Neben dem Gründungskonzept der XL-Monopiles rückt wegen der Kombination aus dem vergleichsweise geringen Materialverbrauch bei gleichzeitig hoher Steifigkeit, auch die Jacketgründung in den Fokus. Der Fertigungsaufwand der Jackets ist verglichen mit Monopiles groß; kann jedoch durch die Nutzung von Standardrohren mit automatisiert gefertigten Jacketknoten reduziert werden. Vor diesem Hintergrund beschreibt der vorliegende Forschungsbericht den Fertigungs-prozess zum automatisierten Schweißen von X-förmigen Hohlprofilknoten für Jacket-Gründungsstrukturen von Offshore-Windenergieanlagen. Ein weiteres wesentliches Ziel dieses Vorhabens war die konsequente Digitalisierung der schweißtechnischen Fertigungskette während der automatisierten Schweißung der X-Knoten. Die so ge-speicherten Prozessparameter können zum einen als Grundlage einer Qualitätskon-trolle dienen, zum anderen ermöglichen sie eine vollständige Nachverfolgbarkeit der automatisiert gefertigten Rohrknoten. Der zweite maßgebend in diesem Bericht behandelte Aspekt umfasst die Quantifizie-rung der Einflüsse aus Fertigungsautomatisierung und Innenschweißen auf die Ermü-dungsfestigkeit der Hohlprofilknoten auf Basis serieller Ermüdungsversuche. Dazu wurden im Projekt zwei Serien mit jeweils 16 Schwingversuchen an ein- und doppel-seitig automatisiert geschweißten X-Knoten durchgeführt und entsprechende Struktur-spannungswöhlerlinien für die automatisiert gefertigten X-Knoten bestimmt. Die Ergebnisse dieser Ermüdungsversuche werden abschließend in numerischen Untersuchungen auf Basis der Strukturspannungs- und des Kerbspannungskonzeptes gegenübergestellt, um die Prognosefähigkeit beider Konzepte für die ermüdungs-kritischen Bereiche des X-Knotens bewerten zu können. Durch die im Forschungsvorhaben durchgeführten schweißtechnischen, experimen-tellen und numerischen Analysen konnte das Potential automatisiert gefertigter Rohr-knoten hinsichtlich Reproduzierbarkeit und Ermüdungsverhalten quantifiziert werden. The results of these fatigue tests are then compared to numerical investigations based on the structural stress as well as notch stress approach. The outcomes are then compared to evaluate the capability of both approaches for the prediction of the fatigue-critical locations within the X-joints. Based on the analysis of the welding procedure as well as the experimental and numerical investigations within this research project, the potential of automatically manufactured tubular joints with regard to reproducibility and fatigue behaviour was investigated. Hence, this paper addresses the welding process chain of automated manufactured tubular joints including digitization of the relevant manufacturing parameters such as laser scanning of the weld seam geometry. Additionally, a methodology for the computation of the notch radius as well as the weld seam angle is presented based on the scanned profiles of three analysis points of an automated manufactured tubular X-joint. Subsequently, these parameters are considered within the notch stress approach based fatigue design and their impact is quantified by a comparison with the structural stress approach using equivalent stress concentration factors. An efficient strategy to minimize efforts in producing nanoscale RMs (nanoRMs) for establishing, validating, and standardizing methods for characterizing nanomaterials are multimethod nanoRMs. Ideal candidates are lanthanide-based, multicolor luminescent, and chemically inert nanoparticles (NPs) like upconversion nanoparticles (UCNPs), which can be prepared in different sizes, shapes, and chemical composition with various surface coatings. This makes UCNPs interesting candidates as standards not only for sizing methods, but also for element-analytical methods like laser ablation-inductively coupled plasma mass spectrometry (LA-ICP-MS), quantitative bioimaging methods like X-ray fluorescence computed tomography (XFCT), and luminescence methods and correlative measurements. Here, we explore the potential of two monodisperse LiYF4:Yb,Tm bipyramids with peak-to-peak distances of (43 ± 2) nm and (29 ± 2) nm as size standards for small-angle X-ray scattering (SAXS) and tools for establishing and validating the sophisticated simulations required for the analysis of SAXS data derived from dispersions of nonspheric nano-objects. These SAXS studies are supplemented by two-dimensional (2D)-transmission electron microscopy measurements of the UCNP bipyramids. Additionally, the particle number concentration of cyclohexane dispersions of these UCNP bipyramids is determined by absolute SAXS measurements, complemented by gravimetry, thermogravimetric analysis (TGA), and inductively coupled plasma optical emission spectrometry (ICP-OES). A1 - Bald, Ilko T1 - Vacuum-UV and low-energy electron induced DNA strand breaks - Influence of the DNA sequence and substrate N2 - DNA is effectively damaged by radiation, which can on the one hand lead to cancer and is on the other hand directly exploited in the treatment of tumor tissue. DNA strand breaks are already induced by photons having an energy below the ionization energy of DNA. At high photon energies, most of the DNA strand breaks are induced by low-energy secondary electrons. In the present study we quantified photon and electron induced DNA strand breaks in four different 12mer oligonucleotides. They are irradiated directly with 8.44 eV vacuum ultraviolet (VUV) photons and 8.8 eV low energy electrons (LEE). By using Si instead of VUV transparent CaF2 as a substrate the VUV exposure leads to an additional release of LEEs, which have a maximum energy of 3.6 eV and can significantly enhance strand break cross sections. Atomic force microscopy is used to visualize strand breaks on DNA origami platforms and to determine absolute values for the strand break cross sections. Upon irradiation with 8.44 eV photons all the investigated sequences show very similar strand break cross sections in the range of 1.7 - 2.3 x 10-16 cm2. The strand break cross sections for LEE irradiation at 8.8 eV are one to two orders of magnitude larger than the ones for VUV photons, and a slight sequence dependence is observed. The sequence dependence is even more pronounced for LEEs with energies < 3.6 eV. The present results help to assess DNA damage by photons and electrons close to the ionization threshold. To increase the DNA damage in carcinogenic cells and preserve healthy tissue at the same time, radiosensitizing molecules such as halogenated nucleobase analogs can be incorporated into the DNA during the cell reproduction cycle. In the present study 8.44 eV photon irradiation induced single strand breaks (SSB) in DNA sequences modified with the radiosensitizer 5-bromouracil (5BrU) and 8-bromoadenine (8BrA) are investigated. 5BrU was incorporated in the 13mer oligonucleotide flanked by different nucleobases. It was demonstrated that the highest SSB cross sections were reached, when cytosine and thymine were adjacent to 5BrU, whereas guanine as a neighboring nucleobase decreases the activity of 5BrU indicating that competing reaction mechanisms are active. This was further investigated with respect to the distance of guanine to 5BrU separated by an increasing number of adenine nucleotides. It was observed that the SSB cross sections were decreasing with an increasing number of adenine spacers between guanine and 5BrU until the SSB cross sections almost reached the level of a non-modified DNA sequence, which demonstrates the high sequence dependence of the sensitizing effect of 5BrU. 8BrA was incorporated in a 13mer oligonucleotide as well and the strand breaks were quantified upon 8.44 eV photon irradiation in direct comparison to a non-modified DNA sequence of the same composition. No clear enhancement of the SSB yield of the modified in comparison to the non-modified DNA sequence could be observed. Additionally, secondary electrons with a maximum energy of 3.6 eV were generated when using Si as a substrate giving rise to further DNA damage. A clear enhancement in the SSB yield can be ascertained, but to the same degree for both the non-modified DNA sequence and the DNA sequence modified with 8BrA. However, the kinetics of these reactions is still poorly understood, which is also due to the complexity of plasmonic nanostructures. We determined the reaction rates of the hot-electron-mediated reaction of 4-nitrothiophenol (NTP) on gold nanoparticles (AuNPs) using fractal kinetics as a function of the laser wavelength and compared them with the plasmonic enhancement of the system. The reaction rates can be only partially explained by the plasmonic response of the NPs. Hence, synchrotron X-ray photoelectron spectroscopy (XPS) measurements of isolated NTP-capped AuNP clusters have been performed for the first time. In this way, it was possible to determine the work function and the accessible valence band states of the NP systems. The results show that besides the plasmonic enhancement, the reaction rates are strongly influenced by the local density of the available electronic states of the system. Previous approaches to coat ZIF particles with polyelectrolytes led to surface corrosion of the template material. This work overcomes previous limitations by performing a Layer-by-Layer (LbL) polyelectrolyte coating onto ZIF-8 and ZIF-67 particles in nonaqueous environment. Using the 2-methylimidazolium salt of polystyrensulfonic acid instead of the acid itself and polyethyleneimine in methanol led to intact ZIF particles after polyelectrolyte coating. This was verified by electron microscopy. Further, zetapotential and atomic force microscopy measurements confirmed a continuous polyelectrolyte multilayer built up. The here reported adaption to the well-studied (LbL) polyelectrolyte selfassembly process provides a facile method to equip ZIF particles with a nanometer thin polyelectrolyte multilayer membrane. ZIF-67 is often used in polymer composite materials e. g. for gas separation membranes. Post-synthesis treatment of ZIF-67 crystals with ultrasound leads to unforeseen plasticity that resulted in sintered ZIF-67 and ZIF-67 nano-flakes. Consequently, ultrasound increases the external surface area of ZIF-67 which might improve e.g. blending with polymers in composite materials. These new morphologies of ZIF-67 were characterized by transmission electron, scanning electron, and atomic force microscopy. The ultrasound treatment of ZIF-67 did not result in the formation of an amorphous framework or a meta-stable crystal structure as indicated by powder x-ray diffraction. In addition, ultra-sonicated ZIF-67 retained the high gas adsorption capacity and pore size compared to synthesized ZIF-67. The morphological changes are hard to detect with standard analytical methods that are usually utilized for MOF characterization. These findings also suggest that sonochemical treatment of ZIFs leads to structural effects beyond increasing the amount of nucleation clusters during sono-chemical synthesis, which is currently not addressed in the field.
