Here we report, for the first time, two effective methods for the immobilization of unspecific peroxygenase (UPO; EC 1.11.2.1). This biocatalyst type comprises heavily glycosylated heme-thiolate proteins that catalyze various biotechnologically relevant oxyfunctionalizations. Both the encapsulation in cryogel and the retention of the enzyme in hollow fiber modules were found to be efficient methods for their immobilization. After encapsulation, the enzyme still exhibited 60\% of its initial activity. Interestingly, we did not find differences in the kinetic parameters of free and immobilized UPOs. In long-term experiments, the conversion of the pharmaceutical diclofenac with immobilized UPOs in different reactor types yielded between 62 mg and 154 mg of the major human drug metabolite 4′-hydroxydiclofenac. The maximal total turnover number was about 60-fold higher compared to the free enzyme. A test over 5 months showed that storage of encapsulated UPOs in non-polar solvents (e.g., cyclohexane) helps to preserve the enzyme stability and increases their relative activity (by about ∼150\%, in the case of diclofenac hydroxylation). In addition to the hydrophilic substrate diclofenac, encapsulated UPOs also oxidized the hydrophobic model compound cyclohexane.}, language = {en} } @misc{DakshaDerzsiWilczeketal., author = {Daksha, Manaswi and Derzsi, Aranka and Wilczek, Sebastian and Trieschmann, Jan and Mussenbrock, Thomas and Awakowicz, Peter and Donk{\´o}, Zolt{\´a}n and Schulze, Julian}, title = {The Effect of Realistic Heavy Particle Induced Secondary Electron Emission Coefficients on the Electron Power Absorption Dynamics in Single- and Dual-Frequency CCPs}, series = {Plasma Sources Science and Technology}, volume = {26}, journal = {Plasma Sources Science and Technology}, number = {8}, issn = {1361-6595}, doi = {10.1088/1361-6595/aa7c88}, pages = {085006}, language = {en} } @misc{WilczekTrieschmannSchulzeetal., author = {Wilczek, Sebastian and Trieschmann, Jan and Schulze, Julian and Donko, Zoltan and Brinkmann, Ralf Peter and Mussenbrock, Thomas}, title = {Disparity between current and voltage driven capacitively coupled radio frequency discharges}, series = {Plasma Sources Science and Technology}, volume = {27}, journal = {Plasma Sources Science and Technology}, number = {12}, issn = {1361-6595}, doi = {10.1088/1361-6595/aae5c1}, pages = {12}, language = {en} } @misc{KlichWilczekJanssenetal., author = {Klich, Maximilian and Wilczek, Sebastian and Janssen, Jesper Frederikus Jacobus and Brinkmann, Ralf Peter and Mussenbrock, Thomas and Trieschmann, Jan}, title = {Ion dynamics in capacitively coupled argon-xenon discharges}, series = {Plasma Sources Science and Technology}, volume = {30}, journal = {Plasma Sources Science and Technology}, number = {6}, issn = {1361-6595}, doi = {10.1088/1361-6595/ac02b0}, language = {en} } @techreport{ThonickeRahnerArnethetal., author = {Thonicke, Kirsten and Rahner, Eva and Arneth, Almut and Bartkowski, Bartosz and Bonn, Aletta and D{\"o}hler, Christian and Finger, Robert and Freitag, Jens and Grosch, Rita and Grossart, Hans-Peter and Gr{\"u}tzmacher, Kim and Hartman Scholz, Amber and H{\"a}user, Christoph and Hickler, Thomas and H{\"o}lker, Franz and J{\"a}hnig, Sonja C. and Jeschke, Jonathan and Kassen, Rees and Kastner, Thomas and Kramer-Schadt, Stephanie and Krug, Cornelia and Lakner, Sebastian and Loft, Lasse and Matzdorf, Bettina and Meakins, Felicity and De Meester, Luc and Monaghan, Michael T. and M{\"u}ller, Daniel and Overmann, J{\"o}rg and Quaas, Martin and Radchuk, Viktoriia and Reyer, Christopher and Roos, Christian and Scholz, Imme and Schroer, Sibylle and Sioen, Giles Bruno and Sommer, Simone and Sommerwerk, Nike and Tockner, Klement and Turk, Zachary and Warner, Barbara and W{\"a}tzold, Frank and Wende, Wolfgang and Veenstra, Tonjes and Voort, Hein van de}, title = {10 Must-Knows aus der Biodiversit{\"a}tsforschung}, publisher = {Potsdam-Institut f{\"u}r Klimafolgenforschung e.V. By contrast, the small-scale and heterogeneous occurrence of small-scale fog (hundreds of meters) particularly during summers is widely unconsidered. However, these events are important for the local vegetation and particularly for the biological soil crusts (BSC) that are widely distributed in this extreme ecosystem. Consequently, a case study in a typical fog oasis in the Pan de Az{\´u}car National Park was conducted to test the feasibility combining field measurements, drone profiling, remote sensing and numerical modeling (i) to investigate fog-type specific differences regarding dynamics, physical properties and formation, (ii) to test the applicability of remote sensing technology for fog monitoring based on existing low-resolution and a proposed new high-resolution product and (iii) to estimate the related fog water input to BSCs. Two types of fog were observed. The well-known fog/LST deck emerging from the Pacific Ocean with high water path and large spatial extent was the first type. Fog of the second type was patchier, small-scale and not necessarily connected to the LST over the ocean. Instead, fog formation of the second type was related to thermal breeze systems, which produced shallow clouds containing less water than those of type 1. In general, such small-scale fog events were not captured well by existing remote sensing products but could be detected with the proposed new high-resolution product which provided promising results. Both fog types were important water resources for the BSCs, with approximately 8\% to 24\% of the fog water flux available to the BSCs at the surface. The results indicated the feasibility of the proposed methods' pool to estimate the water budget of BSCs with a high spatial resolution in the future.}, language = {en} } @inproceedings{LangendoerferAdam, author = {Langend{\"o}rfer, Peter and Adam, Sebastian}, title = {A Graphical Tool for Specification, Rapid Prototyping and Implementation of Location Based Services}, language = {en} } @misc{WilczekSchulzeBrinkmannetal., author = {Wilczek, Sebastian and Schulze, Julian and Brinkmann, Ralf Peter and Donk{\´o}, Zoltan and Trieschmann, Jan and Mussenbrock, Thomas}, title = {Electron dynamics in low pressure capacitively coupled radio frequency discharges}, series = {Journal of Applied Physics}, volume = {127}, journal = {Journal of Applied Physics}, number = {18}, doi = {10.1063/5.0003114.1}, language = {en} } @misc{OenguenerDittmarMeyeretal., author = {{\"O}ng{\"u}ner, Emir and Dittmar, Mirko and Meyer, Peter and Merbold, Sebastian and Egbers, Christoph}, title = {Cross-sectional PIV analysis in turbulent pipe flow at high Reynolds number}, series = {Experimentelle Str{\"o}mungsmechanik, 24. However, whether there is synchronisation of these strategies at the entire community level is unclear. Here, we combine trait data for 2800 above- and belowground taxa from 14 trophic guilds spanning a disturbance and resource availability gradient in German grasslands. The results indicate that most guilds consistently respond to these drivers through both direct and trophically mediated effects, resulting in a 'slow-fast' axis at the level of the entire community. Using 15 indicators of carbon and nutrient fluxes, biomass production and decomposition, we also show that fast trait communities are associated with faster rates of ecosystem functioning. These findings demonstrate that 'slow' and 'fast' strategies can be manifested at the level of whole communities, opening new avenues of ecosystem-level functional classification.}, language = {en} } @misc{RanjanScholzSemmleretal., author = {Ranjan, Amit and Scholz, Julia and Semmler, Torsten and Wieler, Lothar H. and Ewers, Christa and M{\"u}ller, Stefanie and Pickard, Derek J. and Schierack, Peter and Tedin, Karsten and Ahmed, Niyaz and Schaufler, Katharina and Guenther, Sebastian}, title = {ESBL-plasmid carriage in E. coli enhances in vitro bacterial competition fitness and serum resistance in some strains of pandemic sequence types without overall fitness cost}, series = {Gut pathogens}, volume = {10}, journal = {Gut pathogens}, number = {24}, issn = {1757-4749}, doi = {10.1186/s13099-018-0243-z}, pages = {9}, language = {en} } @misc{MohsinGuentherSchieracketal., author = {Mohsin, Mashkoor and Guenther, Sebastian and Schierack, Peter and Tedin, Karsten and Wieler, Lothar H.}, title = {Probiotic Escherichia coli Nissle 1917 reduces growth, Shiga toxin expression, release and thus cytotoxicity of enterohemorrhagic Escherichia coli}, series = {International Journal of Medical Microbiology}, volume = {305}, journal = {International Journal of Medical Microbiology}, number = {1}, issn = {1618-0607}, doi = {10.1016/j.ijmm.2014.10.003}, pages = {20 -- 26}, language = {en} } @misc{GuentherGrothSchieracketal., author = {Guenther, Sebastian and Groth, Ingrid and Schierack, Peter and Grabley, Susanne and Munder, Thomas}, title = {Direct detection of Kitasatospora species with a chaperone oligonucleotide microarray method lacking PCR amplification}, series = {Journal of basic microbiology}, volume = {48}, journal = {Journal of basic microbiology}, number = {4}, issn = {1521-4028}, doi = {10.1002/jobm.200800038}, pages = {315 -- 318}, language = {en} } @misc{MohsinRazaRoschanskietal., author = {Mohsin, Mashkoor and Raza, Shahbaz and Roschanski, Nicole and Guenther, Sebastian and Ali, Aamir and Schierack, Peter}, title = {Description of the First Escherichia coli Clinical Isolate Harboring the Colistin Resistance Gene mcr-1 from the Indian Subcontinent}, series = {Antimicrobial Agents and Chemotherapy}, volume = {61}, journal = {Antimicrobial Agents and Chemotherapy}, number = {1}, issn = {1098-6596}, doi = {10.1128/AAC.01945-16}, pages = {2}, language = {en} } @misc{ZoellnerZaumseilWilkensetal., author = {Z{\"o}llner, Marvin Hartwig and Zaumseil, Peter and Wilkens, Henrik and Gevers, Sebastian and Wollschl{\"a}ger, Joachim and B{\"a}umer, Marcus and Xie, Ya-Hong and Niu, Gang and Schroeder, Thomas}, title = {Stoichiometry-structure correlation of epitaxial Ce1-xPrxO2-δ (x = 0-1) thin films on Si(111)}, series = {Journal of Crystal Growth}, volume = {355}, journal = {Journal of Crystal Growth}, number = {1}, issn = {0022-0248}, pages = {159 -- 165}, language = {en} } @misc{GuentherSchierackGrobbeletal., author = {Guenther, Sebastian and Schierack, Peter and Grobbel, Mirjam and L{\"u}bke-Becker, Antina and Wieler, Lothar H. and Ewers, Christa}, title = {Real-time PCR assay for the detection of species of the genus Mannheimia.}, series = {Journal of Microbiological Methods}, volume = {75}, journal = {Journal of Microbiological Methods}, number = {1}, issn = {1872-8359}, doi = {10.1016/j.mimet.2008.05.008}, pages = {75 -- 80}, language = {en} } @misc{WillitzkiLorenzHiemannetal., author = {Willitzki, Annika and Lorenz, Sebastian and Hiemann, Rico and Guttek, Karina and Goihl, Alexander and Hartig, Roland and Conrad, Karsten and Feist, Eugen and Sack, Ulrich and Schierack, Peter and Heiserich, Lisa and Eberle, Caroline and Peters, Vanessa and Roggenbuck, Dirk and Reinhold, Dirk}, title = {Fully automated analysis of chemically induced γH2AX foci in human peripheral blood mononuclear cells by indirect immunofluorescence}, series = {Cytometry Part A}, volume = {83}, journal = {Cytometry Part A}, number = {11}, issn = {1552-4930}, doi = {10.1002/cyto.a.22350}, pages = {1017 -- 1026}, language = {en} }
