. The aim of the numerical analyses presented here is to simulate the behaviour of rammed earth under different inplane loading conditions. For the wallets, tests under static loading both macro- and micro-modelling approaches were considered for the simulation of the experimental tests. For the walls subjected to cyclic loading only the micro-modelling approach was applied for the simulation of the experimental tests. The respective FEM model was calibrated with the experimental results. The rammed earth layers were represented by continuum elements, the contact surfaces between layers by interface elements. This approach allowed assessing the influence of the apparent weakness of the interfaces between layers on the shear behaviour of rammed earth. The goal of the numerical simulation of the cyclic tests was to establish the adequacy of common analytical methods (e. g. used for masonry) applied to the analysis of rammed earth. Rammed earth exhibits brittle characteristics similar to masonry materials and is used in geometrical typologies, such as walls, common in masonry construction. Für die Wandscheiben wurden Versuche unter linearer Last, sowohl in Makro- als auch Mikromodellansätze für die Simulation der experimentellen Versuche gewählt. Das betreffende FEM Modell wurde mit den experimentellen Ergebnissen kalibriert. Die Stampflehmlagen wurden mit durchgehenden Elementen, die Kontaktflächen zwischen den Lagen mit Oberflächenelementen repräsentiert. Dieser Ansatz erlaubte die Bewertung des Einflusses der auftretenden Schwächung in den Oberflächen zwischen den Lagen auf das Scherverhalten des Stampflehms. Ziel der numerischen Simulation von zyklischen Versuchen war es, die Eignung allgemeiner Analysemethoden (z. B. für Mauerwerk) für die Analyse von Stampflehm einzuführen und anzuwenden. T1 - Different Impact of Gadopentetate and Gadobutrol on Inflammation-Promoted Retention and Toxicity of Gadolinium Within the Mouse Brain N2 - Objectives: Using a murine model of multiple sclerosis, we previously showed that repeated administration of gadopentetate dimeglumine led to retention of gadolinium (Gd) within cerebellar structures and that this process was enhanced with inflammation. This study aimed to compare the kinetics and retention profiles of Gd in inflamed and healthy brains after application of the macrocyclic Gd-based contrast agent (GBCA) gadobutrol or the linear GBCA gadopentetate. Moreover, potential Gd-induced neurotoxicity was investigated in living hippocampal slices ex vivo. Materials and Methods: Mice at peak of experimental autoimmune encephalomyelitis (EAE; n = 29) and healthy control mice (HC; n = 24) were exposed to a cumulative dose of 20 mmol/kg bodyweight of either gadopentetate dimeglumine or gadobutrol (8 injections of 2.5 mmol/kg over 10 days). Magnetic resonance imaging (7 T) was performed at baseline as well as at day 1, 10, and 40 post final injection (pfi) of GBCAs. Mice were sacrificed after magnetic resonance imaging and brain and blood Gd content was assessed by laser ablation-inductively coupled plasma (ICP)-mass spectrometry (MS) and ICP-MS, respectively. In addition, using chronic organotypic hippocampal slice cultures, Gd-induced neurotoxicity was addressed in living brain tissue ex vivo, both under control or inflammatory (tumor necrosis factor α [TNF-α] at 50 ng/μL) conditions. Results: Neuroinflammation promoted a significant decrease in T1 relaxation times after multiple injections of both GBCAs as shown by quantitative T1 mapping of EAE brains compared with HC. This corresponded to higher Gd retention within the EAE brains at 1, 10, and 40 days pfi as determined by laser ablation-ICP-MS. In inflamed cerebellum, in particular in the deep cerebellar nuclei (CN), elevated Gd retention was observed until day 40 after last gadopentetate application (CN: EAE vs HC, 55.06 ± 0.16 μM vs 30.44 ± 4.43 μM). In contrast, gadobutrol application led to a rather diffuse Gd content in the inflamed brains, which strongly diminished until day 40 (CN: EAE vs HC, 0.38 ± 0.08 μM vs 0.17 ± 0.03 μM). The analysis of cytotoxic effects of both GBCAs using living brain tissue revealed an elevated cell death rate after incubation with gadopentetate but not gadobutrol at 50 mM. The cytotoxic effect due to gadopentetate increased in the presence of the inflammatory mediator TNF-α (with vs without TNF-α, 3.15% ± 1.18% vs 2.17% ± 1.14%; P = 0.0345). Conclusions: In the EAE model, neuroinflammation promoted increased Gd retention in the brain for both GBCAs. Whereas in the inflamed brains, efficient clearance of macrocyclic gadobutrol during the investigated time period was observed, the Gd retention after application of linear gadopentetate persisted over the entire observational period. Gadopentetate but not gadubutrol appeared to be neurotoxic in an ex vivo paradigm of neuronal inflammation. T1 - Microbiologically Influenced Corrosion - More than just Microorganisms N2 - Microbiologically influenced corrosion (MIC) is a phenomenon of increasing concern which affects various materials and sectors of society. MIC describes the effects, often negative, that a material can experience due to the presence of microorganisms. Unfortunately, although several research groups and industrial actors worldwide have already addressed MIC, discussions are fragmented, while information sharing and willingness to reach out to other disciplines is limited. A truly interdisciplinary approach, that would be logical for this material/biology/chemistry-related challenge, is rarely taken. In this review we highlight critical non-biological aspects of MIC that can sometimes be overlooked by microbiologists working on MIC but are highly relevant for an overall understanding of this phenomenon. Here, we identify gaps, methods and approaches to help solve MIC related challenges, with an emphasis on the MIC of metals. We also discuss the application of existing tools and approaches for managing MIC and propose ideas to promote an improved understanding of MIC. Carbides formed from the addition of Cr are eutectic of M3C type. The presence of such hard phases can enhance the abrasion wear resistance of ductile iron. A new CADI can be produced by the addition of Nb. Niobium carbide particles are formed in the beginning of solidification and remain stable once they are insoluble in solid iron matrix. The dry sand abrasive wear resistance of ductile irons alloyed with 1.0, 1.8, and 2.4 wt% Nb were tested in both “as-cast” and “heat treated” conditions using standard ASTM G65. Results were compared to abrasive wear data obtained on ductile iron alloyed with 1 wt% Cr, CADI (1 wt% Cr), and the basic composition of iron without carbide stabilizing elements. In the “ascast” condition, the addition of Nb did not lead to a reduction in wear, while CADI with Nb is a promising substitute for CADI with Cr addition, because both materials showed very similar values of abrasion resistance. Micro-ploughing and micro-cutting mechanisms were observed on the worn surfaces of ductile irons. Abrasive wear resistance of these alloys was correlated with the volume fraction of carbides. Tension and compression experiments were performed in a CT scanner employing a dedicated mechanical testing rig. The in-situ microtomographic scans enabled correlating the measured specimen response with relevant microstructural features and fracture processes. The microstructural segmentation of SHCC was performed in the framework of Deep Learning and it targeted an accurate segmentation of pores, fibers and aggregates. Besides their accurate volumetric representation, these phases were quantified in terms of content, size and orientation. The fracture processes were monitored at different loading stages and Digital Volume Correlation (DVC) was employed to spatially map the strains and cracks in the specimens loaded in compression. However, all major host groups are severely undersampled, and the taxonomy of existing lineages is not well developed. Full-length 16S rRNA gene sequences and genomes of arthropod-associated archaea are scarce, reference databases lack resolution, and the names of many taxa are either not validly published or under-classified and require revision. Here, we investigated the diversity of archaea in a wide range of methane-emitting arthropods, combining phylogenomic analysis of isolates and metagenome-assembled genomes (MAGs) with amplicon sequencing of full-length 16S rRNA genes. Our results allowed us to describe numerous new species in hitherto undescribed taxa among the orders Methanobacteriales (Methanacia, Methanarmilla, Methanobaculum, Methanobinarius, Methanocatella, Methanoflexus, Methanorudis, and Methanovirga, all gen. nova), Methanomicrobiales (Methanofilum and Methanorbis, both gen. nova), Methanosarcinales (Methanofrustulum and Methanolapillus, both gen. nova), Methanomassiliicoccales (Methanomethylophilaceae fam. nov., Methanarcanum, Methanogranum, Methanomethylophilus, Methanomicula, Methanoplasma, Methanoprimaticola, all gen. nova), and the new family Bathycorpusculaceae (Bathycorpusculum gen. nov.). Reclassification of amplicon libraries from this and previous studies using this new taxonomic framework revealed that arthropods harbor only CO2 and methyl-reducing hydrogenotrophic methanogens. Numerous genus-level lineages appear to be present exclusively in arthropods,suggesting long evolutionary trajectories with their termite, cockroach, and millipede hosts, and a radiation into various microhabitats and ecological nichesprovided by their digestive tracts (e.g., hindgut compartments, gut wall, or anaerobic protists). Therefore, this paper presents a numerical work aiming at modelling the non-linear behaviour of unstabilised rammed earth under shear loading, resorting to the finite ele-ments method (FEM). The models were used to simulate the behaviour of a set of rammed earth wallets tested under diagonal compression. Both macro- and micro-modelling approach-es were considered, where the objective of this last approach was to evaluate the influence of apparent weakness of the interfaces between layers on the shear behaviour. The total strain ro-tating crack model (TSCRM) was used to simulate the behaviour of the rammed earth material, while the Mohr-Coulomb failure criterion was used to simulate the behaviour of interfaces be-tween layers. A1 - Zscherpel, Uwe T1 - Application of the digital radiography in weld inspection of gas and oil pipelines N2 - The aim of this work is to evaluate the feasibility of the direct radiography on weld inspection in oil pipelines and gas pipeline during the manufacturing process. To that, 6 specimens with 6 different thickness and varied height of reinforced weld with 5 different kinds of defects were made. All samples were radiographied using Class I films and flat panel. For all specimens the inspection length was 8''. Thus, with the flat panel the detector-to-object distance varied so that it may adequate to several diameters of the tubes. The detector-to-object distance was calculated based on the physical size of the detector taking into consideration a safe distance between the tube curvature and the flat panel extremities, keeping the lowest possible magnification factor so that it could be obtained the length of the inspection. Images with 6 integration time for each experimental arrangement were obtained. The images obtained with the Flat Panel/YXLON system were analyzed according to their quality by using the Contrast parameters (essential wire) (DNV 2007/ IS0 12096 – with reinforcement and ISO 10893-7 – basis material), Basic Spatial Resolution – BSR (ISO 10893-7) and normalized signal-to-noise ratio - SNRN (ISO 10893-7) and by detectability using as reference the conventional radiography. The results showed that for all thickness, the exposure time used to meet the image quality requirements were below with direct radiography. However the BSR were not reached for thickness of 4.85, 6.40 and 9.67 mm, therefore the compensation principle established by ISO 10893-7 was considered, that is, one more contrast wire for a less wire pair. The digital technique proved to be more sensitive to real defects found on welds than the conventional technique. Then it can be conclude that the digital radiography utilizing the flat panel can be applicable to the oil and gas segment with advantages over conventional technique as to quality aspects, productivity, environment, safety and health.
