Because of their frequent application in consumer products, the assessment of their toxicological potential has seen a renewed importance. A Major difficulty is the traceability of nanoparticles in in vitro and in vivo experiments. Even if the particles are labeled, for example, by a fluorescent marker, the dynamic exchange of ligands often prohibits their spatial localization. Our study provides an insight into the adsorption and desorption kinetics of two different fluorescent labels on silver nanoparticles with a core radius of 3 nm by dynamic light scattering, small-angle X-ray scattering, and fluorescence spectroscopy. We used BSA-FITC and tyrosine as examples for common fluorescent ligands. It is shown that the adsorption of BSA-FITC takes at least 3 days, whereas tyrosine adsorbs immediately. The quantitative amount of stabilizer on the particle surface was determined by fluorescence spectroscopy and revealed that the particles are stabilized by a monolayer of BSA-FITC (corresponding to 20 ± 9 molecules), whereas tyrosine forms a multilayered structure consisting of 15900 ± 200 molecules. Desorption experiments show that the BSA-FITC-stabilized particles are ideally suited for application in in vitro and in vivo experiments because the ligand desorption takes several days. Depending on the BSA concentration in the particles surroundings, the rate constant is k = 0.2 per day or lower when applying first order kinetics, that is, 50% of the BSAFITC molecules are released from the particle’s surface within 3.4 days. For illustration, we provide a first application of the fluorescence-labeled particles in an uptake study with two different commonly used cell lines, the human liver cell model HepG2 and the human intestinal cell model of differentiated Caco-2 cells. The EU/FP7 research project NanoDefine has been exclusively dedicated to support the implementation of this EC Recommendation for the European Union’s legislation. For the first time, measurement techniques able to determine the size of nanoparticles have been evaluated systematically on a well-defined set of quality control materials (spherical, monodisperse) as well as industrial materials of complex shapes and considerable polydispersity. Particularly based on the analytical performance of the measurement techniques as tested on the challenging real-world particulate materials, it was possible to formulate recommendations for use of a new tiered approach consisting of screening and confirmatory techniques. Thus, a consistent framework of guidance for nanomaterial identification according to the EC Definition has been issued in form of the NanoDefiner e-tool, with a transparent decision flow scheme and an extensive user manual. Selected examples of analysis and classification as nano-/non-nanomaterials will be given, highlighting the limits of applicability of the available measurement techniques in dependence on sample properties. On some recent relevant developments in sample preparation – as a crucial part for an accurate analysis - will be also reported. Further, ISO/TC 229 ‘Nanotechnologies’ activities aiming at establishing accurate TEM and SEM measurement of NP size and shape as robust, traceable, standard procedures are highlighted. With participation of BAM, study groups have been organizing inter-laboratory comparisons on well-selected NP systems according to the market needs, such as aggregated or shape-controlled titania nano-powder for which size and shape distribution of primary particles must be measured accurately. Die Verwendung ultrakurzer Laserimpulse mit Dauern im Femtosekundenbereich ermöglicht dabei neben einer herausragenden Bearbeitungs-präzision auch die selbstorganisierte Erzeugung verschiedener charakteristischer Ober-flächenstrukturen mit Größenskalen im Mikrometer- bis hinunter in den sub-100-nm-Bereich, z.B. sogenannte Ripples („Laser-Induced Periodic Surface Structures“, LIPSS), Grooves, oder Spikes. In dem Vortrag wird ein Überblick über die in den vergangenen Jahren in Zusammenarbeit mit dem BAM Fachbereich 6.3 durchgeführten tribologischen Experimente gegeben. Besonderes Augenmerk liegt dabei auf den tribologischen Eigenschaften (Reibung und Verschleiß) der unterschiedlichen Femtosekunden-Laser-generierten Oberflächen-morphologien auf gängigen Metallen (z.B. Stahl, Titan).
