A1 - Panne, Ulrich A1 - Schneider, Rudolf T1 - Cetirizine as pH-dependent cross-reactant in a carbamazepine-specific immunoassay N2 - High performance liquid chromatography (HPLC) was hyphenated with a previously reported carbamazepine-specific enzyme-linked immunosorbent assay (ELISA) as a screening approach to water analysis in order to identify possible interferences from transformation products. Treated wastewater was analysed and three substances were recognized by the antibody besides carbamazepine: the metabolites 10,11-dihydro-10,11-epoxycarbamazepine and 2-hydroxycarbamazepine plus the structurally not obviously related antihistamine cetirizine. The molar cross-reactivity against cetirizine was found to be pH-dependent and assessed to be 400% at pH 4.5 and 22% at pH 10.5. Performing the ELISA at pH 10.5 greatly improved the accuracy when carbamazepine was determined in surface and wastewater samples. T1 - CCQM-K126: low polarity organic in water: carbamazepine in surface water N2 - This study aimed to assess the measurement capabilities of participating National Metrology Institutes/ Designated Institutes (NMIs/DIs) and expert laboratories in determining of low-polarity organics in surface water. This comparison was organized by Government Laboratory, Hong Kong (GLHK). At the CCQM Organic Analysis Working Group (OAWG) Meeting held in November 2012 in Hong Kong, GLHK initially proposed a CCQM key comparison and a parallel pilot study programme on pharmaceuticals in surface water. Further discussion at the CCQM Meeting held in April 2014 in Paris, the OAWG approved a CCQM Track C comparison (CCQM-K126) on low polarity pharmaceuticals in surface water. In the meeting, the programme was supported by more than six National Metrology Institutes/ Designated Institutes (NMIs/ DIs). CCQM-K126 officially commenced in July 2014 and had registration from nine NMIs/DIs. Participants were provided two bottles (40 mL each) of surface water and were requested to determine the mass fraction of spiked carbamazepinein in surface water. The coordinator received nine sets of results from eight NMIs/DIs in February 2015. Apart from one using an immunoassay technique, all participants applied isotope dilution liquid chromatography-tandem mass spectrometry (ID-LCMS/MS) technique as their determination technique. T1 - Can ocean warming alter sub-lethal effects of antiepileptic and antihistaminic pharmaceuticals in marine bivalves? N2 - The negative effects induced in marine organisms by Climate Change related abiotic factors consequences, namely ocean warming, are well-known. However, few works studied the combined impacts of ocean warming and contaminants, as pharmaceutical drugs. Carbamazepine (CBZ) and cetirizine (CTZ) occur in the marine environment, showing negative effects in marine organisms. This study aimed to evaluate the impacts of Ocean warming on the effects of CBZ and CTZ, when acting individually and combined (drug vs drug), in the edible clam Ruditapes philippinarum. For that, drugs concentration, bioconcentration factors and biochemical parameters, related with clam’s metabolic capacity and oxidative stress, were evaluated after 28 days exposure to environmentally relevant scenarios of these stressors. The results showed limited impacts of the drugs (single and combined) at control and warming condition. Indeed, it appeared that warming improved the oxidative status of contaminated clams (higher reduced to oxidized glutathione ratio, lower lipid peroxidation and Protein carbonylation levels), especially when both drugs were combined. This may result from clam’s defence mechanisms activation and reduced metabolic capacity that, respectively, increased elimination and limited production of reactive oxygen species. At low stress levels, defence mechanisms were not activated which resulted into oxidative stress. The present findings highlighted that under higher stress levels clams may be able to activate defence strategies that were sufficient to avoid cellular damages and loss of redox homeostasis. Nevertheless, low concentrations were tested in the present study and the observed responses may greatly Change under increased pollution levels or temperatures. Further research on this topic is needed since marine heat waves are increasing in frequency and intensity and pollution levels of some pharmaceuticals are also increasing in coastal systems. Im Rahmen der Projekte MARKERIA I & II wurden verschiedene anthropogene Marker wie Koffein, Carbamazepin, Diclofenac, Bisphenol A und Hormone wie Östron untersucht, um deren Eignung für die Abwassersurveillance zu bewerten. Die Ergebnisse zeigen, dass einige Substanzen wie Carbamazepin stabile Konzentrationen aufweisen und sich gut als Marker eignen, während andere wie Koffein starke Schwankungen zeigen. Einige Marker wie Isolithocholsäure werden zwar in hohen Konzentrationen gefunden, zeigen aber methodische Herausforderungen wie instabile Assays. Die Studie hebt hervor, dass es Hotspots mit erhöhten Konzentrationen gibt, aber insgesamt eine relativ geringe Variabilität zwischen verschiedenen Rohabwässern besteht. Zukünftige Entwicklungen sollen sich auf die Verbesserung von Antikörpern, die Entwicklung tragbarer Vor-Ort-Analysemethoden und die Standardisierung immunanalytischer Verfahren konzentrieren. T1 - Application of dispersive liquid-liquid microextraction for estrogens' quantification by enzyme-linked immunosorbent assay N2 - Estrogens, such as 17β-estradiol (E2) and 17α-ethinylestradiol (EE2), are the major responsible for endocrine-disrupting effects observed in aquatic environments due to their high estrogenic potency, even at concentrations ranging from pg L-1 to ng L-1. Thus, it is essential to develop analytical methodologies suitable for monitoring their presence in water samples. Dispersive liquid–liquid microextraction (DLLME) was used as a pre-concentration step prior to the quantification of E2 and EE2 by enzyme-linked immunosorbent assay (ELISA). First, an evaluation of the effect of DDLME on the E2 and EE2 ELISA calibration curves was performed. Since the extraction procedure itself had an influence on the ELISA optical density (OD), it became necessary to subject, not only the samples, but also all the standards to the DLLME process. Working ranges were determined, being between 1.2 and 8000 ng L-1, for E2, and between 0.22 and 1500 ng L-1, for EE2. The influence of organic matter, both in the extraction and quantification, was evaluated and it was observed that its presence in the solution did not affect considerably the calibration curve. Recovery rates were also determined, ranging from 77% to 106% for ultrapure water and from 104% to 115% for waste water samples, the most complex ones in what concerns matrix effects. Results obtained when applying the proposed method to real water samples can be considered quite satisfying. Moreover, the obtained working ranges encompass values generally reported in literature, confirming the practical use of the method for environmental samples. In contrast to classical microtiter plate-based assays, microparticles are a next generation solid support, as they promote automation of immunoassays using flow-based techniques. Antibody immobilization is a crucial step, as these reagentsmare expensive, and inefficient coupling can result in low sensitivities. This paper proposes a general procedure for efficient immobilization of antibodies onto TentaGel particles, via Nhydroxysuccinimide chemistry. The goal was the preparation of solid supports with optimum immunorecognition, while increasing the sustainability of the process. The influence of buffer composition, activation and coupling time, as well as the amount of antibody on the immobilization efficiency was investigated, resorting to fluorophore-labeled proteins and fluorescence imaging. Buffer pH and activation time are the most important parameters for efficient coupling. It is demonstrated, that the hydrolysis of N-hydroxysuccinimide esters occurs at similar rates as in solution, limiting the utilizable time for coupling. Finally, applicability of the generated material for automated affinity extraction is demonstrated on the mesofluidic platform lab-on-valve. Another lingering problem are health-threatening contaminants, such as mycotoxins, that deteriorate food and feed. Analytical methods, suitable for trace analysis, are needed that are desirably also fast, inexpensive and, if possible, robust, and portable. Immunoanalytical, i.e., anti-body-based technologies, which are available in a broad range of formats, can be profitably used here to analyze for the distribution and the concentration trends of contaminants. Some of these formats are single-analyte but high-throughput methods. To use them wisely, indicator substances or sum parameters must be established to be used in screening approaches. Other methods are suitable to be performed on portable in-strumentation in the field (on-site), more precisely at the point-of-need. Furthermore, there are array technologies that allow for parallel analysis of several analytes of interest (multiplexing). The talk reports on methods and data for compounds con-sidered as anthropogenic contamination markers (the antiepileptic carbamazepine, the analgesic diclofenac, the anti-histaminic cetirizine, the steroid hormone estrone, the antimicrobial amoxicillin, the stimulants caffeine and cocaine, the endocrine disruptor bisphenol A, and a bile acid). Some work on mycotoxins (ochratoxin A and ergot alkaloids) is also presented. It is demonstrated that antibody-based technolo-gies have a huge potential to gain analytical insights at the point-of-need.
