Das Systemmodell wurde anhand eines ausgesuchten Spannbetonüberbaus in einzelliger Kastenbauweise entwickelt. Es besteht aus zwei integralen Teilmodellen: ein Modell zur Beschreibung des Systemschädigungszustandes und ein Modell zur Beschreibung der Standsicherheit. Für die Modellierung des stochastischen Systemschädigungszustandes eines Brückensystems werden dynamische Bayes'sche Netze (DBN) vorgeschlagen. Dieser Ansatz ermöglicht es, alle relevanten Schädigungsprozesse und deren stochastische Abhängigkeiten zu berücksichtigen. Ein wesentlicher Vorteil dieses Ansatzes ist es, dass DBN ideal dafür geeignet sind, Bayes'sche Aktualisierungen auf Grundlage von Informationen aus Inspektionen und Überwachungsmaßnahme auf eine effiziente und robuste Art und Weise durchzuführen. Der DBN-Ansatz ist deshalb für die Entwicklung von Software für das Erhaltungsmanagement von alternden Brückenbauwerken, die vom Benutzer keine vertieften Kenntnisse der Zuverlässigkeitstheorie verlangt, ideal geeignet. Für die Modellierung der Standsicherheit eines alternden Kastenträgers wird vereinfachend Biegeversagen des globalen Längssystems betrachtet. Zur Berechnung der maximalen Traglast eines Kastenträgers infolge des Systemschädigungszustandes wird ein plastisch-plastisches Verfahren eingesetzt, wobei die Beanspruchungen mittels der Fließgelenktheorie unter Ausnutzung der plastischen Beanspruchbarkeit der Querschnitte des Kastenträgers ermittelt werden. Ein Kastenträger versagt, wenn sich durch die Ausbildung einer ausreichend großen Anzahl von Fließgelenken eine kinematische Kette ausbildet. Dieser Modellierungsansatz berücksichtigt Redundanzen, die sich aus der plastischen Beanspruchbarkeit der Querschnitte und der statischen Unbestimmtheit eines Kastenträgers ergeben. Zum Nachweis der praktischen Einsetzbarkeit des entwickelten Systemmodells wurde ein Software-Prototyp entwickelt, der eine intuitiv benutzbare graphische Benutzeroberfläche (Front-End) mit einem Berechnungskern (Back-End) koppelt. Die aktuelle Version des Software-Prototyps implementiert ein Modell der chloridinduzierten Bewehrungskorrosion und ein Tragwerksmodell, welches das Verfahrens der stetigen Laststeigerung zur Bestimmung der maximalen Traglast des Kastenträgers auf der Grundlage eines Finite-Elemente-Modells umsetzt. Zur Durchführung von Bayes'schen Aktualisierungen des Systemschädigungszustandes auf der Grundlage des DBN-Modells implementiert der Prototyp den Likelihood-Weighting-Algorithmus. Die entwickelte Architektur des Prototyps ermöglicht eine Erweiterung der Software um weitere Schädigungsprozesse. Der entwickelte Software-Prototyp ermöglicht Benutzern ohne vertiefte Kenntnisse der Zuverlässigkeitstheorie eine Berechnung des Einflusses von Bauwerksinformationen auf den Systemschädigungszustand und die Tragsicherheit eines Kastenträgers. T1 - How life history influences the responses of the clam Scrobicularia plana to the combined impacts of carbamazepine and pH decrease N2 - In the present study, the bivalve Scrobicularia plana, collected from two contrasting areas (pristine location and mercury contaminated area), was selected to assess the biochemical alterations imposed by pH decrease, carbamazepine (an antiepileptic) and the combined effect of both stressors. The effects on oxidative stress related biomarkers after 96 h exposure revealed that pH decrease and carbamazepine induced alterations on clams, with greater impacts on individuals from the contaminated area which presented higher mortality, higher lipid peroxidation and higher glutathione S-transferase activity. These results emphasize the risk of extrapolating results from one area to another, since the same species inhabiting different areas may be affected differently when exposed to the same stressors. Furthermore, the results obtained showed that, when combined, the impact of pH decrease and carbamazepine was lower than each stressor acting alone, which could be related to the defence mechanism of valves closure when bivalves are under higher stressful conditions. In this regard, fast and reliable analytical methods are required to allow quick countermeasures in case of contamination. Here, we report the development of a magnetic bead-based immunoassay (MBBA) for the fast and cost-effective determination of the analgesic diclofenac (DCF) in water samples, based on diclofenac-coupled magnetic beads and a robust monoclonal anti-DCF antibody. A novel synthetic strategy for preparation of the beads resulted in an assay that enabled for the determination of diclofenac with a significantly lower limit of detection (400 ng/L) than the respective enzyme-linked immunosorbent assay (ELISA). With shorter incubation times and only one manual washing step required, the assay demands for remarkably shorter time to result (< 45 min) and less equipment than ELISA. Evaluation of assay precision and accuracy with a series of spiked water samples yielded results with low to moderate intra- and inter-assay variations and in good agreement with LC–MS/MS reference analysis. The assay principle can be transferred to other, e.g., microfluidic, formats, as well as applied to other analytes and may replace ELISA as the standard immunochemical method. The AM process involves repeated rapid melting, solidification and reheating, which exposes the material to non-equilibrium conditions that affect elemental segregation and the subsequent formation of solidification phases, either beneficial or detrimental. These variations are difficult to characterize due to the small length scale within the micron sized melt pool. To understand how the non-equilibrium conditions affect the initial solidification phases and their critical temperatures, a multi-length scale, multi modal approach has been taken to evaluate various methods for identifying the initial phases formed in the as-built Inconel 718 produced by laser-powder bed fusion (L-PBF) additive manufacturing (AM). Using a range of characterization tools from the bulk differential thermal analysis (DTA) and x-ray diffraction (XRD) to spatially resolved images using a variety of electron microscopy tools, a better understanding is obtained of how these minor phases can be properly identified regarding the amount and size, morphology and distribution. Using the most promising characterization techniques for investigation of the as-built specimens, those techniques were used to evaluate the specimens after various heat treatments. During the sequence of heat treatments, the initial as-built dendritic structures recrystallized into well-defined grains whose size was dependent on the temperature. Although the resulting strength was similar in all heat treated specimens, the elongation increased as the grain size was refined due to differences in the precipitated phase distribution and morphology. In this contribution, we propose a machine learning (ML) approach for automated damage detection, based on an ML toolbox for industrial condition monitoring. The toolbox combines multiple complementary algorithms for feature extraction and selection and automatically chooses the best combination of methods for the dataset at hand. Here, this toolbox is applied to a guided wave-based SHM dataset for varying temperatures and damage locations, which is freely available on the Open Guided Waves platform. A classification rate of 96.2% is achieved, demonstrating reliable and automated damage detection. Moreover, the ability of the ML model to identify a damaged structure at untrained damage locations and temperatures is demonstrated. In dem vorliegenden Beitrag werden allgemeine Grundlagen und die zukünftigen Arbeitsfelder des AK 2.15 vorgestellt. Ausgehend von der Einführung relevanter Fachbegriffe wird zunächst die Einbettung zuverlässigkeitsbasierter Verfahren in den aktuellen Normungs- und Regelungskontext aufgezeigt. Anschließend werden anhand des Lebenszyklus eines geotechnischen Bauwerks die Unsicherheiten in den geotechnischen Prognosen und Bewertungen beschrieben. T1 - Acquiring a Machine Learning Data Set for Structural Health Monitoring of Hydrogen Pressure Vessels at Operating Conditions using Guided Ultrasonic Waves N2 - Hydrogen is an energy source of increasing importance. As hydrogen is very reactive to air and needs to be stored under high pressure, it is crucial to provide safe transportation and storage. Therefore, structural health monitoring, based on guided ultrasonic waves and machine learning methods, is used for Composite Overwrapped Pressure Vessels (COPVs) containing hydrogen. To acquire data that allows robust detection of COPV defects, there are two main process parameters to consider. These are the pressurization of the vessel and the temperature conditions at the vessel. This paper will focus on the derivation of a design of experiment (DoE) from the needs of various validation scenarios (e.g. concerning pressure, temperature or excitation frequency). Practical limitations must be considered as well. We designed experiments with multiple reversible damages at different positions. A network of 25 transducers, structured as five rings with five sensors in one line, is installed on a vessel. Guided ultrasonic waves are used via the pitch-catch procedure, which means that the transducers act pairwise as transmitter and receiver in order to measure all transmitterreceiver combinations. This leads to 600 signal paths, recorded by a Verasonics Vantage 64 LF data acquisition system. Finally, the influences of temperature and pressure within the acquired data set are going to be visualized. T1 - Reproducibility and Scattering in Additive Manufacturing: Results from a Round Robin on PBF-LB/M AlSi10Mg Alloy T1 - Reproduzierbarkeit und Streuung bei der additiven Fertigung: Ergebnisse eines Ringversuchs mit einer PBF-LB/M AlSi10Mg-Legierung N2 - The round robin test investigated the reliability users can expect for AlSi10Mg additive manufactured specimens by laser powder bed fusion through examining powder quality, process parameter, microstructure defects, strength and fatigue. Besides for one outlier, expected static material properties could be found. Optical microstructure inspection was beneficial to determine true porosity and porosity types to explain the occurring scatter in properties. Fractographic analyses reveal that the fatigue crack propagation starts at the rough as-built surface for all specimens. Statistical analysis of the scatter in fatigue using statistical derived safety factors concludes that at a stress of 36.87 MPa the fatigue limit of 107 cycles could be reached for all specimen with a survival probability of 99.999 %. N2 - Im Rahmen eines Ringversuchs wurde durch die Untersuchung der Pulverqualität, der Prozessparameter, der Gefügefehler, der Festigkeit und der Ermüdung die Zuverlässigkeit bestimmt, die Nutzer von AlSi10Mg-Proben erwarten können, die mit pulverbettbasiertes Schmelzen mittels Laser (engl. Laser Powder Bed Fusion) gefertigt worden sind. Abgesehen von einem Ausreißer wurden die erwarteten statischen Materialeigenschaften erreicht. Eine optische Gefügeprüfung diente dazu, die tatsächliche Porosität und Arten von Porosität zu ermitteln, um die bei den Eigenschaften auftretende Streuung zu erklären. Fraktographische Unterschungen zeigen eine bei allen Proben von der rauen Oberfläche im As-built-Zustand ausgehende Ermüdungsrissausbreitung. Aus der statistischen Analyse der Streuung bezüglich der Ermüdung unter Anwendung von statistischen abgeleiteten Sicherheitsfaktoren geht hervor, dass alle Proben die Dauerfestigkeit von 107 Zyklen bei einer Spannung von 36,87 MPa mit einer Überlebenswahrscheinlichkeit von 99,999 % erreichten. In the aquatic environment, most of the studies have been devoted to fish and bivalves, while little is known on the impacts induced in polychaetes. The present study evaluated the impacts of carbamazepine and caffeine on the regenerative capacity of Diopatra neapolitana, a polychaete species with high ecological and economic relevance. Under laboratory controlled conditions polychaetes were exposed, during 28 days, to carbamazepine (Ctl-0.0; 0.3; 3.0; 6.0; 9.0 mg/L) and caffeine (Ctl-0.0; 0.5; 3.0; 18.0 mg/L). During the experiment, at days 11, 18, 25, 32, 39 and 46 after amputation, for each specimen, the percentage of the body width regenerated was determined and the number of new segments was counted. The regenerative capacity was assessed considering the number of days needed to achieve full regeneration and the total number of new segments. The obtained results revealed that with the increase of drugs concentrations organisms regenerated less new segments and took longer to completely regenerate. A growing body of evidence demonstrates that ocean acidification can have direct and indirect impacts on marine organisms although combined effects with other stressors, namely with pharmaceuticals, have received very little attention to date. The present study aimed to evaluate the impacts of the pharmaceutical drug Carbamazepine and pH 7.1, acting alone and in combination, on the clamScrobicularia plana. For this, a long-termexposure (28 days)was conducted and a set of oxidative stress markers was investigated. The results obtained showed that S. plana was able to develop mechanisms to prevent oxidative damage when under low pH for a long period, presenting higher survival when exposed to this stressor compared to CBZ or the combination of CBZ with pH 7.1. Furthermore, the toxicity of CBZ on S. plana was synergistically increased under ocean acidification conditions (CBZ + pH 7.1): specimens survival was reduced and oxidative stress was enhanced when compared to single exposures. These findings add to the growing body of evidence that ocean acidification will act to increase the toxicity of CBZ to marine organisms,which has clear implications for coastal benthic ecosystems suffering chronic pollution from pharmaceutical drugs. Different types of supports can be used in the immobilization of photoactive species, such as porphyrins (Pors) and phthalocyanines (Pcs). This mini-review will consider the different methodologies for the immobilization of Pors and Pcs and their photocatalytic performance in the photodegradation of organic pollutants in water, addressing also their recycling ability in successive water treatments. Among the most abundant drugs in the environment is caffeine, which has been reported as an effective chemical anthropogenic marker. However, as for the majority of pharmaceuticals, scarce information is available on the adverse effects of caffeine on marine benthic organisms, namely polychaetes which are the most abundant group of organisms in several aquatic ecossystems. Thus, the present study aimed to evaluate the biochemical alterations induced by environmentally relevant concentrations of caffeine on the polychaete species Diopatra neapolitana and Arenicola marina. The results obtained demonstrated that after 28 days exposure oxidative stress was induced in both species, especially noticed in A. marina, resulting from the incapacity of antioxidant and biotransformation enzymes to prevent cells from lipid peroxidation. The present study further revealed that D. neapolitana used glycogen and proteins as energy to develop defense mechanisms while in A. marina these reserves were maintained independently on the exposure concentration, reinforcing the low capacity of this species to fight against oxidative stress. For this reason, caffeine is continuously released into the environment with potential impacts on inhabiting organisms. The current study evaluated the biochemical alterations induced in the clam species Ruditapes philippinarum after exposure for 28 days to caffeine (0.5, 3.0 and 18.0 mg/L). The results obtained showed that, with the increasing caffeine concentrations, an increase in clams defense mechanisms (such as antioxidant and biotransformation enzymes activity) was induced which was accompanied by an increase in protein content. Nevertheless, although an increase on defense mechanisms was observed, clams were not able to prevent cells from lipid peroxidation that increased with the increase of caffeine concentration. Furthermore, with the increase of exposure concentrations, clams increased their metabolic activity (measured by electron transport activity), reducing their energy reserves (glycogen content), to fight against oxidative stress. Overall, the present study demonstrated that caffeine may impact bivalves, even at environmentally relevant concentrations, inducing oxidative stress in organisms. The present study is an important contribution to address knowledge gaps regarding the impacts of long-term exposures to pharmaceuticals since most of the studies assessed the effects after acute exposures, most of them up to 96 h. These include increasing discharges of pharmaceutical contaminants into the aquatic coastal systems. Pharmaceutical drugs are often detected in aquatic environments but still information on their toxicity impacts on inhabiting species is scarce, especially when acting in combination. Furthermore, almost no information is available on the impacts of pharmaceuticals in polychaetes, often the most abundant taxon in benthic communities and commonly used as indicator species of environmental conditions. Therefore, the present study aimed to evaluate the biochemical alterations induced in the polychaete Hediste diversicolor, from a low contaminated area at the Ria de Aveiro lagoon (Portugal), by the antiepileptic drug carbamazepine (0.0 - control, 0.3, 3.0, 6.0 and 9.0 μg/L) and the stimulant caffeine (0.0 - control, 0.5, 3.0, and 18.0 μg/L), acting alone and in combination (0.3 CBZ + 0.5 CAF and 6.0 CBZ + 3.0 CAF). Glutathione Stransferases (GSTs), superoxide dismutase (SOD) and catalase (CAT) activities was determined in Hediste diversicolor from each condition. Lipid peroxidation (LPO), glutathione reduced and oxidized (GSH and GSSG), glycogen and electron transport system (ETS) were also measured. The results obtained clearly revealed that both drugs induced oxidative stress in H. diversicolor, shown by the increase on LPO levels and decrease on total glutathione and GSH/GSSG ratio with the increase of exposure concentrations. Furthermore, the present findings demonstrated that polychaetes biotransformation capacity as well as antioxidant defense mechanisms were not sufficiently efficient to fight against the excess of reactive oxygen species (ROS) leading to LPO when organisms were exposed to both drugs. Our results also demonstrated that polychaetes tended to decrease the activity of ETSwhen exposed to drugs, avoiding energy expenditurewhich may prevent them fromgreater damages. The present study further revealed that the impacts induced by the combination of both drugswere similar to those obtained at the highest drugs concentrations acting alone. T1 - Toxic effects of the antihistamine cetirizine in mussel Mytilus galloprovincialis N2 - Recent studies have become increasingly focused on the assessment of pharmaceuticals occurrence in aquatic ecosystems, however the potential toxicity to non-target organisms is still largely unknown. The antihistamine cetirizine is a commonly used pharmaceutical, already detected in surface waters of marine aquatic systems worldwide. In the present study Mytilus galloprovincialis mussels were exposed to a range of cetirizine concentrations (0.3, 3.0, 6.0 and 12.0 mu/L), resembling moderate to highly contaminated areas, over 28 days. The responses of different biochemical markers were evaluated in mussels whole soft tissue, and included energy-related parameters (glycogen content, GLY; protein content, PROT; electron transport system activity, ETS), and oxidative stress markers (superoxide dismutase activity, SOD; catalase activity, CAT; glutathione S-transferases activity, GSTs; lipid peroxidation levels, LPO; reduced (GSH) and oxidized (GSSG) glutathione content). The results obtained demonstrated that with the increase of exposure concentrations mussels tended to increase their energy reserves and maintain their metabolic potential, which was significantly higher only at the highest concentration. Our findings clearly revealed that cetirizine inhibited the activity of GSTs and although induced the activity of antioxidant enzymes (SOD and CAT) mussels were not able to prevent cellular damages observed through the increase of LPO associated to the increase of exposure concentrations. Thus, this study confirmed that cetirizine induces toxic effects in Mytilus galloprovincialis, which, considering their trophic relevance, wide use as bioindicator and wide spatial distribution of this species, can result in ecological and economic negative impacts at a large scale. T1 - Physiological and biochemical alterations induced in the mussel Mytilus galloprovincialis after short and long-term exposure to carbamazepine N2 - The bivalve Mytilus galloprovincialis collected in the Ria de Aveiro, was selected to evaluate the acute and chronic effects of carbamazepine (CBZ) at environmentally relevant concentrations. CBZ is an antiepileptic drug widely found in the aquatic environment with toxic effects to inhabiting organisms. However, few studies evaluated the acute and chronic toxicity of this drug. The experiment was performed 'by exposing mussels to 0.0, 0.3, 3.0, 6.0 and 9.0 CBZ mu g/L, for 96 h and 28 days. To assess the toxicity of the drug, a battery of biomarkers related to mussels general physiological health status and oxidative stress was applied. CBZ was quantified in mussel tissues by an Enzyme-Linked Immunosorbent Assay (ELISA). The results obtained show that CBZ did not induce oxidative stress. However, our findings,demonstrated that the drug was taken up by mussels even though presenting low bioconcentration factor (BCF) values (up to 2.2). Furthermore, our results demonstrated that after a chronic exposure the physiological parameters, namely the condition and gonadosomatic indices, were negatively affected which may impair organisms' reproductive capacity with consequences to population sustainability. The easy recovery of these heterogeneous photocatalysts, just by applying an external magnetic field, allows their reuse in multiple treatment cycles. The photocatalytic activity of the non-immobilized photosensitizers and the obtained hybrid materials was evaluated in the degradation of 17 beta-estradiol, as a model organic pollutant present in water, using batch and flow mode treatment systems, assisted by visible light radiation (4 mW cm(-2)). The flow mode system potentiated the photocatalytic capacity of these novel hybrid materials. In order to improve the process, further studies based on different photocatalyst concentration and pH conditions were performed. The various forms of composite materials, due to their high material properties, are widely used as structural materials in the aviation, space, marine, automobile, and sports industries. However, some defects like voids, delamination, or inhomogeneous fiber distribution that form during the fabricating processes of composites can seriously affect the mechanical properties of the composite material. In this study, several imaging NDE techniques such as: thermography, high frequency eddy current, ultrasonic, x-ray radiography, x-ray laminography, and high resolution x-ray CT were conducted to characterize the microstructure of carbon fiber composites. Then, a 3D analysis was implemented by the destructive technique of serial sectioning for the same sample tested by the NDE methods. To better analyze the results of this work and extract a clear volume image for all features and defects contained in the composite material, an intensive comparison was conducted among hundreds of 3D-NDE and multi serial sections’ scan images showing the microstructure variation. In this work, silica-coated Magnetite nanoparticles (NPs) were used as carriers of glycosylated porphyrins and phthalocyanines. Their subsequent cationization resulted in the production of stable antimicrobial photosensitizing materials, effective against E. coli. Suspensions of the photocatalysts in water present bimodal size distributions formed by big clusters and small NPs with hydrodynamic diameters between 8 and 38 nm. The presence of small NPs in the suspensions is related to an effective photodynamic inactivation (PDI) of E. coli cells. Glycosylation of the PS showed a positive effect on the PDI performance, which could be related to a higher accumulation of the photocatalyst over the bacterial cell membrane. In addition, these biocidal agents proved to be photostable and their photoactive performance decreased only between 23% and 28% upon 5 PDI cycles, mostly because of the loss of material between cycles, which makes them promising materials for water disinfection purposes. The underlying system model consists of two integrated sub-models: a condition model for predicting the deterioration state of the box girder and a structural model for evaluating the overall system reliability. The condition model is based on a dynamic Bayesian network (DBN) model which considers the spatial variation of the corrosion process. Inspection data are included in the calculation of the system reliability through Bayesian updating on the basis of the DBN model. To demonstrate the effect of partial inspections, the software prototype is applied to a case study of a typical highway bridge with six spans. It is time-consuming, monoclonality is not guaranteed, and the number of clones that can be screened is limited. Our approach employs a novel hapten-specific labeling technique of hybridoma cells. This allows for fluorescence-activated cell sorting (FACS) and single-cell deposition and thereby eliminates the above-mentioned problems. A two-step staining approach is used to detect antigen specificity and antibody expression: in order to detect antigen specificity, hybridoma cells are incubated with a hapten−horseradish peroxidase conjugate (hapten−HRP), which is subsequently incubated with a fluorophore-labeled polyclonal anti-peroxidase antibody (anti-HRP−Alexa Fluor 488). To characterize the expression of membrane-bound immunoglobulin G (IgG), a fluorophore-labeled anti-mouse IgG antibody (anti-IgG−Alexa Fluor 647) is used. Hundreds of labeled hybridoma cells producing monoclonal antibodies (mAbs) specific for a hapten were rapidly isolated and deposited from a fusion mixture as single-cell clones via FACS. Enzyme-linked immunosorbent assay (ELISA) measurements of the supernatants of the sorted hybridoma clones revealed that all hapten-specific hybridoma clones secrete antibodies against the target. There are significant improvements using this high-throughput technique for the generation of mAbs including increased yield of antibody-producing hybridoma clones, ensured monoclonality of sorted cells, and reduced development times. A1 - Schneider, Thomas T1 - Tribological performance of MoS2 coatings in liquid helium and at high loads N2 - The main part of a narrow support element (NSE) of the W7-X superconducting coil system is an aluminium bronze pad, PVD coated on its spherical surface with MoS2, which slides against the flat surface of the stainless steel coil housing, coated with MoS2 spray. The operational requirements of the NSEs are: vacuum of p < 10-6 mbar, temperature T ~4 K, maximum load P ~1500 kN, typical displacement ≤5 mm, smooth sliding and no stick-slip events. The paper describes test results obtained with a downscaled NSE at T = 4.2 and 77 K. During the test the NSEs were submerged in liquid helium and nitrogen, respectively. Whereas the LN2 test ran smoothly for up to ~15,000 cycles, the test in LHe showed stick-slip from the very first cycle. The stick-slip disappeared after ~50 cycles. Post mortem analysis of the tested parts revealed that in case of LHe the sprayed MoS2 film was removed during the first 30-100 cycles by blistering and flaking. The reason for the loss of adhesion at LHe temperature is not known, several possible causes are under discussion. Further experiments under vacuum and at T ~4 K are being prepared which are expected to help in clarifying the issue. T1 - Modified paramagnetic beads in a microfluidic system for the determination of ethinylestradiol (EE2) in river water samples N2 - In this work, we have developed and characterized a novel microfluidic immunoassay methodology for rapid and sensitive quantification of ethinylestradiol (EE2) in river water samples. The detection of EE2 was carried out using a competitive direct immunoassay method based on the use of anti-EE2 polyclonal antibodies immobilized on magnetic microspheres 3-aminopropyl-modified manipulated for an external removable magnet. The EE2 present in the water sample was allowed to compete with EE2-horseradish peroxidase (HPR) conjugated for the immobilized anti-EE2 antibody. The HPR, in the presence of hydrogen peroxide (H2O2) catalyzes the oxidation of catechol (Q) whose back electrochemical reduction was detected on gold electrode at 0.0 V. The response current obtained from the product of enzymatic reaction is inversely proportional to the amount of EE2 in the water sample. The electrochemical detection can be done within 1 min and total assay time was 30 min. The calculated detection limits for electrochemical detection and the ELISA procedure are 0.09 and 0.32 ng L-1 respectively and the intra- and inter-assay coefficients of variation were below 5.8%. Our electrochemical immunosensor showed higher sensitivity and lower time consumed than the standard spectrophotometric detection ELISA method, which shows the potential for assessment of EE2 in river water samples. Usually, the geological subsoil and the building ground are the dominant sources for enhanced indoor radon levels. Additionally, building materials can increase indoor radon concentrations when these materials contain higher contents of 226Ra (radium), especially in combination with low air exchange rates. For a realistic estimation of indoor radon concentrations, it is helpful to carry out measurements of radon exhalation rates from relevant materials using emission test chambers. In Germany, it is aspired to limit the total indoor radon concentration to 100 Bq/m3, whereby building materials should contribute at most 20 Bq/m3. Within a project financed by the German Institute for Construction Technology (DIBt), a practical oriented measurement procedure of the radon exhalation of building materials in accordance to ISO 16000-9 was developed to have a means for the assessment of these materials with respect to their indoor use. Classic deformation monitoring (performed by strain gauges etc.) and temperature monitoring (Pt100 and alike) deliver data from fixed, single spots of a structure; quasidistributed measurements (fiber bragg gratings) provide a Chain of discrete measurement points along a limited sensing length. In contrast, an opticai fiber connected to a device for distributed strain and temperature sensing (DTSS) will provide a continuous profile of strain and temperature - spatially resolved down to less than 1 m - over a ränge of several tens of kilometers. In DTSS measurements, the nonlinear opticai effect of stimulated Brillouin Scattering (SBS) is employed: two light waves with a stable, tunable frequency offset are injected into opposite ends of the sensing fiber, where they will form a beat pattem, at which parts of the light will be scattered. By matching the frequency offset of the light waves to the propagation of acoustic fluctuation in the opticai fiber, a power transfer from one light wave to the other can be measured; since the acoustic propagation directly shifts with train and temperature of the fiber, these two quantities can be measured by tuning the light waves’ frequency offset. T1 - New possibilities for quality assurance of diaphragm wall joints N2 - Diaphragm walls are used to provide a stable and watertight confinement for the construction of tunnels, Underground railway stations, deep basements and other structures requiring deep excavations. Water seepage and soil loss through the joints have occurred on several occasions, causing extensive damage. This may have been due to uncontrolled concrete flow between the joints, unexpected geologic conditions or just poor workmanship. Different ways of quality assurance are applied on construction sites. Most of them rely on recording Parameters of the construction equipment and conventional concrete quality control. In the frame of the EUREKA/ZIM project DiWaQ we have looked at several ways to perform non-destructive Quality control. Some methods are to be applied after excavation before concreting (phase 1); others after the wall is completed (phase 2). For phase 1 we have used two different principles for checking the shape of the excavation, especially the joint to existing panels. A mechanical device which can be adapted to the excavating Equipment was developed; field tested and validated on test and construction sites. It is described in detail in another paper in the same volume. A sonar-based device with a 10 mm resolution has shown good performance in the lab, but is still under development. For phase 2 different temperature sensors have been lowered into access holes to monitor the hydration heat developed during concrete curing. The same tubes can also be used to perform electromagnetic and ultrasonic crosshole measurements across or along joints to detect flaws. Especially the ultrasonic method has shown promising results on Validation specimen at the BAM test site at Horstwalde, but limitations as well. Stony building materials are an important source of indoor radon. This article describes the determination of the exhalation rate of stony construction materials by the use of commercially available measuring devices in combination with VOC emission test chambers. Five materials – two types of clay brick, clinker brick, light-weight concrete brick, and honeycomb brick – generally used for wall constructions were used for the experiments. Their contribution to real room concentrations was estimated by applying room model parameters given in ISO 16000-9, RP 112, and AgBB. This knowledge can be relevant, if for instance indoor radon concentration is limited by law. It is excreted from humans and released via sewage treatment plant effluents into aquatic environments. EDCs are any environmental pollutant chemical that, once incorporated into an organism, affects the hormonal balance of various species including humans. Its presence in the environment is becoming of great importance in water quality. This paper describes the development of an accurate, sensitive and selective method for capture, preconcentration and determination of EE2 present in water samples using: magnetic particles (MPs) as bioaffinity support for the capture and preconcentration of EE2 and a glassy carbon electrode modified with multi-walled carbon nanotubes (MWCNTs/GCE) as detection system. The capture procedure was based on the principle of immunoaffinity, the EE2 being extracted from the sample using the anti-EE2 antibodies (anti-EE2 Ab) which were previously immobilized on MPs. Subsequently the analyte desorption was done employing a sulfuric acid solution and the determination of the EE2 in the pre-concentrated solution was carried out by square wave voltammetry (SWV). This method can be used to determine EE2 in the range of 0.035–70 ng L-1 with a detection limit (LOD) of 0.01 ng L-1 and R.S.D. < 4.20%. The proposed method has been successfully applied to the determination of EE2 in water samples and it has promising analytical applications for the direct determination of EE2 at trace levels. Main prerequisites for reliable Φf measurements, which are for transparent luminophore solutions commonly done relative to a reference, i.e., a fluorescence quantum yield standard of known Φf, are reliable and validated instrument calibration procedures to consider wavelength-, polarization-, and time-dependent instrument specific signal contributions, and sufficiently well characterized fluorescence quantum yield standards. As the standard’s Φf value directly contributes to the calculation of the sample’s Φf, its accuracy presents one of the main sources of uncertainty of relative Φf measurements. To close this gap, we developed a first set of 12 fluorescence quantum yield standards, which absorb and emit in the wavelength region of 330−1000 nm and absolutely determined their Φf values with two independently calibrated integrating sphere setups. Criteria for standard selection and the configuration of these novel fluorescence reference materials are given, and the certification procedure is presented including homogeneity and stability studies and the calculation of complete uncertainty budgets for the certified Φf values. While some discontinuities in AM microstructures stem from similar mechanisms as observed in more traditional processes such as casting, others are unique to the AM process. Discontinuities in AM are challenging to detect, due to their submicron size and orientation dependency. Toward the goal of improving structural integrity, minimizing discontinuities in an AM build requires an understanding of the mechanisms of formation to mitigate their occurrence. This study utilizes various techniques to evaluate the shape, size, nature and distribution of discontinuities in AM Inconel 718, in a non-hot isostatic pressed (HIPed) as-built, non-HIPed and direct age, and HIPed with two step age samples. Non-destructive synchrotron radiation refraction and transmission radiography (SXRR) provides additional information beyond that obtained with destructive optical microscopy. SXRR was able to distinguish between voids, cracks and lack of melt in, due to its sensitivity to the orientation of the discontinuity. Enzyme-linked immunosorbent assays, with photometric detection of 3,3’,5,5’-tetramethylbenzidine (TMB),form a good basis for sensitive detection. To provide a straightforward approach for the miniaturization of the detectionstep, we have studied the pitfalls of the electrochemical TMB detection. By cyclic voltammetry it was found that the TMB electrochemistry is strongly dependent on the pH and the electrode material. A stable electrode response to TMB could be achieved at pH 1 on gold electrodes. We created a smartphonebased, electrochemical, immunomagnetic assay for the detection of ochratoxin A in real samples, providing a solid basis forsensing of further analytes. Proper knowledge of their levels in wastewater helps to track sources of pollutants in natural waters and allows for calculation of removal efficiencies in wastewater Treatment plants. Here, we describe the development and application of an indirect competitive, multiplexing suspension Array fluorescence immunoassay (SAFIA) for the detection of carbamazepine (CBZ), diclofenac (DCF), caffeine (CAF), and isolithocholic acid (ILA) in wastewater, covering those classes of anthropogenic markers. The assay consists of haptens covalently conjugated to fluorescence-encoded polystyrene core/silica shell microparticles to create a site for competitive binding of the antibodies (Abs). Bound Abs are then stained with fluorophore-labeled Abs. Encoding and signaling fluorescence of the particles are determined by an automated flow cytometer. For compatibility of the immunoassay with the 96-well microtiter plate format, a stop reagent, containing formaldehyde, is used. This enables a wash-free procedure while decreasing time-to-result. Detection limits of 140 ± 40 ng/L for CBZ, 180 ± 110 ng/L for CAF, 4 ± 3 ng/L for DCF, and 310 ± 70 ng/L for ILA are achieved, which meet the sensitivity criteria of wastewater analysis. We demonstrate the applicability of SAFIA to real wastewater samples from three different wastewater Treatment plants, finding the results in good agreement with LC-MS/MS. Moreover, the accuracy in general exceeded that from classical ELISAs. We therefore propose SAFIA as a quick and reliable approach for wastewater analysis meeting the requirements for process analytical technology. For additively manufactured Inconel 718, the recently developed standard ASTM F3301 provides guidance for heat treatment of powder bed fusion specimens. Although this standard is based on standards developed for wrought Inconel 718, it does not include direct aging. Since direct aging reduces the number of processing steps, it can result in a post processing cost reduction if the desired properties are obtained. In this study, we characterized the microstructure and tensile behavior of Inconel 718 specimens produced by a laser powder bed fusion process. The specimens were heat treated according to two different routines after stress relieving: a full heat Treatment versus a one-step direct aging process. Differences in the resulting texture and grain morphology were observed. The ex-situ stress-strain behavior was broadly similar. However, a slight increase in yield strength was observed for the direct aged specimen. In order to understand this behavior, investigations with in-situ synchrotron Energy dispersive X-ray diffraction tensile testing revealed differences in the load partitioning among different Crystal directions. Importantly, the elastic anisotropy expressed by the magnitude of the diffraction elastic constants showed a dependency on the microstructures. Distinguished features like high specific surface area and outstanding electrical conductivity make them suitable for electrochemical biosensing applications. Here, we report the development of a biosensor involving the functionalized MXene−titanium carbide nanosheets (Ti3C2Tx-NS) and monoclonal antibodies against the SARS-CoV-2 nucleocapsid protein (anti-SARS-CoV-2 mAb) to design a point-of-care device for detection of the SARS-CoV-2 nucleocapsid protein (SARS-CoV-2 NP) antigen. Few-layered titanium carbide nanosheets (denoted as FL-Ti3C2Tx-NS) have been synthesized using a single-step etching and delamination method and characterized using optical and electron microscopy techniques revealing the suitability for immunosensing applications. Binding studies revealed the excellent affinity between the biosensor and the SARS-CoV-2 NP. Electrochemical detection of SARS-CoV-2 NP is performed using differential pulse voltammetry and read by a smartphone-based user interface. The proposed FL-Ti3C2Tx-NS based biosensor offers the detection of SARS-CoV-2 NP with a limit of detection of 0.91 nM in a wide detection range in spiked saliva samples. Additionally, there is no cross-reactivity in the presence of potential interferants like SARS-CoV-2 spike glycoprotein and bovine serum albumin. These findings demonstrate the potential of MXenes in developing a rapid and reliable tool for SARS-CoV-2 NP detection. T1 - Pushing up the magnetisation values for iron oxide nanoparticles via zinc doping: X-ray studies on the particle's sub-nano structure of different synthesis routes N2 - The maximum magnetisation (saturation magnetisation) obtainable for iron oxide nanoparticles can be increased by doping the nanocrystals with non-magnetic elements such as zinc. Herein, we closely study how only slightly different synthesis approaches towards such doped nanoparticles strongly influence the resulting sub-nano/atomic structure. We compare two co-precipitation approaches, where we only vary the base (NaOH versus NH3), and a thermal decomposition route. These methods are the most commonly applied ones for synthesising doped iron oxide nanoparticles. The measurable magnetisation change upon zinc doping is about the same for all systems. However, the sub-nano structure, which we studied with Mössbauer and X-ray absorption near edge spectroscopy, differs tremendously. We found evidence that a much more complex picture has to be drawn regarding what happens upon Zn doping compared to what textbooks tell us about the mechanism. Our work demonstrates that it is crucial to study the obtained structures very precisely when “playing” with the atomic order in iron oxide nanocrystals. Das Modell liefert eine sich kontinuierlich ändernde probabilistische Zustandsbewertung. Die Zustandsbewertung gibt eine Aussage über den Zustand und die Zuverlässigkeit des Brückensystems und seiner Bauteile und dient als Grundlage für die Planung und die Optimierung von Maßnahmen. Die Verwendung von Resultaten aus Inspektionen, Prüfungen und Überwachungen erfolgt durch eine Aktualisierung der Modellparameter. Die Aktualisierung beruht auf der Methode der Bayes'schen Aktualisierung und wird auf der Grundlage der entwickelten Klassifizierung der Bauwerksinformationen mit entsprechenden Methoden durchgeführt. Dieses Verfahren erlaubt es, alle Informationen in konsistenter Weise in ein einziges Modell einfließen zu lassen. Dabei wird die Genauigkeit und Aussagekraft der gewonnenen Daten und Beobachtungen explizit berücksichtigt. Durch die Aktualisierung der Modellparameter unter Berücksichtigung von Systemeffekten wird die Zustandsbewertung der Bauteile und des Brückensystems aktualisiert. The central part of the concept is a model describing the bridge system. The overall bridge model is comprised of deterioration models and a mechanical system model of the structure. The model is a-priori characterized by the input data (such as the geometry, the materials, and the utilization of the bridge). Significant uncertainties in the deterioration modeling as well as the load conditions motivate a probabilistic modeling. The model enables a continuously updated probabilistic condition assessment. It provides information about the condition and the reliability of the bridge system and its components and serves as a basis for the planning and the optimization of maintenance actions. Inspection, measurement, and monitoring information are included by updating the model parameters. Bayesian updating is utilized for the updating of the model parameters and is performed on the basis of the developed classification of structural information and the corresponding procedures. These methodologies allow to consistently incorporate all available information into the model and the prediction. Thereby, the precision and the validity of the data and observations are explicitly accounted for. The condition assessment of the bridge system and its components is updated by updating the model parameters, and maintenance actions can be planned and optimized taking into account all available information in a rational and consistent manner. In this way, the intelligent bridge with inspections, measurements, and monitoring information becomes an adaptive system, which has the ability to adjust to changes. The cells containing the non-toxic nanoprobes were shown to be displaceable in an external magnetic field and can be manipulated in microfluidic channels. The distribution of the composite nanostructures that are contained in the endosomal system is discussed on the basis of surfaceenhanced Raman scattering (SERS) mapping, quantitative laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) micromapping, and cryo soft X-ray tomography (cryo soft-XRT). Cryo soft-XRT of intact, vitrified cells reveals that the composite nanoprobes form intra-endosomal aggregates. The nanoprobes provide SERS signals from the biomolecular composition of their surface in the endosomal environment. The SERS data indicate the high stability of the nanoprobes and of their plasmonic properties in the harsh environment of endosomes and lysosomes. The spectra point at the molecular composition at the surface of the Ag–Magnetite and Au–Magnetite nanostructures that is very similar to that of other Composite structures, but different from the composition of pure silver and gold SERS nanoprobes used for intracellular investigations. As shown by the LA-ICP-MS data, the uptake efficiency of the magnetite composites is approximately two to three times higher than that of the pure gold and silver nanoparticles. By our analysis technique we were able to determine the nano- and micro dispersion from the experimental results by our numerical analysis. These results are discussed with respect to crucial advantages of the generically 3-dimensional tomography technique as compared to 2-dimensional methods. Our quantitative analysis of the 3d images demonstrates that in the case of our model composites the filler dispersion is good, independent on the filler concentration. Our new 3-dimensional results evidence that considering only 2-d pictures may lead to ambiguities that can lead to wrong conclusions. Conventional diagnostic approaches often fall short, particularly for dense tissues, and are frequently invasive, costly, and of limited availability. This reinforces the need for a compact, economical, and ultrasensitive assay that is operationally simple and interpretable. We present an efficient electrochemical detection platform for the cancer biomarker mucin 1 (MUC1). A fluorine-doped tin oxide (FTO) surface was modified with an iron-based metal–organic framework (FeMOF) intercalated with palladium nanorods (PdNR). FeMOF was prepared using Fe3+/Fe2+ precursors at a 1.2/1 mmol ratio and dual ligands, i.e. tetrahydroxy-1,4-benzoquinone and 2-aminobenzene-1,4-dicarboxylic acid. AntiMUC1 antibodies were immobilized on a modified electrode via p-phenylenediamine (PDA) (FTO/FeMOF@PdNR/PDA/antiMUC1Ab) and evaluated using electrochemical impedance spectroscopy (EIS) and voltammetry. The designed sensor demonstrated an excellent binding affinity for the MUC1 antigen. Among these techniques, the EIS method stands out for its technical performance, as evidenced by the high sensitivity (detection limit 0.074 fg mL−1), quantification limit 0.24 fg mL−1, and high analytical sensitivity (1.39 × 103 Ω fg−1 mL−1 cm−2). The negligible cross-reactivity with interferent biomolecules, rapid response (10-minute equilibrium), regenerability up to 5 cycles, high reproducibility (RSD ∼1–3%), and long-term stability (up to 35 days) further validate the suitability of the proposed MUC1 immunosensor. This study presents an ultrasensitive biosensor that is compact, cost-effective, and easy for individuals at home to use after further development into a kit-based end product. In this contribution, we thoroughly Review the formulation of the equivalent time-invariant reliability problems and extend this approximation to structures for which inspection and monitoring data is available. Thereafter, we present methods for efficiently evaluating the reliability over time. In particular, we propose the combination of sampling-based methods with a FORM (first-order reliability method) approximation of the series system reliability problem that arises in the computation of the lifetime reliability. The Yeast Estrogen Screen (YES) assay is an excellent method to evaluate the presence of these compounds in water due to its simplicity and capacity to assess the bioaccessible forms/fractions of these compounds. In the presence of a compound with estrogenic activity, Saccharomyces cerevisiae cells, containing a lacZ reporter gene encoding the enzyme β-galactosidase, are induced, the enzyme is synthesised, and released to the extracellular medium. In this work, a YES-based approach encompassing the use of a lacZ reporter gene modified strain of S. cerevisiae, microcarriers as solid support, and a fluorescent substrate, fluorescein di-β-D-galactopyranoside, is proposed, allowing for the assessment of EDCs’ presence after only 2 h of incubation. The proposed method provided an EC50 of 0.17 ± 0.03 nM and an LLOQ of 0.03 nM, expressed as 17β-estradiol. The assessment of different EDCs provided EC50 values between 0.16 and 1.2 × 103 nM. After application to wastewaters, similar results were obtained for EDCs screening, much faster, compared to the conventional 45 h spectrophotometric procedure using a commercial kit, showing potential for onsite high-throughput screening of environmental contamination. Cell clones specific for five different haptens were analyzed. We selectively double stained and analyzed fixed hybridoma cells with fluorophore-labeled haptens to demonstrate the target-selectivity, and with a fluorophore-labeled anti-mouse IgG antibody to characterize the level of surface expression of membrane-bound IgGs. ELISA measurements with the supernatants of the individual hybridoma clones revealed that antibodies from those cells, which showed the highest fluorescence intensities in the flow cytometric analysis, also displayed the highest affinities for the target antigens. The fluorescence intensity of antibody-producing cells corresponded well with the produced antibodies' affinities toward their respective antigens. Immunohistochemical staining verified the successful double labeling of the cells. Our method makes it possible to perform a high-throughput screening for hybridoma cells, which have both an adequate IgG production rate and a high target affinity. T1 - Zerstörungsfreie Materialprüfung in Gegenwart und Zukunft - 50 Jahre Deutsche Gesellschaft für Zerstörungsfreie Prüfung N2 - Die Darstellung der Arbeiten und Aufgaben der DGZfP und ihrer Mitglieder zeigt, daß sie sich auf die Förderung der in die Zukunft gerichteten Forschungs- und Entwicklungsschwerpunkte und auf Arbeiten für wirtschaftlich erfolgreiche Anwendungen konzentriert; intensiv ein Programm der Ausbildung und Qualifizierung von Prüfpersonal durchführt, das den Bedürfnissen aller Partner der Wirtschaft gerecht wird und das hoffentlich in den wichtigsten Teilen international gültig werden kann. Die DGZfP ist sich in ihren Bemühungen in Deutschland der Unterstützung durch alle wichtigen Behörden und Dienststellen sicher und setzt zudem Vertrauen in die ausländischen ZfP-Partner und in die Bereitschaft zur internationalen Zusammenarbeit. Unter Berücksichtigung dessen und des Bewußtseins der eigenen Möglichkeiten glaubt sie, daß sie den gesetzten Verpflichtungen gerecht werden kann, nämlich die zerstörungsfreien Prüfungen und Verfahren, richtig anzuwenden, Aussagen und Folgerungen durch Forschung, Entwicklung, Anwendung und Ausbildung zu intensivieren und zu unterstützen. Die zerstörungsfreie Materialprüfung der Gegenwart und Zukunft wird von zwei menschlichen Eckpfeilern bestimmt: Einmal von den Menschen, die auf der Basis solider Fachkenntnisse die Materialprüfung erfolgreich durchführen, und zum anderen von den Menschen, die als Forscher, Ingenieure und Techniker sich den Anforderungen für Forschung und praxisbezogene Anwendung stellen, die ideenreich Geräte, Anlagen und Verfahren konzipieren und realisieren, mit denen Prüfanforderungen vielfältiger Art erfüllt werden können. Beide Gruppen müssen die gegenseitigen nationalen und internationalen Kontakte pflegen und intensivieren. ED - Resch-Genger, Ute ED - Schneider, Ralf T1 - pH and concentration dependence of the optical properties of thiol-capped CdTe nanocrystals in water and D2O N2 - The optical properties of semiconductor nanocrystals (SC NCs) are largely controlled by their size and surface chemistry, i.e., the chemical composition and thickness of inorganic passivation shells and the chemical nature and number of surface ligands as well as the strength of their bonds to surface atoms. The latter is particularly important for CdTe NCs, which – together with alloyed CdₓHg₁₋ₓTe – are the only SC NCs that can be prepared in water in high quality without the need for an additional inorganic passivation shell. Aiming at a better understanding of the role of stabilizing ligands for the control of the application-relevant fluorescence features of SC NCs, we assessed the influence of two of the most commonly used monodentate thiol ligands, thioglycolic acid (TGA) and mercaptopropionic acid (MPA), on the colloidal stability, photoluminescence (PL) quantum yield (QY), and PL decay behavior of a set of CdTe NC colloids. As an indirect measure for the strength of the coordinative bond of the ligands to SC NC surface atoms, the influence of the pH (pD) and the concentration on the PL properties of these colloids was examined in water and D₂O and compared to the results from previous dilution studies with a set of thiol-capped Cd₁₋ₓHgₓTe SC NCs in D₂O. As a prerequisite for these studies, the number of surface ligands was determined photometrically at different steps of purification after SC NC synthesis with Ellman’s test. Our results demonstrate ligand control of the pH-dependent PL of these SC NCs, with MPA-stabilized CdTe NCs being less prone to luminescence quenching than TGA-capped ones. For both types of CdTe colloids, ligand desorption is more pronounced in H₂O compared to D₂O, underlining also the role of hydrogen bonding and solvent molecules. Here, the authors present a generalized approach of how to extract the isotropic ellipsometric sample parameters from an imaging Mueller-matrix measurement even if the sample is significantly misaligned. They validate the method by layer-thickness calculations based on imaging Mueller-matrix measurements of flat crystalline silicon samples that were misaligned on purpose. Furthermore, they also exploit this method’s capabilities to perform spatially resolved layer-thickness measurements of a single-layer indium-tin-oxide coating on a fused-silica microlens without the need of realignment or repositioning of the sample during the measurement. T1 - Effect of curvature of spherical microscopic indentations on the ellipsometric readout N2 - The authors describe and interpret curvature-related changes to the ellipsometric readout. As model system for a concave curvature, a set of three spherical microscopic indents in silicon (100) of different sizes was prepared by instrumented indentation testing using a spherical indenter. For reference, these samples were characterized by AFM to reveal the topography of each structure. The concavelike indents were analyzed by Mueller-Matrix imaging ellipsometry to extract lateral intensity images of 12 elements of the Mueller-Matrix. As a result of the detailed analysis of the image elements m22, m23, and m14, it was possible to correlate intensity changes and symmetry properties to depolarization and cross polarization induced by the edge threshold and the curved surface of the indent. From fecal pellets of an individual mouse, a sufficient amount of active immunoglobulins or their fragments can be extracted to perform a regular examination of the status of the immune response by immunoassay. Hapten-specific antibodies from the feces of mice from three immunization trials showed very similar characteristics to those obtained from serum at a given date. Therefore, it can be suspected that some serum IgG enters the intestinal lumen and ends up in the feces, where they appear to be considerably stable. Hapten-specific IgAs were not found in the feces. Being able to analyze antibody titers in feces could be an interesting animal welfare refinement to standard practice that does not entail repeated blood sampling.
