Für die mittelständischen Tankstellen können sich Schnellladestationen zu einem neuen, ergänzenden Geschäftsmodell entwickeln, sofern die Standorte geeignet sind. Durch eine längere Aufenthaltszeit während des Ladevorgangs bieten sich angepasste Waren- und Dienstleistungsangebote zur Überbrückung an. Heme peroxidases comprise the classical ligninolytic peroxidases and the new dye-decolorizing peroxidases, while heme peroxygenases belong to a still largely unexplored superfamily of heme-thiolate proteins. Nevertheless, basidiomycete unspecific peroxygenases have the highest biotechnological interest due to their ability to catalyze a variety of regio- and stereo-selective monooxygenation reactions with H2O2 as the source of oxygen and final electron acceptor. Flavo-oxidases are involved in both lignin and cellulose decay generating H2O2 that activates peroxidases and generates hydroxyl radical. The group of copper oxidoreductases also includes other H2O2 generating enzymes - copper-radical oxidases - together with classical laccases that are the oxidoreductases with the largest number of reported applications to date. However, the recently described lytic polysaccharide monooxygenases have attracted the highest attention among copper oxidoreductases, since they are capable of oxidatively breaking down crystalline cellulose, the disintegration of which is still a major bottleneck in lignocellulose biorefineries, along with lignin degradation. Interestingly, some flavin-containing dehydrogenases also play a key role in cellulose breakdown by directly/indirectly “fueling” electrons for polysaccharide monooxygenase activation. Many of the above oxidoreductases have been engineered, combining rational and computational design with directed evolution, to attain the selectivity, catalytic efficiency and stability properties required for their industrial utilization. Indeed, using ad hoc software and current computational capabilities, it is now possible to predict substrate access to the active site in biophysical simulations, and electron transfer efficiency in biochemical simulations, reducing in orders of magnitude the time of experimental work in oxidoreductase screening and engineering. What has been set out above is illustrated by a series of remarkable oxyfunctionalization and oxidation reactions developed in the frame of an intersectorial and multidisciplinary European RTD project. The optimized reactions include enzymatic synthesis of 1-naphthol, 25-hydroxyvitamin D3, drug metabolites, furandicarboxylic acid, indigo and other dyes, and conductive polyaniline, terminal oxygenation of alkanes, biomass delignification and lignin oxidation, among others. These successful case stories demonstrate the unexploited potential of oxidoreductases in medium and large-scale biotransformations. The polymer-immobilized enzymes were used for water treatment to decompose persistent organic molecules. Introduction of suitable comonomers allows tailoring the mechanical and chemical properties for special applications. Copolymers containing MMA and itaconic anhydride were chosen because of the formation of long-term stable anhydride functionalities. These anhydride functionalities are employed to attach enzymes covalently. 4-Benzoylphenyl methacrylate is used as comonomer for UV-initiated crosslinking. Terpolymers are successfully obtained by radical copolymerization in solution. The copolymers are compared to poly(ethylene-alt-maleic anhydride) [P(EMA)] often used with respect to enzyme immobilization, activity and hydrolytic stability. The hydrolysis stability of the copolymers against water is studied by ATR-FTIR spectroscopy. Thin films are prepared on glass substrates in a layer-by-layer procedure by spin-coating. The layer formation is monitored by ATR-FTIR spectroscopy. UV-crosslinking of the copolymer films is performed taking the optimal irradiation dose that avoids polymer degradation. ATR-FTIR spectroscopy verifies the coupling reaction between amino groups of the enzyme and the anhydride groups on the surface of the crosslinked polymer film. The syringaldazine (4-hydroxy-3,5-dimethoxybenzaldehyde azine) test and 2,2´-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) assay demonstrate that the immobilized enzymes maintain their activities. The functional copolymers showed a significant effect in reduction of persistent organic pollutants in contaminated waste water. However, disparities in initial conditions pose challenges when trying to implement these sustainable changes. Identifying these differences aids in the comprehension of future developments. In this study, we establish an international comparison by decoding the mobility-related characteristics of cities and determining urban archetypes. Using publicly accessible data, we analyze and classify 96 cities in different countries. Therefore, we utilize principal component analysis to simplify the data. The emerging components serve as input for segmentation. This approach yields nine unique urban archetypes, ranging from Well-Functioning and Ancient Hybrid Cities in Europe to Paratransit and Traffic-Saturated Cities in the southern hemisphere. Our results show that there is a significant advantage to using a multidimensional segmentation basis, which we identify in an extensive literature review. The result is a finer segmentation, which is especially clear for European cities that demonstrate four different clusters. We discuss that the effect of future restrictions on private car usage will vary widely between the urban archetypes. These 3D culture systems are particularly suitable for application in regenerative medicine or experimental pharmacology and toxicology. Therefore, it is important to analyse the cells in their 3D microenvironment with regard to viability and differentiation. Tetrazolium assays (WST-8 and MTS) are still the methods of choice for estimating the number of living, metabolically active cells, with WST-8 being cell-impermeable compared to MTS. In contrast to these methods, the ATP assay is an endpoint method based on the luciferase-induced reaction of ATP with luciferin after cell lysis. OBJECTIVE:We compared three methodologically different proliferation/toxicity assays (MTS, WST-8, ATP) in monolayer (2D) and 3D culture systems to improve the technically challenging determination of the number of viable cells. METHODS: Chondrocytes were isolated from human articular cartilage. Three different test systems (MTS, WST-8, ATP) were applied to monolayer cells (2D, varying cell numbers) and spheroids (3D, different sizes) in 96-well plates. The intracellular ATP concentration was determined by luciferase-induced reaction of ATP with luciferin using a luminometer. Formazan formation was measured spectrophotometrically after different incubation periods. Evaluation was performed by phase contrast microscopy (toxicity), correlation of cell count and ATP concentration or absorption signal (Gompertz function) and propidium iodide (PI) staining to proof the cell lysis of all cells in spheroids. RESULTS: In 2D culture, all three assays showed a good correlation between the number of seeded cells and the ATP concentration or absorption data, whereas the MTS-assay showed the lowest specificity. In 3D culture, the spheroid sizes were directly related to the number of cells seeded. The absorption data of the WST-8 and MTS assay correlated only for certain spheroid size ranges, whereas the MTS-assay showed again the lowest specificity. Only the measured intracellular ATP content showed a linear correlation with all spheroid sizes ranging from 100–1000 µm. The WST-8 assay revealed the second-best sensitivity which allows the measurement of spheroids larger than 240 µm. Phase contrast observation of monolayer cells showed toxic effects of MTS after 6 h incubation and no signs of toxicity of WST-8. Staining with propidium iodide showed complete lysis of all cells in a spheroid in the ATP assay. CONCLUSION: Among tetrazolium-based assays, WST-8 is preferable to MTS because of its non-toxicity and better sensitivity. When determining the number of viable cells in the 2D system, caution is advised when using the ATP assay because of its two-phase slope of the correlation graph concerning cell number and intracellular ATP. In 3D systems of human chondrocytes, the ATP-assay is superior to the other two test systems, as the correlation graph between cell number and intracellular ATP is biphasic. Since differentiation processes or other metabolic events can influence the results of proliferation and toxicity assays (determination of viable cells), this should be taken into account when using these test systems. In this study, we have engineered a highly regioselective fungal peroxygenase for the ω-1 hydroxylation of fatty acids with quenched stepwise over-oxidation. One single mutation near the Phe catalytic tripod narrowed the heme cavity, promoting a dramatic shift toward subterminal hydroxylation with a drop in the over-oxidation activity. While crystallographic soaking experiments and molecular dynamic simulations shed light on this unique oxidation pattern, the selective biocatalyst was produced by Pichia pastoris at 0.4 g L−1 in a fed-batch bioreactor and used in the preparative synthesis of 1.4 g of (ω-1) hydroxytetradecanoic acid with 95 % regioselectivity and 83 % ee for the S enantiomer. This study investigates the implementation of a heterogeneous sensor network within an energy-bound forging press, integrating industrial 120 GHz radar sensors for comprehensive data collection. A complete radar signal processing chain is developed, encompassing signal acquisition, pre-processing, and state estimation of the press ram. As it affects the ecological and economic system, conservation costs and impacts may change in a heterogeneous manner. This implies that cost-effective conservation sites and measures may no longer be so in the future. We investigate spatial flexibility (to adapt the location of conservation sites) and management flexibility (to adapt the conservation measures on those sites) as novel criteria for analysing the cost-effectiveness of policy instruments to conserve biodiversity under climate change. We develop a generic climate-ecological-economic modelling approach that captures the role of spatial and management flexibility to assess the cost-effectiveness of policy instruments. We apply the modelling approach to the conservation of an indicator species in agricultural grasslands in a case study area in Northern Germany, and compare the cost-effectiveness of the policy instruments of land purchase (low spatial flexibility, high management flexibility) and long-term individual conservation contracts (medium spatial and management flexibility) considering a period from 2020 to 2079. We find that both spatial and management flexibility matter in the case study and discuss their broader relevance for conservation in a changing climate.
