T1 - Bioinformatics meets virology: The European virus bioinformatics center's second annual meeting N2 - The Second Annual Meeting of the European Virus Bioinformatics Center (EVBC), held in Utrecht, Netherlands, focused on computational approaches in virology, with topics including (but not limited to) virus discovery, diagnostics, (meta-)genomics, modeling, epidemiology, molecular structure, evolution, and viral ecology. The goals of the Second Annual Meeting were threefold: (i) to bring together virologists and bioinformaticians from across the academic, industrial, professional, and training sectors to share best practice; (ii) to provide a meaningful and interactive scientific environment to promote discussion and collaboration between students, postdoctoral fellows, and both new and established investigators; (iii) to inspire and suggest new research directions and questions. Approximately 120 researchers from around the world attended the Second Annual Meeting of the EVBC this year, including 15 renowned international speakers. This report presents an overview of new developments and novel research findings that emerged during the meeting. T1 - Unity in defence: honeybee workers exhibit conserved molecular responses to diverse pathogens N2 - Background: Organisms typically face infection by diverse pathogens, and hosts are thought to have developed specific responses to each type of pathogen they encounter. The advent of transcriptomics now makes it possible to test this hypothesis and compare host gene expression responses to multiple pathogens at a genome-wide scale. Here, we performed a meta-analysis of multiple published and new transcriptomes using a newly developed bioinformatics approach that filters genes based on their expression profile across datasets. Thereby, we identified common and unique molecular responses of a model host species, the honey bee (Apis mellifera), to its major pathogens and parasites: the Microsporidia Nosema apis and Nosema ceranae, RNA viruses, and the ectoparasitic mite Varroa destructor, which transmits viruses. Results: We identified a common suite of genes and conserved molecular pathways that respond to all investigated pathogens, a result that suggests a commonality in response mechanisms to diverse pathogens. We found that genes differentially expressed after infection exhibit a higher evolutionary rate than non-differentially expressed genes. Using our new bioinformatics approach, we unveiled additional pathogen-specific responses of honey bees; we found that apoptosis appeared to be an important response following microsporidian infection, while genes from the immune signalling pathways, Toll and Imd, were differentially expressed after Varroa/virus infection. Finally, we applied our bioinformatics approach and generated a gene co-expression network to identify highly connected (hub) genes that may represent important mediators and regulators of anti-pathogen responses. Conclusions: Our meta-analysis generated a comprehensive overview of the host metabolic and other biological processes that mediate interactions between insects and their pathogens. We identified key host genes and pathways that respond to phylogenetically diverse pathogens, representing an important source for future functional studies as well as offering new routes to identify or generate pathogen resilient honey bee stocks. The statistical and bioinformatics approaches that were developed for this study are broadly applicable to synthesize information across transcriptomic datasets. These approaches will likely have utility in addressing a variety of biological questions. A1 - McMahon, Dino Peter T1 - Eating in a losing cause: Limited benefit of modifed macronutrient consumption following infection in the oriental cockroach Blatta orientalis N2 - Background: Host–pathogen interactions can lead to dramatic changes in host feeding behaviour. One aspect of this includes self-medication, where infected individuals consume substances such as toxins or alter their macronutrient consumption to enhance immune competence. Another widely adopted animal response to infection is illness-induced anorexia, which is thought to assist host immunity directly or by limiting the nutritional resources available to pathogens. Here, we recorded macronutrient preferences of the global pest cockroach, Blatta orientalis to investigate how shifts in host macronutrient dietary preference and quantity of carbohydrate (C) and protein (P) interact with immunity following bacterial infection. Results: We fnd that B. orientalis avoids diets enriched for P under normal conditions, and that high P diets reduce cockroach survival in the long term. However, following bacterial challenge, cockroaches signifcantly reduced their overall nutrient intake, particularly of carbohydrates, and increased the relative ratio of protein (P:C) consumed. Surprisingly, these behavioural shifts had a limited efect on cockroach immunity and survival, with minor changes to immune protein abundance and antimicrobial activity between individuals placed on diferent diets, regardless of infection status. Conclusions: We show that cockroach feeding behaviour can be modulated by a pathogen, resulting in an illness-induced anorexia-like feeding response and a shift from a C-enriched to a more P:C equal diet. However, our results also indicate that such responses do not provide signifcant immune protection in B. orientalis, suggesting that the host’s dietary shift might also result from random rather than directed behaviour. The lack of an apparent beneft of the shift in feeding behaviour highlights a possible reduced importance of diet in immune regulation in these invasive animals, although further investigations employing pathogens with alternative infection strategies are warranted. T1 - Honeybee intestines retain low yeast titers, but no bacterial mutualists, at emergence N2 - Honeybee symbionts, predominantly bacteria, play important roles in honeybee health, nutrition, and pathogen protection, thereby supporting colony health. On the other hand, fungi are often considered indicators of poor bee health, and honeybee microbiome studies generally exclude fungi and yeasts. We hypothesized that yeasts may be an important aspect of early honeybee biology, and if yeasts provide a mutual benefit to their hosts, then honeybees could provide a refuge during metamorphosis to ensure the presence of yeasts at emergence. We surveyed for yeast and fungi during pupal development and metamorphosis in worker bees using fungal-specific quantitative polymerase chain reaction (qPCR), next-generation sequencing, and standard microbiological culturing. On the basis of yeast presence in three distinct apiaries and multiple developmental stages, we conclude that yeasts can survive through metamorphosis and in naïve worker bees, albeit at relatively low levels. In comparison, known bacterial mutualists, like Gilliamella and Snodgrassella, were generally not found in pre-eclosed adult bees. Whether yeasts are actively retained as an important part of the bee microbiota or are passively propagating in the colony remains unknown. Our demonstration of the constancy of yeasts throughout development provides a framework to further understand the honeybee microbiota. T1 - Under pressure: offering fundamental insight into structural changes on ball milling battery materials N2 - Synthesis of Li ion battery materials via ball milling has been a huge area of growth, leading to new high-capacity electrode materials, such as a number of promising disordered rocksalt (DRS) phases. In prior work, it was generally assumed that the synthesis was facilitated simply by local heating effects during the milling process. In this work, we show that ball milling Li2MoO4 leads to a phase transformation to the high pressure spinel polymorph and we report electrochemical data for this phase. This observation of the formation of a high pressure polymorph shows that local heating effects alone cannot explain the phase transformation observed (phenakite to spinel) and so indicates the importance of other effects. In particular, we propose that when the milling balls collide with the material, the resulting shockwaves exert a localised pressure effect, in addition to local heating. To provide further support for this, we additionally report ball milling results for a number of case studies (Li2MnO3, Li2SnO3, Nb2O5) which reinforces the conclusion that local heating alone cannot explain the phase transformations observed. The work presented thus provides greater fundamental understanding of milling as a synthetic pathway and suggests potential strategies to prepare such samples without milling (e.g., doping to create internal chemical pressure). In addition, we suggest that further research is needed into the effect of the use of milling as a route to smaller particles, since we believe that such milling may also be affecting the surface structure of the particles through the influence of the shockwaves generated. Biodegradable, alternating copolyesters of terephthalic acid, aliphatic dicarboxylic acids, and alkane diols N2 - Copolyesters with an alternating sequence of terephthalic acid and aliphatic dicarboxylic acids were prepared with three different methods. First, dicarboxylic acid dichlorides were reacted with bis(2-hydroxyethyl)terephthalate (BHET) in refluxing 1,2-dichlorobenzene. Second, the same monomers were polycondensed at 0-20 °C in the presence of pyridine. Third, dicarboxylic acid dichlorides and silylated BHET were polycondensed in bulk. Only this third method gave satisfactory molecular weights. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry revealed that the copolyesters prepared by the pyridine and silyl methods might have contained considerable fractions of cyclic oligoesters and polyesters despite the absence of transesterification and backbiting processes. The alternating sequences and thermal properties were characterized with 1H NMR spectroscopy and differential scanning calorimetry measurements, respectively. In agreement with the alternating sequence, all copolyesters proved to be crystalline, but the crystallization was extremely slow [slower than that of poly(ethylene terephthalate)]. A second series of alternating copolyesters was prepared by the polycondensation of silylated bis(4-hydroxybut- yl)terephthalate with various aliphatic dicarboxylic acid dichlorides. The resulting copolyesters showed significantly higher rates of crystallization, and the melting temperatures were higher than those of the BHET-based copolyesters. © 2001 John Wiley & Sons, Inc. The combination of cyclic load and corrosive aqueous environment leads to corrosion fatigue of pipes and components (e.g. pumps) and thus inevitably to the reduction of the lifetime of these components. To estimate the reliability of components from adjusted in-situ-laboratory experiments a corrosion chamber was designed and tested with CO2 saturated corrosive aqueous media flowing at a steady rate. Unique feature of this special chamber is its installation directly onto the sample and thus providing flexible usability in almost every testing machine. This allows simultaneous mechanical loading of the sample, operation at temperatures up to 100 °C and exposure to fluid flow of corrosive liquids and gases. The lifetime reduction of AISI 420C (X46Cr13, 1.4034) is demonstrated at T=60 °C, geothermal brine: Stuttgart Aquifer flow rate: 9 l/h, CO2. S-N plots, micrographic-, phase-, fractographic- and surface analysis were applied to obtain sustainable information on the corrosion fatigue behavior. The lateral and vertical dimensions of laser ablated craters as well as the laser damage thresholds were evaluated for different pulse numbers applied to the same spot. The joint observation using optical microscopy, atomic force microscopy and scanning electron microscopy revealed the surface morphology of the ablated craters and also showed that the ablation threshold depends significantly on the number of laser pulses applied to the same spot due to incubation effects. The incubation effects change the absorption processes involved in fs-laser ablation of the transparent material from multiphoton absorption to a single-photon absorption. These results are discussed on the basis of recent models of the interaction of fs-laser pulses with dielectrics. However, this method has its limits under standard conditions, particularly in detecting compounds beyond a chain length of around C40. Here, we compare the CHC chain length range detectable by GC-MS with the range assessed by silver-assisted laser desorption/ionization mass spectrometry (Ag-LDI-MS), a novel and rarely applied technique on insect CHCs, in seven species of the order Blattodea. For all tested species, we unveiled a considerable range of very long-chain CHCs up to C58, which are not detectable by standard GC-MS technology. This indicates that general studies on insect CHCs May frequently miss compounds in this range, and we encourage future studies to implement analytical techniques extending the conventionally accessed chain length range. Furthermore, we incorporate 3D scanned insect body surface areas as an additional factor for the comparative quantification of extracted CHC amounts between our study species. CHC quantity distributions differed considerably when adjusted for body surface areas as opposed to directly assessing extracted CHC amounts, suggesting that a more accurate evaluation of relative CHC quantities can be achieved by taking body surface areas into account. Unlike the well-studied immune systems of cells and individuals, little is known about the origins of immunity during the transition to eusociality, a major evolutionary transition comparable to the evolution of multicellular organisms from single-celled ancestors. We aimed to tackle this by characterizing the immune gene repertoire of 18 cockroach and termite species, spanning the spectrum of solitary, subsocial and eusocial lifestyles. We find that key transitions in termite sociality are correlated with immune gene family contractions. In cross-species comparisons of immune gene expression, we find evidence for a caste-specific social defence system in termites, which appears to operate at the expense of individual immune protection. Our study indicates that a major transition in organismal complexity may have entailed a fundamental reshaping of the immune system optimized for group over individual defence. A1 - Hothan, Sascha T1 - European test standard intumescent coatings applied to steel tension bars with solid section N2 - Intumescent coatings are used in civil engineering to improve the fire resistance of steel constructions. Due to the thin coating thickness and the profile-following application, the architectural appearance of the steel structure can be preserved. EN 13381-8 provides regulations to determine the contribution to the fire resistance of intumescent coatings applied to steel beams and columns. The scope of this standard excludes tension members and steel members with solid sections. For products that have already been successfully tested and assessed on beams and columns, EN 13381-10 offers the possibility based on unloaded fire tests to extend the scope of application of intumescent coatings to tension bars with solid sections. This approach contradicts the national safety level in Germany, where mechanically loaded testing is mandatory. Therefore, a new part of the test standard series EN 13381 is currently developed to enable the application of intumescent coatings on steel tension bars with solid section based on mechanically loaded fire tests. BAM has already carried out numerous fire tests on loaded steel tension members with intumescent coatings. Currently, in the BAM research project FIRESTEMIC, the influence of the steel bar orientation and the profile type on the thermal performance of intumescent coatings are investigated. Both questions concerning the thermal protection ability of intumescent coatings were analysed based on three different test sets, which were carried out in the tension furnace (Fig. 1a). The fire exposure corresponds to the standard temperature-time curve according to EN 1363-1. This paper summarises the main findings from the fire tests conducted in the FIRESTEMIC project. The results serve as an experimental background for the proposal of the new standard. Regarding the bar orientation, a new test setup was developed and proposed for the new standard. The two tested commercial water-based intumescent coatings with applied dry film thickness from 1.5 mm to 2.5 mm showed only a slight dependence on the bar orientation. In terms of the steel profile type, circular and rectangular solid sections with identical section factor and applied dry film thickness were tested. It was observed that the circular solid bars with diameter 30 mm heat up faster compared to the solid rectangular bars with dimension 30×30 mm (Fig. 1b). Also at larger steel bars, i.e. diameter 40 mm and dimension 40×40 mm, the same trend occurred. Thus, it is recommended for the new test standard to allow a transfer of the test results from circular to rectangular solid sections. T1 - Elevated virulence of an emerging viral genotype as a driver of honeybee loss N2 - Emerging infectious diseases (EIDs) have contributed significantly to the current biodiversity crisis, leading to widespread epidemics and population loss. Owing to genetic variation in pathogen virulence, a complete understanding of species decline requires the accurate identification and characterization of EIDs. We explore this issue in the Western honeybee, where increasing mortality of populations in the Northern Hemisphere has caused major concern. Specifically, we investigate the importance of genetic identity of the main suspect in mortality, deformed wing virus (DWV), in driving honeybee loss. Using laboratory experiments and a systematic field survey, we demonstrate that an emerging DWV genotype (DWV-B) is more virulent than the established DWV genotype (DWV-A) and is widespread in the landscape. Furthermore, we show in a simple model that colonies infected with DWV-B collapse sooner than colonies infected with DWV-A. We also identify potential for rapid DWV evolution by revealing extensive genome-wide recombination in vivo. The emergence of DWV-B in naive honeybee populations, including via recombination with DWV-A, could be of significant ecological and economic importance. Our findings emphasize that knowledge of pathogen genetic identity and diversity is critical to understanding drivers of species decline. In order to gain knowledge upon the corrosion fatigue strength of materials a corrosion chamber for "in situ" conditions was designed and successfully applied.Two different steels X46Cr13 and X5CrNiCuNb16-4 have been tested and their corrosion fatigue behavior was compared. To simulate the frequency of operating pumps (30 – 40 Hz) a resonant testing machine was used. In addition technical CO2 was introduced into the closed system at a rate close to 9 L/h to keep stable environmental conditions. The samples have a surface roughness of Rz = 4 to simulate technical machined surfaces. The calculated tensile strength of X46Cr13 with soft annealed microstructure (coagulated cementite in ferrite-perlite matrix) is about 680 MPa and the yield strength is about 345 MPa. The tensile strength of X5CrNiCuNb16-4 is about 1078 MPa and the yield strength about 928 MPa. Testing parameters are: corrosion media: saline aquifer water (Stuttgart Aquifer) temperature of the brine at 60 °C and load ratio of R=-1. For X46Cr13 a stress amplitude between 160 MPa to 270 MPa and for X5CrNiCuNb16-4 a stress amplitude between 150 to 500 was chosen. This leads to corrosion fatigue and reduces the lifetime of certain components. Usually underwater power pumps are placed inside the borehole. In this case an installation depth of 600 to 800 meters below ground level is typical. In these depths an additional load due to high pressure and high temperature is introduced which is based on geothermal brine. Therefore the material which is used for the pump is steel.For fatigue testing at loads existing in deep geological layers an appropriate system for fatigue tests under in-situ conditions was set up. The requirements of the design are: temperature of the corrosion medium of 200 °C pressure of 200 bars and the complete immersion of the specimen in flowing corrosion medium containing mixtures of corrosive gasses during the entire test period.The test system has three main components the corrosion chamber the heating module and the reservoir. It is important that the corrosion chamber is only fixed onto the specimen which allows for highest flexibility of the test system operational in many different test machines. The heating module heats the corrosion medium up to 200 °C. The reservoir make sure that the volume of the corrosion medium satisfies requirements of the DIN 50905 Part 1 is conformed providing minimum required corrosive medium of 10 ml/cm² in relation to the sample surface.This innovative fatigue test system aims at performing in- situ fatigue corrosion tests simultaneously at high pressure and high temperature in liquid and gaseous corrosive environment. Results from the tests will allow determining the fatigue life of several materials influenced by pressure and temperature under corrosive conditions. A perspective on insect strategies to avoid metamorphosis N2 - Insects with complete metamorphosis (holometaboly) are extremely successful, constituting over 60% of all described animal species. Complete metamorphosis confers significant advantages because it enables organisms to optimise life-history components through temporal partitioning, and thereby to exploitmultiple ecological niches. Yet holometaboly can also impose costs, and several lineages have evolved life cycle modifications to avoid complete metamorphosis. In this review, we discuss different strategies that have evolved that result in the loss of complete metamorphosis (type I and type II paedomorphosis). In addition, the ecological pressures and developmental modifications that facilitate this avoidance are considered, as well as the importance of life cycle complexity in life-history evolution. Interestingly, only female holometabolous insects have entirely avoided complete metamorphosis, and it is always the ancestrally juvenile morphology that is retained. These findings point to a strong sex-biased trade-off between investment in reproduction and development. While the loss of complete metamorphosis in females has occurred independently on several occasions across holometabolous insects, only a small number of species possessing this ability have been described. Thus, complete metamorphosis, which originated only once in insects, appears to have been almost fully retained. This indicates that significant modifications to the holometabolan metamorphic ground plan are highly constrained, and suggests that the transition to complete metamorphosis is evolutionarily irreversible. In order to serve the needs in antibiotics development we selected untapped bioresources and implemented high throughput approaches suitable for the discovery of strains producing antibiotics with anti-Gram negative activity. Our approaches rely on the hypothesis that Termite associated bacteria are likely to produce potent antibiotics to defend their hosts against entomopathogenic microorganisms. Termite nests and guts harbor suitable, highly diverse microbiomes in which bacterial taxa are present known to potentially produce natural compounds. In a first step the diversity of Coptotermes species nest microbiomes was assessed carefully by using 16S rDNA amplicon sequencing on the Illumina MiSeq platform and nest material was selected to retrieve viable cells by using Nycodenz density gradient centrifugation. In order to analyze the diversity of the culturable termite nest microbiome, bacterial cells were either distributed in 384-well plates (approach 1) or encapsulated in small spheric agarose beads by an high throughput microfluidics technique (approach 2). Cultures obtained from approach 1 were scaled-up in 96-well Duetz-systems for characterization of diversity and for rapid supernatant screening using the bioluminescence-labeled E. coli pFU166. The generated droplets of approach 2 simultaneously received a small population of GFP-tagged Gram negative screening cells and were sorted for low fluorescence using FACS. After elimination of redundancy we performed a fast scale-up of active strains. Implementation of this pipeline allows us to prioritize antibiotics producing strains in a ultra-high throughput fashion and by cultivation of broad diversity in our approches. However, there is little evidence of dust-derived P-containing minerals in soils. Here we studied P forms along a well-described climatic Gradient on Hawaii, which is also a dust deposition gradient. Soil mineralogy and soil P forms from six sites along the climatic gradient were analyzed with bulk (X-ray diffraction and P K-edge X-ray absorption near edge structure) and microscale (X-ray fluorescence, P K-edge X-ray absorption near edge structure, and Raman) analysis methods. In the wettest soils, apatite grains ranging from 5 to 30 μm in size were co-located at the micro-scale with quartz, a known continental dust indicator suggesting recent atmospheric deposition. In addition to co-location with quartz, further evidence of dust-derived P included backward trajectory modeling indicating that dust particles could be brought to Hawaii from the major global dust-loading areas in central Asia and northern Africa. Although it is not certain whether the individual observed apatite grains were derived from long-distance transport of dust, or from local dust sources such as volcanic ash or windblown fertilizer, these observations offer direct evidence that P-containing minerals have reached surface layers of highly-weathered grassland soils through atmospheric deposition. T1 - High-Throughput Cultivation for the Selective Isolation of Acidobacteria From Termite Nests N2 - Microbial communities in the immediate environment of socialized invertebrates can help to suppress pathogens, in part by synthesizing bioactive natural products. Here we characterized the core microbiomes of three termite species (genus Coptotermes) and their nest material to gain more insight into the diversity of termite-associated bacteria. Sampling a healthy termite colony over time implicated a consolidated and highly stable microbiome, pointing toward the fact that beneficial bacterial phyla play a major role in termite fitness. In contrast, there was a significant shift in the composition of the core microbiome in one nest during a fungal infection, affecting the abundance of wellcharacterized Streptomyces species (phylum Actinobacteria) as well as less-studied bacterial phyla such as Acidobacteria. High-throughput cultivation in microplates was implemented to isolate and identify these less-studied bacterial phylogenetic group. Amplicon sequencing confirmed that our method maintained the bacterial diversity of the environmental samples, enabling the isolation of novel Acidobacteriaceae and expanding the list of cultivated species to include two strains that may define new species within the genera Terracidiphilus and Acidobacterium. T1 - Evolution of Termite Symbiosis Informed by Transcriptome-Based Phylogenies N2 - Termitidae comprises 80% of all termite species that play dominant decomposer roles in Tropical cosystems. Two major events during Termite evolution were the loss of cellulolytic gut protozoans in the ancestor of Termitidae and the subsequent gain in the termitid subfamily Macrotermitinae of fungal symbionts cultivated externally in ‘‘combs’’ constructed within the nest. How these symbiotic transitions occurred remains unresolved. Phylogenetic analyses of mitochondrial data previously suggested that Macrotermitinae is the earliest branching termitid lineage, followed soon after by Sphaerotermitinae, which cultivates bacterial symbionts on combs inside its nests. This has led to the hypothesis that comb building was an important evolutionary step in the loss of gut protozoa in ancestral termitids. We sequenced genomes and transcriptomes of 55 termite species and reconstructed phylogenetic trees from up to 4,065 orthologous genes of 68 species. We found strong support for a novel sister-group relationship between the bacterial comb-building Sphaerotermitinae and fungus comb-building Macrotermitinae. This key finding indicates that comb building is a derived trait within Termitidae and that the creation of a comb-like ‘‘external rumen’’ involving bacteria or fungi may not have driven the loss of protozoa from ancestral termitids, as previously hypothesized. Instead, associations with gut prokaryotic symbionts, combined with dietary shifts from wood to other plant-based substrates, may have played a more important role in this symbiotic transition. Our phylogenetic tree provides a platform for future studies of comparative termite evolution and the evolution of symbiosis in this taxon. Hierzu wurden einerseits bereits vorhandene Erfahrungen und Erkenntnisse genutzt und andererseits mittels Brandversuchen neue Prüfdaten zur Beantwortung bisher offengebliebener Aspekte zum Einfluss der Bauteilorientierung und Profilart erzeugt und ausgewertet. Dadurch wird die prüftechnische Grundlage für eine sichere, zulassungsbasierte Anwendung von reaktiven Brandschutzsystemen auf zugbeanspruchten Stahlbauteilen gelegt und ein einheitliches Sicherheitsniveau etabliert. Der im Rahmen des Forschungsprojektes entwickelte Normentwurf wurde mit den Fachexperten der zuständigen nationalen und europäischen Normungsgremien diskutiert und abgestimmt. Das erarbeitete Prüf- und Auswertungsverfahren basiert auf der Durchführung von Brandversuchen an mechanisch belasteten Stahlzuggliedern mit RBS. Ergänzend sind unbelastete Brandversuche zur Beurteilung des Einflusses der Bauteilorientierung sowie der Eignung bei Schwelbrandbeanspruchung möglich. Die im Projekt durchgeführten experimentellen Untersuchungen liefern die Grundlage für den im Normenentwurf vorgeschlagenen Prüfaufbau der mechanisch belasteten und unbelasteten Brandversuche. Ferner konnte durch die Untersuchungen gezeigt werden, dass sich unter bestimmten Voraussetzungen die Prüfergebnisse von Stahlzuggliedern mit Kreisvollprofil ohne zusätzliche Brandversuche auf entsprechende Zugglieder mit Rechteckvollprofil übertragen lassen. Darüber hinaus konnte belegt werden, dass die Leistungsfähigkeit eines RBS aufgrund unterschiedlicher Bauteilorientierungen abnehmen kann und daher zwingend beim Anwendungsbereich des RBS zu berücksichtigen ist. Hierzu wird ein Temperaturdifferenz-kriterium vorgeschlagen, bei dessen Einhaltung die Wirksamkeit des RBS bei von der horizontalen Referenzorientierung abweichenden Bauteilneigungen noch gleichwertig gegeben ist. Andernfalls bietet der Normenentwurf die Möglichkeit die reduzierte thermische Schutzwirkung des RBS im Assessment zu berücksichtigen. Die Ergebnisse aus dem Forschungsvorhaben wurden bereits auf einem wissenschaftlichen Symposium vorgestellt sowie in einer referierten Fachzeitschrift publiziert. Ferner wurden die Informationen zum Forschungsprojekt und dem Normenentwurf im Rahmen eines Anwenderworkshops interessierten Kreisen zugänglich gemacht. Collaborative trial in suspect and non-target screening of organic micropollutants using LC- and GC-HRMS N2 - A collaborative trial involving 16 participants from nine European countries was conducted within the NORMAN network in efforts to harmonise suspect and non-target screening of environmental contaminants in whole fish samples of bream (Abramis brama). Participants were provided with freeze-dried, homogenised fish samples from a contaminated and a reference site, extracts (spiked and non-spiked) and reference sample preparation protocols for liquid chromatography (LC) and gas chromatography (GC) coupled to high resolution mass spectrometry (HRMS). Participants extracted fish samples using their in-house sample preparation method and/or the protocol provided. Participants correctly identified 9-69% of spiked compounds using LC-HRMS and 20-60% of spiked compounds using GC-HRMS. From the contaminated site, suspect screening with participants’ own suspect lists led to putative identification of on average ~145 and ~20 unique features per participant using LC-HRMS and GC-HRMS, respectively, while non-target screening identified on average ~42 and ~56 unique features per participant using LC-HRMS and GC-HRMS, respectively. Within the same sub-group of sample preparation method, only a few features were identified by at least two participants in suspect screening (16 features using LC-HRMS, 0 features using GC-HRMS) and non-target screening (0 features using LC-HRMS, 2 features using GC-HRMS). The compounds identified had log octanol/water partition coefficient (KOW) values ranging from -9.9 to 16 and mass-to-charge ratio (m/z) of 68 to 761 (LC-HRMS and GC-HRMS). A significant linear trend was found between log KOW and m/z for the GC-HRMS data. Overall, these findings indicate that differences in screening results are mainly due to the data analysis workflows used by different participants. Further work is needed to harmonise the results obtained when applying suspect and non-target screening approaches to environmental biota samples.
