Energieforschungsprogramms. Im Projekt MAGGIE sollten beispielhaft für das genossenschaftliche historische Stadtquartier „Margaretenau“ in Regensburg Musterlösungen für energieoptimiertes Wohnen mit innovativen Wandauauten aus solaraktiven Baustoen und einer vorhersagebasierten Versorgungstechnologie entwickelt werden. Für ein bestehendes Wohngebäude als Demonstrationsund Versuchsobjekt wurde beispielhaft ein neuartiges, besonders ezientes Hybridsystem aus Wärmepumpentechnologie und Kraft-Wärme-Kopplung (Blockheizkraftwerk) konzipiert und umgesetzt. Für die denkmalgerechte Modernisierung historischer Fassaden wurde ein solaraktives und solaradaptives Außenputzsystem entwickelt und getestet. Der erhöhte Ausnutzungsgrad solarer Gewinne in Verbindung mit einer verbesserten thermischen Behaglichkeit im Gebäudeinnerendurch innovative Innenputz-/Innenfarbsysteme kann maßgeblich zur Einsparung von Heizwärme beitragen. Dazu wurden mit Blick auf die bauphysikalischen und wärmetechnischen Eigenschaften Versuche an einem eigens aufgebauten Wandprüfstand durchgeführt, ergänzt durch bauphysikalische Modellsimulationen. Die Ergebnisse sind vielversprechend, auch wenn eine serientaugliche Lösung für den Einsatz am Gebäude im Rahmen des Projektes nicht entstehen konnte. Das wichtigste Ziel des Projektes war es, ein hochezientes, skalierbares Versorgungssystem durch die Kombination von Kraft-Wärme-Kopplung (Blockheizkraftwerk, BHKW) mit einer Pvunterstützten Wärmepumpen-Technologie zu entwickeln und im Detail zu modellieren, in Verbindung mit einer innovativenWärmelogistik zur Reduktion der Verteilungsverluste für dieWarm-wasserversorgung. Das Systemsollte neben einem hohen Eigenstrom-Nutzungsanteil auch netzdienliche Schwarmfunktionalität aufweisen. Die Skalierbarkeit des vernetzten Systems („Mitwachsen“) erlaubt eine abschnittsweise Modernisierung des gesamten Ensembles über mehrere Jahre, was für die Finanzierbarkeit und praktische Umsetzung erhebliche Bedeutung hat. Durch die hohe Energieezienz wird die Warmmiete für die Bewohner der genossenschaftlichen Siedlung durch die Modernisierungsmaßnahmen nicht erhöht, so dass auch nach der Modernisierung ein bezahlbares Wohnen sichergestellt wird. Die Auslegung des Systems erfolgte durch ein neu entwickeltes, generisch einsetzbares Planungs-, Optimierungs- und Steuerungstool, das auch die Regelung im Betrieb übernimmt. Die in Echtzeit laufende Nachoptimierung des Systems während des Betriebs greift auf reale Monitoringdaten zu. Die Einbindung von Nutzerbedarfsprolen, Strombörse und Wetterdaten in die Steuerung ist vorbereitet und gestattet einen dynamischen und perspektivischen Anlagenbetrieb zur Maximierung der Solar- und Umweltwärme-Anteile der Energieversorgung. Mithilfe einer intensiven Nutzerinformation in einem partizipativen Planungsprozess wurde die Akzeptanz der experimentellen Maßnahmen sichergestellt. Durch oene Kommunikation der Zielsetzungen und Lösungsansätze für eine klimafreundliche, kostenbewusste Restaurierung des Wohnungsbestands und die aktive Einbindung der Bewohner fand das Projekt das Interesse und die Unterstützung der Menschen, auch in der schwierigen Phase der Umsetzung baulicher Maßnahmen in einem zwangsläug in Abschnitten vorübergehend entmieteten Gebäude. Here we show that the four peroxygenases AaeUPO, MroUPO, rCciUPO and rNOVO catalyze the stepwise hydroxylation of cyclohexane to cyclohexanol and cyclohexanone. The catalytic efficiencies (kcat/Km) for the initial hydroxylation were in the same order of magnitude for all four peroxygenases (∼104 M−1 s−1), whereas they differed in the second step. The conversion of cyclohexanol by AaeUPO and rCciUPO was 1–2 orders of magnitude less efficient (∼102 M−1 s−1) than by MroUPO and rNOVO (∼104 M−1 s−1). The highest conversion rate in terms of H2O2 utilization was accomplished by MroUPO under repeated addition of the peroxide (87% in relation to the total products formed). Using the latter UPO, we successfully established a micro-mixing reaction device (SIMM-V2) for the oxidation of cyclohexane. As cyclohexanone is a chemical of high relevance, for example, as starting material for polymer syntheses or as organic solvent, new enzymatic production pathways for this compound are of interest to complement existing chemical and biotechnological approaches. Stable and versatile peroxygenases, as those presented here, may form a promising biocatalytic platform for the development of such enzyme-based processes. These allow high-quality reconstructions from sparse known data. While they have been explicitly developed with compression in mind, they have not entered actual codecs so far: Storing these optimised data efficiently is a nontrivial task. Since this step is essential for any competetive codec, we propose two new compression frameworks for linear PDEs: Efficient storage of pixel locations obtained from an optimal control approach, and a stochastic strategy for a locally adaptive, tree-based grid. Suprisingly, our experiments show that homogeneous diffusion inpainting can surpass its often favoured biharmonic counterpart in compression. Last but not least, we demonstrate that our linear approach is able to beat both JPEG2000 and the nonlinear state-of-the-art in PDE-based image compression. Here we report, for the first time, two effective methods for the immobilization of unspecific peroxygenase (UPO; EC 1.11.2.1). This biocatalyst type comprises heavily glycosylated heme-thiolate proteins that catalyze various biotechnologically relevant oxyfunctionalizations. Both the encapsulation in cryogel and the retention of the enzyme in hollow fiber modules were found to be efficient methods for their immobilization. After encapsulation, the enzyme still exhibited 60% of its initial activity. Interestingly, we did not find differences in the kinetic parameters of free and immobilized UPOs. In long-term experiments, the conversion of the pharmaceutical diclofenac with immobilized UPOs in different reactor types yielded between 62 mg and 154 mg of the major human drug metabolite 4′-hydroxydiclofenac. The maximal total turnover number was about 60-fold higher compared to the free enzyme. A test over 5 months showed that storage of encapsulated UPOs in non-polar solvents (e.g., cyclohexane) helps to preserve the enzyme stability and increases their relative activity (by about ∼150%, in the case of diclofenac hydroxylation). In addition to the hydrophilic substrate diclofenac, encapsulated UPOs also oxidized the hydrophobic model compound cyclohexane. By contrast, the small-scale and heterogeneous occurrence of small-scale fog (hundreds of meters) particularly during summers is widely unconsidered. However, these events are important for the local vegetation and particularly for the biological soil crusts (BSC) that are widely distributed in this extreme ecosystem. Consequently, a case study in a typical fog oasis in the Pan de Azúcar National Park was conducted to test the feasibility combining field measurements, drone profiling, remote sensing and numerical modeling (i) to investigate fog-type specific differences regarding dynamics, physical properties and formation, (ii) to test the applicability of remote sensing technology for fog monitoring based on existing low-resolution and a proposed new high-resolution product and (iii) to estimate the related fog water input to BSCs. Two types of fog were observed. The well-known fog/LST deck emerging from the Pacific Ocean with high water path and large spatial extent was the first type. Fog of the second type was patchier, small-scale and not necessarily connected to the LST over the ocean. Instead, fog formation of the second type was related to thermal breeze systems, which produced shallow clouds containing less water than those of type 1. In general, such small-scale fog events were not captured well by existing remote sensing products but could be detected with the proposed new high-resolution product which provided promising results. Both fog types were important water resources for the BSCs, with approximately 8% to 24% of the fog water flux available to the BSCs at the surface. The results indicated the feasibility of the proposed methods’ pool to estimate the water budget of BSCs with a high spatial resolution in the future. However, whether there is synchronisation of these strategies at the entire community level is unclear. Here, we combine trait data for 2800 above- and belowground taxa from 14 trophic guilds spanning a disturbance and resource availability gradient in German grasslands. The results indicate that most guilds consistently respond to these drivers through both direct and trophically mediated effects, resulting in a ‘slow-fast’ axis at the level of the entire community. Using 15 indicators of carbon and nutrient fluxes, biomass production and decomposition, we also show that fast trait communities are associated with faster rates of ecosystem functioning. These findings demonstrate that ‘slow’ and ‘fast’ strategies can be manifested at the level of whole communities, opening new avenues of ecosystem-level functional classification.
