T1 - Depletion of Cardiac Cardiolipin Synthase Alters Systolic and Diastolic Function JF - iScience N2 - Cardiolipin (CL) is a major cardiac mitochondrial phospholipid maintaining regular mitochondrial morphology and function in cardiomyocytes. Cardiac CL production includes ist biosynthesis and a CL-remodeling process. Here we studied the impact of CL-biosynthesis and the enzyme Cardiolipin Synthase (CLS) on cardiac function. CLS and cardiac CL-species were significantly downregulated in cardiomyocytes following catecholamine-induced cardiac damage in mice, accompanied by increased oxygen consumption rates, signs of oxidative stress and mitochondrial uncoupling. RNAi-mediated cardiomyocyte-specific knockdown of CLS in Drosophila melanogaster resulted in marked cardiac dilatation, severe impairment of systolic performance and slower diastolic filling velocity assessed by fluorescence-based heart imaging. Finally, we showed that CL72:8 is significantly decreased in cardiac samples from patients with heart failure with reduced ejection fraction (HFrEF). In summary, we identified CLS as a regulator of cardiac function. In addition, the application of non-destructive testing is indispensable, if information about the inner structure - such as the location of rebars and tendon ducts or the damage-related condition assessment to detect grouting defects, honeycombs, delamination, or corrosion - is required. Besides the selection of a suitable NDT method and an appropriate inspection system, the reliability of the results depends largely on the person who applies the non-destructive inspection technique and evaluates the inspection results. To ensure a high quality of non-destructive concrete evaluation as well as to keep the uncertainty caused by the inspection personnel to a minimum, structured, consistent, and regulated theoretical as well as practical training of inspection personnel is essential. To close this gap, the subcommittee of education and training (UA-A) within the committee for NDT-CE of the German Society for Non-Destructive Testing (DGZfP) has been reactivated in 2018 to establish uniform training standards for non-destructive concrete inspections in the long term. The subcommittee consists of scientists, practitioners, authorities, and clients. So far, the national standard DIN 4871 “Non-destructive testing - Qualification of NDT personnel in Civil Engineering (NDT-CE)” was developed. This standard considers the civil-industry-specifics, for example, that standards for NDT of concrete, as well as related product standards with a few exceptions, still do not exist at the moment. The method requires the gravimetric preparation of several blends (bi)—each consisting of roughly the same masses (mx,i) of the sample solution (x) and my,i of a spike solution (y) plus different masses (mz,i) of a reference solution (z). Only these masses and the isotope ratios (Rb,i) in the blends and reference and spike solutions have to be measured. The derivation of the underlying equations based on linear regression is presented and compared to a related concept reported by Pagliano and Meija. The uncertainties achievable, e.g., in the case of the Si blank in extremely pure TMAH of urel (w(Si)) = 90% (linear regression method, this work) and urel (w(Si)) = 150% (the method reported by Pagliano and Meija) seem to suggest better applicability of the new method in practical use due to the higher robustness of regression analysis. This information can be relevant or irrelevant, true or biased, precise or imprecise. The incorporation of relevant and quality-assessed measured information into reliability reassessment offers the chance to extend remaining lifetimes and support decision making about optimal actions or maintenance strategies. This paper shows recent developments in a national research project that aims to produce a guideline on the NDT-based, structure-specific modification of partial safety factors. The general methodology, results from recalculations according to the Eurocodes and metrologically solvable testing tasks relevant in the recalculation of the concrete bridges are shown and compared with the non-destructive testing methods applicable to concrete bridges. A case study is used to demonstrate that as-built drawings, in this case of the positions of tendons and shear reinforcement, can be verified using the radar method. Die Aufnahmekonfiguration kann bei Objekten dieser Art häufig nicht so gewählt werden, dass die unterschiedlichen Materialeigenschaften mit ausreichender Qualität abgebildet werden können. Aus diesem Grund ist eine objektive Bewertung der Oberflächenmessung mit Hilfe eines in dieser Untersuchung eingeführten Qualitätsmaßes erforderlich. Anhand der in dieser Arbeit vorgestellten Prüfkörperserie soll der Einfluss der Materialzusammensetzung auf das dimensionelle Messen mit CT erfasst und charakterisiert werden. Unter Berücksichtigung der Qualitätsbewertung der Oberflächenpunkte erfolgt die Berechnung der Form- und Abstandsabweichungen. Ziel ist es, die Genauigkeit der Oberflächenmessung hinsichtlich unterschiedlicher Aufnahmeparameter und Materialzusammensetzungen darzustellen. Anhand dieser Ergebnisse ist es möglich, den computertomographischen Aufnahme- und Auswertungsprozess quantitativ zu bewerten und Ursachen der Abweichungen zu diskutieren. T1 - Microbial hotspots in lithic microhabitats inferred from DNA fractionation and metagenomics in the Atacama Desert JF - Microorganisms N2 - The existence of microbial activity hotspots in temperate regions of Earth is driven by soil heterogeneities, especially the temporal and spatial availability of nutrients. Here we investigate whether microbial activity hotspots also exist in lithic microhabitats in one of the most arid regions of the world, the Atacama Desert in Chile. While previous studies evaluated the total DNA fraction to elucidate the microbial communities, we here for the first time use a DNA separation approach on lithic microhabitats, together with metagenomics and other analysis methods (i.e., ATP, PLFA, and metabolite analysis) to specifically gain insights on the living and potentially active microbial community. Our results show that hypolith colonized rocks are microbial hotspots in the desert environment. In contrast, our data do not support such a conclusion for gypsum crust and salt rock environments, because only limited microbial activity could be observed. The hypolith community is dominated by phototrophs, mostly Cyanobacteria and Chloroflexi, at both study sites. The gypsum crusts are dominated by methylotrophs and heterotrophic phototrophs, mostly Chloroflexi, and the salt rocks (halite nodules) by phototrophic and halotolerant endoliths, mostly Cyanobacteria and Archaea. The major environmental constraints in the organic-poor arid and hyperarid Atacama Desert are water availability and UV irradiation, allowing phototrophs and other extremophiles to play a key role in desert ecology. Different morphology of the films prepared by thermal vacuum evaporation and drop-casting from solutions has been observed depending on the surface used. It has been found that gold surface has a unique property to influence self-assembly of both monolayer and larger aggregates or crystals of the compounds, in contrast to glass and PTFE substrates. It was found that thiocarbonyl group is able to be chemisorbed to the gold surface. However, it was concluded that in spite of the fact that the thiocarbonyl groups are important for the chemical interaction with the gold surface, physical adsorption on the substrate surface, compound–compound and compound–solvent (when using solution) interactions are of great significance to drive self-assembly of the final film. Brückenkolloquium - Beurteilung, Ertüchtigung und Instandsetzung von Brücken N2 - Die drei Beispiele zerstörungsfreier Prüfungen an Spannbetonbrücken zeigen unterschiedliche Problemstellungen und deren Lösung mit unterschiedlichen Ansätzen. Allen drei Beispielen gemeinsam ist die Verwendung kommerzieller Geräte. Diese werden von verschiedenen Anbietern bereitgestellt. Eine weitere Gemeinsamkeit ist, dass die Grundgeräte durch zusätzliche Maßnahmen ergänzt. Erst dann ist der gewünschte Untersuchungserfolg sicher gestellt. Im ersten Beispiel wurde die vom Auftraggeber geforderte Präzision dadurch erreicht, dass systematische Voruntersuchungen stattfanden. Die Übertragung der Lokalisierung direkt auf die Betonoberfläche stellte sicher, dass keinen Informationsverluste an die nachfolgenden Aufgaben auftraten. Das zweite Beispiel zeigt den Nutzen von Automatisierung und Kombination von Verfahren. Damit können dem Auftraggeber die notwendigen Angaben über Einbauteile mit großer Detaitreue und leicht verständlicher Visualisierung aufbereitet werden. Der abschließende Bericht zeigt, wie vorhandene Auswertungen sinnvoll durch noch in der Forschung befindliche Ansätze bereichert werden. Insgesamt ist die zerstörungsfreie Prüfung in der Lage, an Brücken preisrelevante Fragestellungen zu lösen. Voraussetzung hierfür ist eine sorgfältig geplante Vorgehensweise und ein sicheres Verständnis der Grundlage der eingesetzten Geräte und Auswertungsschritte. T2 - 1. To overcome this problem, polyolefin surfaces were modified thermally, plasmachemically, or by flame treatment with different oxygen-containing groups, however, unfortunately, such treatments were accompanied by undesired, adhesion lowering polymer degradation. To solve this dilemma, solutions of synthetic polymers and copolymers were prepared, sprayed into the barrier discharge or electrosprayed without discharge and deposited as thin adhesion-promoting layers. The deposited polymer layers from poly(vinylamine), poly(ethylene glycol)-poly(vinyl alcohol) copolymers and poly(acrylic acid) were endowed with monotype functional groups. Using the aerosol - dielectric barrier discharge only a fraction of functional groups survived the deposition process in contrast to the electrospray in which all functional groups were retained. The molecular rods have oligospiroketal (OSK) and oligospirothioketal (OSTK) backbones. Contrary to the expectations, cyclic oligomers were always obtained instead of linear rigid-rod polymers. Replacing the OS(T)K rods with a flexible chain yielded polymeric products, suggesting that the OS(T)K structure is responsible for the formation of cyclic products. The reason for the preferred formation of cyclic products is due to the presence of folded conformations, which have already been described for articulated rods. T1 - Assessment of the hepatic tumor extracellular matrix using elastin‑specific molecular magnetic resonance imaging in an experimental rabbit cancer model JF - Scientific Reports N2 - To investigate the imaging performance of an elastin-specific molecular magnetic resonance imaging (MRI) probe with respect to the extracellular matrix (ECM) in an experimental hepatic cancer model. Twelve rabbits with hepatic VX2 tumors were examined using 3 T MRI 14, 21, and 28 days after tumor implantation for two subsequent days (gadobutrol, day 1; elastin-specific probe, day 2). The relative enhancement (RE) of segmented tumor regions (central and margin) and the peritumoral matrix was calculated using pre-contrast and delayed-phase T1w sequences. MRI measurements were correlated to histopathology and element-specific and spatially resolved mass spectrometry (MS). Mixed-model analysis was performed to assess the performance of the elastin-specific probe. In comparison to gadobutrol, the elastin probe showed significantly stronger RE, which was pronounced in the tumor margin (day 14–28: P ≤ 0.007). In addition, the elastin probe was superior in discriminating between tumor regions (χ2(4) = 65.87; P < 0.001). MRI-based measurements of the elastin probe significantly correlated with the ex vivo elastinstain (R = .84; P <0 .001) and absolute gadolinium concentrations (ICP-MS: R = .73, P <0 .01). LA-ICP-MS imaging confirmed the colocalization of the elastin-specific probe with elastic fibers. Elastin-specific molecular MRI is superior to non-specific gadolinium-based contrast agents in imaging the ECM of hepatic tumors and the peritumoral tissue.
