For the effective usage of LTT filler materials, a deeper understanding of the complex processes that lead to the final residual stress state during multipass welding is necessary. Transformation kinetics and the strain evolution of multi-pass welds during welding were investigated in situ at the beamline HEMS@PETRAIII, Germany. Compared to conventional welds, the total strain was reduced and compression strain was achieved when using LTT filler materials. For an optimal use of the LTT effect in the root of multi-pass welds, the alloying concept must be adapted taking care of dilution. In this paper, the Progress made at Physikalisch-Technische Bundesanstalt (PTB) and BAMFederal Institute for Materials Research and Testing is presented and remaining questions are discussed. The projected characterization procedure at PTB, which is based on the established SIE method (sum of the individual estimates), using a new indium fixed-point cell is described as an example. This procedure includes an SI-traceable chemical Analysis of the material in the fixed-point cell with sufficiently low uncertainties, the individual experimental determination of the influence of the quantified impurities on the fixed-point temperature, and the establishment of direct links to the phase-transition temperatures of the national standard and of an assumed material of ideal purity. A characteristic difference to the common practice is the chemical analysis of the fixed-point metal being done after determining the cell’s freezing temperature. This allows for the detection and consideration of contamination and purification effects due to the filling process, or due to the contact with the carbon crucible and other parts of the fixed-point cell. A chemical analysis of an indium fixed-point was carried out by BAM with relative measurement uncertainties below 30 % which have not been previously achieved. The results provide evidence for the precipitation of some impurities, which is apparently inconsistent with the corresponding binary phase diagrams, but was explained in a recent publication. Implications for the use of the SIE method shall be described briefly at the end. T1 - Targeted luminescent near-infrared polymer-nanoprobes for in vivo imaging of tumor hypoxia N2 - Polystyrene nanoparticles (PS-NPs) were doped with an oxygen-sensitive near-infrared (NIR)-emissive palladium meso-tetraphenylporphyrin and an inert reference dye which are both excitable at 635 nm. The nanosensors were characterized with special emphasis on fundamental parameters such as absolute photoluminescence quantum yield and fluorescence lifetime. The PS-NPs were employed for ratiometric dual-wavelength and lifetime-based photoluminescent oxygen sensing. They were efficiently taken up by cultured murine alveolar macrophages, yielding a characteristic and reversible change in ratiometric response with decreasing oxygen concentration. This correlated with the cellular hypoxic status verified by analysis of hypoxia inducible factor-1α (HIF-1α) accumulation. In addition, the surface of PS-NPs was functionalized with polyethylene glycol (PEG) and the monoclonal antibody herceptin, and their binding to HER2/neu-overexpressing tumor cells was confirmed in vitro. First experiments with tumor-bearing mouse revealed a distinctive ratiometric response within the tumor upon hypoxic condition induced by animal sacrifice. These results demonstrate the potential of these referenced NIR nanosensors for in vitro and in vivo imaging that present a new generation of optical probes for oncology. The present work highlights that contour scanning is crucial to ensure geometrical accuracy and thereby the high performance under uniaxial compression of complex Alloy 718 lattice structures. Studies of X-ray computed tomography visualizations of as-built and compression-strained structures reveal the continuous and smooth bending and compression of the walls, and the earlier onset of internal contact appearance in the denser lattices printed with contour. In contrast, the effect of addition of He to the Ar process gas appears to have limited influence on the mechanical response of the lattices and their microstructure as characterized by electron backscattered diffraction. However, the addition of He proved to significantly enhance the cooling rate and to reduce the amount of the generated spatters as evidenced by in situ monitoring of the process emissions, which is very promising for the process stability and powder reusability during laser powder bed fusion. Das Brückensystem wird durch ein Modell beschrieben, welches den zentralen Teil des Konzeptes darstellt. Das Modell wird in Schädigungsmodelle und ein Tragwerkssystem-Modell unterteilt. Dieses Modell wird a-priori durch die Eingangsdaten (welche etwa die Geometrie, die Materialien und die Verwendung der Brücke beschreiben) charakterisiert. Aus diesen ergeben sich dann auch die Ausgangsmodelle. Um die signifikanten Streuungen und Unsicherheiten adäquat abzubilden sind diese Modelle probabilistisch. Das Modell liefert eine sich kontinuierlich ändernde probabilistische Zustandsbewertung. Die Zustandsbewertung gibt eine Aussage über den Zustand und die Zuverlässigkeit des Brückensystems und seiner Bauteile und dient als Grundlage für die Planung und die Optimierung von Maßnahmen. Die Verwendung von Resultaten aus Inspektionen, Prüfungen und Überwachungen erfolgt durch eine Aktualisierung der Modellparameter. Die Aktualisierung beruht auf der Methode der Bayes'schen Aktualisierung und wird auf der Grundlage der entwickelten Klassifizierung der Bauwerksinformationen mit entsprechenden Methoden durchgeführt. Dieses Verfahren erlaubt es, alle Informationen in konsistenter Weise in ein einziges Modell einfließen zu lassen. Dabei wird die Genauigkeit und Aussagekraft der gewonnenen Daten und Beobachtungen explizit berücksichtigt. Durch die Aktualisierung der Modellparameter unter Berücksichtigung von Systemeffekten wird die Zustandsbewertung der Bauteile und des Brückensystems aktualisiert. Das ermöglicht die Planung und die Optimierung von Maßnahmen unter Berücksichtigung der Bauwerksinformationen. The central part of the concept is a model describing the bridge system. The overall bridge model is comprised of deterioration models and a mechanical system model of the structure. The model is a-priori characterized by the input data (such as the geometry, the materials, and the utilization of the bridge). Significant uncertainties in the deterioration modeling as well as the load conditions motivate a probabilistic modeling. The model enables a continuously updated probabilistic condition assessment. It provides information about the condition and the reliability of the bridge system and its components and serves as a basis for the planning and the optimization of maintenance actions. Inspection, measurement, and monitoring information are included by updating the model parameters. Bayesian updating is utilized for the updating of the model parameters and is performed on the basis of the developed classification of structural information and the corresponding procedures. These methodologies allow to consistently incorporate all available information into the model and the prediction. Thereby, the precision and the validity of the data and observations are explicitly accounted for. The condition assessment of the bridge system and its components is updated by updating the model parameters, and maintenance actions can be planned and optimized taking into account all available information in a rational and consistent manner. In this way, the intelligent bridge with inspections, measurements, and monitoring information becomes an adaptive system, which has the ability to adjust to changes. T1 - Exploring polymorphism and stoichiometric diversity in naproxen/proline cocrystals N2 - We present naproxen/proline cocrystals discovered when combining enantiopure and racemic naproxen and proline. Using liquid-assisted grinding as the main method to explore the variety of crystal forms in this system, we found 17 cocrystals, of which the structures of only four of them were previously known. The naproxen/proline system exhibited multiple polymorphs of 1 : 1 stoichiometry as well as more rare cocrystals with 1 : 2 and 2 : 3 stoichiometries, two cocrystal hydrates and one cocrystal solvate. In situ ballmilling, used to monitor liquid-assisted grinding reactions, revealed that the solvent dictates the reaction intermediates even if the final reaction product stays the same. Synchrotron X-ray diffraction data collected in situ upon heating allowed us to monitor directly the phase changes upon heating and gave access to pure diffraction patterns of several cocrystals, thus enabling their structure determination from powder X-ray diffraction data; this method also confirmed the formation of a conglomerate in the RS-naproxen/DL-proline system. Proline in cocrystals kept its ability to form charge-assisted head-to-tail N-H⋯O hydrogen bonds, typical of pure crystalline amino acids, thus increasing the percentage of strong chargeassisted interactions in the structure and consequently providing some of the cocrystals with higher melting points as compared to pure naproxen. The majority of drugs are chiral, and hence, these data are of importance to the pharmaceutical industry as they provide insight into the challenges of chiral cocrystallization. An investigation of gas phase explosion propagation limits on ethene mixtures N2 - A method for the determination of safety properties for micro reactors and micro structured components is presented. Micro structured reactors are not inherently safe but the range of safe operating conditions of micro reactors are extended since the explosion region is reduced. The λ/3 rule was demonstrated to be applicable to micro scale tubes for stoichiometric mixtures of ethane–oxygen and ethane–nitrous oxide. Furthermore first results from an investigation concerning detonation propagation through a micro reactor of non-ideal geometry are shown. T1 - Integration of the 1,2,3-triazole 'click' motif as a potent signalling element in metal ion responsive fluorescent probes N2 - In a systematic approach we synthesized a new series of fluorescent probes incorporating donor–acceptor (D-A) substituted 1,2,3-triazoles as conjugative π-linkers between the alkali metal ion receptor N-phenylaza-[18]crown-6 and different fluorophoric groups with different electron-acceptor properties (4-naphthalimide, meso-phenyl-BODIPY and 9-anthracene) and investigated their performance in organic and aqueous environments (physiological conditions). In the charge-transfer (CT) type probes 1, 2 and 7, the fluorescence is almost completely quenched by intramolecular CT (ICT) processes involving charge-separated states. In the presence of Na+ and K+ ICT is interrupted, which resulted in a lighting-up of the fluorescence in acetonitrile. Among the investigated fluoroionophores, compound 7, which contains a 9-anthracenyl moiety as the electron-accepting fluorophore, is the only probe which retains light-up features in water and works as a highly K+/Na+-selective probe under simulated physiological conditions. Virtually decoupled BODIPY-based 6 and photoinduced electron transfer (PET) type probes 3–5, where the 10-substituted anthracen-9-yl fluorophores are connected to the 1,2,3-triazole through a methylene spacer, show strong ion-induced fluorescence enhancement in acetonitrile, but not under physiological conditions. Electrochemical studies and theoretical calculations were used to assess and support the underlying mechanisms for the new ICT and PET 1,2,3-triazole fluoroionophores. Here, the authors present a generalized approach of how to extract the isotropic ellipsometric sample parameters from an imaging Mueller-matrix measurement even if the sample is significantly misaligned. They validate the method by layer-thickness calculations based on imaging Mueller-matrix measurements of flat crystalline silicon samples that were misaligned on purpose. T1 - Effect of curvature of spherical microscopic indentations on the ellipsometric readout N2 - The authors describe and interpret curvature-related changes to the ellipsometric readout. As model system for a concave curvature, a set of three spherical microscopic indents in silicon (100) of different sizes was prepared by instrumented indentation testing using a spherical indenter. For reference, these samples were characterized by AFM to reveal the topography of each structure. The concavelike indents were analyzed by Mueller-Matrix imaging ellipsometry to extract lateral intensity images of 12 elements of the Mueller-Matrix. As a result of the detailed analysis of the image elements m22, m23, and m14, it was possible to correlate intensity changes and symmetry properties to depolarization and cross polarization induced by the edge threshold and the curved surface of the indent.
