Regulatory test conditions shall cover these severe accident conditions and consist of mechanical tests and a following thermal test. To withstand the mechanical tests heavy weight packages are often designed with impact limiters consisting of wood encapsulated in steel sheets. The thermal test is defined precisely in the IAEA-regulations as a 30 minute fully engulfing 800 °C fire. After the fire phase a pre-damaged impact limiter might continue burning or smouldering and influence the cask thermal behaviour with its energy release. The energy transferred from the impact limiter to the cask is of importance for the safety of transport packages. A full-scale fire test with an impact limiter of 2.3 m in diameter and filled with spruce wood was designed and performed. The impact limiter continued burning for 3 days. For example, variations in the 26Mg/24Mg isotope abundance ratio in natural systems are typically reported as delta26Mg values that represents the relative difference between the 26Mg/24Mg ratio measured in a sample relative to its measurement preferably in an iRM. In the past, such delta26Mg measurements were referenced to NIST SRM 980, the initial zero of the delta26Mg scale. With the development of MC-ICPMS, the detection of small but measurable isotopic differences in different chips of SRM 980 became apparent. It was then replaced by a Mg solution (DSM3), the new zero of the delta26Mg scale. A potential replacement iRM for DSM3 has been developed, ERM-AE143. This iRM has also been measured for its absolute isotope amount ratios1,2 making it traceable to the SI. The results of an delta26Mg intercomparison experiment include the Mg iRMs SRM 980, IRMM-009, ERM-AE143, AE144, AE145 standards DSM3 and Cambridge-1. The intercomparison involved 5 expert laboratories, consisting of 3 national metrological institutes (BAM, NIST, PTB) and 2 scientific research laboratories (GFZ Potsdam, UBremen). The iRMs were measured relative to AE143 and cover a range of ≈5 ‰ in delta26Mg. IRMM-009 has the lowest delta26Mg value while DSM3 has the highest, spanning a range in values that covers natural Mg isotope variations. The 2SD reproducibilities of the individual values from the different laboratories range from 0.02 to 0.26 ‰. The mean delta26Mg values, calculated from the laboratory means however show 2SD reproducibilities varing between 0.025 and 0.093 ‰. Propagated measurement uncertainties suggest a standard uncertainty of about 0.1‰ for delta26Mg determinations. The production of mAbs is routinely performed by hybridoma technique. Hybridomas are generated by fusing antibody-producing B cells with indefinitely proliferating myeloma cells. Both, cell fusion and the indispensable identification and isolation of the desired antibody-producing hybridoma clone is still far from being trivial. We describe a novel method that should help to optimize the cell fusion, and the screening for antibody-secreting hapten-specific hybridoma cells by using fluorescence activated cell sorting and single-cell fusion. Hybridoma cells specific for a hapten were incubated with a hapten-peroxidase conjugate (hapten-HRP), which was subsequently incubated with a fluorophore-labeled polyclonal anti-peroxidase antibody (anti-HRP-Alexa Fluor® 488). To characterize the expression of membrane-bound immunoglobulin G (IgG) a fluorophore-labeled anti-mouse IgG antibody (anti-IgG-Alexa Fluor® 647) was used. Successful staining was verified by confocal laser scanning microscopy (CLSM). We show that it is possible to specifically label hapten-specific hybridoma cells. It should also be possible to use this labelling approach for the isolation of hapten-specific B cells from the spleen and use these cells for single-cell fusion. While their convenient mechanical characteristics predestine these materials for shock absorption, their low thermal conductivity is responsible for their excellent thermal insulation properties. A principal characteristic affecting not only the use properties, but also the burning behaviour, is the density of RPUF. In case of fire, cellular polymers like RPUF behave differently from bulk materials. The reason for their comparatively high ignitability is the high rate of temperature rise on the surface when exposed to heat, which is due to their cellular structure and characterised as low thermal inertia. The heat build-up on the surface results in a short time to ignition. Therefore foams are able to develop fire and large amounts of smoke within a short period of time. Even though their fire load is quite low because of their low density, their heat release rate is high. These burning properties are not only a consequence of the chemical consistence of such materials, but also a result of their morphology. The aim of the work is to investigate the fire phenomena of RPUF. Characterised by simultaneous physical and chemical processes which interact, depend, and compete with each other, the combustion of RPUF is a highly complex occurrence. Therefore it is necessary to study all aspects in detail. The fire phenomena that were examined are gasification, liquefaction, charring and structural integrity as well as collapse. The melting of cellular polymers and dripping of pyrolysis products, together with the ability to form pool fires, are the main hazards in fires involving foams. Structural collapse via melting or decomposition, or the retention of the foam’s morphology through charring, are the main characteristics of their burning behaviour. Cellular structure is a further important factor influencing the materials’ response to fire, and deserves examination in greater depth. Since the cells in closed-cell foam are filled with the blowing agent used to produce the material, and this blowing agent can be inert (carbon dioxide) or highly flammable (pentane), this component also affects the burning properties and will be investigated. Examination of the influence of the macroscopic morphology, namely the cell size and thickness of cell walls, will contribute to the study. In order to provide comprehensive insight into the processes taking place during combustion, and to elucidate structure-property relationships, a set of foams with systematically varied properties was prepared. RPUF was obtained using water and pentane as a blowing agent. Besides RPUF, a flame-retarded RPUF and rigid polyisocyanurate-polyurethane foam were examined, since these foams are known for their higher stability from a thermodanymic point of view. As a flame retardant active in the gas phase, triethylphosphate was used. Using a multi-methodological approach, the complex interaction of fire phenomena was studied in detail. Cone calorimeter measurements provide insight into fire behaviour under forced-flaming conditions. They were carried out in an horizontal and a vertical orientation to account for the effects of melt flow and dripping. Sample-holders were equipped with glass windows to monitor the materials’ response to forced-flaming conditions with a video camera. Thermocouples were inserted into the specimens, giving information about the temperature gradient in the inside of the sample and the temperature of the pyrolysis zone during combustion. Changes in the morphology and thickness of the pyrolysis zone were examined using a hot-stage microscope, as well as SEM images of cross-sections of quenched foam specimens. The determination of the Oxygen Index and the use of thermogravimetry measurements rounded out the investigation. In this paper we give an overview of the latest status regarding the development of a finite element material model for the crush of spruce wood. Although the crush of wood – mainly in longitudinal direction – is a phenomenon governed by macroscopic fracture and failure of wood fibres we smear fracture and failure mechanisms over the continuous voume. In first step we altered an existing LS-DYNA material model for foams, which considers an ellipse shaped yield surface written in terms of the first two stress invariants. The evolution of the yield surface in the existing model depends on the volumetric strain only. For the use with spruce wood, we modified the existing material model to consider the deviatoric strain for the evolution of the yield surface as well. This is in accordance with the results of crush tests with spruce wood specimens, where the crushing deformation was rather deviatoric for uniaxial stress states and rather volumetric for multiaxial stress states We rate the basic idea of this approach to be reasonable, though other problems exist regarding the shape of the yield surface and the assumption of isotropic material properties. Therefore we developed a new transversal isotropic material model with two main directions, which considers different yield curves according to the multiaxiality of the stress state via a multi-surface yield criterion and a non-associated flow rule. The results show the ability to reproduce the basic strength characteristics of spruce wood. Nevertheless, problems with regularization etc. show that additional investigations are necessary. The austenitic stainless steel AISI 304 from a consistent heat was used for all investigations to guarantee the same chemical composition and thus no additional influences caused by the material itself. A wide range of different grinding parameters such as pressure, cooling medium and grain size of the abrasive was varied. Thus, comparison of the corrosion behaviour of different surface states and the evaluation of an optimum grinding process using corundum abrasives were possible. The results will contribute to present discussions and give novel impulses for companies in the metalworking industry.
