Kühlschmierstoffe auf der Basis von Fettsäureestern bieten technologische Vorteile gegenüber mineralölbasierten Produkten. Außerdem sind sie umweltgerechter und leisten einen Beitrag zum nachhaltigen Wirtschaften. Dennoch werden sie in der Praxis kaum eingesetzt, da sie bisher aufgrund von Rohstoffpreisen und Synthesekosten teurer sind, als konventionelle Konkurrenzprodukte. Alternativen hinsichtlich Rohstoffen und Synthesewegen werden betrachtet und die Eigenschaftsbilder resultierender Esterprodukte miteinander verglichen. Methoden. In die Untersuchungen wurden drei auf enzymatischkatalytischem Weg hergestellte Fettsäure-2-Ethylhexylester auf der Basis von Erdnussfett, Rindertalg und Altspeisefett sowie vier konventionell synthetisierte Ester aus Tier- bzw. Pflanzenfetten und 2-Ethyl-1-Hexanol einbezogen. Bestimmt wurden physikalische Eigenschaftswerte, Fettsäurespektren, Gehalte an freien Fettsäuren sowie Oxidations- und Hydrolysestabilitäten. Ferner wurden Thermolyseexperimente durchgeführt. Ergebnisse. Die physikalischen Eigenschaften der sieben Ester, d.h. Dichte, Viskosität, Pourpoint und Flammpunkt, waren vergleichbar und ließen die Anwendbarkeit der Produkte als Basisöle für Kühlschmierstoffe erwarten. Hydrolysebeständigkeiten waren hoch und thermische Belastungen der Ester gaben keine Hinweise auf besondere Schadstoffbildungspotentiale. Defizite wiesen insbesondere die enzymatisch hergestellten Ester hinsichtlich Oxidationsstabilitäten und den Gehalten an freien Fettsäuren auf. Ausblick. Für die Praxis wird es notwendig sein, Mindestqualitäten für Rohstoffe und resultierende Esteröle zu definieren, die als Grundöle im Kühlschmierstoffbereich eingesetzt werden sollen. Außerdem müssen Verfahren ausgearbeitet werden, die es ermöglichen, zu qualitativ hochwertigen Esterprodukten zu gelangen, ohne den Preisvorteil von Rohstoffen wie Altspeise- und Tierfetten durch hohe Produktions- und Veredelungskosten zu verlieren. Ein Beitrag hierzu ist sicherlich die Weiterentwicklung der wenig energieaufwändigen enzymatischen Alkoholyse zur Esterproduktion. In terms of drug testing, a number of devices have been introduced into the market in recent decades, notably for workplace inspection or roadside testing. However, these systems do not always fulfill the requirements in terms of reliability, especially when low cut‐off levels are required. With respect to surface water, the presence of anthropogenic small organic molecules such as prescription and over‐the‐counter pharmaceuticals as well as illicit drugs like cannabinoids, heroin, or cocaine, has become a challenge for scientists to develop new analytical tools for screening and on‐site analysis because many of them serve as markers for anthropogenic input and consumer behavior. Here, a modular approach for the detection of cocaine is presented, integrating an electrochemical enzyme‐linked immunosorbent assay (ELISA) performed on antibody‐grafted magnetic beads in a hybrid microfluidic sensor utilizing flexible tubing, static chip and screen‐printed electrode (SPE) elements for incubation, recognition, and cyclic voltammetry measurements. A linear response of the sensor vs. the logarithm of cocaine concentration was obtained with a limit of detection of 0.15 ng/L. Within an overall assay time of 25 minutes, concentrations down to 1 ng/L could be reliably determined in water, oral fluids, and urine, the system possessing a dynamic working range up to 1 mg/L. A sensitive direct competitive immunoassay was developed and optimized with a limit of detection (LOD) of 5.6 ng/L. Exhaustive cocaine extraction by solvent was tested using different methanol concentrations and buffered solutions. Crossreactivity studies were performed to determine the degree of interference of cocaine metabolites with the immunoassay. Sixty-five Euro banknotes obtained from different districts in Berlin were evaluated. A 100% contamination frequency with cocaine was detected. A comparison between the amount of cocaine extracted by cotton swabbing of one square centimeter of the banknote showed a good correlation for lower contamination levels. This assay showed high sensitivity of detecting pg of cocaine per 1 cm2 of one banknote by swabbing 1 cm2: 0, 14, and 21 pg/cm2. Moreover, three notes of different denominations revealed high cocaine concentration; 1.1 mg/note, and twice 55 μg/note. T1 - Degradation of carbamazepine in environmentally relevant concentrations in water by hydrodynamic-acoustic-cavitation (HAC) N2 - The antiepileptic drug carbamazepine is one of the most abundant pharmaceuticals in the German aquatic environment. The effect of low carbamazepine concentrations (1–50 µg L-1) is discussed controversially, but ecotoxicological studies revealed reproduction toxicity, decreased enzymatic activity and bioaccumulation in different test organisms. Therefore, as a preventive step, an efficient and cost-effective technique for wastewater treatment plants is needed to stop the entry of pharmaceuticals into the aquatic environment. Cavitation, the formation, growth, and subsequent collapse of gas- or vapor-filled bubbles in fluids, was applied to solve this problem. The technique of Hydrodynamic-Acoustic-Cavitation was used showing high synergistic effect. Under optimized conditions carbamazepine (5 µg L-1) was transformed by pseudo-first order kinetics to an extent of >96% within 15 min (27% by hydrodynamic cavitation, 33% by acoustic cavitation). A synergistic effect of 63% based on the sum of the single methods was calculated. Carbamazepine concentrations were monitored by a sensitive and selective immunoassay and after 60 min no known metabolites were detectable by LC-MS/MS. T1 - Thermische Regenerierung von Zeolithpulver Typ X aus der VOC-Luftreinigung N2 - Für die Entfernung von VOC (volatile organic compounds) aus der Luft von Fertigungsstätten werden vielfach mikroporöse Adsorbentien eingesetzt. Die gereinigte Luft wird rezyklisiert. Für die mit VOC beladenen Adsorbentien wird ein Regenerierungsweg für den mehrfachen Gebrauch gesucht. Bei der Regenerierung sollen die VOC möglichst vollständig desorbiert werden und mit einer geringen Gasmenge in einer katalytischen Verbrennung zu CO2 und H2O umgewandelt werden. Die Wirtschaftlichkeit der Luftreinigung wird wesentlich bestimmt durch die Sorptionskapazität des Adsorbens, die erforderliche Energie für die thermische Regenerierung und die Menge des Spülgases bei der Regenerierung. In dieser Arbeit wird der Einsatz von Typ X-Zeolithpulver in der Luftreinigung einer Lackiererei beschrieben und dessen Regenerierung im Labor mittels DTA/TG und Isothermenmessung der Schlüsselkomponenten untersucht. Ferner wurde die Regenerierung des beladenen Zeoliths in einem Labor-Drehrohrofen unter Variation der Betriebsparameter untersucht und anschließend mit ausgewählten Parametern in einem kleintechnischen Drehrohrofen durchgeführt. Die Verfahrensvarianten werden diskutiert. The food industry often runs own lab procedures, for example, for mycotoxin analysis, but it is a major goal to simplify analysis, linking analytical methods with smart technologies. Enzyme-linked immunosorbent assays, with photometric detection of 3,3’,5,5’-tetramethylbenzidine (TMB),form a good basis for sensitive detection. To provide a straightforward approach for the miniaturization of the detectionstep, we have studied the pitfalls of the electrochemical TMB detection. By cyclic voltammetry it was found that the TMB electrochemistry is strongly dependent on the pH and the electrode material. A stable electrode response to TMB could be achieved at pH 1 on gold electrodes. We created a smartphonebased, electrochemical, immunomagnetic assay for the detection of ochratoxin A in real samples, providing a solid basis forsensing of further analytes. In contrast to classical microtiter plate-based assays, microparticles are a next generation solid support, as they promote automation of immunoassays using flow-based techniques. Antibody immobilization is a crucial step, as these reagentsmare expensive, and inefficient coupling can result in low sensitivities. This paper proposes a general procedure for efficient immobilization of antibodies onto TentaGel particles, via Nhydroxysuccinimide chemistry. The goal was the preparation of solid supports with optimum immunorecognition, while increasing the sustainability of the process. The influence of buffer composition, activation and coupling time, as well as the amount of antibody on the immobilization efficiency was investigated, resorting to fluorophore-labeled proteins and fluorescence imaging. Buffer pH and activation time are the most important parameters for efficient coupling. It is demonstrated, that the hydrolysis of N-hydroxysuccinimide esters occurs at similar rates as in solution, limiting the utilizable time for coupling. Finally, applicability of the generated material for automated affinity extraction is demonstrated on the mesofluidic platform lab-on-valve.
