T1 - Mobility particle size spectrometers: Calibration procedures and measurement uncertainties N2 - Mobility particle size spectrometers (MPSS) belong to the essential instruments in aerosol science that determine the particle number size distribution (PNSD)in the submicrometer size range. Following calibration procedures and target uncertainties against standards and reference instruments are suggested for a complete MPSS quality assurance program: a) calibration of the CPC counting efficiency curve (within 5% for the plateau counting efficiency; within 1 nm for the 50% detection efficiency diameter), b) sizing calibration of the MPSS, using a certified polystyrene latex (PSL) particle size standard at 203 nm (within 3%), c) intercomparison of the PNSD of the MPSS (within 10% and 20% of the dN/dlogDP concentration for the particle size range 20 – 200 nm and 200 to 800 nm, respectively), and d) intercomparison of the integral PNC of the MPSS (within 10%). Furthermore, following measurement uncertainties have been investigated: a) PSL particle size standards in the range from 100-500nm match within 1% after sizing calibration at 203 nm. b) Bipolar diffusion chargers based on the radioactive nuclides Kr85, Am241 and Ni63 and a new ionizer based on corona discharge follow the recommended bipolar charge distribution, while soft X-ray-based charges may alter faster than expected. c) The use of a positive high voltage supply show a 10% better performance than a negative one. d) The intercomparison of the integral PNC of an MPSS against the total number concentration is still within the target uncertainty at an ambient pressure of approximately 500 hPa. T1 - Interactions of fullerene-polyglycerol sulfates at viral and cellular interfaces N2 - Understanding the mechanism of interactions of nanomaterials at biointerfaces is a crucial issue to develop new antimicrobial vectors. In this work, a series of water-soluble fullerene-polyglycerol sulfates (FPS) with different fullerene/polymer weight ratios and varying numbers of polyglycerol sulfate branches are synthesized, characterized, and their interactions with two distinct surfaces displaying proteins involved in target cell recognition are investigated. The combination of polyanionic branches with a solvent exposed variable hydrophobic core in FPS proves to be superior to analogs possessing only one of these features in preventing interaction of vesicular Stomatitis virus coat glycoprotein (VSV-G) with baby hamster kidney cells serving as a model of host cell. Interference with L-selectin-ligand binding is dominated by the negative charge, which is studied by two assays: a competitive surface plasmon resonance (SPR)-based inhibition assay and the leukocyte cell (NALM-6) rolling on ligands under flow conditions. Due to possible intrinsic hydrophobic and electrostatic effects of synthesized compounds, pico- to nanomolar half maximal inhibitory concentrations (IC50) are achieved. With their highly antiviral and anti-inflammatory properties, together with good biocompatibility, FPS are promising candidates for the future development towards biomedical applications. T1 - Photoacoustic thermal characterization of low thermal diffusivity thin films N2 - The photoacoustic measurement technique is a powerful yet underrepresented method to characterize the thermal transport properties of thin films. For the case of isotropic low thermal diffusivity samples, such as glasses or polymers, we demonstrate a general approach to extract the thermal conductivity with a high degree of significance. We discuss in particular the influence of thermal effusivity, thermal diffusivity, and sample layer thickness on the significance and accuracy of this measurement technique. These fundamental thermal properties guide sample and substrate selection to allow for a feasible thermal transport characterization. Furthermore, our data evaluation allows us to directly extract the thermal conductivity from this transient technique, without separate determination of the volumetric heat capacity, when appropriate boundary conditions are fulfilled. Using silica, poly(methyl methacrylate) (PMMA) thin films, and various substrates (quartz, steel, and silicon), we verify the quantitative correctness of our analytical approach. Digitale Technologien bieten ein großes Potenzial zur Bewältigung der aktuellen und künftigen Herausforderungen im Infrastrukturmanagement. Im BMBF-geförderten Projekt Bewertung alternder Infrastrukturbauwerke mit digitalen Technologien (AISTEC) wird untersucht, wie unterschiedliche Technologien und deren Verknüpfung gewinnbringend eingesetzt werden können. Am Beispiel der Maintalbrücke Gemünden werden ein sensorbasiertes Bauwerksmonitoring, bildbasierte Inspektion mit durch Kameras ausgestatteten Drohnen (UAS) und die Verknüpfung digitaler Bauwerksmodelle umgesetzt. Die aufgenommenen Bilder dienen u. a. als Grundlage für spätere visuelle Anomaliedetektionen und eine 3D-Rekonstruktion, welche wiederum für die Kalibrierung und Aktualisierung digitaler Tragwerksmodelle genutzt werden. Kontinuierlich erfasste Sensordaten werden ebenfalls zur Kalibrierung und Aktualisierung der Tragwerksmodelle herangezogen. Diese Modelle werden als Grundlage für Anomaliedetektionen und perspektivisch zur Umsetzung von Konzepten der prädiktiven Instandhaltung verwendet. Belastungsfahrten und historische Daten dienen in diesem Beitrag der Validierung von kalibrierten Tragwerksmodellen. N2 - Infrastructure systems of industrialised countries today and in the future require efficient management with an ageing stock, increasing loads while simultaneously maintaining a high level of safety. Digital technologies offer great potential for the current and future challenges in infrastructure management. The BMBF-funded project AISTEC is investigating how the individual technologies and their interconnection can be used beneficially. With the Maintalbrücke in Gemünden as an exemplary application, sensor-based structural monitoring, image-based inspection using unmanned aircraft systems (UAS) equipped with cameras and the integration of digital structural models are being implemented. The recorded images serve, among others, as basis for subsequent anomaly detection and a 3D reconstruction, which in turn are used for updating digital structural models. Continuously recorded sensor data is used to update the parameters of the structural models, which in turn provide the basis for predictive maintenance. Load tests are used to validate the models. A1 - Procop, Mathias A1 - Beck, Uwe T1 - Realisation and metrological characterisation of thickness standards below 100 nm N2 - High-accuracy film thickness measurements in the range below 100 nm can be made by various complex methods like spectral ellipsometry (SE), scanning force microscopy (SFM), grazing incidence X-ray reflectometry (GIXR), or X-ray fluorescence analysis (XRF). The measurement results achieved with these methods are based on different interactions between the film and the probe. A key question in nanotechnology is how to achieve consistent results on a level of uncertainty below one nanometre with different techniques. Two different types of thickness standards are realised. Metal film standards for X-ray techniques in the thickness range 10 to 50 nm are calibrated by GIXR with monochromatised synchrotron radiation of 8048 eV. The results obtained at four different facilities show excellent agreement. SiO2 on Si standards for SE and SFM in the thickness range 6 to 1000 nm are calibrated by GIXR with monochromatised synchrotron radiation of 1841 eV and with a metrological SFM. Consistent results within the combined uncertainties are obtained with the two methods. Surfaces and interfaces of both types of standards are additionally investigated by transmission electron microscopy (TEM). Dabei spielen dünne Schichten und deren Kenndaten eine wesentliche Rolle, z.B. als Röntgenspiegel oder optische und magnetooptische Datenträger. Die Schichtdicke zählt hierbei zu den technologisch wichtigen Parametern. Abgeleitet von Anwenderinteressen, werden für die Röntgenreflektometrie (XRR bzw. GIXR), Elektronenstrahl-Mikroanalyse (EPMA) und Röntgenfluoreszenzanalyse (XRF) einerseits und die Ellipsometrie andererseits zugeschnittene Schichtdickenmaßverkörperungen (SDM) entwickelt, gefertigt und untersucht. Das Ziel ist die Bereitstellung praxistauglicher, kalibrierter SDM, im weiteren Schichtdickennormale (SDN) genannt. SDN sind Maßverkörperungen, für die der Schichtdickenwert mit einer definierten Messunsicherheit bekannt und auf ein metrologisch anerkanntes Längennormal zurückgeführt ist. T1 - A validated robust and automatic procedure for vibration analysis of bridge structures using MEMS accelerometers N2 - Today, short- and long-term structural health monitoring (SHM) of bridge infrastructures and their safe, reliable and cost-effective maintenance has received considerable attention. From a surveying or civil engineer’s point of view, vibration-based SHM can be conducted by inspecting the changes in the global dynamic behaviour of a structure, such as natural frequencies (i. e. eigenfrequencies), mode shapes (i. e. eigenforms) and modal damping, which are known as modal parameters. This research work aims to propose a robust and automatic vibration analysis procedure that is so-called robust time domain modal parameter identification (RT-MPI) technique. It is novel in the sense of automatic and reliable identification of initial eigenfrequencies even closely spaced ones as well as robustly and accurately estimating the modal parameters of a bridge structure using low numbers of cost-effective micro-electro-mechanical systems (MEMS) accelerometers. To estimate amplitude, frequency, phase shift and damping ratio coefficients, an observation model consisting of: (1) a damped harmonic oscillation model, (2) an autoregressive model of coloured measurement noise and (3) a stochastic model in the form of the heavy-tailed family of scaled t-distributions is employed and jointly adjusted by means of a generalised expectation maximisation algorithm. Multiple MEMS as part of a geo-sensor network were mounted at different positions of a bridge structure which is precalculated by means of a finite element model (FEM) analysis. At the end, the estimated eigenfrequencies and eigenforms are compared and validated by the estimated parameters obtained from acceleration measurements of high-end accelerometers of type PCB ICP quartz, velocity measurements from a geophone and the FEM analysis. Additionally, the estimated eigenfrequencies and modal damping are compared with a well-known covariance driven stochastic subspace identification approach, which reveals the superiority of our proposed approach. We performed an experiment in two case studies with simulated data and real applications of a footbridge structure and a synthetic bridge. The results show that MEMS accelerometers are suitable for detecting all occurring eigenfrequencies depending on a sampling frequency specified. Moreover, the vibration analysis procedure demonstrates that amplitudes can be estimated in submillimetre range accuracy, frequencies with an accuracy better than 0.1 Hz and damping ratio coefficients with an accuracy better than 0.1 and 0.2 % for modal and system damping, respectively. T1 - Stochastic subspace-based damage detection of a temperature affected beam structure N2 - Structural health monitoring (SHM) of civil structures often is limited due to changing environmental conditions, as those changes affect the structural dynamical properties in a similar way like damages can do. In this article, an approach for damage detection under changing temperatures is presentedand applied to a beam structure. The used stochastic subspace-based algorithm relies on a reference null space estimate, which is confronted to data from the testing state in a residual function. For damage detection the residual is evaluated by means of statistical hypothesis tests. Changes of the system due to temperature effects are handled with a model interpolation approach from linear parameter varying system theory. From vibration data measured in the undamaged state at some few reference temperatures, a model of the dynamic system valid for the current testing temperature is interpolated. The reference null space and the covariance matrix for the hypothesis test is computed from this interpolated model. This approach has been developed recently and was validated in an academic test case on simulations of a mass-spring-damper. In this paper, the approach is validated experimentally on a beam structure under varying temperature conditions in a climate chamber. Compared to other approaches, the interpolation approach leads to significantly less false positive alarms in the reference state when the structure is exposed to different temperatures, while faults can still be detected reliably. T1 - Wrapping and Blocking of Influenza A Viruses by Sialylated 2D Nanoplatforms N2 - Inhibition of respiratory viruses is one of the most urgent topics as underlined by different pandemics in the last two decades. This impels the development of new materials for binding and incapacitation of the viruses. In this work, we have demonstrated that an optimal deployment of influenza A virus (IAV) targeting ligand sialic acid (SA) on a flexible 2D platform enables its binding and wrapping around IAV particles. A series of 2D sialylated platforms consisting graphene and polyglycerol are prepared with different degrees of SA functionalization around 10%, 30%, and 90% named as G-PG-SAL, G-PG-SAM, and G-PG-SAH, respectively. The cryo-electron tomography (Cryo-ET) analysis has proved wrapping of IAV particles by G-PG-SAM. A confocal-based colocalization assay established for these materials has offered the comparison of binding potential of sialylated and non-sialylated nanoplatforms for IAV. With this method, we have estimated the binding potential of the G-PG-SAM and G-PG-SAH sheets for IAV particles around 50 and 20 times higher than the control sheets, respectively, whereas the low functionalized G-PG-SAL have not shown any significant colocalization value. Moreover, optimized G-PG-SAM exhibits high potency to block IAV from binding with the MDCK cells. T1 - Beyond microplastics ‑ investigation on health impacts of submicron and nanoplastic particles after oral uptake in vitro N2 - The continuously increasing use of plastics is supposed to result in a rising exposure of MNPs to humans. Available data on human health risks of microplastics after oral uptake increased immensely in the past years and indicates very likely only low risks after oral consumption. Concerning nanoplastics, uptake, transport and potential adverse effects after oral uptake are less well understood. This study aims to investigate differences between microplastic particles and particles in the submicron- and nanoscaled size derived from food-relevant polymers with a particle size range consistent with higher potential for cellular uptake, fate, and effects when applied to human intestinal and liver cells. This work includes the development of cellular and subcellular detection methods for synthetic polymeric particles in the micro- and nanometer-range, using Scanning Electron Microscopy, Small-Angle X-ray and Dynamic Light Scattering methods, Asymmetric Flow Field Flow Fractionation, octanol-water fractionation, fluorescence microscopy and flow cytometry. Polylactic acid (250 nm and 2 μm (polydisperse)), melamine formaldehyde (366 nm) and polymethylmethacrylate (25 nm) were thoroughly characterized. The submicro- and nanoplastic test particles showed an increased uptake and transport quantity through intestinal cells. Both types of particles resulted in observed differences of uptake behavior, most likely influenced by different lipophilicity, which varied between the polymeric test materials. Toxic effects were detected after 24 h only in overload situations for the particles in the submicrometer range. This study provides further evidence for gastrointestinal uptake of submicro- and nanoplastics and points towards differences regarding bioavailability between microplastics and smaller plastic particles that may result following the ingestion of contaminated food and beverages. Furthermore, the results reinforce the importance for studying nanoplastics of different materials of varying size, surface properties, polymer composition and hydrophobicity.
