A1 - Prager, Jens T1 - Acoustic-structure interaction in the Scaled Boundary Finite Element Method for primsatic geometries N2 - Due to the short wavelength compared to the dimensions of the structure, the simulation of ultrasonic waves is still a challenging task. A numerical method well suited for this purpose is the semi-analytical Scaled Boundary Finite Element Method (SBFEM). When applying this method, only the boundary of a computational domain is discretized using finite elements, while the interior is described by an analytical ansatz. Hence, the number of degrees of freedom is reduced significantly compared to the classical Finite Element Method (FEM). In recent years, a particular formulation of the SBFEM for the simulation of ultrasonic guided waves was developed. The method constitutes an efficient algorithm for prismatic structures of arbitrary length, such as plates, pipes, or beams. Wave propagation phenomena in such structures can be modeled for isotropic and anisotropic inhomogeneous waveguides. Even though the method is an efficient tool for the simulation of guided waves in solid media, a reliable model for the simulation of acoustic wave propagation in fluids as well as acoustic-structure interaction in terms of SBFEM is still missing. In principle, the fluid can be described by a displacement-based formulation and thus be implemented in existing SBFEM algorithms for solid bodies. However, due to the discretization with classical finite elements, spurious modes occur, which cannot be separated from the physical modes straightforwardly. The spurious modes can be suppressed using a penalty parameter. Although very accurate results were achieved for some problems, this procedure has been proven unreliable for certain cases. For this reason, we propose a different approach in this contribution. We employ a pressure model to simulate the acoustic behavior of fluids. The implementation of the pressure model results in a higher effort due to the necessity of incorporating coupling terms, but it presents a stable alternative without spurious modes. The accuracy of the method is demonstrated in comparison with analytical solutions and results obtained using the FEM. This was achieved by means of Failure Modes and Effects Analysis (FMEA) carried out with eight NDT experts and by a survey of 27 experienced inspectors. The results show that failures can happen throughout the entire NDT process. Prevention of those failures could be improved through the optimization of the organization, technology, documentation and regulations, working conditions and the general process, and through the optimization of the formal education and training. Specialized training of the executives, extended training of the supervisors and the inspectors and improvement of the inspection documentation have been suggested. The study also showed potential for the improvements of the inspection in the field. Prior to the formation of a biofilm on a substrate, planktonic cells attach on the surface. Hence, the properties of the surface play a key role in biofilm formation and are of great importance for the development of strategies to prevent bacterial attachment and biofilm formation. This project aims at clarifying to which extent surface micro-/nanostructuring and chemical functionalization affects bacterial attachment and whether a synergistic combination of the two can be used to control bacterial adhesion. To answer these questions, model surfaces with regular patterns of 5-10 micrometers in size have been prepared, which provide distinct zones differing in terms of their chemistry or nano-roughness. This was achieved by micro contact printing of self-assembled monolayers with different functional groups and deposition of patterned ZnO nanorod arrays for studying the effect of surface chemistry and morphology, respectively. Typical contrasts studied were combinations of positively/negatively charged, hydrophobic/hydrophilic or flat/rough. The attachment behavior of bacteria on tailored surfaces were studied in a flow chamber as a function of time. The strain Pseudomonas fluorescens SBW25 was chosen as a model organism. DNA-intercalating dyes such as Syto9 have a high affinity to adsorb on ZnO nanorods. To overcome this limitation a genetic modification was performed by introducing a gene which expresses a green fluorescent protein in P. fluorescens SBW25 enabling the quantitative evaluation of the flow chamber studies by means of fluorescence microscopy. Further analysis of the attachment behavior was performed by means of scanning electron microscopy. The presentation will summarize the results of our systematic study on the role of individual parameters on bacterial attachment and highlight synergistic combinations, showing an inhibition or enhancing effect. As the investigations with model substrates enable a precise control of the surface parameters, this approach can be applied to different microorganisms and material systems to achieve a correlative description of bacterial adhesion on solid surfaces. Performing SDS-PAGE and using the SDS-PAGE buffer as an eluent showed that CER could be successfully enriched from human serum. Based on the copper determination with ICP-MS, the amount of bound CER on the nanodiamonds can be calculated. We could show the fulfilled mass balance of bound CER and CER in the supernatant after incubation with a known amount of CER. For isotope ratio analysis this method can be applied to compare ratios of the total copper content in human serum to copper ratios from CER enrichment. As, in the past, the advantages in ageing resistance and durability were highly evaluated, we now face the problem of quite stable plastic waste within the environment. However, there is only little knowledge on the real timescales until macroscopic fragmentation for the different kinds of plastic under various environmental conditions. Here, weathering methods are presented, which have been used for the failure prediction in specific outdoor conditions. Issues of uncertainty, reproducibility, or validation are discussed. For the prediction of the plastic’s fragmentation, much longer time scales have to be considered. To do this within a limited project life span, various processes as well as their acceleration potential have to be evaluated separately, such as temperature increase or spectral shift. The possibilities and limitations of such controlled acceleration will be discussed. T1 - Discussion on Weathering Reference Materials N2 - Weathering reference materials are used to characterize the harshness of an exposure, aiming on either reproducibility of a specific exposure or on the comparability between various kinds of weathering exposure. The materials that are used as weathering reference materials differ in their sensitivities (as well as in interactions and interferences of the latter), conditioned by the different processes which lead to the respective property change. It is also essential to take into account the necessary measurement equipment for the respective property change, in order to allow timely intervention. What are the key issues on choosing a weathering reference material? What can be learned from the weathering reference materials, investigated so far? Possibilities and limitations are discussed on the basis of existing weathering reference materials. Conclusions are drawn, for establishing new weathering reference materials. Im Gegensatz dazu wies Grundwasser aus dem Einzugsbereich einer historischen Kontamination durch Fluorchlorkohlenwasserstoffe (FCKW) ein TFA/DFA-Verhältnis von 1:3 auf. Dies und die ungewöhnlich hohe DFA-Konzentration an dieser Stelle wird vor dem Hintergrund der beobachteten mikrobiellen Abbauprodukte des ursprünglichen eingetragenen FCKW 1,1,2-Trichlor-1,2,2-trifluorethan (R113) diskutiert. Eine mikrobielle Umwandlung des bekannten Abbauproduktes Chlortrifluorethylen (R1113) zur DFA wurde bislang nicht in Umweltkompartimenten beobachtet und wird hier auf Basis bekannter Stoffwechselwege vorgeschlagen. TFA wurde in Regenwasser, Oberflächenwasser und Grundwasser in vergleichbaren Größenordnungen von ca. 500 ng/L bestimmt. Im Gegensatz dazu wurde DFA in Grundwasserproben im Bereich der Schadstoffquelle der FCKW-Kontamination mit bis zu 2.000 ng/L nachgewiesen. Die Summe von TFA und DFA im Grundwasser korreliert nicht mit dem Eintrag aus Niederschlag und Oberflächenwasser. Darum liegt die Vermutung nahe, dass es für DFA andere Quellen als den direkten Eintrag oder den möglichen Abbauweg über TFA geben muss. DFA ist vermutlich ein Abbauprodukt von R1113. FCKWs, die im ersten Abbauschritt zu 1,1-Difluorethenen metabolisieren, stehen im Verdacht, in der Folge zu DFA zu hydrolysieren. Entscheidend scheint, dass bereits im Ausgangsprodukt ein CF2-Strukturelement enthalten ist, das dann zur Bildung von DFA führen kann. Somit würde aus dem ursprünglichen Eintrag des Kältemittels R113 über schrittweise Dechlorierung via 1,2-Dichlor-1,2,2-Trifluorethan (R123a) und R1113 die Bildung des beobachteten DFA folgen. Es ist also sinnvoll, DFA in die Diskussion zur Regulierung der Endprodukte des Abbaus fluorhaltiger Kälte- oder Lösungsmittel einzubeziehen. Die Ergebnisse wurden im Normausschuss Wasserwesen (NAW) des DIN vorgestellt und es soll ein Arbeitskreis gegründet werden, der ein Normverfahren zur Quantifizierung von TFA und DFA in Wasser erarbeitet. Die fehlerfreie Bauteilproduktion wird jedoch durch Reibungs- und Verschleißerscheinungen aufgrund hoher thermo-mechanischer Werkzeugbelastungen und fehlender Schmierstoffsysteme limitiert. Als Lösungsansatz wird eine Modifikation der Werkzeugoberfläche mittels Laserimplantation angestrebt, um folglich deren Verschleißbeständigkeit nachhaltig zu erhöhen. Das Verfahren basiert auf einem lokalen Dispergieren keramischer Hartstoffpartikel in die Werkzeugoberfläche, infolgedessen hochfeste und erhabene Strukturen im Mikrometerbereich entstehen. Aufgrund der signifikanten Reduzierung der Kontaktfläche sowie der hohen Verschleißbeständigkeit der eingesetzten TiB2-Hartstoffe wird ein verbessertes tribologisches Einsatzverhalten unter presshärtetypischen Prozessbedingungen erwartet. Zur Verifizierung dieser Annahmen wurden im Rahmen dieser Arbeit modifizierte Pin-on-Disk Tests durchgeführt, um das Reib- und Verschleißverhalten der laserimplantierten Werkzeugoberflächen unter Presshärtebedingungen zu untersuchen. Zur weiteren Vertiefung des Prozess-verständnisses wurden die verschlissenen Platinen via Tastschnittmessungen und Querschliffaufnahmen analysiert. Wärme, Feuchte und atmosphärische Gase beeinflusst. Die Messung einer effektiven Bestrahlungsstärke und ihre zeitliche Integration sind deshalb von großer Bedeutung. Photochemische Prozesse werden beim Sehen oder allgemeiner bei Farbstoffen schon durch sichtbare (VIS) Strahlung, dagegen bei der Erythembildung der Haut und der Photodegradation von Polymeren erst durch die energiereicheren Photonen der ultravioletten (UV) Strahlung initiiert. Die spektrale Empfindlichkeit des Auges und die Erythembildung sind im VIS- bzw. UV-Bereich sehr stark wellenlängenabhängig, was durch die Angabe der mit der spektralen Hellempfindlichkeit V() des Auges bewerteten VIS-Strahlung in Lux (lx) bzw. die mit der Erythem-Wirkfunktion bewertete UV Strahlung als dimensionsloser UV-Index berücksichtigt wird. Spektrale Empfindlichkeiten. Die Photodegradation von Polymeren ist im UV-Bereich ebenfalls stark wellenlängenab-hängig, was bisher nicht berücksichtigt wurde. Seit Mitte der 70er Jahre, als die lx- bzw lxh- Messung bei der Strahlungsbeanspruchung langsam durch die Ermittlung der radiometrischen UV-Bestrahlungsstärke bzw. der UV-Bestrahlung im gesamten UV-Bereich abgelöst wurde, gab es hier bei der Bewitterungsprüfung keine weiteren Fortschritte auf dem Gebiet der Charakterisierung der Strahlungsbeanspruchung im UV-Bereich. Seit Jahren liegen zahlreiche Ergebnisse über die spektrale Empfindlichkeit von Photo-degradationsprozessen vor, die zeigen, dass die spektrale Empfindlichkeit im Bereich der Globalstrahlung bei ca. 300 nm, am größten ist und im langwelligen UV-Bereich bei ca. 400 nm gegen Null geht. Die Messungen der radiometrischen Größen UV-Gesamt-bestrahlungsstärke und UV-Gesamtbestrahlung erfasst deshalb über weite Wellen-längenbereiche Strahlungsanteile, die für die Photodegradation bedeutungslos sind und mittelt über potenziell bedeutsame Änderungen im kurzwelligen UV-Bereich hinweg. Es wird das Konzept eines neuen UV-Radiometers vorgestellt, dessen spektrale Empfindlichkeit einer mittleren spektralen Empfindlichkeit der Photodegradation von Polymeren entspricht. Due to the moisture content being only 50 ppmV no visible acid condensation took place. To simulate stress conditions at the inside pipeline surface due to fluid pressure (10 MPa) specimens were preloaded using a load frame. Experiments conducted at 278 K and at 313 K revealed the highest corrosion rate at lower temperature. Corrosive effect of impurities was strongest applying mixed atmosphere, containing oxidizing and reductive components, closely followed by CO2 streams with pure oxidizing character. By far, the lowest corrosion rate (10x lower) resulted from reductive atmosphere. In general, at constant temperature and pressure the CO2 stream composition strongly influences the morphology, thickness and composition of the corrosion products. Applying oxidizing or mixed impurities, iron hydroxides or oxides (e.g. goethite, hematite) occur as dominating corrosion products, capable to incorporate different amounts of sulfur. In contrast, using reductive atmosphere very thin corrosion layers with low crystallinity were developed, and phase identification by XRD was unfeasible. SEM/EDX analysis revealed the formation of Fe-O compounds, most likely attributed to the oxygen partial pressure in the system induced by CO2 (≥0.985 volume fraction) and volatile H2O. In addition to the surface covering corrosion layer, secondary phases had grown locally distributed on top of the layer. These compounds are characteristic for the applied atmosphere and vary in number, shape and chemical composition. However, an important and almost always present effect that is typically set aside is the material oxidation during or after the laser processing. Usually, this leads to optical, chemical, structural and mechanical alterations that are seldomly studied in detail. Therefore, in this work we present a study that includes the formation of LIPSS on Ti6Al4V alloy for tribological applications were different oxidations layers are involved. The material characterization using different techniques, such as energy dispersive X-ray analyses (EDX), X-ray photoelectron spectroscopy (XPS), micro Raman spectroscopy (µ-RS), and depth-profiling Auger electron microscopy (AEM), and other techniques, confirms the formation of a non-negligible superficial oxide layer – strongly dependent on the specific processing parameters. We perform tribological experiments on laser irradiated samples oxidized by alternative ways to have a comparison on its friction performance. The obtained results show significant differences that depend directly on the thickness and the quality of the oxidized layer. This may open a fertile research area until now barely explored with clear impact on LIPSS based applications. A1 - Bruno, Giovanni T1 - Bestimmung der Mikrostruktur und Simulation des Schädigungs-verhaltens von lasergesintertem Polyamid 12 unter statischer Zugbelastung N2 - Um das Material- und Schädigungsverhalten von additiv gefertigtem Polyamid 12 (PA12) unter quasistatischer Belastung zu charakterisieren, wurden mechanische Tests und Experimente zur Bestimmung der Mikrostruktur durchgeführt. Die Proben wurden nach dem Prinzip des Selektiven Lasersinterns (SLS) hergestellt. Unter quasistatischer Belastung mit Haltezeiten ergab sich ein viskoplastisches Materialverhalten. Im Zugversuch wurde eine maximale Zugfestigkeit von 40.6 MPa und eine Bruchdehnung von 7.4% beobachtet. Mittels Digitaler Bildkorrelation (DIC) wurde die Ausbildung von Scherbändern während des Zugs nachgewiesen. Mittels Röntgenrefraktion wurde eine Erhöhung von inneren Oberflächen beobachtet, die senkrecht zur Zugrichtung orientiert sind. Die Analyse der Gesamtporosität aus Computertomographie-Messungen ergab keine Änderung infolge der Zugbelastung. Jedoch wurde eine bimodale Porengrößenverteilung und eine steigende Sphärizität festgestellt. Das Materialverhalten wurde mit den Ergebnissen des Zugversuchs nach dem Chaboche Modell [3] in sehr guter Übereinstimmung simuliert. Allerdings gestattet dieses Modell nicht, das Bruchverhalten einzuschließen. Daher wurde zur Simulation des Schädigungsverhaltens das Modell gemäß dem Ansatz von Gurson, Tvergaard und Needleman unter Berücksichtigung der mikrostrukturellen Parameter erweitert. Während das Chaboche Modell eine homogene Spannungsverteilung generiert, zeigt das erweiterte Modell die Ausbildung von Scher¬bändern und die korrekte Bruchdehnung. However, the dressing result is highly dependent on bond components, cutting grain size and concentration which lead to local differences in the material removal and irregular topographies. Improvements concerning spatial resolution as well as signal-to-background ratio due to low-dispersion sample chambers make LA-ICP-MS also interesting for single cell analysis. To evaluate the interaction of nanoparticles (NPs) with cells LA-ICP-MS was applied for the imaging of individual cells. Our findings show, that NP aggregates can be localized within cellular compartments. The uptake efficiency depends strongly on the physico-chemical properties of the nanostructures (size, chemical composition, surface modification), as well as on the incubation conditions (concentration, time). Moreover, LA-ICP-MS is increasingly becoming an important complementary technique in bioanalysis by using element-tagging strategies to determine biomolecules indirectly. Based on the specific binding between antibodies and their corresponding antigens, proteins and peptides can be detected in tissue or cells using tagged antibodies. As artificial tags metal chelates loaded with lanthanides, polymer-based elemental tags or metal-containing nanoparticles can be used. Thereby LA-ICP-MS is a sensitive detection tool for multiplexed immuno-histochemistry of tissue and cell samples. Die fehlerfreie Bauteilproduktion wird jedoch durch Reibungs- und Verschleißerscheinungen aufgrund hoher thermo-mechanischer Werkzeugbelastungen und fehlender Schmierstoffsysteme limitiert. Als Lösungsansatz wird eine Modifikation der Werkzeugoberfläche mittels Laserimplantation angestrebt, um folglich deren Verschleißbeständigkeit nachhaltig zu erhöhen. Das Verfahren basiert auf einem lokalen Dispergieren keramischer Hartstoffpartikel in die Werkzeugoberfläche, infolgedessen hochfeste und erhabene Strukturen im Mikrometerbereich entstehen. Aufgrund der signifikanten Reduzierung der Kontaktfläche sowie der hohen Verschleißbeständigkeit der eingesetzten TiB2-Hartstoffe wird ein verbessertes tribologisches Einsatzverhalten unter presshärtetypischen Prozessbedingungen erwartet. Zur Verifizierung dieser Annahmen wurden im Rahmen dieser Arbeit modifizierte Pin-on-Disk Tests durchgeführt, um das Reib- und Verschleißverhalten der laserimplantierten Werkzeugoberflächen unter Presshärtebedingungen zu untersuchen. Zur weiteren Vertiefung des Prozess-verständnisses wurden die verschlissenen Platinen via Tastschnittmessungen und Querschliffaufnahmen analysiert. These novel TOP nanosensors (temperature, oxygen, pH) cover the biologically and physiologically relevant concentration ranges of these parameters/analytes with single wavelength excitation in PBS buffer and in a cell culture medium containing bovine serum albumin (BSA). The response of both nanosensors to all parameters is fully reversible and only minimally affected by the presence of BSA, the most common serum albumin. Moreover, comparative studies with nanosensors containing only a single type of stimuli-responsive molecule and with the respective molecular systems revealed that the different sensor components do not interfere with each other. These surface nanostructures are a universal phenomenon and can be generated on almost any material by irradiation with intense linearly polarized radiation. LIPSS are formed in a “self-ordered” way and are often accompanying laser material processing applications. The structures can be produced in a single-step process and enable surface functionalization through the adaption of optical, mechanical and chemical surface properties. Their spatial periods typically range from several micrometers down to less than 100 nanometers, exhibiting a clear correlation with the polarization direction of the laser radiation used. Various types of LIPSS have been classified, relevant control parameters were identified, and material specific formation mechanisms are analyzed for different types of inorganic solids, i.e., metals, semiconductors, and dielectrics, through time-resolved optical experiments and theoretical simulations. Special attention will be paid to a comparison of the currently available formation theories for LIPSS with a discussion of their respective strengths and weaknesses. Photoluminescence signals are, however, affected by wavelength-, polarization- and time-dependent instrument specific effects. This hampers the comparability of fluorescence measurements and calls for simple tools for instrument characterization and the quantification of measured fluorescence intensities. Well characterized fluorescence standards for instrument calibration and performance validation (IPV) can be used also to reference fluorescence signals. Of special importance is the reliable and accurate determination of photoluminescence quantum yields (Ф f), that equals the number of emitted per absorbed photons and presents the key performance parameter for emitter efficiency and the comparison of different luminophores. The determination of Ф f is typically done with the aid of so-called quantum yield standards with well-known Ф f values. These standards can also be applied to evaluate integrating sphere setups, which are increasingly being used for absolute measurements of Ф f values. In this respect, division biophotonics of BAM has certificated a set of Ф f standards, which absorb and fluorescence in the wavelength range from 350 to 1100 nm. T1 - Post-impact evaluation at RC plates with planar tomography and FEM N2 - Due to the many possible applications, the uncomplicated production and the high application range, reinforced concrete is a widely used building material. This large range of physical material properties still poses an engineering challenge in determining all necessary requirements for predicting dynamic effects under impact load. Many aspects of impact have already been examined and some correlations have been studied intensively. Examples are the work of Lastunen and Booker, who studied the influence of projectile properties on the impact form. Li has also investigated the local effects of an impact. The field of detailed damage analysis has not been in focus so far. This presentation shows some studies in medium-velocity impact with the focus on post-impact damage evaluation. The impactor is modified so that the test plates show low penetration on the top and scabbing on the bottom. With the unique tomography lab test stand at BAM the plate is scanned after the impact and the damage is analyzed. Cracks and scabbing are made visible with a reconstruction. The comparison of simulation and tomography allows to create prognosis models for damage characterization. With the extension of the tested zone to the running surface, rail defect signal types other than head checks can be measured. Due to their most-ly irregular shape, a quantitation based on a calibration against regular test cracks of varying depth may not be linear. Estimates of the expected influence of more complex crack patterns may be obtained by a finite element simulation of sufficiently simple limiting cases, like two displaced or intersecting cracks or a simply branched or flexed crack. Diese Untersuchungen hatten zum Ziel, die wesentlichen Einflussparameter bei der Handprüfung auf das Prüfergebnis zu evaluieren. Im Rahmen der Zuverlässigkeitsbetrachtung wurden die ProbabilityofDetection(POD) und die Nachweisgrenzen ermittelt.Die durchgeführte Studie zeigt, dass die Bewertung der gesamten Zuverlässigkeit der ZfP komplex ist. Dieses verlangt eine detaillierte Versuchsplanung und die Einbeziehung von unterschiedlichen Faktoren, von den Anwendungsparametern bis hin zu den komplexen menschlichen und organisatorischen Faktoren. Die im Rahmen der Studie ermittelten Ergebnisse lassen aufgrund der geringen Anzahl an Testreflektoren keine allgemeingültige Übertragung auf beliebige Korbbogen-geometrienund Reflektorgeometrienzu. T1 - Simulation von mechanisierten Eisenbahnschienenprüfungen mit Ultraschall N2 - Bei der mechanisierten Prüfung von verlegten Eisenbahnschienen mit Schienenprüfzügen werden große Prüfdatenmengen mit einer komplexen Prüfkopfanordnung in kurzer Zeit bei hohen Prüfgeschwindigkeiten aufgenommen. Der Typ und der Zustand des Gleises kann während der Prüfung in schnellem Wechsel streckenabhängig variieren, z.B. unterschiedliche Schienentypen oder Abnutzung sowie beim Durchfahren von Weichenstraßen. Der logistische Aufwand diese Prüfung insgesamt durchzuführen ist dabei sehr hoch und eine Wiederholung der Prüfung in einem kurzen Zeitintervall nicht möglich. Es werden während der Prüfdatenaufnahme bereits Datenreduktionsverfahren, Rekonstruktionsverfahren und Online-Parametervariationen eingesetzt, um die Ergebnisdaten zum einen in Ihrer Menge zu begrenzen zum anderen auf die lokalen Geometrie- und Zustandsänderungen des Prüfobjektes Schiene zu reagieren. Im Regelwerk EN 16729-1 „Zerstörungsfreie Prüfung an Schienen im Gleis -Teil 1: Anforderungen an Ultraschallprüfungen und Bewertungsgrundlagen“ werden Bezugsreflektoren definiert, um die Leistung automatisierter Ultraschallsysteme für die Schienenprüfung zu verifizieren. Um die Einflüsse der komplexen Randbedingungen der mechanisierten Schienenprüfung auf das Prüfergebnis evaluieren zu können, wurde ein Softwarewerkzeug erarbeitet, mit dem eine Prüffahrt auf einer Schiene simuliert werden kann. Basierend auf der Datenaufnahme mit dem Glassy-Rail-Diagramm können alle relevanten Prüfparameter und Artefakte in Schienen sowie Störeinflüsse bei der Simulation berücksichtigt werden. Constantly increasing traffic density and heavy loads have been the motor for the development of new test equipment from handheld devices to rail inspection trains. (Krull 2003)Up to the present most of the system solutions feature conventional ultrasonic transducers housed in wheel-type and slide-type probes. Different tasks have to be carried out during an in-service inspection for flaws in the rail head, rail web and rail foot as well as rolling contact fatigue (Heckel 2018). The more tasks the inspection system has to perform, the more probes are needed. Compared against standard ultrasonic testing methods the application of array probes offers advantages and flexibility by the electronic steering possibilities to control the transmitted and received sound fields. This allows to increase functionality by software while decreasing the number of probes needed in hardware in parallel. One drawback in application of phased array probes is that the repetition frequency of the subsequent measurements will be reduced by the number of virtual probe functions each phased array probe has to perform. This may limit the range of use for phased array probes in high speed applications. To overcome these limits special designs for array probes and signal processing are necessary. T1 - Modellgestützte Bestimmung der Auffindwahrscheinlichkeit (POD) von Reflektoren bei der Ultraschallprüfung großer Schmiedeteile N2 - Bruchmechanische Untersuchungen werden zur Vorhersage der Lebensdauer und der Wartungszyklen benutzt. Dabei wird davon ausgegangen, dass auch Objekte ohne detektierte Anzeigen Materialfehler mit einer Reflektivität kleiner der Nachweisgrenze aufweisen können. Keine detektierte Anzeige bedeutet daher für Bauteilauslegung, dass sie eine konservative Annahme machen müssen: es wird angenommen, dass das Bauteil Materialfehler in Größe der Nachweisgrenze enthält. Dies macht die Nachweisgrenze zu dem die Lebensdauer limitierenden Faktor. Bei probabilistischen Methoden wird zusätzliches Wissen, wie Zuverlässigkeit und Fehlerauffindwahrscheinlichkeiten bei der zerstörungsfreien Prüfung mit in die Berechnung einbezogen, um zu konservative Annahmen zu vermeiden. Ziel ist es eine Model-Assisted-POD aufzubauen mit der die reale volumetrische Ultraschallprüfung großer Schmiedeteile nachgebildet werden kann. Die resultierende POD soll im Weiteren in bruchmechanischen Betrachtungen Verwendung finden. In vorangegangenen Arbeiten [Preißel2015] wurde dazu der Einfluss des Prüfrasters nach der DGZfP-Richtlinie US 07:2014 auf die Fehlerauffindwahrscheinlichkeit (parametrisch) simulativ betrachtet und die Simulationstechnik experimentell validiert. In diesem Paper werden die Ergebnisse von Preißel überprüft und um eine Prüfraster-Summen-POD, die das Szenario der realen Bauteilprüfung beschreibt, erweitert. Zudem wird eine experimentell validierte, simulative, nicht-parametrische POD für schrägliegende Defekte aufgebaut. Schließlich werden erste Untersuchungen zur Defektmorphologie vorgestellt. Diese Arbeit ist im Rahmen eines Studenten-Stipendiums der DGZfP entstanden. T1 - Zuverlässigkeitsbetrachtungen bei der Ultraschallprüfung von Radsatzwellen im Ausbildungsbetrieb N2 - Im Rahmen der UT1-M1 Ausbildung im Ausbildungszentrum Wittenberge der DGZfP wurden statistische Untersuchungen am Beispiel der Radsatzwellenprüfung mit Testradsatzwellen durchgeführt. Diese Untersuchungen hatten zum Ziel, die wesentlichen Einflussparameter bei der Prüfung auf das Prüfergebnis zu evaluieren. Dabei wurde unter anderem die intrinsische Fähigkeit des Prüfverfahrens zum Auffinden verschiedener Reflektorgeometrien ermittelt. Die Ergebnisse der Handprüfung wurden mit Ergebnissen mechanisierter Prüfungen verglichen. Im Rahmen der Zuverlässigkeitsbetrachtung wurden die Probability of Detection (POD) und die Nachweisgrenzen ermittelt. Ziel dieser Arbeit war es auch, menschliche und organisatorische Einflussfaktoren auf die Ultraschall-Handprüfung von Radsatzwellen zu identifizieren, um Empfehlungen zur Optimierung der Ausbildung auszuarbeiten. Dazu wurden prüfungsbezogene Risiken in der Praxis mittels Fehlzustandsart- und –auswirkungsanalyse (FMEA) analysiert sowie von den Prüfern empfundene Probleme während der Ausbildung und in der Praxis mittels Fragebogen identifiziert. Die Ergebnisse der FMEA haben gezeigt, dass während des gesamten Prozesses Fehler auftreten können, für die Gegenmaßnahmen getroffen und implementiert werden sollten. Aus den Hinweisen und Erkenntnissen wurden einige Empfehlungen zur Optimierung der Ausbildung erarbeitet. A1 - Heckel, Thomas A1 - Casperson, Ralf T1 - Aktuelle Entwicklungen der Wirbelstrom- und Ultraschallprüfung an verlegten Eisenbahnschienen N2 - Die kombinierte Schienenprüfung mit Wirbelstrom und Ultraschall ermöglicht eine ganzheitliche Detektion von Inhomogenitäten im Kopf und Steg der Schiene und gewährleistet das Auffinden sowohl von Volumen-als auch Oberflächeneffekten wie z. B. Head Checks. Für die verbesserte Detektion von Rollkontaktermüdung (rolling contact fatique, RCF) und den daraus resultierenden Schienenfehlern (Head-Checks, Squats, Riffel, …) wurde für die Wirbelstromprüfung der Prüfbereich auf die gesamte Rollkontaktfläche erweitert. Der Fokus liegt im Neudesign der Wirbelstrom-Prüfvorrichtung und der damit verbundenen normgerechten Auslegung der Konstruktion für die fahrzeugspezifischen Gegebenheiten. Ferner wurde die Anwendersoftware des kombinierten Prüfsystems vereinheitlicht. Um die wirtschaftliche Effektivität unsere Prüftechnik zu steigern, wird die Multiplex-Gerätetechnologie in die automatisierte Schienenprüfung eingeführt. Damit wird der freizuhaltende Einbauraum innerhalb der Fahrzeuge sowie Kabelwege deutlich reduziert. Durch den Übergang zu der Multiplextechnologie wurde weiterhin die Stromaufnahme um mehr als 50% reduziert. Different methods such as plasma oxidation, chemical or electrolytic etching and chemical vapor deposition have been investigated to increase, for example, the bonding strength. The present study deals with the functionalization of the fiber surface based on laser-induced periodic surface structures (LIPSS). They can be characterized as a modulation of the surface topography on the nano- and microscale that results from the irradiation of the surface with linearly polarized laser radiation close to the ablation threshold. According to their spatial period, LIPSS are classified into low-spatial frequency LIPSS (LSFL) and high-spatial frequency LIPSS (HSFL). The great potential of both types of LIPSS structures regarding functional surface properties was demonstrated in numerous investigations. The objective of the present study was the homogenous manufacturing of both types of LIPSS on large areas of carbon fiber arrangements without damage. The results are discussed based on a detailed analysis of the topographic and chemical surface properties. T1 - Simulation von mechanisierten Eisenbahnschienenprüfungen mit Ultraschall N2 - Bei der mechanisierten Prüfung von verlegten Eisenbahnschienen mit Schienenprüfzügen werden große Prüfdatenmengen mit einer komplexen Prüfkopfanordnung in kurzer Zeit bei hohen Prüfgeschwindigkeiten aufgenommen. Der Typ und der Zustand des Gleises kann während der Prüfung in schnellem Wechsel streckenabhängig variieren, z.B. unterschiedliche Schienentypen oder Abnutzung sowie beim Durchfahren von Weichenstraßen. Der logistische Aufwand diese Prüfung insgesamt durchzuführen ist dabei sehr hoch und eine Wiederholung der Prüfung in einem kurzen Zeitintervall nicht möglich. Es werden während der Prüfdatenaufnahme bereits Datenreduktionsverfahren, Rekonstruktionsverfahren und Online-Parametervariationen eingesetzt, um die Ergebnisdaten zum einen in Ihrer Menge zu begrenzen zum anderen auf die lokalen Geometrie- und Zustandsänderungen des Prüfobjektes Schiene zu reagieren. Im Regelwerk EN 16729-1 „Zerstörungsfreie Prüfung an Schienen im Gleis -Teil 1: Anforderungen an Ultraschallprüfungen und Bewertungsgrundlagen“ werden Bezugsreflektoren definiert, um die Leistung automatisierter Ultraschallsysteme für die Schienenprüfung zu verifizieren. Um die Einflüsse der komplexen Randbedingungen der mechanisierten Schienenprüfung auf das Prüfergebnis evaluieren zu können, wurde ein Softwarewerkzeug erarbeitet, mit dem eine Prüffahrt auf einer Schiene simuliert werden kann. Basierend auf der Datenaufnahme mit dem Glassy-Rail-Diagramm können alle relevanten Prüfparameter und Artefakte in Schienen sowie Störeinflüsse bei der Simulation berücksichtigt werden. Das Papier weist oben die Skizze eines Prismas, im mittleren Bereich am rechten Rand die Beschriftungen „violett/blau“, „weiß“ sowie „gelb/roth“ auf. Interessanterweise ist die Aufschrift „violett/blau“ dem eher rötlichen, die Aufschrift „gelb/roth“ dem eher bläulichen Querstreifen zugeordnet. Die rechte untere Ecke ist mit „Sk“ (Seebeck?) gekennzeichnet. Sowohl die Beschriftungen als auch die Skizze wurden mit einem Graphitstift ausgeführt. One of the promising techniques for the detection and characterisation of such damages is based on ultrasonic guided wave propagation and analysis. However, the multimodal nature and dispersive behaviour of these waves make their analysis difficult. Various signal processing techniques have been proposed for easier interpretation of guided wave signals and extraction of the necessary information about the damage. One of them is the wavenumber mapping which consists of creating a cartography of the wavenumber of a propagating mode over an inspected area, using a dense wavefield acquisition measured for example with a scanning laser Doppler vibrometer. This technique allows both the quantification of the in-plane size and the depth of damage, for example, impact-induced delamination in composite laminates. In this contribution, wavenumber mapping is applied to a delaminated aluminium-CFRP composite structure which corresponds to composite-overwrapped pressure vessels used for storing gases in aerospace and automotive industries. The analysis of experimental data obtained from measurements of guided waves propagating in an aluminium-CFRP composite plate with impact-induced damage is performed. The output of the imaging is a three-dimensional representation of the delamination induced by the impact. Good agreement between conventional ultrasonic testing and guided wave damage mapping can be found. Currently, FTIR or Raman spectroscopy are mainly applied for the analysis of microplastic particles (MP) in environmental samples. However, these methods have great difficulties in determining metrologically traceable MP values, especially with regard to the limiting values, as preferred in regulation. Therefore, we developed a systematic and fast thermoanalytical method called TED-GC-MS (thermal extraction desorption gas chromatography mass spectrometry), which determines mass contents. Now the current goal is the determination of its process parameters. Jedoch wurde beim Schweißen von Duplexstählen eine Verschiebung des Gleichgewichts zwischen Austenit- und Ferritanteil hin zu höheren Ferritgehalten und damit negativen mechanischen und Korrosionseigenschaften beobachtet. Um diesem Umstand entgegenzuwirken, werden häufig spezielle Schweißzusätze mit höheren Nickelanteilen genutzt, welche bei dickwandigen Strukturen jedoch nicht über die gesamte Schweißnahttiefe nachgewiesen werden können. Dies führt zu unterschiedlichen Eigenschaften und einer höheren Korrosionsanfälligkeit im Wurzelbereich. Eine Möglichkeit die gleichmäßige Verteilung von Legierungselementen innerhalb der Naht zu gewährleisten, bietet die Erzeugung von Pufferschichten mittels Laser-Pulver-Auftragschweißen (LPA). In diesem Zusammenhang wurden Duplexpulver mit unterschiedlichen Nickelgehalten und verschiedene LPA-Prozessparameter untersucht, die eine ebene Schichtbildung mit verringerten Ferritgehalten ermöglichen. In einem zweiten Schritt konnte nachgewiesen werden, dass das Laserstrahlschweißen der beschichteten Kanten bei üblichen Prozessparametern möglich ist und zu fehlerfreien Schweißnähten mit ausgeglichenem austenitisch- ferritischem Gefüge führt. Jedoch wurde beim Schweißen von Duplexstählen eine Verschiebung des Gleichgewichts zwischen Austenit- und Ferritanteil hin zu höheren Ferritgehalten und damit negativen mechanischen und Korrosionseigenschaften beobachtet. Um diesem Umstand entgegenzuwirken, werden häufig spezielle Schweißzusätze mit höheren Nickelanteilen genutzt, welche bei dickwandigen Strukturen jedoch nicht über die gesamte Schweißnahttiefe nachgewiesen werden können. Dies führt zu unterschiedlichen Eigenschaften und einer höheren Korrosionsanfälligkeit im Wurzelbereich. Eine Möglichkeit die gleichmäßige Verteilung von Legierungselementen innerhalb der Naht zu gewährleisten, bietet die Erzeugung von Pufferschichten mittels Laser-Pulver-Auftragschweißen (LPA). In diesem Zusammenhang wurden Duplexpulver mit unterschiedlichen Nickelgehalten und verschiedene LPA-Prozessparameter untersucht, die eine ebene Schichtbildung mit verringerten Ferritgehalten ermöglichen. In einem zweiten Schritt konnte nachgewiesen werden, dass das Laserstrahlschweißen der beschichteten Kanten bei üblichen Prozessparametern möglich ist und zu fehlerfreien Schweißnähten mit ausgeglichenem austenitisch- ferritischem Gefüge führt. The goodness of assumed distribution function plays a significant role in the inferential statistics to predict the population properties by using limited test data. Often, GAUSSian and WEIBULL probability nets are empirical methods used to validate the distribution function; Anderson-Darling and KolmogorovSmirnov tests are the mostly favorable approaches for Goodness of Fit. However, the different approaches used to determine the parameters of distribution function lead mostly to different conclusions for safety assessments. In this study, six different methods are investigated to show the variations on the rates for accepting the composite pressure vessels according to GTR No. 13 life test procedure. The six methods are: a) NormLog based method, b) Least squares regression, c) Weighted least squares regression, d) A linear approach based on good linear unbiased estimators, e) Maximum likelihood estimation and f) The method of moments estimation. In addition, various approaches of ranking function are considered. In the study, Monte Carlo simulations are conducted to generate basic populations based on the distribution functions which are determined using different methods. Then the samples are extracted randomly from a population and evaluated to obtain acceptance rate. Here, the “populations” and “samples” are corresponding to the burst strength or load cycle strength of the pressure vessels made from composite material and a plastic liner (type 4) for the storage of hydrogen. To the end, the results are discussed, and the best reliable methods are proposed. One of the promising techniques for the detection and characterisation of such damages is based on ultrasonic guided wave propagation and analysis. However, the multimodal nature and dispersive behaviour of these waves make their analysis difficult. Various signal processing techniques have been proposed for easier interpretation of guided wave signals and extraction of the necessary information about the damage. One of them is the wavenumber mapping which consists of creating a cartography of the wavenumber of a propagating mode over an inspected area, using a dense wavefield acquisition measured for example with a scanning laser Doppler vibrometer. This technique allows both the quantification of the in-plane size and the depth of damage, for example, impact-induced delamination in composite laminates. In this contribution, wavenumber mapping is applied to a delaminated aluminium-CFRP composite structure which corresponds to composite-overwrapped pressure vessels used for storing gases in aerospace and automotive industries. The analysis of experimental data obtained from measurements of guided waves propagating in an aluminium-CFRP composite plate with impact-induced damage is performed. The output of the imaging is a three-dimensional representation of the delamination induced by the impact. Good agreement between conventional ultrasonic testing and guided wave damage mapping can be found. However, comparatively little is known about the evolutionary origins of immunity in social insects. We tackle this by identifying immune genes from 18 cockroach and termite species, spanning a gradient of social lifestyles. Termites have undergone contractions of major immune gene families during the early origin of the group, particularly in antimicrobial effector and receptor proteins, followed by later re-expansions in some lineages. In a comparative gene expression analysis, we find that reproductive individuals of a termite invest more in innate immune regulation than other castes. When colonies encounter immune-challenged nestmates, gene expression responses are weak in reproductives but this pattern is reversed when colony members are immune-challenged individually, with reproductives eliciting a greater response to treatment than other castes. Finally, responses to immune challenge were more comprehensive in both subsocial and solitary cockroaches compared to termites, indicating a reduced overall ability to respond to infection in termites. Our study indicates that the emergence of termite sociality was associated with the evolution of a tapered yet caste-adapted immune system. T1 - Infection stage and pathogen life cycle determine collective termite behaviour N2 - Social insects nesting in soil environments are in constant contact with entomopathogens and have evolved disease resistance mechanisms within a colony to prevent the occurrence and spread of infectious diseases. Among these mechanisms: mutual grooming reduces the cuticular load of pathogens, and burial of cadavers and cannibalism can prevent pathogens from replicating within the group. We explored how the rate and type of collective behavioural response is determined by stepwise infection dynamics operating at the level of the individual. Specifically, we infected the eastern subterranean termite Reticulitermes flavipes with different types of infectious particle and infection route of the entomopathogenic fungus Metarhizium anisopliae and recorded behavioural responses of nestmates to individuals at different times during the progression of infections. As expected, termites groomed conidia-exposed individuals significantly more than controls. Interestingly, grooming was significantly elevated after fungal germination than before, suggesting that pathogen growth cues act as strong stimulators of allogrooming. Conidia-exposed termites were cannibalized, but only after they became visibly ill. By contrast, termites did not groom blastospore-injected individuals more than controls at any time-point following infection. Instead, we found that blastospore-injected individuals were continually cannibalized at a low-level following injection with either viable or heat-killed blastospores, with a marked increase in cannibalism after termites injected with viable blastospores became visibly ill and were close to death. Together, these findings point to the importance of host condition as a cue for social hygienic behavior, and that the host itself appears to emit essential sickness cues that act as targets for its own sacrifice. This demonstrates that termites have independently evolved to both identify and destructively respond to sickness. Understanding the proximal and ultimate drivers of this major evolutionary transition represents an important goal in biology. One outstanding question concerns the evolution of termite immunity, which is thought to have undergone broad-sweeping adaptations in order to enhance group-level immune protection. To understand the evolutionary origins of termite immunity, we conducted qualitative and quantitative transcriptome analyses along a gradient of sociality. Firstly, we aimed to identify large-scale genetic shifts in immune traits linked to eusociality by comparing immune gene repertoires in solitary and subsocial cockroaches and across a range of eusocial termite lineages. Secondly, we compared the responses of a solitary cockroach, a subsocial wood-roach and different castes of a lower termite species to a non-specific immune challenge, in order to understand how sociality may have influenced the evolution of immune gene regulation. Firstly, we found that termites have a broadly representative repertoire of canonical immune genes as compared to gregarious cockroaches and subsocial wood-roaches. Secondly, with respect to immune challenge, the solitary cockroach and the subsocial wood-roach displayed a similarly comprehensive induced response, while the termite response was considerably dampened by comparison and strongly influenced by caste; with reproductives displaying a generally higher constitutive level of immune-gene expression compared to sterile castes. In summary we did not find termite eusociality to be associated with significant changes in immune gene diversity, but rather to be linked with significant modifications to the regulation of immunity following the origin of division of labour. Particularly, squeeze casting production of Al-Si alloys is employed in the conception of metal matrix composites (MMC) for combustion engines. Such materials are of a high interest since they allow combining improved mechanical properties and reduced weight and hence improve efficiency. We investigate two types of MMCs, which can be potentially used for production of combustion engine pistons: 1) a near-eutectic cast AlSi12CuMgNi alloy reinforced with 15%vol. Al2O3 random planar oriented short fibers and 2) the same alloy reinforced with 7%vol. Al2O3 random planar oriented short fibers + 15%vol. SiC particles. Complex 3D microstructure of the samples in as-cast condition, consisting of four and five phases (Al matrix, eutectic Si, intermetallics, Al2O3 fibers and SiC particles) was investigated by synchrotron computed tomography (CT). Advanced methods based on machine learning were applied for segmentation of all phases. This allowed extracting quantitative information such as volume fraction, shape and interconnectivity of every phase. In-situ compression tests during neutron diffraction experiments were used to track the load transfer among phases, while CT on pre-strained samples to monitor and quantify damage. A micromechanical model was developed to simulate the evolution of the internal stress of each phase during uniaxial compression. We showed that in composites with the Al2O3 fiber mat plane perpendicular to the load axis the Al-alloy matrix presents a large hydrostatic stress component, i.e. undergoes compression also in the direction transverse to the external load. This feature holds with and without the addition of SiC ceramic reinforcement and is absent in the case when the Al2O3 fiber mat plane is parallel to the load axis. We show, that the intermetallics play a decisive role at very high loads, when all other reinforcement phases suffer (extensive) damage. The addition of SiC particles does alleviate the load on the Al2O3 fibers, on the eutectic Si, and on the intermetallic phases in both cases of parallel and orthogonal (to the load axis) Al2O3 fiber orientation. Apart from the beneficial addition of ceramic reinforcement, the presence of intermetallic phase and eutectic silicon, forming an interconnected network even at high loads, when single particles break, confers peculiar properties to these multi-phase composites. T1 - The importance of the horizontal stresses on the bearing capacity of a foundation pile N2 - In case one wants to predict or design the bearing capacity of a foundation pile and there are no possibilities to perform an in-situ test, such as a Cone Penetration Test, the pile bearing capacity is in most cases estimated with analytical formulas. The most known and used method is the Meyerhof method published some decades ago. There are also other design methods such as derived from a certain failure mechanism around the pile tip, which is, in most cases, wedge failure mechanism. This failure mechanism was originally developed for a shallow (infinite) strip foundation, though. Therefore, it represents a plane failure mechanism. Numerical simulations on loaded foundation piles performed with the Plaxis software Show however, that the failure mechanism of a foundation pile represents a far more complex threedimensional failure mechanism around the pile tip. In addition, the existing analytical methods for foundation piles are based on the vertical stresses in the soil, as if the failure mechanism is the same as of a shallow foundation. Numerical simulations, performed in Plaxis show that, not the vertical, but the horizontal stresses, play an important role on the pile bearing capacity. Plaxis represents the stresses in the soil by using the procedure. So, different horizontal soil stresses are obtained for different values of the lateral earth pressure coefficient. The results show that the pile tip bearing capacity depends strongly on the horizontal stresses in the soil, but only for. The same results were observed by using a Material Point Method (MPM). Consequently, the analytical methods should estimate the pile bearing. A1 - Gorbushina, Anna T1 - Exploring the genome of the black yeast Knufia petricola N2 - Black yeasts form a polyphyletic group of Ascomycota that colonize bare surfaces like rocks, facades of buildings, and solar panels. Their protective adaptations enable an adequate response to fluctuating and diverse temperature, water and UV radiation stresses. Together with bacteria and algae they form sub-aerial biofilms (SAB) this way discoloring and weathering the surfaces they grow on. Strain A95 of Knufia petricola (Eurotiomycetes, Chaetothyriales) displays both typical yeast-like cell growth and constitutive dihydroxynaphthalene (DHN) melanogenesis. Along with the cyanobacterium Nostoc punctiforme as photobiont, it is already used in a model system for studying SAB formation and bio-weathering. Applying the recently developed tools for the generation of deletion mutants will allow to define gene functions and to identify genes critical for abiotic and biotic interactions. We present a chromosome-level genome assembly and annotation for K. petricola A95. The genome was assembled with MaSuRCA using a hybrid assembly approach of Illumina MiSeq and PacBio SMRT sequencing data. The resulting assembly consists of 17 contigs including the complete mitochondrial genome and five complete chromosomes. It shows indication of repeat-induced point mutations (RIP). Supported by RNA sequencing data from eight different growth conditions, 10,994 genes were predicted with the BRAKER2 pipeline. Functional annotation of genes was obtained from general functional annotation databases and the fungal specific database FungiPath. Comparative analyses are in progress to identify genes specific to black yeasts, that may facilitate the survival on exposed surfaces. In sum, the genome sequence of K. petricola is a valuable resource to gain insight into the protein inventory and functional pathways of extremotolerant and symbiosis-capable fungi. In these niches, MCF have evolved mineral-weathering and symbiotic capabilities as well as mechanisms to cope with multiple stresses such as solar irradiation, temperature extremes and low water activity. Due to their stress tolerance these ascomycetes are prominent in modern terrestrial ecosystems – like man-made material surfaces from roof to solar panels. MCF interactive capabilities support their facultative symbiotic relationships with cyanobacteria and ensure their rock-weathering geochemical activity. Using the rock-inhabiting fungus K. petricola A95 (Chaetothyriales), we developed transformation protocols and deleted genes responsible for production of the protective pigments melanins and carotenoids. To confirm that the mutant phenotypes were not due to hidden mutations, melanin synthesis was restored by complementing the mutants with the respective wild type genes. Strains of K. petricola carrying gene variants for fluorescent proteins EGFP and DsRed are available. We successfully labelled the cytoplasm, nuclei, peroxisomes and mitochondria. Targeted and ectopic integrations result in stable transformants suitable for further phenotypical characterization. As K. petricola is a non-pathogenic fungus with all characteristic features of MCF, including meristematic growth, melanized cell-walls, extracellular polymeric substances and extensive pigment production, our results will shed light on protective role of pigments during cell wall maturation and oxidative stress defence in rock-inhabiting MCF. Genes involved in environmental sensing or substrate and phototroph interactions are currently targeted. With the help of a mutant collection and fluorescently labelled K. petricola we will be able to investigate interactions of MCF with environmental stressors, mineral substrates, soil matrices and phototrophic symbionts. Together with bacteria and algae they may establish sub-aerial biofilms and cause weathering of the surfaces they grow on. Their impressive survival abilities as well as their constitutive protective pigmentation and cluster-like microcolony organisation are similar in environmental isolates as well as in heat-tolerant opportunistic pathogens of animals and humans. The exact genetic properties that ensure their survival in extreme environments can be studied if some black fungi were amenable to genetic manipulations. We selected the rock-inhabiting fungus Knufia petricola (class Eurotiomycetes, order Chaetothyriales) that grows moderately in axenic culture and exhibits all the characteristics of black yeasts such as yeast-like cell growth, absence of reproductive structures and constitutive dihydroxynaphthalene (DHN) melanogenesis. For this environmental strain we developed protocols to efficiently generate and transform protoplasts resulting in stable homokaryotic transformants by targeting genes involved in pigment synthesis and expressing fluorescent reporter genes. Hence, endogenous and foreign genes can be expressed from episomal AMA1-containing plasmids and genome-integrated DNA constructs. Moderate rates of homologous recombination allow for both ectopic and targeted integrations. CRISPR-Cas9 was further validated as a strategy for obtaining selection marker-free mutants and silencing via RNA interference as an approach to study essential genes. For practical and economic reasons, high-quality, costly measurements are currently only carried out sparsely, both in time and space, i.e., measurement data are collected in single day campaigns at selected locations only. Recent developments in sensor technology enable cost-efficient gas monitoring in real-time for long-term intervals. This knowledge of contaminant distribution inside the industrial environment would provide means for better and more economic control of air impurities, e.g., the possibility to regulate the workspace’s ventilation exhaust locations, can reduce the concentration of airborne contaminants by 50%. This paper describes a concept proposed in the project “Robot-assisted Environmental Monitoring for Air Quality Assessment in Industrial Scenarios“ (RASEM). RASEM aims to bring together the benefits of both – low- and high-cost – measuring technologies: A stationary network of low-cost sensors shall be augmented by mobile units carrying high-quality sensors. The supply of spare parts for repair and replacement of lost equipment would be one key pillar of such a strategy. Bearing in mind the absolute distance and flight trajectories for manned missions to Mars, supplying spare parts from Earth is impossible. Thus, in space manufacturing remains the only option for a timely supply. With a high flexibility in design and the ability to manufacture ready to use components directly from a computer aided model, additive manufacturing technologies appear extremely attractive. For metal parts manufacturing the Laser Beam Melting process is the most widely used additive manufacturing process in industrial application. Fungi are known to enhance rock weathering, although the exact mechanisms are not completely understood. These natural processes can be simulated by reproducible experimental systems. Particularly attractive are genetically amenable rock-weathering fungi, where certain traits can be knocked-out to identify weathering-relevant functions. Here, we studied the effect of the rock-inhabiting fungus Knufia petricola A95 and its melanin-deficient mutant (ΔKppks) on the weathering of a Fe-bearing olivine. We examined the olivine dissolution kinetics and looked at the olivine-fungus interface. For the former we inoculated olivine powder with the fungus in batch and mixed flow reactors and analysed the medium solution by ICP-OES, for the latter biofilm-covered olivine sections from flow-through reactors were analysed by FIB-TEM. In general olivine dissolution was lower when olivine was incubated without fungi: the abiotic olivine dissolution rates were lowest and the surface of the abiotically reacted olivine sections showed no etching. Various chemical analyses show Fe (oxyhydr)oxide precipitation on top of the abiotically reacted olivine, indicating its role in inhibiting olivine dissolution. Both the wild type (WT) and ΔKppks solubilised and bound significant amounts of Fe released by olivine dissolution. Fe (oxyhydr)oxide precipitation was thus reduced, explaining the enhanced olivine dissolution in their presence. Particularly efficient at preventing Fe precipitation were attached WT cells: the inhibition of olivine dissolution almost completely disappeared. The attachment capacity of the WT is most likely caused by the production of WT-specific extracellular polymeric substances. Our presented experimental systems allowed the precipitation of Fe (oxyhydr)oxides and included a rock-inhabiting fungus and thus simulated weathering processes relevant to natural ecosystems. Still very important issues remain nowadays, hampering a completely autonomous production of parts and even restricting the freedom of design by means of these technologies. However, resorption of modern materials for bone grafting (hydroxyapatite (HA) and tricalcium phosphate (TCP)) is reported, in some cases, to be not enough, this is why the search for more soluble compounds compared to HA and TCP looks very perspective. A possible way to increase ceramics solubility leads to partial substitution of Ca2+ -ions in Ca3(PO4)2 by alkali cations, like Na+ or/and K+. Improvement of solubility stems from decreasing lattice energy of a substituted phase, as well as the increase in hydration energy of the ions releasing from the phase to ambient solution. From this viewpoint, bioceramics based on compositions from Ca3(PO4)2 - CaKPO4 - CaNaPO4 ternary system seems to be prospective for bone replacement and grafting in the sense of resorption properties. At the same time, one should bear in mind that solubility level (resorbability) is governed not only by reduction of lattice energy but also by microstructure features. Grain sizes and porosity contribute much to dissolution rate making the study of sintering of the ceramics mentioned above highly important. To control Ca3(PO4)2 - CaKPO4 - CaNaPO4 based ceramic microstructure it is necessary to know possible phase transformations in the system and the way to manage microstructure by sintering schedule or sintering process. In this work, an isothermal section for phase diagram of Ca3(PO4)2 - CaKPO4 - CaNaPO4 ternary system is studied with several techniques. According to the XRD of quenched samples, this phase triangle has four single-phase areas at 1200˚C (Figure 1). It was shown that single-phase CaK0.6Na0.4PO4 cannot be sintered to full-dense ceramics by conventional sintering regardless time-temperature schedule. Two-step sintering technique, beneficial in the case of HA-ceramics, was unsuccessful in all cases of calcium-alkali phosphate compositions. However, field-assisted sintering techniques like, e.g. Spark Plasma Sintering (SPS), can overcome this problem due to significant impact on grain boundary diffusion. In connection with this fact, grains grow much slower retaining sintering process in a pore control regime. In this work CaK0.6Na0.4PO4 low-porous ceramics was also fabricated by FAST-methods of sintering. Moreover, other alternative sintering techniques, such as reaction sintering, may be useful in accelerating ions diffusion but stopping excessive grain growth.Strength properties of ceramics were evaluated by B3B-testing, micro- and nanoindentation techniques. Fracture toughness also becomes higher with potassium content increase, guiding porosity level. Resorption properties of sintered ceramics were studied in different solutions with pH=5 and 7.4. T1 - Raman band widths of anhydrite II reveal the burning history of high-fired medieval gypsum mortars N2 - The use of high-fired gypsum as binder for masonry and joint mortars or stuccowork in Central Europe in the Early and High Middle Ages was a regional specific as it depended on local gypsum deposits. The calcination technology possible at the time resulted in an assemblage of calcium sulphate phases dehydrated to different degrees and partly thermally damaged accessory minerals of the raw gypsum. Because of the absence of medieval textbooks, the observation of high-temperature, low-pressure mineral transformations and the correlation of phases coexisting in not hydrated binder relicts in the gypsum matrix to the mineralogy of the raw material and the burning conditions constitute the only source to the historical technological know-how. The CaSO4–H2O system consists of five crystalline phases, which can be discriminated by structural analysis methods, such as Raman spectroscopy, due to obvious differences in their spectroscopic data: gypsum (CaSO4 ⋅ 2 H2O), bassanite (hemihydrate, CaSO4 ⋅ ½ H2O), anhydrite III (CaSO4), anhydrite II (CaSO4), and anhydrite I (CaSO4). Only recently, it was possible to demonstrate that small spectroscopic variations exist also within the relatively large stability range of anhydrite II from approx. 180°C to 1180°C: all Raman bands narrow with increasing burning temperature applied in the synthesis from gypsum powder. The determination of band widths of down to 3 cm-1 and differences between them of a few tenths of a wavenumber is not a trivial task. Thus, this contribution discusses peak fitting and strategies for correction of instrument-dependent band broadening. Raman maps of polished thin sections of gypsum mortars provide access to the burning histories of individual remnant thermal anhydrite grains and enable the discrimination of natural anhydrite originating from the gypsum deposit. This novel analytical method was applied to samples from medieval South Tyrolean stucco decorations and sculptures. Beyond that, Raman microspectroscopy was employed for following pyrometamorphic reactions in natural impurities of the raw material. In the presented examples mineral thermometry indicates process temperatures above 800°C: the breakdown of magnesium-rich chlorite led to the formation of forsterite Mg2SiO4, while the thermal decomposition of dolomite CaMg(CO3)2 yielded – after hydration and carbonation – magnesite MgCO3, CaCO3 polymorphs and magnesian calcite. Lower burning temperatures, which leave the accessory minerals in their pristine form, can be traced by measuring the spectra of anhydrite crystalites in grains of firing products and evaluating Raman band widths. Throughout the applications of this analytical method so far, calcination temperatures ranging from approx. 600°C to 900°C were determined. Besides serving as an anchor point for subsequent functionalization, the surface chemistry of these particles also fundamentally influences their interaction with the surrounding medium and can have a significant effect on colloidal stability, particle uptake, biodistribution, and particle toxicity in biological systems. Moreover, functional groups enable size control and tuning of the surface during the synthesis of particle systems. For these reasons, a precise knowledge of the chemical nature, the total number of surface groups, and the number of groups on the particle surface that are accessible for further functionalization is highly important. Die Glimmentladungs-Massenspektrometrie (GD-MS) stellt hierfür eine schnelle Multielementmethode dar. Aufgrund ihrer höheren Anregungs- und Ionisationsenergien ist die Bestimmung von Nichtmetallen eine weit größere Herausforderung. Außerdem fehlt es an für die direkte Feststofftechnik notwendigen Kalibrierstandards z.B. in Form zertifizierter Referenzmaterialien. Für die quantitative Bestimmung von Verunreinigungen wurden mit Standards dotierte Pulverpresslinge und dotierte, gesinterte Materialien auf ihre Eignung als Kalibrierstandards für die GD-MS untersucht. Dabei kamen unterschiedliche Plasmabedingungen (gepulster/ kontinuierlicher Betrieb; verschiedene Plasmagase) zum Einsatz. Ein Überblick über analytische Kenngrößen und Grenzen der verwendeten Kalibrierstrategien wird gegeben. Zur Validierung des Verfahrens wurde die hochauflösende Continuum Source Absorptionsspektrometrie (HR-CS-MAAS) verwendet. Die analytische Nutzung der Molekül-absorptionsspektrometrie zur Bestimmung von Halogenen basiert auf der Bildung insitu-generierter zweiatomiger Moleküle (AlCl, GaF). Die entstehenden Molekülspektren werden im Echelle-Spektrometer aufgespalten und mittels CCD-Array detektiert, was eine sehr empfindliche Messung und präzise Quantifizierung ermöglicht. Some of these comparisons deal directly with the preparation and characterization of monoelemental solutions or with topics, closely related. The importance of monoelemental solutions lies in the fact that almost every measurement in inorganic analysis relies on the comparison with either a reference material, or references in form of solutions, usually (mono)elemental solutions. All quantitative measurement approaches, e.g. isotope dilution or standard addition, need an accurate reference solution made from a well characterized reference material, prepared under full gravimetric control. These primary (monoelemental) solutions do not only serve as arbitrary references/calibration solutions, but they also link up measurement results to the International System of units (SI), this way establishing the so-called metrological traceability to a measurement unit of the SI. Without such solutions on the highest possible level of accuracy and with the smallest possible associated uncertainties (for e.g. element content and/or impurities), an analysis itself can never be as good as it could be with appropriate reference solutions. Neben den Risiken durch Schadstoffeinträge gibt es in Deutschland Regionen mit einem stark erhöhten Nährstoffüberschuss. Um die Grundwasserbelastung zu verhindern wurde u.a. das Düngerecht verschärft, was die Flächenkonkurrenz zwischen Klärschlamm und Wirtschaftsdünger vergrößern wird. Es ist davon auszugehen, dass sich die neuen Regelungen im Düngerecht und der Klärschlammverordnung massiv auf die landwirtschaftliche Verwertung von Klärschlamm und damit auf die Kreislaufwirtschaft verschiedener Nährstoffe auswirken werden.
