T1 - Printing metal-spiked inks for LA-ICP-MS bioimaging internal standardization: comparison of the different nephrotoxic behavior of cisplatin, carboplatin, and oxaliplatin N2 - The study of the distribution of the cytostatic drugs cisplatin, carboplatin, and oxaliplatin along the kidney may help to understand their different nephrotoxic behavior. Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) allows the acquisition of trace element images in biological tissues. However, results obtained are affected by several variations concerning the sample matrix and instrumental drifts. In this work, an internal standardization method based on printing an Ir-spiked ink onto the surface of the sample has been developed to evaluate the different distributions and accumulation levels of the aforementioned drugs along the kidney of a rat model. A conventional ink-jet printer was used to print fresh sagittal kidney tissue slices of 4 μm. A reproducible and homogenous deposition of the ink along the tissue was observed. The ink was partially absorbed on top of the tissue. Thus, this approach provides a pseudo-internal standardization, due to the fact that the ablation sample and internal standard take place subsequently and not simultaneously. A satisfactory normalization of LA-ICP-MS bioimages and therefore a reliable comparison of the kidney treated with different Pt-based drugs were achieved even for tissues analyzed on different days. Due to the complete ablation of the sample, the transport of the ablated internal standard and tissue to the inductively coupled plasma-mass spectrometry (ICP-MS) is practically taking place at the same time. Pt accumulation in the kidney was observed in accordance to the dosages administered for each drug. Although the accumulation rate of cisplatin and oxaliplatin is high in both cases, their Pt distributions differ. The strong nephrotoxicity observed for cisplatin and the absence of such side effect in the case of oxaliplatin could explain these distribution differences. The homogeneous distribution of oxaliplatin in the cortical and medullar areas could be related with its higher affinity for cellular transporters such as MATE2-k. Pathologists are archiving vast numbers of FFPE samples which can be stored for decades. Conventional immunohistochemical staining (IHC) of biomarkers on FFPE tissue sections is one of the most important analytical techniques for cancer diagnosis and pathology in general. However standardization for IHC samples and quality management is tedious and differs significantly from clinic to clinic. Combining established IHC staining strategies with modern mass spectrometry mediated methods would increase it`s potential and enable access of large FFPE archives for multiplexed quantitation purposes. In this work element mass spectrometry and a new ink-jet printed internal standardization approach was successfully combined with IHC staining to facilitate quantitative multiplex assays for archived FFPE samples. The printing strategy improves elemental image resolution and reproducibility of paraffin embedded breast cancer tissue sections in laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) using conventional IHC staining as a model system to investigate the new capabilities of this technique. For the internal standardization we applied a conventional CD-ink-jet printer to print a metal spiked ink onto the top of thin layer tissue sections with constant density. Printing was carried out in a direct comparison to an iodination of the tissue section as previously described as an alternative standardization method. The use of the printed internal standard allowed correction of the fluctuation during the laser ablation process and compensated instrumental drift effects. Mediated by the ink correction approach we achieved better signal-to-background-ratios (SBR) of 74 and better spatial resolution of 30 µm compared to iodination (SBR=23). This improved performance was demonstrated on tumorous areas in FFPE breast cancer tissue sections and allowing detection of Her-2 in tumorous areas of this tissue with significantly improved contrast. T1 - Towards defining reasonable minimum composition thresholds – Impacts of variable CO2 stream compositions on transport, injection and storage N2 - To set up recommendations on how to define “reasonable minimum composition thresholds” for CO2 streams to access CO2 pipeline networks, we investigated potential impacts of CO2 streams with different and temporally variable compositions and mass flow rates along the CCS chain. All investigations were based on a generic “CCS cluster scenario” in which CO2 streams captured from a spatial cluster of eleven emitters (seven fossil-fired power plants, two cement plants, one refinery and one steel mill) are collected in a regional pipeline network. The resulting CO2 stream (19.78 Mio t impure CO2 per year) is transported in a trunk line (onshore and offshore) and injected into five generic replicate storage structures (Buntsandstein saline aquifers) offshore. Experimental investigations and modeling of selected impacts revealed beneficial as well as adverse impacts of different impurities and their combinations. Overall, no fundamental technical obstacles for transporting, injecting and storing CO2 streams of the considered variable compositions and mass flow rates were observed. We recommend to define minimum composition thresholds for each specific CCS project through limiting i) the overall CO2 content, ii) maximum contents of relevant impurities or elements, iii) acceptable variability of concentrations of critical impurities, and defining impurity combinations to be avoided. T1 - Large-area fabrication of low- and high-spatial-frequency laser-induced periodic surface structures on carbon fibers N2 - The formation and properties of laser-induced periodic surface structures (LIPSS) were investigated on carbon fibers under irradiation of fs-laser pulses characterized by a pulse duration τ = 300 fs and a laser wavelength λ = 1025 nm. The LIPSS were fabricated in an air environment at normal incidence with different values of the laser peak fluence and number of pulses per spot. The morphology of the generated structures was characterized by using scanning electron microscopy, atomic force microscopy and Fast-Fourier transform analyses. Moreover, the material structure and the surface chemistry of the carbon fibers before and after laser irradiation was analyzed by micro Raman spectroscopy and X-ray photoelectron spectroscopy. Large areas in the cm2 range of carbon fiber arrangements were successfully processed with homogenously distributed high- and low-spatial frequency LIPSS. Beyond those distinct nanostructures, hybrid structures were realized for the very first time by a superposition of both types of LIPSS in a two-step process. The findings facilitate the fabrication of tailored LIPSS-based surface structures on carbon fibers that could be of particular interest for e.g. fiber reinforced polymers and concretes. T1 - An approach for quantification of platinum distribution in tissues by LA-ICP-MS imaging using isotope dilution analysis N2 - Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) has been revealed as a convenient technique for trace elemental imaging in tissue sections, providing elemental 2D distribution at a quantitative level. For quantification purposes, in the last years several approaches have been proposed in the literature such as the use of CRMs or matrix matched standards. The use of Isotope Dilution (ID) for quantification by LA-ICP-MS has been also described, being mainly useful for bulk analysis but not feasible for spatial measurements so far. In this work, a quantification method based on ID analysis was developed by printing isotope-enriched inks onto kidney slices from rats treated with antitumoral Pt-based drugs using a commercial ink-jet device, in order to perform an elemental quantification in different areas from bio-images. For the ID experiments ¹⁹⁴Pt enriched platinum was used. The methodology was validated by deposition of natural Pt standard droplets with a known amount of Pt onto the surface of a control tissue, where could be quantified even 50 pg of Pt, with recoveries higher than 90%. The amount of Pt present in the whole kidney slices was quantified for cisplatin, carboplatin and oxaliplatin-treated rats. The results obtained were in accordance with those previously reported. The amount of Pt distributed between the medullar and cortical areas was also quantified, observing different behavior for the three drugs. To preserve the lipid content and the heterogeneous structure of osseous tissue, 4 μm thick human bone sections were prepared via cryoembedding and tape-assisted cryosectioning, circumventing the application of organic solvents and a decalcification step. A protocol for comparative mass spectrometry imaging (MSI) on the same section was established for initial analysis with time-of-flight secondary ion mass spectrometry (TOF-SIMS) at a lateral resolution of 10 μm to <500 nm, followed by atmospheric pressure scanning microprobe matrix-assisted laser desorption/ionization (AP-SMALDI) Orbitrap MSI at a lateral resolution of 10 μm. This procedure ultimately enabled MSI of lipids, providing the lateral localization of major lipid classes such as glycero-, glycerophospho-, and sphingolipids. Bindemittel hinsichtlich des Widerstands gegenüber betonaggressiven Einwirkungen beurteilen zu können. Ein wesentliches Ziel von Prüfverfahren zur Beurteilung der Dauerhaftigkeit von Baustoffen ist, die im Laufe der Nutzungsdauer zu erwartenden Einwirkungen auf den Baustoff realitätsnah abzubilden. Dazu sind die Prüfparameter auf die im jeweiligen Anwendungsbereich vorherrschenden Umgebungs- und Randbedingungen anzupassen. Daneben sind auch die Bewertungskriterien individuell unter Berücksichtigung der im Einzelfall geforderten Nutzungsdauer eines Bauwerks festzulegen. Eine systematische Zuordnung von adäquaten Prüfverfahren bzw. Prüfparametem und Bewertungskriterien für die verschiedenen Anwendungsbereiche im Betonbau, in denen betonaggressive Einwirkungen auftreten, ist mit Ausnahme von Einzelfällen bislang nicht vorhanden. Daneben stellt sich in der Praxis immer wieder das Problem, das Angriffspotenzial der Umgebung eines Bauwerks (prüftechnisch) zu erfassen und zu bewerten. Der Hauptausschuss Baustofftechnik des DBV hat daher den Arbeitskreis „Chemischer Angriff auf Beton“ beauftragt, dieses Merkblatt zu erarbeiten, das auf Basis von einschlägigen Erfahrungen der Baupraxis und wissenschaftlichen Erkenntnissen Grundsätze und bewährte Methoden zur Beurteilung der Einwirkungs- und Widerstandsseite beim chemischen Angriff auf Beton aufzeigt und diesbezüglich Hinweise für verschiedene Anwendungsbereiche im Betonbau liefert. These procedures are based on printing of metal spiked inks onto the top of thin layer samples for simultaneous internal standardization and calibration of LA-ICP-MS. In the case of internal standardization the ink is spiked with indium as an internal standard and homogenously printed over the entire membrane (size 56 cm2) prior to LA-ICP-MS detection, a standard deviation (RSD) value of 2% was achieved. In the second approach the metal content of lanthanide tagged proteins and antibodies after biological work flows was quantified by LA-ICP-MS on nitro-cellulose membranes. In this case the inks spiked with varying metals were printed with different densities on the same nitrocellulose membranes in well-defined squares to produce matrix-matched calibration standards. For validation and calibration the ink squares were excised and the specific metal content was measured by liquid ICP-MS after solubilization of the membrane slice. For the printed calibration standard limits of detection (LOD) of <4 fmol for different metals and relative process standard deviations of 1–2% only were determined via LA-ICP-MS. T1 - Application of MeCAT-Click labeling for protein abundance characterization of E. coli after heat shock experiments N2 - In a proof of concept study, metal-coded affinity tags based on click chemistry (MeCAT-Click) were used to analyze the proteome of Escherichia coli (E. coli) in response to heat stress. This allows high labeling efficiency, high detection sensitivity, and multiplex capabilities, which are pivotal for its application to protein quantification. Two approaches are presented for relative quantification of differentially lanthanide-labeled proteins. The first approach uses isotope-labeling, where ESI-MS was utilized to quantify the differentially labeled proteins from different states of E. coli. With this approach, 14 proteins were found with changed abundance, among them five proteins upregulated. In the second approach, differentially labeled samples were separated by two dimensional gel electrophoresis (2 DE) and scanned by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Comparison of the signal intensities of the different lanthanides was used to quantify different sample states. Based on this information, ESI-MS was used to identify the proteins with different abundance. The sensitivity of LA-ICP-MS allowed us to find one upregulated protein that was nearly invisible by silver staining ("Probable replication endonuclease from retron EC67"). The advantage of this approach is to locate low abundant proteins with differential expression using LA-ICP-MS, which may be overlooked otherwise. Biological significance: This paper demonstrates the successful application of a novel metal labeling strategy to quantify the proteins from complex biological samples. In comparison with former metal labeling strategies, it reduces the steric hindrance and improves the labeling efficiency during the labeling process, which ensure its successful application. This methodology is compatible with both molecular and elemental mass spectrometry. ESI-MS/MS in combination with software-based search allows the identification and relative quantification of labeled proteins. In addition, LA-ICP-MS helps to locate the labeled proteins in 2-DE gels with superior detection capability, thus, target proteins with low abundance can be precisely followed. Thus it can measure the gas concentration in the immediate vicinity of the object which causes the emission. A further stage of extension is to enhance the system for plume tracking and identification of sources of hazardous gases. The overall objective of the research is to reduce the costs of offshore wind energy deployment in deep water. In order to provide all participating research projects with detailed data, the test site will be equipped with extensive measurement instrumentation. This research initiative was named RAVE – Research at Alpha VEntus – and consists of a variety of projects in connection with the installation and operation of alpha ventus. T1 - A validated robust and automatic procedure for vibration analysis of bridge structures using MEMS accelerometers N2 - Today, short- and long-term structural health monitoring (SHM) of bridge infrastructures and their safe, reliable and cost-effective maintenance has received considerable attention. From a surveying or civil engineer’s point of view, vibration-based SHM can be conducted by inspecting the changes in the global dynamic behaviour of a structure, such as natural frequencies (i. e. eigenfrequencies), mode shapes (i. e. eigenforms) and modal damping, which are known as modal parameters. This research work aims to propose a robust and automatic vibration analysis procedure that is so-called robust time domain modal parameter identification (RT-MPI) technique. It is novel in the sense of automatic and reliable identification of initial eigenfrequencies even closely spaced ones as well as robustly and accurately estimating the modal parameters of a bridge structure using low numbers of cost-effective micro-electro-mechanical systems (MEMS) accelerometers. To estimate amplitude, frequency, phase shift and damping ratio coefficients, an observation model consisting of: (1) a damped harmonic oscillation model, (2) an autoregressive model of coloured measurement noise and (3) a stochastic model in the form of the heavy-tailed family of scaled t-distributions is employed and jointly adjusted by means of a generalised expectation maximisation algorithm. Multiple MEMS as part of a geo-sensor network were mounted at different positions of a bridge structure which is precalculated by means of a finite element model (FEM) analysis. At the end, the estimated eigenfrequencies and eigenforms are compared and validated by the estimated parameters obtained from acceleration measurements of high-end accelerometers of type PCB ICP quartz, velocity measurements from a geophone and the FEM analysis. Additionally, the estimated eigenfrequencies and modal damping are compared with a well-known covariance driven stochastic subspace identification approach, which reveals the superiority of our proposed approach. We performed an experiment in two case studies with simulated data and real applications of a footbridge structure and a synthetic bridge. The results show that MEMS accelerometers are suitable for detecting all occurring eigenfrequencies depending on a sampling frequency specified. Moreover, the vibration analysis procedure demonstrates that amplitudes can be estimated in submillimetre range accuracy, frequencies with an accuracy better than 0.1 Hz and damping ratio coefficients with an accuracy better than 0.1 and 0.2 % for modal and system damping, respectively. Ziel des Machine Learning Modells ist das Identifizieren von Bindefehlern in den aufgebauten Volumina mit prozessbegleitend gemessenen Schweißstromquellensignalen als Input. Es werden durch Algorithmen Merkmalsvariablen in der Vorverarbeitung der Daten extrahiert und die Korrelation zwischen den Merkmalsvariablen und den Bindefehlern analysiert. Durch den vorgestellten Algorithmus werden diese automatisiert als Input an ein DNN übergeben. Diese Arbeit untersucht die Genauigkeit der Klassifizierung von verschiedenen DNN-Architekturen mit jeweils 4 verdeckten Schichten. Als Trainings- und Testsatz werden randomisiert extrahierte Merkmale von defektfreien und fehlerhaften Proben verwendet, wobei Bindefehler zum Teil absichtlich provoziert werden. Within CLUSTER, we investigated potential impacts of CO2 streams with different and temporally variable compositions and mass flow rates along the whole CCS chain. Investigations included, amongst others, impacts on: Corrosion of pipeline steel, pipeline network design and related transport costs, alteration of well bore cements, pressure development and rock integrity, geochemical reactions, and petrophysical and geomechanical rock properties. All investigations are based on a generic CCS chain scenario. In this scenario, CO2 streams are captured from a spatial cluster of eleven emitters and collected in a regional pipeline network. Emitters comprise seven fossil fuel-fired power plants equipped with different capture technologies, two cement plants, one refinery and one integrated iron and steel plant. In total, 19.78 Mio t CO2 (including impurities) are captured in the emitter cluster annually. The combined CO2 stream is transported in a trunk line with a length of 400 km (100 km of these offshore) and is injected into five generic storage structures. The storage reservoirs are saline aquifers of the Buntsandstein. The investigations revealed beneficial and deteriorating impacts of different impurities and combinations thereof. Overall, no fundamental technical obstacles for transporting, injecting and storing CO2 streams of the modelled variable compositions and mass flow rates were observed. Based on the results, the CLUSTER project team recommends not to define “minimum composition thresholds” for CO2 streams as strict threshold values for each individual impurity in the stream. Instead, CO2 stream compositions and variabilities for specific CCS projects should be constrained with regard to a set of parameters including i) the overall CO2 content, ii) maximum contents of relevant impurities or elements, iii) acceptable variability of CO2 stream composition, and iv) impurity combinations to be avoided. This challenge is fundamentally tied to the intricate microstructural defects that emerge during the processing. Systematic microstructure-based modelling can offer solutions to address this bottleneck. In this work, we utilize a crystal plasticity model, developed for gamma''-strengthened Ni-base alloys and calibrated with single crystal tensile and creep tests of Inconel 718. By systematically refining the representative volume element complexity---from equiaxed to elongated grain morphologies, and from unimodal to bimodal orientation and grain size distributions---we demonstrate how each layer of structural realism contributes to the model’s predictive capacity. Creep tests of laser-based powder bed fusion (PBF-LB/M) manufactured samples in three orientations (with the tensile axis parallel, perpendicular, and 45° tilted relative to the building direction) were performed at 650° C, accompanied by electron backscatter secondary diffraction measurements. The results of our simulations reveal that the bimodal grain morphology and crystallographical texture significantly influence the observed creep anisotropy. We show that the elongated grain structure combined with grain boundary sliding plays a major role in the creep response, specifically in tilted specimens. T1 - Unravelling the formation histories of placer gold and platinum-group mineral particles from Corrego Bom Successo, Brazil: A window into noble metal cycling N2 - Gold and platinum-group-metals (PGM) are cycled through Earth's environments by interwoven geological, physical, chemical and biological processes leading to the trans/neoformation of metallic particles in placers. The placer deposit at Corrego Bom Successo (CBS, Brazil) is one of the few localities worldwide containing secondary gold- and PGM-particles. Placer gold consists of detrital particles from nearby hydrothermal deposits that were transformed in the surface environment. Processes that have affected these particles include shortdistance transport, chemical de-alloying of the primary Gold silver, and (bio)geochemical dissolution/reprecipitation of Gold leading to the formation of pure, secondary gold and the Dispersion of gold nanoparticles. The latter processes are likely mediated by non-living organic matter (OM) and bacterial biofilms residing on the particles. The biofilms are largely composed of metallophillic β- and γ-Proteobacteria. Abundant mobile gold and platinum nanoparticles were detected in surface waters, suggesting similar mobilities of these metals. Earlier hydrothermal processes have led to the formation of coarsely-crystalline, arborescent dendritic potarite (PdHg). On potarite surfaces, biogeochemical processes have then led to the formation of platinum- and palladium-rich micro-crystalline layers, which make up the botryoidal platinum palladium aggregates. Subsequently potarite was dissolved from the core of many aggregates leaving voids now often filled by secondary anatase (TiO2) containing biophilic elements. The presence of fungal structures associated with the anatase suggests that fungi may have contributed to ist formation. For the first time a primary magmatic PGM-particle comprising a mono-crystalline platinum palladium-alloy with platinum iridium osmium inclusions was described from this locality, finally defining a possible primary source for the PGM mineralisation. In conclusion, the formation of modern-day placer gold- and PGM-particles at CBS began 100s ofmillions of years ago bymagmatic and hydrothermal processes. These provided the metal sources for more recent biogeochemical cycling of PGEs and gold that led to the trans/neoformation of gold- and PGM-particles. T1 - Studies towards the development of a PET radiotracer for imaging of the P2Y1 receptors in the brain: synthesis, 18F-labeling and preliminary biological evaluation N2 - Purine nucleotides such as ATP and ADP are important extracellular signaling molecules in almost all tissues activating various subtypes of purinoreceptors. In the brain, the P2Y1 receptor (P2Y1R) subtype mediates trophic functions like differentiation and proliferation, and modulates fast synaptic transmission, both suggested to be affected in diseases of the central nervous system. Research on P2Y1R is limited because suitable brain-penetrating P2Y1R-selective tracers are not yet available. Here, we describe the first efforts to develop an 18F-labeled PET tracer based on the structure of the highly affine and selective, non-nucleotidic P2Y1R allosteric modulator 1-(2-[2-(tert-butyl)phenoxy]pyridin-3-yl)-3- [4-(trifluoromethoxy)phenyl]urea (7). A small series of fluorinated compounds was developed by systematic modification of the p-(trifluoromethoxy)phenyl, the urea and the 2-pyridyl subunits of the lead compound 7. Additionally, the p-(trifluoromethoxy)phenyl subunit was substituted by carborane, a boron-rich cluster with potential applicability in boron neutron capture therapy (BNCT). By functional assays, the new fluorinated derivative 1-{2-[2-(tert-butyl)phenoxy]pyridin-3-yl}-3-[4-(2-fluoroethyl) phenyl]urea (18) was identified with a high P2Y1R antagonistic potency (IC50 ¼10 nM). Compound [18F] 18 was radiosynthesized by using tetra-n-butyl ammonium [18F]fluoride with high radiochemical purity, radiochemical yield and molar activities. Investigation of brain homogenates using hydrophilic interaction chromatography (HILIC) revealed [18F]fluoride as major radiometabolite. Although [18F]18 showed fast in vivo metabolization, the high potency and unique allosteric binding mode makes this class of compounds interesting for further optimizations and investigation of the theranostic potential as PET tracer and BNCT agent. It has been found that, in this system, the distributions of γ-stabilizing elements (Cr, Co, W, and Re) are described by the Boltzmann solution for diffusion between two semiinfinite plates of a binary alloy. The processing of these distributions has shown that the diffusion coefficients of Cr, Co, W, and Re in the multicomponent system are close to those in binary alloys of these elements with Ni. The diffusion redistribution of the elements leads to the dissolution of the γ′ phase in the nickel-base superalloy, growth of nickel grains toward the superalloy constituent of the diffusion pair, and the formation of porosity on both sides of the migrating interface, which is determined from a crystal misorientation of the alloy single crystal and nickel grains. Wenn auch positive Befunde bei dem Nachweis von Bestandteilen aus Pferd in Lebensmitteln wie Lasagne derzeit so gut wie nicht mehr anzutreffen sind, so hat die Tierartendifferenzierung insgesamt beim Nachweis von Verfälschungen in Lebensmitteln einen hohen Stellenwert. Diese Arbeit fasst daher den aktuellen Stand der Analytik in Deutschland mit Schwerpunkt bei der Standardisierung zusammen.
