The exploitation of EV requires reliable measurements, which is currently very difficult, as most EV are smaller than 200 nm. At present, flow cytometry (FCM) is the most appropriate technique for EV analysis in biological samples, as FCM is readily available in many clinical laboratories and allows to identify cell-specific EV at high throughput. However, due to technical variations between different FCM instruments, EV concentration measurements are currently not well comparable between most laboratories. Therefore, EV reference materials and standardized reference methods are urgently needed to calibrate flow rate, light scattering intensity, and fluorescence intensity of FCM in the sub-micrometer size range. This requires a better matching of the optical properties of calibration beads and EV as can be realized with current polystyrene calibration beads. The EMPIR project 18HLT01 “MetVes II” aims to develop synthetic reference materials and traceable measurement methods to standardize EV measurements. The reference materials should resemble EV properties, so that calibrations are reliable and do not require a change of acquisition settings. Hence, the reference materials should contain particles with a traceable number concentration in the range of 109–1012 particles/mL to calibrate flow rate, a traceable size with discrete diameters between 50–1000 nm and a refractive index (RI) in the range of 1.37–1.42 to calibrate scattering intensity, and a traceable fluorescence intensity between 100–100,000 molecules of equivalent soluble fluorochromes (MESF). At BAM, various approaches to prepare such low-RI nanometer-sized reference materials will be studied, preliminary results of the primary characterization of these candidate reference particles will be presented, and possible applications besides FCM-based EV detection will be outlined. The goodness of assumed distribution function plays a significant role in the inferential statistics to predict the population properties by using limited test data. Often, GAUSSian and WEIBULL probability nets are empirical methods used to validate the distribution function; Anderson-Darling and Kolmogorov-Smirnov tests are the mostly favorable approaches for Goodness of Fit. However, the different approaches used to determine the parameters of distribution function lead mostly to different conclusions for safety assessments. In this study, six different methods are investigated to show the variations on the rates for accepting the composite pressure vessels according to GTR No. 13 life test procedure. The six methods are: a) Norm-Log based method, b) Least squares regression, c) Weighted least squares regression, d) A linear approach based on good linear unbiased estimators, e) Maximum likelihood estimation and f) The method of moments estimation. In addition, various approaches of ranking function are considered. In the study, Monte Carlo simulations are conducted to generate basic populations based on the distribution functions which are determined using different methods. Then the samples are extracted randomly from a population and evaluated to obtain acceptance rate. Here, the “populations” and “samples” are corresponding to the burst strength or load cycle strength of the pressure vessels made from composite material and a plastic liner (type 4) for the storage of hydrogen. Additionally, corrosion fatigue experiments (ambient pressure, technically clean CO2, saline aquifer water of Stuttgart Aquifer) were performed using specimen of X46Cr13 (1.4043, AISI 420C) with regard to the influence of the roughness of technical surfaces on the number of cycles to failure at different stress amplitudes. Specimen of Duplex stainless steel X2CrNiMoN22-3-2 (1.4462) for corrosion fatigue experiments were provided with technical surfaces after machining as well as polished surfaces. Results were obtained at load amplitudes ranging from 175 MPa to 325 MPa in the geothermal brine of the Northern German Basin at 98 °C. The main precipitation phases on the surface as well as within pits reveal carbonates or hydroxides such as siderite (FeCO3) and ferrous hydroxide goethite (FeOOH) independent of the original surface roughness. Corrosion rates for polished and technical surfaces were below 0.005 mm/year compared to corrosion rates of 0.035 mm/year after shot peening. Specimen with technical surfaces tested at high stress amplitudes (>275 MPa) lasted longer (cycles to failure: P50% at Sa 300 MPa=5x105) than specimen with polished surfaces (cycles to failure: P50% at Sa 300 MPa=1.5x105). This behavior is emphasized by the slope coefficient (technical surfaces k = 19.006, polished surfaces k=8.78) meaning earlier failure for polished at high stress amplitude Sa. Although rather low scatter ranges (technical surface: TN=1:1.35, polished surface: TN=1.1.95) indicate no change in failure mechanism it may be assumed that at low stress pitting is the initiating crack growth process whereas at high stress amplitudes the formation of micro cracks is reason for crack propagation and failure. To better understand its behaviour differently heat treated coupons of 1.4542 and for comparison X20Cr13 and X46Cr13 were kept in the artificial brine of the Northern German Basin at T=60 °C. Ambient pressure as well as p=100 bar for 700 h - 8000 h in water saturated supercritical CO2 and CO2-saturated synthetic aquifer Environment was applied. Fatigue tests were performed via push-pull tests with a series of 30 specimens from 150 MPa to 500 MPa (sinusoidal dynamic test loads, R=-1; resonant frequency ~ 30 Hz). FeCO3 and FeOOH are corrosion products also after dynamic corrosion tests. Martensitic microstructure offers good corrosion resistance in geothermal environment. The S-N-curve showing no typical fatigue strength and very steep slopes of possible fatigue strength for finite life. Possible influencing artefacts, such as Al-inclusions could not be correlated to early rupture despite specimens containing inclusions at the fracture surface and cross section reached lower number of cycles. Typologies of Arabic inks and their detection limits N2 - In this poster we will present some results of a research projects dealing with Arabic recipes of black inks. To improve conservation practices of Arabic manuscripts we have assessed the feasibility of the recipes collected from many Arabic sources on bookmaking and reproduced the most common inks. Ink samples were then artificially aged and analysed using an array of analytical techniques, mostly non-invasive, in order to build a database of Arabic inks and their ingredients and also to verify the detection limits of the portable equipment employed. We will show procedures and problems concerning the identification of ink types and ingredients using reflectography (in Vis, IR and UV), X-Ray Fluorescence, Infrared and Raman spectroscopy. The focus will be in particular on squid ink and mixed inks – both in the form of carbon inks with the addition of tannins and of a mixture of carbon and iron gall inks – which are particularly difficult to detect. We will also examine the results of iron gall inks made with iron filings or nails instead of vitriol, the various precursor used to extract gallic acid that are mentioned in the sources as surrogates of gall nuts. While simulations on coarse scales are practical for engineering applications, a deeper understanding of the material is gained on finer scales. This is at the cost of an increased numerical effort that can be reduced by the three methods developed and used in this work, each corresponding to one publication. The coarse spatial scale is related to fully homogenized models. The material is described in a phenomenological approach and the numerous parameters sometimes lack a physical meaning. Resolving the three-phase mesoscopic structure consisting of aggregates, the mortar matrix and the interfaces between them allow to describe similar effects with simpler models. Quarantine of potentially infested objects can be logistically challenging. Anoxia under controlled Nitrogen atmosphere is a most gentle but also time-consuming method to eradicate insect pests in all kinds of different materials. Treatment results are usually affected by duration, temperature, humidity and residual oxygen content. During a two-year research project, 34 relevant pest insect species of all developmental stages were tested in different materials (wood, paper, wool) to monitor treatment success and to determine optimum treatment parameters. Duration of treatment ranged from one to three weeks at temperatures of 20–27 °C. As expected, results showed significant differences in mortality among tested species. Highest tolerance of hypoxic conditions was found in elder larvae of Hylotrupes bajulus. Although this species is an unlikely museum pest, it may serve as an overall most tolerant reference. Anobiids and other wood boring beetles are more often an issue related to cultural heritage. A combination of three weeks exposure time at maximum 0.5% residual oxygen and 24 °C, alternatively 1% residual oxygen and 27 °C are recommended for infested artefacts. T1 - Mussel-Inspired Multivalent Linear Polyglycerol Coatings Outperform Monovalent Polyethylene Glycol Coatings in Antifouling Surface Properties N2 - Biofouling constitutes a major challenge in the application of biosensors and biomedical implants, as well as for (food) packaging and marine equipment. In this work, an antifouling surface coating based on the combination of mussel-inspired dendritic polyglycerol (MI-dPG) and an amine-functionalized block copolymer of linear polyglycerol (lPG−b−OA11, OA = oligo-amine) was developed. The coating was compared to a MI-dPG surface which was postfunctionalized with commercially available amine-terminated Polyethylene glycol (HO−PEG−NH2) of similar molecular weight. In the current work, These coatings were compared in their chemical stability, protein fouling characteristics, and cell fouling characteristics. The lPG−b−OA11-functionalized coating showed high chemical stability in both phosphate buffered saline (PBS) and sodium dodecyl sulfate (SDS) solutions and reduced the adhesion of fibrinogen from human plasma with 99% and the adhesion of human serum albumin with 96%, in comparison to the bare titanium dioxide substrate. Furthermore, the Proliferation of human umbilical vein endothelial cells (HUVECs) was reduced with 85% when the lPG−b−OA11 system was compared to bare titanium dioxide. Additionally, a reduction of 94% was observed when the lPG−b−OA11 system was compared to tissue culture polystyrene. Die Auswahl an verfügbaren Klebstoffen und Vorbehandlungsoptionen ist dabei ebenfalls vielfältig. Einige Polymere weisen darüber hinaus Eigenschaften auf, die eine Herstellung von festen und dauerhaft beanspruchbaren Klebverbindungen erschweren. Mittels Oberflächenbehandlung, z.B. durch Plasmafeinreinigung bzw. –aktivierung, können die Grenzflächern solcher Polymere - bezogen auf die Eignung als Fügeteilpartner – optimiert werden. Schnelle Prüfverfahren zur Ermittlung von Verbundeigenschaften sind somit erforderlich. Das Messprinzip Centrifugal Adhesion Testing Technologie verwendet die radial gerichtete Zentrifugalkraft als Belastungsgröße des zu testenden Verbunds, die durch Variation der Drehzahl gesteuert wird. Bei Erreichen der kritischen Beanspruchung trennt sich ein aufgeklebter Prüfstempel vom Substrat. Zeitgleich wird ein positionsbezogenes IR-Signal aus dem drehenden Rotor an das Basisgerät gesendet und zusammen mit der Drehzahl an die PC-Software übertragen. Für jede Probe wird die Bruchkraft berechnet und in Echtzeit angezeigt. Der Adhesion Analyser LUMiFrac® untersucht bis zu acht Prüfkörper unter identischen Bedingungen. Das neue Messprinzip erfüllt die Anforderungen von DIN EN 15870, DIN EN 14869-2, ISO 4624 sowie DIN EN 13144. Die Multiprobentestung ermöglicht schnelle Charakterisierungen von Klebverbindungen. So kann der Effekt einer Plasmabehandlung von Bauteilen auf die Verbundfestigkeit zeitsparend bestimmt werden. Die gezielte Veränderung unterschiedlicher Plasma-Parameter, wie beispielsweise, die Plasmazusammensetzung, der Abstand vom Plasmabrenner zur Oberfläche oder die Verweildauer unter dem Brenner, beeinflußt die Festigkeit des Klebsystems und ist dem LUMiFrac direkt quantifizierbar. Neben der Festigkeitsbestimmung von Klebverbindungen ist die Haftung von Schichtsystemen ein wichtiger Anwendungsbereich. For this purpose, in this work, a tensile module capable of uniaxial stretching and compression with up to 5 kN force was integrated into an AFM stage. The elimination of the need for sample unmounting and remounting and the resulting possibility of keeping the sample under constant mechanical load during AFM measurements not only enables a precise positioning of the area of interest but also allows for the analysis of processes in the elastic deformation regime. This methodology was demonstrated for two case studies. Scanning Kelvin Probe Force Microscopy (SKPFM) was used as the main tool to characterize the deformation behavior. Moreover, a flexible electrochemical measurement cell was used to enable electrochemical analysis by means of electrochemical impedance spectroscopy (EIS) and Linear Sweep Voltammetry (LSV) during AFM measurements at different levels of strain. The in situ AFM results are complemented by microstructure analysis by means of electron backscatter diffraction (EBSD). In the first case study, the deformation induced delamination of a thin organic coating on AA2024 T3 aluminium alloy was investigated as a function of alloy surface treatment. The formation of cracks in the insulating passive film enabled an early detection of deformation processes by means of SKPFM. The second case study focused on the comparison of corrosion and deformation behavior of conventional and additively manufactured 316 stainless steels. In comparison to the conventional 316 stainless steel, the effect of processing was clearly detectable on the additively manufactured material as zones of inhomogeneous potential, which also affected the initiation of local corrosion processes. With the increasing utilization of multi-material structures in the automotive and aerospace industry, it is of great scientific and technical interest to understand the processes leading to interface degradation and to develop novel strategies to increase corrosion and delamination resistance. The aim of this project is to develop thin epoxy-based films and their carbon nanofiller loaded composites on aluminium alloy AA2024-T3 as a model system and to investigate their interfacial stability under corrosive and coupled corrosive-mechanical load. Spin coating was used for the layer-by-layer deposition of poly[(o-cresyl glycidyl ether)-co-formaldehyde] and poly-(ethylenimine) bi-layers. Atomic force microscopy (AFM) results indicate a very homogeneous and dense film with low surface roughness. Carbon nanofillers were introduced either by mixing into the coating components or in between individual layers to control the separation between the carbon nanofillers and alloy surface. The film chemistry and barrier properties were characterized by means of spectroscopic and electrochemical methods, respectively. The degradation and delamination behavior of the epoxy-based films was characterized by means of in situ AFM corrosion experiments. The quantitative imaging (QI) mode allowed the observation of hydrogen-generation induced blister formation during exposure to corrosive electrolyte and how the local corrosion processes evolved with exposure time. Complementary energy dispersive X-ray spectroscopy (EDX) analysis was performed to correlate the corrosion behavior with the different intermetallic particle chemistries and distributions. Both alloys are single phase fcc prior to oxidation and were simultaneously exposed to different oxygen containing atmospheres at 700° C and 800° C for durations up to 288 h. Cr20Mn20Fe20Co20Ni20 showed poor oxidation resistance at high temperatures due to its high amount of manganese leading to the formation of non-protective Mn-oxide scales. In contrast, a relatively homogeneous and protective chromia layer was found to form on CrCoNi at 700° C and 800° C after each exposure time. Mass change-, SEM- and XRD analysis of the oxide layers were performed in the present study and provide a better understanding of the oxidation mechanisms as well as the impact of the alloy composition on oxidation behavior. The overarching goal of MUMMERING is the creation of a research tool that exploits the wealth of 3D imaging modalities applied in materials engineering. The focus of this work is advanced reconstruction algorithms. The primary objective is the development and implementation of the DIRECTT algorithm. Due to restrictions introduced by the sample or the imaging setup during a measurement, it is not always possible to acquire data that fulfil the Nyquist sampling criterion. The DIRECTT algorithm is intended to produce reconstructions of superior quality for such cases of limited data sets, compared to those of other available algorithms, such as algebraic iterative ones. Although DIRECTT has been developed primarily for the reconstruction of volumes from data acquired by an X-ray Computed Tomography scanner, it can also be applied to the different modalities, such as laminography. The talk describes the reconstruction principle of DIRECTT. Furthermore, reconstructions of measured data are presented and compared to reconstructions produced by other established algorithms. T1 - Analyse von Humboldt Codices aus Mizquiahuala im Vergleich zu Fragmenten in Mexiko und Paris N2 - Unter den von Humboldt in Amerika erworbenen Schriftstücken der Kolonialzeit findet sich eine Tributliste aus dem Jahr 1569, die der Otomí Bevölkerung aus dem Ort Mizquahuala im Staat Hidalgo/ Mexiko zugeordnet wird. Sie ist in Form eines Kalenders ausgeführt, Wochentage und verschiedene Produkte lassen sich unterscheiden. Eduard Seler beschreibt dieses Manuskript zusammen mit einer weiteren Tributliste, die in Inhalt und Zeichnung anders gestaltet ist, jedoch ebenfalls aus dieser Region stammt. Mit den für die Analyse von Manuskripten bewährten Untersuchungsmethoden werden Papier und Farbmittel identifiziert. Hierzu zählen Raman-, FTIR-, Röntgenfluoreszenz- und VIS- Spektroskopie. Charakteristisch für die Herstellung von schriftlichen Dokumenten in der Kolonialzeit von Amerika ist die Verwendung von indigenem Material wie Papier aus Baumrinde und Pflanzenfarben zusammen mit den aus Europa bekannten Materialien. Geklärt wird, welche Auswahl von Farbstoffen zur Herstellung der Dokumente genutzt wurde, die diese als Gruppe klassifizieren könnte. Die den beiden Fragmenten zugeordneten Farbmaterialien sowie formale und inhaltliche Aspekte lassen vermuten, dass zwei weitere Schriftstücke der Berliner Sammlung `Manuscripta Americana´, deren Herkunft nicht bekannt ist, mit ihnen in Beziehung gebracht werden können. Stilistisch gleiche Fragmente befinden sich zudem in Mexiko, Paris und den USA. Im Vergleich dieser Fragmente untereinander wird diskutiert, inwieweit die über die Materialanalyse identifizierten spezifischen Farbstoffe einen Hinweis auf eine gemeinsame lokale Herkunft zulassen. Diese Entwicklung kann durch die fluidische Anregung ermöglicht werden. Das Ziel dieser vorliegenden Arbeit war der Aufbau einer Messapparatur, die auf Schnelligkeit setzt und kontinuierlich misst. Nach der Durchführung der fluidischen Anregung wurden mit Hilfe eines Laservibrometers die Daten aufgenommen. Zur Analysierung der auf diese Weise erhobenen Messwerte wurden sie mittels der Fast-Fourier-Transformation in den Frequenzbereich transformiert. Например, при лазерной сварке со сквозным проплавлением стальной пластины толщиной 2 мм длина кратера шва может быт на 19% больше установившейся длины сварочной ванны. Установлено, что центр кратера, в котором заканчивается затвердевание жидкого металла, смещен в сторону хвостовой части кратера относительно оси теплового источника в момент прекращения его действия. N2 - A functional-analytical solution of the problem of heat conduction after the moment of switch-off of the moving heat source is presented. Different combinations of heat sources and heated bodies are considered. It is demonstrated that an additional melting of the base metal is possible after the switch-off due to the overheated weld pool metal. For example, in laser beam keyhole welding of a 2 mm thick steel plate, the crater can be 19% longer than the weld pool. It is found that the crater centre, where the solidification of the liquid metal ends, is displaced from the heat source axis at the moment of switch-off towards the weld pool tail. The rate and the direction of crystallization of the molten metal in the weld pool and the end crater differ significantly. Static tests at room temperature as well in humid and high temperature conditions are done on the coupon level. Additionally, a full-scale static and fatigue test must be performed on the complete airframe (minimum on the fuselage together with the wing). For each type-certificate the complete building block approach test program must be performed. Traditionally in Germany, the certification of sailplanes (Certification Standard 22) follows rather a family concept. A shared data base was created over the last 50 years based upon a large number of material testing. In addition to static tests at room temperature and hot-humid conditions, fatigue tests are also done on the coupon level. Additional static and fatigue tests were done on complex structures such as spar-beams, fuselages and full-scale wing structures. However, for each type-certificate, only static tests should be performed in full-scale. This concept is determined by the certification memorandum CM-S-006 “Composite Lightweight Aircraft” 2017. Es werden die Möglichkeiten eines speziell für diesen Zweck an der BAM entwickelten Prüfsystems mit einer maximalen Tragkraft von 2 MN aufgezeigt. Durch die konstruktive Gestaltung der Anlage lassen sich in Schweißversuchen schweißbedingte Beanspruchungen nachbilden und die komplexen Einflüsse und Wechselwirkungen durch Schweißprozess, Bauteilgeometrie und -konstruktion sowie durch die eingesetzten Grund- und Zusatzwerkstoffe quantifizieren. The operational temperatures and pressures required today can only be achieved by using new creep-resistant grades of steel. The modified 13CrMoV9-10 vanadium steel shows a good resistance against creep and compressed hydrogen and has been in use for the construction of petrochemical reactors since the mid-1990s. Nevertheless, processing of this type of steel requires extreme care during the welding procedure. This is due to its low toughness and high strength in the welded state when not post weld heat treated combined with increased susceptibility to cracking during stress relaxation. Previous research into crack formation in creep-resistant steels has largely focused on thermal and metallurgical factors; however, little knowledge has been gathered regarding the influence of the welding procedure on crack formation during post weld heat treatment considering real-life manufacturing conditions. The influence of heat control on the mechanical properties has been investigated by simulating the welding and subsequent post weld heat treatment operations during the construction of petrochemical reactors using a special 3-D testing facility on the laboratory scale. This work is subdivided in two parts. In part I of this study the stresses resulting from preheating, welding, dehydrogenation heat treatment and the final post weld heat treatment were analyzed during experiments under varied heat control. In all experiments stress relief cracks formed during post weld heat treatment could be observed. The total crack lengths correlated with the welding induced stresses. Part II of this work is dedicated to the characterization of the cracks and the microstructure. The application of a special acoustic emission analysis indicated that the cracks formed in a temperature range between 300 °C and 500 °C during the post weld heat treatment. In comparison to small scale specimens welded without additional shrinkage restraint, the toughness of the restrained welds was significantly decreased. SEM and TEM analyses of all samples revealed accelerated aging due to early precipitation of special carbides during post weld heat treatment under component relevant restraint.
