The aim is to assess their impact on the fatigue behavior. Therefore, a combination of methods, including image processing of micro‐computed tomography (CT) scans, fatigue testing, and machine learning, was applied. A workflow was established to contribute to the nondestructive assessment of component quality and mechanical properties. Additionally, this study illustrates the application of machine learning to address a classification problem, specifically the categorization of pores into gas pores and lack of fusion pores. Although it was shown that internal defects exhibited a reduced impact on fatigue behavior compared with surface defects, it was noted that surface defects exert a higher influence on fatigue behavior. A machine learning algorithm was developed to predict the fatigue life using surface defect features as input parameters. The O2 mole fraction (XO2) in the gas mixtures varied from 21% to 50% with the CH4/H2 mole fraction from 0 to 2%. Experimental result showed that MIE of coal dusts significantly decreases even by three orders of magnitude in mJ with increasing XO2 and the addition of CH4/H2. Compared with CH4, H2 had a relatively strong promotion effect on the spark ignition of coal dusts. The inhibiting effect of CO2 was found to be much stronger than N2, but this inhibiting effect of CO2 could be eliminated by 9% increment of XO2. The effect on MIE of coal dusts thus followed by the order: 9% increment of XO2 > CO2 replacing N2 > 2% CH4 or H2 addition. In eukaryotes, the multiple steps of biogenesis are accomplished by complex protein machineries in both mitochondria and cytosol. The underlying biochemical pathways have been elucidated over the past decades, yet the mechanisms of cytosolic [2Fe-2S] protein assembly have remained ill-defined. Similarly, the precise site of glutathione (GSH) requirement in cytosolic and nuclear Fe/S protein biogenesis is unclear, as is the molecular role of the GSH-dependent cytosolic monothiol glutaredoxins (cGrxs). Here, we investigated these questions in human and yeast cells by various in vivo approaches. [2Fe-2S] cluster assembly of cytosolic target apoproteins required the mitochondrial ISC machinery, the mitochondrial transporter Atm1/ABCB7 and GSH, yet occurred independently of both the CIA system and cGrxs. This mechanism was strikingly different from the ISC-, Atm1/ABCB7-, GSH-, and CIA-dependent assembly of cytosolic–nuclear [4Fe-4S] proteins. One notable exception to this cytosolic [2Fe-2S] protein maturation pathway defined here was yeast Apd1 which used the CIA system via binding to the CIA targeting complex through its C-terminal tryptophan. cGrxs, although attributed as [2Fe-2S] cluster chaperones or trafficking proteins, were not essential in vivo for delivering [2Fe-2S] clusters to either CIA components or target apoproteins. Finally, the most critical GSH requirement was assigned to Atm1-dependent export, i.e. a step before GSH-dependent cGrxs function. Our findings extend the general model of eukaryotic Fe/S protein biogenesis by adding the molecular requirements for cytosolic [2Fe-2S] protein maturation. To obtain approval, these transport packages must adhere to the specification-based criteria of the international transport regulations SSR-6 of the International Atomic Energy Agency (IAEA). To ensure compliance with these requirements, specific mechanical and thermal tests need to be addressed with respect to the package type. Typically, SSR-6 prescribes a mechanical test followed by a thermal fire test as part of different testing scenarios. To approve the latter test of the sequence, BAM performs calorimeter tests in advance with so-called fire reference packages for characterizing the actual fire and its impact on the package to be tested. Packages are designed with different geometry sizes depending on their purpose. For previous tests, the implemented test setups in the fire test stand were sufficiently dimensioned and could cover all requirements in this respect. However, to cover additional testing needs in the future, BAM is expanding the test setup for the purpose of testing significantly larger packages. In previous test setups one ring burner for propane surrounding the test specimen was sufficient. The limiting size in this configuration was the design height of the transport package to be tested. According to the thermal test of the IAEA-Regulations [1], a 30-minute fully engulfing 800°C pool fire or an equally severe fire, e.g., propane gas fire, must be applied to the transport package. Possible adjustments such as nozzle cross-section and propane mass flow can be adjusted to a certain extent. Further modifications, to cover significantly higher and larger packages all-around with a fully engulfing fire, must be accomplished with an additional, second burner ring. Both burner rings had the same dimensions and were mounted on top of each other at different heights to create a significantly larger volume of fire that completely engulfs the package including its impact limiter. To meet the IAEA regulatory boundary conditions, the enhanced fire test stand with the second burner ring is tested with a large fire reference package and will then be used for real-size transport packages after all parameters are successfully met. This fire reference package represents the external geometry of a generic transport cask for radioactive material and is equipped with numerous temperature sensors to record temperature curves at the interior wall surfaces. However, its widespread adoption in various use cases faces challenges due to the absence of interfaces to established Manufacturing Execution Systems (MES) that support customers in the predominantly data-driven quality assurance. Current state-of-the-art PBF-LB/M machines utilize communication architectures, such as OPC Unified Architecture (OPC UA), Message Queuing Telemetry Transport (MQTT) and Representational State Transfer Application Programming Interface (REST API). In the context of the Reference Architecture Model Industry 4.0 (RAMI 4.0) and the Internet of Things (IoT), the assets, particularly the physical PBF-LB/M machines, already have an integration layer implemented to communicate data such as process states or sensor values. Missing is an MES component acting as a communication and information layer. To address this gap, the proposed Extract Transform Load (ETL) pipeline aims to extract relevant data from the fabrication of each build cycle down to the level of scan vectors and additionally to register process signals. The suggested data schema for archiving each build cycle adheres to all terms defined by ISO/TC 261—Additive Manufacturing (AM). In relation to the measurement frequency, all data are reorganized into entities, such as the AM machine, build cycle, part, layer, and scan vector. These scan vectors are stored in a runtime-independent format, including all metadata, to be valid and traceable. Packages for the transportation of radioactive material must demonstrate that the package can withstand certain prescribed tests from the IAEA Regulations [1]. In addition to mechanical tests, a thermal test in form of a fire test must be carried out. As packages to be tested at BAM are signifi-cantly larger than previous package designs, BAM has expanded an existing fire test stand. A modular concept is chosen, which means that the arrangement of the burner nozzles can be adapted to the test specimen. The dimensions of the burner rings, the type, the orientation and the number of burner nozzles can be varied depending on the test specimen. In addition, various pumps can be used to set the corresponding mass flow. With the help of a calorimeter test, the fire test stand can be qualified for a specific size of packages regarding the boundary conditions of the IAEA Regulations [1]. Due to the typically wood filled impact limiters in German package designs, a fire test is necessary, as experiments have shown that possible openings that occurred during a mechanical test contributed to the igni-tion of the wood filled impact limiters within the prescribed 30 minutes of the IAEA Regulations [1]. From a series of experiments, two experiments are pre-sented to show the possibilities to obtain different temperatures and temperature rates in the test specimen. In addition to sensor data, the heat flux into the package is calculated to verify that the IAEA boundary conditions are satisfied. To investigate the combustion behaviour of densely packed layers of spruce wood, encapsulated in pre-damaged cylindrical metal enclosures, a test set-up has been realised. The set-up consists of a combustion chamber to perform these tests under defined boundary conditions. The temperature development of the test specimens will be observed fromoutside with a thermographic imager, with HD-Cameras, and the mass loss will be measured during the entire test. Airflow conditions in the combustion chamber are analysed using Computational Fluid Dynamics (CFD) calculations in OpenFOAM. The planned combustion test setup is described. To be able to monitor this pollutant, highly efficient analytical methods are needed, including immunoassays. In a medical research project, monoclonal antibodies against diclofenac and its metabolites had been produced. Based on this monoclonal anti-DCF antibody, a new indirect competitive enzyme-linked immunosorbent assay (ELISA) was developed and applied for environmental samples. The introduction of a spacer between diclofenac and the carrier protein in the coating conjugate led to higher sensitivity. With a test midpoint of 3 mg L−1 and a measurement range of 1–30 mg L−1, the system is not sensitive enough for direct analysis of surface water. However, this assay is quite robust against matrix influences and can be used for wastewater. Without adjustment of the calibration, organic solvents up to 5%, natural organic matter (NOM) up to 10 mg L−1, humic acids up to 2.5 mg L−1, and salt concentrations up to 6 g L−1 NaCl and 75 mg L−1 CaCl2 are tolerated. The antibody is also stable in a pH range from 3 to 12. Cross-reactivity (CR) of 1% or less was determined for the metabolites 40-hydroxydiclofenac (40-OH-DCF), 5-hydroxydiclofenac (5-OH-DCF), DCF lactam, and other NSAIDs. Relevant cross-reactivity occurred only with an amide derivative of DCF, 6-aminohexanoic acid (DCF-Ahx), aceclofenac (ACF) and DCF methyl ester (DCF-Me) with 150%, 61% and 44%, respectively. These substances, however, have not been found in samples. Only DCF-acyl glucuronide with a cross-reactivity of 57% is of some relevance. For the first time, photodegradation products were tested for cross-reactivity. With the ELISA based on this antibody, water samples were analysed. In sewage treatment plant effluents, concentrations in the range of 1.9–5.2 mg L−1 were determined directly, with recoveries compared to HPLC-MS/MS averaging 136%. N2 - A new approach in electrochemical corrosion testing includes the use of gel-type electrolytes as an alternative to aqueous test electrolytes. Gel-type electrolytes enable minimally invasive studies of naturally formed zinc coatings. Thus, corrosion-relevant parameters can be determined that allow conclusions on layer formation and layer stability. This contribution will clarify the value of gel-type electrolytes for corrosion research at the example of various coatings on zinc. Their naturally formed protective layers are electrochemically characterized and corrosion relevant values are determined by using a gel pad based on polysaccharide. The corrosion relevant values allow the differentiation of various zinc coating systems and can describe the current protective effect provided by the coating. It is shown that gel-type electrolytes influence protective layers and coatings considerably less than corresponding bulk electrolytes. From the results it is evident that gel-type electrolytes represent a viable and promising field in corrosion research.
