Geomorphological processes in (pre)history can have resulted in a modification of the local relief around archaeological sites, especially in areas that are prone to sediment erosion and relocation, so that models of past surfaces are necessary for a correct interpretation of archaeological survey results. In this study, we combined terrestrial survey and remote sensing methods to reconstruct the palaeorelief around two archaeological excavation sites in an inland dune area in southern Brandenburg, Germany. Remains from two Mesolithic hunter-gatherer camps were documented in archaeological excavations and found to be associated with a buried soil horizon. To gather information on the relief of the buried soil surface, we used a combination of sedimentological and pedological profile description of archaeological survey trenches and geophysical prospection with Ground-Penetrating Radar; supplemented by microdrone photography and photogrammetry, GPS surveys and analysis of LIDAR-based elevation models. A digital elevation model of the buried surface was created by combining point data from these sources. The buried surface morphology and the position of the archeological remains within the reconstructed landscape were analyzed in GIS. The comparison of the generated paleosurface model with the recent surface elevation model shows that sand remobilization resulted in a considerable reshaping of the relief. Analysis of the buried surface model further shows that the relief position of the two archaeological sites in the study area was considerably more prominent in relation to the corresponding buried soil surface than in relation to the recent surface morphology. Results affirm the importance of considering Holocene relief modifications in archaeological surveys. We aimed to establish protocols for the detection of KCNJ3 in formalin-fixed, paraffin-embedded (FFPE) breast cancer tissue. Several antibodies were tested for sensitivity and specificity by western blot, followed by optimisation of the immunohistochemistry (IHC) procedure and establishment of KCNJ3 mRNA in situ hybridisation (ISH). Methods were validated by processing 15 FFPE breast cancer samples for which microarray data were available. Spearman's rank correlation analysis resulted in borderline significant correlation for IHC versus ISH (rS: 0.625; p<0.05) and IHC versus microarray (rS: 0.668; p<0.01), but in significant correlation for ISH versus microarray (rS: 0.861; p<0.001). The ISH method was superior to IHC, regarding robustness, sensitivity and specificity and will aid to further study expression levels of KCNJ3 in both malignant and physiological conditions. Amongst these, the potassium channel gene KCNJ3 (encoding for GIRK1 proteins) has been reported to be upregulated in tumors of patients with breast cancer and to correlate with positive lymph node status. We aimed to study KCNJ3 levels in different breast cancer subtypes using gene expression data from the TCGA, to validate our findings using RNA in situ hybridization in a validation cohort (GEO ID GSE17705), and to study the prognostic value of KCNJ3using survival analysis. In a total of > 1000 breast cancer patients of two independent data sets we showed a) that KCNJ3 expression is upregulated in tumor tissue compared to corresponding normal tissue (p < 0.001), b) that KCNJ3 expression is associated with estrogen receptor (ER) positive tumors (p < 0.001), but that KCNJ3 expression is variable within this group, and c) that ER positive patients with high KCNJ3 levels have worse overall (p < 0.05) and disease free survival probabilities (p < 0.01), whereby KCNJ3 is an independent prognostic factor (p <0.05). In conclusion, our data suggest that patients with ER positive breast cancer might be stratified into high risk and low risk groups based on the KCNJ3 levels in the tumor. Escherichia coli carries a broad range of virulence-associated genes (VAGs) which contribute to intestinal (inVAGs) and extraintestinal (exVAGs) infection. Moreover, initial evidence indicates that inVAGs and exVAGs support intestinal colonization. We developed new screening tools to genotypically and phenotypically characterize E. coli isolates originating in humans, domestic pigs, and 17 wild mammal and avian species. We analyzed 317 isolates for the occurrence of 44 VAGs using a novel multiplex PCR microbead assay (MPMA) and for adhesion to four epithelial cell lines using a new adhesion assay. We correlated data for the definition of new adhesion genes. inVAGs were identified only sporadically, particularly in roe deer (Capreolus capreolus) and the European hedgehog ( Erinaceus europaeus). The prevalence of exVAGs depended on isolation from a specific host. Human uropathogenic E. coli isolates carried exVAGs with the highest prevalence, followed by badger (Meles meles) and roe deer isolates. Adhesion was found to be very diverse. Adhesion was specific to cells, host, and tissue, though it was also unspecific. Occurrence of the following VAGs was associated with a higher rate of adhesion to one or more cell lines: afa-dra, daaD, tsh, vat, ibeA, fyuA, mat, sfa-foc, malX, pic, irp2, and papC. In summary, we established new screening methods which enabled us to characterize large numbers of E. coli isolates. We defined reservoirs for potential pathogenic E. coli. We also identified a very broad range of colonization strategies and defined potential new adhesion genes. Little is known about adhesion patterns and host- and tissue adaption of commensal E. coli and about E. coli originating in clinically healthy hosts carrying pathotype-specific virulence-associated genes. FINDINGS: Adhesion pattern of E. coli (n = 282) from humans and from 18 animal species were verified on intestinal human Caco-2 and porcine IPEC-J2 cells and, furthermore, for comparison on human urinary bladder 5637, porcine kidney PK-15 epithelial and HEp-2 cells. The analysis was carried out on 150,000 images of adhesion assays.Adhesion patterns were very diverse; 88 isolates were completely non-adherent, whereas 194 adhered to at least one cell line with the dominant adhesion patterns "diffusely distributed" and "microcolony formation". Adhesion patterns "chains" and "clumps" were also visible. Chain formation was mediated by the presence of epithelial cells. Clump formation was very specific on only the 5637 cell line. All enteropathogenic (eae+) E. coli (EPEC; n = 14) were able to form microcolonies which was cell line specific for each isolate. Most EPEC formed microcolonies on intestinal IPEC-J2 and Caco-2 but several also on urinary tract cells. Shigatoxin-producing (stx+) E. coli (n = 10) showed no specific adhesion patterns. CONCLUSIONS: E. coli isolates were highly diverse. Commensal and pathogenic isolates can adhere in various forms, including diffuse distribution, microcolonies, chains and clumps. Microcolony formation seems to be a global adhesion strategy also for commensal E. coli. The superior objective of the research project is to contribute to a reliable but simultaneously less conservative design of future high pressure blade integrated disks (blisk). It is planned to achieve trend-setting advances based on a close combination of both numerical and experimental analyses. The analyses are focused on the second rotor of this research compressor, which is the only one being manufactured as blisk. The comprehensive test program is addressing both surge and forced response analyses e.g. caused by low engine order excitation. Among others the interaction of aeroelastics and blade mistuning is demanding attention in this regard. That is why structural models are needed, allowing for an accurate forced response prediction close to reality. Furthermore, these models are required to support the assessment of blade tip timing (BTT) data gathered in the rig tests and strain gauge (s/g) data as well. To gain the maximum information regarding the correlation between BTT data, s/g-data and pressure gauge data, every blade of the second stage rotor (28 blades) is applied with s/g. However, it is well known that s/g on blades can contribute additional mistuning that had to be considered upon updating structural models. Due to the relevance of mistuning, efforts are made for its accurate experimental determination. Blade-by-blade impact tests according to a patented approach are used for this purpose. From the research point of view, it is most interesting to determine both the effect s/g-instrumentation and assembling the compressor stages on blade frequency mistuning. That is why experimental mistuning tests carried out immediately after manufacturing the blisk are repeated twice, namely, after s/g instrumentation and after assembling. To complete the pre-test program, the pure mechanical damping and modal damping ratios dependent on the ambient pressure are experimentally determined inside a pressure vessel. Subsequently the mistuning data gained before is used for updating subset of nominal system mode (SNM) models. Aerodynamic influence coefficients (AICs) are implemented to take aeroelastic interaction into account for forced response analyses. Within a comparison of different models, it is shown for the fundamental flap mode (1F) that the s/g instrumentation significantly affects the forced response, whereas the impact of assembling the compressor plays a minor role. Kolloquium Fördertechnikim Bergbau - Tagungsband N2 - Während im Tagebau früher das Augenmerk primär auf einen zuverlässigen und wirtschaftlichen Abbau und Transport von Rohstoffen gerichtet war, spielen heutzutage noch einige andere Aspekte eine entscheidende Rolle. Da die Abbaugebiete immer weiter in besiedelte Gegenden vordringen und die Vorgaben im Umweltschutz sich verschärfen, ist der Betreiber in zunehmendem Maße bestrebt, unter anderem die Geräuschemissionen der Tagebaugroßgeräte zu senken. Da sich speziell Gurtförderanlagen im Bereich des Vorschnitts über große Entfernungen erstrecken und an den Rändern der Tagebaue verlaufen, tragen diese zu einem großen Teil zur Geräuschemission bei. Tragrollen sind oft zu Tausenden in den Förderanlagen verbaut und bei der akustischen Wirkung von entscheidender Bedeutung. Um eine Gesundheitsgefährdung für die beschäftigten Mitarbeiter durch Lärm zu vermeiden und die Auswirkungen auf angrenzende Siedlungen auf ein Mindestmaß zu begrenzen, existieren gesetzliche Vorschriften und Richtlinien. Die Gesetze und Verordnungen zum Umweltschutz und Arbeitsschutz sollen eine Reduzierung der Lärmimmissionen erreichen. Laut den Immissionsrichtwerten1 der Technischen Anleitung zum Schutz gegen Lärm (TA Lärm) darf nachts in Kerngebieten, Dorf- und Mischgebieten der Beurteilungspegel von 45 dB(A) nicht überschritten werden, in reinen Wohngebieten liegt die Grenze sogar bei 35 dB(A). Zum Vergleich: Eine leise Unterhaltung oder leise Musik wird mit einem Schalldruckpegel von 40 dB(A) angegeben. Der durch den Betrieb der Gurtförderanlagen entstehende meterbezogene Schallleistungspegel LW beträgt im Durchschnitt etwa 80 bis 100 dB(A). Durch Reduktion der Schallemission könnten die Planungen der Streckenverläufe optimiert werden, was zu wirtschaftlichen Einsparungen führt. Weiterhin ist, wenn die für Tagebauanlagen zu beachtenden Richtwerte eingehalten werden, auch die Genehmigung für die Einrichtung und den Betrieb solcher Anlagen einfacher abzusichern. Um genaue Kenntnisse über die Schallemission von Tragrollen zu erlangen, wurde in Zusammenarbeit der ehemaligen VEM AG (jetzt LEAG) und der ehemaligen Hochschule Lausitz (jetzt BTU Cottbus-Senftenberg) ein Tragrollenprüfstand entwickelt. Im Rahmen einer Baugruppenprüfung auf diesem Prüfstand werden akustische und mechanische Kenngrößen von Tragrollen untersucht. Bestandteile dieser Untersuchungen sind: - Hochfahrprüfläufe – Dabei wird eine Tragrolle unter Prüflast auf eine Umfangsgeschwindigkeit von 10 m/s beschleunigt und der Schalldruckpegel aufgezeichnet. Dies ermöglicht die Berechnung der Schallemission in Abhängigkeit von der Umfangsgeschwindigkeit. - Ermittlung des Abklingverhaltens – Durch Anschlagversuche wird das Eigenschwing- und Dämpfungsverhalten einer Tragrolle erfasst. - Erfassung der Tragrollengeometrie – Die präzise Beschreibung der Geometrie des Tragrollenmantels ermöglicht Rückschlüsse auf mögliche Anregungsursachen. Methods A group of 18 patients with TGA performed an established real-space navigation paradigm, requiring allo- and egocentric spatial orientation abilities, 3 days (postacute stage) and 3 months (follow-up) after symptom onset. Visual exploration behavior and navigation strategy were documented by a gaze-controlled, head-fixed camera. Allo- and egocentric spatial orientation performance was compared to that of 12 age-matched healthy controls. Navigation-induced brain activations were measured using [18F]-fluorodeoxyglucose-PET in a subgroup of 8 patients in the postacute stage and compared to those of the controls. Results In the postacute stage, the patients navigated worse and had higher error rates than controls in allocentric (p = 0.002), but not in egocentric, route planning (p = 0.30), despite complete recovery of verbal (p = 0.58) and figural memory (p = 0.11). Until follow-up, allocentric navigation deficits improved, but higher error rates and reduced use of shortcuts persisted (p < 0.0001). Patients still exhibited relatively more fixations of unique landmarks during follow-up (p = 0.05). PET measurements during the postacute stage showed increased navigation-induced brain activations in the right hippocampus, bilateral retrosplenial, parietal, and mesiofrontal cortices, and cerebellar dentate nucleus in patients compared to controls (p < 0.005). Conclusions Patients with TGA show selective and prolonged deficits of allocentric spatial orientation. Activations in right hippocampal and extrahippocampal hubs of the cerebral navigation network functionally substitute for the deficit in creating and updating the internal cognitive map in TGA. Objective: Melting behavior and autofluorescence of materials are decisive for optimal printing and applicability of the product which are influenced by varying unknown additives. Methods: We have produced devices for bioanalytical applications from commercially available thermoplastic polymers using a melt-layer process. We characterized them by differential scanning calorimetry, fluorescence spectroscopy and functional assays (DNA capture assay, model for cell adhesion, bacterial adhesion and biofilm formation test). Results: From 14 tested colored, transparent and black materials we found only deep black acrylonitrile-butadiene-styrene (ABS) and some black polylactic acid (PLA) useable for fluorescence-based assays, with low autofluorescence only in the short-wave range of 300-400 nm. PLA was suitable for standard bioanalytical purposes due to a glass transition temperature of approximately 60°C, resistance to common laboratory chemicals and easy print processing. For temperature-critical methods, such as hybridization reactions up to 90°C, ABS was better suited. Conclusions: Autofluorescence was not a disadvantage per se but can also be used as a reference signal in assays. The rapid development of individual protocols for sample processing and analysis required the availability of a material with consistent quality over time. For fluorescence-based assays, the use of commercial standard materials did not seem to meet this requirement. A recent hypothesis based on the predictive coding theory of brain function suggests that in functional disorders, internal expectations do not match the actual sensory body states, leading to perceptual dysregulation and symptom perception. To test this hypothesis, we investigated the account of internal expectations and sensory input on gaze stabilization, a physiologically relevant parameter of gaze shifts, in functional dizziness. Methods: We assessed gaze stabilization in eight functional dizziness patients and 11 healthy controls during two distinct epochs of large gaze shifts: during a counter- rotation epoch (CR epoch), where the brain can use internal models, motor planning, and resulting internal expectations to achieve internally driven gaze stabilization; and during an oscillation epoch (OSC epoch), where, due to terminated motor planning, no movement expectations are present, and gaze is stabilized by sensory input alone. Results: Gaze stabilization differed between functional patients and healthy controls only when internal movement expectations were involved [F(1,17) = 14.63, p = 0.001, and partial η2 = 0.463]: functional dizziness patients showed reduced gaze stabilization during the CR (p = 0.036) but not OSC epoch (p = 0.26). Conclusion: While sensory-driven gaze stabilization is intact, there are marked, well- measurable deficits in internally-driven gaze stabilization in functional dizziness pointing at internal expectations that do not match actual body states. This experimental evidence supports the perceptual dysregulation hypothesis of functional disorders and is an important step toward understanding the underlying pathophysiology. However, small-scale changes of sediments and other soil forming factors can result in high spatial variation of soil properties in these landscapes; and relocation of soil material by geomorphic processes further increases the spatial heterogeneity of the soil landscape. These spatial variations in soil properties and conservation need to be considered in order to correctly decipher and interpret the buried soils as records of past environmental conditions. In the forefield of the open-cast mine Cottbus-Nord, archaeological excavations in a dune and drift sand area revealed widespread buried soils of different characteristics. The densely spaced excavation trenches give exceptionally good insights into the pedosphere, allow for reconstructing the distribution of fossil and recent soils in a high spatial resolution, and offer good opportunities to improve the understanding of spatial and temporal patterns of soil formation. Remains of postpleistocene hunter-gatherer campsites were documented in archaeological excavations and found to be associated with a buried soil horizon. We recorded the stratigraphy of soil profiles along more than 15 trenches within an area of about 20 ha, and additionally described the position of buried soils from GPR surveys and microdrone photogrammetry. In a recently opened, 455 m long trench, all characteristic soils and sediments of the study region could be observed and stratigraphically connected. Within this trench, four profiles were identified for further research. Each profile was classified and described according to WRB and German Guidelines for Soil Mapping; and was sampled and investigated through laboratory analyses including determination of organic matter, total carbon and nitrogen contents, soil colour identification, texture analysis, magnetic susceptibility measurement, metal content analysis using X-ray fluorescence analyzer and pH measurement. The compaction and cementation of soil horizons was assessed in situ with a pocket penetrometer. Results show a small-scale mosaic of soils developed on fluvio-aeolian, limnic and aeolian sediments, high spatial variations in pedogenesis due to varying groundwater influence and intensity of horizontal and vertical leachate transport, and high variations in the erosion or conservation of fossil soils. The soil and sediment stratigraphy reflects several phases of landscape development: i) the formation of a Late Pleistocene soil on fluvio-aeolian deposits, ii) a fossilization by aeolian sands, iii) a stability phase with intensive podsolization and peat formation, and iv) a land use-induced aeolian remobilization of the sands. Most representatives of the plants have evolved to be highly energy-efficient in their survival strategies, despite their limited resources. Plants have to deal with a variety of threats originating from different herbivores and microbial pathogens while handling a number of abiotic stress factors. For plants and WSNs, different defence mechanisms impose different fitness costs and therefore the costs and benefits have to be well-balanced. This way, plants are able to not only conquer a large variety of different attacks but also handle several different simultaneous attacks adequately. To react early and adapt to upcoming threats, plants within a network are capable of signalling each other within a certain distance about ongoing attacks. The biological strategies of plants in terms of signalling and defence, as well as their energy and resource limitations, share significant similarities with WSNs. This study aims to explore these similarities and demonstrates the correspondence between the biological concepts and WSNs. Additionally, we present a novel approach to enhancing security in WSNs through a cooperative distributed security scheme inspired by key factors of plant defence mechanisms, with the goal of reducing the energy constraints and maximising the longevity of the network.
