Nanoplastic toxicity studies are complicated by the lack of available, well-characterized test and reference materials. Further, many nanoplastic test materials are inherently more polydisperse and heterogenous in shape compared to polystyrene beads, making accurate and representative size distribution measurements particularly challenging. The aim of this study was to conduct an inter-laboratory comparison of dynamic light scattering measurements, the most commonly used particle sizing method for nanomaterials. Using a published standard operating procedure, size measurements in water and a standardized cell culture medium (CCM) were generated for spherical, carboxy-functionalized polystyrene nanoparticles (PS-COOH; 50 nm; benchmark material), and for increasingly complex in-house produced spherical poly(ethylene terephthalate) (nanoPET) and irregular shaped polypropylene (nanoPP) test materials. The weighted mean of hydrodynamic diameters of PS-COOH dispersed in water (55 ± 5 nm) showed moderate variation between labs (coefficient of variation, CV = 8.2%) and were similar to literature reports. Measurements of nanoPET (82 ± 6 nm) and nanoPP (182 ± 12 nm) in water exhibited similar CV values (nanoPET: 7.3% and nanoPP; 6.8%). Dispersion of PS-COOH and nanoPET in CCM increased the CV to 15.1 and 14.2%, respectively, which is lower than literature reports (CV = 30%). Vacuum matrix-assisted ionization onan intermediate pressure matrix-assisted laser desorption/ionization sourcewithout the use of a laser, high voltages, or any other added energy wassimply unbelievable, at first. Individually and as a whole, the variousdiscoveries and inventions started to paint, inter alia, an exciting new pictureand outlook in mass spectrometry from which key developments grew thatwere at the time unimaginable, and continue to surprise us in its simplisticpreeminence. We, and others, have demonstrated exceptional analyticalutility. Our current research is focused on how best to understand, improve, and use these novel ionization processes throughdedicated platforms and source developments. These ionization processes convert volatile and nonvolatile compounds from solid orliquid matrixes into gas-phase ions for analysis by mass spectrometry using, e.g., mass-selected fragmentation and ion mobilityspectrometry to provide accurate, and sometimes improved, mass and drift time resolution. The combination of research anddiscoveries demonstrated multiple advantages of the new ionization processes and established the basis of the successes that lead tothe Biemann Medal and this Perspective. How the new ionization processes relate to traditional ionization is also presented, as wellas how these technologies can be utilized in tandem through instrument modification and implementation to increase coverage ofcomplex materials through complementary strengths. T1 - Metabolic specialization of denitrifiers in permeable sediments controls N2O emissions N2 - Coastal oceans receive large amounts of anthropogenic fixed nitrogen (N), most of which is denitrified in the sediment before reaching the open ocean. Sandy sediments, which are common in coastal regions, seem to play an important role in catalysing this N‐loss. Permeable sediments are characterized by advective porewater transport, which supplies high fluxes of organic matter into the sediment, but also leads to fluctuations in oxygen and nitrate concentrations. Little is known about how the denitrifying communities in these sediments are adapted to such fluctuations. Our combined results indicate that denitrification in eutrophied sandy sediments from the world's largest tidal flat system, the Wadden Sea, is carried out by different groups of microorganisms. This segregation leads to the formation of N2O which is advectively transported to the overlying waters and thereby emitted to the atmosphere. At the same time, the production of N2O within the sediment supports a subset of Flavobacteriia which appear to be specialized on N2O reduction. If the mechanisms shown here are active in other coastal zones, then denitrification in eutrophied sandy sediments may substantially contribute to current marine N2O emissions. T1 - The human factor - Results of a small-angle scattering data analysis round robin N2 - A round-robin study has been carried out to estimate the impact of the human element in small-angle scattering data analysis. Four corrected datasets were provided to participants ready for analysis. All datasets were measured on samples containing spherical scatterers, with two datasets in dilute dispersions and two from powders. Most of the 46 participants correctly identified the number of populations in the dilute dispersions, with half of the population mean entries within 1.5% and half of the population width entries within 40%. Due to the added complexity of the structure factor, far fewer people submitted answers on the powder datasets. For those that did, half of the entries for the means and widths were within 44 and 86%, respectively. This round-robin experiment highlights several causes for the discrepancies, for which solutions are proposed. T1 - Substrate and electron donor limitation induce phenotypic heterogeneity in different metabolic activities in a green sulphur bacterium N2 - Populations of genetically identical cells can display marked variation in phenotypic traits; such variation is termed phenotypic heterogeneity. Here, we investigate the effect of substrate and electron donor limitation on phenotypic heterogeneity in N2 and CO2 fixation in the green sulphur bacterium Chlorobium phaeobacteroides. We grew populations in chemostats and batch cultures and used stable isotope labelling combined with nanometer‐scale secondary ion mass spectrometry (NanoSIMS) to quantify phenotypic heterogeneity. Experiments in H2S (i.e. electron donor) limited chemostats show that varying levels of NH4+ limitation induce heterogeneity in N2 fixation. Comparison of phenotypic heterogeneity between chemostats and batch (unlimited for H2S) populations indicates that electron donor limitation drives heterogeneity in N2 and CO2 fixation. Our results demonstrate that phenotypic heterogeneity in a certain metabolic activity can be driven by different modes of limitation and that heterogeneity can emerge in different metabolic processes upon the same mode of limitation. In conclusion, our data suggest that limitation is a general driver of phenotypic heterogeneity in microbial populations. Nach ersten Anwendungen in der Astronomie und der Atmosphärenforschung wurden in letzter Zeit Anwendungsfelder in der Sicherheitstechnik, der Medizintechnik, der Nachrichtentechnik und der zerstörungsfreien Prozess- und Qualitätskontrolle erschlossen. Dielektrische Materialien wie Kunststoffe und Keramiken können von Terahertz-Wellen durchdrungen werden. Das ermöglicht Untersuchungen des Objektinneren ohne spezielle Schutzmaßnahmen, wie sie z.B. beim Röntgen notwendig sind. Die Richtlinie erleichtert die Verständigung zwischen Nutzern, Herstellern und dem Vertrieb von Terahertz-Systemen. In der Richtlinie werden bekannte Einsatzgebiete sowie die eingesetzten Verfahren und Funktionsprinzipien aufgeführt, um den Leser bei der Auswahlentscheidung zu unterstützen. Nach einer kurzen Einführung in die physikalischen Grundlagen und Hinweisen zur elektromagnetischen Verträglichkeit, zum Strahlenschutz und zur regulierten Frequenzvergabe werden die verschiedenen Geräteklassen von Terahertz-Systemen ausführlich vorgestellt. Die Richtlinie gibt Hinweise zur messtechnischen Charakterisierung von Terahertz-Wellen. T1 - Short-pulse laser induced transient structure formation and ablation studied with time-resolved coherent XUV-scattering N2 - XUV- and X-ray free-electron-lasers (FEL) combine short wavelength, ultrashort pulse duration, spatial coherence and high intensity. This unique combination of properties opens up new possibilities to study the dynamics of non-reversible phenomena with ultrafast temporal and nano- to atomic-scale spatial resolution. In this contribution we wish to present results of time-resolved experiments performed at the XUV-FEL FLASH (HASYLAB/Hamburg) aimed to investigate the nano-scale structural dynamics of laser-irradiated materials. Thin films and fabricated nano-structures, deposited on Si3N4-membranes, have been excited with ultrashort optical laser pulses. The dynamics of the non-reversible structural evolution of the irradiated samples during laser-induced melting and ablation has been studied in an optical pump - XUV-probe configuration by means of single-shot coherent scattering techniques (i.e. diffraction imaging [1]). In a first set of experiments we investigated the formation of laser induced periodic surface structures (LIPSS) on the surface of thin Si-films (thickness 100 nm). In a simplified view LIPPS are generated as a result of interference between the incident laser pulse and surface scattered waves which leads to a periodically modulated energy deposition. Time-resolved scattering using femtosecond XUV-pulses (with a wavelength of 13.5 nm and 7 nm) allowed us to directly follow LIPSS evolution on an ultrafast time-scale and with better than 40 nm spatial resolution. The observed scattering patterns show almost quantitative agreement with theoretical predictions [2] and reveal that the LIPSS start to form already during the 12 ps pump pulse. In the second set of measurements we studied picosecond and femtosecond laser induced ablation and disintegration of fabricated nano-structures. Correlations of coherent diffraction patterns measured at various time delays to the pattern of the undisturbed object show that order in the structure is progressively lost starting from short length scales. This structural rearrangement progresses at close to the speed of sound in the material. Under certain circumstances (e.g. adequate sampling) it became also possible to reconstruct real-space images of the object as it evolves over time [3]. The possibility of femtosecond single-shot imaging of ultrafast dynamic processes with nanoscale resolution provides yet more details of the physical processes involved. [1] H. N. Chapman et al. T1 - The human factor - Results of a small-angle scattering data analysis round robin N2 - A Round Robin study has been carried out to estimate the impact of the human element in small-angle scattering data analysis. Four corrected datasets were provided to participants ready for analysis. All datasets were measured on samples containing spherical scatterers, with two datasets in dilute dispersions, and two from powders. Most of the 46 participants correctly identified the number of populations in the dilute dispersions, with half of the population mean entries within 1.5 % and half of the population width entries within 40 %, respectively. Due to the added complexity of the structure factor, much fewer people submitted answers on the powder datasets. For those that did, half of the entries for the means and widths were within 44 % and 86 % respectively. This Round Robin experiment highlights several causes for the discrepancies, for which solutions are proposed. T1 - Acidity and Stability of Bronsted Acid Sites in Green Clinoptilolite Catalysts and Catalytic Performance in the Etherification of Glycerol N2 - Natural zeolite clinoptilolite CLIN with a framework ratio of Si/Al ≥ 4 containing mainly potassium and calcium ions in its internal channel system was used as a starting material. The acidic HCLIN catalysts were prepared under soft conditions avoiding the use of environmental less benign mineral acids. The starting material was ion exchanged using a 0.2 M aqueous ammonium nitrate solution at a temperature 80 ◦C for 2 h. The obtained NH4CLIN was converted into the acid HCLIN catalyst by calcination at 300–600 ◦C. The obtained samples were characterized by XRD, FTIR, SEM/TEM, AAS, and EDX element mapping. The state of aluminium and silicon was studied by 27Al- and 29SiMAS NMR spectroscopy. The textural properties of the catalysts were investigated by nitrogen adsorption and desorption measurements. The Brønsted acidity of the HCLIN catalysts was studied by temperature-programmed decomposition of the exchanged ammonium ions releasing ammonia as well as 1H MAS NMR, {1H–27Al} Trapdor, and {1H–27Al} Redor experiments. The strongly agglomerated samples were crystalline and thermally stable up to >500 ◦C. Although a part of the clinoptilolite framework is maintained up to 600 ◦C, a loss of crystallinity is already observed starting from 450 ◦C. The specific surface areas of the starting CLIN and ammonium exchanged NH4CLIN are low with ca. 26 m2/g. The pores are nearly blocked by the exchangeable cations located in the zeolite pores. The thermal decomposition of the ammonium ions by calcination at 400 ◦C causes an opening of the pore entrances and a markable increase in the specific micropore area and micropore volume to ca. 163 m2/g and 0.07 cm3/g, respectively. It decreases with further rising calcination temperature indicating some structural loss. The catalysts show a broad distribution of Brønsted acid sites (BS) ranging from weak to strong sites as indicated the thermal decomposition of exchanged ammonium ions (TPDA). The ammonium ion decomposition leaving BS, i.e., H+ located at Al–O–Si framework bridges, starts at ≥250 ◦C. A part of the Brønsted sites is lost after calcination specifically at 500 ◦C. It is related to the formation of penta-coordinated aluminium at the expense of tetrahedral framework aluminium. The Brønsted sites are partially recreated after repeated ammonium ion exchange. The catalytic performance of the acidic HCLIN catalysts was tested in the etherification of glycerol as a green renewable resource with different C1 -C4 alcohols. The catalysts are highly active in the etherification of glycerol, especially with alcohols containing the branched, tertiary alkyl groups. Highest activity is observed with the soft activated catalyst HCLIN300 (300 ◦C, temperature holding time: 1 min). A total of 78% conversion of glycerol to mono and di ether were achieved with tert-butanol at 140 ◦C after 4 h of reaction. The mono- and di-ether selectivity were 75% and 25%, respectively. Authoritative, focused reviews help to address this problem but become so granular that the overall directions of the field are lost. This broader perspective can be provided partially by general overviews but then the thinking, experimental details, theoretical underpinnings and instrumental innovations of the original work must be sacrificed. In the present compilation, this dilemma is overcome by assembling the most impactful publications in the area of analytical atomic spectrometry. Each entry was proposed by at least one current expert in the field and supported by a narrative that justifies its inclusion. The entries were then assembled into a coherent sequence and returned to contributors for a round-robin review. Such metabolic specialization could be driven by stochastic gene expression and could provide individual cells with growth benefits of specialization. We measured the degree of phenotypic specialization in two parallel metabolic pathways, the assimilation of glucose and arabinose. We grew Escherichia coli in chemostats, and used isotope-labeled sugars in combination with nanometer-scale secondary ion mass spectrometry and mathematical modeling to quantify sugar assimilation at the single-cell level. We found large variation in metabolic activities between single cells, both in absolute assimilation and in the degree to which individual cells specialize in the assimilation of different sugars. Analysis of transcriptional reporters indicated that this variation was at least partially based on cell-to-cell variation in gene expression. Metabolic differences between cells in clonal populations could potentially reduce metabolic incompatibilities between different pathways, and increase the rate at which parallel reactions can be performed. The particles are dispersible in chloroform resulting in stable suspensions. Magnetic response against an external magnetic field proved the superparamagnetic nature of the particles with a low coercivity (Bc) value of 297 µT. The combination of the advantageous superparamagnetism of the mgNP with the exceptional stability of OPDL makes these novel hybrid mgNP promising candidates as multifunctional building blocks for magnetic nanocomposites with tunable physical properties. T1 - Stowage during transport: a proposal for harmonization N2 - For stowage and retention during transport, the International Atomic EnergyAgency (IAEA) Transport regulations (SSR-6) only require that the package shall be securely stowed. Additional information is given in the IAEA guidance material (SSG-26), that includes specific load factors that should be considered in the safety demonstrations. Nevertheless, applicants commonly use other load factors to justify the design of the packaging attachment points. In particular, acceleration values vary between different countries and this may lead to difficulties during the validation of foreign approval certificates. Discussions with applicants identified that the load factors presented in the IAEA guidance material are not necessarily relevant for the different transport modes. For some modes, the load factors are representative of situations occurring in routine conditions of transport. For other modes, the load factors are more representative of situations occurring in normal or accident conditions of transport. Furthermore, the origins of the specified values are not clear. In this context, some IAEA TRANSSC members decided to review the stowage guidance. An international working group was constituted in 2013, including representatives of competent authorities, technical support organizations, and transport stakeholders. Several topics were discussed and many questions were raised during the two year review. For example, some discussions focused on Paper No.1031 2 the conditions which have to be considered for stowage design, both as relevant to the load factors used for strength and fatigue analysis, as well as the criteria which have to be considered for the package attachment points. In addition, related questions on operational aspects were also discussed. Overall, the working group concluded that new guidance material was warranted as input into SSG-26. As a result of those discussions, the international working group produced new guidance material for stowage in transport, addressing each of these topics. The proposal to modify the IAEA guidance material (SSG-26, Appendix IV), presented by France to the TRANSSC in the 2015 initiated Review cycle of the Regulations, was unanimously accepted and will be implemented in the next edition of the guidance material. T1 - Model peptides uncover the role of the beta-secretase transmembrane sequence in metal ion mediated oligomerization N2 - The β-secretase or β-site amyloid precursor protein cleaving enzyme 1 (BACE1) is the enzyme responsible for the formation of amyloid-β peptides, which have a major role in Alzheimer pathogenesis. BACE1 has a transmembrane sequence (TMS), which makes it unique among related proteases. We noticed that the BACE1 TMS contains an uncommon sulfur-rich motif. The sequence MxxxCxxxMxxxCxMxC spans the entire TMS, resembles metal ion binding motifs, and is highly conserved among homologues. We used a synthetic 31-mer model peptide comprising the TMS to study metal ion binding and oligomerization. Applying diverse biochemical and biophysical techniques, we detected dimer and trimer formation of the TMS peptide with copper ions. Replacement of the central Cys466 by Ala essentially abolished these effects. We show that the peptide undergoes a redox reaction with copper ions resulting in a disulfide bridge involving Cys466. Further, we find peptide trimerization that depends on the presence of monovalent copper ions and the sulfhydryl group of Cys466. We identified Cys466 as a key residue for metal ion chelation and to be the core of an oligomerization motif of the BACE1-TMS peptide. Our results demonstrate a novel metal ion controlled oligomerization of the BACE1 TMS, which could have an enormous therapeutic importance against Alzheimer disease. We explored the potential of recombinant human liver cytochrome P450 monooxygenases CYP1A2, CYP2C9, and CYP3A4, heterologously expressed in Escherichia coli, to convert mono- and sesquiterpenoids to human metabolites. This natural product group is a diverse class of secondary metabolites and includes several industrially and pharmaceutically interesting compounds. Incubation of cedrol with CYP3A4 resulted in a bioconversion of 74% (±8.9%) after 1 h of the unknown metabolites 2-hydroxycedrol and 4-hydroxycedrol, which have been structurally elucidated by 1H and 13C NMR and GC-MS. We conclude that recombinant human cytochrome P450 enzymes can be useful tools in a combinatorial biosynthesis strategy for the production of new natural products and for in vitro metabolization studies. It allows to realize states of extreme electronic excitation and/or very high temperature and pressure and to drive materials close to and beyond fundamental stability limits. As a consequence, structural changes and phase transitions often occur along unusual pathways and under strongly nonequilibrium conditions. Due to the inherent multiscale nature—both temporally and spatially—of these irreversible processes, their direct experimental observation requires techniques that combine high temporal resolution with the appropriate spatial resolution and the capability to obtain good quality data on a single pulse/event basis. In this respect, fourth-generation light sources, namely, short wavelength and short pulse free electron lasers (FELs), are offering new and fascinating possibilities. To address this issue, we suggest intersectoral collaboration to achieve zero plastic pollution. The outcomes of the project P-LEACH demonstrated the enormous complexity and range of potential toxic effects of plastic-associated chemicals and micro-/nanoplastics released into water from UV-weathered plastics. We initiated an intersectoral dialogue amongst scientists, manufacturers, regulators and representatives of civil society about how to alleviate the negative impacts of plastic pollution. Circular economy offers a framework for selecting non-toxic chemicals, extending product (re)use, and waste reduction, which act to alleviate pollution when applied to plastics. We suggest three measures to advance a circular economy of plastics: 1.) Increase simplicity of chemicals in virgin plastics combined with transparent information on the contents; 2.) Consider recyclability already in plastic material and product design; 3.) Foster communication through intersectoral dialogue. Major cornerstones are the provision of standardized, easy-to-use tools to characterize plastics and plastic leachates chemically and (eco)toxicologically, the enhancement of citizen awareness enabling them to make informed choices, the creation of economic incentives for manufacturers, and sector-specific regulations to provide products that safeguard environmental and human health. Bei noch steigendem Bedarf an Phosphor (P) geht man derzeit davon aus, dass die weltweiten P-Reserven noch über 200 Jahre ausreichen. Um den P-Vorrat langfristig abzusichern, sollte der in Reststoffen enthaltene Phosphor möglichst umfänglich genutzt werden. Dieser Forderung trägt das von der Bundesregierung 2012 erstmals beschlossene Deutsche Ressourceneffizienzprogramm ProgRess Rechnung. Rohphosphat wird auch seitens der Europäischen Kommission aufgrundseiner ökonomischen Bedeutung und des unsicheren Angebots als „kritischer Rohstoff“ eingestuft. Vor diesem Hintergrund hat die Bundesregierung in der Verordnung zur Neuordnung der Klärschlammverwertung vom 27. September 2017 in Deutschland eine verpflichtende technische Rückgewinnung von Phosphor für Abwasserbehandlungsanlagen ab einer Ausbaugröße von 100.000 Einwohnerwerten (12 Jahre nach Inkrafttreten der Verordnung) bzw. ab einer Ausbaugröße von über 50.000 Einwohnerwerten (15 Jahre nach Inkrafttreten der Verordnung) vorgeschrieben. Die Verordnung enthält keine Vorgaben hinsichtlich der anzuwendenden Technologie bei der Phosphorrückgewinnung. Ausgenommen von der Rückgewinnungspflicht sind Klärschlämme mit niedrigen Phosphorgehalten (weniger als 20 Gramm Phosphor je Kilogramm Klärschlamm-Trockenmasse). Auch die im Jahre 2017 verabschiedeten neuen Rechtsvorschriften für die Düngung verlangen zukünftig einen nachhaltigen und ressourceneffizienten Umgang mit Nährstoffen bei der landwirtschaftlichen Erzeugung und schränken die P-Zufuhr insbesondere auf hoch versorgten Böden ein (Düngeverordnung vom 26. Mai 2017). Dies wird zu einem mehr am Pflanzenbedarf orientierten und ggf. vermehrt überbetrieblichen Einsatz von wirtschaftseigenen Düngern führen. Um den politischen und gesetzlichen Anforderungen gerecht zu werden, müssen einerseits wirksame und kosteneffiziente technische Verfahren zum P-Recycling (weiter)entwickelt werden. Andererseits müssen die Recyclingprodukte die Anforderungen für eine direkte Verwendung in der Landwirtschaft oder als Rohstoff für die Herstellung von Düngemitteln erfüllen. The description of the content, considering the UF6 origin, i.e. natural or reprocessed, shall be clearly justified especially when the UF6 isotopic composition exceeds the limits specified in ASTM standards. Concerning the containment of the UF6, the applicant shall demonstrate in all conditions of Transport the leak-tightness of the valve and plug of the cylinders filled with enriched UF6. In this regard, when justifications are based on numerical calculations, the absence of contact between These components of the cylinder and the internal surfaces of the overpack after the regulatory drop tests shall be shown. In particular, absence of contact between the valve and any other component of the packaging shall be confirmed to respect the current IAEA regulations [3]. If complementary calculations show a contact between the plug and the internal surfaces of the overpack, additional tests are required to confirm that the strength resulting from this contact will not affect the plug leak-tightness. It can be noticed that the future revision of the IAEA regulations will include additional provision in case of contact of the plug with any other component of the packaging. In addition, the applicant shall demonstrate that the melting temperature of the valve, including the tinned joint, will not be exceeded during the regulatory fire test. Therefore, the degradation of the microstructure is characterized through ultrasonic wave measurements as well as microscopic crack analysis. In order to numerically model these processes, experiments on the penetration behavior of water into the concrete were carried out. The penetration behavior over time as well as the influence of degradation on the water transport were investigated. To predict the influence of concrete degradation on alkali diffusivity, a multi-scale continuum micromechanics model is incorporated into the numerical model, which accounts for the topology and the three-dimensional distribution of microcracks. As expected, the numerical simulation predicts larger alkali-penetration in pre-damaged concrete. Regarding the micro-crack distribution, an anisotropic distribution of micro-cracks tangential to the direction of the alkali and water flux increases their penetration depth. Die Ergebnisse der zunächst erfolgten großflächigen Durchmusterung mit Radar zeigen, dass in den Bereichen mit einer Dunkelfärbung der Betonfahrbahndecke erhöhte Laufzeiten der Radarimpulse zwischen der Oberfläche und der Rückseite der Betonfahrbahndecke auftreten, was auf eine erhöhte integrale Durchfeuchtung der Betonfahrbahndecke schließen lässt. Vertiefend durchgeführte NMR-Feuchtemessungen in der Betonrandzone zeigen eine gute Korrelation zwischen erhöhtem Feuchtegehalt und der dunkel gefärbten Fahrbahnoberfläche. Die Ergebnisse beider zerstörungsfreien Prüfmethoden lassen den Schluss zu, dass die Betonfahrbahndecke in der Nähe des Fugenkreuzes eine höhere Durchfeuchtung als im Bereich der Querscheinfuge aufweist. Die geringste Durchfeuchtung besitzt die Betonfahrbahndecke in der Plattenmitte. Die mit den zerstörungsfreien Prüfverfahren ermittelten Befunde stimmen prinzipiell gut mit den punktuell mittels Darr-Wäge-Verfahren an Bohrmehlproben gewonnenen Ergebnissen überein. N2 - Darkening of the surface within alkali-silica reaction context - Root cause analysis with non-destructive measurement methods The damage evolution in concrete pavements due to alkali-silica reaction is characterized in the initial phase by a darkening of the surface along joint regions. It is generally assumed that an increased moisture content of the concrete boundary zone is responsible for this visual appearance. Against this background, a concrete pavement slab of a representative German motorway section was systematically investigated in-situ using different non-destructive measurement methods. The results of the high-resolution, planar radar showed an increased time of flight of the radar pulse between the pavement surface and the base of the concrete slab in the darkened areas. This indicated that the overall moisture content is increased near joints in the slab. Additional NMR moisture measurements in the concrete boundary zone confirmed the correlation between increased moisture content and the dark-colored surface of the concrete pavement. The results of both non-destructive measurement methods indicated that the moisture content near the intersection of two joints was higher than the moisture content near the area around the transverse joint. The lowest moisture content was measured in the center of the concrete pavement slab. The results obtained with the two non-destructive testing methods were in overall good agreement with moisture measurements of boring dust analyzed using the gravimetric method. Für die Visualisierung und Quantifizierung der von außen zugänglichen Oberflächenanteile der Einzelkörner wurde ein Softwaretool entwickelt, erprobt und erfolgreich angewandt. Es gelang so u.a. den Nachweis zu erbringen, dass die gebrochenen Einzelkörner der Grauwacke und des Rhyoliths einen deutlich geringeren von außen zugänglichen Oberflächenanteil als die gleichartigen Gesteinskörner aus dem Kies besitzen. Aufgrund der limitierten Ortsauflösung der 3D-CT von 11 bis 16,5 μm wurde mit BET eine um drei Zehnerpotenzen höhere spezifische Oberfläche ermittelt. Zu den Porositätsuntersuchungen an den Korngemengen sei angemerkt, dass zum Teil gegenläufige Trends zwischen offener Porosität und spezifischer Oberfläche festgestellt wurden. Dieses ist beim Bezug des bei den Löseversuchen in 0,1 M KOH mit definierter NaCl-Zugabe aus der Gesteinskörnung gelösten SiO2 und Al2O3 von zentraler Bedeutung. Bei den Löseversuchen an den fraktionsspezifischen Korngemengen selbst zeigte sich, dass die stufenweise Erhöhung des NaCl-Gehalts von 0 auf 10 M.-% im Eluat eine erhöhte SiO2-Löslichkeit und eine verminderte Auslaugung von Al2O3 zur Folge hat. Auffallend war hierbei, dass die Al2O3-Löslichkeit bei GK4 ohne und mit geringer NaCl-Zugabe die mit Abstand höchsten Werte annimmt. Bei den zusätzlich durchgeführten Löseversuchen an den tomografierten Einzelkörnern in 1 M KOH-Lösung mit Zugabe von 1 M.-% NaCl zeigte sich, dass nur vereinzelt bei den untersuchten Gesteinsarten eine Korrelation zwischen dem SiO2- bzw. Al2O3-Gehalt im Eluat und der absoluten BET-Oberfläche nachweisbar ist. Dies ist vermutlich darauf zurückzuführen, dass neben der Oberfläche weitere Parameter (z.B. die mineralogische Zusammensetzung der Einzelkörner) das Löseverhalten maßgebend beeinflussen. Bei den parallel zu den Löseversuchen durchgeführten Betonversuchen nach dem ARS 04/2013 zeigte sich, dass die Alkaliempfindlichkeit der GK4 bei beiden Betonversuchen mit Alkalizufuhr unterschiedlich zu bewerten ist. So ist unabhängig von der vorgegebenen pessimalen Betonrezeptur des Fahrbahndeckenbetons ausschließlich bei der Klimawechsellagerung die GK4 als alkaliempfindlich einzustufen. Allerdings konnte der Befund der stark ausgeprägten AKR bei der GK4 mit der Dünnschliffmikroskopie nicht bestätigt werden. Analoges gilt für die Bewertung der Alkaliempfindlichkeit von GK2 mit den Betonversuchen nach der Alkalirichtlichtlinie des DAfStb. So ist auch hier die GK2 nicht im 60 °C-Betonversuch sondern ausschließlich im 40 °C-Betonversuch als alkaliempfindlich einzustufen. Dienach den verschiedenartigen Betonversuchen durchgeführten LIBS-Analysen am Vertikalschnitt der Prüfkörper zeigen, dass im Kernbereich nach dem 60 °C-Betonversuch mit 10 %iger NaCl-Lösung und nach der Klimawechsellagerung mit NaCl-Beaufschlagung annähernd gleich hohe Natrium- und Chloridgehalte vorgefunden werden. Wider Erwarten treten an den beaufschlagten Prüfflächen Abreicherungen von Natrium und Anreicherungen von Chlorid auf. Im 40°C- und 60 °C-Betonversuch wurde das Natrium bis in eine maximale Tiefe von ca. 30 mm bzw. 20 mm ausgelaugt. Die aufbauenden korrelativen Betrachtungen zwischen den Ergebnissen des 60°C Betonversuchs mit Alkalizufuhr und der Löseversuche an den fraktionsspezifischen Korngemengen in 1 M KOH-Lösung mit definierter NaCl-Zugabe sind durchaus vielversprechend. So zeigt sich beispielsweise beim Waschbeton, dass der fraktionsspezifisch gewichtete SiO2-Überschuss im Eluat nach 56 Tagen mit Zugabe von 0,5 M.-% NaCl ohne Bezug auf die BET-Oberfläche zur gleichen Einstufung der vier betrachteten Gesteinskörnungen wie bei den Dehnungen im 60 °C-Betonversuch mit 3 %-iger NaCl-Lösung führt. Außerdem korrelieren die im 60 °C-Betonversuch mit 10 %-iger NaCl-Lösung ermittelten Dehnungen mit dem gewichteten SiO2-Überschuss im Eluat bei Zugabe von 2 M.-% NaCl ohne Bezug auf die BET-Oberfläche. Ausschließlich bei den Betonversuchen nach der Alkali-Richtlinie erwies sich der Bezug des im Eluat ermittelten SiO2-Überschusses auf die BET-Oberfläche als zielführend. Aufgrund des Potenzials der Löseversuche mit NaCl-Zugabe für die Bewertung der Alkaliempfindlichkeit der Gesteinskörnung im Fahrbahndeckenbeton wird empfohlen, die vergleichenden Untersuchungen zwischen den Löseversuchen an Korngemischen der einzelnen Fraktionen und den Betonversuchen nach dem ARS 04/2013 mit einer möglichst hohen Anzahl an Gesteinskörnungen und einem präzisierten Untersuchungsumfang fortzuführen. N2 - The following four aggregates (GK) of varying grades of alkali sensitivity were examined for their microstructure-dependent solubility behavior: • GK1 (EIII-S) gravel with high quarz content • GK2 (EIII-S) greywacke (crushed stone) • GK3 (EI-S) quarz-feldspar-porphyry (crushed stone) • GK4 (EI-S) gravel with high sandstone content Pore structure analysis was carried out both on single grains using comparative 3D-CT and BET tests, and on grain samples of individual grain fractions (excluding the 16/22 mm fraction) using water absorption and BET. A software tool for visualizing and quantifying the externally accessible surfaces of single grains was developed, tested and successfully applied. This enabled us, among other things, to prove that single broken grains of greywacke and rhyolite have a much smaller externally accessible surface than grains of gravel of the same aggregate fraction. Due to the 3D-CT’s limited spatial resolution of 11 to 16.5 μm, BET was used to determine a two to three powers of ten higher specific surface. In relation to the porosity tests of the grain samples, it should be noted that in some cases, counterdirectional trends between open porosity and specific surface were determined. This is of central importance in relation to the SiO2 and Al2O3 dissolved from the aggregate in the solubility tests in 1 M KOH with defined addition of NaCl. The solubility tests on the grain samples of specific fractions showed that the gradual increase of the NaCl content in the eluate from 0 to 10 M.-% resulted in increased SiO2 solubility and reduced leaching of Al2O3. It was striking in this context that the solubility of Al2O3 in GK4 with no or little addition of NaCl gives by far the highest values. The additional solubility tests carried out on single grains, that were previously subjected to CT testing, in 1 M KOH solution with 1 M.-% added NaCl showed that in the types of aggregate examined, a correlation between the SiO2 and Al2O3 in eluate and the absolute BET surface is only occasionally demonstrable. This is probably due to the fact that solubility behavior is decisively influenced by other parameters in addition to the surface (e.g. by the mineralogical composition of individual grains). The concrete tests according to ARS 04/2013 carried out parallel to the solubility tests showed that GK4’s alkali sensitivity in both concrete tests with added alkali must be evaluated differently. For example, independent of the prescribed pessimal road surface concrete formula, GK4 can only be classified as alkali sensitive under cyclic storage. However petrographic microscopy could not confirm the findings of very marked ASR in GK4. The same applies to the evaluation of alkali sensitivity in GK2 with the concrete tests according to the German Committee for Structural Concrete (DAfStb)’s Alkali Guidelines. In this case too, GK2 should not be classified as alkali sensitive in 60 °C concrete testing, but only in 40 °C concrete testing. The LIBS analyses carried out on the vertical cross-section of the test specimens after the various concrete tests showed that almost equally high sodium and chloride levels were found in the core area after the 60 °C concrete prism test with 10 % NaCl solution and after cyclic storage with NaCl application. Contrary to expectations, the surfaces of samples after NaCl application showed a decrease in sodium and an increase in chloride. In the 40°C and 60°C concrete prism tests, sodium was depleted up to a maximum depth of approx. 30 mm and 20 mm respectively. Subsequent correlative observations between the findings of the 60°C concrete prism test with alkali application, and the solubility tests on the samples of specific grain fractions in 1 M KOH solution with defined NaCl addition, are very promising. In the case of exposed aggregate concrete, for example, the fraction-specific weighted excess SiO2 to in the eluate after 56 days with added 0.5 M.-% NaCl unrelated to the BET surface results in the same classification of the four aggregates examined as the expansions in the 60 °C concrete prism test with 3% NaCl solution. In addition, the expansions found in the 60 °C concrete prism test with 10% NaCl solution correlated with the weighted excess SiO2 in eluate with added 2 M.-% NaCl without relation to the BET surface. Only in the case of the concrete test in accordance with the Alkali Guidelines did the relationship of the excess SiO2 in the eluate to the BET surface prove to be productive. Due to the potential of the solubility test with added NaCl for the evaluation of alkali sensitivity of aggregates in road surface concrete, we recommend continuing with comparative tests between solubility tests of grain samples of the various fractions and the concrete tests in accordance with ARS O4/13 with the greatest possible number of aggregates and a precisely defined investigation scope. The goal is to determine their influence on the fluid transport processes within the material on the basis of experimental results and numerical simulations. The overall question was, to which extent the ingress of externally supplied alkalis and subsequently an alkali-silica reaction are affected by such modifications in the microstructure. Degradation in the concrete microstructure is characterized by ultrasonic wave measurements as well as by microscopic crack analysis. Furthermore, experiments on the penetration behavior of water into the investigated materials were performed. The penetration behavior into predamaged concrete microstructures was examined by the classical Karsten tube experiment, nuclear magnetic resonance method, and time domain reflectometry techniques. In order to create an appropriate model of the material's degradation on the water transport, the Darcy law was applied to describe the flow in partially saturated concrete. Material degradation is taken into account by an effective permeability that is dependent on the state of degradation. This effective permeability is obtained by the micromechanical homogenisation of the flow in an Representative Elementary Volume (REV) with distributed ellipsoidal microcracks embedded in a porous medium. The data gained in the microscopic crack analysis is used as input for the micromechanical model. Finite element simulations for unsaturated flow using the micromechanical model were compared with the experimental results showing good qualitative and quantitative agreement. Für eine empirische Herangehensweise ist die Verfügbarkeit adäquater Prüfverfahren von entscheidender Bedeutung. In Deutschland existiert aktuell jedoch kein standardisiertes oder genormtes Verfahren für die prüftechnische Bestimmung des thermischen Ausdehnungskoeffizienten von Beton. Daher wurden unter Beachtung straßenbauspezifischer Gesichtspunkte zwei Prüfansätze entwickelt, die in diesem Beitrag vorgestellt sowie hinsichtlich möglicher Messunsicherheiten und Messungenauigkeiten diskutiert werden. Außerdem werden ausgewählte Ergebnisse aus Analysen an Bestandsbetonen aus dem BAB-Netz dargestellt. Im Ergebnis sollen die Untersuchungen einen Beitrag zur Schaffung der prüftechnischen Voraussetzungen für eine abgesicherte Quantifzierung der thermischen Dehnung von Fahrbahndeckenbetonen leisten. Hydroformylation in microemulsions, which is currently under investigation at Technische Universität Berlin on a mini-plant scale, was identified as a cost efficient approach which also enhances product selectivity. Herein, we present the application of online Raman spectroscopy on the reaction of 1-dodecene to 1-tridecanal within a microemulsion. To achieve a good representation of the operation range in the mini-plant with regard to concentrations of the reactants a design of experiments was used. Based on initial Raman spectra partial least squares regression (PLSR) models were calibrated for the prediction of 1-dodecene and 1-tridecanal. Limits of predictions arise from nonlinear correlations between Raman intensity and mass fractions of compounds in the microemulsion system. Furthermore, the prediction power of PLSR models becomes limited due to unexpected by-product formation. Application of the lab-scale derived calibration spectra and PLSR models on online spectra from a mini-plant operation yielded promising estimations of 1-tridecanal and acceptable predictions of 1-dodecene mass fractions suggesting Raman spectroscopy as a suitable technique for process analytics in microemulsions. Recently, the hydroformylation in micro¬emulsions, which allows for the efficient recycling of the expensive rhodium catalyst, was found to be feasible. However, the high sensitivity of this multi-phase system with regard to changes in temperature and composition demands a continuous observation of the reaction to achieve a reliable and economic plant operation. For that purpose, we tested the potential of both online NMR and Raman spectroscopy for process control. The lab-scale experiments were supported by off-line GC-analysis as a reference method. A fiber optic coupled probe of a process Raman spectrometer was directly integrated into the reactor. 25 mixtures with varying concentrations of olefin (1-dodecene), product (n-tridecanal), water, n-dodecane, and technical surfactant (Marlipal 24/70) were prepared according to a D-optimal design. Online NMR spectroscopy was implemented by using a flow probe equipped with 1/16” PFA tubing serving as a flow cell. This was hyphenated to the reactor within a thermostated bypass to maintain process conditions in the transfer lines. Partial least squares regression (PLSR) models were established based on the initial spectra after activation of the reaction with syngas for the prediction of unknown concentrations of 1-dodecene and n-tridecanal over the course of the reaction in the lab-scale system. The obtained Raman spectra do not only contain information on the chemical composition but are further affected by the emulsion properties of the mixtures, which depend on the phase state and the type of micelles. Based on the spectral signature of both Raman and NMR spectra, it could be deduced that especially in reaction mixtures with high 1-dodecene content the formation of isomers as a competitive reaction was dominating. Similar trends were also observed during some of the process runs in the mini-plant. The multivariate calibration allowed for the estimation of reactants and products of the hydroformylation reaction in both laboratory setup and mini-plant. T1 - Application of MeCAT-Click labeling for protein abundance characterization of E. coli after heat shock experiments N2 - In a proof of concept study, metal-coded affinity tags based on click chemistry (MeCAT-Click) were used to analyze the proteome of Escherichia coli (E. coli) in response to heat stress. This allows high labeling efficiency, high detection sensitivity, and multiplex capabilities, which are pivotal for its application to protein quantification. Two approaches are presented for relative quantification of differentially lanthanide-labeled proteins. The first approach uses isotope-labeling, where ESI-MS was utilized to quantify the differentially labeled proteins from different states of E. coli. With this approach, 14 proteins were found with changed abundance, among them five proteins upregulated. In the second approach, differentially labeled samples were separated by two dimensional gel electrophoresis (2 DE) and scanned by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Comparison of the signal intensities of the different lanthanides was used to quantify different sample states. Based on this information, ESI-MS was used to identify the proteins with different abundance. The sensitivity of LA-ICP-MS allowed us to find one upregulated protein that was nearly invisible by silver staining ("Probable replication endonuclease from retron EC67"). The advantage of this approach is to locate low abundant proteins with differential expression using LA-ICP-MS, which may be overlooked otherwise. Biological significance: This paper demonstrates the successful application of a novel metal labeling strategy to quantify the proteins from complex biological samples. In comparison with former metal labeling strategies, it reduces the steric hindrance and improves the labeling efficiency during the labeling process, which ensure its successful application. This methodology is compatible with both molecular and elemental mass spectrometry. ESI-MS/MS in combination with software-based search allows the identification and relative quantification of labeled proteins. In addition, LA-ICP-MS helps to locate the labeled proteins in 2-DE gels with superior detection capability, thus, target proteins with low abundance can be precisely followed. Its excellent sensitivity allows one to track the proteins of interest that are barely visible by silver staining. T1 - Experimental Analysis of a Pressurized Vessel Exposed to Fires: an Innovative Representative Scale Apparatus N2 - A novel deign of test equipment has been commissioned to investigate thermal stratification and boiling during fire exposure of pressure vessels. Extensive temperature measurements and video of the internal conditions during fire exposure are possible, and the equipment has been designed for future compatibility with laser-based velocity measurement techniques. It is expected to generate data large quantities of data that will be of use in validation of two- and three-dimensional CFD models for the prediction of pressure vessel behaviour in fire. Future work will seek to characterize the boundary layer conditions in detail for a range of test fluids, fill levels and fire-induced thermal boundary conditions. Initial tests undertaken during commissioning may indicate that fire exposure of the vessel wall just above the liquid level can have a notable influence on the pressurization rate, by increasing the degree of superheat. Further experimental and modelling work is required to confirm and quantify this effect, or to rebut this conclusion. The measurement results achieved with these methods are based on different interactions between the film and the probe. A key question in nanotechnology is how to achieve consistent results on a level of uncertainty below one nanometre with different techniques. Two different types of thickness standards are realised. Metal film standards for X-ray techniques in the thickness range 10 to 50 nm are calibrated by GIXR with monochromatised synchrotron radiation of 8048 eV. The results obtained at four different facilities show excellent agreement. SiO2 on Si standards for SE and SFM in the thickness range 6 to 1000 nm are calibrated by GIXR with monochromatised synchrotron radiation of 1841 eV and with a metrological SFM. Consistent results within the combined uncertainties are obtained with the two methods. Surfaces and interfaces of both types of standards are additionally investigated by transmission electron microscopy (TEM). The examples were obtained in the research group FOR 384 funded by DFG (German Research Foundation). The co-operation of experimental research and modeling allowed imaging and identification of grouted and ungrouted areas of tendon ducts (including strands) in a large test specimen (40 m²). In addition to the criteria for indicating grouting faults in post-tensioned ducts known until now the phase evaluation of reflected ultrasonic pulses is described. Experiments and modeling of wave propagation are presented for reflections at metal plates in concrete (thickness range 0.5 mm to 40 mm) and for tendon ducts including strands. The main part of the progress was achieved by automated measurements using dry contact transducers, 3D-SAFT reconstruction including phase evaluation and modeling considering wave propagation for typical elastic parameters and exact experimental site conditions. The results for shear waves as well as for pressure waves are compared in the frequency range from 50 kHz to 120 kHz. Hierfür wurde die Leistungsfähigkeit der folgenden Messverfahren hinsichtlich ihrer Aussageschärfe, Anwendungsgrenzen und Messgeschwindigkeit bei praxisnahen Randbedingungen untersucht: - Impakt-Echo, manuelle Impaktauslösung - Impakt-Echo, automatisierte Impaktauslösung - Ultraschall-Echo mit multistatischem Array - Impuls-Thermografie - Radar Als Versuchskörper dienten sechs liegend hergestellte Betonplatten der Abmessungen 200/200/30 cm aus zwei verschiedenen Betongüten und mit unterschiedlichem Bewehrungsgehalt (Bewehrungsgrade 0 %, 0 ,9 %, 2 ,6 %), von denen vier Platten auf trockenem und zwei Platten auf ständig nass gehaltenem Sand gelagert waren. Alle sechs Platten enthielten jeweils dieselben künstlich eingebrachten Fehlstellen: vier quadratische Kiesnester in unterschiedlicher Tiefe mit Kantenlängen zwischen 10 und 15 cm sowie drei oberflächennahe Hohlstellen (Ø 4/6/8 cm). Zur Erfassung eines möglichen Einflusses des Betonalters wurden die Messungen 4 Tage, 28 Tage sowie 180 Tage nach Herstellung der Platten durchgeführt. Um aus den Untersuchungen einen breiten Nutzen für die Praxis zu erzielen, basierten die eingesetzten Messverfahren auf handelsüblichen Geräten, welche jedoch bezüglich ihres Messprinzips und insbesondere der Datenerfassung und -auswertung weiterentwickelt wurden. Die Fehlstellengröße wurde bewusst klein gewählt. Mit Ausnahme des manuellen Impakt-Echo-Verfahrens, mit welchem unter Verwendung der Originalsoftware des Herstellers keine Fehlstellen nachweisbar waren, zeigten sich alle Messverfahren geeignet, unter verschiedenen Randbedingungen Gefügestörungen zu lokalisieren. Insgesamt wurde jeder Fehlstellentyp von zumindest einem Verfahren bei mindestens zwei Versuchskörpern detektiert. Es konnten jedoch an keiner Betonplatte mit einem einzelnen Messverfahren alle vorhandenen Fehlstellen an allen Messterminen geortet werden. Am häufigsten wurden die beiden großen oberflächennahen Fehlstellen (Hohlstelle Ø 8 cm, Kiesnest 10 cm) erkannt. Am schwersten zu lokalisieren waren die kleine Hohlstelle mit 4 cm Durchmesser sowie die beiden auf der Unterseite gelegenen Kiesnester. Die Untersuchungen zeigen, dass für eine erfolgreiche Anwendung zerstörungsfreier Messverfahren in der Praxis die Auswahl des am besten geeigneten Verfahrens bzw. einer Kombination verschiedener Verfahren jeweils auf die vorliegende Problemstellung abgestimmt sein muss. Für Betonbodenplatten mit hohem Feuchtegehalt (junger Beton) oder starker Bewehrung ist entweder das Ultraschall- oder das Impakt-Echo-Verfahren geeignet. Das Ultraschall-Verfahren wird von hohen Bewehrungsgehalten zwar stärker beeinflusst als das Impakt-Echo-Verfahren, besitzt aber eine bessere Auflösung und ermöglicht auch im oberflächennahen Bereich eine quantitative Bestimmung der Fehlstellentiefe. Das Impakt-Echo-Verfahren kann wegen der niederfrequenten Impulsanregung Fehlstellen erst in größeren Tiefen und mit schlechterer Auflösung nachweisen, ist aber ideal geeignet zur Bestimmung der Plattendicke aufgrund seines signifikanten Rückwandechos. Sowohl das Radar-Verfahren als auch die Impuls-Thermografie können bei hohen Feuchte- und Bewehrungsgehalten nur sehr oberflächennah eingesetzt werden. Bei trockenem Beton und geringen bis mittleren Bewehrungsgehalten liefert das Radar-Verfahren sehr gute Ergebnisse. In kurzer Zeit ermöglicht es eine flächendeckende Ortung der Tiefenlage und Größe von Inhomogenitäten bei guter Auflösungsgenauigkeit. Vorhandene Bewehrung wird deutlich abgebildet, kann aber darunterliegende Fehlstellen verschatten. Alternativ kann die Impuls-Thermografie für die Ortung oberflächennaher Gefügestörungen eingesetzt werden. Das Verfahren eignet sich insbesondere bei Oberflächen, die nicht direkt zugänglich sind, da zwischen Infrarotkamera und Messobjekt nur Sichtkontakt bestehen muss. Die Betongüte hatte bei keinem eingesetzten Verfahren einen erkennbaren Einfluss auf die Messergebnisse. Streng genommen ist dies jedoch nur für den Einfluss des Wasserzementwertes, der Zementart und des Zementgehalts bei gleichbleibender Sieblinie und gleicher Kornart gültig. Bei Verwendung von Leichtzuschlag sind abweichende Ergebnisse denkbar.
